The results of previous studies on the effect of diazepam on palate formation in animals have been inconclusive. Teratogen-induced cleft palate is usually caused by a delay in palatal shelf elevation. The present study investigated the effect of diazepam on palate formation in the Sprague-Dawley rat. Five groups of dams received subcutaneous doses of either 10, 20, 30, 40, or 50 mg/kg body weight of diazepam. Control dams received propylene glycol (vehicle). Dams in each dosage group were killed at 16.9 (16 d 9 h); 16.16, and 17.9 days of gestation, respectively, to assess delay in palatal shelf elevation. Crown rump length (CRL) of 1,283 fetuses collected from 105 dams was measured. Fetuses in each time/dosage group showed a reduction in CRL (P less than .01). With increasing dosage the number of fetuses showing delayed palatal shelf elevation was significantly increased (P less than .01). These results demonstrate that with an increase in dose there is an increased delay in palatal shelf elevation and a decrease in CRL. However, in this strain there seems to be a rapid prenatal recovery, resulting in a marked reduction in the incidence of delayed palatal shelf elevation.  相似文献

AIMS: Selection of suitable surfactants for enhancing and stabilizing alpha-amylase of Geobacillus thermoleovorans. METHODS AND RESULTS: Geobacillus thermoleovorans was cultivated in shake flasks containing 50 ml of starch-yeast extract-tryptone (SYT) medium with/without surfactants. Titres of the enzyme in media were monitored. The enzyme was also preserved at 4 degrees C with/without surfactants and enzyme activities were determined. Among polyethylene glycol (PEGs) of different molecular weights, PEG 8000 (0.5%, w/v) caused a slight increase in the enzyme titre, while Tween-20, Tween-40 and Tween-60 (0.03%, w/v) exerted a significant stimulatory effect on enzyme secretion. In the presence of SDS, Tween-80 and cholic acid (0.03%, w/v), the enzyme production was nearly twofold higher than that in the control. The anionic (SDS, cholic acid) and non-ionic (Tweens) detergents increased the cell membrane permeability, and thus, enhanced alpha-amylase secretion. Furthermore, anionic surfactants exhibited stabilizing effect on the enzyme during preservation at 4 degrees C. CONCLUSIONS: PEG 8000 and the ionic detergents (SDS, cholic acid and Tween-80) were more effective in the solubilization of cell membrane components, and enhancing enzyme yields than the cationic detergents such as CTAB (N,Cetyl-N,N,N-trimethyl ammonium bromide). Further, these surfactants were found to stabilize the enzyme at 4 degrees C. SIGNIFICANCE AND IMPACT OF THE STUDY: The secretion of Ca2+-independent hyperthermostable alpha-amylase was enhanced in the presence of certain anionic and non-ionic detergents in the medium. Furthermore, the surfactants stabilized the enzyme during preservation at 4 degrees C. The use of this enzyme in starch hydrolysis eliminates the addition of Ca2+ in starch liquefaction and its subsequent removal by ion exchange from sugar syrups.  相似文献

The present study was performed to develop a suitable cryoprotectant solution for cryopreservation of rat two-cell stage embryos. First, we examined the cell permeability of several cryoprotectants; propylene glycol had the fastest permeability compared to dimethyl sulfoxide, ethylene glycol, and glycerol. Embryos were then exposed to a solution containing propylene glycol to evaluate its effects on fetal development. As the development was similar to that of fresh embryos, P10 (10% v/v propylene glycol in PB1) was used as a pretreatment solution. Next, the effects of the vitrification solution components (sucrose, propylene glycol, ethylene glycol, and Percoll) were examined by observing the vitrification status; 10% v/v propylene glycol, 30% v/v ethylene glycol, 0.3 mol sucrose, and 20% v/v Percoll in PB1 (PEPeS) was the minimum essential concentration for effective vitrification without the formation of ice crystals or freeze fractures.  相似文献

MultiLure traps were deployed in a Hawaiian orchard to compare the attraction of economically important fruit flies and nontarget insects to the three-component BioLure and torula yeast food lures. Either water or a 20% propylene glycol solution was used to dissolve the torula yeast or as capture fluid in BioLure traps. Torula yeast in water was more attractive than BioLure for male and female Bactrocera cucurbitae (Coquillett) and Bactrocera dorsalis (Hendel) and as attractive for Ceratitis capitata (Wiedemann), and the addition of propylene glycol significantly inhibited the attractiveness of torula yeast. The known synergistic effect of propylene glycol with BioLure, resulting in increased captures of Anastrepha flies, was not observed with Bactrocera. Nontarget Drosophilidae, Neriidae, Phoridae, Calliphoridae, Sarcophagidae, and Muscidae were more strongly attracted to BioLure, and both lures collected Chloropidae equally. As with fruit flies, propylene glycol in torula yeast significantly decreased nontarget captures. The results therefore suggest that torula yeast in water is a more effective attractant than BioLure for pest Bactrocera while minimizing nontarget captures.  相似文献

An investigation was carried out to evaluate the protective effect of polyhydric alcohols, such as propylene glycol and glycerol on the inactivation of an alkaline protease by sodium dodecyl sulfate (SDS) and H2O2. Addition of polyols increased the stability of a Bacillus clausii I-52 alkaline protease towards not only the thermal-induced, but also the SDS and H2O2-induced inactivation. Among the polyols examined, the best results were obtained with propylene glycol. The half-life of the enzyme was increased by 43- and >105-fold by the addition of 10% (v/v) propylene glycol to the enzyme preparations containing 5% (w/v) SDS and 5% (v/v) H2O2 at 50 °C, respectively. Besides the protection effect of propylene glycol from enzyme inactivation by SDS and H2O2, it also improved the hydrolytic efficiency towards substrate like BSA during the protease reaction containing SDS or H2O2. This result suggests that propylene glycol has a significant potential as a good stabilizer of an alkaline protease preparation, which finds use as an additive in industrial applications, especially, the detergent industry.  相似文献

Cryoprotectors (propylene glycol), ethylene glycol, polyethylene glycol-1500 and dimethyl sulphoxide) are studied for their effect on permeability of liposomes for incorporated molecules of 5,5-dithiobis-2-nitrobenzoic acid (DTNB) under cooling within a temperature range from 0 degree C to -25 degrees C. A similarity is found in the way of ethylene glycol and propylene glycol, dimethyl sulphoxide and polyethylene glycol-1500 effect on the liposome permeability way. Cooling in the presence of ethylene glycol and propylene glycol causes changes in liposome permeability with a local maximum at -18 degrees C. In the medium with 2M NaCl and ethylene glycol, liposomes were resistant to cooling. Dimethyl sulphoxide and polyethylene glycol-1500 induced a two-phase kinetics of changes in liposome permeability, the first phase being within the 0 = -9 degrees C and the second--within -9--25 degrees C temperature ranges. The found differences are supposed to be associated with the effect of the cryoprotective compounds on the lipid crystallization in a lower-temperatures range.  相似文献

Ionic liquids containing lithium methylsulfonyl group were prepared from the precursors poly(propylene glycol)-block-(ethylene glycol)-block-(propylene glycol)-bis(2-aminopropyl ether) with different molecular weight. These liquids revealed excellent electrical conductivity in the temperature range -25 to 85 degrees C. Also, they exhibited a high boiling temperature and hence a low vapor pressure in ambient condition. Additionally, they showed a high fluidity with their viscosities being comparative with that of water. To determine the sensitivity of an ethanol sensor by using these ionic liquids, these liquids were subjected into a sequential electrochemical tests with nickel electrodes which performed a high sensitivity for the ethanol sensor. It was found that only the derivative with low molecular weight could detect ethanol. Furthermore, a linear relationship between the response current and the concentration of ethanol was constructed. The detection limit was found to be 0.13% (v/v) and its response time was 336 s.  相似文献

Experiments on alcohol addicts blood were made to study the time course of the endogenous ethanol level after a single administration of mebicar (1.5 g), a derivative of bicyclic bisuria, 50 ml of 5% sodium hydroxybutyric syrup, a derivative of gamma-hydroxybutyric acid, and 20 mg diazepam, a derivative of 1,4-benzodiazepines. The clinical effect was recorded simultaneously. It was established that different tranquilizers stimulate the increase in the endogenous ethanol level as regards the spectrum of psychotropic activity. This effect was the most pronounced with mebicar and to a less measure with diazepam.  相似文献

Summary Methane-grown cells ofMethylococcus capsulatus andMethylosinus trichosporium readily oxidized propene and various isomers of butene to their respective epoxides. When examined in a proton NMR spectrum using tris([3-trifluoromethylhydroxymethylene]-d-camphorato), europium III derivative as an optically active chemical shift reagent, the products propylene oxide and 1,2-epoxybutane were found to contain equal amounts of both isomers. Methane-grown cells of both bacteria had considerable levels of reducing equivalents to catalyze the epoxidation of gaseous olefins. Cells depleted of reductants catalyzed the oxidation in the presence of low levels of methanol or formaldehyde with a stoichiometry of about 2:1. The rates of epoxidation of propene and 1-butene in a continuous reactor were 2–3-times that of a batch-wise reaction; the epoxidation activity, however, was lost within 3 h. The inactivation was attributed to the reactivity of the accumulated epoxides in the reactor. Propene and 1-butene oxidation by both bacteria were drastically inhibited by the respective products. Thus, the major problem in the application of microorganisms for production of epoxides from gaseous olefins is the rapid separation of the reactive products.  相似文献

An extracellular protease isolated fromBacillus subtilis AG-1 was investigated with respect to various detergents and formulation components. The enzyme had optimum at pH 8.0 and 60 °C temperature while zymographic study revealed two activity bands of 24.9 and 18 kDa. It showed high stability towards non-ionic (Tween 20, Tween 80, Triton X-100) and anionic surfactants sodium dodycyl sulfate (SDS), retaining 100 and 71% of its original activity. Another distinctive feature of the enzyme was its efficient stability towards hydrogen peroxide (H2O2) and sodium perborate and different commercial detergent brands. AG-1 protease was also examined for its activity/performance in combination with different stabilizers like glycerol, propylene glycol and polyethylene glycol (PEG). Enzyme showed a promising activity in the presence of this polyols especially PEG (8000). Whilst its compatibility with different commercially available powder and liquid detergents was also very interesting. These results suggest AG-1 protease as a good detergent compatible and can be utilized in the formulation of an environment friendly bio-detergent.  相似文献

合成了水杨酸铜·乙醇络合物,提纯了Tween-20、Tween-40,Tween-60.Tween-80,并测定了它们在pB7.4的磷酸缓冲液中的CMCO。用Cyt,c-HX-XO法分别测定了水杨酸铜·乙醇络合物清除超氧阴离子自由基的活性,不同浓度的单纯表面活性剂对O2的影响,以及表面活性剂与水杨酸铜·乙醇络合物协同清除O2的作用。观察到在胶束体系中水杨酸铜·乙醇络合物的SOD样活性明显高于缓冲液体系无表面活性剂时的SOD样活性,表面活性剂本身也具有清除O2的功能。抗红细胞膜脂质过氧化实验也得到类似结果。  相似文献

Propylene glycol, a compound metabolically active as a carbohydrate, is often employed as part of the vehicle for pharmacological preparations. Since the latter may be administered to experimental animals which are used in studies concerned with carbohydrate metabolism, the effects of small doses of propylene glycol on gluconeogenesis were determined.The intramuscular administration of propylene glycol provoked a dose dependent increase in liver glycogen, rate of glycogen synthesis, and blood glucose concentration. Maximal effects occured within 90 minutes and the values returned to control levels within 3 hours. Quinolinic acid, a weak inhibitor of basal gluconeogenesis, was found to markedly inhibit the increased gluconeogenesis resulting from propylene glycol administration.These findings suggest that the elevated gluconeogenesis produced by propylene glycol does not follow the same metabolic pattern as the basal gluconeogenesis and that rats receiving this compound cannot be considered as metabolic equivalents to untreated animals with respect to carbohydrate metabolism.  相似文献

Eosin B is used to estimate proteins above 1 microg/ml concentration [Waheed AA, Gupta, PD. Anal. Biochem. 1996:233:249-256; Waheed AA, Gupta PD. J. Biochem. Biophys. Meth. 1996;33:187-196]. In the present report we describe a method for estimating submicrogram quantities of proteins using the dye eosin Y. The increase in sensitivity of this assay is approximately two fold under optimal assay condition. The optimum concentration of eosin Y and citric acid for submicrogram assay is 0.01 and 0.05%; (final concentration) respectively. The protein-dye complex formation is completed within 2 min and its absorbance is stable up to 60 main with a variation of +/-4.0%. The interference due to sugars, reducing agents, glycerol and some neutral detergents like Triton X-100, NP-40 and Tween-20 is less than 12% whereas Brij-35, ethanol, acetone and chelators like EGTA and EDTA suppress the absorbance by about 12-18%. However, basic buffers like Tris, urea, CHAPS and NaN, interfere with the formation of the protein-dye complex. The increase in absorbance of protein-eosin Y complex compared to that of protein-eosin B complex is due to the higher extinction co-efficient of eosin Y compared to eosin B.  相似文献

Pregnancy rates after transfer of frozen bovine embryos: a field trial   总被引:1,自引:0,他引:1  
An efficient and practical technique for bovine embryo cryopreservation is a fundamental issue in the widespread use of embryo transfer. The present study shows results obtained in field experiments. In the first experiment, two slow-freezing methods using glycerol and a one-step method using ethylene glycol were compared: glycerol added in two steps (5 and then 10%), glycerol added in one step (10%) and 1.5 M ethylene glycol with direct transfer. The three methods were equally effective; pregnancy rates of 40.4, 39.1 and 45.4%, respectively were achieved. In the second experiment, using 1.5 M ethylene glycol with direct transfer, 20 and 5 min of equilibration of the cryoprotectant were tested. There were no observed significant differences in pregnancy rates. In the third experiment, ethylene glycol and propylene glycol were combined with three sucrose concentrations (0, 0.1 or 0.3 M) in a one-step method. It was observed that ethylene glycol and 0.1 M sucrose yielded the highest pregnancy rate, not differing from fresh controls. Similar pregnancy rates were noted after using multiple-step or one-step methods, but the one-step method is preferable due to its simplicity and applicability to field conditions.  相似文献

Body mass is a frequently used trait in ecological and evolutionary research. In the present study, I demonstrate that sampling and storage conditions affect wet and dry weights in an insect predator, Anchomenus dorsalis (Pontoppidan) (Coleoptera: Carabidae). Live beetles were placed in one of five preservative fluids for 1 month to simulate sampling by pitfall traps. Sodium chloride solution, ethylene glycol, ethylene glycol + detergent, and propylene glycol caused significant increases in both wet and dry weights compared with control (short‐term frozen) specimens, whereas formaldehyde did not. In a separate experiment, four methods of long‐term (6 months) sample storage (freezing, ethanol, propylene glycol, and ethyl acetate vapour) all caused significant changes in wet weight compared with the control treatment. The dry weight of the specimens preserved in ethanol decreased significantly in contrast to the long‐term frozen specimens and those in propylene glycol and ethyl acetate vapour, whose dry weight did not differ significantly from the control specimens. The combination of formaldehyde as the preservative fluid and freezing as the storage method thus appears to be an optimal combination for studies in which the body mass of dead insects is considered.  相似文献

Methoxyflurane (Metofane) has been widely used as an open-circuit anaesthetic in small laboratory animals for several decades. Its low vapour pressure and high blood solubility have permitted its use in convenient and simple drop-chamber/nose-cone setups. Recently, following the decision by the primary manufacturer to discontinue production, it has become increasingly difficult to obtain methoxyflurane. We describe here a simple and effective adaptation of isoflurane, an excellent inhalation anaesthetic, to open-circuit drop-chamber/nose-cone anaesthesia. It was found that the vapour concentration of isoflurane could be continuously varied by dissolving the anaesthetic in propylene glycol and that a 20% solution produced effective anaesthesia such that in adult mice, 2 ml of 20% isoflurane in propylene glycol induced anaesthesia within 2 min in a one-litre drop chamber. Furthermore, anaesthesia maintenance with 20% isoflurane was tested in two sets of mice. In one set, surgical plane anaesthesia was maintained for 10 min in a head chamber. After removal of the chamber, the animals awoke within one minute and recovered without any indication of post-anaesthetic distress. The second set contained pregnant mice; here anaesthesia was maintained for between 10 and 12 min, during which laparotomy, exposure of one uterine horn, intrauterine injection and wound closure were completed. The recovery from anaesthesia was also within a minute and with no signs of distress. Healthy litters were delivered after a normal gestation. This isoflurane/propylene glycol procedure is simple, effective and humane, and is a good substitute for methoxyflurane.  相似文献

We studied the ability of frozen-thawed mouse morulae to develop in vitro when the cryoprotectant proteins were substituted with one of the following nonorganic macromolecules: polyvinyl alcohol (PVA), polyvinyl pyrrolidone (PVP), and ficoll. We also determined how these agents interacted with 3 different cryoprotectants: glycerol (GLY), propylene glycol (PG), and ethylene glycol (EG). The influence of both of the above factors was measured on the basis of post-thaw morphological appearance, the percentage of development to the expanded blastocyst stage and the total cell count. Morulae (n=950) were collected from superovulated mice. Those classified as good or excellent were distributed among the 12 different freezing solutions, obtained by combining the 3 cryoprotectants with the 4 macromolecules (the 3 mentioned above, plus a control of 5% fetal calf serum) in phosphate buffered saline (PBS). Embryos frozen in PVA, PVP and ficoll tended to be a little difficult to recover from the straws. Development to the expanded blastocyst stage was significantly lower (P<0.05) in propylene glycol (43.6%) than in ethylene glycol (79.5%) or in glycerol (76.1%). Polyvinyl alcohol provided a higher survival rate when combined with glycerol (90.3) or ethylene glycol (95.0), but when it was combined with propylene glycol, only 56.5% of embryos survived after thawing. A positive interaction was observed between glycerol and PVA and between ethylene glycol and PVA or ficoll. The results indicate that fetal serum could be successfully substituted for any of the 3 chemically defined macromolecules. However, our findings also suggest that the use of PG as a cryoprotectant should be avoided when mouse morulae are frozen using the quick freezing method.  相似文献

The immunoreactivity of human plasma apolipoprotein C-II was investigated using a specific radioimmunoassay. In whole plasma, the mean value quantitated was 2.21 ± 0.415 mg/dl, while in delipidated plasma, a mean value of 3.84 ± 1.186 mg/dl was obtained, suggesting that the antigenic sites of the apolipoprotein were not fully detected in unmodified plasma by our antibody preparation. Two detergents, Tween-20 and Triton X-100, were studied to determine if they could enhance the immunoreactivity of apolipoprotein C-II in whole plasma. At concentrations of 0.012–0.06%, Tween-20 markedly increased the immunoreactivity of whole plasma, but not of delipidated plasma, indicating that antigenic sites of plasma apolipoprotein C-II has been exposed by Tween-20. In contrast, Triton X-100 had no effect on the immunoreactivity of whole plasma apolipoprotein C-II. A radioimmunoassay conducted in the presence of 0.06% Tween-20, resulted in a mean value in whole plasma (3.39 ± 1.11 mg/dl) that was not significantly different from that obtained when the assay was done on delipidated samples. The immunoreactivity of VLDL apolipoprotein C-II was also drastically enhanced following lipolysis by bovine milk lipoprotein lipase, supporting the hypothesis that antigenic sites are masked by the lipids. Finally, the mechanism responsible for the effect of Tween-20 on apolipoprotein C-II immunoreactivity was investigated. The results obtained from circular dichroism and ultracentrifugation suggest that the detergent may dissociate the apolipoprotein from lipoprotein particles, thus fully exposing the antigenic sites for reaction with antibodies.  相似文献

With the method of dynamic light scattering it was shown that the average size of micelles in the series of formulations based on various clindamycin salts, i. e. ClindHCl+Tween-20, ClindBz+Tween-20, ClindHCl+Cremafor-EL and ClindBz+Cremafor-EL increased from 6 to 20 nm. Investigations with the agar diffusion method revealed that the bactericidic action of the micelle-capsulated therapeutics did not depend on the micelle size within 6 to 20 mn. The concentration of the micellar clindamycin or gentamicin equal to 0.05 mcg/ml was bacteriostatic with respect to Micrococcus (Sarsina) luteus.  相似文献
