The diversity of the fungal community associated with adults of Dryocosmus kuriphilus following emergence was examined using HTS analysis. Ascomycota dominated the fungal core-biome community. The functional guilds of the 90 taxa forming the core-biome were assessed, demonstrating three main groups: saprotrophs, plant pathogens and entomopathogens. Twenty-nine OTUs out of 90 were resolved to species level identifying 26 different fungal species. Among these species, many were cosmopolitan or previously recorded in Europe. Ten taxa were previously recorded on chestnut, including some recognized plant pathogens associated with foliage and green tissues such as Epicoccum nigrum, Gnomoniopsis castanea, Colletotrichum acutatum, Stromatoseptoria castaneicola, Ramularia endophylla. Beauveria bassiana; within the core microbiome, Fusarium larvarum represented the most abundant entomopathogenic species. Some of these species are known to impact directly or indirectly the vitality of the insects in the galls. The chestnut blight pathogen, Cryphonectria parasitica, was never found associated with D. kuriphilus. Based on the present study, an active role for D. kuriphilus as a vector of chestnut fungal endophyte/pathogens cannot be demonstrated but neither ruled out.  相似文献

The Asian chestnut gall wasp Dryocosmus kuriphilus (Hymenoptera: Cynipidae) is one of the most serious pests of chestnut trees worldwide. Wasps lay eggs into chestnut buds from mid Jane to late July, depending on latitude, and galls develop the following spring on new vegetation, reducing photosynthesis and nut production. We observed that modification of tissues surrounding D. kuriphilus eggs, which differentiate to form the larval chamber, started approximately 1 month after oviposition, shortly after eggs hatch. The larval chambers continued to increase slightly in size throughout the autumn months until January. After that, a period of stagnation, which corresponds to the plant's dormancy, occurred, followed by rapid growth from March to May, during the period from bud swelling to bud break. Galls continued to grow during the leaf expansion after bud break and stopped when plant organs achieved their final size. Our results have implications for the management of the pest, providing a better understanding of the critical time periods for the effective control.  相似文献

  • 1 The cynipid gall wasp Dryocosmus kuriphilus Yasumatsu (Hymenoptera: Cynipidae) is an invasive wasp in Europe and is one of the major pests of chestnuts worldwide. An investigation of this cynipid was started in Campania, Southern Italy, in 2008, with the aim of providing guidelines for its sustainable control.
  • 2 The number of larvae/gall increased in the second year of colonization relative to the first one but thereafter decreased in the third year of colonization, whereas the volume of the galls after the initial increase in the second year of colonization became stable. Adult emergence generally peaked between the middle of June and July.
  • 3 The only reliable method for assessing the presence of the pest at its earliest stage was through dissecting the buds because oviposition scars on the buds tended to disappear during the winter.
  • 4 Yellow traps were shown to be an effective method for assessing the emergence of cynipid adults and could potentially be used to time chemical applications in those cases in which this control method is feasible.
  • 5 No correlation was recorded between the relative durations of the final stages (pupae, pharate adults and emergence).
  • 6 Possible control strategies for this invasive pest are discussed.

In the 2015–2016 growing seasons, two novel symptoms were assessed on the crown of trees in orchards and coppices of chestnut groves in Central Italy. The first symptom was flagging of annual shoots with green leaves undergoing sudden wilt and turning brown later in the season. The second symptom consisted of leaves on annual shoots turning yellow before wilting in absence of flagging represented the second symptom. Samples were collected along transects in early summer, late summer and winter, and processed in the laboratory. The flagging symptom was associated in early summer with the presence of C. parasitica in cryptic dried buds on stems from the previous year's growth. The pathogen was also found in dormant buds in winter, suggesting that the infection could take place in summer during the Chinese gall wasp oviposition period. Cryphonectria parasitica was also isolated from abandoned galls in winter supporting the hypothesis that galls are a potential source of inoculum for crown infections. Aetiology of yellowing was not clarified and no fungal taxa were specifically associated with this symptom. Gnomoniopsis castanea, C. parasitica and, in early summer, Colletotrichum acutatum were the most abundant fungal taxa isolated from chestnut shoots and buds.  相似文献

Dryocosmus kuriphilus Yasumatsu (Hymenoptera: Cynipidae), is one of the most serious pests attacking chestnut trees. Recently it was incidentally introduced into Northwest Italy and it is now spreading throughout Europe. This pest was successfully controlled in Japan by introducing a parasitoid, Torymus sinensis Kamijo (Hymenoptera: Torymidae), from China’s mainland. Following this successful experience, the parasitoid was introduced into Italy from Japan. One year of preliminary studies led to a successful method of rearing imported galls with a synchronization between the parasitoid’s emergence and the presence of the target galls in the field. In two consecutive years, a total of 2,117 individuals were released in several sites covering most of the infested area. There are encouraging data about the settlement of the parasitoid and its synchrony with the host’s cycle: from about 64,000 host galls collected in the field over 200 T. sinensis were reared. The role of native parasitoids associated with the chestnut gall pest in its Italian distribution range is also discussed.  相似文献

We describe here the parasitoid wasps Torymus sinensis Kamijo and T. beneficus Yasumatsu & Kamijo (early-spring and late-spring strains), which are introduced and indigenous natural enemies of the chestnut gall wasp Dryocosmus kuriphilus Yasumatsu, an invasive chestnut pest in Japan. We recently discovered specimens of T. koreanus Kamijo, endemic in Korea, among Torymus parasitoids collected from D. kuriphilus galls in a Japanese chestnut orchard. In this study we compare the composition of Torymus parasitoids emerging from D. kuriphilus galls before and after the release of T. sinensis. Before the release of T. sinensis, early-spring and late-spring strains of T. beneficus predominated (58.3 and 20.8% of specimens collected). However, a few years after the release, both T. beneficus strains had been almost completely displaced by T. sinensis. In contrast to the rapid decrease in T. beneficus, T. koreanus did not decrease drastically before and even after the release of T. sinensis (approximately 10–20% of specimens collected). These results suggest that not a few T. koreanus were present in the Japanese chestnut orchard investigated at least several years after the release of T. sinensis, although both the T. beneficus strains were rapidly displaced by T. sinensis during this period.  相似文献

  • 1 Preliminary investigations were carried out on Dryocosmus kuriphilus Yasumatsu on Castanea sativa Miller in Tuscany to assess variations in gall characteristics in coppice and high forest at two crown heights (height < 2 or 2–6 m), influence of bud size and bud position on oviposition rates and susceptibility of three cultivars.
  • 2 Gall size may depend on various factors, including wasp population density. In the present study area, small galls (with one or two cells) were the most numerous in 2008, whereas larger galls (with more than three cells) prevailed in 2009.
  • 3 Dryocosmus kuriphilus oviposition occurrence was influenced by both bud size and bud position. Buds with eggs tended to be larger in size compared with bud without eggs, suggesting that D. kuriphilus females prefer to lay eggs in larger buds (approximately 6 mm3) compared with smaller buds (approximately 3 mm3). The mean number of eggs per bud tended to decrease from the apical bud toward the basal bud.
  • 4 Three C. sativa cultivars, Carpinese, Fusca and Cesurone, were examined. Fusca grafts had significantly more galls compared with Carpinese and Cesurone, whereas Cesurone grafts had more larvae per bud compared with Carpinese and Fusca. Overall, the Carpinese cultivar may be less susceptible to D. kuriphilus galling compared with the Fusca and Cesurone cultivars.

The entomopathogenic fungus Beauveria bassiana is widely used for insect pest control and can produce three distinct infective unit types under different nutritional and environmental conditions: aerial conidia, blastospores, and submerged conidia. Here, we identified endophytic colonization ability and existing forms of the three types of B. bassiana infective units after inoculating Arabidopsis plants via soil drenching, and tested their effects on their presence mold disease caused by Botrytis cinerea. We found that all B. bassiana infective unit types colonized Arabidopsis leaves, with germinating and producing hyphae by hydrophilic blastopores and submerged conidia; further, we showed that blastospores were more effective in defending against B. cinerea, compared with aerial conidia. These findings suggest that in addition to aerial conidia, the colonization of other two types of entomopathogenic fungal infective units could also have important impacts on plant resistance. This study contributes to better understanding on the function of B. bassiana as fungal endophytes, which could lead to a new paradigm for how to successfully use these organisms in biological control against plant diseases.  相似文献

Pochonia chlamydosporia (Pc123) is a fungal parasite of nematode eggs which can colonize endophytically barley and tomato roots. In this paper we use culturing as well as quantitative PCR (qPCR) methods and a stable GFP transformant (Pc123gfp) to analyze the endophytic behavior of the fungus in tomato roots. We found no differences between virulence/root colonization of Pc123 and Pc123gfp on root-knot nematode Meloidogyne javanica eggs and tomato seedlings respectively. Confocal microscopy of Pc123gfp infecting M. javanica eggs revealed details of the process such as penetration hyphae in the egg shell or appressoria and associated post infection hyphae previously unseen. Pc123gfp colonization of tomato roots was low close to the root cap, but increased with the distance to form a patchy hyphal network. Pc123gfp colonized epidermal and cortex tomato root cells and induced plant defenses (papillae). qPCR unlike culturing revealed reduction in fungus root colonization (total and endophytic) with plant development. Pc123gfp was found by qPCR less rhizosphere competent than Pc123. Endophytic colonization by Pc123gfp promoted growth of both roots and shoots of tomato plants vs. uninoculated (control) plants. Tomato roots endophytically colonized by Pc123gfp and inoculated with M. javanica juveniles developed galls and egg masses which were colonized by the fungus. Our results suggest that endophytic colonization of tomato roots by P. chlamydosporia may be relevant for promoting plant growth and perhaps affect managing of root-knot nematode infestations.  相似文献

Crown sheath rot, caused by the ascomycete Gaeumannomyces graminis var. graminis that infects the root and the base of the culm of rice, causes early grains maturation, tiller death and reduced yield. As a paucity of information exists in the literature on the rice‐G. graminis var. graminis interaction at the microscopic level, this study aimed to gain novel insights into the infection process of this pathogen in the root and culm of rice using both light and scanning electron microscopy. In the roots, the fungus initially colonized the epidermal, exodermal and sclerenchyma cells. At 15 days after inoculation (dai), fungal hyphae colonized the cortex and clusters of perithecia were observed in the roots. At 20 dai, the fungus reached the central cylinder, and an intense fungal colonization at the base of the culm was observed that resulted in the formation of a mycelial mat on both adaxial and abaxial surfaces of the leaf sheaths. At 25 dai, fungal growth was noticed in the parenchyma cells, vascular bundles and airspaces. Perithecia emerged through the base of prophyllum and from the first leaf sheath at 30 dai. The results of this study provide new insights into the infection process of G. graminis var. graminis in rice and may help to find better control measures in reducing crown sheath rot development.  相似文献

We investigated the association between a gall midge, Illiciomyia yukawai, and its symbiotic fungi on Japanese star anise, Illicium anisatum. The number of fungal species isolated from the galls increased with development of the galls, whereas those from the leaves showed a different trend. Botryosphaeria dothidea was dominant in the galls from June to October, and after that Phomopsis sp. 1, Colletotrichum sp., and Pestalotiopsis sp. became dominant. Although B. dothidea was not isolated from the leaves, it was detected from mycangia (abdominal sternite VII) of egg-laying adults at a high isolation frequency (>90%). However, B. dothidea was not isolated from mycangia of adults emerging from galls that were enclosed by plastic bags. This indicates that I. yukawai is closely associated with B. dothidea and that its newly emerged adults do not take the fungus into mycangia directly from the galls where they had developed. Also, the fungus from the fungal layers of ambrosia galls has less ability to propagate on artificial media despite the presence of its mycelial mass in mature galls.  相似文献

American chestnut (Castanea dentata [Marsh.] Borkh.) dominated the eastern forests of North America, serving as a keystone species both ecologically and economically until the introduction of the chestnut blight, Cryphonectria parasitica, functionally eradicated the species. Restoration efforts include genetic transformation utilizing genes such as oxalate oxidase to produce potentially blight-resistant chestnut trees that could be released back into the native range. However, before such a release can be undertaken, it is necessary to assess nontarget impacts. Since oxalate oxidase is meant to combat a fungal pathogen, we are particularly interested in potential impacts of this transgene on beneficial fungi. This study compares ectomycorrhizal fungal colonization on a transgenic American chestnut clone expressing enhanced blight resistance to a wild-type American chestnut, a conventionally bred American-Chinese hybrid chestnut, and other Fagaceae species. A greenhouse bioassay used soil from two field sites with different soil types and land use histories. The number of colonized root tips was counted, and fungal species were identified using morphology, restriction fragment length polymorphism (RFLP), and DNA sequencing. Results showed that total ectomycorrhizal colonization varied more by soil type than by tree species. Individual fungal species varied in their colonization rates, but there were no significant differences between colonization on transgenic and wild-type chestnuts. This study shows that the oxalate oxidase gene can increase resistance against Cryphonectria parasitica without changing the colonization rate for ectomycorrhizal species. These findings will be crucial for a potential deregulation of blight-resistant American chestnuts containing the oxalate oxidase gene.  相似文献

Ten fungal isolates belonging to the genera Beauveria (3), Hypocrea (1), Gibberella (1), Metarhizium (2), Trichoderma (1) and Fusarium (2) were evaluated in the laboratory to determine whether they could become endophytic in two pea leafminer (Liriomyza huidobrensis) host plants (Vicia faba and Phaseolus vulgaris), and to assess their possible negative effects on leafminers. Beauveria (ICIPE279), Hypocrea, Gibberella, Fusarium and Trichoderma isolates colonized roots, stems and leaves of both host plant species. Beauveria isolates G1LU3 and S4SU1 colonized roots, stems, and leaves of P. vulgaris but only the root and stem of V. faba. Isolates of Metarhizium failed to colonize the two host plants. The effects of endophytically colonized fungal pathogens on mortality, oviposition, emergence and longevity of L. huidobrensis were investigated after endophytic colonization of V. faba plants. All the fungal isolates that succeeded in colonizing the host plant were pathogenic to L. huidobrensis, causing 100 % mortality within 13.2 ± 0.7–15.0 ± 0.6 d. However, Hypocrea outperformed the other isolates (p < 0.0 001) in reducing longevity of the progeny (11.2 ± 1.0 vs. 17.8 ± 1.4 d in the control), the number of pupae (80.0 ± 6.7 vs. 387.0 ± 21.7 pupae in the control), and adult longevity (3.8 ± 1.0 vs. 9.9 ± 1.8 d in the control). Adult emergence was significantly reduced (p < 0.0 001) in Hypocrea (21.4 %) and Beauveria (38.0 %) treatments compared to the control (82.9 %).  相似文献

The impact of gall-inducing aphids on shoot development was analyzed in 900 shoots from 20 pistachio trees, Pistacia atlantica Desf. (Anacardiaceae): 600 in which the axillary—lateral buds were galled by Slavum wertheimae HRL during the previous growth season, and 300 ungalled shoots. Although P. atlantica is a compensating tree, and the aphids do not attack the apical buds, further development of shoots from the apical buds was stopped in 62% of the galled shoots, while only 8.7% of nongalled shoots stopped their growth. Further development was stopped more often on shoots carrying two or more galls than on shoots supporting only one gall. To assess the hypothesis that bud destruction by the aphids explains this pattern, a field experiment was conducted in 140 shoots, distributed across seven trees. One, two or three axillary buds from five shoots of each tree were removed for each treatment, and five other shoots were marked as controls. Only 14 shoots (10%) of the 140 did not develop. The growth of the other shoots was not very different among the treatments. The colonization of the apical shoots, which developed on previously treated shoots, by three other galling aphid species was monitored. Removing lateral buds considerably reduced the establishment of Geoica sp. galls (70% of them colonized control shoots), but weakly influenced Forda riccobonii (Stefani). It also contributed only 5% of the total variance of the distribution of Smynthurodes betae West. The different results of the survey and the experiment show that the impact of S. wertheimae galls on the future growth of shoots from apical buds is more complex than the simple physical destruction of the axillary buds. Optimal clutch size of the chestnut gall-wasp, Dryocosmus kuriphilus Yasumatsu (Hymenoptera: Cynipidae), was examined in galls on wild and resistant chestnut trees in 1988 and 1989. The rate of escape success of newly-emerged adults from galls was an average of 60%, irrespective of cell numbers per gall. Dry mass per cell of a gall (as an index of nutritive condition) decreased with increasing cell number per gall, but was proportional to the mean number of mature eggs of new adults per gall. The number of cells per gall that occurred most frequently did not agree with that attained by the maximum survival rate from young larva to adult emergence of the gall-wasp. This discrepancy was examined from the viewpoint of three factors: 1) quality of offspring, 2) defensive response of the host plant causing mortality of the gall-wasp before cell formation, and 3) fitness per gall vs. fitness per egg. It is concluded that the third factor is most likely to be the one best in explaining the discrepancy.  相似文献

The effect of fungal infection by Metarhizium anisopliae on feeding and oviposition of adult Liriomyza huidobrensis was examined on three host plants, faba bean (Vicia faba), French bean (Phaseolus vuklgaris) and snow pea (Pisum sativum) in the laboratory. Flies were contaminated with dry conidia and allowed to feed and oviposit on the different host plants. Mortality in L. huidobrensis varied between 14% and 20% in the controls and between 77% and 100% in fungal treatments 120 h post-infection for the three host plants. L. huidobrensis made more punctures (47.3–52.6 cm?2) in the control than in the fungal treatments (23.1–26.9 cm?2) for the three host plants. The cumulative average number of punctures cm?2/female by L. huidobrensis was higher in the controls than in fungal treatments from 72 h post-treatment in faba bean (12.2 vs. 8.2) and French bean (14.8 vs. 8.9), and from 48 h post-inoculation in snow pea (8.5 vs. 5.7). Female L. huidobrensis laid more eggs in the control (0.6–6.1) than in fungal treatments (0.2–1.5) across the host plants tested. The cumulative mean number of eggs cm?2/female was significantly higher in the controls than in fungal treatments from 48 h post-treatment in faba bean (0.4 vs. 0.2) and French bean (0.1 vs. 0), and 96 h post-inoculation in snow pea (0.2 vs. 0.1). The host plant did not affect the average total number of punctures but had a significant effect on egg laying, with faba bean harboring greater number of eggs in both control and fungal treatments. A proper timeline application of the fungus before onset of feeding and oviposition peaks will be crucial in field suppression of the pest using M. anisopliae. In addition, a great consideration must be given to the target host plants prior to application of the fungus.  相似文献

Foliar endophytic fungi are present in almost all vascular plants. The composition of endophyte communities varies among plant individuals. Likely, but understudied, sources of this variation are the species composition of the plant community and initial attacks by insect herbivores. We addressed these issues by characterizing fungal endophyte communities on leaves of chestnut (Castanea sativa) grown in pure vs. mixed stands. We used ITS metabarcoding methods to identify endophytic fungi associated with galls caused by the invasive gall wasp, Dryocosmus kuriphilus, and with surrounding chestnut leaf tissues. We found 1378 different OTUs. The richness, diversity and composition of endophyte communities differed between galls and surrounding leaf tissues but were independent of forest stand composition. Fungal endophyte richness was lower in galls than in surrounding leaf tissues. Most differences in the composition of fungal endophyte communities between galls and foliar tissues were due to OTU turnover. These results suggest that insect-induced galls provide a particular habitat condition for endophytic microorganisms, regardless of forest species composition. A better understanding of endophyte biology is important to improve their use as biocontrol agents of galling insects.  相似文献

Metarhizium is an insect pathogenic fungus and a plant root symbiont. Here the root association patterns (rhizoplane or endophytic colonization) were analyzed in common beans (Phaseolus vulgaris) and sweet corn (Zea mays) using M. robertsii and M. brunneum under various vermiculite treatments (control, with sucrose, with an insect) at two time points of plant growth (10 and 20 days). We observed that M. brunneum and M. robertsii preferentially endophytically colonized the hypocotyl, however, greater rhizoplane colonization was observed at the regions proximal to the hypocotyl in both plants. Vermiculite amended with an infected insect resulted in greater endophytic and rhizoplane colonization at 20 days compared to 10 days, for both plants as well as for both Metarhizium species. Regardless of the vermiculite treatment, corn was preferentially colonized compared to bean. Sucrose amendment in the vermiculite and infected insect amended vermiculite only showed differences in rhizoplane colonization. The greatest root association occurred with M. brunneum with an infected insect and that in corn after 20 days.  相似文献
