The efficiencies of mono gradient elution and dual salt‐pH gradient elution for separation of six mAb charge and size variants on a preparative‐scale ion exchange chromatographic resin are compared in this study. Results showed that opposite dual salt‐pH gradient elution with increasing pH gradient and simultaneously decreasing salt gradient is best suited for the separation of these mAb charge and size variants on Eshmuno® CPX. Besides giving high binding capacity, this type of opposite dual salt‐pH gradient also provides better resolved mAb variant peaks and lower conductivity in the elution pools compared to single pH or salt gradients. To have a mechanistic understanding of the differences in mAb variants retention behaviors of mono pH gradient, parallel dual salt‐pH gradient, and opposite dual salt‐pH gradient, a linear gradient elution model was used. After determining the model parameters using the linear gradient elution model, 2D plots were used to show the pH and salt dependencies of the reciprocals of distribution coefficient, equilibrium constant, and effective ionic capacity of the mAb variants in these gradient elution systems. Comparison of the 2D plots indicated that the advantage of opposite dual salt‐pH gradient system with increasing pH gradient and simultaneously decreasing salt gradient is the noncontinuous increased acceleration of protein migration. Furthermore, the fitted model parameters can be used for the prediction and optimization of mAb variants separation in dual salt‐pH gradient and step elution. © 2018 American Institute of Chemical Engineers Biotechnol. The apparent tendency of atherosclerotic lesions to form in complex blood flow environments has led to many theories regarding the importance of hemodynamic forces in endothelium-mediated atherosusceptibility. The effects of shear stress magnitude and spatial shear stress gradient on endothelial cell gene expression in vitro were examined in this study. Converging-width flow channels were designed to impose physiological ranges of shear stress gradient and magnitude on porcine aortic endothelial cells, and real-time quantitative PCR was performed to evaluate their expression of five genes of interest. Although vascular cell adhesion molecule-1 expression was insensitive to either variable, each of the remaining genes exhibited a unique dependence on shear stress magnitude and gradient. Endothelial nitric oxide synthase showed a strong positive dependence on magnitude but was insensitive to gradient. The expression of c-jun was weakly correlated with magnitude and gradient, without an interaction effect. Monocyte chemoattractant protein-1 expression varied inversely with gradient and also depended on the interaction of gradient with magnitude. Intercellular adhesion molecule-1 expression also exhibited an interaction effect, and increased with shear magnitude. These results support the notion that vascular endothelial cells are able to sense shear gradient and magnitude independently.  相似文献

以野外径流小区的次降雨产流产沙数据为基础,对不同次降雨条件下坡面产流产沙随坡度的变化规律进行研究.结果表明:降雨性质的差异对坡面产流随坡度而变化的影响较小,在研究区的6场次降雨中,坡面产流量随坡度的变化规律基本一致,坡面径流量均随坡度增大而增大,但坡面径流量随坡度增加而增加的趋势较弱,这主要由降雨量与入渗量之间的转化程度所决定;次降雨性质对不同坡度的坡面产沙规律有重要影响,一般情况下存在临界侵蚀坡度,但临界坡度不是唯一值,而是随着降雨特性的不同而不同,临界侵蚀坡度随径流量的增大而增大,临界坡度较大时,坡面产沙量随坡度变化而变化的趋势也往往较大.  相似文献

Reversed-phase gradient LC-MS/MS bioanalytical methods of 5-100% organic solvent in a 1-3 min gradient time are common in today's bioanalytical laboratory. The goal of this work was to develop a theory-guided systematic strategy for maximizing resolution and speed in rapid gradient LC-MS/MS bioanalysis. We studied the effect of gradient time (t(G)), initial and final eluent strength (% B=% organic), and flow rate (F) on the separation of multiple critical pairs (R(s)) and peak capacity (n(c)) in a gradient elution of a mixture of five structurally related compounds. By optimizing the gradient time t(G), the initial and final percentages of the organic component of the mobile phase, comparable resolution and peak capacity could be achieved in a shorter run time. More importantly, we demonstrated that higher flow rates improved resolution, peak capacity and reduced run time in rapid gradient separations on a 5 μm particle column. A straightforward mathematical explanation of the phenomenon was provided applying basic resolution equations in gradient elution theory. A systematic approach to execute a rapid gradient LC-MS/MS bioanalytical method to shorten run time and improve resolution is proposed, taking into consideration not only the analytes of interest but also potential matrix effects from the dosing vehicle and biological matrix.  相似文献

Predicting the effect of a changing environment, e.g., caused by climate change, on realized niche dynamics, and consequently, biodiversity is a challenging scientific question that needs to be addressed. One promising approach is to use estimated demographic parameters for predicting plant abundance and occurrence probabilities. Using longitudinal pinpoint cover data sampled along a hydrological gradient in the Marais poitevin grasslands, France, the effect of the gradient on the demographic probabilities of colonization and survival was estimated. The estimated probabilities and calculated elasticities of survival and colonization covaried with the observed cover of the different species along the hydrological gradient. For example, the flooding tolerant grass A. stolonifera showed a positive response in both colonization and survival to flooding, and the hydrological gradient is clearly the most likely explanation for the occurrence pattern observed for A. stolonifera. The results suggest that knowledge on the processes of colonization and survival of the individual species along the hydrological gradient is sufficient for at least a qualitative understanding of species occurrences along the gradient. The results support the hypothesis that colonization has a predominant role for determining the ecological success along the hydrological gradient compared to survival. Importantly, the study suggests that it may be possible to predict the realized niche of different species from demographic studies. This is encouraging for the important endeavor of predicting realized niche dynamics.  相似文献

High-field, high-speed Magnetic Resonance Imaging (MRI) generates high sound levels within and nearby the scanner. The mechanism and process that produces the gradient magnetic field (a cylindrical electro-magnet, called the gradient coil cylinder, which produces a spatially and temporally varying magnetic field inside a static background magnetic field) is the primary source of this noise. This noise can cause difficulties in verbal communication in and around the scanner, heightened patient anxiety, temporary hearing loss and possible permanent hearing impairment for health care workers and patients. In order to effectively suppress the sound radiation from the gradient coil cylinder the sound field within and nearby the gradient coil needs to be characterized This characterization may be made using an analytical solution of the sound pressure field, computational simulation, measurement analysis or some combination of these three methods. This paper presents the computational simulation and measurement results of a study of the sound radiation from a head and neck gradient coil cylinder within a 4 Tesla MRI whole body scanner. The measurement results for the sound pressure level distribution along the centerline of the gradient coil cylinder are presented. The sound pressure distributions predicted from Finite Element Analysis of the gradient coil movement during operation and subsequent Boundary Element Analysis of the sound field generated are also presented. A comparison of the measured results and the predicted results shows close agreement. Because of the extremely complex nature of the analytical solution for the gradient coil cylinder, a treatment of the analytical solution and comparison to the computational results for a simple cylinder vibrating in a purely radial direction are also presented and also show close agreement between the two methods thus validating the computational approach used with the more complex gradient coil cylinder.  相似文献

The effect of the transmembrane proton gradient (delta pH) and potential gradient (delta psi) upon the rate and extent of amine accumulation was investigated in chromaffin ghosts. The chromaffin ghosts were formed by hypo-osmotic lysis of isolated bovine chromaffin granules and extensive dialysis in order to remove intragranular binding components and dissipate the endogenous electrochemical gradients. Upon ATP addition to suspensions of chromaffin ghosts, a transmembrane proton gradient alone, a transmembrane gradient alone, or both, could be established, depending upon the compositions of the media in which the ghosts were formed and resuspended. When chloride was present in the medium, addition of ATP resulted in the generation of a transmembrane proton gradient, acidic inside of 1 pH unit (measured by [14C]methylamine distribution), and no transmembrane potential (measured by [14C]-thiocyanate distribution). When ATP was added to chromaffin ghosts suspended in a medium in which chloride was substituted by isethionate, a transmembrane potential, inside positive, of 45 mV and no transmembrane proton gradient, was measured. In each medium, the addition of agents known to affect proton or potential gradients, respectively, exerted a predictable mechanism of action. Accumulation of [14C]epinephrine or [14C]5-hydroxytryptamine was over 1 order of magnitude greater in the presence of the transmembrane proton gradient or the transmembrane potential than in the absence of any gradient and, moreover, was related to the magnitude of the proton or potential gradient in a dose-dependent manner. When ghosts were added to a medium containing chloride and isethionate, both a delta pH and delta psi could be generated upon addition of ATP. In this preparation, the maximal rate of amine accumulation was observed. The results indicate that amine accumulation into chromaffin ghosts can occur in the presence of either a transmembrane proton gradient, or a transmembrane potential gradient, and that the maximal rate of accumulation may exist when both components of the protonmotive force are present.  相似文献

Summary The climatic and physiographic features of the Western Desert of Egypt vary gradually from the Mediterranean coast inland. The present study provides an analysis of the gradients exhibited by these environmental variations and their relationships with vegetational gradients and the phytosociological behaviour of species in a sector along the Cairo-Alexandria desert road. The multivariate analyses of the vegetation lead to the recognition of six interrelated vegetational groupings which were distributed along a climatic gradient of increasing aridity, and an edaphic gradient of increasing sandiness and decreasing salinity and level of total phosphorus. These groupings were distinguished into three sets: a set ofConvolvulus-Helianthemum-Artemisia, andHelianthemum-Echiochilon, a set ofAnabasis, andSalsola-Anabasis, and a set ofAsphodelus-Noaea andThymelaea-Anabasis-Noaea groupings. The first set occupies the more arid end of the climatic gradient and the end of the coarse texture, low salinity and low levels of total phosphorus of the edaphic gradient. The second set occupies the opposite end of the edaphic gradient and the middle position of the climatic gradient and the middle position of the climatic gradient, and the third, the middle position of the edaphic gradient and the less arid end of the climatic gradient. Taxonomic nomenclature is according to T?ckholm (1974). Abstract. A new measure of compositional turnover rate at any gradient point is proposed. The absolute values of the derivatives of estimated species response curves are used to estimate the instantaneous rate of change of the community. The total gradient length can be found by integrating the instantaneous turnover rates over the gradient span. Both the instantaneous rate and total gradient length have been known under the name of beta diversity. Our method is elaborated for the Gaussian response function. It is compared to Hill's SD measure, much used for estimating the gradient length in connection of Detrended Correspondence Analysis with nonlinear rescaling for axes. The turnover rate along a canopy cover gradient in forest vegetation in Finland was used as an example. Forest site type and stand age were used as subsidiary gradients. The two ways to compute Hill's scaling gave very different results and were not comparable with the new method. The turnover rate was dependent on the gradient position. However, the turnover rate along the gradient of interest and its total length were dependent on the subsidiary gradients.  相似文献

The physical and functional properties of Leydig cell populations obtained by centrifugation of testicular cells in two different density gradient media, Percoll and Metrizamide, were compared. Percoll-gradient centrifugation yielded two Leydig cell bands (Peak I and Peak II) that were comparable, as to their density and testosterone-producing capacity, to the respective Leydig cell bands, Population I and Population II, isolated in a Metrizamide gradient. The denser Leydig cell band (II) had a greater capacity for testosterone production than the less dense band (I), regardless of the type of gradient used for its isolation. Metrizamide gradient centrifugation separated the majority of germ cells from the "light" (Population I) Leydig cells, whereas in the Percoll gradient, germ cells comigrated with Peak I Leydig cells. Leydig cell separation by Percoll gradients was highly dependent on the presence of Ca2+ and Mg2+ in the medium, while these cations had no effect on the separation of Leydig cells by Metrizamide. In conclusion, Metrizamide gradient centrifugation yielded two Leydig cell populations of similar functional and physical properties to the respective populations isolated in Percoll gradients.  相似文献

Chlorotetracyclin (10-4M) has been used to observe the distribution of membrane-associated calcium during pollen germination of Lilium longiflorum. For comparison, the general membrane distribution has been determined with 4·10-5 M fluorescamine. The pollen grains show a calcium gradient with either weak or strong chlorotetracycline-fluorescence intensity, but always increasing toward the germination colpus. This gradient intensifies during germination, reaching a maximum before the pollen tube emerges. The typical tip-to-base calcium gradient of the tube does not change during growth. Independent of the developmental stage, the pollen grains show a flat fluorescamine-fluorescence gradient with the highest intensity in one half of the grain. Pollen tubes reveal a tip-to-base membrane gradient, independent of their length. As an additional marker for membrane distribution, the distribution of phosphorus, measured by proton-induced X-ray emission in chemically fixed tubes, has been used. Decapitated pending vines are the most responsive parts of vanilla plants. Inflorescence localization along training vines showed a decreasing gradient from the decapitated end to the base in sun locality and was irregular and showed opposite gradient in shade locality. Soluble and ionic peroxidase activities, determined in the leaves and in the internodes, showed a marked peak during the end of June and a gradient along training vines which was inverse to the gradient of inflorescence localization and opposite between sun or shade conditions, supporting the utility of peroxidases as spatial and temporal biochemical markers of flowering processes.  相似文献

In a recent publication, we identified a novel F-box protein, encoded by fates-shifted (fsd), that plays a role in targeting Bcd for ubiquitination and degradation. Our analysis of mutant Drosophila embryos suggests that Bcd protein degradation is important for proper gradient formation and developmental fate specification. Here we describe further experiments that lead to an estimate of Bcd half-life, <15 min, in embryos during the time of gradient formation. We use our findings to evaluate different models of Bcd gradient formation. With this new estimate, we simulate the Bcd gradient formation process in our own biologically realistic 2-D model. Finally, we discuss the role of Bcd-encoded positional information in controlling the positioning and precision of developmental decisions.  相似文献

This paper examines the relationship between protected and endangered riverine species (target species) and hydrodynamics in river-floodplain ecosystems, combining ecological and policy-legal aspects of biodiversity conservation in river management. The importance of different hydrodynamic conditions along a lateral gradient was quantified for various taxonomic groups. Our results show that (i) target species require ecotopes along the entire hydrodynamic gradient; (ii) different parts of the hydrodynamic gradient are important to different species, belonging to different taxonomic groups; (iii) in particular low-dynamic parts are important for many species and (iv) species differ in their specificity for hydrodynamic conditions. Many species of higher plants, fish and butterflies have a narrow range for hydrodynamics and many species of birds and mammals use ecotopes along the entire gradient. Even when focussing only on target species, the entire natural hydrodynamic gradient is important. This means that the riverine species assemblage as a whole can benefit from measures focussing on target species only. River reconstruction and management should aim at re-establishing the entire hydrodynamic gradient, increasing the spatial heterogeneity of hydrodynamic conditions.  相似文献

Streptococcus cremoris was grown in pH-regulated batch and continuous cultures with lactose as the energy source. During growth the magnitude and composition of the electrochemical proton gradient and the lactate concentration gradient were determined. The upper limit of the number of protons translocated with a lactate molecule during lactate excretion (the proton-lactate stoichiometry) was calculated from the magnitudes of the membrane potential, the transmembrane pH difference, and the lactate concentration gradient. In cells growing in continuous culture, a low lactate concentration gradient (an internal lactate concentration of 35 to 45 mM at an external lactate concentration of 25 mM) existed. The cell yield (Ymax lactose) increased with increasing growth pH. In batch culture at pH 6.34, a considerable lactate gradient (more than 60 mV) was present during the early stages of growth. As growth continued, the electrochemical proton gradient did not change significantly (from -100 to -110 mV), but the lactate gradient decreased gradually. The H+-lactate stoichiometry of the excretion process decreased from 1.5 to about 0.9. In nongrowing cells, the magnitude and composition of the electrochemical proton gradient was dependent on the external pH but not on the external lactate concentration (up to 50 mM). The magnitude of the lactate gradient was independent of the external pH but decreased greatly with increasing external lactate concentrations. At very low lactate concentrations, a lactate gradient of 100 mV existed, which decreased to about 40 mV at 50 mM external lactate. As a consequence, the proton-lactate stoichiometry decreased with increasing external concentrations of protons and lactate at pH 7.0 from 1 mM lactate to 1.1 at 50 mM lactate and at pH 5.5 from 1.4 at l mM lactate to 0.7 at 50 mM lactate. The data presented in this paper suggest that a decrease in external pH and an increase in external lactate concentration both result in lower proton-lactate stoichiometry values and therefore in a decrease of the generation of metabolic energy by the end product efflux process.  相似文献

The role of the proton gradient and external pH in the motility and chemotaxis of Bacillus subtilis was investigated. Presence of a substantial proton gradient is not necessary for motility or chemotaxis, as long as the electrical potential is sufficient to maintain motility. Changes in the proton gradient do, however, lead to changes in swimming behavior, and these changes are mediated by two processes. One is sensitive to external pH and probably operates through a pH receptor. The second is sensitive to changes in the proton gradient. When the level of the protonmotive force is high enough to maintain motiligy, changes in the components of the protonmotive force are sensed by the bacteria and lead to behavioral changes, but changes in the protonmotive force are not necessary for chemotaxis.  相似文献

Rhythmic production of vertebral precursors, the somites, causes bilateral columns of embryonic segments to form. This process involves a molecular oscillator--the segmentation clock--whose signal is translated into a spatial, periodic pattern by a complex signalling gradient system within the presomitic mesoderm (PSM). In mouse embryos, Wnt signalling has been implicated in both the clock and gradient mechanisms, but how the Wnt pathway can perform these two functions simultaneously remains unclear. Here, we use a yellow fluorescent protein (YFP)-based, real-time imaging system in mouse embryos to demonstrate that clock oscillations are independent of beta-catenin protein levels. In contrast, we show that the Wnt-signalling gradient is established through a nuclear beta-catenin protein gradient in the posterior PSM. This gradient of nuclear beta-catenin defines the size of the oscillatory field and controls key aspects of PSM maturation and segment formation, emphasizing the central role of Wnt signalling in this process.  相似文献

Drosophila Wingless (Wg) is the founding member of the Wnt family of secreted proteins. During the wing development, Wg acts as a morphogen whose concentration gradient provides positional cues for wing patterning. The molecular mechanism(s) of Wg gradient formation is not fully understood. Here, we systematically analyzed the roles of glypicans Dally and Dally-like protein (Dlp), the Wg receptors Frizzled (Fz) and Fz2, and the Wg co-receptor Arrow (Arr) in Wg gradient formation in the wing disc. We demonstrate that both Dally and Dlp are essential and have different roles in Wg gradient formation. The specificities of Dally and Dlp in Wg gradient formation are at least partially achieved by their distinct expression patterns. To our surprise, although Fz2 was suggested to play an essential role in Wg gradient formation by ectopic expression studies, removal of Fz2 activity does not alter the extracellular Wg gradient. Interestingly, removal of both Fz and Fz2, or Arr causes enhanced extracellular Wg levels, which is mainly resulted from upregulated Dlp levels. We further show that Notum, a negative regulator of Wg signaling, downregulates Wg signaling mainly by modifying Dally. Last, we demonstrate that Wg movement is impeded by cells mutant for both dally and dlp. Together, these new findings suggest that the Wg morphogen gradient in the wing disc is mainly controlled by combined actions of Dally and Dlp. We propose that Wg establishes its concentration gradient by a restricted diffusion mechanism involving Dally and Dlp in the wing disc.  相似文献

Stable isotope probing (SIP) of nucleic acids allows the detection and identification of active members of natural microbial populations that are involved in the assimilation of an isotopically labelled compound into nucleic acids. SIP is based on the separation of isotopically labelled DNA or rRNA by isopycnic density gradient centrifugation. We have developed a highly sensitive protocol for the detection of 'light' and 'heavy' nucleic acids in fractions of centrifugation gradients. It involves the fluorometric quantification of total DNA or rRNA, and the quantification of either 16S rRNA genes or 16S rRNA in gradient fractions by real-time PCR with domain-specific primers. Using this approach, we found that fully 13C-labelled DNA or rRNA of Methylobacterium extorquens was quantitatively resolved from unlabelled DNA or rRNA of Methanosarcina barkeri by cesium chloride or cesium trifluoroacetate density gradient centrifugation respectively. However, a constant low background of unspecific nucleic acids was detected in all DNA or rRNA gradient fractions, which is important for the interpretation of environmental SIP results. Consequently, quantitative analysis of gradient fractions provides a higher precision and finer resolution for retrieval of isotopically enriched nucleic acids than possible using ethidium bromide or gradient fractionation combined with fingerprinting analyses. This is a prerequisite for the fine-scale tracing of microbial populations metabolizing 13C-labelled compounds in natural ecosystems.  相似文献
