Sunflower necrosis virus disease (SNVD) was reported to cause a significant damage to sunflower production in India. Based on the biocontrol efficacy against SNVD observed in the previous greenhouse experiments, two plant growth promoting microbial consortia (PGPMCs) viz., PGPMC-1 consisting of Bacillus licheniformis strain MML2501 + Bacillus sp. strain MML2551 + Pseudomonas aeruginosa strain MML2212 + Streptomyces fradiae strain MML1042 and PGPMC-2 consisting of B. licheniformis MML2501 + Bacillus sp. MML2551 + P. aeruginosa MML2212 were used in the present study. Powder and liquid formulations of the above plant growth promoting microorganisms (PGPMs) were evaluated along with farmers’ practice (imidacloprid + mancozeb) and control in farmers’ fields. Significant disease reduction, increase of seed germination, plant height and yield parameters with an additional seed yield of 840 kg/ha, an additional income of Rs. 10,920/ha and benefit cost ratio of 6.1 were recorded following treatment with a powder formulation of PGPMC-1 compared to control. Further, liquid formulation of PGPMC-1 significantly reduced SNVD up to 51.4% compared to control. Besides, this treatment improved the seed germination (24.4%), plant height (61.3%), yield parameters with an additional seed yield of 936 kg/ha and an additional income of Rs. 12,168/ha. Benefit cost ratio was calculated as 6.8 in this treatment compared to 4.6 and 3.7, respectively for PGPMC-2 liquid formulation and farmers’ practice compared to control. This is the first report of PGPMCs mediated biological control of SNVD under field conditions.  相似文献

An efficient and improved method for in vitro propagation of mature tree of Dalbergia sissoo, an ecologically and commercially important timber yielding species, has been developed through axillary shoot proliferation. Bud breaking occurred from nodal shoot segments derived from rejuvenated shoots produced during early spring from a 20–25-year-old lopped tree, on MS medium containing 8.88 μM benzylaminopurine (BAP). Multiple shoots differentiated (20–21shoots/node) on re-culture of explants on half-strength agar gelled amended MS medium with a combination of 2.22 μM of BAP and 0.002 μM of thidiazuron (TDZ) with 1.0 mM each of Ca(NO3)2, K2SO4, KCl, and NH4(SO4)2. The maximum shoot multiplication (29–30 shoots/node) was achieved on subculturing in the above mentioned but liquid medium. Furthermore, the problem of shoot tip necrosis and defoliation observed on solid medium were overcome by the use of liquid medium. Ex vitro rooting was achieved on soilrite after basal treatment of microshoots with 984 μM of indole-3-butyric acid (IBA) for 2 min. About 90 % microshoots were rooted on soilrite within 2–3 weeks under the greenhouse conditions. From 20 nodal shoot segments, about 435 hardened plants were acclimatized and transplanted. This is the first report for rapid in vitro propagation of mature trees of D. sissoo on liquid medium followed by ex vitro rooting.  相似文献

The respiratory quotient (RQ) was found to be a suitable control parameter for optimum oxygen supply for the production of 2,3-butanediol + acetoin under microaerobic conditions. In laboratory scale continuous cultures optimum production of 2,3-butanediol + acetoin was obtained at an RQ value between 4.0 to 4.5. This agreed well with optimum RQ value (4.0) stoichiometrically derived from the bioreactions involved. In fed-batch cultures product concentrations as high as 102.9 g/L (96.0 g/L butanediol + 6.9 g/L acetoin) can be achieved within 32 h cultivation with an RQ control algorithm for oxygen supply. Under similar conditions only 85.7 g/L product (77.6 g/L butanediol + 8.1 g/L acetoin) was obtained with control of constant oxygen supply rate throughout the cultivation.In pilot scale batch cultures under identical oxygen supply rate the achievable RQ value was found to be strongly influenced by the reactor type and scale. The initial oxygen supply rate influenced the achievable RQ as well. However, in all the reactors studied the specific product formation rate of cells in the exponential growth phase was only a function of RQ. The same optimum RQ value as found in continuous cultures was obtained. It was thus concluded that RQ can be used as a control parameter for optimum production of 2,3-butanediol + acetoin in both laboratory and pilot plant scale reactors. In anticipation of large-scale distribution of a Turkish isolate of Puccinia jaceae var. solstitialis in California for biological control of yellow starthistle (Centaurea solstitialis, YST), susceptibility of YST within the state was determined and a protocol for bulk inoculum production was developed. Inoculation was made of 62 field accessions of YST representative of the range of habitats in California. These were determined to be equally susceptible to infection by the isolate approved for release in the United States in 2003. To support a program to speed establishment by release at many locations statewide, protocols for artificial increase of inoculum were developed. Over 64 g of urediniospores were produced with a mass-production system under greenhouse conditions from 2003 to 2006. Yield of inoculum varied by season, with peak production occurring from early spring through early summer. A large-scale urediniospore harvest also was made from a field plot at Davis, California. Our results show that susceptibility of YST in California is not likely to limit establishment of P. jaceae for biological control, and that production of this or other obligate pathogenic fungi (biological control agent) is possible for support of statewide release and research programs.  相似文献
