Ecological explanations for the prevalence of resource specialists are abundant, whereas phylogenetic evidence on their origins is scarce. In this paper, we provide a molecular phylogenetic study of the 19 specialist or generalist species in the bark beetle genus Dendroctonus, which collectively attack species in four different genera in the conifer family Pinaceae. Given substantial variation in diet breadth, we asked two general questions concerning the evolution of resource use in this group. How conservative is the evolution of host use in these insects? Does specialization tend to be derived (i.e., a “dead end”)? To answer these questions, we estimated the phylogeny of Dendroctonus using mitochondrial DNA sequences and mapped transitions in resource use on the resulting phylogeny estimate. The evolution of affiliations with Pinus and Picea hosts in Dendroctonus was conservative among beetle species (PTP test; P < 0.012), but there was no significant correspondence between the phylogeny of these beetles and the phylogeny among their Pinaceae hosts (among genera, P = 0.28; among Pinus species, P = 0.82). Degree of specialization, as measured in the proportion of hosts used, was bimodally distributed with “generalist” species utilizing < 60% of the congeneric hosts within their range and six specialist species utilizing < 40% of the available hosts. Among the generalists, we found a strong correlation between the number of hosts encountered and the number of hosts utilized (R = 0.97, P < 0.0001), whereas there was no significant correlation among the specialists (R = 0.27, P = 0.59). The evolution of specialization in Dendroctonus proved highly labile—specialists arose from generalists at least six separate times (without reversal) all in derived positions, and closer examination of some specialists revealed instances where they appear to have lost particular host species from their diet. However, evidence from the ecological literature also suggests that several Dendroctonus generalists may have increased their range of host genera within the Pinaceae.  相似文献

The family Zetorchestidae is a morphologically and ecologically diverse group assigned to the higher oribatid mites (Brachypylina). We addressed the phylogeny of the family by including species of the genera Belorchestes, Litholestes, Microzetorchestes and Zetorchestes. We also analysed the affinities of the putative sister taxon (Eremaeidae), investigating Eremaeus and Eueremaeus. Zetorchestidae, Eremaeidae and Niphocepheidae were recently combined in one superfamily (Zetorchestoidea). These taxa were placed into a wider phylogenetic context by adding other presumably closely related taxa. Phylogenetic analyses based upon nuclear and mitochondrial DNA‐sequences revealed the monophyly of the Zetorchestidae as well as of all investigated species and genera of this family. Ancestral state reconstruction of jumping ability in latter family, moreover, suggested reverse character evolution within the studied zetorchestid taxa. Genetic diversity of the genera Eremaeus and Eueremaeus turned out to be higher than known, suggesting the existence of cryptic species. However, none of our analyses supported a sister group relationship among Zetorchestidae and Eremaeidae. Moreover, all calculated trees show a paraphyletic position between Zetorchestidae respectively Eremaeidae and Niphocepheidae.  相似文献

The advantages of nucleotide sequence data for studying phylogeny have been shown to include number of potential characters available for comparison, rate independence between molecular and morphological evolution, and utility of molecular data for modeling patterns of nucleotide substitution. Potential pitfalls have also been revealed and include difficulties of inferring positional homology, incongruence between organismal and gene genealogies, and low likelihood of recovering the correct phylogeny given certain patterns in the timing of speciation events. Statistical methods for comparing phylogenetic hypotheses have been used to assess the reliability of alternative trees for ascaridoid nematodes. Based on partial ribosomal RNA sequences, tree topologies inconsistent with monophyly of the Ascaridinae were significantly worse by maximum likelihood inference. The topology of the maximum parsimony tree based on full-length sequences of 18S rRNA and 300 nucleotides of Cytochrome oxidase II for 13 ascaridoid species was generally consistent with traditional taxonomic expectations at lower ranks, but inconsistent with most proposed arrangements at higher taxonomic levels.  相似文献

An adult male gorilla was donated from private ownership in 1994 to Zoo Atlanta and became part of the American Zoo and Aquarium Association's Gorilla Species Survival Plan. This animal, Ivan, was captured as an infant in Africa in 1964. Ambiguity regarding origin and concomitant sub‐species designation was resolved by analyzing the cytochrome oxidase II mitochondrial gene known to contain eight diagnostic sites for gorilla sub‐species. 细胞色素氧化酶Ⅱ(cytochromeoxidaseⅡ,COⅡ)基因位于线粒体DNA(mtDNA)上,编码细胞色素氧化酶亚基Ⅱ,该亚基为细胞色素c提供重要的结合位点。COⅡ基因进化速率较快,是昆虫分子系统学研究中理想的分子标记。目前,已经利用该基因从各个分类水平对昆虫系统发育关系、物种形成与分化、种群遗传与变异及生物地理等方面做了广泛的研究。研究表明,利用该基因可以很好地解决昆虫属、种及种下分类单元的系统发育问题,但是在解决科、亚科等高级阶元的系统发育关系时仍存在一些局限,COⅡ基因与其他mtDNA及核基因的联合分析能够更好地解决昆虫的系统发育问题。  相似文献

The blow fly genus Lucilia is composed largely of saprophages and facultative myasis agents, including the economically important species Lucilia cuprina (Wiedemann) (Diptera: Calliphoridae) and Lucilia sericata (Meigen). Only one species is generally recognized as an obligate agent of myiasis, Lucilia bufonivora Moniez, and this is an obligate parasite of toads. Lucilia silvarum (Meigen), a sister species, behaves mainly as a carrion breeder; however, it has also been reported as a facultative parasite of amphibians. Morphologically, these species are almost identical, and historically this has led to misidentification, taxonomic ambiguity and a paucity of studies of L. bufonivora. In this study, dipterous larvae were analysed from toad myiasis cases from the U.K., The Netherlands and Switzerland, together with adult specimens of fly species implicated in amphibian parasitism: L. bufonivora, L. silvarum and Lucilia elongata Shannon (from North America). Partial sequences of two genes, cox1 and ef1α, were amplified. Seven additional blow fly species were analysed as outgroups. Bayesian inference trees of cox1, ef1α and a combined‐gene dataset were constructed. All larvae isolated from toads were identified as L. bufonivora and no specimens of L. silvarum were implicated in amphibian myiasis. This study confirms L. silvarum and L. bufonivora as distinct sister species and provides unambiguous molecular identification of L. bufonivora.  相似文献

The Leptynia hispanica stick insect species complex includes bisexuals, triploid and tetraploid parthenogenetic populations, suggesting that polyploidy has played a central role in the evolution of this complex. An analysis of karyotype, mitochondrial DNA (cox2) and nuclear DNA (ef1-alpha) markers was carried out to clarify phylogenetic relationships and microevolutionary/phylogeographical patterns of the L. hispanica complex. Our analyses suggested a subdivision of bisexual populations into four groups, tentatively proposed as incipient species. Moreover, triploids and tetraploids showed two independent origins, the latter being more ancient than the former. From ef1-alpha analysis, triploids showed hybrid constitution, while the hybrid constitution of tetraploids is likely, but more data are needed. We suggest that L. hispanica is a case of 'geographical parthenogenesis' with parthenogenetic strains colonizing large peripheral ranges, and bisexuals confined to glacial refuge areas. Moreover, the age, wide distribution and competitive advantage of polyploids over diploids, demonstrate their significance in the evolution of the L. hispanica species complex.  相似文献

Catfishes Sperata are popular, known for its taste and nutritional value, and are found naturally in wide variety of freshwaters in South Asia. The taxonomy of Sperata spp., sampled from Hakaluki Haor in Bangladesh, was re‐evaluated based on morphological variation and DNA barcoding. The collective variation in morphometric characters and mitochondrial DNA revealed an undescribed old and well‐separated lineage under the genus Sperata along with two previously known Sperata aor and Sperata seenghala in Bangladesh. Analyses of morphological traits suggested significant differentiation among Sperata species. The variation in mitochondrial DNA supported the distinct lineage and taxonomical discrimination. Sperata aor diverged earlier from the new lineage and Sperata seenghala with a divergence of 5.39 (CI: 3.91–7.19) Mya (PP > 90). Sperata seenghala and the new lineage shared a most recent common ancestry, which diverged from each other around 3.41 (CI: 2.24–4.62) Mya (PP > 90). Thus, the newly identified lineage could be a subspecies of S. seenghala or even a species under the genus Sperata. The information of the study will be useful for conservation, sustainable management and selective breeding of the putative species, including previously reported Saor and S. seenghala in Bangladesh.  相似文献

Although aquatic caterpillars are a globally rare lifestyle, we have found them in multiple, independent lineages of the endemic moth genus Hyposmocoma across all of the Hawaiian Islands with flowing water. We formally describe 11 new species of Hyposmocoma that belong to four different larval case types: cone, bugle, medium burrito, and large burrito: Hyposmocoma kahamanoa sp. nov. from Oahu Island, Hyposmocoma kamakou sp. nov. from Molokai Island, Hyposmocoma kahaiao sp. nov. , Hyposmocoma waihohonu sp. nov. , and Hyposmocoma moopalikea sp. nov. from Maui Island, and Hyposmocoma aumakuawai sp. nov. , Hyposmocoma eepawai sp. nov. , Hyposmocoma ipowainui sp. nov. , Hyposmocoma kawaikoi sp. nov. , Hyposmocoma uhauiole sp. nov. , and Hyposmocoma wailua sp. nov. from Kauai Island. We also illustrate and describe in detail the aquatic case‐bearing larva of Hyposmocoma kahamanoa. Despite having similar ecologies as algae and lichen grazers at and below the water line of streams, prior research indicates that species with each case type constitute an independent lineage, with terrestrial sister taxa, and therefore the different groups of species bearing unique case types each represent an independent aquatic invasion. The case‐bearing larvae often occur sympatrically, and on Kauai even species with similar case‐types occur together, suggesting complex patterns of speciation and either past periods of isolation or sympatric speciation. Phylogenetic analysis of 2243 base pairs from two nuclear and one mitochondrial gene for 18 species confirm that each species is endemic to a single volcano, and that morphological divergence within case‐types has not been dramatic. Diversification has been complex, and superficially similar case type lineages are not all monophyletic. Kauai, the oldest but smallest of the major high islands, supports more species in the aquatic guild than any other island, thus island age, rather than size, may be important in generating diversity in this group. © 2011 The Linnean Society of London, Zoological Journal of the Linnean Society, 2011, 162 , 15–42.  相似文献

Introgression is the effective exchange of genetic information between species through natural hybridization. Previous genetic analyses of the Drosophila yakuba—D. santomea hybrid zone showed that the mitochondrial genome of D. yakuba had introgressed into D. santomea and completely replaced its native form. Since mitochondrial proteins work intimately with nuclear‐encoded proteins in the oxidative phosphorylation (OXPHOS) pathway, we hypothesized that some nuclear genes in OXPHOS cointrogressed along with the mitochondrial genome. We analyzed nucleotide variation in the 12 nuclear genes that form cytochrome c oxidase (COX) in 33 Drosophila lines. COX is an OXPHOS enzyme composed of both nuclear‐ and mitochondrial‐encoded proteins and shows evidence of cytonuclear coadaptation in some species. Using maximum‐likelihood methods, we detected significant gene flow from D. yakuba to D. santomea for the entire COX complex. Interestingly, the signal of introgression is concentrated in the three nuclear genes composing subunit V, which shows population migration rates significantly greater than the background level of introgression in these species. The detection of introgression in three proteins that work together, interact directly with the mitochondrial‐encoded core, and are critical for early COX assembly suggests this could be a case of cytonuclear cointrogression.  相似文献

Carrot (Daucus carota L. subsp. sativus) cultivation is important in Argentina, with the Garay department in Santa Fe province being a key production area. In recent years, dry rot symptoms have afflicted various carrot cultivars, resulting in substantial field losses and postharvest discards. Several soilborne pathogens, including Fusarium species, have been implicated in similar carrot diseases globally. This study aimed to identify the specific causal agent responsible for dry rot in carrots in central Argentina to enable targeted disease management strategies. Samples of symptomatic carrots were collected from fields in the Garay department. Fungal isolates were obtained from the infected tissue and subjected to morphological and molecular analyses. Pathogenicity tests were conducted to confirm the ability of the isolates to cause disease symptoms in carrots. Based on morphological and molecular identification, isolates were identified as Fusarium nirenbergiae, Fusarium triseptatum and Fusarium solani. Pathogenicity tests demonstrated that the three Fusarium species were capable of causing dry rot symptoms in carrots. This study represents the first report of Fusarium nirenbergiae, Fusarium triseptatum and Fusarium solani causing dry rot in carrots in Argentina.  相似文献

Accurate delimitation of species boundaries is especially important in cryptic taxa where one or more character sources are uninformative or are in conflict. Rather than relying on a single marker to delimit species, integrative taxonomy uses multiple lines of evidence such as molecular, morphological, behavioural and geographic characters to test species limits. We examine the effectiveness of this approach by testing the delimitation of two cryptic Nearctic species of Dioryctria (Lepidoptera: Pyralidae) using three independent molecular markers [cytochrome c oxidase I (COI), second internal transcribed spacer unit (ITS2), and elongation factor 1alpha (EF1alpha)], forewing variation and larval host plant association. Although mitochondrial DNA (mtDNA) haplotypes do not form reciprocally monophyletic clades, restricted gene flow between COI haplotype groups, and concordance with ITS2 genotypes, forewing variation and host plant associations support delimitation of two Nearctic species: eastern Dioryctria reniculelloides and western Dioryctria pseudotsugella. Conversely, EF1alpha genotype variation was incongruent with the two previous markers. A case of discordance between COI and ITS2 was detected, suggesting either introgression due to hybridization or retained ancestral polymorphism due to incomplete coalescence. This study is consistent with other similar literature where molecular loci in closely related species progress from shared to fixed haplotypes/alleles, and from polyphyletic to reciprocally monophyletic relationships, although loci may vary in these characteristics despite maintenance of genomic integrity between distinct species. In particular, mtDNA in other studies generally showed a lower rate of fixation of differences than did X-linked or autosomal loci, reinforcing the need to use an integrative approach for delimiting species.  相似文献

The green lacewing Chrysoperla carnea is a complex of cryptic species whose differentiation has been so far based upon morphology, ecophysiology, behaviour and preliminary mitochondrial DNA data using cytochrome oxidase subunit II (COII) and NADH dehydrogenase subunit II. In this work we extended the DNA data by screening nucleotide sequences of COII, cytochrome oxidase I, cytochrome b and the large ribosomal subunit of the mtDNA. These new data suggest that C. carnea s.s. is a well-supported, separate taxon, but that other taxa of the complex are not consistently differentiated by the current DNA data.  相似文献

Several species of the genus Oligoryzomys are natural hosts of different hantavirus genotypes affecting humans. The systematics of the genus is confusing, which complicates the identification of the rodent host and hence the potential endemic areas of hantavirus pulmonary syndrome. In this study, we analyse molecular data to infer phylogenetic relationships among Central and South American specimens of Oligoryzomys, and compare our results with previously published data on karyotypic, geographic distribution and host–virus associations to solve contradictory taxonomic reports. We identified 25 clades, each one corresponding to a different putative species. The phylogenetic trees show that Oligoryzomys longicaudatus is strongly related to the Oligoryzomys flavescens complex, which comprises four clades; Oligoryzomys nigripes is related to Oligoryzomys stramineus, Oligoryzomys vegetus is related to Oligoryzomys fulvescens from Central America, and Oligoryzomys brendae is the sister species of Oligoryzomys aff. destructor. We identified the following rodent host–hantavirus genotype relationships: O. longicaudatus–Andes; O. flavescens ‘West'–Bermejo; O. flavescens ‘East'–Lechiguanas; O. nigripes–Juquitiba; Oligoryzomys microtis–Rio Mamore and Rio Mamore‐3; Oligoryzomys chacoensis–Oran; Oligoryzomys costaricencis–Choclo; Oligoryzomys delicatus–Maporal; Oligoryzomys utiaritensis–Castelo dos Sonhos; Oligoryzomys sp. RT2012–Rio Mamore‐4; Oligoryzomys sp. (and not Oligoryzomys fornesi)–Anajatuba. In this study we tested the use of mucus from five species of Neotropical marine batoid elasmobranchs to extract genomic DNA for barcoding and phylogenetic analysis. The DNA from all individuals sampled was successfully amplified and sequenced for molecular barcode, allowing 99–100% accuracy to the species level. This method proved to provide reliable and good-quality DNA for barcoding and phylogenetic analysis of Neotropical elasmobranchs, through rapid handling and with low disturbance to animals.  相似文献

通过与从GenBank中下载的8种隼属鸟类的Cytb和COⅠ部分序列进行同源比对,并以最大简约法(MP)和贝叶斯推断法(BI)构建系统树,对1只涉案的隼科鸟类进行了分子鉴定。结果表明:样品与猎隼Cytb和COⅠ的序列同源性最高,分别为98.91%~99.41%和99.67%~100%,样品与猎隼Cytb和COⅠ序列的遗传距离最小,分别为0.003~0.007和0.000~0.003;同时,样品和猎隼在两种系统树上都始终聚在一起。由此可推断该样品为猎隼,为2010年5月四川省丹巴县森林公安局受理的非法盗猎和贩运隼形目鸟类一案的审理提供了有力的证据。  相似文献

The most genetically divergent populations of the European snow vole Chionomys nivalis occupy the extreme eastern range of the Black Sea and the Caspian Sea. It was recently suggested that subspecies C. n. spitzenbergerae from the Central Taurus Mountains (Turkey) represents a highly divergent lineage of C. nivalis from the Alada? Range which induced us to address its status by examining topotype specimens. Two females karyotyped displayed 54 mitotic chromosomes of 25 acrocentric autosomal pairs. One of the smallest autosomal pairs was heteromorphic in both specimens, consisting of a submetacentric and an acrocentric chromosomes (NFa = 53). Cytochrome b sequences however unambiguously clustered both individuals with reference sequences from C. nivalis from Turkey. We conclude that a deviant haplotype from the Alada? Range does not represent C. n. spitzenbergerae. Its taxonomic identity was not resolved in our study. Further attention should be devoted to snow voles from Central Anatolia and Western Iran, which are characterized by cranial peculiarities.  相似文献

Leafcutting ants of the genus Atta are the most conspicuous members of the tribe Attini, the fungus-growing ants. Atta species have long attracted the attention of naturalists, and have since become a common model system for the study of complex insect societies as well as for the study of coevolutionary dynamics due to their numerous interactions with fungi and other microbes. Nevertheless, systematics and taxonomy of the 15 species in the genus Atta have proven challenging, due in part to the extreme levels of worker polymorphism these species display, leading to disagreements about the validity of as many as five different subgenera and calling into question the monophyly of the genus. Here, we use DNA sequence information from fragments of three mitochondrial genes (COI, tRNA leucine and COII) and one nuclear gene (EF1-αF1), totaling 1070 base pairs, to reconstruct the phylogenetic relationships of Atta species using maximum parsimony, maximum likelihood and Bayesian inference techniques. Our results provide support for monophyly of the genus Atta, and suggest that the genus is divided into four monophyletic groups, which correspond to four of the five previously erected Atta subgenera: Atta sensu stricto and Archeatta, each with species composition identical to earlier proposals; Neoatta and Epiatta, with major differences in species composition from earlier proposals. The current geographic ranges of these species suggest that the historical separation of South America from Central and North America has played a role in speciation within this genus.  相似文献
