Conclusions Except for the pronounced adaptation-hysteresis effect, the pulse experiments exhibited the expected trend: deviation from the steady feed reference curve was greatest at lowest dilution rates. Under conditions of lowest glucose level the effect of pulsing would be expected to cause the largest fluctuations in glucose, causing a certain fraction of the cells to ferment. Generally over the entire dilution rate range the biomass production was decreased and the ethanol was increased by pulsing the feed stream. There is, however, some evidence that pulse feeding can trigger quite unexpected results. Point (6) at D=0.3 h–1 in Fig. 1 exhibit a biomass productivity which was about 20% greater than the continuous feeding reference value (DX=3.6 kg m–3 h–1 as compared with 3.0 kg m–3 h–1). Such performance would be of significant commercial value, but the poor reproducibility due to adaptation, as seen here, certainly would preclude its application.The results obtained should also be applicable to fed batch operation at the corresponding glucose level. Further experiments including the variation of the glucose feeding period would be necessary to obtain a conclusive picture. A bacterial consortium with complementary metabolic capabilities was formulated and specific removal rates were 0.14, 0.35, 0.04, and 0.39 h–1 for benzene, toluene, o-xylene, and m,p-xylene, respectively. When immobilized on a porous peat moss biofilter, removal of all five (= BTX) components was observed with rates of 1.8–15.4 g m–3 filter bed h–1. Elimination capacities with respect to the inlet gas concentrations of BTX and airflow rates showed diffusive regimes in the tested concentration range of (0.1–5.3 g m–3) and airflow (0.55–1.82 m3 m–2 h–1) except for o-xylene which reached its critical gas concentration at 0.3 g m–3.  相似文献

The biological removal of ammonia and butanal in contaminated air was investigated by using, respectively, a laboratory-scale filter and a scrubber-filter combination. It was shown that ammonia can be removed with an elimination efficiency of 83% at a volumetric load of 100 m3·m–2·h–1 with 4–16 ppm of ammonia. During the experiment percolates were analysed for nitrate, nitrite, ammonium and pH. It was found that the nitrification in the biofilter could deteriorate due to an inhibition of Nitrobacter species, when the free ammonia concentration was rising in the percolate. It should be easy to control such inhibition through periodic analysis of the liquid phase by using a filter-scrubber combination. Such a combination was studied for butanol removal. Butanal was removed with an elimination efficiency of 80% by a scrubber-filter combination at a volumetric load of 100 m3·m–2·h–1 and a high butanal input concentration. Mixing the filter material with CaCO3 and pH control of the liquid in the scrubber resulted in an increase of the elimination efficiency. These results, combined with previous results on the biofiltration of butanal and butyric acid, allow us to discuss the influence of odour compounds on the removal efficiency of such systems and methods for control. Ethanol was produced with Zymomonas mobilis Z6 (ATCC 29191), in batch culture with synthetic medium on glucose as substrate and in the presence of aspartate. The concentrations of glucose, phosphate, ammonium, ethanol and dissolved O2 and CO2 in the medium and O2 and CO2 in the outlet gas as well as the cell mass by culture fluorescence were measured on-line. Cell mass, glucose and aspartate concentrations were measured off-line. In the presence of a sufficient amount of aspartate, the ethanol inhibition effect can be reduced considerably. However, the improvement with yeast extract is more incisive. Rhodospirillum rubrum was grown continuously and photoheterotrophically under light limitation using a cylindrical photobioreactor in which the steady state biomass concentration was varied between 0.4 to 4 kg m–3 at a constant radiant incident flux of 100 W m–2. Kinetic and stoichiometric models for the growth are proposed. The biomass productivities, acetate consumption rate and the CO2 production rate can be quantitatively predicted to a high level of accuracy by the proposed model calculations. Several biofilters and biotrickling filters were used for the treatment of a mixture of formaldehyde and methanol; and their efficiencies were compared. Results obtained with three different inert filter bed materials (lava rock, perlite, activated carbon) suggested that the packing material had only little influence on the performance. The best results were obtained in a biotrickling filter packed with lava rock and fed a nutrient solution that was renewed weekly. A maximum formaldehyde elimination capacity of 180 g m–3 h–1 was reached, while the methanol elimination capacity rose occasionally to more than 600 g m–3 h–1. Formaldehyde degradation was affected by the inlet methanol concentration. Several combinations of load vs empty bed residence time (EBRTs of 71.9, 46.5, 30.0, 20.7 s) were studied, reaching a formaldehyde elimination capacity of 112 g m–3 h–1 with about 80% removal efficiency at the lowest EBRT (20.7 s).  相似文献

Outdoor chemostat cultures of the marine microalgaIsochrysis galbana at constant dilution rate (0.034 h–1 ) have been carried out under different weather conditions. Steady-state biomass concentrations were 1.61±0.03 kg m–3 in May and 0.95±0.04 kg m–3 in July, resulting in biomass output rates of 0.54 kg m–3 d–1 and 0.32 kg m–3 d–1 in May and July, respectively. Two patterns of daily variation with the solar cycle were observed in the fatty acid content. Saturated and mono-unsaturated fatty acids (16:0 and 16:1n7) show significant variation with the solar cycle, associated with short-term changes in environmental factors. Palmitic and palmitoleic acids are generated during daylight and consumed during the dark period. However, polyunsaturated fatty acids do not show a significant response to the solar cycle and their changes are associated with long-term variation in environmental factors. The maximum EPA productivity was obtained in May, 14.1 g m–3 d–1, which is close to that found in the literature for indoor continuous cultures. Nonetheless, the outdoor EPA content (up to 2.61 % d.wt) was lower than the indoor EPA content from a previous study (5% d.wt).  相似文献

Ethanol production in a continuous fermentation/membrane pervaporation system   总被引:12,自引:0,他引:12  
The productivity of ethanol fermentation processes, predominantly based on batch operation in the U.S. fuel ethanol industry, could be improved by adoption of continuous processing technology. In this study, a conventional yeast fermentation was coupled to a flat-plate membrane pervaporation unit to recover continuously an enriched ethanol stream from the fermentation broth. The process employed a concentrated dextrose feed stream controlled by the flow rate of permeate from the pervaporation unit via liquid-level control in the fermentor. The pervaporation module contained 0.1 m2 commercially available polydimethylsiloxane membrane and consistently produced a permeate of 20%–23% (w/w) ethanol while maintaining a level of 4%–6% ethanol in a stirred-tank fermentor. The system exhibited excellent operational stability. During continuous operation with cell densities of 15–23 g/l, ethanol productivities of 4.9–7.8 gl–1 h–1 were achieved utilizing feed streams of 269–619 g/l glucose. Pervaporation flux and ethanol selectivities were 0.31–0.79 lm–2 h–1 and 1.8–6.5 respectively.  相似文献

Removal of toluene in waste gases using a biological trickling filter   总被引:12,自引:0,他引:12  
The removal of toluene from waste gas was studied in a trickling biofilter. A high level of water recirculation (4.7 m h–1) was maintained in order to keep the liquid phase concentration constant and to achieve a high degree of wetting. For loads in the range from 6 to 150 g m–3 h–1 the maximum volumetric removal rate (elimination capacity) was 35±10 g m–3 h–1, corresponding to a zero order removal rate of 0.11±0.03 g m–2 h–1 per unit of nominal surface area. The surface removal was zero order above the liquid phase concentrations of approximately 1.0 g m–3, corresponding to inlet gas concentrations above 0.7–0.8 g m–3. Below this concentration the surface removal was roughly of first order. The magnitude of the first order surface removal rate constant, k1A , was estimated to be 0.08–0.27 m h–1 (k1A a=24–86 h–1). Near-equilibrium conditions existed in the gas effluent, so mass transfer from gas to liquid was obviously relatively fast compared to the biological degradation. An analytical model based on a constant liquid phase concentration through the trickling filter column predicts the effluent gas concentration and the liquid phase concentration for a first and a zero order surface removal. The experimental results were in reasonable agreement with a very simple model valid for conditions with an overall removal governed by the biological degradation and independent of the gas/liquid mass transfer. The overall liquid mass transfer coefficient, KLa, was found to be a factor 6 higher in the system with biofilm compared to the system without. The difference may be explained by: 1. Difference in the wetting of the packing material, 2. Mass transfer occurring directly from the gas phase to the biofilm, and 3. Enlarged contact area between the gas phase and the biofilm due to a rough biofilm surface.  相似文献

Fermentation in tubular recycle reactors with high biomass concentrations is a way to boost productivity in alcohol production. A computer model has been developed to investigate the potential as well as to establish the limits of this process from a chemical engineering point of view. The model takes into account the kinetics of the reaction, the nonideality of flow and the segregation in the bioreactor. In accordance with literature, it is shown that tubular reactors with biomass recycle can improve productivity of alcohol fermentation substantially.With the help of the computer based reactor model it was also possible to estimate the detrimental effects of cell damage due to pumping. A diauxic fermentation was observed during batch fermentation of enzyme-hydrolyzed whey permeate to ethanol by Saccharomyces cerevisiae. Glucose was consumed before and much faster than galactose. In the continuous membrane recycle bioreactor (MRB), sugar utilization was a function of dilution rate and concentration of sugars. At a cell concentration of 160 kg/m3, optimum productivity was 31 kg/(m3 · h) at ethanol concentration of 65 kg/m3. Low levels of acetate (0.05–0.1 M) reduced cell growth during continuous fermentation, but also reduced galactose utilization.  相似文献

Summary The effect of a single pulse of ammonium sulphate or of citrate upon the progress and final outcome of a batch citric acid fermentation was studied. It was found that the optimum addition time for the supplemental N was in the range of 40 to 75 h. Final citric acid concentration achieved was significantly increased when the concentration of N source added was between 0.25 and 0.5 kg m–3. The mechanism of the observed stimulation seemed to be an indirect one. Addition of exogenous citric acid to the broth, led to an increase in citrate production by the culture. Membrane technologies were investigated with the aim to improve stability of C-Phycocyanin extracts resulting from ultrasonic breakage of Spirulina platensis. Five membranes, ranging from microfiltration to reverse osmosis, were utilized both for clarification and concentration steps. Nanofiltration with tubular organic membranes exhibited good performances: pigment recovery was 100%, mean permeation flux was 85 l h–1 m–2 for achieving a concentration factor of 7 with 30×105 Pa pressure and 1.5 m s–1 tangential velocity (turbulent flow).  相似文献

An upflow velocity of 0.21 m h–1 was optimal to minimize the effect of organic shocks (from 6 to 30 kg COD m–3 d–1) when operating an upflow anaerobic filter for the treatment of an oleic acid-based effluent (50% w/v COD). This value represented the transition between a mass transfer controlled regime and a kinetic regime. Under hydraulic shock loads, an increase in upflow velocity from 0.3 to 0.4m h–1 decreased the removal efficiency from 68 to 51%.  相似文献

Summary Continuous fermentation fed by 150 kg/m3 of glucose with total cell recycling by tangential microfiltration enabled yeasts concentration of 300 kg/m3 (dry weight) to be reached with a dilution rate of 0,5h–1 and a cell viability greater than 75%. Summary A high performance fermentation process for the continuous production of citric acid from sugarcane molasses by using the combination of submerged calcium alginate-immobilized and surface-stabilized cultures of Aspergillus niger KCU 520 in a continuous flow horizontal bioreactor is described. The citric acid productivity was dependent on the dilution rate with an optimum value of 0.015/h. Presaturation of fermentation medium with sterile air, in addition to surface aeration, before feeding into the bioreactor enhanced the citric acid productivity. The highest productivity, citric acid product concentration and yield obtained were 1.7 kg M–3h–1, 110kg M–3 and 91% respectively. The cultures were continuously used for 30 days without any apparent loss in citric acid productivity.  相似文献

Two biofilters fed toluene-polluted air were inoculated with new fungal isolates of either Exophiala oligosperma or Paecilomyces variotii, while a third bioreactor was inoculated with a defined consortium composed of both fungi and a co-culture of a Pseudomonas strain and a Bacillus strain. Elimination capacities of 77 g m–3 h–1 and 55 g m–3 h–1 were reached in the fungal biofilters (with removal efficiencies exceeding 99%) in the case of, respectively, E. oligosperma and Paecilomyces variotii when feeding air with a relative humidity (RH) of 85%. The inoculated fungal strains remained the single dominant populations throughout the experiment. Conversely, in the biofilter inoculated with the bacterial–fungal consortium, the bacteria were gradually overgrown by the fungi, reaching a maximum elimination capacity around 77 g m–3 h–1. Determination of carbon dioxide concentrations both in batch assays and in biofiltration studies suggested the near complete mineralization of toluene. The non-linear toluene removal along the height of the biofilters resulted in local elimination capacities of up to 170 g m–3 h–1 and 94 g m–3 h–1 in the reactors inoculated, respectively, with E. oligosperma and P. variotii. Further studies with the most efficient strain, E. oligosperma, showed that the performance was highly dependent on the RH of the air and the pH of the nutrient solution. At a constant 85% RH, the maximum elimination capacity either dropped to 48.7 g m–3 h–1 or increased to 95.6 g m–3 h–1, respectively, when modifying the pH of the nutrient solution from 5.9 to either 4.5 or 7.5. The optimal conditions were 100% RH and pH 7.5, which allowed a maximum elimination capacity of 164.4 g m–3 h–1 under steady-state conditions, with near-complete toluene degradation.  相似文献

The accumulation of biofilm by Acetobacterium sp. during continuous culture in an upflow anaerobic filter (UAF) growing on methanol-formate was the result of space velocity and inlet concentrations of substrate and Co+2. To achieve good development of biofilm, a space velocity of 0.38 h–1, inlet substrate concentrations of 125 mM of both methanol and formate, and Co+2 at 0.16 mM were required. Cell productivities in the effluent of the UAF-reactor were about 6-fold higher than in chemostat cultures (0.20 g l–1 h–1 for UAF and 0.035 g l–1 h–1 for chemostat) (previous studies), and the maximum vitamin B12 specific concentration was 5.1 mg g cell–1.  相似文献

Pseudomonas sp. SR-5 was isolated as a styrene-degrading bacterium. In liquid culture containing 1% (v/v) styrene, more than 90% styrene was degraded in 53 h and the doubling time of SR-5 was 2 h. The removal of styrene gas was investigated in biofilters for 31 days using an organic packing material of peat and an inorganic packing material of ceramic inoculated with SR-5. The maximum-styrene-elimination capacities for peat and ceramic packing materials were 236 and 81 g m–3 h–1, respectively. The percentage of styrene converted to low molecular weight compounds including CO2 in the peat and ceramic biofilters during a 10-day operation were estimated to be 90.4 and 36.7%, respectively. As the pressure drop in the peat bioflter at the end of experiment was significantly higher than that in ceramic biofilter, a biofilter using a mixture of peat and ceramic was tested. We determined that the maximum elimination capacity was 170 g m–3 h–1 and the production of low molecular weight compounds was 95% at a low pressure drop for this mixed packing material filter.  相似文献
