The objective of the study was to determine the coefficients of ileal apparent digestibility (CIAD) of sorghum protein and amino acids (AA) in weaned piglets and growing pigs. Digestibility coefficients were estimated using the regression and difference methods for the weaned piglets; and the direct and difference methods for the growing pigs. To test the hypothesis that CP and AA digestibility of sorghum is lower in weaned piglets than in growing pigs, two experiments were conducted. In experiment one, 20 weaned piglets were fitted with a 'T' cannula at 21 days of age and were fed for 2 weeks one of five dietary treatments: a reference or control diet providing 200 g of CP/kg from casein (C) as the sole protein source, and four casein-sorghum (C-S) diets kept isoproteic to C by the appropriate adjustment of C and maize starch proportions; the amount of sorghum (S) in these diets was 135, 307, 460 and 614 g/kg. In experiment 2, fifteen castrated pigs weighing 57.8 ± 2.8 kg were used and randomly allotted to one of three dietary treatments: a reference casein-maize starch diet containing C as the sole protein source, a C-S diet, both diets containing 160 g of CP/kg, and a fortified S diet containing 68 g of CP/kg. In piglets the CIAD for CP and AA decreased linearly (P < 0.05) as the amount of S in the diet increased. The average ileal digestibility of AA from C was 0.858 ± 0.111, and decreased to 0.663 ± 0.191 at the higher S level. The CIAD estimated using the regression or difference methods were similar for leucine, cysteine, glutamic acid, serine, alanine and tyrosine, and different for the other AA. In growing pigs the CIAD of protein and AA (except alanine and cysteine) were similar (P > 0.05) for the C and the C-S diets, but higher (P < 0.05) than those for the S diet. The CIAD for S obtained by the difference method were higher (P < 0.05) than those obtained using the direct method, except for lysine, isoleucine, valine, methionine, threonine and cysteine. The results indicate that except for lysine and cysteine, growing pigs' ability to digest AA and protein is superior than weaned piglets.  相似文献

The influence of two commercial xylanase preparations on the apparent metabolisable energy (AME) and, ileal and excreta amino acid digestibilities in wheat for broilers was investigated. The following three dietary treatments were tested: a basal diet containing 918 g kg−1 wheat, basal diet plus enzyme 1 and basal diet plus enzyme 2. Each diet was offered ad libitum to four pens (four birds/pen) from day 35 to day 42 post-hatching. Total collection of excreta was carried out during the last 4 days. All birds were euthanatised on day 42 and the contents of the lower half of the ileum were collected. Apparent ileal and excreta amino acid digestibilities were calculated using acid-insoluble ash as the indigestible marker. The AME values of wheat were markedly improved by exogenous enzymes. Addition of enzymes 1 and 2 improved the AME values by 12.6% and 18.6%, respectively. Supplementation of enzymes resulted in significant (p<0.05) improvements in ileal and excreta amino acid digestibilities in wheat. Mean excreta digestibility coefficients of the 15 amino acids in wheat, wheat plus enzyme 1 and wheat plus enzyme 2 were 0.70, 0.78 and 0.79, respectively. The corresponding values at the ileal level were 0.78, 0.84 and 0.85, respectively. Excreta digestibility of amino acids, with the exception of tyrosine, were found to be consistently lower than the ileal estimates. In the case of unsupplemented wheat, apparent digestibilities of individual amino acids measured in excreta were 5–22% units lower (p<0.10–0.01) than those measured at the terminal ileum. The ileal-excreta differences were generally lowered by the addition of enzymes.  相似文献

The aim of this study was to test the hypothesis of an improved gut environment of post-weaning piglets when administered a blend of essential oils (EO; thymol and cinnamaldehyde) and an enzyme combination (xylanase and β-glucanase (XB)) either alone or in combination. To assess the effect of dietary treatments, faecal nutrient digestibility and microbial counts, as well as ileum histology and gene expression of inflammatory mediators were evaluated. One hundred and ninety-two weaned piglets were allocated into four experimental treatments, and fed the basal diet (CTRL) either without or with EO, XB or their combination (EO+XB) for a 42-day period. The experiment concerning digestibility was designed with two periods (period I: days 15 to 21; period II: days 29 to 35) and the faeces were collected on days 20, 21, 34 and 35. On day 42, six piglets from each treatment were slaughtered. It was found that EO, XB and EO+XB supplementation did not affect (P>0.05) the growth performance of the piglets from days 0 to 42. Moreover, no dietary effect on faecal score was observed. Faecal digestibility of dry matter, organic matter, ash, dietary fibre, lipid, CP and NDF were increased from period I to period II (P<0.01 to P=0.06), while no effect (P>0.05) of EO, XB or their combination on the faecal digestibility was observed at both periods. Compared with the CTRL diet, dietary XB reduced the faecal Lactobacillus and Escherichia coli counts but increased the Lactobacillus to Coliforms ratio on day 42 (P=0.02, 0.03 and 0.03, respectively), and all the additives supplementations decreased the counts of faecal Coliforms on day 42 (P<0.01). XB supplementation increased the villus to crypt ratio (P=0.04) and reduced the mucosal macrophages number (P<0.01) in the ileum compared with the CTRL group, and dietary EO or EO+XB decreased the number of lymphatic follicles (P=0.01 and P<0.01, respectively) and mucosal macrophages (P=0.02 and P<0.01, respectively). In addition, the interleukin (IL)-1α was downregulated in piglets treated with EO+XB compared with the EO group (P=0.02). In conclusion, the administration of EO, XB or their combination was effective in improving ileum histology, and EO+XB supplementation might benefit the modulation of the expression of ileum inflammatory cytokines in piglets.  相似文献

In pigs, digestive disorders associated with weaning lead to antibiotic use to maintain intestinal health. Microalgae have been studied in humans and rodents for their beneficial effects on health. The nutritional value of microalgae in animal diets has been assessed, but results were not conclusive. Dietary supplementation with microalgae as an alternative to antibiotic use was studied in two trials (72 piglets with initial BW=9.1±1.1 kg in trial 1 and 24 piglets with initial BW=9.1±0.9 kg in trial 2). All piglets were weaned at 28 days of age and then housed in individual cages. Piglets were randomly allocated to one of the four diets during 2 weeks after weaning: a standard diet with no supplementation (NC) or the standard diet supplemented with 1% Spirulina (SP), with 1% Chlorella (CV), or with 0.2% of colistin as positive control (PC). Trial 1 was performed to determine the effect of microalgae supplementation from 28 to 42 days on performance and incidence of diarrhoea. Animals received then a standard diet from 42 to 56 days of age. Trial 2 was performed from 28 to 42 days of age to assess nutrient digestibility of the experimental diets and to determine inflammatory status and intestinal morphology at 42 days of age. In trial 1, 94% of the pigs had diarrhoea in the 1st week after weaning with no beneficial effect of colistin on diarrhoea incidence, average daily feed intake (ADFI), average daily gain (ADG), and gain : feed (G : F) ratio. This suggests that the diarrhoea was due to digestive disorders that did not result from enterotoxigenic Escherichia coli infection. Supplementation with either Spirulina or Chlorella did not affect ADFI, ADG and G : F in trials 1 and 2 (P>0.10). Diarrhoea incidence was reduced in CV pigs compared with NC, SP and PC pigs (P<0.05). Total tract digestibility in pig receiving microalgae was greater for gross energy (P<0.05), and tended to be greater for dry matter, organic matter and NDF (P<0.10) compared with NC and PC pigs. Villus height at the jejunum was greater in SP and CV pigs compared with NC and PC pigs (P<0.05). This study shows a potential effect of both Spirulina and Chlorella supplementation on intestinal development and a potential of Chlorella supplementation to manage mild digestive disorders. Further investigation is necessary to determine the mechanism action of Spirulina and Chlorella on gut health and physiology.  相似文献

Protein sources from cereals are used in broiler diets, usually in order to reduce feeding costs. However, their efficient use in poultry diets is limited by the level of fiber whose compounds are resistant to digestion in the small intestine; due to this sugars are not digested by endogenous poultry enzymes. The aim of this study was to determine the effect of multi-carbohydrase (MC) and phytase (Phy) on the total retention of nutrients, retention of apparent metabolizable energy corrected for nitrogen (AMEN) (trial 1) and apparent and standardized ileal digestibility of amino acids (trial 2) of rice bran (RB). A total of 245-day-old male broilers (Cobb 500) was distributed at 21-day-old in a completely randomized design in a 2 × 2 + 1 (0 and 200 mg/kg MC; 0 and 50 mg/kg Phy, and basal diet – BD) factorial arrangement of treatments, to give seven replicates and seven birds per replicate. The BD based on corn (trial 1) and cornstarch and casein (trial 2) was used only to determine the coefficients of retention of nutrients and energy, and coefficients of digestibility of amino acids of the RB. The test diets were made by mixing BD and RB 7 : 3 wt/wt basis. There was interaction (P<0.05) between MC × Phy for DM, nitrogen and AMEN, retention and no interaction (P>0.05) for ash, calcium, phosphorous and NDF was observed. Enzymes interacted (P<0.05) on standardized ileal digestibility of arginine, histidine, leucine, methionine, phenylalanine, threonine, valine, aspartic acid, glutamic acid, proline and serine. Dietary combination of MC and Phy resulted in higher (P<0.05) standardized digestibility of arginine, histidine, methionine and threonine relative to single enzyme supplementation or control diet without enzymes. Enzyme isolated inclusions in the diets improved (P<0.05) standardized digestibility of methionine. The supplementation of carbohydrases and Phy in RB will improve the nitrogen, energy and amino acids utilization for broiler chickens.  相似文献

Sixteen female piglets (58 d of age, 16.8 ± 0.8 kg body weight [BW]) were assigned to two groups (n = 8) and received until day 100 of age (50.3 ± 1.2 kg BW) ad libitum either a diet with a standard (diet C) or low (diet L) total phosphorus (P) content (5.38 and 4.23 g/kg, respectively). Diet C was supplemented with mineral P (1.15 g/kg) and did not contain microbial phytase. Diet L did not contain any inorganic P but 750 FTU/kg of microbial phytase. Despite these treatments, both diets were composed with the same ingredients. Body mineralisation of each gilt was assessed by determining the bone mineral content (BMC), area bone mineral density (BMD) by the dual-energy X-ray absorptiometry (DXA) at days 58, 72, 86 and 100 of age. Feeding diet L caused a higher P digestibility (p = 0.008) measured from days 72 to 86 of age and at 100 days of age a higher BMC and BMD (p ≤ 0.01). Furthermore, the gilts of group L deposited more minerals in the body than control pigs (by 2.4 g/d, p = 0.008). It was found that BMD and BMC were positively correlated with body lean mass and digestible P intake. The results indicated that, even for very young pigs, the addition of microbial phytase instead of inorganic P increases the amount of digestible P covering the requirements of piglets for proper bone mineralisation. Furthermore, it was proved that the DXA method can be successfully applied to measure body fat and lean mass contents as well as bone mineralisation of growing pigs using the same animals.  相似文献

Two experiments were conducted to determine the content of digestible energy (DE) and metabolisable energy (ME) as well as the apparent ileal digestibility (AID) and standardised ileal digestibility (SID) of amino acids in peanut meal (PNM) for growing pigs. In Experiment 1, 78 growing pigs (46.8 ± 2.6 kg) were randomly allotted to 1 of 13 diets, including a corn–soya bean meal basal diet and 12 PNM test diets. In Experiment 2, 12 growing barrows (48.7 ± 2.8 kg) were allotted to one of two 6 × 6 Latin squares. The treatments include a N-free diet and 10 PNM test diets. The results of Experiment 1 showed that the DE and ME differed (p < 0.05) among the 12 PNM samples. On a dry matter basis, the DE and ME content ranged from 14.5 to 16.4 MJ/kg (mean 15.6 MJ/kg) and from 12.7 to 15.5 MJ/kg (mean 13.9 MJ/kg), respectively. The apparent total tract digestibility (ATTD) of gross energy (GE) was 82.2%. The DE of PNM could be precisely predicted by equations including NDF combined with GE or crude protein (CP) with an R2 value of 0.91 and 0.92, respectively. For the AID and SID for lysine, the results of Experiment 2 indicated variations among PNM sources ranging from 59.6% to 76.7% and 64.8% to 80.9%, respectively. However, for CP, variations for AID and SID were lower and ranged from 70.2% to 81.9% and 75.7% to 85.6%, respectively. The results indicate that the concentration of lysine was the best single predictor to estimate the digestibility of amino acids. However, further work is needed to investigate the reason for the variation in the digestibility of lysine and avoid processing procedures that are detrimental to lysine digestibility.  相似文献

Two experiments were conducted to estimate the digestibility of energy, nitrogen and amino acids (AA) in growing pigs fed diets containing one of five corn distillers' dried grains with solubles (DDGS), including three normal oil DDGS (NO-DDGS) and two low oil DDGS (LO-DDGS) samples. Exp. 1 was conducted to determine the digestible energy (DE) and metabolisable energy (ME) content. Six growing barrows (initial body weight [BW]: 35.1 +/- 2.2 kg) were allotted to a 6 x 6 Latin square design, with six periods and six diets. One diet was a corn soybean meal basal diet and the other five diets were based on corn, soybean meal and 28.8% DDGS. The average DE and ME values for the three NO-DDGS samples were 16.0 and 14.9 MJ/kg dry matter (DM). These values were 9 and 13% greater than the LO-DDGS values of 14.7 and 13.2 MJ/kg DM respectively. Exp. 2 was conducted to determine and compare apparent (AID) and standardised (SID) ileal digestibility for crude protein and AA in the five DDGS samples. Six growing barrows (initial BW, 32.2 +/- 1.9 kg) fitted with a simple T-cannula were allotted to a 6 x 6 Latin square design with six periods and six diets. Five of the diets were based on the five DDGS samples, and the remaining one diet was nitrogen-free diet based on cornstarch and sucrose. Titanium dioxide (0.1%) was used as inert marker. The results of the experiment showed the largest variation among the different samples in AID and SID for lysine (from 41.8 to 65.8% and 53.8 to 73.9% respectively) and threonine (from 54.3 to 73.8% and 65.2 to 79.5% respectively). Also, among the indispensable AA, the SID values for arginine, histidine, threonine and tryptophan observed in LO-DDGS were not different from the values derived from NO-DDGS. In conclusion, LO-DDGS may have decreased energy compared with NO-DDGS because of its lower fat content. However, oil removal during the production of DDGS may not affect amino acid digestibility.  相似文献

On day 28, 30 piglets were killed to measure concentrations and molar ratios of short-chain fatty acids in the hindgut. From day 0-14, digestibility of DM, N and GE was increased linearly and diarrhea frequency was decreased due to the increasing inclusion levels of extruded corn. From day 14-28, digestibility of GE and N was enhanced linearly due to the increasing proportion of extruded corn. On day 28, as proportion of extruded corn increased, the level of butyrate in the caecum and colon was increased linearly and the relative proportion of propionate was reduced. Results indicated that increasing proportions of extruded corn had no effect on growth performance, but significantly improved digestibility of N and energy at day 10 of trial and may be advantageous to the intestinal health of piglets.  相似文献

Crossbred pigs (n=720; average age=28±3 days and weight=9.5±0.3 kg) were used in a 20-day trial in order to determine the influence of phosphorus (P) source and various doses of pharmacological zinc (Zn) on growth performance, plasma minerals and mineral digestibility. Pigs (five intact males and five females per pen) were randomly allotted to treatments in a 3×3 factorial arrangement with three sources of dietary P (4.5 g/kg digestible P, 4.5 g/kg digestible P plus 2500 phytase units (FTU)/kg, or 5.5 g/kg digestible P) and three dietary levels of supplemental Zn (0, 1750 or 3500 mg/kg) from ZnO (82% Zn) with eight pens per treatment. Diets were formulated to exceed all nutrient requirements, including calcium (Ca), P and Zn from day 0 to 20. Zn supplementation increased (quadratic P<0.05) average daily feed intake. There was a significant Zn level×P source interaction on average daily gain and feed conversion ratio (FCR). Pigs fed 4.5 g/kg digestible P without or with 2500 FTU/kg phytase gained more per day (quadratic P<0.05) and had better FCR (quadratic P<0.05) when they were fed 1750 mg/kg supplemental Zn. However, pigs fed 5.5 g/kg digestible P gained more per day (linear P<0.05) and were more efficient (linear P<0.05) when they were fed 3500 mg/kg supplemental Zn. Plasma Zn and Zn digestibility increased (linear P<0.05) as pharmacological Zn supplementation increased from 0 to 3500 mg/kg, irrespective of P source. However, Ca, P, sodium (Na), potassium (K) and copper (Cu) digestibility were reduced (P<0.05) as pharmacological Zn supplementation increased, and this was mitigated or exacerbated by the supplementation of 5.5 g/kg digestible P or phytase. In conclusion, increasing the dietary inclusion of pharmacological Zn may impact growth performance in young pigs through the interaction with minerals such as Ca, P, Na and K. Pharmacological Zn may reduce Na or K digestibility and indirectly reduce water secretion into the lumen, resulting in an increase in faecal dry matter as pharmacological Zn supplementation in the diet increased.  相似文献

Today, weaner diets are optimised using digestibility coefficients obtained from grower-finisher pigs, which may overestimate the digestibility in weaners. The aim of this study was to evaluate the standardised ileal digestibility (SID) of CP and amino acids (AAs), and the intestinal morphology in pigs 0–4 weeks postweaning when fed different protein sources. The experiment included 128 pigs weaned at day 28 and the protein sources were wheat, soybean meal (SBM), enzyme-treated soybean meal (ESBM), hydrothermally treated rapeseed meal (HRSM) and casein. The experiment was conducted as a difference method study including wheat in all diets. Eight pigs were slaughtered on the day of weaning (day 0) and six pigs/treatment were slaughtered at days 7, 14, 21, and 28 postweaning. The SID of CP and AA, as average over the four weeks, was lowest for ESBM and highest for wheat and casein, with SBM and HRSM being intermediate. The SID of CP and AA increased (both linear and quadratic, P < 0.05) over time after weaning. The average SID of CP for all protein sources postweaning was 0.38, 0.59, 0.76, and 0.71 on days 7, 14, 21, and 28, respectively. These differences were significant (P < 0.05) between days 7 and 21, and between days 7 and 28 (P < 0.05), whereas there tended to be a difference between days 7 and 14 (P = 0.06). Protein source did not affect the small intestinal morphology response parameters, whereas time after weaning did. Villous height and villous height to crypt depth ratio differed (P < 0.05) between the days 0 and 7, with shorter villi and a higher ratio at day 7. Crypt depth was not altered between days 0 and 7, or between days 7 and 14. For villi density, crypt density and small intestinal length, a significant increase from days 7 to 14 was observed, but there was no further increase to or difference between days 21 and 28. In conclusion, the low SID of CP in casein on day 7 (0.50) illustrates the challenges related to protein digestion in weanling pigs. The SID of CP and AA is very low during the first two weeks postweaning and time after weaning is more important for protein digestibility, than the source of protein. Fewer mature epithelial cells and less absorptive area in the small intestine in the early postweaning period may partly explain the poor protein digestibility.  相似文献

Two experiments, a performance experiment and a mineral balance study, were conducted on grower-finisher pigs (42 to 101 kg live weight) to investigate the effects of Peniophora lycii phytase enzyme and 25-hydroxyvitamin D3 (25-OHD3) on growth performance, carcass characteristics, nutrient retention and excretion, and bone and blood parameters. The two experiments were designed as a 2 × 2 factorial (two levels of phytase and two levels of 25-OHD3). The four diets were T1, low-phosphorous diet; T2, T1 + phytase; T3, T1 + 25-OHD3 and T4, T1 + phytase + 25-OHD3 diet. In all, 25 μg of 25-OHD3 was used to replace 1000 IU of vitamin D3 in diets T3 and T4. Diets were pelleted (70°C) and formulated to contain similar concentrations of energy (13.8 MJ DE/kg), lysine (9.5 g/kg) and digestible phosphorus (P; 1.8 g/kg). Neither the inclusion of phytase nor 25-OHD3 in the diet had any effect on pig performance. There was an interaction between phytase and 25-OHD3 on calcium (Ca) and P retention (P < 0.01) and on the apparent digestibility of ash (P < 0.01), P (P < 0.001) and Ca (P < 0.001). Pigs offered phytase diets only, had a higher retention of Ca and P and digestibility of ash (P < 0.01), P (P < 0.001) and Ca (P < 0.01) compared with pigs offered unsupplemented diets. However, when the combination of phytase and 25-OHD3 were offered, no effects were detected compared with 25-OHD3 diets only. Pigs fed phytase diets had higher bone ash (P < 0.01), bone P (P < 0.01) and bone Ca (P < 0.05) concentrations compared with pigs offered non-phytase diets. In conclusion, pigs offered phytase diets had a significantly increased bone ash, Ca and P than pigs offered unsupplemented phytase diets. However, there was no advantage to offering a combination of phytase and 25-OHD3 on either bone strength or mineral status compared to offering these feed additives separately.  相似文献

Two experiments were conducted to evaluate the digestible (DE) and metabolisable energy (ME), apparent (AID) and standardised ileal digestibility (SID) of amino acids (AA) in yeast extract (YE) and spray-dried porcine plasma (SDPP). In Experiment 1, 18 barrows (25.1 ± 1.2 kg body weight [BW]) were randomly allotted to three treatments with six replicates per treatment. The DE and ME of YE was 20.64 and 19.31 MJ/kg, respectively, which were not significantly different with the DE and ME of SDPP (18.74 and 18.05 MJ/kg, respectively). In Experiment 2, six barrows (20.6 ± 2.6 kg BW) fitted with ileal T-cannulas were fed three diets in a repeated 3-period Latin square design. For Met and Glu, the AID tended to be, while the SID was significantly higher (< 0.05), in YE than in SDPP. The AID of Cys tended to be lower in YE (p = 0.07), while the SID of Phe tended to be higher in YE than in SDPP (p = 0.06). Accordingly, YE could be a potential substitute for SDPP as a superior protein ingredient in diets for pigs in terms of the available energy and AA digestibility.  相似文献

Around 70% of total seed phosphorus is represented by phytate which must be hydrolysed to be bioavailable in non-ruminant diets. The limited endogenous phytase activity in non-ruminant animals make it common practice to add an exogenous phytase source to most poultry and pig feeds. The mature grain phytase activity (MGPA) of cereal seeds provides a route for the seeds themselves to contribute to phytate digestion, but MGPA varies considerably between species and most varieties in current use make negligible contributions. Currently, all phytases used for feed supplementation and transgenic improvement of MGPA are derived from microbial enzymes belonging to the group of histidine acid phosphatases (HAP). Cereals contain HAP phytases, but the bulk of MGPA can be attributed to phytases belonging to a completely different group of phosphatases, the purple acid phosphatases (PAPhy). In recent years, increased MGPAs were achieved in cisgenic barley holding extra copies of barley PAPhy and in the wheat HIGHPHY mutant, where MGPA was increased to ~6200 FTU/kg. In the present study, the effect of replacing 33%, 66% and 100% of a standard wheat with HIGHPHY wheat was compared with a control diet with and without 500 FTU of supplemental phytase. Diets were compared by evaluating broiler performance, ileal Ca and P digestibility and tibia development, using nine replicate pens of four birds per diet over 3 weeks from hatch. There were no differences between treatments in any tibia or bird performance parameters, indicating the control diet did not contain sufficiently low levels of phosphorus to distinguish effect of phytase addition. However, in a comparison of the two wheats, the ileal Ca and P digestibility coefficients for the 100% HIGHPHY wheat diets are 22.9% and 35.6% higher, respectively, than for the control diet, indicating the wheat PAPhy is functional in the broiler digestive tract. Furthermore, 33% HIGHPHY replacement of conventional wheat, significantly improved Ca and P digestibility over the diet-supplemented exogenous phytase, probably due to the higher phytase activity in the HIGHPHY diet (1804 v. 1150 FTU). Full replacement by HIGHPHY gave 14.6% and 22.8% higher ileal digestibility coefficients for Ca and P, respectively, than for feed supplemented with exogenous HAP phytase at 500 FTU. This indicates that in planta wheat PAPhys has promising potential for improving P and mineral digestibility in animal feed.  相似文献

We have generated transgenic maize plants expressing Aspergillus phytase either alone or in combination with the iron-binding protein ferritin. Our aim was to produce grains with increased amounts of bioavailable iron in the endosperm. Maize seeds expressing recombinant phytase showed enzymatic activities of up to 3 IU per gram of seed. In flour paste prepared from these seeds, up to 95% of the endogenous phytic acid was degraded, with a concomitant increase in the amount of available phosphate. In seeds expressing ferritin in addition to phytase, the total iron content was significantly increased. To evaluate the impact of the recombinant proteins on iron absorption in the human gut, we used an in vitro digestion/Caco-2 cell model. We found that phytase in the maize seeds was associated with increased cellular iron uptake, and that the rate of iron uptake correlated with the level of phytase expression regardless of the total iron content of the seeds. We also investigated iron bioavailability under more complex meal conditions by adding ascorbic acid, which promotes iron uptake, to all samples. This resulted in a further increase in iron absorption, but the effects of phytase and ascorbic acid were not additive. We conclude that the expression of recombinant ferritin and phytase could help to increase iron availability and enhance the absorption of iron, particularly in cereal-based diets that lack other nutritional components.  相似文献

Abstract

A balance experiment was carried out to investigate the effects of betaine monohydrate (BET) or betaine derived from condensed molasses solubles (CMS) as a substitute for methionine and choline on nitrogen (N) balance and total tract nutrient digestibility in weaned piglets. The experiment included four treatments with 32 barrows with an average initial body weight (BW) of 13.5 kg. The supplementation of DL-methionine and choline (positive control = PC) to the basal diet, which was deficient in methionine and low in compatible osmolytes in the form of betaine or its precursor choline (negative control = NC) resulted in a significant increase in N retention of 0.8 g/d. The substitution of DL-methionine and choline with BET or CMS did not affect N retention compared to the PC and the NC treatment either. Feeding the PC diet increased the digestibilities of organic matter, NDF, ADF, NFE, crude ash, Ca, P, methionine, tryptophan and cystine by 1.9%, 7.3%, 9.7%, 1.1%, 6.3%, 13.9%, 7.7%, 15.9%, 4.3% and 2.8%, respectively, and tended (p < 0.20) to increase the digestibilities of most other amino acids by 1.6 – 3.4%. Digestibility of CP, EE (HCl), Mg and Na was 3.1% (p = 0.09), 5.1% (p = 0.09), 5.1% (p = 0.06) and 3.3% (p = 0.17) higher, respectively, when compared to the NC treatment. BET and CMS supplementation increased most nutrient digestibilities in the same magnitude as for the PC treatment. In summary, the supplementation of betaine, originating from different sources, to a diet with low contents of compatible osmolytes increased in particular the fermentation of fibre and enhanced mineral absorption. The supplementation of the NC with DL-methionine was more efficient in improving N retention than the replacement of DL-methionine by betaine originating from BET or CMS.  相似文献

Feeding high-protein diets in animals can lead to a decrease of nitrogen utilization efficiency, and then promote the environmental pollution. Recently, more reports have demonstrated that lowering protein level in diets supplemented with specific amino acids can address these problems. However, the whole proteome alteration in the skeletal muscle of weaned piglets fed low-protein diets is poorly understood. Here, we applied the isobaric tags for relative and absolute quantification approach to investigate this alteration. We fed weaned piglets with normal protein diet (20% crude protein) and low-protein diet supplemented with lysine, methionine, threonine, and tryptophan (17% crude protein) for 25 days. Then proteomic profiling of skeletal muscles was performed. In total, 1354 proteins were quantified in the porcine skeletal muscle proteome. 132 proteins were identified as differentially expressed proteins between the two groups. Differentially expressed proteins were significantly enriched in various nutrient metabolism including lipid, carbohydrate, and amino acid metabolism. Interestingly, a total of 20 differentially expressed proteins, which are involved in the oxidative phosphorylation pathway, were all down-regulated by the low-protein diet feeding. Further immunoblotting confirmed the down-regulations of MT-ATP8, COX2, NDUFA6, and SDHB, four selected proteins among these 20 proteins. Meanwhile, the ATP level in the low-protein diet group was also reduced. These findings for the first time reveal that oxidative phosphorylation pathway is suppressed in longissimus dorsi muscle of weaned piglets fed low-protein diet supplemented with limiting amino acids, which may provide new insights into further formula design and the choice of limiting amino acids in diets.  相似文献
