GUS activities were evaluated in eight replicates of each of eight sugar beet hairy root clones, which had been derived from a single seed. Statistical analysis by the Tukey test demonstrated that 19/28 and 16/28 inter-clone comparisons were significantly different when normalised for protein concentration and DNA content respectively. Possible causes of this variation and its implications for the genetic manipulation of plant growth and development are discussed.  相似文献

Sugar-beet plants ( Beta vulgaris L. cv. Monohill) were cultivated for 4 weeks in a complete nutrient solution. Indirect effects of cadmium were studied by adding 5, 10 or 20 μ M CdCl2 to the culture medium while direct effects were determined by adding 1, 5, 20, 50 or 2 000 μ M CdCl2 to the assay media. The photosynthetic properties were characterized by measurement of CO2 fixation in intact plants, fluorescence emission by intact leaves and isolated chloroplasts, photosystem (PS) I and PSII mediated electron transport of isolated chloroplasts, and CO2-dependent O2 evolution by protoplasts. When directly applied to isolated leaves, protoplasts and chloroplasts. Cd2+ impeded CO2 fixation without affecting the rates of electron transport of PSI or PSII or the rate of dark respiration. When Cd2+ was applied through the culture medium the capacity for, and the maximal quantum yield of CO2 assimilation by intact plants both decreased. This was associated with: (1) decreased total as well as effective chlorophyll content (PSII antennae size), (2) decreased coupling of electron transport in isolated chloroplasts, (3) perturbed carbon reduction cycle as indicated by fluorescence measurements. Also, protoplasts isolated from leaves of Cd2+-cultivated plants showed an increased rate of dark respiration.  相似文献

Plant regeneration from protoplasts of sugar beet (Beta vulgaris)   总被引:1,自引:0,他引:1  
Mesophyll protoplasts from two of five sugar beet lines tested were regenerated into plants. Mesophyll protoplasts of all lines showed high plating efficiencies up to 4.0% developed hard compact callus, and two of the lines also developed white, soft and friable callus consisting of starch grain-containing cells. Whereas the compact callus never regenerated into plants, the white friable ones frequently developed globular structures, which became green in the light and formed adventitious shoots after cytokinin (BAP or thidiazuron) treatment. Genetic analysis by PCR-fingerprinting and flow cytometry showed uniformity and unchanged ploidy levels in 15 independently regenerated plantlets in line NF. but altered ploidy level (from diploid to triploid) in a regenerated plantlet of clone VRB.  相似文献

通过对旱地甜菜叶片生长特性及摘除不同叶组对块根产量,含糖量,显微结构的影响研究,结果表明:甜菜第10-20片叶的叶龄最长,积温最高,是甜菜的主要功能叶;甜菜从第20片叶期起进入块根,糖份增长期,从第55叶期起进入糖份积累期;摘除不同叶组的叶片对甜菜块根产量,含糖量及显微结构均有不同程度降低作用,摘除前期叶组对甜菜块根产量,产糖量,根径减幅较大,摘除后期叶组对块根含糖量,维管束环数,维管束环密度减幅较大;摘除第1-30片叶对甜菜影响最大。  相似文献

The consumption of fructans as a low caloric food ingredient or dietary fibre is rapidly increasing due to health benefits. Presently, the most important fructan source is chicory, but these fructans have a simple linear structure and are prone to degradation. Additional sources of high-quality tailor-made fructans would provide novel opportunities for their use as food ingredients. Sugar beet is a highly productive crop that does not normally synthesize fructans. We have introduced specific onion fructosyltransferases into sugar beet. This resulted in an efficient conversion of sucrose into complex, onion-type fructans, without the loss of storage carbohydrate content.  相似文献

Members of the Chenopodiaceae, such as sugar beet and spinach, accumulate glycine betaine in response to salinity or drought stress. The last enzyme in the glycine betaine biosynthetic pathway is betaine aldehyde dehydrogenase (BADH). In sugar beet the activity of BADH was found to increase two- to four-fold in both leaves and roots as the NaCl level in the irrigation solution was raised from 0 to 500 mM. This increase in BADH activity was paralleled by an increase in level of translatable BADH mRNA. Several cDNAs encoding BADH were cloned from a gt10 libary representing poly(A)+ RNA from salinized leaves of sugar beet plants, by hybridization with a spinach BADH cDNA. Three nearly full-length cDNA clones were confirmed to encode BADH by their nucleotide and deduced amino acid sequence identity to spinach BADH; these clones showed minor nucleotide sequence differences consistent with their being of two different BADH alleles. The clones averaged 1.7 kb and contained an open reading frame predicting a polypeptide of 500 amino acids with 83% identity to spinach BADH. RNA gel blot analysis of total RNA showed that salinization to 500 mM NaCl increased BADH mRNA levels four-fold in leaves and three-fold in the taproot. DNA gel blot analyses indicated the presence of at least two copies of BADH in the haploid sugar beet genome.  相似文献

Reciprocal gene exchange between cultivated sugar beet and wild beets in seed production areas is probably the reason for the occurence of weed beets in sugar beet production fields. Therefore, when releasing transgenic sugar beet plants into the environment, gene transfer to wild beets ( Beta vulgaris ssp. maritima ) has to be considered. In this study the transfer of BNYVV- (beet necrotic yellow vein virus) resistance and herbicide-tolerance genes from two transgenic sugar beet lines that were released in field experiments in 1993 and 1994 in Germany to different wild beet accessions was investigated. In order to evaluate the consequences of outcrossing, manual pollinations of emasculated wild beet plants with homozygous transgenic sugar beet plants were performed. In the resulting hybrids the transgenes were stably inherited according to Mendelian law. Gene expression in leaves and roots of the hybrids was in the same range as in the original transgenic sugar beet plants. Moreover, it was found that in one of the wild beet accessions, transfer and expression of the BNYVV resistance gene did considerably increase the level of virus resistance.  相似文献

Surveys of the principal yellowing viruses of sugar beet, beet yellows virus (BYV) and beet mild yellowing virus (BMYV) in Spain were carried out from 1990–1993. Beet yellowing viruses were detected in all provinces, although the mean percentages of plants infected with BYV and BMYV were practically zero in the southern zone. Within the northern zone high variations from one province to another could be observed. The mean percentages of plants infected with BYV were higher in the Ebro Valley than in the Duero Valley. Areas infected with BYV were very restricted, while BMYV could be found to a variable extent all over Spain, although the infection levels were lower. The incidence and distribution of these viruses in the Spanish sugar beet crop makes the study and application of control measures for beet yellowing viruses necessary.  相似文献

Bruising of sugar beet roots and the consequential sugar loss do not receive the attention they deserve within the sugar beet industry. Recent harvester tests have indicated that current levels of bruising damage could be decreased with existing technology. There is, however, little understanding of biological factors affecting susceptibility to bruising of sugar beet roots. This paper examines the available information on causes of bruising, the significance of some tissue characteristics, the processes of sugar loss following bruising and agronomic, physiological and biochemical considerations relevant to bruising and the sugar loss that follows. The effect of field margins on the yield of sugar beet, wheat and barley was studied on commercial farms and in a series of field experiments from 1992–1997. There was always a trend of increasing yield from the edge of the field to the centre, with a marked reduction around the ‘tramlines’ and the area where machinery turns. In the studies on commercial farms, headland yield loss varied widely. In sugar beet the headlands yielded 19–41% less than the centre, with a mean reduction of 26%. In cereals the range was 3–19%, with a mean loss of 7%. Headland yield reductions were generally smaller in the field experiments than those found on commercial farms. These headland effects did not move towards the centre of the field when grass margins were planted at the edge of the field; there was no significant effect on the yield of the adjacent crop. The presence of boundary trees had the greatest effect on yield: in the outer 9 m of the field, the area shaded by trees produced 4.4 t ha-1 of wheat, and the area that was not shaded 8.1 t ha-1. Turning of machinery also significantly reduced yield, while grazing by rabbits and hares surprisingly had no effect. Following the reform of the Common Agricultural Policy in 1992, the main effect of which was to change from a price support policy to direct payments to producers, farmers in the European Union who produce more than a specified tonnage of ‘eligible crops’ per year, are required to fallow a given percentage of their land (currently 5%), to qualify for Arable Area Payments. Growers can elect to fallow fields on a rotational basis, or permanently. Headland set-aside is a term used to describe strips of set-aside, a minimum of 20 m wide around the edges of fields. In these experiments, the headland effect did not extend beyond 20 m from the field edge. Therefore, particularly in fields with boundary hedges or trees, headland set-aside could effectively remove the poor-yielding area at the field margin.  相似文献

In sugar beet genotypes with the Holly type of resistance to rhizomania, a disease due to infection of the beet necrotic yellow vein virus (BNYVV), the major gene rrl is responsible for resistance. Twelve RAPD markers linked to rrl were selected by BSA and mapped on linkage group IV using a segregating population previously analysed by the same group. Markers F61050 and N9600 were tightly linked, respectively in coupling and repulsion, to the Rrl allele (recombination values of 1.4 cM for both markers). After sequencing the products amplified by F61050 and N9600, new PCR primers were used to generate the two SCAR markers F6 and N9. The simultaneous use of these markers in a PCR reaction allows the correct fingerprinting of rrl rrl, Rrl rrl and Rrl Rrl sugar beet plants in populations segregating for the Holly resistance. In a group of sugar beet elite lines containing the Holly type of rhizomania resistance, SCAR F6 is always present whereas the SCAR N9 fragment is absent. Thus, in marker-assisted selection with coupling-repulsion-phase markers, SCAR F6 can be used in combination with N9, or together with any other RAPD marker linked in repulsion to the Rrl allele.  相似文献

AIMS: To determine the fermentation profiles by human gut bacteria of arabino-oligosaccharides of varying degree of polymerization. MATERIALS AND METHODS: Sugar beet arabinan was hydrolyzed with a commercial pectinase and eight fractions, of varying molecular weight, were isolated by gel-filtration chromatography. Hydrolysis fractions, arabinose, arabinan and fructo-oligosaccharides were fermented anaerobically by gut bacteria. Total bacteria, bifidobacteria, bacteroides, lactobacilli and the Clostridium perfringens/histolyticum sub. grp. were enumerated using fluorescent in situ hybridization. RESULTS: Bifidobacteria were stimulated to different extents depending on molecular weight, i.e. maximum increase in bifidobacteria after 48 h was seen on the lower molecular weight fractions. Lactobacilli fluctuated depending on the initial inoculum levels. Bacteroides numbers varied according to fraction; arabinan, arabinose and higher oligosaccharides (degree of polymerization, dp > 8) resulted in significant increases at 24 h. Only carbohydrate mixtures with dp of 1-2 resulted in significant increases at 48 h (log 8.77 +/- 0.23). Clostridia decreased on all substrates. CONCLUSIONS: Arabino-oligosaccharides can be considered as potential prebiotics. Significance and Impact of the Study: Arabinan is widely available as it is a component of sugar beet pulp, a co-product from the sugar beet industry. Generation of prebiotic functionality from arabinan would represent significant added value to a renewable resource.  相似文献

Rhizoctonia damping-off caused by Rhizoctonia solani Kühn, is one of the most damaging sugar beet diseases. It causes serious economic damage wherever sugar beets are grown. Biological control is an efficient and environmentally friendly way to prevent damping-off disease. Suppression of damping-off disease caused by R. solani was carried out by four isolates of Bacillus subtilis (Ehrenberg) Cohn as well as three isolates of each of Trichoderma harzianum Rifai and Trichoderma hamatum (Bonord.) Bainier. The effect of Bacillus and Trichoderma isolates against R. solani was investigated in vitro and tested on sugar beet plants under greenhouse conditions. Isolates of Bacillus and Trichoderma were able to inhibit the growth of R. solani in dual culture. Furthermore, Trichoderma isolates gave high antagonistic effect than isolates of B. subtilis. Under greenhouse conditions, coating seeds by T. harzianum and B. subtilis separately, reduced seedling damping-off significantly. However, applications of T. harzianum increased the percentage of surviving plants more than B. subtilis in comparison to control. The obtained results indicate that T. harzianum and B. subtilis are very effective biocontrol agents that offer potential benefit in sugar beet damping-off and should be harnessed for further biocontrol applications.  相似文献

Beets are a crop of particular concern regarding invasiveness questions because they commonly become feral due to unintentional hybridization with annual forms of wild beets. In this study the performance of transgenic beets resistant to Beet Necrotic Yellow Vein Virus (BNYVV) was compared to the performance of unmodified material from the same breeding line. Both transgenic and control genotypes were also compared to a conventionally bred variety carrying a similar phenotypic trait. Field tests were developed in a step by step fashion in order to study seed emergence and competitiveness in early life stages. The tests quantified the potential ecological advantage of virus resistance under virus and non-virus infestation conditions. In experimental field releases in 1993 and 1994 in Germany, a small but increasingly clear 'additive' ecological advantage of the genetically engineered trait was detected. In both years and all competition treatments, the conventional tolerant variety performed best. An impact of naturalization on natural, nonagricultural habitats may appear in wild beet populations in Italian seed beet production areas. However, a survey of coastal areas of North-Eastern Italy found no virus infestation in 1994, suggesting that an increase in wild beet fitness is unlikely to occur.  相似文献

The separate effects of beet yellows virus (BYV) and beet mild yellowing virus (BMYV) on yield of sugar-beet cultivars inoculated at different growth stages were assessed in field trials in 1985 and 1987. Early or mid-season inoculation decreased sugar yield by up to 47% for BYV, and up to 29% for BMYV. Infections after the end of July had no significant effect on yield. Both viruses caused significant increases in the juice impurities sodium, potassium and amino-nitrogen after infecting plants early in the season. Yield losses associated with infection were determined by the causative virus, the time of infection, and susceptibility of the sugar-beet cultivars.  相似文献

The principal storage reserve of sugar beet seeds is starch, which is localised in the perisperm. Additional storage reserves include the seed proteins, albumins, globulins and glutclins, which are exclusively located in the embryo. Soluble sugars are also detectable in all the organs of the mature seed. The time-course of reserve mobilisation in the different organs of the sugar beet ( Beta vulgaris L. cv. Regina) seed during germination and early seedling growth is documented, with particular reference to changes in (a) activities of hydrolases: a-amylase, β-amylase, and α-glucosidases; (b) levels of carbohydrates and (c) proteins. Amylase activities increase substantially in both cotyledons, as well as the perisperm, whereas the increase in α-glucosidase activities is largely confined to the perisperm.  相似文献
