Patients with chronic kidney disease (CKD), who usually display low serum 25-hydroxyvitamin D (25D) and 1,25-dihydroxyvitamin D (1,25D), are at high risk of infection, notably those undergoing peritoneal dialysis (PD). We hypothesized that peritoneal macrophages from PD patients are an important target for vitamin D-induced antibacterial activity. Dialysate effluent fluid was obtained from 27 non-infected PD patients. Flow cytometry indicated that PD cells were mainly monocytic (37.9±17.7% cells CD14+/CD45+). Ex vivo analyses showed that PD cells treated with 25D (100 nM, 6 hrs) or 1,25D (5 nM, 6 hrs) induced mRNA for antibacterial cathelicidin (CAMP) but conversely suppressed mRNA for hepcidin (HAMP). PD cells from patients with peritonitis (n = 3) showed higher baseline expression of CAMP (18-fold±9, p<0.05) and HAMP (64-fold±7) relative to cells from non-infected patients. In 12 non-infected PD patients, oral supplementation with a single dose of vitamin D2 (100,000 IU) increased serum levels of 25D from 18±8 to 41±15 ng/ml (p = 0.002). This had no significant effect on PD cell CD14/CD45 expression, but mRNA for HAMP was suppressed significantly (0.5-fold, p = 0.04). Adjustment for PD cell CD14/CD45 expression using a mixed linear statistical model also revealed increased expression of CAMP (mRNA in PD cells and protein in effluent) in vitamin D-supplemented patients. These data show for the first time that vitamin D supplementation in vitro and in vivo promotes innate immune responses that may enhance macrophage antibacterial responses in patients undergoing PD. This highlights a potentially important function for vitamin D in preventing infection-related complications in CKD.  相似文献

Spontaneously hypertensive rats (SHR), like patients with sleep apnea, have hypertension, increased sympathetic activity, and increased chemoreceptor drive. We investigated the role of carotid chemoreceptors in cardiovascular responses induced by obstructive apnea in awake SHR. A tracheal balloon and vascular cannulas were implanted, and a week later, apneas of 15 s each were induced. The effects of apnea were more pronounced in SHR than in control rats (Wistar Kyoto; WKY). Blood pressure increased by 57±3 mmHg during apnea in SHR and by 28±3 mmHg in WKY (p<0.05, n = 14/13). The respiratory effort increased by 53±6 mmHg in SHR and by 34±5 mmHg in WKY. The heart rate fell by 209±19 bpm in SHR and by 155±16 bpm in WKY. The carotid chemoreceptors were then inactivated by the ligation of the carotid body artery, and apneas were induced two days later. The inactivation of chemoreceptors reduced the responses to apnea and abolished the difference between SHR and controls. The apnea-induced hypertension was 11±4 mmHg in SHR and 8±4 mmHg in WKY. The respiratory effort was 15±2 mmHg in SHR and 15±2 mmHg in WKY. The heart rate fell 63±18 bpm in SHR and 52±14 bpm in WKY. Similarly, when the chemoreceptors were unloaded by the administration of 100% oxygen, the responses to apnea were reduced. In conclusion, arterial chemoreceptors contribute to the responses induced by apnea in both strains, but they are more important in SHR and account for the exaggerated responses of this strain to apnea.  相似文献

Total body irradiation of mice is a commonly used research technique; however, humane endpoints have not been clearly identified. This situation has led to the inconsistent use of various endpoints, including death. To address this issue, we refined a cageside observation-based scoring system specifically for mice receiving total body irradiated. Male and female C57BL/6 mice (age, 8 wk) received 1 of 3 doses of radiation from 1 of 2 different radiation sources and were observed for progression of clinical signs. All mice were scored individually by using cageside observations of their body posture (score, 0 to 3), eye appearance (0 to 3), and activity level (0 to 3). Retrospective analysis of the observation score data indicated that death could be predicted accurately with total scores of 7 or greater, and observation scores were consistent between observers. This scoring system can be used to increase the consistent use of endpoint criteria in total body murine irradiation studies and ultimately to improve animal welfare.Abbreviations: TBI, total body irradiation; ARS, acute radiation syndromeTotal body irradiation (TBI) of mice is a widely used technique with numerous applications. Mice undergoing TBI followed by stem cell transplant are a model of engraftment kinetics13,14,17,18 and graft-versus-host disease.10,28 Mice may undergo multiple rounds of TBI followed by gene transfer to create chimeras to study various diseases.9,11,19,36 More basically, the biologic effects of radiation in mice are a useful model for the various manifestations of human radiation sickness15,29,35,37 and can be used to develop and evaluate treatments.5,23,37Despite the widespread irradiation of mice, studies typically do not report criteria used for humane euthanasia or the mortality observed due to irradiation with failure of the graft, transplant, or treatment. This practice has resulted in the frequent and widespread use of death or the moribund state as the experimental endpoint.5,6,35 The moribund condition, an unresponsive and immobile animal, is a commonly used endpoint for a variety of research protocols associated with high mortality or progressive and severe disease states.32,33 Using the moribund criterion requires the animal to progress through all potential phases of pain and distress associated with the chosen model to a near-death state before the animal is euthanized.25The ability to predict death with a high probability and high accuracy for mice receiving TBI would allow preemptive euthanasia. This action would ameliorate terminal pain and distress associated with acute radiation syndrome (ARS) and markedly improve animal welfare yet maintain scientific integrity in accordance with the recommendation of the Guide for the Care and Use of Laboratory Animals.12 Although objective endpoint criteria are generally preferable, this option is not always possible. For mice receiving TBI, neither weight loss26 nor decreased food and water consumption29 are good predictors of death, because many animals that survive demonstrate marked weight loss and reduced consumption. Furthermore, mice receiving TBI should be handled minimally to avoid increasing mortality.15,31 As an alternative approach to objective endpoint criteria, subjective behavioral criteria may be used, providing that they are adequately described, monitored, and applied so that trends can be documented.24,34The current study sought to refine an observation-based scoring system to assess the health status of irradiated mice and determine the broad applicability of endpoints across radiation doses, radiation sources, and animal sexes. It was performed in conjunction with an IACUC-approved study designed to establish survivability curves for mice undergoing TBI with no other experimental manipulation. Male and female C57BL/6 mice that received 1 of 3 TBI doses from 1 of 2 different radiation sources were evaluated by using daily cageside observational scoring of body posture, activity level, and eye appearance. The results were analyzed to verify consistency between observers and identify the predictive nature of these criteria of impending death, with the goal of establishing useful endpoint criteria for all mice receiving TBI.  相似文献

ObjectiveTo characterize the metabolic phenotype of 2 cases of normal weight young women who developed type 2 diabetes (T2D), severe insulin resistance (insulin requirement >200 units/day), marked hypertriglyceridemia (>2000 mg/dL), and hepatic steatosis beginning 9 years after undergoing total body irradiation (TBI) and bone marrow transplantation for childhood cancer.MethodsFasting plasma glucose, insulin, free fatty acids (FFAs), leptin, adiponectin, resistin, TNFα, and IL-6 were measured in each case and in 8 healthy women; Case 1 was also assessed after initiating pioglitazone. Coding regions and splice junctions of PPARG, LMNA, and AKT2 were sequenced in Case 1 and of PPARG in Case 2 to evaluate for familial partial lipodystrophies. Genotyping of APOE was performed in Case 1 to rule out type III hyperlipoproteinemia.ResultsBoth cases had elevated plasma levels of insulin, leptin, resistin, and IL-6, high-normal to elevated TNFα, and low to low-normal adiponectin in keeping with post-receptor insulin resistance and adipose tissue inflammation. Case 1 experienced a biochemical response to pioglitazone. No causative mutations for partial lipodystrophies or type III hyperlipoproteinemia were identified.ConclusionThough metabolic derangements have previously been reported in association with TBI, few cases have described insulin resistance and hypertriglyceridemia as severe as that seen in our patients. We speculate that early childhood TBI may impede adipose tissue development leading to metabolic complications from an attenuated ability of adipose tissue to accommodate caloric excess, and propose that this extreme metabolic syndrome be evaluated for as a late complication of TBI. RECENT progress in the definition of the DL-A antigens within a closely bred colony of beagles maintained at the Mary Imogene Basset Hospital (Cooperstown colony) has shown that these antigens are components of the principal system of histocompatibility (DL-A) in the dog1–3. Dausset and associates4 have described the segregation of DL-A antigens in 679 offspring of 151 consecutive matings within the Coopers-town colony. Through methods of backcross analysis, 23 DL-A haplotypes (that is, specificities determined by the DL-A region of one single chromosome) have been identified and the DL-A genotypes of 1,302 offspring of 517 matings in this colony have been defined4. This combined genetic and serological information has provided an opportunity to reassess the reported survival rates of allogeneic bone marrow transplantation in serologically matched littermate dogs5. We describe here the long term survival of bone marrow transplants and renal allografts obtained from the same donors in nine littermate and three non-littermate recipients selected on the basis of genetic and serological criteria of DL-A compatibility in the Cooperstown colony. The results are contrasted with the survival of twenty-two renal allografts performed in untreated recipients selected on the basis of similar criteria.  相似文献

Increases in the terrestrial levels of ultraviolet-B (UV-B) radiation (280 to 320 nm) due to diminished stratospheric ozone have prompted an investigation of the protective mechanisms that contribute to UV-B tolerance in plants. In response to UV-B stress, flowering plants produce a variety of UV-absorptive secondary products derived from phenylalanine. Arabidopsis mutants with defects in the synthesis of these compounds were tested for UV-B sensitivity. The transparent testa-4 (tt4) mutant, which has reduced flavonoids and normal levels of sinapate esters, is more sensitive to UV-B than the wild type when grown under high UV-B irradiance. The tt5 and tt6 mutants, which have reduced levels of UV-absorptive leaf flavonoids and the monocyclic sinapic acid ester phenolic compounds, are highly sensitive to the damaging effects of UV-B radiation. These results demonstrate that both flavonoids and other phenolic compounds play important roles in vivo in plant UV-B protection.  相似文献

Acute radiation syndrome is a life-threatening condition that has the potential to affect large populations of humans. Although several animal models of this syndrome are available, the total-body–irradiated mouse has emerged as an important tool to evaluate the efficacy of prospective prophylaxis, mitigation, and treatment compounds. Despite the widespread use of this model, humane endpoints have not been clearly identified. To address this issue, we developed a cageside observation-based scoring system specifically for total-body–irradiated mice to assess the progression of clinical signs associated with acute radiation syndrome. Male C57BL/6 mice (n = 175; age, 8 to 9 wk) received an anticipated LD50 dose of radiation and were observed for progression of clinical signs of acute radiation syndrome for 30 d. All mice were scored individually through cageside observation of their body posture (score, 0 to 3), eye appearance (0 to 3), and activity level (0 to 3). Retrospective analysis of the score data indicated that death could be predicted accurately by using increasing cumulative scores (0 to 9). Total scores of 6, 7, 8, and 9 were associated with mortality rates of 78.6%, 86.4%, 93.3%, and 100%, respectively. Furthermore, scores of 6, 7, and 8 predicted death within 3, 1.5, and 0.5 d, respectively. The use of this scoring system provides investigators and IACUCs with predictive humane, surrogate endpoints for total-body–irradiated mice. This system allows preemptive euthanasia of mice before they become moribund, thereby minimizing pain and distress associated with acute radiation syndrome and improving animal welfare.Abbreviations: ARS, acute radiation syndrome; LD50/30, the dose of radiation that is lethal for 50% of the test subjects within 30 d; TBI, total body irradiationAcute radiation syndrome (ARS) due to either accidental radiation exposure or nuclear attack is a life-threatening condition that has the potential to affect a large population of people. The severity of ARS is dependent on the overall dose, dose rate, radiation quality, and proportion of the body that is irradiated. ARS typically progresses through 4 clinical phases: prodrome, latency, illness, and either recovery or death.1,7 The prodromal period is characterized by nausea, vomiting, and fatigue and can include autonomic instability and loss of consciousness at high doses in humans.1 After the latent phase, which can be absent with high doses of radiation, the illness phase manifests in various organ systems as particular syndromes of the hematopoietic, gastrointestinal, skin, and neurovascular systems.1,8 The hematopoietic system is the most sensitive to radiation, and decreased blood cell counts can be detected even in asymptomatic patients.1 Increasing radiation doses cause complete destruction of bone marrow, sloughing of the mucosal layer of the gastrointestinal system, skin burns, and breakdown of the neurologic and cardiovascular systems, ultimately resulting in death.1,8Preparatory planning for the medical management of ARS is essential and requires the use of total body irradiation (TBI) models in animals to evaluate the efficacy of prospective prophylaxis, mitigation, and treatment compounds.8,28 The 2 body systems most sensitive to TBI are the hematopoietic and gastrointestinal systems, and mice are an excellent model for both; the C57BL/6 and C3H/He strains are used most often.28 To study the hematopoietic syndrome, the most widely used TBI condition for acute radiation damage is the LD50, because of the temporal predictability of the development of subsequent clinical signs. The temporal sequence and outcome of the hematopoietic syndrome occur considerably more rapidly in mice than in humans, and the experimental outcome is defined as the LD50/30 (50% population death within 30 d of irradiation) in mouse models.28 In addition, because mice have been deemed an appropriate species for testing radioprotectors or mitigators, the clinical efficacy of various drugs can be evaluated easily in a mouse TBI model.28Although mice are an exceedingly common model for ARS, there are currently no established humane endpoint criteria for these types of studies. Similar to sepsis models, TBI and ARS studies typically use LD50/30 as the standard of comparison, and this practice has resulted in the frequent and widespread use of death or moribund state as the experimental endpoint.5,7,18,26 The moribund condition, an unresponsive and immobile animal, is a commonly used endpoint for a variety of research protocols associated with high mortality or progressive and severe disease states.23,24 Using this criterion requires the animal to progress through all potential phases of pain and distress associated with the chosen model to a near-death state before the animal is euthanized.17The ability to predict death with a high probability and high accuracy in TBI studies would allow for preemptive euthanasia, with the intent to (1) ameliorate terminal pain and distress associated with ARS and (2) improve animal welfare. Swine generate reassortant influenza viruses because they can be simultaneously infected with avian and human influenza; however, the features that restrict influenza reassortment in swine and human hosts are not fully understood. Type I and III interferons (IFNs) act as the first line of defense against influenza virus infection of respiratory epithelium. To determine if human and swine have different capacities to mount an antiviral response the expression of IFN and IFN-stimulated genes (ISG) in normal human bronchial epithelial (NHBE) cells and normal swine bronchial epithelial (NSBE) cells was evaluated following infection with human (H3N2), swine (H1N1), and avian (H5N3, H5N2, H5N1) influenza A viruses. Expression of IFNλ and ISGs were substantially higher in NHBE cells compared to NSBE cells following H5 avian influenza virus infection compared to human or swine influenza virus infection. This effect was associated with reduced H5 avian influenza virus replication in human cells at late times post infection. Further, RIG-I expression was lower in NSBE cells compared to NHBE cells suggesting reduced virus sensing. Together, these studies identify key differences in the antiviral response between human and swine respiratory epithelium alluding to differences that may govern influenza reassortment.  相似文献

ABSTRACT. Mice infected with the protozoan parasite Trypanosoma cruzi , the causative agent of human Chagas'disease, develop immunosuppressed responses to heterologous antigens. Experiments were performed using infected mice in the acute stage of infection to assess immunoregulatory activities during induction of direct plaque-forming cells (DPFC) to sheep erythrocytes (SRBC), hapten-conjugated SRBC (TNP-SRBC), and horse erythrocytes (TNP-HRBC). Studies in vivo demonstrated that anti-SRBC responses were best enhanced when T. cruz -infected mice were injected with primed T cells derived from normal or infected mice immunized four days previously. The presence of enhancing capacities for DPFC responses by T cells from T. cruzi -infected mice were also supported by experiments examining the hapten-carrier effect. Preimmunization of infected mice with SRBC or HRBC four days before injection of hapten-homologous (TNP-SRBC or TNP-HRBC) carrier resulted in markedly augmented anti-hapten antibody responses. These results show that functional help provided by T cells activated during priming and exposed to a challenge dose of antigen (SRBC) in a time-dependent mode can overcome the effect of immunosuppression in T. cruzi -infected mice.  相似文献

Direct-current, low-intensity, electric fields were suggested as a minimally invasive treatment for various cancers. The tumor microenvironment may affect treatment efficacy, albeit it has not generally been considered when evaluating novel anti-cancer treatments. We evaluate the effects of electric treatment on epithelial, breast-cancer cells, co-cultured with non-cancerous fibroblasts, a simplified model for the tumor-microenvironment. We evaluate changes in morphology, cytoskeleton, and focus on dynamic intracellular structure and mechanics. Multiple-particle tracking was used within living cells to quantify time-dependent structural and mechanical changes. Cancer cells suffer severe cell death and exhibit transient rounding and changes in internal structural and mechanics. Interestingly, treating cancer cells in co-culture with fibroblasts delays and reduces their responses to treatment. Our particle-tracking data indicates a mechanism relating the observed changes in intracellular transport to transient changes in the microtubule network and its motors. In contrast, fibroblasts are only minimally affected by treatment, separately or in co-culture. To conclude, intracellular mechanics reveal time-dependent responses after treatment, unavailable by bulk measurements. This time-dependence could provide a window of opportunity for continued treatment. We demonstrate the importance of evaluating anti-cancer treatments within their microenvironment, which can affect response magnitude and time-course.  相似文献

One of the few bacteria that have been consistently linked to colorectal cancer (CRC) is the opportunistic pathogen Streptococcus gallolyticus. Infections with this bacterium are generally regarded as an indicator for colonic malignancy, while the carriage rate of this bacterium in the healthy large intestine is relatively low. We speculated that the physiological changes accompanying the development of CRC might favor the colonization of this bacterium. To investigate whether colon tumor cells can support the survival of S. gallolyticus, this bacterium was grown in spent medium of malignant colonocytes to simulate the altered metabolic conditions in the CRC microenvironment. These in vitro simulations indicated that S. gallolyticus had a significant growth advantage in these spent media, which was not observed for other intestinal bacteria. Under these conditions, bacterial responses were profiled by proteome analysis and metabolic shifts were analyzed by 1H-NMR-spectroscopy. In silico pathway analysis of the differentially expressed proteins and metabolite analysis indicated that this advantage resulted from the increased utilization of glucose, glucose derivates, and alanine. Together, these data suggest that tumor cell metabolites facilitate the survival of S. gallolyticus, favoring its local outgrowth and providing a possible explanation for the specific association of S. gallolyticus with colonic malignancy.The human intestine is the habitat for several hundred different bacterial species with an increasing bacterial concentration and variability toward the distal colon (1). The resident gut microbiota is essential for human health by making dietary nutrients available to the host and preventing the invasion of pathogens by competitive colonization and nutrient competition (2, 3). Strikingly, the part of the intestine with the highest bacterial colonization, the colon, is also most affected by cancer, with 146,970 annual cases in the United States of America (4). In a healthy colonic environment, the host has several defense mechanisms to shield itself from bacterial infection, such as the viscous mucus layer overlaying the epithelium. However, the progression of CRC1 is accompanied by changes in the integrity of the colon, including reduced mucus production (5) and increased epithelial permeability (6). These physiological changes can drive the intestinal ecosystem, which is relatively stable during adult life, into dysbiosis (7, 8). As a consequence, the host may become more susceptible to opportunistic bacterial infections (911).One of the few bacteria that have been consistently linked to CRC is the opportunistic pathogen Streptococcus gallolyticus (previously known as Streptococcus bovis biotype I). In CRC patients the fecal carriage rate of this bacterium is increased from 10% to about 50% (12), which suggests that this disease facilitates the colonic survival of S. gallolyticus. Importantly, ∼60% of patients that present with S. gallolyticus endocarditis have concomitant CRC (both adenomas and carcinomas) (13, 14), which largely exceeds the rates reported in the general population (∼25%) (15). These patients had no gastro-intestinal signs or clinical symptoms of malignancy and CRC was only detected because this bacterial infection guided the physician to perform a colonoscopy.Several mechanisms for this apparent association between S. gallolyticus and CRC can be envisaged. Recently, we postulated a model in which the collagen binding ability of S. gallolyticus contributes to the specific colonization of malignant colonic sites (16). However, the altered microenvironment of the tumor may also provide conditions that favor survival and outgrowth of S. gallolyticus in this newly formed intestinal niche. For example, Hirayama et al. have shown that glucose-1-phosphate and fructose-1-phophate levels as well as amino acid concentrations were significantly higher in tumor tissue than in normal tissue (17). To investigate if this altered nutritional status of the CRC microenvironment could facilitate the foraging of S. gallolyticus, we simulated the influence of colon tumor cell metabolites on S. gallolyticus growth by incubating this bacterium in spent medium of malignant cells. Subsequently, the bacterial responses were profiled by two-dimensional proteome analysis, and metabolic shifts in the culture medium were assessed by 1H-NMR-spectroscopy. In silico pathway analysis and further in vitro simulations showed that, unlike other intestinal bacteria, S. gallolyticus had a growth advantage under these conditions, which could mainly be attributed to increased glycolysis. These results provide the first molecular support that tumor metabolites may facilitate the local outgrowth of tumor-foraging bacteria, such as S. gallolyticus.  相似文献

We examined the effects of dietary vitamin E (VE) on oxidative damage to DNA and lipids in the liver a few days after total body irradiation (TBI). ODS rats, which lack vitamin C synthesis, were fed either a low VE diet (4.3 &#117 mg &#117 VE/kg) or a basal VE diet (75.6 &#117 mg &#117 VE/kg) for 5 weeks while vitamin C was supplied in the drinking water. The VE level in the liver of the low VE group was lower and the levels of lipid peroxides were higher compared to those of the basal VE group: the relative levels in the two groups were 1:30 for VE, 18:1 for 4-hydroxynonenal (HNE), and 10:1 for hexanal (HA). The level of 8-hydroxydeoxyguanosine (8OHdG), a marker of oxidative DNA damage, did not differ between the low VE and the basal VE groups. When the rats received TBI at the dose of 3 &#117 Gy and were killed on day 6, the levels of HNE, HA and 8OHdG increased by 2.2-, 2-, and 1.5-times, respectively, in the low VE group, but TBI did not cause such increases in the basal VE group. Changes in antioxidative enzymes (glutathione peroxidase, catalase, and Cu/Zn-SOD) in the liver could not explain the different responses of the two diet groups to TBI-induced oxidative damage. The concentrations of vitamin C and glutathione in the liver did not differ between the two groups. These results suggest that dietary VE can prevent the oxidative damage to DNA and lipids in the liver which appear a few days after TBI at dose of 3 &#117 Gy.  相似文献

Although most people can identify facial expressions of emotions well, they still differ in this ability. According to embodied simulation theories understanding emotions of others is fostered by involuntarily mimicking the perceived expressions, causing a “reactivation” of the corresponding mental state. Some studies suggest automatic facial mimicry during expression viewing; however, findings on the relationship between mimicry and emotion perception abilities are equivocal. The present study investigated individual differences in emotion perception and its relationship to facial muscle responses - recorded with electromyogram (EMG) - in response to emotional facial expressions. N° = °269 participants completed multiple tasks measuring face and emotion perception. EMG recordings were taken from a subsample (N° = °110) in an independent emotion classification task of short videos displaying six emotions. Confirmatory factor analyses of the m. corrugator supercilii in response to angry, happy, sad, and neutral expressions showed that individual differences in corrugator activity can be separated into a general response to all faces and an emotion-related response. Structural equation modeling revealed a substantial relationship between the emotion-related response and emotion perception ability, providing evidence for the role of facial muscle activation in emotion perception from an individual differences perspective.  相似文献
