为了明确盐对种子发芽影响的渗透效应和离子效应共同作用方式以及量化种子发芽对盐的响应, 以两个大麦(Hordeum vulgare)品种‘Cask’和‘County’为研究对象, 设置4个恒定温度(5、12、20和27 ℃)、5个等渗的NaCl和聚乙二醇(PEG)浓度梯度(-0.45、-0.88、-1.32、-1.76和-2.20 MPa, 蒸馏水作对照), 做常规发芽实验。结果显示: (1)两个品种在NaCl溶液中比在等渗的PEG溶液中发芽率高且发芽速度快; (2) NaCl和PEG分别作为渗透剂计算出的水势模型参数值差异很大, 说明水势模型不能用来描述种子发芽对盐的响应; (3)大麦种子在盐溶液中的发芽速率与盐浓度成显著的负相关直线关系, 因此我们修订了水势模型, 将修订后的模型命名为盐度模型, 用来量化盐对大麦种子发芽的影响。与水势模型计算出的发芽时间相比, 盐度模型计算出的50%种子发芽时间与大麦种子实际发芽时间更接近; (4)大麦种子在等渗的NaCl和PEG溶液中发芽速率差异随着水势降低, 先增加后降低。据此我们提出盐的渗透效应和离子效应共同作用于种子发芽的3种情况: 第一种在低盐条件下, 主要是渗透效应起负作用; 第二种情况在中盐条件下, 渗透效应和离子效应共同起作用, 离子效用的正作用强于渗透效应的负作用; 第三种情况在高盐条件下, 离子效应逐渐开始起离子毒害的负作用。  相似文献

Seeds of the crisp lettuce cultivar Pennlake were germinated using all combinations of six ‘initial’ solutions of polyethylene glycol 6000 (PEG) with osmotic potentials ranging from 0 to -8 bars and seven ‘secondary’ solutions of PEG with osmotic potentials ranging from 0 to -10 bars, to which seeds were moved after 24 or 48 h in the ‘initial’ solution. The number of seeds germinating decreased at more negative osmotic potentials of both ‘initial’ and ‘secondary’ solutions but there was an interaction between germination temperature and the osmotic potential of the ‘initial’ solution. At an ‘initial’ solution osmotic potential of 0 bars germination at 20°C exceeded that at 10°C. As the osmotic potential of the ‘initial’ solution decreased germination at 20°C decreased more than at 10°C so that at the more negative osmotic potentials germination at 10°C exceeded that at 20°C. However seeds ungerminated after 14 days germinated normally when transferred back to water, so that the average final germination was 99.5%. The results suggest that major fluctuations in soil water potential in a seedbed are unlikely to influence seed germination per se provided that a period of 24 to 48 h at 0 bars tension is available at some time. The timing of such a period relative to sowing will have a considerable effect on the time of germination and hence the time of emergence. It is concluded that factors other than the direct effect of soil moisture content on germination are involved in reducing seedling emergence under fluctuating soil moisture conditions in the field.  相似文献

研究不同浓度(0、1、2、3、6、9、14、18g.L-1)的盐溶液(NaCl、MgSO4、盐渍土壤)对河西走廊中部荒漠边缘的黑果枸杞种子吸胀、萌发和幼苗生长的影响,并观察胁迫解除后种子的反应。结果表明:黑果枸杞种子吸胀速率随NaCl、MgSO4和土壤溶液浓度的增大呈先升后降的趋势,吸水速度随胁迫时间的延长而减慢;种子萌发率随3种盐浓度的增大而降低,盐胁迫解除后种子仍具有较高的萌发率;发芽指数、活力指数、根长、下胚轴随3种盐浓度的增大而降低或先升后降,根轴比随盐胁迫的增强先升后降;随3种盐浓度的增大,种苗损害率增大,3种盐的胁迫效应依次NaCl>MgSO4>盐渍土壤溶液。黑果枸杞种子萌发和幼苗生长对NaCl胁迫较为敏感,其耐受的临界阈值是6g.L-1;种子萌发能耐受较高浓度的MgSO4的胁迫,幼苗生长对MgSO4胁迫较敏感,其耐受的临界阈值是9g.L-1;种子萌发和幼苗生长对生境盐渍土壤具有较强的耐受能力和适应性。  相似文献

A comparison of different chemicals for osmotic treatment of vegetable seed   总被引:2,自引:0,他引:2  
Samples of carrot, celery, leek and onion seed were treated before sowing by imbibition in osmotic solutions of polyethylene glycol 6000 (PEG), glycerol and potassium dihydrogen orthophosphate (KH2PO4). The solutions were sufficiently concentrated to prevent germination during treatment, and within each species, the amount of water taken up by the seeds during treatment did not vary greatly between solutions. All treatments increased the rates of seed germination and seedling emergence compared to untreated controls, but glycerol-treated seeds germinated and emerged significantly more slowly than did PEG- and KH2PO4-treated seeds. The effects of PEG and glycerol treatments on percentage germination and seedling emergence were small, but KH2PO4 treatment tended to reduce percentage germination and emergence, most markedly in leek and one cultivar of celery. It is concluded that PEG treatment gives the most consistently beneficial effects for the range of species tested.  相似文献

Salinization is one of the most important factors affecting agricultural land in the world. Salinization occurs naturally in arid and semiarid regions where evaporation is higher than rainfall. Sugar beet yield declines with an increase in salinity, but the sensitivity to salts varies with salt composition in water and sugar beet growth stage. The aim of this study was to determine the effect of water salinity levels and salt composition on germination and seedling root length of four sugar beet cultivars (PP22, IC2, PP36, and 7233). The experiments were undertaken with irrigation water with two salt compositions (NaCl alone and mixture of MgSO4 + NaCl + Na2SO4 + CaCl2) in three replicates. Thirteen salinity levels with electrical conductivity (EC) of the irrigation water ranging from 0 to 30 dS/m were applied to each cultivar in both experiments. Seed germination percentage and seedling root length growth were determined in 13 days. Statistical analysis revealed that germination and root length were significantly affected by salt composition, cultivars and salinity levels. Regardless of salt composition, seed germination and seedling root length were significantly affected by the irrigation water with EC up to 8 dS/m and 4 dS/m, respectively. Except for cultivar PP22, the adverse effect of salinity of the irrigation water on seed germination and seedling root length was higher for NaCl alone than for the salt mixture, which refers to lower salt stress in field conditions with natural salt composition. 角果碱蓬(Suaeda corniculata)是藜科一年生盐生植物, 在我国分布于北方盐碱滩涂和盐碱荒漠地区。角果碱蓬具有棕色和黑色两种异型体种子(简称棕色和黑色种子)。对采自内蒙古鄂托克前旗盐渍化生境的角果碱蓬二型种子的形态、休眠和萌发特性开展对比研究, 测定了二型种子休眠和萌发行为对温度、光照和盐分(NaCl)的响应, 以揭示盐生植物异型种子对温带盐漠生境的适应对策。结果表明: (1)二型性种子在大小、种皮特性和结实比例方面有显著差异。与黑色种子相比, 棕色种子个体较大, 种皮透水性强。黑色种子与棕色种子的结实比例约为5.6 : 1。(2)新成熟的棕色种子的萌发对各温度梯度和光照条件不敏感, 萌发率较高(84%-100%); 而新成熟的黑色种子萌发率较低(8%-78%), 萌发对光照敏感。(3)黑色种子具有浅度生理休眠, 种皮划破、赤霉素处理和低温层积均可有效地提高种子的萌发率。(4)二型种子萌发对土壤盐分的胁迫具有不同的响应。与黑色种子相比, 棕色种子对盐分胁迫不敏感, 在较高的盐分浓度下仍有较高的萌发率, 低温层积处理能够降低黑色种子对盐胁迫的敏感性, 有效地提高种子的初始萌发率、萌发恢复率和最终萌发率。角果碱蓬二型种子不同的形态、休眠和萌发特性, 提高了该物种在高度异质性生境中的适合度, 对种群成功地适应温带盐漠环境具有重要的意义。  相似文献

Soil moisture and temperature, sowing depth and penetration resistance affect the time and percentage of seedling emergence, which are crucial for the simulation of drought‐limited crop production. The aim of this research was to measure the effect of soil water potential on germination and emergence, shoot and root elongation rates (SER and RER) of two different seed/crop types. Sugar beet and durum wheat seeds were sown into two soils (clay and loam), submitted to five matric potentials (?0.01, ?0.1, ?0.2, ?0.4 and ?0.8 MPa) and incubated at constant temperature (25°C) and humidity. Cumulative count analysis was used to estimate parameters of the distribution of germination or emergence times for each box of beet or wheat seeds and to derive estimates for base potentials (ψb), hydrothermal times (H) and numbers of viable units. In a second experiment, NaCl solution was used to mimic the soil matric potentials to estimate potential RER and SER. Germination of sugar beet was slightly more sensitive to matric potential than durum wheat (ψb of ?1.13 and ?1.23 MPa, respectively). H(g) was longer for sugar beet than for durum wheat (67 vs 47 MPa °Cd). For emergence ψb was similar for both seed types and soils but hydrothermal times (H(e)) were 40 MPa °Cd higher for sugar beet than for wheat. Emergence was about 20 MPa °Cd earlier in loam than in clay. SER measured in soils were similar for both crops and for durum wheat it agreed with those determined in NaCl solution. RER and SER fell with decreasing osmotic potential to approximately 20% of their maximum values (1.03 mm h?1 and 0.57 mm h?1, respectively). Seedling viability decreased with decreasing matric potential and more in clay than in loam soil and more for sugar beet than durum wheat. Seed and soil aggregate size are discussed with respect to the effects of water diffusion and soil–seed contact on germination and emergence modelling.  相似文献

Germination inhibitors in methanol and water extracts of redbeet fruits (Beta vulgaris cv. rubra L.) have been studied usinglettuce and red beet seed germination as bioassays. The methanolextracts contained substances which inhibited lettuce seed germination,but had no effect on the germination of red beet seeds. Germinationof both lettuce seeds and of water-leached or sulphuric acid-treatedred beet seed balls were inhibited by the water extracts. Theconcentrations of ammonia, ferulic acid, and oxalate in thewater extracts were much lower than required for inhibitionof red beet seed germination. The water extracts contained,however, large amounts of inorganic ions, and the results clearlydemonstrated that the inhibitory effect of the water extractson red beet seed germination was mainly due to the content ofsuch inorganic ions.  相似文献

The nature and importance of endogenous germination inhibitorsin rubbed triploid monogerm sugar beet fruits were studied.Experimental evidence suggests that germination inhibitors donot play an important role in the prevention of germinationof rubbed fruits. Putative inhibitory compounds known to occurin sugar beet extracts were found to have little effect on germination.Measures to leach out water soluble compounds also had littleeffect on germination. Concentrated fruit extracts were foundto inhibit the germination of sugar beet seeds by virtue oftheir osmotic potentials, and this was related to the salt contentof the fruit extracts. However, a positive relationship wasfound between salt content and the original germination of thefruit lines. This relationship could have important consequencesfor assessment of fruit maturity and subsequent germinationbehaviour at harvest. The causes of poor germination in rubbedfruit lie not in the presence of inhibitory compounds in thefruit coat but in some other property of the fruit. Two seed lots of Calluna vulgaris were obtainedfrom English Nature (seed of Cornish provenance) (EN) and John ChambersWildflower Seeds (JCWS). In laboratory tests, under continuous light untreatedseeds of both seed lots were partially dormant at temperatures between14–35 °C, but JCWS seeds were more deeply dormant thanENseeds. The optimum temperature for germination for both lots was ca 18°C. Germination of EN seeds was much lower in the dark than inthe light at all temperatures; JCWS seeds did not germinate in the dark. In thelight at 22 °C, dormancy of both seed lots was broken whenseeds were incubated in GA4/7 solution(2×10–4 M). Dormancy ofJCWSseeds at 22 °C in the light was broken when seeds wereincubated in four different smoke solutions but more so when used incombinationwith GA4/7. Soaking seeds for 4h insmoke/GA4/7solutions before sowing improved both the speed andpercentage germination in pot experiments on a mist bench in the glasshouse byat least 10-fold. Soaking with GA4/7 alone produced a 5-fold increasein germination but seedlings were more etiolated than with thesmoke/GA4/7 mixtures. A seed advancement treatment modified from thatused commercially on sugar beet seeds also promoted germination in bothlaboratory and glasshouse tests. This entailed soaking seeds in 0.2% thiramsuspension for 4h followed by incubation in excess solution at 22°C for 4 days. This treatment was not as effective as thesmoke/GA4/7 seed soaks.  相似文献

The effects of different osmotic stresses (from 0 to –8× 105 Pa) obtained with NaC or polyethylene glycol 6000 solutions on the germination of flax, sesame and onion seeds were investigated. The effect of presoaking with gibberellic acid (GA3) on the germination of the above mentioned seeds was also studied. It was found that the rate of seed germination and the final germination percentages as well as the amount of water absorbed by the seeds were considerably lowered with the rise of osmotic stress levels whatever the stress agent used, more considerable reduction was obtained under polyethylene glycol 6000 than under NaCl. Presoaking with gibberellic acid increased the rate and the final germination percentage of osmotically stressed flax and sesame seeds, while those of stressed onion seeds were slightly retarded.  相似文献

根据乌拉尔甘草种子吸胀时间的长短将种子硬实程度分为H0、H1、H2、H3、H4和Hmax6个等级,测定其活力指标;同时于盐胁迫和水分胁迫下测定硬实和非硬实种子的发芽率、发芽指数、胚根长度及幼苗的抗氧化酶活性。结果表明:随着硬实程度的增大,种子的发芽率、发芽指数、胚根长度、脱氢酶活性和ATP含量均呈上升趋势,丙二醛(MDA)含量则表现出下降趋势;在逆境条件下,硬实种子比非硬实种子的适应能力强。硬实种子的活力高于非硬实种子,硬实程度与种子活力呈正相关。  相似文献

The response to drought stress on germination was investigated on three hybrids of ornamental sunflower, ‘Hadar’, ‘Pazit’, and ‘Zohar’. Different levels of water potential [Ψ: 0.0 (control), ?0.15, ?0.30, ?0.45, ?0.60, ?0.75, and ?0.90 MPa] were adopted using polyethylene glycol-6000 (PEG6000) at four germination temperatures (15, 20, 25, and 30 °C). Final germination percentage, mean germination time, germination index, germination rate index, and germination stress tolerance index were used to evaluate the genotype response to PEG-induced water stress. Shoot and root length and fresh and dry weight were measured on seeds germinated at 20 °C under the different levels of water potentials. During germination, the three ornamental sunflowers showed to be more sensitive to suboptimal temperature than to supraoptimal. Decreasing water potential of imbibition solution progressively inhibited and delayed seed germination. Among cultivars, ‘Hadar’ and ‘Pazit’ performed better at temperature lower than 30 °C. ‘Zohar’ showed a lower sensitivity to PEG-induced water stress at all temperature conditions. Water stress during seed germination depressed the following seedling growth under favourable conditions. As a result, shoot and root length and fresh and dry weight was significantly lower in seedlings from seed germinated at ψ ≤ 0.45 MPa.  相似文献

Seed priming (controlled imbibition) is a widely used technique for improving crop establishment, because it allows a reduction of the time to radicle emergence following seed imbibition and synchronization of individual seeds within seed lots with respect to germination timing. The major problem encountered in seed priming is the control of seed imbibition to a level permitting pre-germinative processes to proceed but that blocks radicle emergence. If not, the consequence of drying back the seeds to initial moisture content for storage purposes could be a total loss of the treated batch. This is because, as long as radicle growth has not begun, seeds may be re-dried without any permanent deleterious effects upon subsequent germination or growth. Recently, we reported the discovery of a molecular marker of sugar beet seed priming, corresponding to the basic B-subunit of the seed storage protein 11S globulin. An ELISA based upon this molecular marker has been used to analyse how different sugar beet seed lots respond to a priming treatment. The results demonstrate that this ELISA allows us to readily distinguish between the primed seeds and the corresponding untreated seeds.  相似文献

Excessive water in the substratum inhibited germination of monogerm sugar beet seed but it was not lethal, and other crop seeds tested were not as sensitive as beet. The inhibition was not evident in a pure oxygen atmosphere and was lessened when the fruit cap was removed. Although bacterial populations on fruits absorbed appreciable quantities of oxygen, differences between populations in optimum and excessive water were insufficient to account for the inhibition. Anatomical observations revealed the presence of a pore through the sclerified fruit wall at the point of attachment to the mother plant. Evidence indicated that the pore was the route for oxygen and water uptake to the seed, and theoretical calculations of gas flux suggested that oxygen supply would be insufficient to sustain germination when the pore was filled with water.  相似文献

Low soil temperatures and low water potentials reduce and delay the seed germination of canola (Brassica rapa L., B. napus L.) in western Canada. Germination is also very sensitive to the salinity effects of nitrogen fertiliser placed with the seed, especially when the seed bed is relatively dry. The effects of pre-hydration and re-drying treatment on canola (Brassica rapa L. cv. Tobin) seed germination and seedling emergence at 10°C subjected to either a water or salt stress were determined. Low water potentials, induced by polyethylene glycol (PEG 8000), low soil moisture, or high concentrations of salts, reduced both germination and seedling emergence, and increased the time to 50% germination and emergence of seeds at 10°C. At equal osmotic potentials, Na2SO4 was less inhibitory on low temperature germination than either NaCl or PEG, suggesting that the sulphate ion partially alleviated the inhibitory effects of low water potential. Solutions of NaCI produced more abnormal seedlings compared to Na2SO4, suggesting that NaCl was more toxic than Na2SO4 during seedling development. Pre-hydration and re-drying partially overcame the inhibitory effects of both low water potential and salts on seed germination and seedling emergence at 10°C. The seed treatment increased the germination rate in Petri dishes and seedling emergence from a sandy loam soil. Water potentials or soil water contents required to inhibit 50% germination or emergence at 10°C were lower for treated seeds compared to control seeds. Salt concentrations inhibiting 50% emergence were higher for treated seeds than control seeds. Neither treated nor control seeds produced seedlings which emerged if the soil water content was lower than 9% or when the soil was continuously irrigated with salt solutions of 100 mmol kg-1 of NaCl or 50 mmol kg-1 of Na2SO4. These results suggest that the pre-hydration and re-drying treatment did not lower the base water potentials at which seedling emergence could occur. Abnormal seedlings were observed in both treated and control seeds, particularly if the soil was watered with NaCl solutions; however, the seed treatment reduced the number of abnormal seedlings.  相似文献

采用砂培方式,研究了外源5-氨基乙酰丙酸(ALA)对盐胁迫下菘蓝种子的萌发、幼苗叶片的可溶性糖含量、丙二醛(MDA)含量及其抗氧化酶活性的影响,探讨ALA缓解菘蓝受盐胁迫伤害的响应机制。结果显示:(1)菘蓝种子萌发及幼苗生长在100 mmol·L-1 NaCl胁迫下受到明显的抑制,种子发芽率、发芽势、发芽指数、活力指数与自然含水量均显著降低,丙二醛含量、可溶性糖含量以及超氧化物歧化酶(SOD)、过氧化物酶(POD)活性显著升高。(2)盐胁迫下适宜浓度的ALA处理显著提高了种子萌发率、自然含水量及SOD、POD和CAT活性,降低了可溶性糖和丙二醛的含量,并以16.7 mg·L-1 ALA处理盐胁迫下菘蓝种子的发芽率、发芽势最大,其幼苗的SOD、POD、CAT活性最强。研究表明,盐胁迫显著抑制菘蓝种子的萌发及幼苗生长,适宜浓度的ALA能够有效缓解盐胁迫对菘蓝种子萌发及幼苗生长的伤害,提高植株的抗盐性,并以16.7 mg·L-1 ALA处理效果最佳。  相似文献

对34种作物1.4万多份的国家库贮存种子进行了生活力监测,结果表明,贮存20年后,92.9%被监测种子的发芽率仍保持在85%以上,但有155份种子(占被监测份数的1.1%)出现了明显的下降(发芽率从80%以上降至70%以下)。发芽率显著下降的作物包括蚕豆、红小豆、黄麻、蓖麻、甜菜、西瓜、烟草、牧草。小麦等8种作物2078份种子的田间出苗率调查表明,所有种质均有出苗,但有8份种质的出苗率低于10%。出苗率与入库初始平均发芽率存在差别,平均出苗率最高的作物为普通菜豆86.2%,比该批入库初始发芽率平均值低9.3%;平均出苗率最低作物为谷子39.2%,比该批种子的入库初始发芽率平均值低51.3%。总体而言,国家库内保存的作物种子中,多数可安全保存20年以上,尤其是水稻等禾谷类作物种子,但对于蚕豆、红小豆等平均监测发芽率出现显著下降的作物种子需要增加监测频率,以确保种子的长期安全保存。  相似文献

Seeds of pokeweed (Phytolacca americana L.), an underinvestigated species, were studied to determine optimal conditions for laboratory germination. Soaking seeds in acid solutions prior to sowing increased rates of germination. Concentrated (conc) H2SO4 was more effective than conc NO3 or conc HCl acids. Physiological evidence from seed germination studies suggests that autotoxicity, or intraspecific inhibition, exists in pokeweed, a species known to possess several biologically active compounds. Seed germination was investigated in the laboratory with aqueous extracts of vegetative and reproductive structures of the plant. The presence of extracts from most plant parts correlated with reduced or no germination by seeds of its own species, whereas the presence of distilled water correlated with high percentages of germination by control seeds. Whether diluted with water by 5-fold (20% v/v) or undiluted, juice of pokeweed fruits completely inhibited the laboratory germination of pokeweed seeds. Also, extracts of freshly harvested mature leaves, stems, and immature fruits inhibited seed germination. However, results with root extracts, obtained from a single individual, depended more on concentration, since the highest concentration (50% v/v) inhibited germination, and lower concentrations (10 and 20% v/v) increased germination percentages over control samples. Results with extracts of juvenile leaves correlated with neither inhibition nor promotion of germination. Thus, except for juvenile leaves and the root, most extracts of the pokeweed plant inhibited seed germination with more mature structures exerting more inhibition and less mature structures exerting less or no inhibition. The ecological implication of autotoxicity is that seeds are more dispersed through time and space. In regard to seed germination by other species, especially those taxa known to possess biologically active compounds, these and other results suggest that the phenomenon of autotoxicity might be more widespread than previously suspected.  相似文献

恶劣环境下,人工海防林因面临养分胁迫而经营困难。为探讨盐、磷胁迫对主要海防林树种木麻黄和台湾相思种子萌发及生长的影响,该研究分别用不同浓度的NaCl(盐)和KH2PO4(磷)溶液处理种子和浇灌幼苗,测定种子萌发和幼苗生长指标。结果表明:(1)高盐胁迫显著抑制种子萌发,对幼苗生长有一定影响,但两种植物影响程度不同;台湾相思种子萌发耐盐性高于木麻黄,前者相对盐害率最大值为23.03%,后者为89.15%;随着盐浓度增加,木麻黄和台湾相思种子的发芽率、发芽势、发芽指数和活力指数均降低,对应最大值分别为38.70%、34.67%、18.70、0.055和76.67%、62.22%、48.46、6.11。(2)两种植物的株高和根长随盐浓度增加而降低,木麻黄和台湾相思株高分别为12.29~6.01 mm和48.27~17.33 mm,根长分别为8.57~1.45 mm和33.41~5.88 mm;台湾相思根、茎、叶生物量及根冠比均随盐浓度的增加逐渐减小,木麻黄各处理差异较小。(3)台湾相思的种子和幼苗较木麻黄更耐低磷环境,二者最适磷浓度存在差异;木麻黄种...  相似文献
