PTN作为重要的细胞因子,存在于多种生物体内,参与多种生物学过程,具有刺激细胞增殖、分化、粘附、迁移等多种功能。根据PTN的基因序列及大肠杆菌密码子的偏爱性,人工合成ptn基因,构建pMAL-his-sumo-ptn重组表达载体,将重组表达载体转化到E.coli(Rosetta)中并进行优化表达。融合蛋白在0.4 mmol/L IPTG、37℃条件下诱导3 h表达量最高。收集菌体、纯化、酶切、SDS-PAGE可检测到分子量为18 kD左右的重组蛋白。  相似文献

摘要 目的:探讨不明原因复发性流产(URSA)再次妊娠早孕期患者平均红细胞体积(MCV)、人绒毛膜促性腺激素(HCG)、γ-干扰素(IFN-γ)/白介素-10(IL-10)比值与妊娠结局的关系。方法:选择2019年9月~2021年5月期间兰州大学第二医院生殖医学科收治的105例URSA再次妊娠早孕期患者作为研究组,将研究组根据妊娠结局分为妊娠成功组(成功分娩,n=71)和妊娠失败组(再次流产,n=34)。选取同期来兰州大学第二医院生殖医学科产检健康的80例孕妇作为对照组。对比研究组、对照组的MCV、HCG、IFN-γ/IL-10比值,多因素Logistic回归分析妊娠结局的影响因素,采用受试者工作特征(ROC)曲线评估MCV、HCG、IFN-γ/IL-10比值单独及联合检测URSA再次妊娠早孕期患者妊娠结局的预测效能。结果:研究组的MCV、IFN-γ/IL-10比值大于对照组,HCG小于对照组(P<0.05)。URSA再次妊娠早孕期患者妊娠结局与年龄、就诊时孕周、胰岛素抵抗指数(HOMA-IR)、孕早期宫腔积液、孕前调理、内分泌异常、抗精子抗体(ASA)阳性、抗心磷脂抗体(ACA)阳性、宫颈机能不全、MCV、HCG、IFN-γ/IL-10比值有关(P<0.05)。多因素Logistic回归分析显示,年龄偏大、就诊时孕周较长、高MCV、低HCG、高IFN-γ/IL-10比值和URSA再次妊娠早孕期患者妊娠不良结局密切关联(P<0.05)。MCV、HCG、IFN-γ/IL-10比值联合检测对URSA再次妊娠早孕期患者妊娠结局预测的效能较高。结论:MCV、HCG、IFN-γ/IL-10比值在URSA再次妊娠早孕期患者妊娠结局具有一定的预测价值,且联合检测效果更好。此外,年龄偏大、就诊时孕周较长、高MCV、低HCG、高IFN-γ/IL-10比值为URSA再次妊娠早孕期患者妊娠结局的独立危险因素。  相似文献

奶牛和山羊的卵母细胞经过体外成熟、体外受精和体外发育至桑椹期,在显微操作下,吸取4~6个胚胎细胞,应用巢式PCR技术对其DNA进行SRY基因的测定以进行胚胎性别鉴定。共有27枚转有外源基因的试管牛胚胎和207枚转基因试管羊胚胎进行了SRY基因检测。选取10枚经过性别鉴定的牛胚胎移植于8头受体母牛和124枚已知性别的羊胚胎移植于44头受体母羊,移植后妊娠率分别为37.5%(牛,3/8)和61.4%(羊,27/44)。最后产生1头分娩牛犊和2头流产胎牛以及14头分娩羊羔和2头流产胎羊,它们的性别与胚胎性别鉴定的结果完全相符。 Abstract: The oocytes of bovines and goats were subjected to in vitro maturation,in vitro fertilization(IVF)and in vitro development upto morula stage.4~6 embryonic cells were aspirated with micro-manipulating and the embryo sexes were identified by detecting SRY gene with nested PCR procedure.A total of 27 transgenic IVF bovine and 207 transgenic IVF goat embryos were performed SRY gene detection.10 bovine and 124 goat sexed embryos were selected to be transferred into 8 bovine and 44 goat recipients,respectively,leading to the pregnant rates of 37.5%( in bovine,3/8)and 61.4%(in goat,27/44).In the end,1 cattle and 14 goatlets were born at term but 2 fetal bovine and 2 fetal goats were miscarried.Their sexes were fully in accordance with the embryonic sex predetermination.  相似文献

An in vitro fertilization and culture system was used to determine the effect of multiple nuclear vacuoles in bovine spermatozoa on fertilization and early embryonic development. After swim-up, semen parameters were similar between 2 bulls except that 60% of spermatozoa from bull A contained multiple nuclear vacuoles, whereas no spermatozoa from bull B (control) contained vacuoles. In Experiment 1, in vitro–matured (IVM) oocytes were inseminated with frozen-thawed semen from the 2 bulls to determine the ability of vacuolated sperm to bind with the zona pellucida. The mean number of spermatozoa bound to the zona pellucida was less (P< 0.05) for bull A (85.7 ± 5.7; n = 112) than for bull B (108.9 ± 5.4; n = 130). In Experiment 2, the percentages of zonae penetrated by spermatozoa from bull A (151 of 201; 75%) and bull B (116 of 150; 77%) were not different. However, the percentages of vacuolated spermatozoa from bull A bound to (43%) and penetrating the zona pellucida (34%) were lower than those in the inseminate (60%). In Experiment 3, fertilization rates, as evidenced by the presence of two pronuclei, were not different for bull A (101 of 136; 74%) and bull B (89 of 115; 77%). In Experiment 4, there was no significant difference in percentage cleavage (72.1% versus 76%) and morulae (29.2% versus 34.8%) or blastocyst production (7.2% versus 8.4 %) for bulls A and B, respectively. Data suggest that spermatozoa with multiple nuclear vacuoles are defective in zona binding. However, vacuolated spermatozoa gaining access to the ooplasm apparantly participate in fertilization and early embryonic development. Mol. Reprod. Gap junctional coupling between cumulus cells is required for oocytes to reach developmental competence. Multiple connexins, which form these gap junctions, have been found within the ovarian follicles of several species including bovine. The aim of this study was to determine the role of connexin 43 (CX43) and its relationship to embryo development, after in vitro fertilization (IVF). Cumulus?oocyte complexes (COCs) were obtained from abattoir sourced, mixed breed, bovine ovaries. COCs were isolated from follicles ranging from 2 to 5 mm in size, representing the preselected follicle pool. Immediately after isolation, two cumulus cell biopsies were collected and stored for analysis pending determination of developmental outcomes. Using in vitro procedures, COCs were individually matured, fertilized, and cultured to the blastocyst stage. Biopsies were grouped as originating from COCs that arrested at the two‐cell stage (low developmental competence [LDC]) or having developed to the late morula/blastocyst stage (high developmental competence [HDC]), after IVF and embryo culture. The expression level of CX43 was found to be significantly higher in cumulus cells from COCs that had an HDC when compared with those that had an LDC. Moreover, the gap junctional intercellular coupling rate was significantly higher in cumulus from COCs deemed to have an HDC. Significantly higher expression of the cumulus health markers luteinizing hormone receptor and cytochrome p450 19A1 was found in the cumulus originating from oocytes with HDC, suggesting that this system may provide a mechanism for noninvasively testing for oocyte health in preselected bovine follicles.  相似文献

Summary

A study of sperm penetration of the egg of the house cricket, Acheta domesticus, using a fluorescent microscope technique, showed that sperm penetration of the chorion, prior to fertilization, is not restricted to a specialised area of the egg surface, such as the micropyle. An acrosomal filament is seen on penetrating sperm. Polyspermy (multiple sperm attachment) is seen under normal conditions.

Eggs fertilized in vitro developed to the 4 day (pre-katatrepsis) stage, but did not undergo katatrepsis. Development was confirmed by cytogenetic studies. The percentage of eggs showing cleavage nuclei (i.e. initial development) was 59% after in vitro fertilization and 5% in ovarian eggs incubated in a hypotonic medium.  相似文献

研究卵泡内环境对猪卵母细胞体外成熟、受精及受精卵体外发育的影响。主要结果如下:直径≥5mm、4-4.9mm、3-3.9mm和2-2.9mm的卵泡卵母细胞体外成熟率分别为90.5%、89.7%、85.4%和67.4%,体外受精后,卵母细胞的发育能力随卵泡直径的增大而增强,直径≥5mm和4-4.9mm卵泡卵的2-细胞、3-4-细胞发育率显著高于直径2-2.9mm的卵泡卵(P<0.05或0.01)。体外成熟培养36h、42h和48h,直径2-2.9mm卵泡卵的体外成熟率,体外受精后的卵裂率差异不显著(P>0.05)。在体外成熟培养液中添加5%或15%的不同直径卵泡的卵泡液,各组间卵母细胞的体外成熟率,受精卵的体外发育率均无显著差异,结果表明:卵泡大小对猪卵母细胞体外成熟、受精及受精卵体外发育有重要影响。  相似文献

A retrospective case control study was undertaken at the molecular biology department of a private center for reproductive medicine in order to determine whether any correlation exists between mitochondrial DNA (mtDNA) content of cleavage-stage preimplantation embryos and their developmental potential. A total of 69 couples underwent IVF treatment (averaged women age: 36.5, SD 4.9) and produced a total of 314 embryos. A single blastomere was biopsied from each embryo at the cleavage stage (day-3 post-fertilization) subjected to low-pass next generation sequencing (NGS), for the purpose of detecting aneuploidy. For each sample, the number of mtDNA reads obtained after analysis using NGS was divided by the number of reads attributable to the nuclear genome. The mtDNA copy number amount was found to be higher in aneuploid embryos than in those that were euploid (mean mtDNA ratio ± SD: 6.3 ± 7.5 versus 7.1 ± 5.8, p < 0.004; U Mann–Whitney test), whereas no statistically significant differences in mtDNA content were seen in relation to embryo morphology (6.6 ± 4.8 vs. 8.5 ± 13.6, p 0.09), sex (6.6 ± 4.1 vs. 6.2 ± 6.8, p 0.16), maternal age (6.9 ± 7.8 vs. 6.7 ± 4.5, p 0.14) or its ability to implant (7.4 ± 6.6 vs. 5.1 ± 4.6, p 0.18). The mtDNA content cannot serve as a useful biomarker at this point in development. However, further studies investigating both quantitative and qualitative aspects of mtDNA are still required to fully evaluate the relationship between mitochondrial DNA and human reproduction.  相似文献

目的 探讨不同品系小鼠的体外受精、胚胎和精子的低温保存效果。方法 本实验分别在中国科学院上海实验动物中心 (SLAC)和日本熊本大学动物资源开发中心 (CARD)对 13个品系小鼠 (C57BL 6J、BALB c、C3H HeJ、ICR、KM、FVB、MRL、NOD、CBA、DBA 2、CD 1、BDF1、B6C3F1)的体外受精 (IVF)率、胚胎培养及移植成绩进行了比较研究。结果 各品系小鼠新鲜精子的IVF率 15 1%~ 87 9% ,冻融精子的IVF率 8%~ 80 % ;冷冻胚胎的复苏率4 2 6 %~ 83 9% ;冻融胚胎移植后的产仔率在 17 8%~ 5 1 8%。结论 遗传背景不同的小鼠体外受精率、冷冻胚胎复苏率和胚胎移植的产仔率差异有显著性。但同一品系两个实验室间的新鲜精子的IVF率、冷冻胚胎的复苏率及移植产仔率差异无显著性 (P >0 0 5 ) ;冻融精子的体外受精率CARD明显高于SLAC(P <0 0 1)。  相似文献

Cdc25C is a dual specificity phosphatase essential for dephosphorylation and activation of cyclin-dependent kinase 1 (cdk1), a prerequisite step for mitosis in all eucaryotes. Cdc25C activation requires phosphorylation on at least six sites including serine 214 (S214) which is essential for metaphase/anaphase transit. Here, we have investigated S214 phosphorylation during human meiosis with the objectives of determining if this mitotic phosphatase cdc25C participates in final meiotic divisions in human oocytes. One hundred forty-eight human oocytes from controlled ovarian stimulation protocols were stained for immunofluorescence: 33 germinal vesicle (GV), 37 metaphase stage I (MI), and 78 unfertilized metaphase stage II (MII). Results were stage dependent, identical, independent of infertility type, or stimulation protocol. During GV stages, phospho-cdc25C is localized at the oocyte periphery. During early meiosis I (MI), phosphorylated cdc25C is no longer detected until onset of meiosis I. Here, phospho-cdc25C localizes on interstitial microtubules and at the cell periphery corresponding to the point of polar body expulsion. As the first polar body reaches the periphery, phosphorylated cdc25C is localized at the junction corresponding to the mid body position. On polar body expulsion, the interior signal for phospho-cdc25C is lost, but remains clearly visible in the extruded polar body. In atresic or damaged oocytes, the polar body no longer stains for phospho-cdc25C. Human cdc25C is both present and phosphorylated during meiosis I and localizes in a fashion similar to that seen during human mitotic divisions implying that the involvement of cdc25C is conserved and functional in meiotic cells.  相似文献

人类透明带(zona pellucida,ZP)是在卵泡发生过程中由卵母细胞和颗粒细胞共同分泌的由ZP1-ZP4四种糖蛋白分子组成的高度有序结构,它与卵母细胞的成熟、受精、胚胎发育及妊娠结局等预后紧密关联。许多生殖中心实验室发现有些患者的卵出现全部或部分的透明带异常,而且不同实验室发现的透明带异常类型各异。研究发现这些透明带异常与卵母细胞受精、胚胎发育及临床结局有一定的相关性。本文综述了关于透明带异常及其对卵母细胞受精、胚胎发育潜能和临床结局的影响的研究进展。  相似文献

HCG是由胎盘的滋养层细胞分泌的一种糖蛋白,其测定可应用于诊断异位妊娠、先兆流产、葡萄胎及妊娠滋养细胞肿瘤(GTN)等不良妊娠结局。HCG水平可用于预测子痫前期一子痫、流产等。D-二聚体是交联的纤维蛋白被纤溶酶水解后产生的一种特异性分子标志物,是继发性纤溶的重要指征,对诊断血栓性疾病具有重要意义。孕妇D-二聚体与妊娠期高血压疾病、产后大出血、妊娠合并心脏病等有关。为了探讨妊娠中、晚期HCG、D-二聚体变化对妊娠结局的影响,通过复习国内外文献,分析孕中、晚期HCG、D2聚体变化与妊娠结局的相关性.为临床预测妊娠结局提供临床依据。  相似文献
