从苹果果实上分离获得的50余种酵母菌中筛选出了能够有效地抑制苹果青霉病(Penicilium expansum Link)的丝孢酵母(Trichosporon pullulans (Lindner.) Diddens and Lodder)、罗伦隐球酵母(Cryptococcus laurentii (Kuffer.) Aureobasidium pullulans (de Bary) Arnaud (Ach 1-1) was grown in a glucose fed-batch fermentor to 106 g dry wt l−1 in 48 h. The cells were dried in a fluidized bed dryer with a final viability of 62%. After 7 months at 4°C, the viability was 28% of the initial value (= 2.3 × 1010 c.f.u. g−1 dry matter). A protection level of 89% was achieved with the biomass preparation at 1 × 108 c.f.u. ml−1 after 28 and 7 days for apples stored respectively at 5 and 25°C against Penicillium expansum. Our process is suitable to produce large quantities of the strain Ach 1-1 as biological control agent for apple preservation.  相似文献

Biological control of apple blue mold with Pseudomonas fluorescens   总被引:3,自引:0,他引:3  
Pseudomonas fluorescens isolate 1100-6 was evaluated as a potential biological control agent for apple blue mold caused by Penicillium expansum or Penicillium solitum. Both the wild-type isolate 1100-6 and a genetically modified derivative labeled with the gene encoding the green fluorescent protein (GFP) were compared. The P. fluorescens isolates with or without GFP equally reduced the growth of Penicillium spp. and produced large zones of inhibition in dual culture plate assays. Cell-free metabolites produced by the bacterial antagonists reduced the colony area of Penicillium isolates by 17.3% to 78.5%. The effect of iron chelate on the antagonistic potential of P. fluorescens was also studied. The use of iron chelate did not have a major effect on the antagonistic activity of P. fluorescens. With or without GFP, P. fluorescens significantly reduced the severity and incidence of apple decay by 2 P. expansum isolates after 11 d at 20 degrees C and by P. expansum and P. solitum after 25 d at 5 degrees C when the biocontrol agents were applied in wounds 24 or 48 h before challenging with Penicillium spp. Populations of P. fluorescens labeled with the GFP were determined 1, 9, 14, and 20 d after inoculation at 5 degrees C. The log CFU/mL per wound increased from 6.95 at the time of inoculation to 9.12 CFU/mL (P < 0.05) 25 d after inoculation at 5 degrees C. The GFP strain did not appear to penetrate deeply into wounds based on digital photographs taken with an inverted fluorescence microscope. These results indicate that P. fluorescens isolate 1100-6 could be an important new biological control for apple blue mold.  相似文献

The capability of yeast Trichosporon sp., an antagonist isolated from peach fruit, in biological control was evaluated in apple ( Malus domestica Borkh. cv. Fuji) fruits, when inoculated with different concentrations of Botrytis cinerea Pers. and Penicillium expansum (Link) Thom, as well as in combination with calcium and fungicide. The concentrations of the yeast cells and pathogen spores obviously influenced disease incidence and lesion development in apples. There was a significant negative correlation between concentrations of the yeast cells and infectivity of the pathogens. When the yeast cell suspensions reached the concentration of 108 colony-forming units (CFU)/mL, there was no infection caused by B. cinerea and P. expansum with spore concentrations below 106 spores/mL in apple fruits. The yeast at concentrations of 106-107 CFU/mL in combination with fungicide (iprodione at 50 μL/L) provided control of decay caused by B. cinerea and P.expansum better than separate application. Effect of controlling gray mould and blue mould rots was enhanced when Trichosporon sp., even at low concentration of 105 CFU/mL, was applied in the presence of 1%-2% CaCl2 in an aqueous suspension.  相似文献

The adhesion and germination of conidia of nine fungal species were assessed on polycarbonate membranes or on the skin of apple fruit in sealed glass bottles injected or not injected with acetate esters. Adhesion was determined after dislodging conidia from surfaces using a sonication probe. Adhesion and germination of conidia of Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Penicillium citrinum, Penicillium claviforme, or Trichoderma sp. on membranes after 48 h were not increased in a 1.84 microg mL(-1) headspace of butyl acetate (BA), ethyl acetate, hexyl acetate, 2-methylbutyl acetate, pentyl acetate, or propyl acetate. Adhesion and germination of Botrytis cinerea, Penicillium expansum, and Penicillium roquefortii conidia were stimulated by all esters. Only conidia of B. cinerea and P. expansum exhibited increased adhesion and germination on the skin of apple fruit in bottles exposed to 0.92 microg mL(-1) of BA. Only conidia of B. cinerea and P. expansum produced decay in inoculated puncture wounds on fruit. Freshly made puncture wounds or 24-h-old puncture wounds in fruit were more adhesive than the unpunctured skin of fruit to conidia of B. cinerea or P. expansum. Fresh wounds were more adhesive to both fungi than 24-h-old puncture wounds. The skin and wounds of fruit were as adhesive to B. cinerea conidia as they were to P. expansum conidia. A 4-h exposure to 1.43 microg mL(-1) of BA increased adhesion of B. cinerea and P. expansum conidia in 24-h-old wounds. Results suggest that acetate-ester stimulation most likely is not a rare phenomenon in the fungi. For nutrient-dependent decay pathogens of apple fruit, acetate esters may be an alternative chemical cue used to maintain adhesion of conidia to wound surfaces.  相似文献

The modes of action of the antagonistic yeast Pichia anomala (strain K) have been studied; however, thus far, there has been no clear demonstration of the involvement of exo-beta-1,3-glucanase in determining the level of protection against Botrytis cinerea afforded by this biocontrol agent on apple. In the present study, the exo-beta-1,3-glucanase-encoding genes PAEXG1 and PAEXG2, previously sequenced from the strain K genome, were separately and sequentially disrupted. Transfer of the URA3-Blaster technique to strain K, allowing multiple use of URA3 marker gene, first was validated by efficient inactivation of the PaTRP1 gene and recovery of a double auxotrophic strain (uracil and tryptophan). The PAEXG1 and PAEXG2 genes then were inactivated separately and sequentially with the unique URA3 marker gene. The resulting mutant strains showed a significantly reduced efficiency of biocontrol of B. cinerea when applied to wounded apple fruit, the calculated protection level dropping from 71% (parental strain) to 8% (mutated strain) under some experimental conditions. This suggests that exo-beta-1,3-glucanases play a role in the biological control of B. cinerea on apple. Furthermore, biological control experiments carried out in this study underline the complexity of the host-antagonist-pathogen interaction. Two experimental parameters (yeast inoculum concentration and physiological stage of the fruit) were found to influence dramatically the protection level. Results also suggest that, under some conditions, the contribution of exo-beta-1,3-glucanase to biological control may be masked by other modes of action, such as competition.  相似文献

AIM: Test of Bacillus subtilis strain GA1 for its potential to control grey mould disease of apple caused by Botrytis cinerea. METHODS AND RESULTS: GA1 was first tested for its ability to antagonize in vitro the growth of a wide variety of plant pathogenic fungi responsible for diseases of economical importance. The potential of strain GA1 to reduce post-harvest infection caused by B. cinerea was tested on apples by treating artificially wounded fruits with endospore suspensions. Strain GA1 was very effective at reducing disease incidence during the first 5 days following pathogen inoculation and a 80% protection level was maintained over the next 10 days. Treatment of fruits with an extract of GA1 culture supernatant also exerted a strong preventive effect on the development of grey mould. Further analysis of this extract revealed that strain GA1 produces a wide variety of antifungal lipopeptide isomers from the iturin, fengycin and surfactin families. A strong evidence for the involvement of such compounds in disease reduction arose from the recovery of fengycins from protected fruit sites colonized by bacterial cells. CONCLUSIONS: The results presented here demonstrate that, despite unfavourable pH, B. subtilis endospores inoculated on apple pulp can readily germinate allowing significant cell populations to establish and efficient in vivo synthesis of lipopeptides which could be related to grey mould reduction. SIGNIFICANCE AND IMPACT OF THE STUDY: This work enables for the first time to correlate the strong protective effect of a particular B. subtilis strain against grey mould with in situ production of fengycins in infected sites of apple fruits.  相似文献

Geosmin, an off-flavour of some rotten grapes, has been implicated in wine defects. Botrytis cinerea and Penicillium expansum were the most common among the numerous microorganisms isolated from rotten grapes. P. expansum produces geosmin on model media but not healthy grape juice. However, geosmin synthesis by P. expansum was demonstrated in grape juice and on crushed grapes that had been pre-cultured with certain B. cinerea strains. 34 out of 156 B. cinerea strains ([bot +] phenotype) isolated from the centre of grape bunches were able to induce high geosmin production, up to 494 ng/l, by P. expansum in grape juice. A study of the impact of grape juice composition on geosmin synthesis by P. expansum revealed the importance of nitrogen composition, particularly amino-acid deficiency. Metabolism of amino acids by B. cinerea was shown to be favourable to geosmin synthesis by P. expansum. However, the amino-acid and ammonium concentrations in grape juices pre-cultured with B. cinerea [bot -] and [bot +] strains were very similar implying that other factors are involved as well. Indeed, an ethanol-precipitable fraction, probably a polysaccharide, synthesized by B. cinerea [bot -], but not [bot +] strains, inhibited geosmin production by P. expansum.  相似文献

AIM: To improve the efficacy of Candida sake by adding ammonium molybdate to control post-harvest decay in Golden Delicious apples. METHODS AND RESULTS: In laboratory trials, C. sake 2 x 10(6) cfu ml(-)1 plus 5 mmol l(-1) ammonium molybdate greatly reduced Penicillium expansum, Botrytis cinerea or Rhizopus stolonifer in apples stored at 20 degrees C for 7 days, and reduced by more than 90% blue and grey mould in apples stored at 1 degrees C for 60 days. The consistency of these results was maintained in semi-commercial trials at 1 degrees C in air and in a low oxygen atmosphere for 120 days. The pre-harvest application of C. sake 2 x 10(7) cfu ml(-1) plus 1 mmol l(-1) ammonium molybdate did not improve post-harvest biocontrol of blue mould. The population of C. sake significantly decreased in the presence of ammonium molybdate in apple wounds. CONCLUSION: The addition of ammonium molybdate at 5 mmol l(-1) to C. sake enhanced the efficacy of the antagonist to control post-harvest diseases on apples. SIGNIFICANCE AND IMPACT OF THE STUDY: Ammonium molybdate significantly reduces the amount of C. sake biomass required to achieve post-harvest disease control, with a consequent reduction in costs. AIMS: To investigate the effects of internal trehalose on viability and biocontrol efficacy of antagonistic yeast Cryptococcus laurentii under stresses of low temperature (LT), controlled atmosphere (CA) and freeze drying. METHODS AND RESULTS: The content of trehalose in C. laurentii was increased by culturing the yeast in trehalose-containing medium. Compared with yeast cells with low trehalose level, the yeast cells with high level of internal trehalose not only obtained higher viability, but also showed higher population and better biocontrol efficacy against Penicillium expansum on apple fruit both at 1 degrees C and in CA condition (5% O(2), 5% CO(2), 1 degrees C). After freeze drying, survival of the yeast with high trehalose level was markedly increased when stored at 25 degrees C for 0, 15 and 30 days. Meanwhile, high integrity of plasma membrane was detected in the freeze-dried yeast with high trehalose level by propidium iodide staining. CONCLUSIONS: Induced accumulation of internal trehalose could improve viability and biocontrol efficacy of C. laurentii under stresses of LT and CA. Moreover, survival of the yeast was also increased as internal trehalose accumulation after freeze drying, and one of the reasons might be that trehalose gave an effective protection to plasma membrane. SIGNIFICANCE AND IMPACT OF THE STUDY: The results of this experiment show a promising way to improve the biocontrol performance of antagonistic yeasts under the commercial conditions.  相似文献

Candida oleophila strain O was previously selected for its high and reliable antagonistic activity against Botrytis cinerea and Penicillium expansum, two important wound pathogens on post-harvest apples. The application of these antagonistic strains on wound pathogens of Citrus was more recently undertaken. The efficacy of yeast (applied at several concentrations from 10(5) to 10(8) CFU/ml) was assessed against P. digitatum and P. italicum inoculated after one hours (at a concentration of 10(5), 106 and 10(7) spores/ml) on 'Clementine' and 'Valencia late' varieties. The protective levels were positively correlated with high concentration of antagonist and low concentration of pathogen. The antagonistic activity of this strain was also dependent on the incubation time before pathogen inoculation. The protective level increased with time between application of the antagonist and inoculation of fungal spores. Finally, the efficacy of biomass of C. oleophila strain O (produced at an industrial scale), and two different formulations of that biomass was assessed in comparison with fungicidal treatment (Thiabendazole) under semi-practical conditions against P. digitatum. This efficacy of strain O (whatever its formulation) was statistically comparable to that for TBZ at commercial dose, indicating that both formulations could be used as an alternative for conventional fungicide in postharvest treatments.  相似文献

A real-time PCR assay using a 3'-Minor Groove Binding (MGB) probe was developed for specific detection and monitoring of Candida oleophila (strain O), a biocontrol agent against Botrytis cinerea and Penicillium expansum, on harvested apples. The application of the RAPD technique on C. oleophila strains followed by reproducible sequence characterized amplified region (SCAR) amplifications allowed the identification of a semi-specific fragment of 244 bp, observed in the profiles of strain O and three other C. oleophila strains. After sequencing, polymorphisms (3%) were observed between the strain O sequence and the three other sequences. A 3'-Minor Groove Binding probe was designed to specifically match a region of the strain O sequence and was able to discriminate a single base mutation or a two-base difference in the corresponding sequences of the non-target strains. This specific detection method was applied to monitor strain O population, recovered by a washing buffer, from harvested apples. Population densities were calculated using an external standard curve consisting in a serial dilution of strain O cells in the washing buffer from untreated apples. Linearity in the standard curve was kept between 1.64 x 10(2) and 1.64 x 10(5) cfu cm(-2) of apple surface. During a first practical experiment, the calculated population densities were similar to those obtained by plating on semi-selective media. This new real-time PCR method is a promising tool to monitor quickly and specifically strain O population on apple surface in middle- or large-scale experiments.  相似文献

More than 200 yeasts were selectively isolated from microbial populations on the surface of different fruits. Fifty of these isolates were tested against blue mould ( Penicillium expansum ) on wounded apples. Isolates LS-11 of Rhodotorula glutinis and LS-28 of Cryptococcus laurentii were the most effective antagonists. They were further evaluated at 20IC on different fruits (apples, pears, strawberries, kiwi fruits and table grapes) against several of the main post-harvest pathogens ( Botrytis cinerea, Penicillium expansum, Rhizopus stolonifer and Aspergillus niger ) and at 4IC on apples inoculated with P. expansum . At 20IC the antagonists significantly reduced rot incidence and showed a wide range of activity on different hostpathogen combinations; isolate LS-28 exhibited a higher and more stable activity than LS-11. Both yeasts were also effective against P. expansum in cold storage conditions. Populations of the two yeasts were assessed on wounded and unwounded surfaces of apples kept at both 20 and 4IC. At either temperature, isolate LS-28 reached greater densities in wounded tissues than LS-11, but had a lower ability to colonize unwounded apple skin. The two yeasts were able to grow in culture at temperatures ranging from 0 to 35IC. In assays performed in vitro at 24IC, the antagonists showed low sensitivity towards several fungicides commonly applied on fruits and vegetables.  相似文献

Penicillium expansum and Botrytis cinerea are among the pathogens most frequently affecting apples and grapes after harvest, respectively. We studied the behaviour of these moulds when subjected to different concentrations of methanol (MeOH) and dimethyl sulfoxide (DMSO) as a alternative method to fungicides in controlling postharvest decay of horticultural products. The experiments were performed with 5 cm Petri dishes containing PDA amended with 0, 5, 10, 20, 30, 40 or 50 microL/mL of the two tested chemicals. Freshly prepared conidia of B. cinerea and P. expansum were sown onto the media and then kept into an incubation chamber at 21 degrees C up to 3 and 6 days, respectively. Daily, the colony forming units (cfu), the colony diameter and the degree of sporification were monitored. Compared to the control, both chemicals affected the growth rate of the two pathogens. The P. expansum and B. cinerea cfu value was not significantly inhibited but the colony diameter and the sporification degree decreased when concentration was raised. B. cinerea cultured on DMSO showed a significant drop of sporification up to the tested concentration of 10 microL/mL, and a complete inhibition of cfu when the concentration was higher than 20 microL/mL.  相似文献

The yeast Candida oleophila (strain O) presents a high level of protective activity against Botrytis cinerea (gray mold) on postharvest apples. The cDNA-AFLP technique allows the comparison of mRNA populations extracted from cells grown in different conditions. In order to isolate yeast genes potentially involved in biological control properties, that technique was applied on strain O cells growing on apple wounds. The biological control properties of 8 C. oleophila strains and strain O were assessed in order to compare the gene expression of a non antagonistic strain against gene expression of strain O. In the absence of a non-antagonistic strain, an other comparison model was designed. It was based on the growth of strain O in different in situ conditions: strain O applied on apple wounds (O), strain O applied on apple wounds in presence of B. cinerea (B) and B. cinerea alone on apple wounds (F). A recovering technique, based on the washing of cells in the wound and a RNA extraction method followed by a DNase treatment were optimised before cDNA-AFLP application. Thirteen primer pairs were used. Their application resulted in an average of 54 and 55 bands for O and B respectively whereas no bands were observed for F. Among these bands, 8 were expressed more intensely in presence of the pathogen (1.1% of the fragments).  相似文献

Botrytis cinerea has been found to be highly pathogenic to 'Chardonnay' and 'Pinot noir' cultivars of the grape-vine producing the characteristic grey mould symptoms within 7 days of inoculation to the vitro-plants. The yeast Pichia anomala (strain FY-102), isolated from apple skin, was found to be antagonistic to B. cinerea as it completely inhibited the appearance of the grey mould symptoms when grown together. The yeast was responsible for morphological changes such as coagulation and leakage of the cytoplasm of B. cinerea. The pathogen, when applied together with P. anomala, failed to bring about the grey mould symptoms on the grape-vine, suggesting that the yeast could control the expression of this disease. An account of the interaction between B. cinerea and P. anomala, as well as the sequences of the complete ITS region of the ribosomal DNA of the yeast are described here.  相似文献

【目的】从银杏(Ginkgo biloba)茎叶中分离鉴定内生细菌, 测定其体外抑菌活性及对辣椒果疫病的防治效果。【方法】采用平板对峙法筛选出对辣椒疫霉菌(Phytophthora capsici)有拮抗作用的内生细菌, 并用平板对扣法测定其中一株防治效果较好的内生细菌产生的挥发性物质对辣椒疫霉菌生长的影响。通过生防菌液和病原菌孢子悬浮液喷雾接种辣椒果测定该菌株对辣椒果疫病的防治效果。基于形态特征、生理生化特性、16S rDNA和gyrA基因序列同源性分析鉴定该生防菌株。【结果】从银杏的茎和叶中分离获得9株内生细菌。平板对峙生长试验结果表明, 菌株W5对供试的辣椒疫霉菌、稻瘟病菌(Pyricularia grisea)、水稻纹枯菌(Rhizoctonia solani)、黄瓜枯萎病菌(Fusarium oxysporum)、荔枝霜疫霉菌(Peronophythora litchi)、荔枝酸腐菌(Geotrichum candidum)均有抑制作用, 其中对辣椒疫霉菌、稻瘟病菌和荔枝霜疫霉菌的抑菌效果显著, 抑菌率分别为88.9%、86.3%和90.2%。其产生的挥发性物质能明显抑制辣椒疫霉菌菌丝的生长。对辣椒采后果疫病的防治效果表明, 先喷雾接种W5菌悬液24 h后再接种辣椒疫霉病菌孢子悬浮液的防治效果最好, 可将辣椒果的保鲜期延长2?3 d。该菌株被鉴定为解淀粉芽胞杆菌(Bacillus amyloliquefaciens)。【结论】获得了一株对植物病原菌物有良好防治效果的银杏内生解淀粉芽胞杆菌W5, 对辣椒采后果疫病及其他病原真菌的防治具有潜在应用价值。  相似文献

AIMS: The aim of this study was to assess the opportunities of Penicillium expansum to develop and produce patulin in apples during cold storage and in the steps prior to processing of apple products. METHODS AND RESULTS: Two lots of apples var. Golden with different ripeness degree were used. Half of each lot was fungicide treated. Apples were inoculated with P. expansum and stored at 1 degrees C for 6 weeks. The extent of lesions and patulin accumulation both at the end of cold storage and after 3 days at 20 degrees C were assessed. Short storage at 20 degrees C aimed to simulate the transport and storage steps at room temperature before processing. Lesion size significantly increased during the storage at 20 degrees C. An interaction between fungicide treatment and ripeness degree was found; efficiency of fungicide treatment was higher for ripe apples. Although lesions were evident after cold storage, no patulin was detected. Patulin was detected only when fruits were further stored at 20 degrees C. Neither ripeness degree nor fungicide treatment affected patulin accumulation. CONCLUSIONS: Cold storage periods of 6 weeks do not lead to patulin accumulation. SIGNIFICANCE AND IMPACT OF THE STUDY: Shortening preprocessing times at warm temperatures would result into a reduction in patulin content at initial steps of fruits entering the processing plants.  相似文献

【背景】苹果树腐烂病发生面积广、危害严重,目前对于该病的防治以化学防治为主,但存在一定的弊端,因此迫切需要寻找一种对该病高效、安全的防治措施。【目的】从敦煌地区盐碱土壤中筛选出对苹果树腐烂病有良好抑制作用的芽孢杆菌菌株。【方法】采用平板对峙培养法和菌丝生长速率法对盐碱土中分离筛选的芽孢杆菌进行拮抗苹果树腐烂病菌的筛选,并对优良拮抗效果菌株进行离体组织防效测定、发酵培养基选择以及结合培养形态、生理生化及16S rRNA基因序列分析进行菌种鉴定。【结果】初筛阶段菌株7-2-1-2对病原菌的抑制率为86.18%,复筛试验中发酵液对病原菌的抑制率为73.2%;在离体枝条创面涂抹菌株7-2-1-2发酵原液后,苹果树腐烂病菌的生长可被完全抑制;菌株7-2-1-2的最优生长培养基为NBY培养基;经初步鉴定,菌株7-2-1-2为枯草芽孢杆菌亚种(Bacillus subtilis subsp. spizizenii)。【结论】菌株7-2-1-2对苹果树腐烂病有较好的生防效果,为该病生防菌剂的选择提供了必要的菌种资源。  相似文献
