Boophilus ticks (n = 25,987) were collected from cattle at 30 communal dip tanks and five commercial farms in the Soutpansberg region, Limpopo Province, South Africa, between May 1999 and December 2001. Only 6.6% were Boophilus decoloratus, while 93.4% were Boophilus microplus, a tick that had not been reported from this area previously. B. microplus was the most common Boophilus tick on communal dip tank cattle, while B. decoloratus was more commonly found on commercial farms. Where the two species occurred together, B. microplus tended to displace B. decoloratus. The displacement was almost complete at the communal dip tanks, while on the commercial farms the population change was still in progress at the end of the survey. The present study demonstrated that a postulated reproductive interference was insufficient in preventing B. microplus from spreading when the climatic conditions were favourable to this species, as the displacement in most of the areas appeared to be rapid and complete.  相似文献

BACKGROUND AND AIMS: Genetic structure and variability were examined in the only three extant populations of the narrow-endemic tree Antirhea aromatica (Rubiaceae, Guettardeae), an endangered species of the tropical forest of eastern Mexico. Patterns of genetic diversity within and among populations for adult plants and seedlings were obtained. METHODS: Allozyme electrophoresis of 15 loci was conducted and the data analysed with statistical approximation for obtaining genetic diversity, structure and gene flow. KEY RESULTS: The mean expected heterozygosity (He) in the adult and seedling populations was 0.18 +/- 0.08 and 0.20 +/- 0.09, respectively. The genetic variation explained by differences among populations was 51 and 35 %, for adult and seedling populations, respectively. On average, gene flow between paired adult populations was low (Nm = 0.26 +/- 0.09), compared with other trees from the tropical forest. CONCLUSIONS: The results indicated that the populations evaluated have high genetic variability, compared with other endemic and geographically narrowly distributed plant species, in areas with high levels of environmental heterogeneity (e.g. tropical forests). The conservation implications of the results are discussed, and in this regard it is proposed that A. aromatica should be considered as an indicator species with economic potential. It is suggested that sustainable management practices should be implemented and that the areas where the species is distributed should be declared a natural reserve to ensure the species conservation.  相似文献

Boophilus microplus infestation is one of the most serious limitations to cattle industry in tropical regions, even though bovines show natural resistance to ticks. This resistance was evaluated in Cross-bred Carora Bulls (CCB) a tropicalized dairy breed from Venezuela. Seven CCB were experimentally infested with B. microplus larvae, "Mozo" strain, they were considered tick-naive because they had never been infested with ticks. The mean inoculum size applied on each bull was 6,477 larvae. After life cycle was completed adult female body weight (BW), egg mass weight (EW), egg hatching rate (%EH), and reproductive index (RI) were recorded. Results revealed a high variability in the levels of resistance to B. microplus. Thus, one animal showed greater resistance (Dunnett, p < 0.05) for the analyzed parameters in contrast with three non-resistant bulls. The others had moderate resistance. The trait "resistance" should be included together with other traits often used in genetic selection of cattle.  相似文献

The pathogenicity of five species of entomopathogenic fungi (Deuteromycetes, species: Beauveria bassiana, Metarhizium anisopliae, Metarhizium flavoviride, Paecilomyces fumosoroseus and Verticillium lecanii ) to the various developmental stages of Boophilus annulatus ticks was compared under laboratory conditions. M. anisopliae and B. bassiana strains were most virulent to engorged females and caused 85-100% mortality within 7-10 days post-inoculation (PI). The highest mortality of engorged females caused by other fungi reached only 25-60%. All tested fungi prevented or reduced the egg laying capability of the ticks several days before their death. Females surviving after treatment with the most virulent M. anisopliae strain (Ma-7) reached only 7-8% of their egg laying capacity as compared with the control. Other fungi caused a reduction of the weight of laid eggs by 35.4-80.8% as compared with untreated females. Only M. anisopliae and B. bassiana strains caused 70-98% mortality of the treated eggs. Unfed larvae of Boophilus annulatus were sensitive to M. anisopliae and M. flavoviride strains. The Ma-7 strain was most virulent to unfed larvae, with a mortality rate of 80.4% at a concentration of 1 ×107 spores ml -1 and 100% mortality at a concentration of 1 ×108 spores ml -1 .  相似文献

The Wood Stork (Mycteria americana) is a colonial wading bird of the tropical and lower subtropical zones. We assessed genetic structure within and among five stork colonies from the Brazilian Pantanal and compared our data with those from North American populations. Samples of 234 individuals were studied using protein electrophoresis to evaluate genetic variability and interpopulation differentiation. Of 22 loci examined, 7 were polymorphic (mean heterozygosity = 0.068). The low Fst value (0.005) indicated little intraspecific variation among breeding colonies. Estimated number of migrants per generation based on private alleles (Nm = 11.3) and on Fst (48.8) suggests high gene flow. Nei's genetic distance values among Pantanal colonies ranged from 0.0001 to 0.0034, demonstrating low genetic divergence among populations. Our data can be explained by supposing high gene flow levels among Pantanal colonies, and between North and South American populations, intermediated by a probable interbreeding population in Central America.  相似文献

We sequenced across all of the gene boundaries in the mitochondrial genome of the cattle tick, Boophilus microplus, to determine the arrangement of its genes. The mtDNA of B. microplus has a coding region, composed of tRNA(Glu) and 60 bp of the 3' end of ND1, that is repeated five times. Boophilus microplus is the first coelomate animal known to have more than two copies of a coding sequence. The mitochondrial genome of B. microplus has other unusual features, including (1) reduced T arms in tRNAs, (2) an AT bias in codon use, (3) two control regions that have evolved in concert, (4) three gene rearrangements, and (5) a stem-loop between tRNA(Gln) and tRNA(Phe). The short T arms and small control regions (CRs) of B. microplus and other ticks suggest strong selection for small genomes. Imprecise termination of replication beyond its origin, which can account for the evolution of tandem repeats of coding regions in other mitochondrial genomes, cannot explain the evolution of the fivefold repeated sequence in the mitochondrial genome of B. microplus. Instead, slipped-strand mispairing or recombination are the most plausible explanations for the evolution of these tandem repeats.  相似文献

Siderastrea stellata and S. radians are scleractinian coral species that present a remarkable overlap of diagnostic characteristics and sympatric distribution. Moreover, both are viviparous with similar reproductive strategies and with a gregarious larval behavior. Samples of both species from the Brazilian coast were analyzed using 18 isozymic loci to quantify their genetic variability and populational structure. Results confirmed species identity, high intrapopulational variability and revealed moderate genetic structuring among all samples (S. stellata: F (ST) = 0.070; S. radians: F (ST) = 0.092). Based on genotypic diversity analysis, there was evidence that local recruitment may have a minor role in the populations (mean, G ( o ):G ( e )= 1.00 +/- 0.0003 SD for S. stellata and 0.99 +/- 0.0023 SD for S. radians). Deviations towards heterozygote deficiencies found in both Siderastrea species could be explained by the Wahlund effect, since there was evidence that populations might be composed of colonies of different ages. In S. radians it is also likely that there is some inbreeding occurring in the studied populations. Despite the brooding pattern and the gregarious larval behavior, our data suggest the occurrence of gene flow along the Brazilian coast. This is the first study on population genetics of Brazilian reef corals.  相似文献

We describe the isolation and characterisation of two putatively new acetylcholinesterase genes from the African cattle ticks Boophilus decoloratus and Rhipicephalus appendiculatus. The nucleotide sequences of these genes had 93% homology to each other and 95% and 91% identity, respectively, to the acetylcholinesterase gene from an Australian strain of another cattle tick, Boophilus microplus. Translation of the nucleotide sequences revealed putative amino acids that are essential for acetylcholinesterase activity: the active site serine, and the histidine and glutamate residues that associate with this serine to form the catalytic triad. All known acetylcholinesterases have three sets of cysteines that form disulfide bonds; however, the acetylcholinesterase genes of these three species of ticks encode only two sets of cysteines. Acetylcholinesterases of B. microplus from South Africa, Zimbabwe, Kenya and Mexico had 98-99% identity with acetylcholinesterase from B. microplus from Australia, whereas acetylcholinesterase from B. microplus from Indonesia was identical to that from Australia. Preliminary phylogenetic analyses surprisingly indicate that the acetylcholinesterases of ticks are closer phylogenetically to acetylcholinesterases of vertebrates than they are to those of other arthropods.  相似文献

BACKGROUND AND AIMS: The monoecious, bird-pollinated epiphytic Tillandsia achyrostachys E. Morr. ex Baker var. achyrostachys is an endemic bromeliad of the tropical dry forests of Mexico with clonal growth. In the Sierra de Huautla Natural Reserve this species shows a host preference for Bursera copallifera (Sessé & Moc ex. DC) Bullock. As a result of deforestation in the study area, B. copallifera has become a rare tree species in the remaining forest patches. This human-induced disturbance has directly affected the population densities of T. achyrostachys. In this study the genetic consequences of habitat fragmentation were assessed by comparing the genetic diversity, gene flow and genetic differentiation in six populations of T. achyrostachys in the Sierra de Huautla Natural Reserve, Mexico. METHODS: Allozyme electrophoresis of sixteen loci (eleven polymorphic and five monomorphic) were used. The data were analysed with standard statistical approximations for obtaining diversity, genetic structure and gene flow. KEY RESULTS: Genetic diversity and allelic richness were: HE = 0.21 +/- 0.02, A = 1.86 +/- 0.08, respectively. F-statistics revealed a deficiency of heterozygous plants in all populations (Fit = 0.65 +/- 0.02 and Fis = 0.43 +/- 0.06). Significant genetic differentiation between populations was detected (Fst = 0.39 +/- 0.07). Average gene flow between pairs of populations was relatively low and had high variation (Nm = 0.46 +/- 0.21), which denotes a pattern of isolation by distance. The genetic structure of populations of T. achyrostachys suggests that habitat fragmentation has reduced allelic richness and genetic diversity, and increased significant genetic differentiation (by approx. 40 %) between populations. CONCLUSIONS: The F-statistic values (>0) and the level of gene flow found suggest that habitat fragmentation has broken up the former population structure. In this context, it is proposed that the host trees of T. achyrostachys should be considered as a conservation priority, since they represent the limiting factor to bromeliad population growth and connectivity.  相似文献

Since its immigration in the Pacific island of New Caledonia in 1942 (i.e. about 240 tick-generations ago), the cattle tick Boophilus microplus has experienced a remarkable adaptive diversification there. In order to better understand the population factors involved, we have investigated the B. microplus population structure on that main host-species, Bos taurus. This study was based microsatellite loci and confirmed that the island colonization came along with a significant bottleneck. Knowledge on B. microplus biology led us to expect B. microplus populations to be composed of highly inbred lineages irregularly dispatched among the individual hosts belonging to the same herds. Instead, this study evidenced a weak inbreeding level and an absence of genetic differentiation within herds. Complementarily, a significant signal of isolation by distance exhibited that human-traffic of cattle does not promote high tick dispersal within the island. Finally, the tick density was found to be about a few hundreds of reproducing adults per squared kilometre, for a gene dispersal range of about a few hundred metres per tick generation. Results are discussed with regard to the evolution of new adaptive changes.  相似文献

Genetic variation and differentiation in six island populations of two species ( Cynopterus brachyotis Müller and Haplonycteris fischeri Lawrence) of Philippine fruit bats (Chiroptera, Pteropodidae) were analysed using allozyme electrophoresis. Cynopterus is eurytopic and widespread in southeast Asia; Haplonycteris is stenotopic and endemic to the Philippines. Genetic variability within populations is consistently higher in Cynopterus , but differentiation between populations is much more pronounced in Haplonycteris. Genetic variation is not significantly correlated with island size in either species, but a positive trend is present in both. Levels of gene flow are sufficiently low to allow differentiation by genetic drift alone in Haplonycteris ( Nm = 0.05), but not in Cynopterus ( Nm = 7.5). There is no significant association between genetic distance and distance between sampling sites; however, between-population differentiation is positively related to degree of geographic isolation during Pleistocene periods of low sea level and to vagility and consequent levels of gene flow among populations. Significant effects of population size on genetic differentiation were not found. Genetic distance matrices for the two species share a common structure that is similar to patterns of mammalian faunal similarity for the Philippines as a whole, suggesting similar effects of geographic and/or environmental factors.  相似文献

The levels of genetic diversity assessed from allozyme data were investigated in 25 populations of Mediterranean Bromus intermedius, B. squarrosus, B. lanceolatus, and B. hordeaceus from Algeria. The geographically restricted diploids B. intermedius and B. squarrosus displayed less genetic diversity (the mean population gene diversity of Nei (Hu) ranged from 0.03 to 0.12) than the widespread tetraploid colonizers B. lanceolatus and B. hordeaceus (Hu = 0.07-0.27). Deviations from Hardy-Weinberg expectations in diploid populations of B. intermedius and B. squarrosus were observed owing to heterozygote excess at several loci and suggested that these self-fertilizing species may have substantial amounts of allogamy. Tetraploid populations of B. lanceolatus and B. hordeaceus were largely homozygous at homologous loci and frequently exhibited intergenomic fixed heterozygosity in accordance with their alloploid origin. Genetic variation at the infraspecific level was mostly distributed within populations in the four species, B. hordeaceus showing the lowest level of interpopulation differentiation (Gst = 0.06) and the highest level of gene flow (Nm = 3.75). Consistent gene flows are in agreement with the strongest intercontinental invasive behaviour of B. hordeaceus. Less differentiation was reported in the literature among later introduced B. hordeaceus populations from England and Australia, indicating reduced differentiation under the process of colonization. Moderate divergence occured among the four taxa, with interspecific genetic identities ranging from 0.87 to 0.93. In spite of substantial genetic similarity, species were clearly differentiated, with each tetraploid being more closely related to a diploid: B. hordeaceus to B. squarrosus and B. lanceolatus to B. intermedius.  相似文献

Highly specialized obligatory cave beetles endemic to the French Pyrenees offer an opportunity to investigate the relative importance of environmental conditions and ecological characteristics on the organization of genetic variability, to describe the genetic structure of populations, and to assess the extent of gene flow between local populations in relation to geologic structure. Twenty-three geographically close populations of the beetle Speonomus hydrophilus occurring both in caves (reduced fluctuations in many abiotic parameters) and under the deepest layer of soil in mountains (more exposed to climatic variations) were studied. Significant genetic differentiation at 17 allozyme loci was found among populations in close proximity, as well as among those from distant parts of range. On a larger scale, genetic differences among populations appear to result from low dispersal rates between populations. The spatial patterning observed suggests that allozyme frequencies are not responding to environmentally controlled selection. Substantial genetic divergence (F(ST) = 0.112) occurred throughout the range, with important variation in levels of genetic variability (H: 0.065-0.184) among populations. A significant level of substructuring has occurred among the populations with four major geographic areas of similarity indicated. The substructuring of the species into regions suggests an influence of paleoclimatic gradient and paleoenvironment on the population's genetic structure. Also, founder effect and reduced gene flow appear to have influenced populations in the southeastern portion of the range.  相似文献

Ballota undulata, Ballota kaiseri, and Ballota saxatilis are very rare (and endemic--B. kaiseri), threatened species growing in St. Catherine Protectorate, southern Sinai, Egypt. They are subjected to a number of threats that have caused populations to decline in both number and size. For the long-term survival of these species, an appropriate conservation strategy for the maintenance of their genetic variation should be developed. This study measures genetic diversity within and among populations of these Ballota species and determines the conservation implications of the results. The genetic analyses demonstrated that the three Ballota species maintain relatively high levels of genetic diversity (He = 0.195-0.317) and that most of the their genetic diversity was found within populations (GST = 0.045-0.099). Indirect estimates of historical gene flow for B. undulata and B. saxatilis were relatively high (Nm(W) = 5.25 and 3.37, respectively) but suggest that there is somewhat less gene movement among B. kaiseri populations (Nm(W) = 2.29). The levels of genetic diversity maintained within populations of the three Ballota species indicate that an appropriate sampling design for ex situ safeguarding should capture the majority of the genetic diversity found within these taxa.  相似文献

We report the cloning, sequence characterization and expression analysis of a calreticulin (CRT) coding cDNA of Boophilus microplus. CRT is a calcium-binding protein involved in multiple cell functions and possibly implicated in parasites host immune system evasion. The CRT cDNA sequence and its molecular characterization are described. Sequence similarity and phylogenetic analyses indicate a close relationship to other arthropod CRT sequences. The CRT cDNA was also expressed in a procariotic system and the recombinant protein (rBmCRT) was used to raise antibodies in a rabbit. Expression analyses of the corresponding gene in different developmental stages and tissues were performed by RT-PCR and Western-blot, which indicated a ubiquitous expression of the B. microplus calreticulin gene and demonstrated its presence in saliva. Sera of tick-infested bovines suggested that this protein may not be able to induce an IgG-based humoral response in its natural host.  相似文献

A survey was carried out to investigate the prevalence of hard tick species (Acari: Ixodidae) on cattle in Mazandaran province, Iran. A total of 953 ticks were collected from 86 infested cattle during activating seasons of ticks during 2004-2005. Nine species were identified: Boophilus annulatus (51.3%), Rhipicephalus bursa (16.8%), Haemaphysalis punctata (6.3%), Ixodes ricinus (6.8%), Hyalomma marginatum (12.5%), Hyalomma anatolicum excavatum (5.2%), Hyalomma asiaticum (0.6%), Hyalomma detritum (0.2 %), and Dermacentor spp. (0.1%). The results show that Boophilus annulatus, Rhipicephalus bursa, and Hyalomma species are dominant tick species in the surveyed area.  相似文献

The suitability of white-tailed deer (Odocoileus virginianus) as hosts for the cattle ticks Rhipicephalus (Boophilus) microplus and Rhipicephalus (Boophilus) annulatus, has been well documented. These ticks have a wide host range, and both transmit Babesia bovis and Babesia bigemina, the agents responsible for bovine babesiosis. Although this disease and its vectors have been eradicated from the United States and some states in northern Mexico, it still is a problem in other Mexican states. It is not known if wild cervids like white-tailed deer can act as reservoirs for bovine babesiosis. The purpose of this study was to determine if B. bovis and B. bigemina or antibodies against them occur in white-tailed deer in the states of Nuevo Leon and Tamaulipas, Mexico. Twenty blood samples from white-tailed deer from two ranches were collected and tested with a nested polymerase chain reaction (nested PCR) and indirect immunofluorescence antibody test (IFAT) for B. bovis and B. bigemina. Eleven samples were positive for B. bigemina and four for B. bovis by nested PCR; amplicon sequences were identical to those reported in GenBank for B. bovis (Rap 1) and B. bigemina. Results of the IFA test showed the presence of specific antibodies in serum samples. This is the first report of the presence of B. bovis and B. bigemina in white-tailed deer using these techniques and underscores the importance of cervids as possible reservoirs for bovine babesiosis.  相似文献

The genetic variability of eight fish-farm and three natural populations of turbot was studied by electrophoretic analysis of 35 enzymatic loci. The results showed low genetic variability in natural populations of turbot ( H T = 0·029 ± 0·013) in comparison with other flatfish species. Great genetic similarity was revealed among the natural populations studied, which indicates high rates of gene flow in this species. The hatchery stocks showed less genetic variation than the wild populations analysed, which suggests genetic drift phenomena involved in the foundation and management of broodstocks. In addition, the heterozygosity differences detected among the hatchery stocks analysed are correlated with inverted levels of fluctuating asymmetry, which supports the existence of inbreeding depression phenomena in turbot culture.  相似文献
