Acclimation of crucian carp and goldfish to temperatures below 15°C causes covering of the gill lamellae by a mass of cells termed the interlamellar cell mass (ILCM). Here we explore the cues underlying gill remodeling (removal or growth of an ILCM) and specifically test the hypotheses that 1) depletion of internal O(2) stores in the absence of any change in external O(2) status can trigger the removal of the ILCM in goldfish acclimated to 7°C, 2) exposing fish acclimated to 25°C to an abundance of O(2) (hyperoxia) can reverse the gill remodeling (i.e., cause the covering of lamellae by an expansion of the ILCM), and 3) neuroepithelial cells (NECs) are involved in signaling the shedding of the ILCM. Hypoxemia induced by phenylhydrazine (anemia) or 5% CO caused a decrease in the ILCM from 80% to 23% and 35%, respectively. Hyperoxia exposure at 25°C caused an increase to 67% of total ILCM and a smaller decrease in the size of the ILCM when fish were transferred from 7 to 25°C. Daily sodium cyanide injections were used to stimulate NECs; this treatment led to a significant decrease in the ILCM. Thus, the three major conclusions of this study are 1) that gill remodeling can occur during periods of internal hypoxemia, 2) that O(2) supply and demand may be a significant driving force shaping gill remodeling in goldfish, and 3) the NECs may play a role in triggering the shedding of the ILCM during hypoxia.

Under cold acclimated conditions, goldfish (Carassius auratus) express an interlamellar cell mass (ILCM) which limits diffusive ion loss but may also impede branchial ammonia excretion (J(amm)). In the present study, goldfish were subjected to a 2-week 5 or 25 °C acclimation in order to modulate the degree of ILCM gill coverage and determine potential effects on J(amm). 25 °C-fish displayed gill coverage which was significantly lower than the 5 °C-fish, though the ILCM was not completely absent in these fish. 5 °C-fish demonstrated J(amm) values approximately 60% lower than those of 25 °C-fish. The magnitude of anterior (branchial) J(amm) strongly correlated with gill coverage (r(2)=0.83), suggesting that the ILCM may impede branchial J(amm). Divided chamber experiments demonstrated that relative to the 25 °C-fish, 5 °C-fish relied more upon posterior routes of excretion. In response to high external ammonia (HEA; 1.5mM NH(4)HCO(3)) exposures, 25 °C-fish displayed ammonia uptake while 5 °C-fish maintained excretion against HEA, suggesting that the ILCM may act as a barrier preventing ammonia uptake. In summary, the ILCM appears to impede branchial J(amm), such that 5 °C-rely more on extra-branchial routes of excretion. We hypothesize that gill remodeling in these fish may be intimately tied to physiological adjustments on the whole-body scale.

Goldfish, Carassius auratus, adaptively remodel their gills in response to changes in ambient oxygen and temperature, altering the functional lamellar surface area to balance the opposing requirements for respiration and osmoregulation. In this study, the effects of thermal- and hypoxia-mediated gill remodeling on branchial Na+ fluxes and the distribution of putative Na+-transporting ionocytes in goldfish were assessed. When assessed either in vitro (isolated gill arches) or in vivo at a common water temperature, the presence of an interlamellar cell mass (ILCM) in fish acclimated to 7°C clearly decreased Na+ efflux across the gill relative to fish maintained at 25°C and lacking an ILCM. However, loss of the ILCM in 7°C-acclimated fish exposed to hypoxia led to a decrease in Na+ efflux (assessed under hypoxic conditions) despite the apparent large increases in functional lamellar surface area. Goldfish possessing an ILCM were able to sustain Na+ uptake, albeit at a lower rate matched to efflux, owing to the re-distribution of ionocytes expressing genes thought to be involved in Na+ uptake [Na+/H+ exchanger isoform 3 (NHE3) and V- type H+-ATPase] to the edge of the ILCM where they can establish contact with the surrounding environment. NHE-expressing cells co-localized with Na+/K+-ATPase expression, suggesting a role for NHE in Na+-uptake in the goldfish. Implications of the ILCM on ion fluxes in the goldfish are discussed.

Gill remodeling can be extensive in crucian carp, where up to a 7.5-fold increase in gill surface area has been observed during exposure to hypoxia through a reduction in the interlamellar cell mass (ILCM) and increased lamellar protrusion that has been hypothesized to be signaled by the need to maximize oxygen uptake under a given condition. Sustained aerobic exercise may have the greatest influence on oxygen demand in fish; however, its effect on gill remodeling in crucian carp has not been investigated. The specific objectives of this study were to determine (i) whether sustained aerobic exercise induces gill remodeling in the crucian carp, (ii) whether gill remodeling following sustained exercise affects the maximum critical swimming speed (U(crit)) and maximal oxygen consumption rate ([Formula: see text]), and (iii) whether gill remodeling following sustained exercise is associated with trade-offs related to ionoregulation. We measured [Formula: see text] in crucian carp at each step during an initial U(crit) test (U(crit1)), forced them to swim at 70% of U(crit) for 40 h, and then conducted a second U(crit) test (U(crit2)). From rest to U(crit1) (7-8 h), we observed a significant increase in protruding lamella height and area of the gills and a reduction in ILCM height and volume, likely associated with partial shedding of the ILCM, indicating that gill remodeling during exercise is rapid. Further changes were observed between U(crit1) and U(crit2), with statistically significant increases in protruding lamellar height, basal length and area, and a statistically significant reduction in protruding lamellar thickness and ILCM height and volume. Interestingly, there was no significant difference between U(crit1) and U(crit2) values, nor in maximal [Formula: see text] measured at U(crit1) and U(crit2). Furthermore, there was no significant difference in plasma osmolarity, [Na(+)], or [Cl(-)] in fish at rest, following U(crit1) or U(crit2). Thus, while these data support the hypothesis that the need to maximize oxygen uptake is an important signal for gill remodeling, which can occur quite rapidly (within 7 h at 15°C), the physiological implications of remodeling during exercise are less clear.

Temperature acclimation may be a critical component of the locomotor physiology and ecology of ectothermic animals, particularly those living in eurythermal environments. Several studies of fish report striking acclimation of biochemical and kinetic properties in isolated muscle. However, the relatively few studies of whole-animal performance report variable acclimation responses. We test the hypothesis that different types of whole-animal locomotion will respond differently to temperature acclimation, probably due to divergent physiological bases of locomotion. We studied two cyprinid fishes, tinfoil barbs (Puntius schwanenfeldii) and river barbels (Barbus barbus). Study fish were acclimated to either cold or warm temperatures for at least 6 wk and then assayed at four test temperatures for three types of swimming performance. We measured voluntary swimming velocity to estimate routine locomotor behavior, maximum fast start velocity to estimate anaerobic capacity, and critical swimming velocity to estimate primarily aerobic capacity. All three performance measures showed some acute thermal dependence, generally a positive correlation between swimming speed and test temperature. However, each performance measure responded quite differently to acclimation. Critical speeds acclimated strongly, maximum speeds not at all, and voluntary speeds uniquely in each species. Thus we conclude that long-term temperature exposure can have very different consequences for different types of locomotion, consistent with our hypothesis. The data also address previous hypotheses that predict that polyploid and eurythermal fish will have greater acclimation abilities than other fish, due to increased genetic flexibility and ecological selection, respectively. Our results conflict with these predictions. River barbels are eurythermal polyploids and tinfoil barbs stenothermal diploids, yet voluntary swimming acclimated strongly in tinfoil barbs and minimally in river barbels, and acclimation was otherwise comparable.

Fathead minnows were subjected to an incremental velocity test using swim tunnel respirometry for the analysis of aerobic scope and swimming performance, as critical aerobic swim speed (U(crit)), following chronic exposures (33-57 ) to 0.9±0.4, 157±18 or 689±66 nmol L?1 Pb and an acute exposure (24 h) to 672±35 nmol L?1 Pb (mean±SEM). Assessment of Pb-induced anemia and neurological impairment were evaluated by blood hemoglobin (Hb) concentrations and a cost of transport (COT) analysis, respectively. Fish from the acute 672±35 nmol L?1 Pb (24.4±1.2 BL s?1) and chronic 689±66 nmol L?1 Pb (24.6±0.9 BL s?1) treatments exhibited reduced U(crits) compared to control fish (27.6±0.8 BL s?1). Aerobic scope was reduced by acute Pb exposure (8.6±2.6 μmol O? g?1 h?1 vs. 22.6±3.8 μmol O? g?1 h?1 from controls) owing to a decrease in maximum oxygen consumption rate (38.8±0.8 μmol O? g?1 h?1 vs. 54.0±4.2 μmol O? g?1 h?1 from controls). However, no effect on aerobic scope was observed with fish chronically exposed to Pb. Significant differences were not observed for Hb concentrations or COT. These findings suggest that the impaired swimming performances arising from acute and chronic Pb exposures reflect different mechanisms of toxicity.

Marine organisms living at low temperatures tend to have larger genomes and larger cells which suggest that these traits can be beneficial in colder environments. In fish, triploidy (three complete sets of chromosomes) can be induced experimentally following fertilization, which provides a model system to investigate the hypothesis that larger cells and genomes offers a physiological advantage at low temperatures. We tested this hypothesis by measuring metabolic rates and swimming performance of diploid and triploid Atlantic salmon (Salmo salar) post smolts acclimated to 3 or 10.5 °C. At 10.5 °C, triploids had significantly lower maximum metabolic rates which resulted in a lower aerobic scope compared to diploids. In addition, triploids initiated ram ventilation at lower swimming speeds, providing further evidence of a reduced capacity to meet oxygen demands during strenuous activity at 10.5 °C. However, at 3 °C, metabolic rates and critical swimming speeds were similar between both ploidies, and as expected substantially lower than at 10.5 °C. Therefore, triploidy in colder environments did not provide any advantage over diploidy in terms of metabolic rate traits or swimming performance in Atlantic salmon. We therefore conclude that traits, other than aerobic scope and swimming performance, contribute to the trend for increased cell and genome size in marine ectotherms living in cold environments.

To test whether the effects of temperature on the metabolic mode changed among different fish species, we investigated the specific dynamic action (SDA) and swimming performance of fasting and fed fish at 15 and 25°C in three juvenile Cyprinidae fish species: goldfish (Carassius auratus), common carp (Cyprinus carpio) and qingbo (Spinibarbus sinensis). Both taxon and temperature had significant effects on the resting oxygen consumption rate (M˙O(rest)), SDA and swimming performance (p<0.05). In addition, the effect of temperature differed significantly among the different species (interaction effect, p<0.05). Under the low temperature condition, digestion had no effect on either critical swimming speed (U(crit)) or the active MO(2) (MO(active)) for all fish species (additive metabolic mode). When the temperature was increased from 15 to 25°C, the metabolic scope (MS) for digestion increased approximately 182, 49 and 17%, and the MS for locomotion increased approximately 129, 58 and 138% in goldfish, common carp and qingbo, respectively. The total metabolic demands for both digestion and locomotion (i.e., the sum of digestive MS and locomotive MS) increased approximately 143, 56 and 112% in goldfish, common carp and qingbo, respectively. The total MS for both digestion and locomotion (the difference between MO(active) in fed fish and MO(rest) in fasting fish) increased approximately 106, 58 and 78% in goldfish, common carp and qingbo, respectively. Thus, the MS for locomotion in fed goldfish decreased due to the large increase in digestive function at the high temperature, and the U(crit) of fed goldfish decreased by 11% compared to that of fasting fish (p<0.05) (digestion-priory metabolic mode). The metabolic mode of qingbo changed to locomotion-priority mode, as illustrated by the large increase in locomotive MS in response to the increase in temperature. In the common carp, temperature had no effect on metabolic mode as illustrated by the parallel increases in cardio-respiratory capacity and metabolic capacity of digestive and locomotive organs. A discussion on the changes in metabolic mode in response to temperature and its possible relationship with the metabolic characteristics of a given fish species was also documented in this paper.

Critical swimming speed ( U crit) and rate of oxygen consumption of Pacific cod Gadus macrocephalus acclimated to 4 and 11° C were determined to assess the influence of water temperature on performance. The physiological effect of exercise trials on fish held at two temperatures was also assessed by comparing haematocrit and plasma concentrations of cortisol, metabolites and ions collected from fish before and after testing. The U crit of fish acclimated and exercised at 4° C did not differ from those acclimated and exercised at 11° C [1·07 body lengths (total length) s−1]. While the standard metabolic rate of 11° C acclimated fish was 28% higher than that of 4° C fish, no significant difference was observed between fish acclimated at the two temperatures. Plasma concentrations of cortisol, glucose and lactate increased significantly from pre- to post-swim in both groups, yet only concentrations of cortisol differed significantly between temperature treatments. Higher concentrations of cortisol in association with greater osmoregulatory disturbance in animals acclimated at the lower temperature indicate that the lower water temperature acted as an environmental stressor. Lack of significant differences in U crit between temperature treatments, however, suggests that Pacific cod have robust physiological resilience with respect to swimming performance within temperature changes from 4 to 11° C.

Fish need to balance their energy use between digestion and other activities, and different metabolic compromises can be pursued. We examined the effects of fasting (7days) on metabolic strategies in goldfish and common carp at different swimming levels. Fasting had no significant effect on swimming performance (U(crit)) of either species. Feeding and swimming profoundly elevated total ammonia (T(amm)) excretion in both species. In fed goldfish, this resulted in increased ammonia quotients (AQ), and additionally plasma and tissue ammonia levels increased with swimming reflecting the importance of protein contribution for aerobic metabolism. In carp, AQ did not change since oxygen consumption (MO(2)) and T(amm) excretion followed the same trend. Plasma ammonia did not increase with swimming suggesting a balance between production and excretion rate except for fasted carp at U(crit). While both species relied on anaerobic metabolism during exhaustive swimming, carp also showed increased lactate levels during routine swimming. Fasting almost completely depleted glycogen stores in carp, but not in goldfish. Both species used liver protein for basal metabolism during fasting and muscle lipid during swimming. In goldfish, feeding metabolism was sacrificed to support swimming metabolism with similar MO(2) and U(crit) between fasted and fed fish, whereas in common carp feeding increased MO(2) at U(crit) to sustain feeding and swimming independently.

Groups of common carp were acclimated to either 10°C or 28°C for 6 weeks. Fish were then exercised at 10°C or 20°C, and the critical swimming speed (fatigue velocity) was measured. At 10°C, cold-acclimated carp were capable of significantly higher swimming speeds. When exercised at 20°C. however, the situation was reversed, and warm-acclimated carp exhibited improved swimming ability. These results provide direct evidence that acclimation of the contractile proteins is beneficial across a wide temperature range. Following acclimation to low environmental temperatures the critical swimming speed exhibited a Q10 of only 1.1 for the temperature range 10–20°C. compared to a value of 2.9 for fish acclimated to the higher temperature.

The effects of acclimation temperature (30 degrees, 20 degrees, and 15 degrees C) and swimming speed on the aerobic fuel use of the Nile tilapia (Oreochromis niloticus; 8-10 g, 8-9-cm fork length) were investigated using a respirometric approach. As acclimation temperature was decreased from 30 degrees C to 15 degrees C, resting oxygen consumption (Mo2) and carbon dioxide excretion (Mco2) decreased approximately twofold, while nitrogenous waste excretion (ammonia-N plus urea-N) decreased approximately fourfold. Instantaneous aerobic fuel usage was calculated from respiratory gas exchange. At 30 degrees C, resting Mo2 was fueled by 42% lipids, 27% carbohydrates, and 31% protein. At 15 degrees C, lipid use decreased to 21%, carbohydrate use increased greatly to 63%, and protein use decreased to 16%. These patterns at 30 degrees C and 15 degrees C in tilapia paralleled fuel use previously reported in rainbow trout acclimated to 15 degrees C and 5 degrees C, respectively. Temperature also had a pronounced effect on critical swimming speed (UCrit). Tilapia acclimated to 30 degrees C had a UCrit of 5.63+/-0. 06 body lengths/s (BL/s), while, at 20 degrees C, UCrit was significantly lower at 4.21+/-0.14 BL/s. Tilapia acclimated to 15 degrees C were unable or unwilling to swim. As tilapia swam at greater speeds, Mo2 increased exponentially; Mo2min and Mo2max were 5.8+/-0.6 and 21.2+/-1.5 micromol O2/g/h, respectively. Nitrogenous waste excretion increased to a lesser extent with swimming speed. At 30 degrees C, instantaneous protein use while swimming at 15 cm/s ( approximately 1.7 BL/s) was 23%, and at UCrit (5.6 BL/s), protein use dropped slightly to 17%. During a 48-h swim at 25 cm/s (2.7 BL/s, approximately 50% UCrit), Mo2 and urea excretion remained unchanged, while ammonia excretion more than doubled by 24 h and remained elevated 24 h later. These results revealed a shift to greater reliance on protein as an aerobic fuel during prolonged swimming.

Transformation of malathion by Lysinibacillus sp. isolated from soil
An axenic bacterial strain, Lysinibacillus sp. KB1, was isolated from malathion-contaminated soil. It tolerated malathion up to 0.15?% and, under aerobic conditions, utilized it as sole carbon source. 20?% malathion and 47?% malaoxon were degraded out of the initially provided malathion. Two metabolites, mal-monocarboxylic acid and mal-dicarboxylic acid, were detected within 7?days at 30?°C. Esterase activity of the strain was 240?±?2.5?U/ml after 7?days of growth. Sterilized soil mixed with malathion showed rapid degradation of malathion when inoculated with strain KB1 as compared to the uninoculated soil.

Different species respond differently to environmental change so that species interactions cannot be predicted from single-species performance curves. We tested the hypothesis that interspecific difference in the capacity for thermal acclimation modulates predator-prey interactions. Acclimation of locomotor performance in a predator (Australian bass, Macquaria novemaculeata) was qualitatively different to that of its prey (eastern mosquitofish, Gambusia holbrooki). Warm (25°C) acclimated bass made more attacks than cold (15°C) acclimated fish regardless of acute test temperatures (10-30°C), and greater frequency of attacks was associated with increased prey capture success. However, the number of attacks declined at the highest test temperature (30°C). Interestingly, escape speeds of mosquitofish during predation trials were greater than burst speeds measured in a swimming arena, whereas attack speeds of bass were lower than burst speeds. As a result, escape speeds of mosquitofish were greater at warm temperatures (25°C and 30°C) than attack speeds of bass. The decline in the number of attacks and the increase in escape speed of prey means that predation pressure decreases at high temperatures. We show that differential thermal responses affect species interactions even at temperatures that are within thermal tolerance ranges. This thermal sensitivity of predator-prey interactions can be a mechanism by which global warming affects ecological communities.

Summary The effect of hypoxia was studied in cold (15°C) and warm (30°C) acclimated goldfish. The hypoxic thresholds, defined as the lowest sustainablePO2 were found to be 1.6 and 4.0 kPa O2 at, respectively, 15°C and 30°C. At these levels the fish did not loose either weight or appetite over a 2-months period. While during starvation under normonic conditions a significant weight loss and breakdown of lactate dehydrogenase (90%) was observed, no such changes were found in fed hypoxic animals. In red lateral muscle, white epaxial muscle and liver of goldfish from 4 differently acclimated groups the maximal activities were measured of: glycogen phosphorylase, hexokinase, malate dehydrogenase, glycerol-3-P dehydrogenase, glucose-6-P dehydrogenase, malic enzyme, succinate oxidase, pyruvate carboxylase, phosphoenol-pyruvate carboxykinase, fructose-bisphosphatase and glucose-6-phosphatase. Thermal compensation, according to Precht's typology, was predominantly observed in red muscle and to a lesser extent in white muscle. The liver glucose-6-P dehydrogenase showed a strong inverse response, which points to enhanced synthetic activity at the higher temperature. Hypoxia acclimation exerted weaker responses at 15°C than at 30°C. Changes in liver enzyme activities suggest depressed protein synthesis and enhanced gluconeogenesis in hypoxic animals. In muscle of 30°C-acclimated goldfish hypoxia induces a significant increase of succinate oxidase activity, indicating adaptation of the aerobic energy metabolism. The occurrence of pyruvate carboxylase, never before observed in vertebrate muscle, probably plays an important role in pyruvate catabolism. Because its action produces oxalo-acetate, the enzyme may stimulate pyruvate oxidation and thus prevent early lactate accumulation. Since all gluconeogenic enzymes were shown to be active in goldfish muscle, the possible occurrence of gluconeogenesis in muscle (albeit at low rate) must be accepted. Enzyme activities in goldfish muscle were compared with literature data for a number of other fish species. This comparison indicates that maximal glycolytic flux in goldfish muscle tissue is rather low, although muscular glycogen levels are very high. It is suggested that this is part of the gold-fish's strategy to cope with hypoxia.

A study has been made of the maximum sustained swimming speed of Crucian carp Carassius carassius (L.) using a fixed velocity technique. The data obtained from swimming tests on 214 carp have been analysed using the method of probit analysis. The 50% fatigue level for 13–16 cm fish acclimated to 9.5±0.6°C has been estimated to be 3.35 lengths/sec. Biochemical measurements have been made on the red and white myotomal muscles and liver of fish subjected to both varying intensities of sustained swimming and short periods of vigorous swimming. Free creatine was found to increase only during high speed swimming in the white muscle. Elevated lactate concentrations occurred at both low and high sustained swimming speeds in the red superficial muscle but not during short periods of strenuous exercise. Glycogen depletion from the red musculature also only took place at the sustained swimming speeds investigated. The reverse situation was operative in the white muscle, significant glycogen depletion occurring only at the highest swimming speed studied. Lactate levels were only significantly different from non-exercised fish in the fish swimming at the higher velocities. The effects of periods of recovery following 200 min of sustained swimming were also investigated. White muscle lactate was at a higher level than non-exercise fish 5 h post-exercise, while both red muscle glycogen and lactate rapidly returned to pre-exercise concentrations. Biochemical measurements on the myotomal muscle types have been discussed in relation to the swimming performance of the fish and the division of labour between red and white fibres.

A flow-through respirometer and swim tunnel was used to estimate the gait transition speed ( U p-c) of striped surfperch Embiotoca lateralis , a labriform swimmer, and to investigate metabolic costs associated with gait transition. The U p-c was defined as the lowest speed at which fish decrease the use of pectoral fins significantly. While the tail was first recruited for manoeuvring at relatively low swimming speeds, the use of the tail at these low speeds [as low as 0·75 body (fork) lengths s−1, L F s−1) was rare (<10% of the total time). Tail movements at these low speeds appeared to be associated with occasional slow manoeuvres rather than providing power. As speed was increased beyond U p-c, pectoral fin (PF) frequencies kept increasing when the tail was not used, while they did not when PF locomotion was aided by the tail. At these high speeds, the tail was employed for 40–50% of the time, either in addition to pectoral fins or during burst-and-coast mode. Oxygen consumption increased exponentially with swimming speeds up to gait transition, and then levelled off. Similarly, cost of transport ( C T) decreased with increasing speed, and then levelled off near U p-c. When speeds ≥ U p-c are considered, C T is higher than the theoretical curve extrapolated for PF swimming, suggesting that PF swimming appears to be higher energetically less costly than undulatory swimming using the tail.
