以苏云金芽胞杆菌拟步行甲亚种菌株(Bacillus thuringiensis subsp.tenebrionis)YBT-1765作为出发菌株,克隆了一个包含复制子的EcoRI酶切片段,大小约为11kb,称为oril65。这是国内外从此亚种中克隆到的第一个复制子,缩小到8kb左右后仍然能够复制。杂交结果显示,此复制子来源于菌株YBT-1765可以检测到的分子量最大的质粒,以此复制子构建的穿梭载体pBMB6071在不同受体菌中的稳定性差异很大,其中在以色列亚种无晶体突变株4Q7中,传40后代,稳定性100%,质粒pBMB6071与含ori1030和ori2062在库斯塔克亚种无晶体突变株BMB171中是相容的。  相似文献

A series of truncated gene products from an alkaline cellulase from an alkalophilic Bacillus sp. No. 1139 has been prepared. The variously sized proteins were products of in vitro insertional mutagenesis constructs made by gene inserts containing translational terminators. One product, a 46-kDa protein, which had about half the Mr of the original cellulase, had a similar enzyme activity and pH optimum to the original 92-kDa protein. In contrast, a slightly smaller product protein (43 kDa) did not show cellulase activity.  相似文献

从土壤中分离到9株产生β-甘露聚糖酶的芽孢杆菌(Bacillus sp.)。Bacillus sp.M50250mL三角瓶摇瓶培养试验,以4%的魔芋粉为碳源,1.0%(NH4)2SO4为氮源,0.35%Na2CO3,30~34℃培养60h产酶达到高峰。酶活力为180~220u/mL。100L罐发酵,在30~32℃,1:0.75vvm通气量,200r/min条件下,发酵液酶活力高达330u/mL。  相似文献

为综合利用烟草废料,筛选得到能有效降解烟碱与绿原酸的两株芽胞菌,考察了所选菌株对烟碱与绿原酸的降解特性。实验结果表明,菌株Bacillus sp.X6表现出较高降解烟碱能力,培养36h可将含烟碱6.04mg/g烟草物料中的烟碱降解60.3%,72h降解率可达87.6%;对绿原酸降解效果最好的是菌株Bacillus sp.Xy,培养48h将含绿原酸10.57mg/g烟草物料中的绿原酸降解了50.5%,72h可将绿原酸降解62.2%,  相似文献

A highly efficient procedure for the transformation of Bacillus thuringiensis and Bacillus subtilis using covalently closed circular plasmid DNA was developed by using the small Staphylococcus aureus plasmid pC194 and electroporation. We have achieved transformation efficiencies in B. thuringiensis subsp. kurstaki (HD-73) greater than 5 x 10(6) transformants/micrograms plasmid DNA. The electro-transformation (or electroporation) procedure also worked with B. subtilis 168 although at a 200-fold less level of efficiency. The results indicated that the plasmid exists in double and single-stranded forms both in B. subtilis and B. thuringiensis. A second single-stranded species was also observed in both species. This technique may prove to be applicable to other members of the genus Bacillus.  相似文献

A bleach-stable, thermotolerant, alkaline protease for detergent formulation from a newly isolated Bacillus SB5 is reported. Most (85%) activity of the enzyme was retained in the presence of 10% (v/v) H2O2 and 1% SDS (w/v) at 40°C, after 1 h. The enzyme was optimal at pH 10 and 60°C to 70°C. Enzyme activity was enhanced 30 to 80% in presence of ionic and non-ionic detergents, surfactants and commercial detergents or bleach.  相似文献

In this study, Ni (II) biosorption capacity of immobilized cells of Bacillus sp. was investigated. Biosorption of Ni (II) was carried out in batch experiments and the important environmental conditions were optimized. The uptake of metal was rapid, and equilibrium was attained within 270 min. Bacillus strains (ten cultures) were isolated from nickel electroplating effluent by heat shock method. These isolates were grown up in nutrient broth supplemented with Ni (II)(50 mg/L). The culture, exhibiting maximum biosorption capacity (qmax: 118 mg/g), was selected and labeled Bacillus Bio‐4. In order to develop an economical biosorption process cell mass of Bacillus, Bio‐4 was immobilized in Na‐alginate. It was concluded from the results that biosorption of nickel is highly dependent on the type of sorbent and experimental conditions employed. Our results demonstrate that 6.0 mg immobilized cells (18 mg cell biomass in 3.0 mL of 1% Na alginate) had a maximum biosorption capacity of 113 mg Ni(II) per liter of suspension at pH 8.0, 100 rpm and 25°C. The Ni (II) removal was estimated to be 97.4%.  相似文献

In vitro culture experiments using three bacterial strains CSJC1, CSJC2, and CSJC3 isolated from speleothems, Rani cave, Chhattisgarh, India, were studied to examine their biomineralization potential. These speleothems showed high microbial cell enumerations on nutrient agar and iron agar (9 × 104 CFU/g) followed by thiosulfate agar (7 × 104 CFU/g), and 60 diverse strains were isolated. The BLASTn sequence search of 16S rRNA sequences with the NCBI database to establish the identity of CSJC1, CSJC2, and CSJC3 strains yielded similarity scores of ≥99% with the respective organisms, and the strains were identified as CSJC1 – Bacillus sp., CSJC2 – Cupriavidus sp., CSJC3 – Bacillus sp. The phylogenetic analysis of CSJC2 strain suggests that it formed a separate major cluster with Cupriavidus sp. and Cupriavidus necator. The phylogenetic analysis of CSJC1 and CSJC3 strains revealed that it formed a major cluster with several strains of Bacillus sp. and Bacillus subtilis. The biominerals induced by Cupriavidus sp. CSJC2 strain imaged with an ultra high-resolution field emission scanning electron microscope (FE-SEM) were seen as calcified coccoid shells that transformed into calcified dumbbells. FE-SEM imaging of biominerals induced by B. subtilis CSJC1 and CSJC3 tested both on B4 media and sheep blood agar individually showed that the precipitates formed calcified dumbbells that were almost similar but not identical phenotypically, indicating that strain-specific morphologies and crystal formation is easier when Ca is present in the media. This is the first comprehensive report on the possible evidences about the role of Cupriavidus sp. in calcite precipitation isolated from speleothems in the Indian caves. These results allow us to postulate that the identified strains may have a role in the biogenic influences in mineral formations at Rani cave.  相似文献

A simple approach to test the ability of bacteria to undergo natural genetic transformation is suggested. The basic feature of the approach is the cultivation of bacterial cells in the presence of exogenous (plasmid) DNA on cellophane membranes placed successively on nutrient and selective agar. Using this approach the ability of Bacillus cereus for "natural" genetic transformation was detected. Transformation frequencies varied from 10(-8) to 10(-6).  相似文献

A DNA recombination-deficient Rec mutant of Bacillus stearothermophilus was obtained via mutagenesis from a parental strain B. stearothermophilus MO-3 deficient in neutral protease npr. The Rec status was confirmed by the fact that no recombinational revertant appeared when a hybrid plasmid pNP13 carrying npr was used. The performance of the temperature-dependent integrative and excisable plasmid pTRA117 was further studied in this Rec host. Its integration into host chromosome was found to be dependent on Rec, although flanking-homology integration had been disproved. Consequently, the excision product of pTRA117, a thermostable plasmid pTRZ117, did not emerge in the Rec host.  相似文献

Twenty-six strains of Bacillus cereus from different sources were determined to be either mesophilic or psychrotrophic by growth at 6 and 42 degrees C. The strains were also screened by two polymerase chain reaction (PCR) methods designed to discriminate between mesophilic and psychrotrophic types. Seventeen of the 26 strains were able to grow at 6 degrees C, but only four conformed to the new psychrotolerant species Bacillus weihenstephanensis. Among the 26 strains were two which caused outbreaks of food poisoning in Norway, and three others that were isolated from food suspected of causing illness. The presence of the gene components encoding production of enterotoxins Nhe, Hbl, EntT and a recently described cytotoxin K was determined by PCR. All the strains possessed genes for at least one of these toxins, and 19 of the 26 strains were cytotoxic in a Vero cell assay. We conclude that there are psychrotrophic B. cereus strains which cannot be classified as B. weihenstephanensis, and that intermediate forms between the two species exist. No correlation between cytotoxicity and the growth temperature of the strains was found.  相似文献

The nucleotide sequences of three independent fragments (designated no. 3, 4, and 9; each 15–20 kb in size) of the genome of alkaliphilic Bacillus sp. C-125 cloned in a λ phage vector have been determined. Thirteen putative open reading frames (ORFs) were identified in sequenced fragment no. 3 and 11 ORFs were identified in no. 4. Twenty ORFs were also identified in fragment no. 9. All putative ORFs were analyzed in comparison with the BSORF database and non-redundant protein databases. The functions of 5 ORFs in fragment no. 3 and 3 ORFs in fragment no. 4 were suggested by their significant similarities to known proteins in the database. Among the 20 ORFs in fragment no. 9, the functions of 11 ORFs were similarly suggested. Most of the annotated ORFs in the DNA fragments of the genome of alkaliphilic Bacillus sp. C-125 were conserved in the Bacillus subtilis genome. The organization of ORFs in the genome of strain C-125 was found to differ from the order of genes in the chromosome of B. subtilis, although some gene clusters (ydh, yqi, yer, and yts) were conserved as operon units the same as in B. subtilis. Mesophilic strains producing a new bacteriocin: Megacin BII, have been isolated from strains of Bacillus megaterium. Facultatively thermophilic strains producing Megacin BI were less sensitive to this new activity than nonproducing mesophiles and strains producing Megacin BII were also also more resistant to Megacin BI. Strains producing Megacin BII contained a large plasmid of 36·106:pSE 203. This plasmid was introduced into non-megacinogenic acceptor strains by protoplast transformation, they then became megacin producers and immune to Megacin BII. Plasmid pSE 203 has been mapped with endonucleases. No similarity to the Megacin A plasmids pBM 309 [Rostás et al. A low-cost medium, MGRS, has been developed for growth and lipase production from Bacillus THL027 at 65 degrees C and pH 7.0. MGRS was composed of 2% (v/v) buffer solution (7.3% (w/v) Na(2)HPO(4), 3.2% (w/v) KH(2)PO(4), pH 7.2), 40 microg ml(-1) FeSO(4) and 40 microg ml(-1) MgSO(4), 0.1% (w/v) (NH(4))(2)SO(4) supplemented with 3% NaCl, 0.1% glucose, 1.0% rice bran oil and 0.5% (w/v) rice bran. The lipase was purified 2.6-fold to apparent homogeneity by ultrafiltration and gel filtration chromatography. Its molecular mass was 69 kDa. The purified enzyme was characterized for its general physical properties.  相似文献

AIMS: In this study, some physical properties of Bacillus sp. exo-polysaccharide were investigated. METHODS AND RESULTS: An extracellular polysaccharide was purified by sequential precipitations after homogenization of the diluted culture supernatant of Bacillus sp. CP912. Its physical properties were examined such as lipid emulsifying effect on several vegetable oils and flocculating activity against the activated carbon suspension. The melting point and endothermic calories of the polysaccharide were 128.7 degrees C and 50.864 kCal mol-1, respectively. Its pyrolysis temperature was 284.58 degrees C. The polysaccharide showed high lipid emulsifying activity on oil-water emulsion, against olive, peanut, sunflower and corn oils. It exhibited high flocculating activity as well against activated carbon. CONCLUSIONS: The present findings suggest that the extracellular polysaccharide produced by Bacillus sp. CP912 has a great industrial potential because of its high lipid emulsifying and flocculating activity. SIGNIFICANCE AND IMPACT OF THE STUDY: These data represent a novel Bacillus sp. extracellular polysaccharide possessing high emulsifying and flocculating effects.  相似文献

The molecular mass of esterases usually falls in the range of 20–160 kDa, although an esterase of 5.7 kDa from Candida lipolytica has been described. Three other enzymes smaller than 10 kDa have been reported, all of which were more thermostable than their higher molecular mass counterparts. This paper describes the purification of an extracellular esterase hydrolysing fluorescein dibutyrate from Bacillus stearothermophilus NCIMB 13335. The esterase had a molecular mass of 1.57 kDa when analysed by SDS-PAGE, gel filtration and MALDI-TOF spectrometry. This enzyme retained more than 90% of its activity after incubation at 90°C for 2 h.  相似文献
