A simple model is presented where, by an iterative procedure, the forces delivered by the power strokes are summed up to overcome the load. The system is moderated by the viscous hindrance. The model reproduces the features of muscle contraction as defined by the data of He et al. [1997. ATPase kinetics on activation of permeabilized isometric fibres from rabbit and frog muscle: a real time phosphate assay. J. Physiol. 501, 125-148] on rabbit psoas muscle fibres. According to the model power strokes are random. Energy summation take place if the subsequent power stroke occurs before the energy delivered by the previous power stroke is completely used. In order the sarcomere to be competent to contract initial driving force must reach a threshold whose value increases with the load. The step size of the power stroke decreases with the increase of the load. The viscous regime is simulated by the equation, where 1/k measures the viscous hindrance of the system. The relationship between water activity, viscosity and stiffness is discussed. It is concluded that the three parameters vary cyclically and that when water activity decreases (sarcomere shortening, cross-bridge attachment) viscosity and stiffness increase.  相似文献

The hydrodynamic characteristics of the polysaccharide pullulan (polymaltotriose) in water have been investigated and its molecular characteristics have been determined. Experimental values varied over the following ranges: velocity sedimentation coefficient (S): 0.9 < S < 11.2, translational diffusion coefficient (107 cm2 s−1): 1.1 < D < 14.7 and intrinsic viscosity (cm3 g−1): 6.7 < [η] < 164, which corresponds to a change in molecular weight (× 103) in the range 3.9 < MSD < 644. On the basis of analysis of the literature and our experimental data, excluded volume effects have been shown to have a prevailing influence on the chain length of these polysaccharides. The equilibrium rigidity and hydrodynamic chain diameter of pullulan were evaluated on the basis of the theory of hydrodynamic properties of a wormlike necklace, taking into account excluded volume effects. At low M (< 30 × 103) the translation friction data (in contrast to viscometric data) cannot be described in the framework of the theory of linear molecules.  相似文献

Galanin has numerous effects on gastrointestinal motility in different species; however, its cellular basis of action in mediating these effects is unclear. Dispersed gastric smooth muscle cells have been shown to possess high-affinity galanin receptors that increase cAMP and cause relaxation. Recent studies show some smooth muscle relaxants such as VIP cause relaxation by both cAMP-dependent and -independent mechanisms. It is unknown if galanin's cellular basis of relaxation is similar or different from that of VIP. To investigate galanin's relaxant effect and compare it to VIP's effect, dispersed smooth muscle cells from guinea pig stomach were prepared by collagenase digestion. The mean length in resting cells was 110 ± 2 μm and, with carbachol treatment, contracted to 89 ± 2 μm. VIP and galanin alone had no effect on cell length, but each caused a dose-dependent inhibition of carbachol-induced contraction and both had an EC50 of 3–7 nM. Galanin (1 μM) and VIP (1 μM) increased cellular cAMP from 118 ± 10 pmol/106 cells in control to 212 ± 14 and 214 ± 12 pmol/106 cells, respectively. The protein kinase A inhibitor, Rp-cAMPS, at 100 μM, completely inhibited the relaxant effect of an EC50 concentration of galanin (3 nM), but only inhibited that by VIP by 80% (p < 0.05). Adding the nitric oxide inhibitor, -NNA ( ), at 100 μM did not alter the length of resting cells or inhibit carbachol-induced contraction. However, -NNA (100 μM) decreased VIP-induced relaxation by 45%, whereas it had no effect on galanin-induced relaxation. To determine the ability of each peptide to activate nitric oxide, the incorporation of [3H]arginine into [3H]citrulline was determined. Galanin (1 μM) did not cause nitric oxide generation whereas VIP (1 μM) increased nitric oxide generation above the control by 97 ± 14% (p < 0.01). These results demonstrated that with galanin, in contrast to VIP, nitric oxide is not involved in its ability to cause gastric smooth muscle cell relaxation. The relaxant action of galanin can be accounted for completely by its ability to activate protein kinase A and therefore resembles recent results with β-adrenergic agents.  相似文献

Dihydropyridine-sensitive Ca2+ channels and the relationship between binding of dihydropyridine derivatives and depolarization-induced Ca2+ uptake have been studied in aneurally cultured human muscle. Analysis of the equilibrium binding of the 1,4-dihydropyridine derivative (+)-PN200-110 revealed a single high-affinity binding site with a Kd of 0.15±0.05 nM and a Bmax of 87±12 fmol/mg protein. Inhibition of (+)-[3H]PN200-110 binding by nitrendipine revealed a Ki of 0.8 nM for the nitrendipine-receptor complex. Depolarization of cultured human muscle achieved by elevating the K+ concentration increased the uptake 45Ca2+ which was inhibited by nitrendipine with an IC50 of 1.1 nM. This study demonstrates that aneurally cultured human muscle has dihydropyridine-sensitive voltage-dependent Ca2+ channels which are functional when the fibers are depolarized.  相似文献

The viscous forces acting on a DNA macromolecule in a fiber are calculated. The DNA polymer is modeled as an infinite rod of elliptical cross section with a grooved surface. The viscous solvent is hydrodynamic water. Appropriate boundary conditions for determining the viscous forces on the acoustic vibrational modes are discussed. The viscous forces acting on each mode are then calculated as functions of both frequency and amount of water in the fiber. The mass loading of the DNA due to water in the grooves is shown to reduce the longitudinal acoustic velocity, which agrees with recent experimental results. The longitudinal modes are determined to be underdamped and correspondingly sharp over a range of frequencies and humidities appropriate to experimental conditions. The torsional and transverse acoustic modes are still strongly overdamped.  相似文献

Shear flow, dynamic oscillation and extensional viscosity measurements were used to compare the rheological performance of several hylan samples (Mv 1.6, 3.2, 3.7, 4.7 and 5.6×106) and hyaluronan (Mv 1.4 and 1.8×106) before and after hydroxyl radicals (√OH) induced degradation. It was found that the higher molecular weight cross-linked structure of hylan was more resistant to degradation than hyaluronan and that this superior stability was reflected in various rheological parameters. The √OH degradation of the initial hylan and hyaluronan samples produced a range of polysaccharides based on hylan and hyaluronan with molecular weight covering a range from 0.5–5.6×106. The rheological parameters associated with the polysaccharides could then also be studied. Zero shear values of the complex viscosity (η*), dynamic viscosity (η′) and shear viscosity (η) were calculated using the method of Morris1 and shown to approach the same value at zero shear or frequency. An adaptation of the method of Gibbs et al.2 gave a ‘master curve’ for the storage and loss modulus of hyaluronan and hylan, which encompasses a 10-fold molecular weight and a 5-fold concentration variation. In all instances for hylan, the storage modulus predominates over the loss modulus, whereas for hyaluronan, the reverse is true, demonstrating the greater elasticity of hylan throughout the whole experimental range of molecular weights and concentrations.  相似文献

2010年对滴水湖及其引水河道沉积物中可培养有机磷细菌(OPB)、无机磷细菌(IPB)的分布变化规律进行了为期1年的调查.结果表明,闸外引水河沉积物中OPB、IPB年平均含量分别为2.76×104±1.31×104CFU·g-1和7.19×103±3.98×103CFU·g-1;闸内引水河中分别为1.05×104±3.56×103 CFU·g-1和2.54×103±8.77×102 CFU·g-1;滴水湖中则为6.69×103±2.63×103 CFU·g-1和1.66×103±5.83×102 CFU·g-1.滴水湖及其引水河道沉积物中OPB、IPB数量与水体中磷元素含量密切相关,各采样点OPB、IPB数量与总磷(TP)含量均呈极显著正相关,且均高于与总溶解性磷(TDP)、活性磷(AP)的相关系数.  相似文献

对广东珠三角地区,凡纳滨对虾低盐度高产虾池环境微生物调查结果,养殖水体异养菌平均数量为5.15×104cfu·mL-1,致病性弧菌为5.00×103cfu·mL-1,池底泥浆中异养细菌平均数量为2.41×106cfu·mL-1,致病性弧菌为1.45×105cfu·mL-1。不同虾池异养细菌的数量差别小而稳定,致病性弧菌的数量差别及波动大。淤泥较深的老化虾池,水体和泥浆中异养细菌平均分别为6.10×104cfu·mL-1和2.89×106cfu·mL-1,致病性弧菌为6.00×103cfu·mL-1和2.14×105cfu·mL-1;无淤泥虾池水体和泥浆中异养细菌平均分别为4.53×104cfu·mL-1和2.08×106cfu·mL-1,致病性弧菌为4.34×103cfu·mL-1和9.86×104cfu·mL-1。老化虾池底泥致病性弧菌明显偏高,成为老化虾池易爆发虾病的重要原因。低盐度虾池水体异养菌和致病性弧菌的数量变化,表现为养殖前期高,中后期低而稳定;池底泥浆中异养菌和致病性弧菌的数量变化,呈不规则波动。高的养殖水温对养殖环境异养菌和致病性弧菌表现抑制作用,养殖环境微生物与水体浮游藻类也有一定的关系。  相似文献

The rate of electron transfer between reduced cytochrome ƒ and plastocyanin (both purified from parsley) has been measured as k = 3.6 · 107 M−1 · s−1, at 298 °K and pH 7.0, with activation parameters ΔH = 44 kJ · mole−1 and ΔS = +46 J · mole−1 · °K−1. Replacement of cytochrome ƒ with red algal cytochrome c-553, Pseudomonas cytochrome c-551 and mammalian cytochrome c gave rates at least 30 times slower: k = 5 · 105, 7.5 · 105 and 1.0 · 106 M−1 · s−1, respectively.

Similar measurements made with azurin instead of plastocyanin gave k = 6 · 106 and approx. 2 · 107 M−1 · s−1 for reaction of reduced azurin with cytochrome ƒ and algal cytochrome respectively.

Rate constants of 115 and 80 M−1 · s−1 were found for reduction of plastocyanin by ascorbate and hydroquinone at 298 °K and pH 7.0. The rate constants for the oxidation of plastocyanin, cytochrome ƒ, Pseudomonas cytochrome c-551 and red algal cytochrome c-553 by ferricyanide were found to be between 3 · 104 and 8 · 104 M−1 · s−1.

The results are discussed in relation to photosynthetic electron transport.  相似文献

The viscous energy dissipation in a two generation model of the human bronchial tree is determined from inspiratory velocity and static pressure data obtained for large Reynolds numbers (104 < Re < 105). This dissipation is found to be an increasing function of both Re and distance downstream from the inlet of the model. The ratio of the dissipation in the model to the energy dissipation in an equivalent straight pipe system is determined. This ratio, Z*, for the model is compared to values in the literature for lower (laminar) Re. There is more dissipation in the branched model than in a straight pipe (Z* > 1) and turbulence keeps Z* at roughly a fixed value for large Reynolds numbers (104 < Re < 105). Z* values for curved pipes are also compared to the branching system values. It is found that the energy dissipation for the branched model behaves similarly to that in curved pipes.  相似文献

Sixty sea-water and marine sediment samples have been collected in the clear and dredged coral waters of a lagoon in Guadeloupe. Total counts of 10 to 100 living bacteria were obtained by counting colonies on membranes after filtration and pour plate count techniques in the clear water coral zones. Living bacterial populations ranging from 2 × 103 to 20 × 103/ml of water were detected in the dredged coral zone. Suspended sandy particulate matter resulting from dredging seems to be the direct cause of the abnormal increase in bacteria. The mean values were ≈ 103 living bacteria/ml for the coastal sea water and 104 for the brackish waters of channels in the mangrove swamp. It seems that the bacterial sediment population has been under-estimated.  相似文献

Response times with electrical fields of gellan and xanthan dry powder suspensions of 25, 32 and 53 μm average diameter and concentrations of 1.0, 1.5 and 2.0% (w/w) dispersed in commercial corn oil were optically measured through a specifically designed set up. In all cases, the delay time was proportional to 1/Ea, where E is the applied field and a is an adjustable parameter. The values of parameter a were very different from the typical value of some known electrorheolgical fluids. Response time of gellan suspensions was shorter than the one obtained for xanthan and it is comparable to the time found by using silica particles in silicon oil. Response times for cellulose were very large and the fibrillation phenomenon was negligible for E<1.0 kv/mm.

Viscosity measurements of semidiluted xanthan, gellan and cellulose suspensions (1.0 and 1.5% w/w) under the influence of electrical fields, were performed in a parallel plates rheometer. Results in the range of stress <70 Pa showed that viscosity values of gellan suspensions were larger than those obtained with xanthan or cellulose under the same applied electric field at shear rates higher than 10 s−1. However, cellulose suspensions showed larger viscosity values compared with the ones measured with xanthan and gellan suspensions at very low shear rates. Dielectric measurements of cellulose, xanthan and gellan 1.5% w/w suspensions were performed in the range 100–8×104 Hz. Results agree with a Maxwell–Wagner type relaxation model.  相似文献

The time required for the completion of the process of strand separation in λ bacteriophage DNA preparations exposed to strongly alkaline conditions in aqueous glycerol solutions was directly proportional to the viscosity of the solvent. This finding supports the idea that the rate-limiting step in the denaturation of high molecular weight DNA is the uncoiling of the strands and in the course of this uncoiling the forces arising from electrostatic and entropic factors must overcome the hydrodynamic viscous drag of the solvent resisting the translation of the polynucleotide strands.  相似文献

为明确协同提高冬小麦产量和水分利用效率的适宜灌水量和种植密度,选用大穗型品种‘泰农18’(T18)和中穗型品种‘山农22’(S22)为试验材料,设置4个灌溉水平(不灌水、每次灌水45、60、75 mm)和4个种植密度,其中泰农18选用135×104、270×104、405×104、540×104 株·hm-2,山农22选用90×104、180×104、270×104、360×104株·hm-2,研究了籽粒产量、麦田耗水特性和水分利用效率对灌水量和密度互作效应的响应。结果表明: 籽粒产量、总耗水量、土壤贮水消耗量和水分利用效率均受到灌溉水平、种植密度及两者互作效应的显著影响。每次灌水量为45 mm,泰农18种植密度为405×104株·hm-2、山农22种植密度为270×104株·hm-2时,两品种籽粒产量均达到最高,拔节后棵间蒸发量占阶段农田总耗水量的比例最小,1 m以下土壤水消耗比例、水分利用效率高。种植密度与灌溉量合理组合,有利于降低水分无效损耗,提高水分利用效率。  相似文献

Calcium-dependent regulation of tension and ATPase activity in permeabilized porcine ventricular muscle was lost after incubation with 10 mM vanadate. After transfer from vanadate to a vanadate-free, low-Ca2+ solution (pCa> 8), the permeabilized muscle produced 84.8% ± 20.1% (± S.D., n=98) of the isometric force elicited by high Ca22+ (pCa 4.5 prior to incubation with vanadate. Transfer back to a high Ca2+ solution elicited no additional force (83.2% ± 18.7% of control force). SDS-PAGE and immunoblot analysis of fibers and solutions demonstrated substantial extraction (>90%) of Troponin I (TnI). Calcium dependence was restored after incubation with solutions containing either whole cardiac troponin or a combination of TnI and troponin C subunits. This reversible extraction of troponin directly demonstrates the role of TnI in the regulation of striated muscle contractility and permits specific substitution of the native TnI with exogenously supplied protein.  相似文献

目的探讨亲缘供者外周血红细胞参数对COBE Spectra血细胞分离机的自动外周血干细胞采集程序(AutoPBSC程序)与单个核细胞采集程序(MNC程序)的影响及经验分析。 方法选取河北燕达陆道培医院2019年6月至2021年2月小红细胞亲缘供者31例45次采集为小红细胞组,选取同期非小红细胞亲缘供者51例60次采集为非小红细胞组,分别应用AutoPBSC程序和MNC程序,比较两组采集情况及采集产品相关指标。采用独立样本t检验和Mann-Whitney U检验分析2组计量资料的差异。 结果与小红细胞AutoPBSC程序组比较,小红细胞MNC程序组血小板(PLT)降低率[(25.88±15.83)﹪比(36.64±10.22)﹪]、采集效率[32.65﹪(23.60﹪,73.82﹪)比63.74﹪ (59.83﹪,68.55﹪)]、采集物体积[(158.83±34.39)比(222.91±63.9)mL]、MNC总数[(218.04±117.57)×108/L比(350.24±127.64)×108/L]、CD34+细胞总数[113.83×106/L (79.25×106/L,154.10×106/L)比233.26×106/L (177.18×106/L,392.51×106/L)]、MNC计数[(4.04±2.61)×108/kg比(5.54±2.22)×108/ kg]、CD34+计数[1.84×106/kg (1.16×106/kg,4.41×106/kg)比3.64×106/kg (2.49×106/kg,6.37×106/kg)]均升高,差异有统计学意义(P < 0.05);与非小红细胞AutoPBSC程序组比较,非小红细胞组MNC程序组采集物体积[(162.83±51.74)比(242.56±43.25)mL]升高,差异有统计学意义(P < 0.05)。 结论对造血干细胞移植供者红细胞体积偏小时,应用MNC程序采集外周血造血干细胞,比应用AutoPBSC程序更有优势。  相似文献

2H2O (99.8%) Ringer's solution greatly reduces the twitch and tetanus of frog sartorius muscle and, as specially shown here, slows the onset features of the mechanical output of the twitch by: (a) increasing the time (LR) from stimulus to start of latency relaxation; (b) slowing the developmet of the latency relaxation, and (c) greatly decreasing the rate of onset of tension development. These changes reflect effects of 2H2O on excitation-contraction coupling and they represent the critical direct effects of 2H2O on muscle since it does not depress either the action potential or the intrinsic myofibrillar contractility. The increase in LR is attributed to slowed inward electrical propagation in the T-tubule. But the critical effect of 2H2O on frog muscle is to greatly depress mobilization of activator Ca2+. The depression of the Ca2+ mobilization and of its effects on the activation of contraction evidently result from (a) a lowered rate of release of Ca2+ from the sarcoplasmic reticulum, as indicated by the slowed development of the latency relaxation, (b) a decreased amount of Ca2+ released in a twitch, and (c) a reduced speed of diffusion of the Ca2+ to the contractile filaments. The depressed mobilization of Ca2+ is apparently the essential cause of 2H2O's general depression of twitch and tetanus output.  相似文献

Low-molecular-weight chitosan were prepared using 85% phosphoric acid at different reaction temperatures and reaction time. At room temperature, the viscosity average-molecular weights (Mv) of chitosan decreased to 7.1×104 from 21.4×104 after 35 days treatment. The degradation rate decreased with increasing hydrolysis time. The yields of chitosan also continuously decreased from 68.4 to 40.2% after 35 days. At 40, 60 and 80 °C, the molecular weight decreased to 3.70×104, 3.50×104 and 2.00×104 on 8 h hydrolysis, respectively. The yields of chitosan remain at a high level compared with that at room temperature and were 86.5, 71.4 and 61.3% at 40, 60 and 80 °C treatment, respectively. The different reaction time gave chitosan with different molecular weights. At 60 °C, the molecular weight of products decreased to 7.40×104 from 21.4×104 within 4 h, then decreased slowly to 1.90×104 in 15 h. It was also found that the water-solubility of chitosan increased as the molecular weight decreased. Results show the changes in yields and molecular weight of chitooligomers were strongly dependent on the reaction temperature and reaction time.  相似文献

The physical properties of three novel acidic exopolysaccharides obtained from P. marginalis types A, B and C, one from P. ‘gingen’, one from P. andropogenis and one from P. fluorescens have been partially characterized. These EPSs were chromatographed on three serially placed SE Shodex OH pak columns covering a molar mass range for pullulans from about 4 × 107 to 1 × 103. The mobile phase was 0.05 M NaNO3. Physical measurements were performed on about 30 mg of sample for each EPS. The weight average molar mass of these EPSs ranged from about 0.71 to 2.85 × 106, the weight average intrinsic viscosity from 7.15 to 35.3 dl/ g and the radius of gyration from 62 to 123nm. The polydispersities of these EPSs ranged from 1.01 to 1.37. The large molar mass, size and viscosities of these EPSs may indicate that they have potential for use as thickeners, stabilizers, emulsifiers, and gelling agents in the food and non-food industries.  相似文献
