The antigenic heterogeneity of the reticular framework of the white pulp (WP) and marginal zone (MZ) is well documented in the human adult spleen. The ontogeny of the WP and MZ of human fetal spleens was examined with special reference to the heterogeneity of the reticular framework. In the spleen of the 17th gestational week (gw), α-smooth muscle actin (α-SMA)-positive reticulum cells were scattered around the arterioles. From the 20th to 23rd gw, α-SMA-positive reticulum cells increased in number and began to form a reticular framework. An accumulation of T and B lymphocytes occurred within the framework, and a primitive WP was observed around the arterioles. At the 24th gw, antigenic diversity of the reticular framework was observed, and T and B lymphocytes were segregated in the framework. T lymphocytes were sorted into the α-SMA-positive reticular framework, and the periarteriolar lymphoid sheath (PALS) was formed around the arteriole. B lymphocytes aggregated in eccentric portions to the PALS and formed the lymph follicle (LF). The reticular framework of the LF was α-SMA-negative. MZ appeared in the α-SMA-positive reticular framework around the WP at the 26th gw. The PALS, LF, and MZ developed with gestational time. The reticular framework of the PALS, LF, and MZ is thus heterogeneous in the fetal spleen, and the development of the heterogeneity is related to the ontogeny of the PALS, LF, and MZ. The relationship between attachment of mRNA to the cytoskeletal framework and its translation was examined using the mRNA for a polypeptide of 40 kDa (P-40) which is translated in rat L6 myoblasts but not in the myotubes. In both myoblasts and myotubes this mRNA was found to be associated with the cytoskeletal framework. Furthermore, the stability of the association between P-40 mRNA and the cytoskeletal framework in absence of RNA and protein synthesis was examined by using actinomycin D and NaF to block RNA and protein synthesis, respectively. In absence of RNA synthesis portions of both nontranslated P-40 mRNA and translated actin mRNA of myotubes were released into the soluble fraction. In myoblasts, however, both mRNAs remained associated with the cytoskeletal framework following inhibition of RNA synthesis. Inhibition of protein synthesis, on the other hand, had a more dramatic effect on the association between the cytoskeletal framework and P-40 mRNA in myoblasts but not in myotubes. In contrast, the association between actin mRNA and cytoskeletal framework was unaffected by inhibition of protein synthesis in both myoblasts and myotubes. The results of these studies show that the molecular nature of association between cytoskeletal framework and mRNA may differ among mRNAs and may also depend on whether the cells are dividing or are terminally differentiated. Furthermore, no direct relationship between the translation of mRNA and its attachment to the cytoskeletal framework was observed.  相似文献

Forests are among the most important ecosystems for the provision of hydrological services. These include water supply and water damage mitigation, in the dimensions of quantity, timing and quality. Although the hydrological role of forests is well documented in the literature, a conceptual framework integrating these three dimensions is still missing. In this study, a comprehensive conceptual framework to improve the assessment of hydrological services provided by forests was developed. In addition, the framework was tested by an illustration for northern Portugal, a region with both Mediterranean and Atlantic climatic influences. The TEEB (The Economics of Ecosystems and Biodiversity) framework of ecosystem services was adapted to the relation between forests and water. Then, this new framework was complemented with a set of spatially-explicit indicators that quantify the supply and demand of hydrological services. In addition, the implications of the framework were discussed in the context of the social-ecological systems, using the DPSIR (Drivers, Pressures, State, Impacts, and Responses) model. Finally, the framework and the indicators were illustrated for northern Portugal using the water supply (quantity) and soil erosion control as examples. Results show that the proposed conceptual framework is a useful tool to support land planning and forest management, adapting the provision of hydrological services to the regional biophysical and social conditions. The test of the framework across a heterogeneous region suggests that a spatially explicit combination of system property, function, service and benefit indicators can be an effective way of analysing and managing the supply and demand of the hydrological services.  相似文献

Cytochalasin D was shown to be a reversible inhibitor of protein synthesis in HeLa cells. The inhibition was detectable at drug levels typically used to perturb cell structure and increased in a dose-dependent manner. The drug also released mRNA from the cytoskeletal framework in direct proportion to the inhibition of protein synthesis. The released mRNA was unaltered in its translatability as measured in vitro but was no longer translated in the cytochalasin-treated HeLa cells. The residual protein synthesis occurred on polyribosomes that were reduced in amount but displayed a normal sedimentation distribution. The results support the hypothesis that mRNA binding to the cytoskeletal framework is necessary although not sufficient for translation. Analysis of the cytoskeletal framework, which binds the polyribosomes, revealed no alterations in composition or amount of protein as a result of treatment with cytochalasin D. Electron microscopy with embedment-free sections shows the framework in great detail. The micrographs revealed the profound reorganization effected by the drug but did not indicate substantial disaggregation of the cytoskeletal elements.  相似文献

The polyomavirus proteins VP1, VP2, and VP3 move from their cytoplasmic site of synthesis into the nucleus, where virus assembly occurs. To identify cellular or viral components which might control this process, we determined the distribution of VP1, VP2, and VP3 in a soluble fraction, a cytoplasmic cytoskeleton fraction, and a nuclear framework fraction of infected cells. All three proteins were detected in a detergent-extractable form immediately after their synthesis in polyomavirus-infected cells. Approximately 50, 25, and 40% of pulse-labeled VP1, VP2, and VP3, respectively, associated with the skeletal framework of the nucleus within 10 min after their synthesis. The remaining portion of each labeled protein failed to accumulate on the nuclear framework during a 40-min chase and was degraded. When expressed separately by recombinant vaccinia viruses, VP1 and VP2, but not VP3, accumulated on the nuclear framework. This association was not dependent on other polyomavirus proteins or viral DNA. The amount of total VP1 and VP2 which was bound to the nuclear framework approximated 45 and 20%, respectively. Indirect immunofluorescence demonstrated an exclusive nuclear localization of VP1 in situ. In coinfection experiments, a greater percentage of total VP2 and VP3 was bound to the nuclear framework of cells which cosynthesized VP1. These results indicate that although VP1 and VP2 can bind independently to the insoluble nuclear framework, the association of VP3 with this nuclear structure is promoted by the presence of VP1.  相似文献

Summary The three-dimensional organization of microtubules in generative cells during their development in pollen grains of Hippeastrum vittatum and the dynamic changes that occur were studied by collecting large quantities of fixed and isolated generative cells for immunofluorescence microscopy. The framework configuration and the arrangement pattern of the microtubule organization was investigated. The microtubule framework changed in shape from being spherical at an early stage to being long spindle-shaped at maturity: various transitional forms were observed: ellipsoidal, pear-shaped and short spindle-shaped. The microtubule arrangement making up this framework changed correspondingly from the original network, which was random in distribution, to axially oriented long bundles via an intermediate pattern composed of a mixture of networks with long bundles. However, cells with the same framework configuration might be heterogeneous in microtubule arrangements.  相似文献

A novel conceptual framework is presented that proposes to apply trait‐based approaches to predicting the impact of environmental change on ecosystem service delivery by multi‐trophic systems. Development of the framework was based on an extension of the response–effect trait approach to capture functional relationships that drive trophic interactions. The framework was populated with worked examples to demonstrate its flexibility and value for linking disparate data sources, identifying knowledge gaps and generating hypotheses for quantitative models.  相似文献

An indicator framework was designed as an operational science-based tool for the evaluation of the environmental aspects of sustainable forest management at stand level in Flanders (Belgium). The framework aims to assess the effects of forest management on forest composition, structure and functioning. It consists of seven principles and 19 criteria, to which 157 potential indicators, selected from literature, were assigned; 40 of these were considered as suitable by an expert panel, based on 10 operational selection criteria. All indicators were quantitative variables measurable in the field.After elaboration of a measurement protocol, the indicator framework was validated in 115 forest stands, distributed over the three main forest types of Flanders. The new indicator framework exhibited greater sensitivity to forest management practices and demonstrated better discriminating power than the method that is currently used by the Flemish forest administration to estimate the naturalness and environmental quality of a forest stand. Following a detailed cost calculation of each indicator and based on the sensitivity of each indicator to forest management practices, the indicator framework was further reduced to a final set of 29 indicators. This framework can also be applied in other regions, provided that local threshold values are defined to convert indicator values to indicator scores.The selection procedure and the possible contribution of this set to the forest management in Flanders are discussed.  相似文献

Current routing services for sensor networks are often designed for specific applications and network conditions, thus have difficulty in adapting to application and network dynamics. This paper proposes an autonomic framework to promote the adaptivity of routing services in sensor networks. The key idea of this framework is to maintain some feature functions that are decoupled from originally-integrated routing services. This separation enables significant service changes to be done by only tuning these functions. Measures including parameterization are taken to save the energy for changing these functions. Further, this framework includes a monitoring module to support a policy-based collaborative adaptation. This paper shows an example autonomic routing service conforming to this framework. The association of poliovirus metabolism with the cytoskeleton was investigated. Infected cells were extracted by using the nonionic detergent Triton X-100 in the physiological cytoskeleton buffer. The skeletal framework obtained was examined by transmission electron microscopy of resinless sections. The fibers of the framework were grossly distorted in infected cells. No virions or procapsids were seen but many virus-specific spheroidal bodies were associated with the framework. They had a diameter of 40 to 70 nm, were characterized by a dense core and a translucent periphery, and occurred in strings, often near the remnants of flattened vesicles. These spheres may correspond to virus-synthesizing bodies. The metabolism of poliovirus RNA was shown to be associated with the skeletal framework by pulse-labeling cells with [3H]uridine and measuring the RNA retained on the framework. 20S double-stranded RNA, a form of poliovirus RNA found only in the replication complex, was attached to the skeleton throughout a 60-min pulse-label. 35S single-stranded viral RNA, a form found in virions, in polyribosomes, and in the replication complex, appeared first on the framework but after a few minutes was also found in the soluble cytoplasmic phase, encapsidated in virions. In contrast to viral RNA, viral proteins exhibited a varied association with the skeletal framework. Viral proteins were pulse-labeled with [35S]methionine and chased with unlabeled methionine. Although all of the virus-specific proteins were found, to some extent, in the skeletal fraction, the derivatives of P2 (P2-X and P2-5) and a derivative of P3 (P3-2) showed a preferential association with the skeletal framework. Virions and procapsids, on the other hand, were not associated with the cytoskeleton; both they and their component proteins (P1-VP0, P1-VP1, P1-VP2, and P1-VP3) were found dominantly in the soluble cytoplasmic phase. The pathway of poliovirus assembly can be inferred from the above data. It is different from that found previously for the enveloped vesicular stomatitis virus and may be representative of encapsidated cytoplasmic virus assembly.  相似文献

To adequately analyze the impacts associated with the rising use of automobiles, an assessment framework is needed that includes environment, health, economic, and sociocultural impacts. Such a framework was developed and applied to a proposed freeway-widening project in Edmonton, Canada. The assessment framework was developed using both Multi-Criteria Analysis and the Ecosystem Approach to Human Health (Ecohealth). Community participation was vital in the application of the assessment framework to this case study. Six stakeholder groups, including community members, City Councillors, and health, environment, and transportation experts, provided needed qualitative data for the assessment framework. Quantitative data were gathered from an ecological study design that associated traffic volumes with respiratory conditions in Edmonton. Community members perceptions about the impacts of the freeway widening differed from those of the expert groups in a number of areas. Environmental and health degradation was more of an issue to community members than to expert groups. Though respiratory conditions were not projected to increase by a significant amount because of the freeway widening, further analysis is necessary on other biophysical and socioeconomic impacts listed in the assessment framework. The divergence in opinion between community members and experts suggests that more communication is needed between these groups in relation to transportation planning. The Ecohealth approach ensures that community concerns are addressed in transportation planning.  相似文献

Multi-atlas segmentation propagation has evolved quickly in recent years, becoming a state-of-the-art methodology for automatic parcellation of structural images. However, few studies have applied these methods to preclinical research. In this study, we present a fully automatic framework for mouse brain MRI structural parcellation using multi-atlas segmentation propagation. The framework adopts the similarity and truth estimation for propagated segmentations (STEPS) algorithm, which utilises a locally normalised cross correlation similarity metric for atlas selection and an extended simultaneous truth and performance level estimation (STAPLE) framework for multi-label fusion. The segmentation accuracy of the multi-atlas framework was evaluated using publicly available mouse brain atlas databases with pre-segmented manually labelled anatomical structures as the gold standard, and optimised parameters were obtained for the STEPS algorithm in the label fusion to achieve the best segmentation accuracy. We showed that our multi-atlas framework resulted in significantly higher segmentation accuracy compared to single-atlas based segmentation, as well as to the original STAPLE framework.  相似文献

An object-oriented modelling framework for the arterial wall is presented. The novelty of the framework is the possibility to generate customisable artery models, taking advantage of imaging technology. In our knowledge, this is the first object-oriented modelling framework for the arterial wall. Existing models do not allow close structural mapping with arterial microstructure as in the object-oriented framework. In the implemented model, passive behaviour of the arterial wall was considered and the tunica adventitia was the objective system. As verification, a model of an arterial segment was generated. In order to simulate its deformation, a matrix structural mechanics simulator was implemented. Two simulations were conducted, one for an axial loading test and other for a pressure–volume test. Each simulation began with a sensitivity analysis in order to determinate the best parameter combination and to compare the results with analogue controls. In both cases, the simulated results closely reproduced qualitatively and quantitatively the analogue control plots.  相似文献

This article describes the current state of the development toward a common European framework for site-specific ecological risk assessment (SS-ERA) Although common progression has been slow in the past two years, earlier activities were very promising. Results are presented of a 2001 workshop to discuss the scientific development and policy needs in preparation of such a common framework. The framework was recommended to follow a tiered approach. Other important elements for a common European framework for SS-ERA were identified to be the use of generic values in the first tier and bioassays in later tiers, to address bioavailability in the assessment, to differentiate for land use. Also, the framework should allow for negotiation between stakeholders specific to the site. These aspects are present in the Dutch approach to SS-ERA, and this article further presents some experience with the application of this framework in a large case of SS-ERA in The Netherlands. The derivation of suitable ecological parameters and assessment criteria in view of land use in a tiered approach risk assessment is focused on, and the interactive process between stakeholders and ongoing discussions concerning references and criteria for assessment are illustrated.  相似文献

This paper presents a new computational framework for automatic foot classification from digital plantar pressure images. It classifies the foot as left or right and simultaneously calculates two well-known footprint indices: the Cavanagh's arch index (AI) and the modified AI. The accuracy of the framework was evaluated using a set of plantar pressure images from two common pedobarographic devices. The results were outstanding, as all feet under analysis were correctly classified as left or right and no significant differences were observed between the footprint indices calculated using the computational solution and the traditional manual method. The robustness of the proposed framework to arbitrary foot orientations and to the acquisition device was also tested and confirmed.  相似文献
