Previous studies of Partula land snails from the Society Islands, French Polynesia, have shown that populations within species are highly differentiated in terms of their morphology, behaviour, ecology and molecular genetic variation. Despite this level of variability, differences between species are sometimes small, possibly reflecting the fact that reproductive isolation is not always complete and there exists the opportunity for genetic exchange between taxa through hybridization. The present study uses sequence data from a mitochondrial gene to further investigate genetic variation in Society Island Partula. Most populations are found in this study to be highly differentiated, but within individual species there seems to be no simple relationship either between genetic distance and geographical proximity, or between variation in mitochondria and that in allozymes or morphological characteristics. Among species there appears to be no simple correlation between degrees of reproductive isolation and genetic relatedness according to mitochondrial DNA. The results suggest that past events as well as ongoing drift and selection may have been important in affecting patterns of variation. Similarities among species at specific localities suggest that there must have been some genetic exchange in the past, although this may not necessarily reflect ongoing rates of hybridization. The discrepancy between results for different markers probably reflects the differential effects of drift and selection on mitochondrial and nuclear genes.  相似文献

Genetic stock structure is atypical in tuna species, with most species demonstrating geographically‐broad, panmictic populations. Here, genetic data suggest a distinct pattern for Thunnus tonggol across the Indo‐Pacific region. The genetic variation in the coastal tuna T. tonggol sampled from across the South China Sea was examined using the highly variable mitochondrial DNA displacement loop (D‐loop) gene region. One hundred and thirty‐nine specimens were sampled from four locations in Indonesia, Vietnam and the Philippines. Phylogenetic reconstruction of genetic relationships revealed no significant ?ST statistics and hence no population structure within the South China Sea. However, subsequent analysis with sequence data from coastal northwest India infers discrete genetic stocks between the Indian Ocean and the South China Sea. Consistent with previous genetic analyses of tuna species in the Indo‐Pacific, the findings in this study infer no population structure within each basin, but rather show a significant partitioning across the wider region. Furthermore, these results have implications for the management of the commercially valuable Thunnus tonggol across national boundaries, and thus requiring collaboration among countries to ensure its sustainable use.  相似文献

Anopheles gambiae and An. arabiensis are mosquito species responsible for most malaria transmission in sub-Saharan Africa. They are also closely related sibling species that share chromosomal and molecular polymorphisms as a consequence of incomplete lineage sorting or introgressive hybridization. To help resolve these processes, this study examined the partitioning of mtDNA sequence variation within and between species across Africa, from both population genetic and phylogeographic perspectives. Based on partial gene sequences from the cytochrome b, ND1 and ND5 genes, haplotype diversity was high but sequences were very closely related. Within species, little or no population subdivision was detected, and there was no evidence for isolation by distance. Between species, there were no fixed nucleotide differences, a high proportion of shared polymorphisms, and eight haplotypes in common over distances as great as 6000 km. Only one of 16 shared polymorphisms led to an amino acid difference, and there was no compelling evidence for nonneutral variation. Parsimony networks constructed of haplotypes from both species revealed no correspondence of haplotype with either geography or taxonomy. This trend of low intraspecific genetic divergence is consistent with evidence from allozyme and microsatellite data and is interpreted in terms of both extensive gene flow and recent range expansion from relatively large, stable populations. We argue that retention of ancestral polymorphisms is a plausible but insufficient explanation for low interspecific genetic divergence, and that extensive hybridization is a contributing factor.  相似文献

Six endemic and two widely distributed species living in Lake Ohrid were studied. In general, these hermaphroditic animals displayed no signs of departure from Hardy-Weinberg equilibrium. Genetic variation in all but one of the endemic species was of the same extent as that in geographically wide ranging invertebrates. On the other hand, the Lake Ohrid population of the common European species Dendrocoelum lacteum was monomorphic at all loci examined. D. sanctinaumi, one of the endemic species, exhibited a clear genetic subdivision into spring and littoral subpopulations. The genetic differentiation of Crenobia alpina alpina and C. a. montenigrina proved commensurable to that of well separated species from other genera. The data suggest that the separation of particular lineages in the set of Lake Ohrid endemics was widely dispersed over time.  相似文献

Proposals for genetic thresholds for species delimitation assume that simple genetic data sets (e.g. mitochondrial sequence data) are correlated with speciation; i.e. such data sets accurately reflect organismal lineage divergence. We used taxonomically stratified phenotypic levels of differentiation (populations, subspecies and species) among nine avian lineages using paired, trans-Beringian samples from three lineages each in three orders (Anseriformes, Charadriiformes, and Passeriformes) to test this assumption. Using mitochondrial DNA sequence data and nuclear genomic data (amplified fragment length polymorphisms), we found a lack of concordance between these two genomes in their respective estimates of divergence and little or no relationship between phenotype (taxonomic relatedness) and genetic differentiation between taxon pairs. There are several possible reasons for the discord observed (e.g. selection on one of the genomes or perhaps lineage sorting), but the implications are that genetic estimates of lineage divergence may not be correlated with estimates from other parts of the genome, are not well correlated with the speciation process and are thus not reliable indicators of species limits.  相似文献

Twenty-two loci were re-evaluated to assess genetic variation and differentiation in three natural populations (two from Gran Canaria and one from Tenerife) of Gelidium (G. canariensis and G. arbuscula). The new data using exclusively the diploid subpopulation gene frequencies confirm that dispersal was restricted over short distances for the two species, but contrary to previous conclusions, the data reveal that these two closely related species differed markedly by their mating systems and patterns of genetic differentiation. Genetic differentiation among populations was twice as high in G. arbuscula as in G. canariensis. It was confirmed that the mean way of reproduction is asexual in G. arbuscula and a discussion included as to how clonal propagation may explain the difference in haploid and diploid allele frequencies in this species. There was no evidence for asexual reproduction in G. canariensis. Heterozygote deficiency could be explained simply by spatial sub-structuring within populations. The importance of the sampling design in determining the level and pattern of genetic differentiation within a species is discussed. 用PCR产物直接测序法对入侵害虫西花蓟马和其他8种蓟马的线粒体 COⅠ基因433 bp片段测序,获得62个个体的序列。分子数据分析显示: 种内个体间平均遗传距离在0~0.005之间,2003年在北京发现的西花蓟马与欧洲等地区报导的西花蓟马不存在明显的遗传差异; 9种蓟马种间平均遗传距离为0.213。构建的NJ树可以很好的显示蓟马的聚类,物种各单元型最初分支自展值均达到100%。结果表明,基于PCR及直接测序技术的分子鉴定可以达到准确鉴定蓟马物种之目的。  相似文献

Analyses among animal species have found that reproductive isolation increases monotonically with genetic distance, evolves more quickly for prezygotic than postzygotic traits, and is stronger among sympatric than allopatric species pairs. The latter pattern is consistent with expectations under the reinforcement hypothesis. To determine whether similar trends are found among plant species, patterns of reproductive isolation (postpollination prezygotic, postzygotic, and "total" isolation) in three plant genera (Glycine, Silene, Streptanthus) were examined using data from previously published artificial hybridization experiments. In Silene, all measures of reproductive isolation were positively correlated with genetic distance. In contrast, in Glycine and Streptanthus, correlations between reproductive isolation and genetic distance were weak or nonsignificant, possibly due to the influence of biologically unusual taxa, variable evolutionary forces acting in different lineages, or insufficient time to accumulate reproductive isolation. There was no evidence that postpollination prezygotic reproductive isolation evolved faster than postzygotic isolation in Glycine or Silene. We also detected no evidence for faster accumulation of postmating prezygotic isolation between sympatric than allopatric species pairs; thus we found no evidence for the operation of speciation via reinforcement. In Silene, which included six polyploid species, results suggest that changes in ploidy disrupt a simple monotonic relationship between isolation and genetic distance.  相似文献

By surveying variation at allozyme loci in several phytophagous lepidopteran species (Geometridae), we have tested two hypotheses about the relationship of genetic variation to environmental heterogeneity: (1) that allozyme polymorphisms may exist because of associations between genotypes and "niches" (different host plants, in this instance), and (2) that the overall genetic variation of a species is correlated with environmental heterogeneity (or breadth of the species' overall ecological niche).—Genetic differentiation among samples of oligophagous or polyphagous species taken from different host species was observed in one of three species, at only one of seven polymorphic loci. The data thus provide no evidence for pronounced genetic substructuring, or "host race" formation in these sexually reproducing species, although host plant-genotype associations in a parthenogenetic moth give evidence of the potential for diversifying selection.—In a comparison of allozyme variation in polyphagous ("generalized") and oligophagous ("specialized") species, heterozygosity appeared to be higher in specialized species, at all polymorphic loci but one. It is possible that this unexpected result arises from a functional relation between breadth of diet and genetic variation.  相似文献

A north/south discontinuity along the northeastern coast of North America in the genetic structure of the American lobster ( Homarus americanus ) was detected using a suite of 13 microsatellite loci assessed using spatial analyses. Population genetic data laid over existing data on physiographic changes and sea-surface temperatures were used to reconstruct the Pleistocene distribution of this species. A postglacial northern-edge colonization model best explains the relative genetic homogeneity of the northern region compared to the southern region centred in the Gulf of Maine. Population genetic analyses identified significant structure (range of standardized theta 0–0.02) but no significant evidence for isolation by distance. The novel application of spatial genetic analyses to a marine species allowed us to interpret these results by providing a greater insight into the evolutionary factors responsible for shaping the genetic structure of this species throughout is natural range.  相似文献

The North American wood turtle, Glyptemys insculpta, is a semi-aquatic species that is considered rare, threatened, or endangered over much of its range. In this study, a particularly vulnerable peripheral isolate population in Iowa has been monitored over a period of 7 years. Population census size, estimated from mark-recapture data, and age structure determined from morphology are compared with genetic variation assessed using microsatellites. For reference, the genetics and demographics of this peripheral isolate are compared to data from a more dense population nearer the core of the species range in West Virginia. Geneflow between the Iowa population and a nearby population in Minnesota also is assessed. Genetic data indicate that the Iowa population is isolated, unique, and diverse. Although the Iowa population has lower allelic richness, lower heterozygosity, and smaller genetic effective population size than does the West Virginia population, the difference is not dramatic despite its lower population size, position at the periphery of the species range, and biogeographic history. The Iowa population is not inbred, and there is no genetic signature of a recent population bottleneck. However, interpretations of recent population dynamics based on genetic data may be unduly encouraging in long-lived species such as G. insculpta. Field data suggest a nearly complete lack of recruitment in Iowa. A number of environmental and anthropogenic factors, including recent increases in summer flooding during egg incubation, may have a more negative impact on the Iowa population than on the West Virginia population.  相似文献

The golden takin is an endangered species, listed as a First Grade Protected animal and found only in the Qinling Mountains in China. A great deal of research on the golden takin's living habitat, population size, and home range has been conducted. Here, we employed sequence analysis of the mitochondrial DNA control region to study the genetic diversity of the golden takin from three separate nature reserve parks in the Qinling Mountains. We also compared the results of our study with previously published data on the genetic diversity of mixed takin species located in the Qinling Mountains and the Minshan area. Based on 62 sampled golden takin individuals, we found an overall mean genetic haplotype diversity of 0.687. There is no significant geographic genetic diversity across different golden takin populations within the Qinling Mountains. However, we did show significant diversity between golden takin from the Qinling Mountains and from Minshan. These original data provide a foundation for the genetic diversity of golden takin, and will yield comprehensive information for better supporting the management in the national reserve parks.  相似文献

The species we have studied the spatiotemporal genetic change in the northern dragonhead, a plant species that has experienced a drastic population decline and habitat loss in Europe. We have added a temporal perspective to the monitoring of northern dragonhead in Norway by genotyping herbarium specimens up to 200 years old. We have also assessed whether northern dragonhead has achieved its potential distribution in Norway. To obtain the genotype data from 130 herbarium specimens collected from 1820 to 2008, mainly from Norway (83) but also beyond (47), we applied a microfluidic array consisting of 96 SNP markers. To assess temporal genetic change, we compared our new genotype data with existing data from modern samples. We used sample metadata and observational records to model the species'' environmental niche and potential distribution in Norway. Our results show that the SNP array successfully genotyped all included herbarium specimens. Hence, with the appropriate design procedures, the SNP array technology appears highly promising for genotyping old herbarium specimens. The captured genetic diversity correlates negatively with distance from Norway. The historical‐modern comparisons reveal similar genetic structure and diversity across space and limited genetic change through time in Norway, providing no signs of any regional bottleneck (i.e., spatiotemporal stasis). The regional areas in Norway have remained genetically divergent, however, both from each other and more so from populations outside of Norway, rendering continued protection of the species in Norway relevant. The ENM results suggest that northern dragonhead has not fully achieved its potential distribution in Norway and corroborate that the species is anchored in warmer and drier habitats.  相似文献

The echinoid, Asthenosoma ijimai belonging to the order Echinothurioida from Japanese waters shows the geographical variation in morphological and ecological characters. The echinothurioid from Ryukyu Islands in southern Japan is cleary different from that of Sagami Bay and Suruga Bay in the middle part of Japan at non-molecular level.Their phylogenetic and taxonomic relationships were studied at the molecular level by allozyme analysis. The results demonstrated that the two echinothurioids from Ryukyu Islands and Sagami Bay do not share gene pools with each other, and they were fixed for different alleles at five genetic loci (Mdh, G6pd, Po, Alk-3 and Est-7) in a total of 23 enzyme genetic loci scored. This indicates no gene flow between the two echinothurioids, and is a molecular evidence for that they are reproductively isolated and genetically distinct species. The Nei's genetic distance (D=0.181) between the two were significantly higher than those between conspecific local populations, and comparable to those between closely related species in many other animals containing echinoderms. The present molecular data are well consistent with the non-molecular evidence from morphology, developmental biology and ecology. Putting these data together, we propose that the two echinothurioids should be classified as two sibling species of the genus Asthenosoma and would like to give the following scientific names: the echinothurioid species from Sagami Bay is Asthenosoma ijimai and that from Ryukyu Islands is A. ijimai R.  相似文献

The genus Epilobium L. (Family Onagraceae) with about 200 species is taxonomically complicated due to wide inter-specific gene flow. There is no report on biosystematics of Epilobium species in Iran, therefore we studied 5 species of this genus using a combination of morphological, molecular (ISSR) and palynological data to delimit these species. All data revealed high degree of morphological and genetic overlap among the studied species. However, they could be separated when all studied features were used together. Morphological characters could delimit 3 species while genetic data separated them in two major groups. However, palynological data could delimit all five species studied. The species relationship was highly compatible in all three data sources used.  相似文献

Non-vagile taxa such as terrestrial molluscs are susceptible to stochastic environmental events that can cause local extinctions or population declines, and extinctions of terrestrial mollusc species are among the highest documented. Many terrestrial snails are habitat specialists, and genetic studies using both allozyme and DNA sequence data have indicated that many species contain substantial and geographically structured genetic variation. In this study, we assess the genetic variation within two species of the rare terrestrial snail genus Prestonella, an inhabitant of rocky areas along water courses in the southern Great Escarpment of South Africa, and correlate genetic diversity to climatic variables. DNA sequence data from mitochondrial 16S rDNA and partial cytochrome oxidase I genes indicate that neither species is monophyletic, and that populations are deeply divergent, even over distances of a few hundred metres. Principal components anaylsis of climatic variables derived from two databases indicates that genetic diversity of the populations is correlated to moisture-related climatic variables. Populations with little or no diversity occur in more arid regions, and are thus most at risk from any future climatic changes that would increase aridification. These moisture variables are thus potent drivers of genetic bottlenecks and may have resulted in historical climate filtering or limitation of the distribution of these species. Temperature appears to be a less important variable, a finding supported by physiological data based on heart rates that show that death occurs only at temperatures far higher than found in their environment.  相似文献

Phytophagous insects of the genus Bactrocera are among the most economically important invasive fruit fly pests. In 2003, an unknown Bactrocera species was found in Kenya. First identified as an ‘aberrant form’ of the Asian B. dorsalis complex, it was later recognized as a new species, Bactrocera invadens. Within 2 years of its discovery, the species was recorded in several African countries, becoming an important quarantine pest. As this invasive fly was discovered only recently, no data are available on its invasion pattern in Africa. This pilot study attempts to infer from genetic data the dynamic aspects of the African invasion of this pest. Using microsatellite markers, we evaluated the level of genetic diversity and the extent of common ancestry among several African populations collected across the invaded areas. A sample from the Asian Sri Lankan population was analysed to confirm the Asian origin of this pest. Genetic data cast no doubt that Sri Lanka belongs to the native range, but only a small percentage of its genotypes can be found in Africa. African populations display relatively high levels of genetic diversity associated with limited geographical structure and no genetic footprints of bottlenecks. These features are indicative of processes of rapid population growth and expansion with possible multiple introductions. In the span of relatively few years, the African invasion registered the presence of at least two uncorrelated outbreaks, both starting from the East. The results of the analyses support that invasion started in East Africa, where B. invadens was initially isolated.  相似文献

The yellow-breasted capuchin monkey, Cebus xanthosternos, is one of the most endangered species of the Brazilian Atlantic Forest. In situ conservation for this species is problematic due to habitat destruction; therefore, captive conservation has been considered as an alternative strategy. A Studbook for C. xanthosternos has been kept for more than 20 years; however, no genetic data has been collected. Our aim was to provide a preliminary assessment of the genetic variability of C. xanthosternos in captivity in Brazil and compare it with data from the wild. Microsatellite and mtDNA sequencing were carried out in 40 samples from five Brazilian institutions registered in the international Studbook and compared with 8 samples collected in a wild population from REBIO-Una/BA. DNA for analysis was extracted from hair, feces and blood. Our results showed that two of the five captive groups assessed had a genetic variability comparable to wild animals. However, the other three groups apparently require urgent management to improve its genetic variability. Considering that inbreeding effects are more pronounced in captivity due to lack of gene flow, our data indicate a need to increase population size by introducing newly rescued individuals into these captive groups. Our results are the first attempt to provide genetic information for captive C. xanthosternos in Brazil.  相似文献

 利用9对SSR引物对中华猕猴桃(Actinidia chinensis)和美味猕猴桃(A. deliciosa)两近缘种的5个同域分布复合体和各自1个非同域分布居群进行了遗传多样性、居群遗传结构的分析以及种间杂交渐渗的探讨。结果表明:1)两物种共有等位基因比例高达81.13%,物种特有等位基因较少(中华猕猴桃:13.27%,美味猕猴桃:5.61%),但共享等位基因表型频率在两近缘种间存在差异,而且与各同域复合体中两物种样本的交错程度或间距存在关联;2)两种猕猴桃均具有极高遗传多样性,美味猕猴桃的遗传多样性(Ho=0 .749, PIC=0.818)都略高于中华猕猴桃(Ho=0.686,PIC=0.799);3)两猕 猴桃物种均具有较低的Nei’s居群遗传分化度,但AMOVA分析结果揭示种内异域居群间(FST=0.091 5)和同域复合体种间(FST=0.111 5)均存在一定程度的遗传分化;中华猕猴桃居群遗传分化(GST=0.086; FST=0.212 1)高于美味猕猴桃(GST= 0.080;FST=0.142 0);4)同域分布复合体两物种间的遗传分化(GST=0.020)低于物种内异域居群间的遗传分化(中华猕猴桃:GST=0.086; 美味猕猴桃:GST=0.080),同域复合体物种间的基因流(Nm=7.89 -29.75)远远高于 同种异域居群间(中华猕猴桃:Nm =2.663; 美味猕猴桃:Nm=2.880); 5)居群UPGMA聚类揭示在同一地域的居群优先聚类,个体聚类结果显示多数个体聚在各自居群组内,但各地理居群并不按地理距离的远近聚类,这与Mantel相关性检测所揭示的居群间遗传距离与地理距离没有显著性相关的结果一致。进一步分析表明两种猕猴桃的遗传多样性和居群遗传结构不仅受其广域分布、远交、晚期分化等生活史特性的影响,同时还与猕猴桃的染色体基数高 (x=29)、倍性复杂和种间杂交等因素密切相关,其中两种猕猴桃的共享祖先多态性和同域分布种间杂交基因渗透对两猕猴桃的居群遗传结构产生了重要影响。  相似文献
