Models of the effects of atmosphericN deposition in forested watersheds have notadequately accounted for the effects of aquatic andnear-stream processes on the concentrations and loadsof NO in surface waters. This studycompared the relative effects of aquatic andnear-stream processes with those from the terrestrialecosystem on the retention and transport ofNO in two contrasting stream reaches ofthe Neversink River, a forested watershed in theCatskill Mountains of New York that receives among thehighest load of atmospheric N deposition in thenortheastern United States. Stream water samples werecollected every two hours and ground-water andtributary samples were collected daily at base flowconditions during four 48-hour periods from April toOctober 1992, and NO mass balances werecalculated for each site. Results indicated diurnalvariations in stream NO concentrations inboth reaches during all four sampling periods; this isconsistent with uptake of NO byphotoautotrophs during daylight hours. Mass-balanceresults revealed significant stream reach losses ofNO at both sites during all samplingperiods. The diurnal variations in NO concentrations and the retention of NO relative to terrestrial contributions to the streamreaches were greater downstream than upstream becausephysical factors such as the head gradients ofinflowing ground water and the organic matter contentof sediment are more favorable to uptake anddenitrification downstream. The mass retention ofNO increased as the mean 48-hr streamdischarge increased at each site, indicating that theresponsible processes are dependent onNO supply. Low stream temperatures duringthe April sampling period, however, probably reducedthe rate of retention processes, resulting in smallerlosses of NO than predicted from streamdischarge alone. Water samples collected from thestream, the hyporheic zone, and the alluvial groundwater at sites in both reaches indicated that the neteffect of hyporheic processes on downstreamNO transport ranged from conservativemixing to complete removal by denitrification. Therelative effects of biological uptake anddenitrification as retention mechanisms could not bequantified, but the results indicate that bothprocesses are significant. These results generallyconfirm that aquatic and near-stream processes causesignificant losses of NO in the NeversinkRiver, and that the losses by these processes atdownstream locations can exceed the NO contributions to the stream from the terrestrialenvironment during summer and fall base-flowconditions. Failure to consider these aquatic andnear-stream processes in models of watershed responseto atmospheric N deposition could result inunderestimates of the amount of NO leaching from forested ecosystems and to an inabilityto unequivocally relate geographic differences inNO concentrations of stream waters tocorresponding differences in terrestrial processes.  相似文献

During the last 60 years, pollution of the groundwater with has greatly increased in many parts of Europe, as a consequence of excessive use of manure and synthetic fertilisers. Monitoring of groundwater-fed wetlands indicated that sediments with high concentrations had the lowest Fe and concentrations in the pore water. A comparison of two restored open water fens, differing in supply via the groundwater, indicated that the redox potential and the sulphate ( ) reduction rate were lower when the groundwater contained not only but also high concentrations. The lower reduction rates in the -rich open water fen were associated with lower concentrations and the presence of plant species characteristic of clear water. In contrast, the higher reduction rates in the -poor open water fen were associated with very high concentrations and massive development of plant species characteristic of eutrophic environments. Investigations at -rich seepage sites in black alder carrs, showed that high concentrations in the pore water caused chlorosis in the alder carr vegetation, due to lower availability of Fe in the pore water and less Fe uptake by the plants. Experimental desiccation of sediments proved that the -rich seepage sites contained no oxidisable FeS x , contrary to -poor locations, which became acidified and mobilised extremely high amounts of due to FeS x oxidation. A laboratory experiment showed that addition to sediments led to reduced releases of Fe, and S2–, very likely due to the oxidation of reduced Fe and S compounds. Overall, the results confirmed that is an energetically more favourable electron acceptor in anaerobic sediments than Fe and , and that high loads function as a redox buffer, preventing reduction of Fe and . Limited reduction prevents S2– -mediated mobilisation of from Fe- complexes. At a higher redox potential, reduced Fe, including FeS x , was oxidised, increasing the content of Fe(III) capable of binding . This prevented increased availability and the concomitant massive development of plant species characteristic of eutrophic environments.  相似文献

Avena sativa (oats) and Lycopersiconesculentum (tomatoes) were grown in dilute nutrientsolutions supplemented with Al only, F only or acombination of both. In solutions containing Al andF, shoot growth was limited when predicted Alr(Al3+, AlOH2+ and Al(OH) )activities were < 0.1 M, activities three orders ofmagnitude lower than the critical value determinedwith Al only. The data suggest that at the activitiesused in these experiments, Alr is most toxic,AlF2+ and AlF are toxic to a lesserextent, and AlF3, AlF and F-are least toxic. Fluoride concentrations in shootsgrown in solutions correlated best with positivelycharged AlF species (i.e.AlF2+, AlF ) and the molar ratio ofF:Al in most plants shoots was about 3:2. However,when activities of positively chargedAlF species were low (< 50 M)and theactivity of AlF3 species high (500 M) the molarratio of F:Al in plant shoots suggested AlF3 wastaken up. These findings are discussed in relation toplant uptake and toxicity. Measured concentrations of Alr and F- insolutions containing Al and F were compared withconcentrations predicted by a computer model(GEOCHEM-PC). The method for measuring F-concentrations, using a non-complexing buffer and Fion-selective electrode, gave good agreement withpredicted F- concentrations. The8-hydroxyquinoline method for measuring Alrconcentrations did not agree with predictedconcentrations, highlighting the limitations of thismethod when measuring Alr in the presence of F ina multi-ligand system with high concentrations of Fand Al.  相似文献

A long-term field experiment was initiated to simulate chronic atmospheric N deposition, a widespread phenomenon in industrial regions of the world. Eight years of experimental nitrate ( ) additions (3 g -N m–2 per year) to four different northern hardwood forests located along a 500 km geographic gradient dramatically increased leaching losses of -N, dissolved organic carbon (DOC), and dissolved organic nitrogen (DON). During the last two water years, the average increase in solution -N and DON leaching from the -amended plots was 2.2 g N m–2, equivalent to 72% of the annual experimental N addition. Results indicate that atmospheric N deposition may rapidly saturate some northern hardwood ecosystems across an entire biome in the upper Great Lakes Region of the USA. Changes in soil C and N cycling induced by chronic N deposition have the potential in this landscape to significantly alter the flux of DOC and DON from upland to aquatic ecosystems. Michigan Gradient study site characteristics are similar to those of European forests most susceptible to N saturation.  相似文献

A small catchment on the Swedish West Coast has been studied over four years to determine S dynamics by using S isotope ratios. A Norway spruce dominated forest covers the catchment, and small peat areas occur in the lower parts of the catchment. The runoff values varied both during the year, and from year to year. Over the period from February 1990 to December 1993, the values ranged from — 1%. to +11%. Over the same period, the throughfall values ranged from +1%. to +15%. There was no correlation (r 2= 0.01; Pr(F)=0.57) between values in throughfall and runoff. Since the only input of S to the catchment is atmospheric deposition, the long-term runoff S mass flux is controlled by the deposition. Therefore, processes in the catchment are responsible for the variation in the runoff values. During periods with enriched runoff, bacterial dissimilatory SO 4 2– reduction occurs in the catchment. After very dry periods, oxidation of this reduced S, which is32S-enriched, can be traced in runoff. Previous studies of the catchment have not been able to distinguish between: 1) oxidation of reduced S and dry deposition, and 2) reduction and adsorption. From the current study, it can be concluded that adsorption and dry deposition cannot cause the observed variation in runoff .  相似文献

Summary Resting rates of O2 consumption against , exercise endurance times and during recovery from vigorous exercise were measured inSceloporus occidentalis captured near sea level and inS. graciosus captured above 2850 m. Oxygen consumption against was also measured inS. occidentalis captured above 2850 m. When was recorded continuously, as ambient was slowly reduced from 155 Torr, it became directly dependent upon ambient between 110 and 120 Torr. The critical for the high altitude lizards was lower than that for the lowland lizards, which enabled the former to maintain relatively higher 's when ambient was reduced below 120 Torr. The high altitude lizards also had significantly greater endurance when stimulated to exercise at 1600 m ( 130 Torr). Both the higher under hypoxia and the greater endurance roughly parallel a significantly greater maximum in the high altitude lizards. At a simulated altitude of 3600 m ( 100 Torr), maximum and rate of recovery of the O2 debt calculated from post active were significantly reduced in the lowland but not the high altitude lizards. The effects of simulated altitude conditions on the lowland but not the mountaine animals indicate adaptations to altitude in these sceloporine lizards. We did not find any consistent relationship between organ/body weight ratios or hematocrit and our measures of endurance or the altitude at which the lizards were captured.  相似文献

In aqueous medium etiolated wheat seedlings release superoxide anion ( ). Interaction of a synthetic antioxidant, butylated hydroxytoluene (BHT, ionol), with oxygen in the aqueous medium is accompanied by formation. This suggests that under certain conditions BHT behaves as a prooxidant. A natural antioxidant, superoxide dismutase (SOD), and also a wound healing preparation, emulsified denatured placenta (EDP), do not exhibit the prooxidant properties. A simple linear relationship between the J coupling constant and the linewidth (1/2) of in-phase NMR peaks has been identified. This relationship permits the rapid and accurate determination of polypeptide J coupling constants from a simple inspection of amide cross peaks in homonuclear 1H TOCSY or 1H NOESY spectra. By using the appropriate set of processing parameters we show that J = 0.5(1/2) – MW/5000 + 1.8 for TOCSY spectra and J = 0.6(1/2) – MW/5000 – 0.9 for NOESY spectra, where 1/2 is the half-height linewidth in Hz and MW is the molecular weight of the protein in Da. The simplicity of this relationship, combined with the ease with which 1/2 measurements can be made, means that J coupling constants can now be rapidly determined (up to 100 measurements in less than 30 min) without the need for any complex curve-fitting algorithms. Tests on 11 different polypeptides involving more than 650 separate J measurements have shown that this method yields coupling constants with an rmsd error (relative to X-ray data) of less than 0.9 Hz. Furthermore, the correlation coefficient between the predicted NMR coupling constants and those derived from high-resolution X-ray crystal structures is typically better than 0.89. These simple linear relationships have been found to be valid for peptides as small as 1 kDa to proteins as large as 20 kDa. Despite the method's simplicity, these results are comparable to the accuracy and precision of the best techniques published to date.  相似文献

Gross and net nitrogen (N) ammonification and nitrification were measured in soils from an uncut and recently cut upland and peatland conifer stand in northwestern Ontario, Canada. Rates of gross total inorganic N immobilization were similar to gross mineralization, resulting in low net mineralization rates in soils from all four upland and peatland conifer stands. Gross ammonification rates were variable but similar in soils from uncut and cut peatland hollows (18–19mgNkg–1day–1) and upland forest floor soils (14–19mgNkg–1day–1). Gross ammonium ( ) immobilization rates were also variable but similar to ammonification rates. Median gross nitrification rates were within 0–2mgNkg–1day–1 in soils from all four upland and peatland cut and uncut stands, although rates were consistently higher for the soils from the cut stands. Large variability in gross nitrification rates were observed in peatland soils, however the highest gross nitrification rates were measured in saturated peatland soils. Net rates remained low in the soils from all four stands due to high nitrate ( ) immobilization and very fast turnover (<0.2 day). Our results suggest that potential losses may be negated by high immobilization in uncut and cut boreal forest stands. This study reveals the potential for the interaction of N production and consumption processes in regulating N retention in upland and peatland conifer forests, and the resilience of the boreal forest to disturbance.  相似文献

The tetrameric heart isozyme of lactate dehydrogenase (H4) is modified by p-chloromercuribenzoate (PCMB) to produce the inactive tetramer and then hybridized with native tetrameric muscle isozyme (M4). The hybrid mixture was isolated by polyacrylamide gel electrophoresis (PAGE) and then stained for enzyme activity and with Coomassie brilliant blue. Only three bands were found on the gels in either case. The hybrid enzymes as isolated by PAGE have half the specific activity of the native muscle enzyme. The electrophoresis properties of HM3 are very similar to those of HM3, while the electrophoresis properties of are very similar to those of H2M2. The above results strongly suggest that the tetramer having enzymatic activity contains at least two native subunits, and the di-subunit in the tetrameric enzyme is the minimal functional unit.  相似文献

Canadian populations of D. radicum differ in their response to temperature during postdiapause development. This population varied from 31 to 90% early type over a 10-year period and the rate of postdiapause development at 20 °C was directly related to the percentage early. In the year with the most rapid development (90% early), development was significantly slower than in the populations from other locations with predominantly early populations, and the year with the slowest development (31% early) showed significantly faster development than that from Kildare, Prince Edward Island (100% late). Therefore the parameters for early and late types of development will not be accurate for use in mixed populations, and the parameters in mixed populations will change among years. Populations of D. radicum in North America and Europe (67 locations by years) varied from 0–100% early. At Winnipeg, the percentage early was directly related to the annual temperature accumulation ( ) during the growing season. The calculation of developmental parameters for the early-emergers of mixed populations provides a more accurate basis for estimating the times of first emergence and the first peak of emergence than parameters based on the whole population. Since postdiapause developmental rates vary both among and annually within locations, developmental models should be designed to include such variations.  相似文献

The effect of inhibitors of the cytochrome pathway and alternative oxidase on the rate of respiration and generation of reactive oxygen species by pea mitochondria was studied. Respiration of mitochondria from pea cotyledons was inhibited by 70-80% by salicylhydroxamate (SHAM). The rate of hydrogen peroxide production by pea cotyledon mitochondria during succinate oxidation was 0.15 nmol/min per mg protein. SHAM considerably accelerated the hydrogen peroxide production. The SHAM-dependent H2O2 production was stimulated by 2 M antimycin A and inhibited by 5 mM KCN and 1 M myxothiazol. The study of the rate of generation by pea mitochondria using EPR spin traps and epinephrine oxidation showed that H2O2 accumulation can be accounted for by a significant increase in the rate of production.  相似文献

In the biosphere plants are exposed to different forms of N, which comprise mineral and organic N forms in soils as well as gaseous NH3, NOx, and molecular N2 in the atmosphere. The form of N uptake is mainly determined by its abundance and accessibility, which make and the most important N forms for plant nutrition under agricultural conditions. With minor importance, the form of N uptake is also subject to plant preferences, by which plants maintain their cation/anion balance during uptake. However, some species seem to have an obligatory preference which even prevents their growth on certain other N sources. In general, uptake of a certain N form closely matches the growth-related demand of the plant, at least when N transport to the root surface is not limiting. In addition, many plants accumulate large pools of N during vegetative growth which are remobilized in the generative stage. As a consequence, systems responsible for N transport need to be tightly regulated in their expression and activity upon sensing N availability and plant demand. Employing the tools of molecular genetics, the first plant genes encoding transporters for inorganic N have recently been isolated and characterized. These data can now complete the wealth of physiological and nutritional studies on N uptake. The present article will focus on the uptake of and into root cells and tries to link data derived from physiological, genetic and molecular studies.  相似文献

The vertical distribution of dissolved greenhouse gases (CH4,CO2 and N2O), NO ,and 13C of CO2 in Lake Biwa during a stagnantperiod was precisely determined. CO2 as well as NO was accumulated in the hypolimnion, whereas NO and CH4concen\-trations were generally higher in theepilimnion than in the hypolimnion. In August, NO andCH4 were ephemerally accumulated at the thermocline. Theconcentration of CH4 always exceeded equilibrium with respectto air/water exchange. N2O was rather uniformly distributed inboth time and space, and remained near equilibrium with respect toair/water exchange. All of these observations are similar to otherstratified, oligotrophic lakes, in which the hypolimnia were welloxygenated. The 13C of CO2 became morenegative with increasing depth, and showed a strong negativecorrelation with apparent oxygen utilization. From the data, the 13Cvalue of organic matter decomposed into CO2 inthe hypolimnion was calculated by isotope mass-balance, and found tobe in a similar range to 13C of phytoplankton and benthic algaeand distinctively higher than 13C of both terrestrial andsedimentary organic matters. This suggests that autochthonous organicmatter was the major source of CO2.  相似文献

Summary Oxygen binding properties of the hemocyanin-containing blood ofBuccinum undatum were examined in vitro and in vivo under normoxic ( 150 mmHg) and hypoxic ( 50 mmHg) conditions at 10°C. Blood pH and showed a decrease in vivo under hypoxic conditions. Oxygen uptake at high water , was about 18 ml O2/kg·h (wet weight) and the critical oxygen tension between 25 and 50 mm Hg. In vitro the O2 binding to hemocyanin showedn-values independent of pH, while both O2 affinity and oxygen carrying capacity were strongly pH dependent. Oxygen affinity increased below pH=8.1 and thus showed a pronounced reversed Bohr shift in the physiological pH range (7.5<pH<8.1). The oxygen carrying capacity similarly increased markedly with falling pH in the physiological pH range (reversed Root shift). Astrup titration curves showed a metabolic and respiratory acidosis under hypoxic conditions ( 50 mm Hg). The role of hemocyanin in the transport of oxygen in relation to ambient O2 availability is discussed.  相似文献

Summary In the intertidal shore crab,Carcinus maenas, pH and values of the prebranchial venous hemolymph, and , the PO2 values of the arterialized cardiac hemolymph, , were measured, and the ventilatory activity was assessed by measuring the hydrostatic pressures at the exits of the epibranchial cavities, under four environmental conditions: normoxic water, normoxic air, hypoxic gas, hyperoxic gas.In the crab breathing normoxic air, was lower and higher than in animals breathing normoxic water. With the switch to hypoxic gas, ventilation increased and and decreased. With the switch to hyperoxic gas, ventilation decreased, and increased and decreased.Thus these crabs breathing air were not as well oxygenated as when breathing air-equilibrated water, and because of their low , they were relatively hypervetilating and hypocapnic compared to air-breathing vertebrates. Hyperoxic breathing, increasing and reducing ventilatory drive, led to increased . Conversely, was reduced by hypoxic breathing. These observations suggest that the gas exchanger of intertidal crabs is not as successfully designed for air breathing as that of land-colonizing insects and air-breathing vertebrates.  相似文献

Summary Nest humidity ( ) under an incubating bantam hen was measured at ambient ranging from 1.3 to 25.0 mmHg. Weight loss of eggs was recorded in clutches of varying size. Nest and ambient were also measured in wild incubating willow ptarmigan nests in dry and wet habitats.Nest increased linearly with ambient in a way predictable on the assumption that the water vapour conductance ( ) of brood patch skin, plumage and eggs were constant and independent of ambient . Nest was also dependent of clutch size. Egg dehydration was quantitatively predicted from measured values and the laws of diffusion.Our findings confirm earlier conclusions that the adult bird does not actively regulate nest at varying ambient . The aim of this study was to measure running times to exhaustion (Tlim) on a treadmill at 100% of the minimum velocity which elicits max max in 38 elite male long - distance runners max = 71.4 ± 5.5 ml.kg–1.min–1 and max = 21.8 ± 1.2 km.h–1). The lactate threshold (LT) was defined as a starting point of accelerated lactate accumulation around 4 mM and was expressed in max. Tlim value was negatively correlated with max (r = -0.362, p< 0.05) and max (r = –0.347, p< 0.05) but positively with LT (%v max) (r = 0.378, p < 0.05). These data demonstrate that running time to exhaustion at max in a homogeneous group of elite male long-distance runners was inversely related to max and experimentally illustrates the model of Monod and Scherrer regarding the time limit-velocity relationship adapted from local exercise for running by Hughson et al. Summary The genetic behaviour of a human serum -lipoprotein factor, called Ag(a1), was studied by agar micro-diffusion technique, utilizing an antibody detected in the serum from a transfused thalassemia patient. It behaves as an inherited, dominant, autosomal character, with complete penetrance at birth. It is controlled by a gene and is closely linked to the Ag x and Ag y genes.The existence of a gene Ag b , allelic to , is postulated but the Ag(b) antigen has not so far been detected by specific antisera.The frequency of the gene in a Milan population was found to be 0,43, and in a Berne population was 0,46. The frequencies of the four possible gene combinations in the sample group from Milan were: Ag yb =0,53; =0,22; =0,21; Ag xb =0,04.The observed frequencies of the factor Ag(a1) were 0,676 and 0,713, respectively among the Milan and Berne populations.  相似文献

Summary The resting oxygen consumption and breathing pattern of nine newborn and adult species (ranging in body size from mouse to human) have been compared on the basis of data collected from the literature. Minute ventilation is similarly linked to at both ages, the percent of extracted as O2 about 2.2. Tidal volume/kg is an interspecies constant in newborns and adults, approximately 8 ml/kg. Breathing frequency decreases with the increase in size in a different way at the two ages: large species have newborns breathing at rates 2–3 times above the corresponding adults' values, while in the small species newborns and adults breathe at almost the same rate. Therefore the newborns of the smallest species have both and below the expected values, implying a greater inability to cope with the external demands than newborns of larger species. Several considerations indicate that in the smallest newborns the mechanical properties of the respiratory system could be a constraint to resting ventilations larger than observed. It is therefore possible that their low is the cause, and not the effect, of the relatively small .  相似文献
