The Ashe magnolia (Magnolia ashei) is a deciduous small tree most noted for its large 1–2 foot long leaves and fragrant creamy white flowers. Although the species is adapted to and used in landscapes in many parts of the U.S., it is endemic only to Northwest Florida where it is limited to ten counties growing on undisturbed bluffs and ravine banks. The populations are highly fragmented and are threatened by degradation of habitat, leading the species to be listed as endangered in the state of Florida. SSR markers were developed to determine the genetic diversity of wild populations of M. ashei in order to guide long-term conservation strategies. 18 marker loci identified a total of 82 alleles that were used to characterize allelic diversity of M. ashei from 11 wild populations, 14 cultivated sources, five accessions of M. macrophylla, and three interspecific hybrids. Results indicated a higher than expected level of heterozygosity within populations, and a clear distinction between Eastern and Western populations; conservation efforts should therefore focus on maintaining these distinct groups in corresponding ex situ seed orchards to counteract pressures due to overcollection, pollution, and loss of habitat due to development. Clustering of individuals was similar using several analytical methods, indicating that despite relatively small sample sizes, our analysis is a valid reflection of the diversity among and relationships between these populations.  相似文献

Identification of population units is crucial for management and monitoring programs, especially for endangered wild species. The roughskin sculpin (Trachidermus fasciatus Heckel) is a small catadromous fish and has been listed as a second class state protected aquatic animal since 1988 in China. To achieve sustainable conservation of this species, it is necessary to clarify the existing genetic structure both between and within populations. Here, population genetic structure among eight populations of T. fasciatus were analyzed by using 16 highly polymorphic microsatellites. High levels of genetic variation were observed in all populations. All pairwise F ST estimates were significant after false discovery rate correction (overall average F ST = 0.054). Furthermore, both STRUCTURE and discriminant analysis of principal components (DAPC) analysis showed that the eight populations were grouped into six clusters. BAYESASS analysis showed generally low recent and asymmetric migration among populations. All these results suggested significant genetic structure across populations. However, there was no isolation by distance relationship among populations, likely resulting from barriers to gene flow created by habitat fragmentation. Our results highlight the need for in situ conservation efforts for T. fasciatus across its entire distribution range, through maximizing habitat size and quality to preserve overall genetic diversity and evolutionary potential.  相似文献

Cycas debaoensis is a critically endangered cycad species endemic to China. This species is found on two kinds of habitats according to the edaphic differences, sand and karst. A previous chloroplast DNA (cpDNA) study indicated that C. debaoensis had low genetic variation within populations and high genetic differentiation among populations. Because maternally inherited cpDNA does not fully characterize genetic structure of the species, we screened seven low-copy nuclear genes and 17 nuclear microsatellite loci to detect the nuclear genetic diversity, differentiation, and the population structure of C. debaoensis. The nuclear genes revealed higher level of genetic diversity. There were both the same and region-specific haplotypes or alleles between the karst and sand regions. Nuclear gene flow among all the populations was much greater than that of cpDNA, which indicated that pollen-mediated gene flow was much greater than seed-mediated gene flow. This promoted low nuclear genetic differentiation among populations of C. debaoensis. The study suggests that both genetic and anthropogenic disturbances have resulted in the critically endangered status of C. debaoensis.  相似文献

The Patagonian silverside Odontesthes hatcheri is a native fish restricted to streams and lakes of Patagonia (Argentina and Chile). Stocking programs to enhance recreational fisheries in man-made reservoirs have introduced a nonnative, closely-related species (the pejerrey O. bonariensis) in Patagonia almost a century ago, and yet little is known about the invasiveness of this species. To evaluate the impact of these introductions we analyze genetic data (microsatellite markers and mitochondrial DNA) to quantify the incidence of hybridization between these two species and assess potential effects on native population structure. Phylogeographic analyses reveal weak geographic differentiation among populations of O. hatcheri, in agreement with previous studies for other freshwater fishes in Patagonia strongly influenced by Quaternary glaciations and hydrographic basin changes since the last glaciation. However, many populations have unique genetic pools. In several areas, introductions resulted in extensive hybridization, with high frequencies of F2 and backcrossed hybrids in natural populations, and in some cases O. bonariensis has completely displaced the native populations. The negative impact of these introductions on native populations is correlated to temperature, a critical parameter in the face of global warming, suggesting that invasiveness of O. bonariensis may increase in the future. Our results advise against continuing stocking programs to preserve the integrity of natural populations of the Patagonian silverside.  相似文献

In the Azores Islands, two Euphrasia L. (Orobanchaceae) endemic species are recognized: Euphrasia azorica H.C.Watson, an annual herb, in Flores and Corvo, and Euphrasia grandiflora Hochst. ex Seub., a semi-shrub, in Pico, São Jorge and Terceira. Both species are highly endangered and protected by the Bern Convention and Habitats Directive. A population genetics study was conducted with new microsatellite primer pairs in 159 individuals of E. azorica and E. grandifolia, sampled from populations in Flores, Corvo, Pico and São Jorge. Allele sizing suggested that E. azorica is a diploid while E. grandiflora is a tetraploid. Euphrasia grandiflora revealed higher genetic diversity then E. azorica. The E. grandiflora population of Morro Pelado in São Jorge, displayed higher genetic diversity when compared with all others, while the E. azorica population of Madeira Seca in Corvo, showed the lowest. Private and less common bands were also overall higher in E. grandiflora populations. Population genetic structure analysis confirmed a distinctiveness between the two Azorean endemic Euphrasia, in addition to island-specific genetic patterns in E. azorica. The genetic structure obtained for E. grandiflora was complex with the populations of Cabeço do Mistério in Pico Island and of Pico da Esperança in São Jorge sharing the same genetic group, while a putative spatial barrier to gene flow was still retrieved between both islands. Although some populations of both species might benefit from propagation actions, studies are needed on plant host species and translocations between islands or between some populations of a same island should be avoided, due to the occurrence of putative ESUs. Eradication of invasive species and control of grazing will be fundamental to promote in situ restauration.  相似文献

Limonium vulgare and related species form a complex group, but until now cytological and genetic studies have been based on single species and specific geographical areas. We investigated genome size, karyological and genetic diversity in samples from Western Mediterranean and evaluated the phylogenetic relationships among the species of this complex. Genome size was assessed using flow cytometry on samples from natural populations of L. vulgare, L. maritimum and L. narbonense. Chromosome counts were conducted in plants obtained from seeds collected in the field. The internal transcribed spacer ITS1 of the nuclear rDNAs was used to assess ITS polymorphisms as well as the phylogenetic relationships within the L. vulgare complex. Our analyses showed that all species were tetraploid, with the chromosome number of L. maritimum being presented here for the first time. Significant differences were observed in genome size, with L. narbonense having lower genome sizes than the other two species, and possible aneuploids being detected. Ten new ITS sequences from L. vulgare, L. narbonense and L. maritimum were provided. Most species’ populations showed unique ribotypes, and L. narbonense has the highest ribotype diversity. One of the L. maritimum populations presented a closer genetic relationship with L. vulgare, whereas the other two seemed to be more related with L. narbonense. Phylogenetic analyses confirmed that L. vulgare and L. narbonense form a monophyletic group, sister to the remaining Limonium species. Our results put into evidence that the studied species may represent a relatively early stage of divergence.  相似文献

Freshwater fish diversity in Algeria with emphasis on alien species   总被引:1,自引:0,他引:1  
Little is known about freshwater fish diversity in Algeria, especially after the broad national program of introduction of exotic species applied for over 20 years. This paper is an attempt to describe the current situation emphasizing the characteristics of the various introductions of non-autochthonous species, their current status and their possible impacts. The freshwater fish fauna of Algeria is composed of 48 species belonging to 15 families. Twenty-one species are autochthonous, of which, three are endemic (Haplochromis desfontainii, Aphanius saourensis, Aphanius apodus), two are endangered (H. desfontainii, Aphanius iberus), and one is critically endangered (Anguilla anguilla). Twenty-seven species were introduced, of which, 18 never became established and 9 are established. Four species are reported for the first time: Abramis brama, Aspius aspius, Carassius carassius, Perca fluviatilis. Since 1860, 303 introduction events have been recorded at almost 107 different sites in Algeria. The number of introduction events per site was between 1 and 10. Most of the introduced fish are found in Oubéïra Lake (seven species). Aquaculture development was the main reason for fish introductions (37.0%). The five most often introduced species were Cyprinus carpio (86 records), Hypophthalmichthys molitrix (65 records), Aristichthys nobilis (51 records), Oreochromis niloticus niloticus (33 records), and Ctenopharyngodon idella (18 records). Since the adverse effects of introduced fish are risky and demonstrated in several countries, the adoption of the precautionary principle is recommended when new introductions are planned. It is important, therefore, to adopt regulations and procedures which will minimize the risks arising from introductions.  相似文献

Knowledge and analysis of the genetic structure of an endangered species is important for its conservation and evolutionary process. Simple sequence repeats (SSRs) and amplified fragment length polymorphisms (AFLPs) were used in evaluation of the genetic diversity and population differentiation in Limonium bicolor (Plumbaginaceae), an endangered herb with high medicinal and horticulture value. A total of 117 alleles were detected with an average 5.85 alleles per locus using SSR and 222 bands from AFLP were amplified in six populations. It was found that L. bicolor was characterized by high levels of genetic polymorphism (100 and 83.78%), low levels of total genetic diversity (\(H_{\mathrm{t}}=\) 0.2824 and 0.2424), and moderate degrees of genetic differentiation among populations (\(\Phi _{\mathrm{ST}} =\) 0.284 and 0.251). Analysis of molecular variance (AMOVA) revealed that the main variation component existed within populations (71.56%; 74.93%) rather than among populations (28.44%; 25.07%). Four main clusters were displayed in the UPGMA using TFPGA, which was consistent with the result of principal coordinate analysis (PCA) using NTSYS. Mutations or infrequent gene flow among populations can increase the plant slowly, thus in situ conservation policies should be implemented first for effective and sustainable development. At the same time, ex situ measures, such as those individuals with rare alleles, to maintain the relationships between individuals and populations are also proposed.  相似文献

The freshwater pearl mussel Margaritifera margaritifera L. is one of the most endangered freshwater mussels in the world. Effective conservation of threatened species requires not only ecological, but also genetic information from the target species and populations. Since low genetic diversity can reduce the ability of a species to adapt to environmental changes, maintaining genetic diversity has been identified as one of the key elements in successful conservation programs. We examined genetic variation of the freshwater pearl mussel from the River Vuokkijoki, Karelia, Russia. We sequenced a fragment of the cytochrome c oxidase subunit I gene (COI) from 22 individuals and compared the data to 32 previously published COI sequences available in GenBank. We identified 10 different COI haplotypes in the sequenced samples, three of which had not been previously reported. Our results show that the River Vuokkijoki has high genetic diversity and suggest that the colonization of this northern freshwater pearl mussel population might have occurred from multiple and even distant refugia. Therefore, the freshwater pearl mussel population of the River Vuokkijoki is valuable for the conservation of the whole species.  相似文献

Patterns of genetic structure and diversity are largely mediated by a species’ ecological niche and sensitivity to climate variation. Some species with narrow ecological niches have been found to exhibit increased population differentiation, limited gene flow across populations, and reduced population genetic diversity. In this study, we examine patterns of population genetic structure and diversity of four bumble bee species that are broadly sympatric, but do not necessarily inhabit the same ecological niche in the Pacific Northwest of the United States. Testing for the effect of isolation by geographic distance (IBD) with linearized F st and D est found that Bombus sylvicola and B. mixtus exhibited significant IBD across populations. In contrast, both B. melanopygus and B. flavifrons, two species that are distributed across a broad elevation gradient, exhibited no IBD, a result further corroborated by Bayesian a priori population assignment tests. Furthermore, we discovered that B. sylvicola populations distributed on the Olympic Peninsula have significantly less average allelic diversity than populations distributed in the Cascade Mountains. Our results suggest that populations distributed in the Olympic Mountains represent a distinct genetic cluster relative to the Cascade Mountains, with B. sylvicola and B. mixtus likely experiencing the greatest degree of population genetic differentiation relative to B. flavifrons and B. melanopygus. While bumble bees are known to co-exist across a diversity of habitats, our results demonstrate that underlying population genetic structure and diversity may not necessarily be similar across species, and are largely governed by their respective niches.  相似文献

Evaluating the consequences of the decline of threatened species on their population genetic structure is crucial for establishing effective conservation strategies in the strongly fragmented landscapes of Central Europe. Laserpitium prutenicum is a bi- to perennial forb occurring in intermittently wet meadows and light oak forests throughout central to eastern and south-eastern Europe. During the past 70 years, the western limit of its distributional range retracted dramatically, the number of populations decreased and the remaining populations faced a considerable increase of fragmentation. To study the effects of this decline on the genetic diversity of L. prutenicum, we conducted an AFLP study on 20 populations from Germany, Poland and the Czech Republic. For comparison, we collected the same data on Selinum carvifolia, a taxonomically related and both ecologically and morphologically similar species, which is still more common in the study area. Both species showed similarly weak spatial genetic structuring and intermediate genetic diversities. We attribute this result to the loss of habitat being faster than the loss of genetic diversity in smaller and fragmented populations. Depending on the ecological characteristics of a species, even a gradual disappearance is not necessarily accompanied by any detectable effect at the population genetic level (“silent goodbye”). In the case of L. prutenicum, habitat preservation should be given priority over all other conservation measures.  相似文献

The genetic diversity and population structure of the endemic species of Baikal Siberia Oxytropis triphylla, O. bargusinensis, and O. interposita were studied for the first time on the basis of the nucleotide polymorphism of intergenic spacers psbA–trnH, trnL–trnF, and trnS–trnG of chloroplast DNA. All populations of these species were characterized by a high haplotype (0.762–0.924) and relatively low nucleotide (0.0011–0.0022) diversity. Analysis of the distribution of variability in O. triphylla and O. bargusinensis showed that there was no significant genetic differentiation between populations of each species; the gene flow was 4.43 and 8.91, respectively. The high level of genetic diversity in the studied populations indicates a relatively stable state of these populations. A study of the phylogenetic relationships of closely related species confirms the concept of the origin of O. bargusinensis and O. tompudae as a result of intersectional hybridization of the species of the sections Orobia and Verticillares.  相似文献

To establish a management plan for endangered and rare species, genetic assessment must first be conducted. The genetic characteristics of plant species are affected by demographic history, reproductive strategy, and distributional range as well as anthropological effects. Abies koreana E. H. Wilson (Pinaceae), Korean fir, is endemic to Korea and found only in sub-alpine areas of the southern Korean Peninsula and Jejudo Island. This species has been designated as critically endangered by the International Union for Conservation of Nature due to a continuous decline in its range and population fragmentation. We genotyped 176 individuals from seven natural populations and two afforested populations on the Korean Peninsula using 19 microsatellite loci. STRUCTURE analysis revealed two genetic clusters in natural populations (F st  = 0.040 and R st  = 0.040) despite low differentiation. We did not detect a significant reduction in genetic diversity or the signature of a genetic bottleneck despite population fragmentation and small population size. We deduced that this species exhibits a metapopulation structure, with the population on Jirisan Mountain acting as a source of genetic diversity for other local small populations on the Korean Peninsula, through contemporary asymmetric gene flow. However, the majority of afforested individuals on the Korean Peninsula originated from a different gene cluster. Thus, we recommend a conservation strategy that maintains two genetically unique clusters.  相似文献

Human introductions of exotic amphibians can have catastrophic effects on native species. However, they usually remain unnoticed without genetic tools when species are difficult to distinguish morphologically. In Western Europe, pool frogs (Pelophylax sp.) make a worrisome case: recent genetic data showed the presence of Italian (Pelophylax bergeri) mtDNA haplotypes within the range of the threatened European Pelophylax lessonae, two morphologically similar taxa. Here we conduct a multilocus phylogeographic and population genetic survey of European and Italian pool frogs (combining present and historic samples), to investigate the origin(s) and consequences of potential introductions. Results are unequivocal: we show that the alien P. bergeri have extensively invaded France and north-Alpine Switzerland, and have also deeply introgressed with P. lessonae, which has led to the complete replacement of most populations. Alien specimens have probably been translocated multiple times from Central Italy at least prior to the 1960s. Based on our dense sampling, only two areas, north and south of the Alps still host native pool frogs in Switzerland, the Joux Valley near the French border and the canton of Ticino, respectively. Importantly, we show that these last P. lessonae populations are remnants of a private genetic diversity specific to Western Europe, which vanished during the P. bergeri’s invasion. Our study emphasizes the risk of genetic pollution during invasion by human-introduced taxa and brings alarming concern regarding uncontrolled amphibian translocations. Moreover, it demonstrates the necessity for genetic surveys to detect and monitor these invasions, especially where species determination is problematic.  相似文献

We isolated and characterized microsatellite loci in Viola websteri (Violaceae), an endangered species from Korea and endemic to Northeast Asia. A total of 27 microsatellite loci were developed and tested in Korean and Chinese populations. The number of alleles per locus varied from two to eight. The observed and expected heterozygosities within two populations were 0.000 to 1.000 and 0.080 to 0.816, respectively. Korean and Chinese populations were clearly distinguished by the private alleles from 16 loci. A total of 21 loci out of the 27 developed loci were successfully cross-amplified in 39 other Viola species. We believe that these microsatellite loci will be useful for future studies on genetic diversity and population structure of V. websteri, as well as other Viola species.  相似文献

Historical events such as colonisation, spatial distribution across different habitats, and contemporary processes, such as human-mediated habitat fragmentation can leave lasting imprints on the population genetics of a species. Orchids currently comprise 17% of threatened flora species in Australia (Environment Protection and Biodiversity Conservation Act 1999) due to the combination of fragmentation and illegal harvesting (Benwell in Recovery plan, swamp orchids Phaius australis, Phaius tancarvilliae, NSW National Parks and Wildlife Service, Sydney, 1994; Jones in A complete guide to native orchids of Australia including the island territories, 2nd edn, Reed Natural History, Sydney, 2006; DE in Phaius australis in species profile and threats database, Department of the Environment. http://www.environment.gov.au/sprat, 2015). The federally endangered Swamp Orchid Phaius australis has a disjunct distribution across an almost 2000 km latitudinal range along Australia’s east coast but it was estimated that 95% of the populations have been lost since European settlement (Benwell 1994). Phaius australis is endangered due to illegal collection and habitat loss that has resulted in limited connectivity between populations, in ecosystems that are vulnerable to climate change. Thus the genetic impacts of its history combined with more recent fragmentation may have impacts on its future viability especially in light of changing environmental conditions. Thirty-four populations were sampled from tropical north Queensland to the southern edge of the subtropics in New South Wales. Population genetics analysis was conducted using 13 polymorphic microsatellite markers developed for the species using NextGen sequencing. Spatial genetic patterns indicate post-colonisation divergence from the tropics southwards to its current climate niche limits. Genetic diversity is low across all populations (A?=?1.5, H e  = 0.171), and there is little evidence of genetic differentiation between regions. Consistent with population genetic theory, the historic loss of populations has resulted in significantly lower genetic diversity in small populations compared to large (P, A, He; p?<?0.05). The viability and persistence of P. australis populations now and in a changing climate are discussed in the context of conservation priorities.  相似文献

Spread of smooth cordgrass (Spartina alterniflora) in China is an exceptional example of unanticipated outcomes arising from intentional introductions. It has been proposed that in China, management strategies used to establish S. alterniflora inadvertently promoted evolutionary outcomes that have contributed to other Spartina invasions. In this study, we assessed whether S. alterniflora in China exhibits genetic signatures of mechanisms known to promote invasion success, including large founding populations, evolved self-fertility, ‘superior source ecotypes’, and post-introduction admixture. This involved comparing microsatellite genotype and chloroplast haplotype variation among Chinese populations to other invasive S. alterniflora populations as well as native range populations, inclusive of samples from all reported source areas. We found distinct signatures of source population contributions to Chinese populations, as well as evidence of post-introduction admixture, and no evidence of limitations from a genetic bottleneck. Measures of inbreeding were well below what has been found in other non-native populations that have evolved self-fertility. Differences in genetic diversity among sites were similar to latitudinal patterns in the native range, but could be attributable to introduction history. Comparisons to other invasive populations indicate that a combination of common and idiosyncratic processes have contributed to the success of S. alterniflora in China and elsewhere, with intentional introductions promoting mechanisms that accelerate rates of spread and widespread invasion.  相似文献

We studied the invasive warty cabbage Bunias orientalis (Brassicaceae) in three geographically distinct areas. Using inter-simple sequence repeat fingerprinting, we analyzed warty cabbages, including non-native populations, from the eastern Baltic and western Siberian regions and native populations from southwestern Russia. The eastern Baltic region and western Siberia represent the two opposite directions of B. orientalis spread in climatically different zones. The genetic structures of the native and non-native B. orientalis populations were assessed through analysis of molecular variance (AMOVA) and the Bayesian clustering method and by determining the main measures of genetic diversity. AMOVA revealed considerable population differentiation in both the native and invasive ranges. Our results did not indicate a decrease in genetic diversity in the non-native populations of B. orientalis. Similar measures of genetic diversity and genetic structure were determined in the invasive populations in two geographically and ecologically distinct, non-native regions located in Europe and Asia. In both of these regions, higher genetic diversity was detected in the non-native populations than in the native region populations, which may be due to multiple introductions. However, Bayesian clustering analysis revealed slightly different sources of invasive populations in the two non-native regions. Genetic diversity patterns revealed the lack of isolation by distance between populations and confirmed the influence of anthropogenic factors on the spread of B. orientalis. The significance of native populations as germplasm resources for breeding is discussed.  相似文献

The variability of the mtDNA locus COI in the Florida crab (Rhithropanopeus harrisii) of coastal populations of the northern Black Sea is studied. The introduction of this crab species is demonstrated to originate from European populations, most likely the Netherlands. The studied populations experienced a strong decline in the level of genetic variability based on the haplotype diversity, but to a lesser extent based on the nucleotide diversity. Analysis of the structure of decapod community of Crimea makes it possible to discuss the reasons underlying invasive success of the Florida crab in the region.  相似文献

RAPD and ISSR analyses revealed genetic diversity and relationships among 11 populations of two closely related northeast China Vicia species, Vicia ramuliflora and V. unijuga. Both methods yielded similar and complementary results, showing high genetic diversity. Vicia ramuliflora had 100% polymorphic loci in both RAPD and ISSR, and V. unijuga had 100% polymorphic loci for RAPD and 98.96% for ISSR. Genetic differentiation was moderate among populations of each species. Genetic variation was distributed mainly within populations for the two species. The high level of gene flow was important for the allocation of genetic variation. The UPGMA dendrogram and principal coordinates analysis at the level of individuals and populations showed that V. ramuliflora and V. unijuga were more closely related than either of them was to the outgroup species, V. cracca. The small molecular variance of V. ramuliflora and V. unijuga supports the conclusion that these two species had a common ancestor.  相似文献
