Summary The course of the Nervus connectivus (N.c.), its branches, and synaptic connections within the frontal ganglion (FG) were investigated electron microscopically after cobalt iontophoresis of the N.c. The subsequent treatment of ultrathin sections with Timm's method was found to be very suitable for identifying the smallest branches. In the neuropil, fibers of the N.c. form Gray-I-type synapses, but also dyads are abundant, whereby the N.c. fibers occur exclusively in postsynaptic position with neurosecretory fibers. Summary In the lamina ganglionaris, the first optic ganglion of the fly, the inventory of cell types as well as the patterns of their connections are well known from light microscopic investigations. Even the synaptic contacts are known with relative completeness. However, the structural details visible on electron micrographs are very difficult to interpret in functional terms. This paper concentrates on two aspects: 1) the synaptic complex between a retinula cell axon and four postsynaptic elements, arranged in a constant elongated array (it is suggested that all synapses in which the retinula cell is presynaptic are of this kind), and 2) the gnarl complex in which a presynaptic specialization in one neuron is separated from another neuron by a complicated glial invagination. The participation of glia at postsynaptic sites seems to be quite common in this ganglion. Occasionally it seems that a glia cell is the only postsynaptic partner facing a presynaptic specialization within a neuron.  相似文献

A cellular (LPS I) and extracellular (LPS II) lipopolysaccharide were isolated from Moraxella glucidolytica cells grown on ethanol and from the culture fluid, respectively. Both LPS were toxic when injected to mice and chick embryos. These LPS contained glucose, galactose, glucosamine, galactosamine, 2-keto-3-deoxyoctonate and lipids. By permethylation studies, glucose was found to be linked (16) and (13) in LPS I and only (16) in LPS II. Galactose was the terminal non-reducing sugar. Branching occurred at positions 3 and 4 of galactose residues. LPS I was rich in - and -hydroxylauric and -hydroxymyristic acids and LPS II contained mainly stearic and -hydroxymyristic acids. LPS I was detoxified by mild acid and alkaline treatments. It was also dissociated by sodium deoxycholate and chromatographed on Sephadex G-75. The main fraction was reassociated by removing the surfactant by dialysis. The morphology of LPS I and LPS II was examined by electron microscopy. Summary Two types of nerve cells, namely, neurosensory and neurosecretory cells have been identified and described in the gastrodermis of Hydra pseudoligactis. The morphological criteria used for the identification of gastrodermal nerves are based on those presented previously for epidermal nerves. The third type of nerve cell in the epidermis, ganglionic cells, was not observed in these studies. Summary The structure of the salivary gland of the dipteran insect Rhynchosciara angelae in a defined stage of the larval development, characterized by the synthesis and storage of secretion product, is described. Observations were made with both Nomarski optics and electron microscopy. Filiform projections extending into the lumen of the gland were observed in the apical portion of the cells. At the basal region junctions, characterized as hemidesmosomes, were observed between the membrane of the cell and the basal lamina. The plasma membrane presents numerous infoldings into the cell increasing considerably the surface area at this region. Throughout the cytoplasm of the gland cells numerous mitochondria, Golgi complexes, microtubules, profiles of endoplasmic reticulum, secretion granules and glycogen granules were observed. Carbohydrates were detected on ultrathin sections by using the periodic acid-silver methenamine and the periodic acid-thiosemicarbazide-silver proteinate techniques.  相似文献

Summary A loose network of catecholamine-containing nerves was demonstrated with a fluorescence histochemical method (Falck-Hillarp) in the coiled portion of eccrine sweat glands in the digital pads of macaques after the injection of nialamide and noradrenaline. In the skin of untreated control animals, fluorescent fibers appear only in some of the glands. A systemic administration of reserpine and a local injection of 6-hydroxydopamine (6-OHDA) or 5-hydroxydopamine (5-OHDA) into the digital pad cause a complete disappearance of fluorescent fibers around the glands and blood vessels. Electron micrographs reveal many unmyelinated varicose axon profiles outside the basement membrane of secretory tubules. Most of these profiles contain many small agranular vesicles and a few large dense-cored vesicles (cholinergic terminal), and some have numerous small granular and a few large densecored vesicles (adrenergic terminal).The local injection of 6-OHDA causes various degenerative changes in the adrenergic terminals but the cholinergic ones and the rest of the cellular structure remain intact. Summary The ventral epidermis of adult Necturus maculosus has been studied using electron and light microscopy. Many larval characteristics of amphibian epidermal structure are retained in adult Necturus. The epidermis is a stratified epithelium consisting of four cell layers and five cell types. Major differences compared with other adult amphibians are: (1) the absence of a well defined moulting cycle together with an apparently diminished synthetic and mitotic activity in the stratum germinativum; (2) an outermost cell layer (stratum mucosum) that is unkeratinized and appears to synthesize a mucous layer; and (3) numerous large club-shaped Leydig cells which span the epidermis between the cells of the stratum germinativum and stratum mucosum. The apical region of the stratum granulosum and stratum mucosum cells shows evidence of extensive synthesis. The stratum mucosum appears to be involved in the secretion of vesicular contents onto the outermost surface of the epithelium. The external surfaces of the stratum mucosum cells possess numerous microridges which are supported by an intricate network of cytofilaments in the apical region of these cells. The significance of these features is discussed in relation to the physiology and ecology of this species.  相似文献

Antibodies targeted to a highly conserved tetradecapeptide region of the pivotal biological clock protein PER detect in the firebrat Thermobia domestica a 115-kDa protein and in the cockroach Periplaneta americana a 110-kDa protein that are present in the cytoplasm of a small set of brain cells. A similar cytoplasmic reaction occurs with antisera to the whole PER protein of Drosophila melanogaster, but these antisera also react with numerous cell nuclei. On western blots, they detect an 80-kDa antigen in T. domestica and 70- and 80-kDa antigens in P. americana. No indication of antigen translocation between cell nuclei and cytoplasm was found. Nuclear staining is maintained at a high constant level in T. domestica held at a 12:12 h light:dark photoperiod (LD) or in continuous light, but disappears rapidly in response to extended darkness. In P. americana under LD conditions, the number of immunoreactive nuclei and their staining intensity fluctuate in parallel, with maximal staining late in the day. The circadian changes are maintained in continuous light but all staining vanishes in continuous darkness. A 6-h light pulse in early night of an LD cycle induces maximal staining after about 10 h, suggesting that the effect of light on nuclear PER-like expression is indirect. The behaviour of nuclear antigens is opposite to that of the cytoplasmic PER-like proteins that persist in constant darkness and disappear in constant light. Under LD conditions, the cytoplasmic PER-like antigen cycles in T. domestica but remains at a steady level in P. americana. The sensitivity to photoregime suggests that both the nuclear and the cytoplasmic PER-like antigens are components of the biological clock.R. Závodská and H. Summary A morphogenetic process that transforms spermatids from a syncytial state to a state in which each spermatid is invested in its own membrane, is initiated at the head region of the spermatid bundle and traverses through the entire length of the bundle in the testis of Drosophila melanogaster. This process not only eliminates the syncytial bridges between spermatids but also removes unneeded organelles and the excess parts of the nuclear membrane, nucleoplasm and cytoplasm. It also brings about structural modifications to flagellar elements. The propagation of this process is seen as the caudal movement of a fusiform swelling of the spermatid bundle, 100 or more in length. Spermatids are individualized in the basal half of the swelling, whereas they remain syncytial in the apical half. The swelling increases its volume as it accumulates cytoplasmic debris while traversing the sperm bundle, from about 15 in maximum diameter in the basal testicular region to as large as 30 at the apical end where it becomes a bag of wastes. A variation of the process in a mutant stock which is known to inactivate up to half of the products of meiosis is briefly described. Summary In all blastomeres of Nassarius from 4- to 16-cell stage yolk nuclei occur. Most of them are spherical bodies, lying juxtanuclearly between the nucleus and the apical plasmalemma. Strangely they are not ultrastructurally uniform but fall into two categories (Fig. 5): Type I is a massive spherical accumulation of mitochondria embedded in a dense intermitochondrial substance, which appears to contain both granules and filaments. Type II is a ball of radially arranged small Golgi stacks clustered around a centre of Golgi vesicles and other organelles embedded in a ground cytoplasm structurally similar to the intermitochondrial substance of type I. The function of both types of yolk nuclei is unknown. These segmentation yolk nuclei have nothing to do with yolk synthesis any more. On the other hand there are no indications that yolk nuclei occurrence is correlated with the break-down of yolk because neither lipid droplets nor protein yolk granules are observed in or beside the yolk nuclei.We wish to thank Mrs. Chr. Summary An indirect immunofluorescence study of the frozen sections of the spleen of an anuran amphibian, Xenopus laevis, showed that lymphocytes bearing a small amount of immunoglobulin (Ig) were localized mostly in the white pulp of non-immunized toads. There were fewer fluorescent cells in the red pulp. In the toads hyperimmunized with human gamma globulin (HGG), cells with strong cytoplasmic fluorescence increased significantly in the outer part of the white pulp. Electron microscopy of spleens from these toads showed that plasma cells at different stages of maturation were abundant in the white pulp, whereas in the red pulp, a smaller number of maturer plasma cells were observed. These results indicate that, in contrast with its mammalian counterpart, the splenic white pulp of this anuran is the site where thymusin-dependent lymphocytes commence blast formation and transformation into plasma cells.  相似文献

Summary The excretory organs of Amphioxus occur as segmentally arranged structures throughout the pharyngeal region and may be divided into three components: the solenocytes, the renal tubule, and the renal glomerulus.The solenocytes possess foot processes that rest upon the coelomic surface of the ligamentum denticulatum. The tubular apparatus of the solenocytes consists of ten triangular rods surrounding a central flagellum. The distal end of the tubular apparatus enters branches of the renal tubule. The renal tubule eventually opens into the atrial cavity of Amphioxus.The renal glomerulus is a sinus within the connective tissue of the ligamentum dentieculatum where it connects elements of the branchial circulation with the dorsal aorta. The renal glomerulus, like other blood vessels of Amphioxus, lacks an endothelial lining.If Amphioxus is adapted to artificial sea water at different concentrations there is no change in kidney morphology suggesting that Amphioxus is either is osmotic with its environment or is osmoregulating with other organs.This work was supported by U.S. Public Health Service Grant 5-T01-GM 00582.  相似文献

Zusammenfassung Zwischen den eigentlichen Ependymzellen des Zentralkanals adulter Feuersalamander kommen amphorenartig gestaltete Elemente vor, die sich aufgrund ihrer synaptischen Kontakte mit Axonendigungen als Neurone identifizieren lassen. Diese intraependymalen Nervenzellen weisen einen apikalen Fortsatz auf, der sich mit einer warzen- oder knotenförmigen Protrusion in das Lumen des Zentralkanals erstreckt. Die Protrusion ist gewöhnlich mit stereozilienartigen Ausläufern besetzt. Die funktionelle Bedeutung der beschriebenen neuronalen Elemente konnte bisher nicht geklärt werden.
On peculiar neuronal cells in the ependyma of the canalis centralis in the salamander
Summary The ependyma of the canalis centralis of adult salamanders was examined by electron microscopy. Between the ependymal cells occur amphora-like elements identifiable as neurons by their synaptic contacts with axon terminals. These intraependymal nerve cells exhibit an apical outgrowth extending into the lumen of the canalis centralis with a wart-like or knob-like protrusion. The latter usually bears extensions resembling stereocilia. The functional significance of the neuronal elements is still unknown.

Two types of rhythmic foregut movements are described in fifth instar larvae of the moth, Manduca sexta. These consist of posteriorly-directed waves of peristalsis which move food toward the midgut, and synchronous constrictions of the esophageal region, which appear to retain food within the crop. We describe these movements and the muscles of the foregut that generate them.The firing patterns of a subset of these muscles, including a constrictor and dilator pair from both the esophageal and buccal regions of the foregut, are described for both types of foregut movement.The motor patterns for the foregut muscles require innervation by the frontal ganglion (FG), which lies anterior to the brain and contains about 35 neurons. Eliminating the ventral nerve cord, leaving the brain and FG intact, did not affect the muscle firing patterns in most cases. Eliminating both the brain and the ventral nerve cord, leaving only the FG to innervate the foregut, generally resulted in an increased period for both gut movements and muscle bursts. This manipulation also produced increases in burst durations for most muscles, and had variable effects on the phasing of muscle activity. Despite these changes, the foregut muscles still maintained a rhythmic firing pattern when innervated by the FG alone.Two nerves exit the FG to innervate the foregut musculature: the anteriorly-projecting frontal nerve, and the posteriorly-directed recurrent nerve. Cutting the frontal nerve immediately and irreversibly stopped all muscle activity in the buccal region, while cutting the recurrent nerve immediately stopped all muscle activity in the pharyngeal and esophageal regions. Recordings from the cut nerves leaving the FG showed that the ganglion was spontaneously active, with rhythmic activity continuing within the nerves. These observations indicate that all of the foregut muscle motoneurons are located within the FG, and the FG in isolation produces a rhythmic firing pattern in the motoneurons. In both freeze-etched and critical-point dried preparations examined by transmission and scanning electron microscopy, respectively, the outer surfaces of the cells of Spirillum serpens VHL assume a wrinkled appearance 10–15 min after challenge by Bdellovibrion bacteriovorus 109D. This wrinkling effect is believed (on circumstantial evidence) to be caused by the bdellovibrio's disruption of the cell wall lipoprotein of the Spirillum. With the exception of those topological changes caused by wrinkling, the outer membrane of the Spirillum cell wall retains a normal appearance as viewed in freeze-etched preparations, even after the Spirillum cell has been converted into a bdelloplast. Although the peptidoglycan layer of the Spirillum cell presumably is weakened somewhat by the invading Bdellovibrio, evidence obtained from freeze-fractured preparations of Spirillum bdelloplasts suggests that the peptidoglycan remains as a discrete cell wall layer, even though the Spirillum cell wall apparently has lost much of its rigidity. That the peptidoglycan backbone remains essentially intact, even after the Spirillum cell has been entered by the Bdellovibrio, is supported by the observation that the soluble amino sugar content of the culture medium, as determined by chemical analysis, does not rise even 5.0 h after the association of the Bdellovibrio with the Spirillum has begun.  相似文献
