A Gram-positive bacterium, designated SCSIO 15042T, was isolated from a sediment of the South China Sea and was subjected to a polyphasic taxonomic study. The isolate grew at 20–60 °C, pH 6.0–10.0 and it could grow with up to 10 % (w/v) NaCl. The cell-wall diamino acid was found to be meso-diaminopimelic acid. Polar lipids were found to be diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine and an unknown polar lipid. The only menaquinone was determined to be MK-7. The major fatty acids were identified as C16:1 ω7c/C16:1 ω6c, C16:0, iso-C15:0, anteiso-C15:0, and iso-C16:0. The DNA G+C content of strain SCSIO 15042T was determined to be 43.1 mol%. Phylogenetic analysis based on 16S rRNA gene sequences affiliated strain SCSIO 15042T to the genus Bacillus. Levels of 16S rRNA gene sequence similarities between strain SCSIO 15042T and Bacillus herbersteinensis D-1-5aT, Bacillus infantis SMC 4352-1T, Bacillus novalis LMG 21837T and Bacillus drentensis LMG 21831T were 96.2, 96.2, 96.1 and 96.1 %, respectively. Based on the evidence of the present polyphasic study, strain SCSIO 15042T is considered to represent a novel species of the genus Bacillus, for which the name Bacillus abyssalis sp. nov. is proposed. Two gram-positive, aerobic, spore-forming, rod-shaped bacteria, designated HB09003T and HB12160, were isolated from seawater and sediment in the northern South China Sea, respectively. Cells were found to be motile by means of peritrichous flagella. The strains were found to grow with 0–15 % (w/v) NaCl, at 10–45 °C and pH 5.0–10.7, with an optimum of 3 % NaCl, 28 °C and pH 8.5, respectively. The predominant isoprenoid quinone of strain HB09003T, selected as the representative strain, was identified as MK-7. This strain was found to possess anteiso-C15:0, iso-C15:0, anteiso-C17:0 and C16:0 as the major fatty acids. The G+C contents of strain HB09003T and HB12160 were determined to be 34.1 and 34.3 mol%, respectively. Analysis of the 16S rRNA gene sequences of the two strains showed an affiliation with the genus Gracilibacillus, with Gracilibacillus kekensis CGMCC 1.10681T (similarity of 97.4, 98.0 %, respectively) and Gracilibacillus ureilyticus CGMCC 1.7727T (similarity of 97.1, 97.8 %, respectively) as their closest relatives. The DNA–DNA hybridization values between strain HB09003T and the two type strains were 42.2 and 54.1 %, respectively. On the basis of phenotypic and genotypic data, strain HB09003T and HB12160 are proposed to represent a novel species of the genus Gracilibacillus, for which the name Gracilibacillus marinus sp. nov. is proposed. A novel non-sporulating, non-motile, catalase-positive, oxidase-negative, facultatively anaerobic, Gram-positive coccus, designated strain JSM 078151T, was isolated from an intertidal sediment sample collected from Naozhou Island in the South China Sea, China. Growth was found to occur in the presence of 0–15 % (w/v) NaCl (optimum 0.5–3 % (w/v) NaCl), at pH 6.5–10.5 (optimum pH 7.0–8.0) and at 5–35 °C (optimum 25–30 °C). The peptidoglycan type was determined to be A3a, containing lysine, glutamic acid and alanine. The major cellular fatty acid identified was anteiso-C15:0 and the predominant menaquinones are MK-7 and MK-8. The polar lipids were found to consist of diphosphatidylglycerol, phosphatidylglycerol, glycolipid and one unidentified phospholipid. The genomic DNA G+C content of strain JSM 078151T was determined to be 55.2 mol%. A phylogenetic analysis based on 16S rRNA gene sequence comparisons revealed that strain JSM 078151T should be assigned to the genus Rothia, and was most closely related to Rothia nasimurium CCUG 35957T (98.3 % sequence similarity), followed by Rothia amarae J18T (97.5 %) and Rothia terrae L-143T (97.3 %). A combination of phylogenetic analysis, DNA–DNA relatedness values, phenotypic characteristics and chemotaxonomic data supports the suggestion that strain JSM 078151T represents a novel species of the genus Rothia, for which the name Rothia marina sp. nov. is proposed. A novel Gram-stain-positive, motile, hemolytic, endospore-forming and rod-shaped bacterium was isolated and designated as strain GIESS003T. The strain grew optimally at 35 °C, at pH 7.0–7.5, and with 3.0–3.5 % (w/v) NaCl. The 16S rRNA gene sequence analysis indicated that strain GIESS003T was associated with the genus Ornithinibacillus and was most closely related to the type strain of Ornithinibacillus contaminans (96.5 % similarity). The major cellular fatty acids were iso-C15:0 and anteiso-C15:0. The polar lipids were diphosphatidylglycerol and phosphatidylglycerol. The major respiratory quinone was menaquinone-7. Strain GIESS003T contained a peptidoglycan of type A4β l-Orn-d-Asp. The G+C content of genomic DNA was 40.1 mol%. On the basis of polyphasic evidence from this study, a new species of the genus Ornithinibacillus, Ornithinibacillus heyuanensis sp. nov., is proposed, with strain GIESS003T (=KCTC 33159T=CCTCC 2013106T) as the type strain.  相似文献

A Gram-stain positive, facultative anaerobic, motile, spore-forming rod-shaped bacterium with peritrichous flagella, designated DX-5T, was isolated from an electroactive biofilm. Growth was observed to occur at 35–60 °C, at pH 7.0–10.0 and with 0.5–10 % (w/v) NaCl (optimum growth: 50 °C, pH 8.0 and 0.5–3 % NaCl). Cells were determined to be catalase- and oxidase-positive. The predominant respiratory quinone was identified as MK-7; the major polar lipids were determined to be diphosphatidylglycerol, phosphatidylglycerol, glycolipid, aminoglycolipid and an unidentified phospholipid; the DNA G+C content was determined to be 46.6 mol%; and the major fatty acids (>5 %) were identified as anteiso-C15:0 (33.6 %), iso-C15:0 (24.1 %) and anteiso-C17:0 (13.4 %). The phylogenetic analysis based on 16S rRNA gene sequence comparisons indicated that strain DX-5T should be assigned to the genus Bacillus, and was related most closely to the type strains of B. fortis DSM 16012T (96.3 %), B. composti KACC 16872T (96.3 %) and B. fordii DSM 16014T (95.8 %). Results of phenotypic, chemotaxonomic and genotypic analysis indicated that strain DX-5T represents a novel species, for which the name B. sediminis sp. nov. is proposed. An actinomycete capable of lysing cyanobacteria, strain JXJ 0074T, was isolated from a soil sample collected from Jiangxi province, south China, and characterized by using polyphasic taxonomy. The new isolate showed morphological and chemotaxonomic properties typical of members of the genus Streptomyces. Phylogenetic analysis of the near-complete 16S rRNA gene sequence indicated that strain JXJ 0074T should be affiliated to the genus Streptomyces and exhibited highest similarities to Streptomyces shenzhenensis DSM 42034T (98.99 %) and Streptomyces lucensis NBRC 13056T (98.60 %), while the similarities to other members of the genus are lower than 98.22 % similarity. However, the DNA–DNA hybridization values between strain JXJ 0074T and S. shenzhenensis DSM 42034T or S. lucensis NBRC 13056T were 46.2 ± 2.6 and 32.6 ± 3.1 %, respectively. Thus, on the basis of the polyphasic data, strain JXJ 0074T represents a novel species of the genus Streptomyces, for which the name Streptomyces jiujiangensis sp. nov. is proposed. A Gram-negative, non-motile, short rod-shaped or spherical bacterial strain that accumulates poly-β-hydroxybutyrate (PHB) granules was isolated from the Beibu Gulf in the South China Sea. Cells have no polar or subpolar flagella, dividing by binary fission. Phylogenetic analyses based on 16S rRNA gene sequences showed that the strain formed a monophyletic branch at the periphery of the evolutionary radiation occupied by the genus Paracoccus, family Rhodobacteraceae, order Rhodobacterales, class Alphaproteobacteria. The closest neighbours were Paracoccus aestuarii strain B7(T) (97.2% similarity), Paracoccus zeaxanthinifaciens ATCC 21588(T) (97.1% similarity) and Paracoccus homiensis DD-R11(T) (96.8%). The predominant fatty acids were C(18:1) ω7c (82.1%), and significant amounts of C(18:0) (5.6%), C(10:0) 3-OH (2.3%) and C(16:0) (1.5%) were present. The predominant respiratory ubiquinone of strain JLT1284(T) was Q-10 and the DNA G+C content of strain JLT1284(T) was 67.0 mol%. The isolate was also distinguishable from members of the genus Paracoccus on the basis of phenotypic and biochemical characteristics. It is evident from the genotypic, chemotaxonomic and phenotypic data, therefore, that strain JLT1284(T) represents a novel species of the genus Paracoccus, for which the name Paracoccus beibuensis sp. nov. is proposed. Psychrotolerant Bacillus-like strains BR035(T) and BR011 were isolated from seawater of the Bering Sea and were characterized by means of a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences revealed that these strains were related to the members of the genus Bacillus and had the highest 16S rRNA gene sequence similarity with Bacillus korlensis ZLC-26(T). DNA-DNA hybridization experiments confirmed that strains BR035(T) and BR011 belonged to the same species and were distinct from their closest relatives. The cells were Gram-positive, rods, motile, spore-forming and psychrotolerant. The temperature range for growth was 4-42°C. The main respiratory quinone was MK-7. The polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, an unknown aminolipid and two unknown phospholipids. The major cellular fatty acids were iso-C15:0, anteiso-C15:0, iso-C14:0 and C16:1ω7c alcohol. The diagnostic diamino acid in the cell-wall peptidoglycan was meso-diaminopimelic acid. The genomic DNA G + C content was 37.6-37.8 mol%. On the basis of the phenotypic characteristics, phylogenetic analysis and DNA-DNA relatedness data, a novel species Bacillus beringensis is proposed and the type strain is BR035(T) (=CGMCC 1.9126(T)=DSM 22571(T)).  相似文献

A novel bacterial strain, designated SGD-1123T was isolated from Chorao Island, in Goa Province, India. The strain was found to be able to grow at 15–42 °C, pH 5–12 and 0–12 % (w/v) NaCl. The whole cell hydrolysates were found to contain meso-diaminopimelic acid, galactose and arabinose. The major fatty acids were identified as iso-C15:0 and anteiso-C15:0, MK-7 was identified as the predominant menaquinone and the predominant polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and an unidentified aminolipid. The genomic DNA G+C content was determined to be 44.6 mol%. Phylogenetic analysis based on 16S rRNA gene sequences placed the isolate within the genus Bacillus and further revealed that strain SGD-1123T had highest sequence similarity with Bacillus aquimaris, and forms a separate clade with its closest relatives i.e. B. aquimaris, Bacillus vietnamensis and Bacillus marisflavi, with which it shares 94.5, 94.1 and 94.1 % similarity respectively. The phylogenetic, chemotaxonomic and phenotypic analyses indicated that strain SGD-1123T represents a novel species within the genus Bacillus, for which the name Bacillus enclensis is proposed. A Gram-positive, rod-shaped, endospore-forming organism, strain BL3-6T, was isolated from tidal flat sediments of the Yellow Sea in the region of Tae-An. A 16S rRNA gene sequence analysis demonstrated that this isolate belongs to the Bacillus cereus group, and is closely related to Bacillus mycoides (99.0% similarity), Bacillus thuringiensis (99.0%), Bacillus weihenstephanensis (99.0%), Bacillus cereus (98.9%), Bacillus anthracis (98.8%), and Bacillus pseudomycoides (98.1%). The phylogenetic distance from any validly described Bacillus species outside the Bacillus cereus group was less than 95.6%. The DNA G+C content of the strain was 39.4 mol% and the major respiratory quinone was menaquinone-7. The major cellular fatty acids were iso-C14:0 (17.8%), iso-C16:0 (15.8%), and iso-C12:0 (11.3%). The diagnostic amino acid of the cell wall was meso-diaminopimelic acid and the major cell wall sugar was galactose. The results of DNA-DNA hybridization (<55.6%) and physiological and biochemical tests allowed genotypic and phenotypic differentiation of strain BL3-6T from the published Bacillus species. BL3-6T therefore represents a new species, for which the name Bacillus gaemokensis sp. nov. is proposed, with the type strain BL3-6T (=KCTC 13318T =JCM 15801T).  相似文献

A Gram-stain negative, motile, rod-shaped bacterium, designated strain WM-2T, was isolated from a forest soil in Sihui City, South China, and characterized by means of a polyphasic approach. Growth occurred with 0–5 % (w/v) NaCl (optimum 0–1 %) and at pH 5.0–10.5 (optimum pH 8.5) and 4–40 °C (optimum 30 °C) in Luria–Bertani medium. Comparative 16S rRNA gene sequence analyses showed that strain WM-2T is a member of the genus Pseudomonas and most closely related to P. guguanensis, P. oleovorans subsp. lubricantis, P. toyotomiensis, P. alcaliphila and P. mendocina with 97.1–96.6 % sequence similarities. In terms of gyrB and rpoB gene sequences, strain WM-2T showed the highest similarity with the type strains of the species P. toyotomiensis and P. alcaliphila. The DNA–DNA relatedness values of strain WM-2T with P. guguanensis and P. oleovorans subsp. lubricantis was 48.7 and 37.2 %, respectively. Chemotaxonomic characteristics (the main ubiquinone Q-9, major fatty acids C18:1 ω7c/C18:1 ω6c, C16:0 and C16:1 ω7c/C16:1 ω6c and DNA G+C content 65.2 ± 0.7 mol%) were similar to those of members of the genus Pseudomonas. Polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, an unknown aminophospholipid, an unknown phospholipid and five unknown lipids. According to the results of polyphasic analyses, strain WM-2T represents a novel species in the genus Pseudomonas, for which the name Pseudomonas sihuiensis sp. nov. is proposed. A novel Gram-negative, aerobic, slightly halophilic, yellow-pigmented, oxidase-negative, Voges–Proskauer positive, non-spore-forming bacterium, designated YIM M 13059T, was isolated from a sediment sample collected from the South China Sea at a depth of 310 m. Optimal growth was found to occur at 28–30 °C, pH 7.0 and in the presence of 3–4 % (w/v) NaCl. Cells were observed to be rod-shaped and motile by peritrichous flagella. The polar lipids of strain YIM M 13059T were found to be diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, a ninhydrin-positive phospholipid, one glycolipid and two unknown phospholipids. The predominant respiratory quinone was determined to be Q-9. The major fatty acids were identified as C18:1 ω7c, C16:1 ω6c/C16:1 ω7c, C16:0 and C12:0 3-OH. The genomic DNA G+C content was determined to be 54.4 mol%. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that the isolate belongs to the genus Halomonas in the family Halomonadaceae. The 16S rRNA gene sequence similarities between strain YIM M 13059 T and the type strains of members of the genus Halomonas were in the range 93.3–98.3 %. However, the levels of DNA–DNA relatedness values between YIM M 13059 and the type strains of the most closely related species, Halomonas zhangjiangensis, Halomonas variabilis, Halomonas neptunia, Halomonas boliviensis and Halomonas sulfadieris were 50.2 ± 0.68 %, 46.8 ± 1.9 %, 28.5 ± 0.74 %, 42.9 ± 0.55 % and 37.1 ± 0.68 %, respectively. Based on phylogenetic, chemotaxonomic and phenotypic data, the strain YIM M 13059T is proposed to represent a novel member of the genus Halomonas, with the name Halomonas nanhaiensis sp. nov. 目的】研究来源于南海海水的一株速生杆菌属菌株NH195T 的多相分类。【方法】采用表型、基因型和化学分类方法,并综合系统发育关系结果,分析菌株NH195T 的分类学地位。【结果】菌株NH195T 是一株革兰氏阴性、好氧、杆状、无运动性细菌;能积累Poly-β-hydroxybutyrate (PHB);能在0.5%-10.0% (质量体积比) NaCl 浓度,pH 5.0-9.0 和20-40 ℃ 条件下生长,最适NaCl 生长浓度为1.0%-3.0%;氧化酶、触酶和脲酶反应结果阳性。菌株NH195T 主要呼吸醌为Q-10,主要脂肪酸为C18:1ω7c、C18:1ω6c 和11 methyl C18:1ω7c,主要极性脂为磷脂酰胆碱、磷脂酰甘油、双磷脂酰甘油、一个未知的氨基脂和两个未知脂。基因组G+C 含量为61.3 mol%。基于16S rRNA 基因的系统发育结果显示,菌株NH195T 隶属于速生杆菌属;其与速生杆菌属标准菌株的16S rRNA 基因相似性范围为94.4%-97.7%。菌株NH195T 与速生杆菌属标准菌株C. halophilus ZXM137TC. indicus P73T 的平均核苷酸一致性(ANI)分别为78.6%和78.0%;基于基因组数据计算所得的DNA 杂交同源率分别为26.1%和23.0%。【结论】基于表型和基因型结果,菌株NH195T 代表了速生杆菌属一个新物种,命名为Celeribacter ethanolicus,标准菌株为NH195T (CGMCC 1.15406T=JCM 31095T)。  相似文献

A non-pigmented, motile, Gram-negative bacterium designated H 17T was isolated from a seawater sample collected in Port Phillip Bay (the Tasman Sea, Pacific Ocean). The new organism displayed optimal growth between 4 and 37 °C, was found to be neutrophilic and slightly halophilic, tolerating salt water environments up to 10 % NaCl. Strain H 17T was found to be able to degrade starch and Tween 80 but unable to degrade gelatin or agar. Phosphatidylglycerol (27.7 %) and phosphatidylethanolamine (72.3 %) were found to be the only associated phospholipids. The major fatty acids identified are typical for the genus Alteromonas and include C16:0, C16:1ω7, C17:1ω8 and C18:1ω7. The G+C content of the DNA was found to be 43.4 mol%. A phylogenetic study, based on the 16S rRNA gene sequence analysis and Multilocus Phylogenetic Analysis, clearly indicated that strain H 17T belongs to the genus Alteromonas. The DNA?DNA relatedness between strain H 17T and the validly named Alteromonas species was between 30.7 and 46.4 mol%. Based on these results, a new species, Alteromonas australica, is proposed.
