Pseudomonas putida 33 wild strain, subjected to gamma ray mutagenesis and designated as P. putida 300-B mutant was used as microbial rhamnolipid-producer by using distant carbon sources (viz. hydrocarbons, waste frying oils ‘WFOs’, vegetable oil refinery wastes and molasses) in the minimal media under shake flask conditions. The behavior of glucose as co-substrate and growth initiator was examined. The 300-B mutant strain showed its ability to grow on all the substrates tested and produced rhamnolipid surfactants to different extents however; soybean and corn WFOs were observed to be preferred carbon sources followed by kerosene and paraffin oils, respectively. The best cell biomass (3.5 g l−1) and rhamnolipids yield (4.1 g l−1) were obtained with soybean WFO as carbon source and glucose as growth initiator under fed-batch cultivation showing an optimum specific growth rate (μ) of 0.272 h−1, specific product yield (qp) of 0.318 g g−1 h and volumetric productivity (PV) of 0.024 g l−1 h. The critical micelle concentration of its culture supernatant was observed to be 91 mg rhamnolipids l−1 and surface tension as 31.2 mN m−1.  相似文献

Pure glycerol and glycerol-rich product (GRP) obtained from the biodiesel industries were used as carbon source for the production of a new extracellular polysaccharide (EPS) by Pseudomonas oleovorans NRRL B-14682. The influence of temperature (20–40 °C) and pH (6.0–8.0) was studied. A temperature of 30 °C and pH control at 6.8 gave the maximum cell growth and EPS production. The culture attained a maximum cell dry weight (CDW) of 9.55 g l−1 and an EPS concentration of 11.82 g l−1 when cultivated with pure glycerol. GRP was a suitable carbon source, as shown by the slightly higher EPS concentration (12.18 g l−1). The EPS productivity obtained with GRP (3.85 g l−1 d−1) was almost twice that obtained with pure glycerol (2.00 g l−1 d−1). Also, the yield on glycerol was higher for the cultivation with GRP (0.36 g g−1) than for pure glycerol (0.28 g g−1). The EPS was a high molecular weight heteropolysaccharide, composed by neutral sugars (37–80 wt% galactose, 2–30 wt% glucose, 0.5–25 wt% mannose and 0.5–20 wt% rhamnose) and containing acyl group substituents (pyruvil, acetyl and succinyl were identified). The EPS forms highly viscous aqueous dispersions with many potential commercial applications.  相似文献

The aim of this study was to develop a bioprocess for l- and d-lactic acid production from raw sweet potato through simultaneous saccharification and fermentation by Lactobacillus paracasei and Lactobacillus coryniformis, respectively. The effects of enzyme and nitrogen source concentrations as well as of the ratio of raw material to medium were investigated. At dried material concentrations of 136.36–219.51 g L−1, yields of 90.13–91.17% (w/w) and productivities of 3.41–3.83 g L−1 h−1 were obtained with lactic acid concentrations as high as 198.32 g L−1 for l-lactic acid production. In addition, d-lactic acid was produced with yields of 90.11–84.92% (w/w) and productivities of 2.55–3.11 g L−1 h−1 with a maximum concentration of 186.40 g L−1 at the same concentrations of dried material. The simple and efficient process described in this study will benefit the tuber and root-based lactic acid industries without requiring alterations in plant equipment.  相似文献

There are currently few successful examples of using straw hemicellulose as a carbon source in the fermentation industry. In this paper, hemicellulose hydrolysates were recovered from steam-exploded wheat straw (SEWS) and used to produce microbial oil. The effects of the steam explosion treatment conditions, the elution temperature and the ratio of elution water to SEWS on sugar recovery were examined. A broth with 3.8 g l?1 of reducing sugar and 22.3 g l?1 of total soluble sugars was obtained with a 10-fold excess (w/w) of water at 40 °C to wash the SEWS treated under steam explosion conditions at 200 °C for 5 min. This broth was used to produce microbial oil by the oleaginous fungus Microsphaeropsis sp., which was able to secrete xylanase to degrade oligosaccharides from straw hemicellulose and accumulate microbial oil. Under optimized conditions, the oil concentration was 2.6 g l?1. The yield of oil from sugar consumed was 0.14 g g?1. The microbial oil produced by this research could be used as feedstock for biodiesel production because the microbial oil was primarily composed of neutral lipids. This research establishes a novel protocol for microbial oil production from straw hemicellulose.  相似文献

Lacto-N-tetraose (Gal(β1-3)GlcNAc(β1-3)Gal(β1-4)Glc) is one of the most abundant oligosaccharide structures in human milk. We recently described the synthesis of lacto-N-tetraose by a whole-cell biotransformation with recombinant Escherichia coli cells. However, only about 5% of the lactose was converted into lacto-N-tetraose by this approach. The major product obtained was the intermediate lacto-N-triose II (GlcNAc(β1-3)Gal(β1-4)Glc).In order to improve the bioconversion of lactose to lacto-N-tetraose, we have investigated the influence of the carbon source on the formation of lacto-N-tetraose and on the intracellular availability of the glycosyltransferase substrates, UDP-N-acetylglucosamine and UDP-galactose. By growth of the recombinant E. coli cells on D-galactose, the yield of lacto-N-tetraose (810.8 mg L−1 culture) was 3.6-times higher compared to cultivation on D-glucose.Using fed-batch cultivation with galactose as sole energy and carbon source, a large-scale synthesis of lacto-N-tetraose was demonstrated. During the 26 h feeding phase the growth rate (μ = 0.05) was maintained by an exponential galactose feed. In total, 16 g L−1 lactose were fed and resulted in final yields of 12.72 ± 0.21 g L−1 lacto-N-tetraose and 13.70 ± 0.10 g L−1 lacto-N-triose II. In total, 173 g of lacto-N-tetraose were produced with a space-time yield of 0.37 g L−1 h−1.  相似文献

Continuous anaerobic fermentations were performed in a novel external-recycle, biofilm reactor using d-glucose and CO2 as carbon substrates. Succinic acid (SA) yields were found to be an increasing function of glucose consumption with the succinic acid to acetic acid ratio increasing from 2.4 g g−1 at a glucose consumption of 10 g L−1, to 5.7 g g−1 at a glucose consumption of 50 g L−1. The formic acid to acetic acid ratio decreased from an equimolar value (0.77 g g−1) at a glucose consumption of 10 g L−1 to a value close to zero at 50 g L−1. The highest SA yield on glucose and highest SA titre obtained were 0.91 g g−1 and 48.5 g L−1 respectively. Metabolic flux analysis based on the established C3 and C4 metabolic pathways of Actinobacillus succinogenes revealed that the increase in the succinate to acetate ratio could not be attributed to the decrease in formic acid and that an additional source of NADH was present. The fraction of unaccounted NADH increased with glucose consumption, suggesting that additional reducing power is present in the medium or is provided by the activation of an alternative metabolic pathway.  相似文献

Anaerobic bioenergy production processes including fermentative biohydrogen (BioH2), anaerobic digestion (AD) and bioelectrochemical system have been investigated for converting municipal waste or various biomass feedstock to useful energy carriers. However, the performance of a microbial fuel cell (MFC) fed on the effluent from a two-stage biogas production process has not yet been investigated extensively in continuous reactor operation on complex substrates. In this study we have investigated the extent to which a microbial fuel cell (MFC) can reduce COD and recover further energy from the effluent of a two-stage biohydrogen and biomethane system. The performance of a four-module tubular MFC was determined at six different organic loadings (0.036–6.149 g sCOD L−1 d−1) in terms of power generation, COD removal efficiency, coulombic efficiency (CE) and energy conversion efficiency (ECE). A power density of 3.1 W m−3 was observed at the OLR = 0.572 g sCOD L−1 d−1, which resulted in the highest CE (60%) and ECE (0.8%), but the COD removal efficiency decreased at higher organic loading rates (35.1–4.4%). The energy recovery was 92.95 J L−1 and the energy conversion efficiency, based on total influent COD was found to be 0.48–0.81% at 0.572 g sCOD L−1 d−1. However, the energy recovery by the MFC is only reported for a four-module reactor and improved performance can be expected with an extended module count, as chemical energy remained available for further electrogenesis.  相似文献

The influence of organic acids on growth and dithiolopyrrolone antibiotic production by Saccharothrix algeriensis NRRL B-24137 was studied. The production of dithiolopyrrolones depends upon the nature and concentration of the organic acids in the culture medium. Study of the nature of organic acids showed that the most effective organic acids for thiolutin specific production were maleic, 4-hydroxybenzoic, benzentetracarboxylic, pantothenic, pivalic and pyruvic acids (which yielded almost five-fold over the starting medium) and pimelic acid (more than three-fold). 4-Bromobenzoic acid showed the best production of senecioyl-pyrrothine (59 mg g−1 DCW). Tiglic acid showed the best production of tigloyl-pyrrothine (22 mg g−1 DCW). The highest yield of isobutyryl-pyrrothine (7.6 mg g−1 DCW) was observed in the presence of crotonic acid. Sorbic acid yielded the best production of butanoyl-pyrrothine (26 mg g−1 DCW). Methacrylic, butyric, pyruvic and 4-bromobenzoic acids also exhibited the best production of butanoyl-pyrrothine (27–11-fold).Study of organic acid concentration showed that among the selected organic acids, pimelic acid yielded the highest specific production of thiolutin (91 mg g−1 DCW) at 7.5 mM; and senecioyl-pyrrothine (11 mg g−1 DCW), tigloyl-pyrrothine (9 mg g−1 DCW) and butanoyl-pyrrothine (3.5 mg g−1 DCW) at 5 mM. Pyruvic acid at 1.25 mM enhanced the production of senecioyl-pyrrothine (4.3 mg g−1 DCW). The maximum production of tigloyl-pyrrothine (18.6 mg g−1 DCW) was observed in the presence of tiglic acid at 2.5 mM. Maximum production of isobutyryl-pyrrothine was observed in the presence of 7.5 mM tiglic acid. In addition, methacrylic acid (at 5 mM) and butyric acid (at 2.5 mM) enhanced the production of butanoyl-pyrrothine (26 and 20 times, respectively).The above results can be employed in the optimisation of the culture medium for the production of dithiolopyrrolone in higher quantities.  相似文献

A strategy that optimization of medium compositions for maximum biomass followed by feeding of sucrose for maximum polysaccharide synthesis was developed for enhancing polysaccharide production in suspension culture of protocorm-like bodies (PLBs) of Dendrobium huoshanense C.Z. Tang et S.J. Cheng. In growth stage, the original half-strength MS medium was optimized with carbon sources, nitrogen sources and metal ion combinations. The effects of different carbon sources on PLBs growth were remarkable and sucrose at 35 g l−1 was the most suitable. Sole nitrate nitrogen of 30 mmol l−1 was the best for PLBs growth. Metal ions (Ca2+, Fe2+, Mn2+ and Zn2+) showed different influences on PLBs growth. The optimal concentration of Ca2+, Fe2+, Mn2+ and Zn2+ was 4.5 mmol l−1, 0.1 mmol l−1, 0.5 mmol l−1 and 0.06 mmol l−1, respectively. In the optimized medium (sucrose, nitrate, Ca2+, Fe2+, Mn2+ and Zn2+ concentration as described above, the other component concentration seen in half-strength MS), 33.9 g DW l−1 PLBs were harvested after 30 days of culture and biomass increase was improved 245% as compared with that in the original medium. In production stage, polysaccharide synthesis was significantly improved by the feeding sucrose. The maximum polysaccharide production (22 g l−1) was obtained in the case of 50 g l−1 sucrose feeding at day 30 of culture, which was about 109-fold higher than that in the original medium without feeding of sucrose.  相似文献

Production of pyruvate from lactate through biocatalysis is a valuable process for its simple composition of reaction system and convenience of recovery. Biocatalyst with lactate-induced NAD-independent lactate dehydrogenases (iLDHs) can effectively catalyze lactate into pyruvate. To reduce the cost of biocatalyst preparation caused by indispensable lactate addition, the mutants with constitutive iLDH of Pseudomonas sp. XP-M2 were screened. Mutant XP-LM exhibited high iLDHs activities in minimal salt medium with cheap substrate glucose as the carbon source. The biocatalyst (8.2 g dry cell weight l−1) containing 169.8 U l−1 l-iLDH was prepared with 20 g 1−1 glucose. The cost-effective biocatalyst prepared from the mutant XP-LM could efficiently catalyze lactate into pyruvate with high yield (0.961 mol mol−1). Based on the different thermostability of d-iLDH and l-iLDH in the biocatalyst, whole cells of the strain might also have the potential in production of pyruvate and d-lactate from racemic lactate.  相似文献

Microbial biolipids/biodiesels derived from volatile fatty acids (VFAs) can be a valuable alternative to plant oils if optimum fermentation conditions are determined. VFAs were used for cell mass and microbial lipid production by Cryptococcus curvatus. The lipid content in the cells increased up to 48% and 28% in batch cultures with the use of 20 g/L glucose and 6 g/L of VFAs as the carbon source, respectively. In this study, C. curvatus used VFAs as a carbon source via anaerobic digestion of rice straw hydrolysates. VFAs produced from rice straw resulted in yield of 0.43 g VFAs/g substrate and 40% higher specific growth rate(0.305 h−1) than synthetic VFAs. The highest fatty acid composition observed was C18:1, was obtained using glucose and VFAs as the carbon source to yield a cetane number of 56–59, which is suitable for biodiesel production. The cost of microbial lipids was estimated to be 0.30–1.15 USD/L given 0–150 USD/ton of VFAs cost for a yield of 0.17 g/g of lipids. Thus, VFAs can be a suitable carbon source for economical biodiesel production.  相似文献

A new bioprocess for the synthesis of lactosucrose was studied using a covalently immobilized β-galactosidase on macrospheres of chitosan. The effects of temperature and pH on the production of lactosucrose and other oligosaccharides were evaluated. At 30 °C and pH 7.0, the maximum concentration of lactosucrose reached to 79 g L−1. The change of the reaction conditions allowed to modify the qualitative profile of the final products without quantitative change in the total of oligosaccharides produced. At pH 7 and 30 °C, products profile was 79 g L−1 of lactosucrose, 37 g L−1 of galactooligosaccharides and 250 g L−1 of total oligosaccharides, while at pH 5 and 64 °C the concentrations for the same compounds were 40, 62 and 250 g L−1, respectively. The immobilization increased the thermal stability up to 260-fold. Using 300 g L−1 of sucrose and 300 g L−1 of lactose, and 8.5 mg of chitosan mL−1, 30 cycles of reuse were performed and the biocatalyst kept the maximal lactosucrose synthesis. These results fulfill some important aspects for the enzyme immobilization and oligosaccharides synthesis: the simplicity of the protocols, the high operational stability of the enzyme and the possibility of driving the final products.  相似文献

In this study, the soap stock as a sole carbon source was used for growing a carotenoid producing yeast (Rhodotorula rubra). The application of soap stock resulted in increase of carotenoids yield up to 5.36 folds when compared with the grown cultures on glucose. On the best Monod equation fitted on the specific growth rate (μ) data, the maximum specific growth rate (μm) and half-saturation concentration (KS) were respectively determined at 0.064 h−1 and 3.26 g L−1 for total fatty acids presented in soap stock. Further tests on the carotenogenesis process were carried out in a cell-immobilized airlift photobioreactor where the natural loofa sponge was used for immobilization of the cells. The performance of the bioreactor was statistically studied by the response surface methodology (RSM) where aeration rate of 0.11 vvm and light irradiation intensity of 2517 Lx provided an optimum condition for producing β-carotene with a specific production rate of 22.65 mg gcell−1 day−1.  相似文献

The relationship between light intensity, nitrogen availability and pigmentation was investigated in mixotrophic and heterotrophic cultures of the unicellular red alga Galdieria sulphuraria 074G, a potential host for production of the blue pigment, phycocyanin (PC). During the exponential growth phase of batch cultures, G. sulphuraria 074G contained 2–4 mg phycocyanin per g dry weight. In carbon-limited and nitrogen-sufficient batch cultures grown in darkness, this value increased to 8–12 mg g−1 dry weight during the stationary phase, whereas the phycocyanin content in nitrogen-deficient cells decreased to values below 1 mg g−1 dry weight during stationary phase. Light intensities between 0 and 100 μmol photons m−2 s−1 had no influence on phycocyanin accumulation in mixotrophic cultures grown on glucose or fructose, while light stimulated phycocyanin synthesis in cultures grown on glycerol, in which the phycocyanin content in stationary phase was increased from 10 mg g−1 dry weight in darkness to 20 mg g−1 dry weight at a light intensity of 80 μmol photons m−2 s−1. At higher light intensities, less phycocyanin accumulated than at lower intensities, irrespective of the carbon substrate used. In carbon-limited continuous flow cultures grown on glucose or glycerol at a dilution rate of 0.63 day−1, corresponding to 50% of the maximum specific growth rate, the highest steady-state phycocyanin content of 15–28 mg g−1 dry weight was found at 65 μmol photons m−2 s−1. In contrast to the apparent glucose repression of light-induced PC synthesis observed in batch cultures, no glucose repression of the light stimulation was observed in continuous flow cultures because the glucose concentration in the culture supernatant always remained at limiting levels. Despite the fact that G. sulphuraria 074G contains less phycocyanin than some other microalgae and cyanobacteria, the ability of G. sulphuraria 074G to grow and synthesize phycocyanin in heterotrophic or mixotrophic cultures makes it an interesting alternative to the cyanobacterium, Spirulina platensis presently used for synthesis of phycocyanin.  相似文献

An annual cycle of biomass and productivity of wild celery (Vallisneria americana) was studied in Kings Bay, FL, USA. In situ growth rates were measured monthly between March 2001 and June 2002 in high-density stands, using a modified hole-punching technique, and applied to shoot density data to obtain areal estimates of production. Mean shoot density varied greatly over the study period, ranging between 200 and 800 shoots m−2. Mean total biomass ranged between 162 and 1013 g m−2, with aboveground material comprising, on average, 70% of total biomass. Total annual estimated production of new attached shoots was 519 g m−2. Leaf growth rates peaked at >50 mg shoot−1 d−1, and mass-specific leaf growth ranged 0.6–1.8% d−1. Annually, individual shoots produced 7.4 g of leaf material and completely replaced standing leaf biomass 3.5 times. Areal leaf production was highest in late spring/summer of 2001, and ranged between 3.6 and 23.0 g m−2 d−1. Annual total leaf production was 2704 g m−2. Seasonality was not apparent in most variables monitored monthly; only 1 of the 64 relationships we examined between environmental variables (nutrients, chlorophyll a, and irradiance) and Vallisneria biological variables were significant, with relative growth rate increasing linearly with irradiance. Peak biomass and productivity of Vallisneria in Kings Bay were high compared to literature values for other Vallisneria populations as well as global averages for well-studied seagrasses, emphasizing the potential importance of Vallisneria to whole ecosystem functioning in springs, lakes, and oligohaline reaches of many estuaries.  相似文献

Marine microorganisms are a potential source of enzymes with structural stability, high activity at low temperature and unique substrate selectivity. Thraustochytrids are marine heterotrophic microbes, well known for the production of omega-3 fatty acids. In this study the effect of Tween 80 as a carbon source was investigated with regard to biomass, lipase and lipid productivity in Schizochytrium sp. S31. Tween 80 (1%) and 120 h of incubation were the optimum condition period for biomass, lipid and lipase productivity in a stirred tank reactor. The yields obtained were 0.9 g L−1 of biomass, 300 mg g−1 of lipid and 39 U/g of lipase activity. Sonication was optimised in terms of time and acoustic power to maximise the yield of extracted lipase. The extracted lipase from Schizochytrium S31 was observed to hydrolyse long chain polyunsaturated fatty acids DHA and EPA.  相似文献

We previously reported a metabolic engineering strategy to develop an isopropanol producing strain of Cupriavidus necator leading to production of 3.4 g L−1 isopropanol. In order to reach higher titers, isopropanol toxicity to the cells has to be considered. A toxic effect of isopropanol on the growth of C. necator has been indeed observed above a critical value of 15 g L−1. GroESL chaperones were first searched and identified in the genome of C. necator. Native groEL and groES genes from C. necator were over-expressed in a strain deleted for PHA synthesis. We demonstrated that over-expressing groESL genes led to a better tolerance of the strain towards exogenous isopropanol. GroESL genes were then over-expressed within the best engineered isopropanol producing strain. A final isopropanol concentration of 9.8 g L−1 was achieved in fed-batch culture on fructose as the sole carbon source (equivalent to 16 g L−1 after taking into account evaporation). Cell viability was slightly improved by the chaperone over-expression, particularly at the end of the fermentation when the isopropanol concentration was the highest. Moreover, the strain over-expressing the chaperones showed higher enzyme activity levels of the 2 heterologous enzymes (acetoacetate carboxylase and alcohol dehydrogenase) of the isopropanol synthetic operon, translating to a higher specific production rate of isopropanol at the expense of the specific production rate of acetone. Over-expressing the native chaperones led to a 9–18% increase in the isopropanol yield on fructose.  相似文献

Phosphogypsum (CaSO4), a primary by-product of phosphoric acid production, is accumulated in large stockpiles and occupies vast areas of land. It poses a severe threat to the quality of water and land in countries producing phosphoric acid. In this study, the potential of sulfate-reducing bacteria for biodegradation of this sulfur-rich industrial solid waste was assessed. The effect of phosphogypsum concentration, carbon and nitrogen sources, temperature, pH and stirring on the growth of sulfate-reducing bacteria was investigated. Growth of sulfate-reducing bacteria was monitored by measuring sulfide production. Phosphogypsum was shown to be a good source of sulfate, albeit that the addition of organic carbon was necessary for bacterial growth. Biogenic sulfide production occurred with phosphogypsum up to a concentration of 40 g L−1, above which no growth of sulfate-reducing bacteria was observed. Optimal growth was obtained at 10 g L−1 phosphogypsum. Both the gas mixture H2/CO2 and lactate supported high amounts of H2S formation (19 and 11 mM, respectively). The best source of nitrogen for sulfate-reducing bacteria was yeast extract, followed by ammonium chloride. The presence of nitrate had an inhibitory effect on the process of sulfate reduction. Stirring the culture at 150 rpm slightly stimulated H2S formation, probably by improving sulfate solubility.  相似文献

The addition of soybean oil and Tween 80 was evaluated with the objective of increasing the production of botryosphaeran, an exopolysaccharide (EPS) of the (1  3;1  6)-β-d-glucan type produced by the fungus Botryosphaeria rhodina MAMB-05. Factorial design and analysis by response surface methodology was developed to select the main factors that would affect and enhance EPS production. The optimized culture conditions were: 40 g l−1 glucose with 10 ml l−1 soybean oil, and 4.5 ml l−1 Tween 80, during 72 h cultivation at 28 °C (180 rpm) and initial pH 5.7. The predicted result for botryosphaeran production was 8.22 ± 1.36 g l−1, and compared with the experimental value of 7.74 ± 0.13 g l−1. Partial characterization of the botryosphaeran produced under the optimized conditions showed one type of polysaccharide with β-glycosidic linkages containing glucose as monosaccharide.  相似文献
