The aim of this study was to (a) investigate the influence of tackling on repeated-sprint performance; (b) determine whether repeated-sprint ability (RSA) and repeated-effort ability (REA) are 2 distinct qualities; and (c) assess the test-retest reliability of repeated-sprint and repeated-effort tests in rugby league. Twelve rugby league players performed a repeated-sprint (12 × 20-m sprints performed on a 20-second cycle) and a repeated-effort (12 × 20-m sprints with intermittent tackling, performed on a 20-second cycle) test 7 days apart. The test-retest reliability of these tests was also established. Heart rate and rating of perceived exertion were recorded throughout the tests. There was a significantly greater (p ≤ 0.05) and large effect size (ES) differences for total sprint time (ES = 1.19), average heart rate (ES = 1.64), peak heart rate (ES = 1.35), and perceived exertion (ES = 3.39) for the repeated-effort test compared with the repeated-sprint test. A large difference (ES = 1.02, p = 0.06) was detected for percentage decrement between the 2 tests. No significant relationship was found between the repeated-sprint and repeated-effort tests for any of the dependent variables. Both tests proved reliable, with total sprint time being the most reliable method of assessing performance. This study demonstrates that the addition of tackling significantly increases the physiological response to repeated-sprint exercise and reduces repeated-sprint performance in rugby league players. Furthermore, RSA and REA appear to be 2 distinct qualities that can be reliably assessed with total time being the most reliable measure of performance.  相似文献

The purposes of this investigation were to establish the reliability of selected physical performance tests in women athletes and nonathletes and to determine performance differences between groups. Fifty women (25 athletes, 25 nonathletes) performed 5 tests in 2 sessions. The performance tests included the figure-eight hop test, up-and-down hop test, side-to-side hop test, hexagon hop test, and zigzag run test. Intraclass correlation coefficients (ICC [2, 1]) were calculated for trial-to-trial, intertester, and day-to-day reliability. Independent t-tests with Bonferroni adjustment (alpha = 0.01) were used for each individual test to compare differences between groups. All tests showed good reliability values (ICC > or = 0.76) in the nonathlete group for all conditions and varied reliability values (0.48-0.99) among conditions in the athlete group. The independent t-tests showed a statistically significant group effect (t > or = 3.041; p < or = 0.004) for all tests. The results showed that these physical performance tests are reliable measurement tools in the female population.  相似文献

Mitochondrial DNA (mtDNA) control-region sequences and microsatellite loci length polymorphisms were used to estimate phylogeographical patterns (historical patterns underlying contemporary distribution), intraspecific population structure and gender-biased dispersal of Phocoenoides dalli dalli across its entire range. One-hundred and thirteen animals from several geographical strata were sequenced over 379 bp of mtDNA, resulting in 58 mtDNA haplotypes. Analysis using F(ST) values (based on haplotype frequencies) and phi(ST) values (based on frequencies and genetic distances between haplotypes) yielded statistically significant separation (bootstrap values P < 0.05) among most of the stocks currently used for management purposes. A minimum spanning network of haplotypes showed two very distinctive clusters, differentially occupied by western and eastern populations, with some common widespread haplotypes. This suggests some degree of phyletic radiation from west to east, superimposed on gene flow. Highly male-biased migration was detected for several population comparisons. Nuclear microsatellite DNA markers (119 individuals and six loci) provided additional support for population subdivision and gender-biased dispersal detected in the mtDNA sequences. Analysis using F(ST) values (based on allelic frequencies) yielded statistically significant separation between some, but not all, populations distinguished by mtDNA analysis. R(ST) values (based on frequencies of and genetic distance between alleles) showed no statistically significant subdivision. Again, highly male-biased dispersal was detected for all population comparisons, suggesting, together with morphological and reproductive data, the existence of sexual selection. Our molecular results argue for nine distinct dalli-type populations that should be treated as separate units for management purposes.  相似文献

An F1 individual derived from a cross between two distinct lines of spring oilseed rape (Brassica napus) was used to produce a pair of complementary backcross populations, each consisting of 90 individuals. The F1 donated male gametes to the Male population and female gametes to the Female population. Genetic maps were generated from both populations and aligned using 117 common loci to form an integrated genome map of B. napus with 243 RFLP-defined loci. A comparison of the frequency and distribution of crossovers in the two populations of F1 gametes (assayed in the Male and Female populations) detected no differences. The genetic maps derived from the Male and Female populations each consisted of 19 linkage groups spanning 1544 and 1577 cM, respectively. The maps were aligned with other B. napus maps, and all 19 equivalent linkage groups were unambiguously assigned. The genetic size and general organisation of the new maps were comparable with those of pre-existing B. napus maps in most respects, except that the levels of polymorphism in the constituent A and C genomes were unusually similar in the new cross.  相似文献

Twelve diverse strains of Drosophila melanogaster have been examined with respect to their individual fitness components and with respect to their relative performance under competitive and noncompetitive conditions. Individual fitness components included estimates of time until successful copulation (t), fecundity (f) and egg-to-adult viability (v), and a composite index of overall fitness of the form fv/t was used for comparisons among strains. Noncompetitive performance was assessed in terms of the biomass (standing crop) and productivity of equilibrium experimental populations. Competitive performance was assessed in terms of relative competitive ability vis-à-vis a standard compound-autosome-bearing strain in single-generation tests. A significant correlation was found between the composite index of individual fitness components and the competitive compound-autosome test. Although the biomass and productivity of equilibrium populations were correlated with each other, neither of these noncompetitive measures was correlated with individual fitness components or with the composite index. We suggest that the performance of strains in such noncompetitive tests may be related to what Wright has called the "mean selective value" of the populations. Judging from their association with the composite index of individual fitness components, competitive tests such as the compound-autosome test seem to be related more nearly to the average Darwinian fitness of the populations.  相似文献

Susceptibility of the sugarcane borer, Diatraea saccharalis (F.), to tebufenozide was measured using a feeding bioassay, and values obtained were compared with baselines generated before the use of this insecticide in Louisiana sugarcane, Saccharum spp. Results from our study suggest that susceptibility to tebufenozide is decreasing in field-collected sugarcane borers. Inflections in the log dosage-probit lines were detected for many of the field collections, indicating increased heterogeneity within these populations in response to tebufenozide. Where appropriate, probit transformation was used to estimate susceptibility, and significant differences (1.6 - 2.7-fold) were measured in LC50 values between some field-collected cohorts and the previously measured baseline. In addition, a discriminating concentration (0.5 ppm) was used to estimate resistance frequencies in cohorts for which probit transformation was not appropriate. Results from these tests suggest that frequencies of resistance were high (49% in one cohort) in populations from some locations. Lighter weight pupae of the survivors from one of the more resistant cohorts suggests that tebufenozide resistance mechanisms may have a biological cost in terms of ecological fitness at early stages of resistance development. As a result of continued resistance monitoring, alternation of management chemistry is expected to help preserve this valuable sugarcane integrated pest management tactic.  相似文献

The architecture of the pod wall and dehiscence zone (DZ) was studied in populations of a resynthesized, shatter-resistant, oilseed rape line, DK142, and the commercial cultivar Apex. The dimensions of the pod wall and its component tissues were significantly larger in DK142. However, the variation in the pod architecture of Apex, DK142 and F2 populations derived from crosses of DK142 and Apex was found to have little or no role in pod shatter. By contrast, variation in the dimensions of the DZ characters correlated strongly and positively with shatter resistance. The size of the main vascular bundle (MVBV) of DK142 as it exited the valve and joined the vascular tissue of the replum was, on average, 60% larger than in Apex, the DZ was 40% wider and there was a high preponderance of vascular tissue other than the MVBV. The variation in the size of the MVBV accounted for much of the variation in shatter resistance of all populations, including shatter-susceptible Apex. The DZ width was also found to be important in explaining the limited range of shatter values in Apex, but in populations of DK142 and F2, where the amount of vascular intrusion into the DZ was much greater, the variation in DZ width was not important. The importance of the vascular tissue to shatter resistance was further highlighted by a novel microfracture test (MFT). By contrast, no significant difference between DK142 and Apex in the ease of separation of the thin-walled DZ cells was detected using the MFT.  相似文献

The existence of monoecious and dioecious populations within plant species is rare. This limits opportunities to investigate the ecological mechanisms responsible for the evolution and maintenance of these contrasting sexual systems. In Sagittaria latifolia, an aquatic flowering plant, monoecious and dioecious populations exist in close geographic proximity but occupy distinct wetland habitats differing in the relative importance of disturbance and competition, respectively. Life-history theory predicts contrasting evolutionary responses to these environmental conditions. We propose that the maintenance of monoecy and dioecy in S. latifolia is governed by ecological selection of divergent life-history strategies in contrasting habitats. Here we evaluate this hypothesis by comparing components of growth and reproduction between monoecious and dioecious populations under four conditions: natural populations, a uniform glasshouse environment, a common garden in which monoecious and dioecious populations and their F1 progeny were compared, and a transplant experiment using shaded and unshaded plots in a freshwater marsh. Plants from dioecious populations were larger in size and produced heavier corms in comparison with monoecious populations. Monoecious populations flowered earlier and produced more flowers, clonal ramets, and corms than dioecious populations. The life-history differences between the sexual systems were shown to have a quantitative genetic basis, with F1 progeny generally exhibiting intermediate trait values. Survival was highest for each sexual system in field plots that most closely resembled the habitats in which monoecious (unshaded) and dioecious (shaded) populations grow. These results demonstrate that monoecious and dioecious populations exhibit contrasting patterns of investment in traits involved with growth and reproduction. Selection for divergent life histories between monoecious and dioecious populations of S. latifolia appears to be the principal mechanism maintaining the integrity of the two sexual systems in areas of geographic overlap.  相似文献

During a study aimed at the specificity of binding of phosphorylase and glycogen synthase to glycogen particles we investigated the separation of these particles with the technique of high-performance liquid chromatography using several media. The technique allowed relatively rapid fractionations with little, if any, loss of enzyme activity. The basis of some separations was molecular weight of the particle. Of the molecular exclusion media used, Superose 6 had an exclusion limit large enough to accommodate most glycogens but excluded mouse liver and bovine liver glycogen particles. Toyopearl HW-75 F was a medium that could accommodate liver glycogen and gave us reasonably well-defined separation of the particles that carried phosphorylase and glycogen synthase. Separations on ion-exchange columns, that were obviously based on the overall charge of proteins associated with the glycogen particles, were not very effective in terms of allowing the separation of different types of particle. Of all the media used the hydrophobic interaction column was best in terms of separating what appeared to be distinct populations of particles that had definite preferences in terms of binding phosphorylase and glycogen synthase. As expected, these separations were based on the hydrophobic interaction of the proteins associated with the glycogen particles, since particles that had been deproteinized by a cold water extraction procedure did not bind to the column.  相似文献

Intra- and inter-population genetic variability and the demographic history of Heliothis virescens (F.) populations were evaluated by using mtDNA markers (coxI, coxII and nad6) with samples from the major cotton- and soybean-producing regions in Brazil in the growing seasons 2007/08, 2008/09 and 2009/10. AMOVA indicated low and non-significant genetic structure, regardless of geographical scale, growing season or crop, with most of genetic variation occurring within populations. Clustering analyzes also indicated low genetic differentiation. The haplotype network obtained with combined datasets resulted in 35 haplotypes, with 28 exclusive occurrences, four of them sampled only from soybean fields. The minimum spanning network showed star-shaped structures typical of populations that underwent a recent demographic expansion. The recent expansion was supported by other demographic analyzes, such as the Bayesian skyline plot, the unimodal distribution of paired differences among mitochondrial sequences, and negative and significant values of neutrality tests for the Tajima's D and Fu's F(S) parameters. In addition, high values of haplotype diversity (?) and low values of nucleotide diversity (π), combined with a high number of low frequency haplotypes and values of θ(π)<θ(W), suggested a recent demographic expansion of H. virescens populations in Brazil. This demographic event could be responsible for the low genetic structure currently found; however, haplotypes present uniquely at the same geographic regions and from one specific host plant suggest an initial differentiation among H. virescens populations within Brazil.  相似文献

Standard metabolic rate ( R S), specific growth rate ( G ) and aggressiveness were investigated in three Finnish populations of Atlantic salmon Salmo salar (Neva, Saimaa and Teno), which were reared in identical hatchery conditions. The populations differed in their geographical origin and native habitat. There was a significant difference between populations in R S: the southernmost Neva population had higher values in R S than the northernmost Teno population. No difference was found in G or aggressiveness between the populations. G was found to have a significant positive association with aggressiveness and R S among the three populations, however, these results were not statistically significant after correction for multiple tests. There was no significant association between R S and aggressiveness. Higher metabolic rate of the most southern population Neva is suggested to be an adaptation to the more abundant food sources of the southern stream.  相似文献

Methamidophos resistance and acetylcholinesterase (AChE) insensitivity to methamidophos, dichlorvos, and carbofuran were determined in the field populations of Plutella xylostella (L.) (Lepidoptera: Yponomeutidae) and its parasitoid Cotesia plutellae Kurdjumov (Hymenoptera: Braconidae) collected from the corresponding hosts between October 1998 and December 2003 in Fuzhou and Minhou, Fijian, China. Resistance levels to methamidophos and AChE insensitivity to the three insecticides in the two species of insects were high during autumn and spring and low during summer. Resistance to methamidophos was 15.3- and 12.6-fold higher in resistant F0 parents of P. xylostella and C. plutellae than in their susceptible F11 progeny, respectively. The bimolecular rate constant (k(i)) values of AChE to methamidophos, dichlorvos, and carbofuran were 4.6-, 6.3-, and 7.7-fold higher in F11 progeny of P. xylostella, and 3.7-, 4.5-, and 3.7-fold higher in F11 progeny of C. plutellae than those in their F0 parents, respectively. Compared with susceptible F11 progeny, the resistance ratios for methamidophos were 4.2-29.8 and 3.8-13.1 in 21 field populations of P. xylostella and C. plutellae, respectively. The k(i) values of AChE to methamidophos, dichlorvos, and carbofuran were 2.0-21.6-, 3.6-9.5-, and 2.6-9.2-fold higher in F11 progeny of P. xylostella, and 1.8-7.6-, 1.9-4.6-, and 2.2-7.6-fold higher in F11 progeny of C. plutellae than those in 21 field populations, respectively. Significant correlative variations of methamidophos resistance as well as significant correlative variations of k(i) values of AChE to insecticides between the two species of insects also were found in space and time. The k(i) values of AChE to insecticides in C. plutellae were far higher than those in P. xylostella. There were no obvious differences in the Km and Vmax of AChE between F0 parents and F11 progeny of P. xylostella and C. plutellae, respectively. But carboxylesterase activity was 1.6-fold higher in F0 parents of C. plutellae than in F11 progeny, and glutathione S-transferase activity was 1.5-fold higher in F0 parents of P. xylostella than in F11 progeny. The results suggested that the AChE insensitivity to insecticides might play the most important role in methamidophos resistance in the two species of insects. From these results, a spatial and temporal correlative evolution of methamidophos resistance and insensitive AChE was found to exist between P. xylostella and C. plutellae.  相似文献

We investigated genetic diversity and phylogenetic relationships among three morphologically distinct populations of Dall's porpoise (Phocoenoides dalli) in Japanese waters by analyzing mitochondrial DNA variation. These populations, the Sea of Japan-Okhotsk dalli-type population, the truei-type population and the standard dalli-type population in the northwestern North Pacific, are clearly discriminated from each other by differences in the size of their white flank patch. A total of 479 bp of the mitochondrial control region and flanking tRNA genes was sequenced for 103 individuals. Haplotypic diversity was high (h = 0.968), but these haplotypes differed by only a few nucleotides (pi = 0.0106). Although many haplotypes were shared between populations, analysis of molecular variance (AMOVA) indicated genetic subdivision among the three populations (overall F (ST) = 0.023, P < 0.001; phi(ST) = 0.026, P = 0.029). Pairwise comparisons indicated a low but significant difference between the Sea of Japan-Okhotsk and the other two populations, whereas there was no significant difference between the latter. These results suggest that there is a close evolutionary relationship among these populations despite their consistent differences in coloration. This may reflect genetic polymorphism in the common ancestral population, which subsequently underwent a rapid divergence. The low genetic variability and haplotypic differentiation of the Sea of Japan-Okhotsk population suggest that it originated from a small population that colonized the Sea of Japan or that experienced population reduction when this Sea was isolated from the North Pacific in the last glacial period.  相似文献

The differences of bionomics and ecologies of the two populations of Callosobruchus chinensis on mung bean (Vigna radiatus) at six constant temperatures (20, 25, 28, 30, 32, 35°C) and 75% r. h. were researched. ANOVA showed that temperature affected the developmental period and fertility significantly (P < 0.05). T‐test indicated there was significant difference of the combined immature stages between the two geographical populations at 25 and 32°C (P < 0.05). The average oviposition between the two geographical populations differed significantly at the same temperature (P < 0.05). At 30°C and 75% r. h., effects of 5 commercially grown legume seeds on development and reproduction of the two geographical populations of Callosobruchus chinensis (L.) were researched. The results showed that broad bean and soybean were more suitable for increase for Xichang population in terms of rm values. The adult weight and diameter of emergency hole of the two populations were also compared. According to the differences in the characteristics of development, reproduction, and damage to host seeds between the two geographical populations, conclusion can be drawn that both are different geographical populations.  相似文献

We have used sol-gel encapsulation protocols to trap kinetically and spectroscopically distinct conformational populations of native horse carbonmonoxy myoglobin. The method allows for direct comparison of functional and spectroscopic properties of equilibrium and non-equilibrium populations under the same temperature and viscosity conditions. The results implicate tertiary structure changes that include the proximal heme environment in the mechanism for population-specific differences in the observed rebinding kinetics. Differences in the resonance Raman frequency of nu(Fe-His), the iron-proximal histidine stretching mode, are attributed to differences in the positioning of the F helix. For myoglobin, the degree of separation between the F helix and the heme is assigned as the conformational coordinate that modulates both this frequency and the innermost barrier controlling CO rebinding. A comparison with the behavior of encapsulated derivatives of human adult hemoglobin indicates that these CO binding-induced conformational changes are qualitatively similar to the tertiary changes that occur within both the R and T quaternary states. Protein-specific differences in the time scale for the proposed F helix relaxation are attributed to variations in the intra-helical hydrogen bonding patterns that help stabilize the position of the F helix.  相似文献

The chicozapote ( Manilkara zapota ) is a tropical fruit tree that occurs in two morphologically distinct populations in the Yucatan peninsula of Mexico. Forest populations consist of tall, straight trees, while swamp populations have a short, shrub-like growth form. Swamp populations also have smaller leaves, fruit and seeds. We performed a random amplified polymorphic DNA (RAPD) analysis on four different populations of chicozapote to test if there was a genetic component to this variation. The populations differed in respect to habitat type (swamp vs. forest) and geographical location (east vs. west). We surveyed 80 random primers, nine of which revealed interpopulation band differences (28 band differences in total). Unweighted pair group method analysis ( UPGMA ) and neighbour-joining dendrograms showed no separation of individuals between the different populations. Analysis of the RAPD data showed no significant differences between swamp and forest populations ( P > 0.1). The lack of genetic differentiation suggests a failure to find a correlation between the RAPD loci and adaptive traits. The observed morphological differences between the swamp and forest populations of chicozapote may either be that gene flow has prevented a build-up of neutral marker differences or a plastic response to differences in habitat.  相似文献

Allozyme variation at six polymorphic loci was examined in 10 populations of Tridacna maxima from reefs in the Western Coral Sea, to test whether patterns of relatedness previously reported for foraminiferan populations reflected a fundamental structuring of the fauna in the region. Genetic distances (Nei's D) among populations of T. maxima ranged from 0–0.065 and increased with increasing geographical separation. No significant differences in gene frequencies were observed among populations within two groups of reefs identified by cluster analysis: the Great Barrier Reef (GBR), and among the offshore reefs excluding Lihou and Osprey. Significant genetic differences among these groups and the outliers Lihou and Osprey were consistent with the greater geographical separation of populations between areas than within areas. There was no evidence of differentiation along a north-south axis as reported for the foraminiferan Marginopora vertebralis, nor did populations from offshore reefs on the Queensland Plateau form a well-defined group that was genetically distinct from the GBR. The patterns observed for M. vertebralis do not appear to reflect a fundamental structuring of biota in the region. The differences in the pattern of genetic variation for M. vertebralis as compared with those for T. maxima may be due to several differences in the biological characteristics of the two species. The time of breeding in particular may influence the extent to which the divergence of the East Australian Current restricts larval dispersal among reefs in the central Queensland Plateau.  相似文献

In recent years, several F2 crosses between outbred lines of livestock have been developed to identify quantitative trait loci (QTL). These populations are valuable for further genetic analysis, including positional candidate gene loci (CGL). Analysis of CGL in F2 populations is, however, hindered by extensive between-breed linkage disequilibrium (LD). The objectives here were to develop and evaluate three tests for CGL in simulated F2 breed-cross populations. 1) A standard association test, based on the fixed effect of CGL genotype. This test was significant for CGL at considerable distances from the QTL. 2) A marker-assisted association test, based on a test at the CGL of the fixed effect of CGL genotype in a breed-cross QTL interval mapping model. This removed the impact of between-breed LD, but was not powerful in detecting CGL closely linked to the QTL, unless the CGL was the QTL. 3) An F-drop test, comparing F ratios for a QTL at the CGL with and without the CGL included as fixed effect. It had low power to distinguish close from distant CGL. Power to distinguish two CGL within 10 cM from the QTL was limited and little improved by including QTL effects associated with markers to remove between-breed LD, although the power was greater when one of the CGL was the causative mutation. Therefore, while we conclude that candidate gene tests in QTL mapping populations must be interpreted with caution, we now have a clearer picture of the value of candidate gene tests in these populations.  相似文献

Artificial selection for short wing was performed in two Drosophila melanogaster populations with partially different gene pools: the C populations were derived from a Canton stock while the H lines were derived from a cross between Canton and a b, cn, vg strain. It is shown that in both populations selection on females (CF, HF) was more effective than selection on males (CM, HM). This difference cannot be explained in terms of differences in additive genetic variability between the two sexes because: (1) both sexes contribute to the genetic variability utilized by selection applied to one sex only, and (2) switching selection pressure on females in the M lines does not result in a response comparable to that obtained in the F populations; this rules out almost completely recombination as the responsible agent for the differences between the selection limits reached by M and F selections.-These results, together with several additional observations concerning sexual dimorphism, fitness and the effect of natural selection, suggest that a complex interaction should be involved in the differential response of M and F lines, controlling the wing length phenotype.  相似文献
