Experiments were performed with field-grown artichoke ( Cynara scolymus L.) cv. Capitan to determine whether the black spot disorder was associated with calcium deficiency in receptacles. Ageing of the plants and size of heads contributed to lower Ca levels of receptacles. Receptacles with lower amounts of Ca were more affected by the disorder. Globe artichoke infection by artichoke latent virus (ALV) and/or broad bean wilt virus strain french artichoke (BBWV-FA) resulted in decreases in both the relative water content (RWC) of leaves and the Ca content of receptacles, and in increased black spot incidence. In addition, a wind-screen and an anti-trans-pirant spray (composed of a wax emulsion and a spreader/sticker surfactant) increased the RWC of leaves and bracts and the Ca content of receptacles, and also significantly reduced the incidence of necrosed buds.
These data support the view that Ca deficiency is involved in the occurrence of black spot disorder.  相似文献

Virus-free and artichoke latent virus (ALV) infected plants of ‘Brindisino’ artichoke obtained by in vitro propagation, were studied over a period of three years to evaluate the effect of ALV infection on artichoke field performance and to determine the infection rate of healthy plants. ALV infection caused qualitative and quantitative changes ‘Brindisino’ artichoke such as leaf and bract discoloration, opening of head apex, delay of first harvest, shortening of head stalk, reduction of head width and a dramatic decrease of yield. Due to the high infection rate of healthy plants during the trial period, the differences in the production of heads between the two groups of plants decreased in the last year of experiment.  相似文献

An isolate of artichoke latent virus (ALV-I) obtained from a symptomless artichoke plant in Southern Italy was characterised and compared with ALV isolates from other countries. ALV occurs in California and throughout the western part of the Mediterranean basin but of Mediterranean countries east of Italy, it was found only in Israel and Turkey. ALV-I was readily transmissible by inoculation of sap to a moderate range of hosts, was transmitted in a non-persistent manner by Aphis fabae, Brachicaudus cardui and Myzus persicae, but was not seed transmitted. The virus has flexuous rod-shaped particles measuring c. 12 nm × 746 nm with a sedimentation coefficient of 145 S and a buoyant density of 1·31 g/cm3. The particles contain single stranded RNA with a mol. wt of 3 × 106 and protein composed of a single polypeptide species with a mol. wt of 33 000. Cylindrical cytoplasmic inclusions consisting of pinwheels and laminated aggregates were present in cells of naturally and artificially infected plants. ALV isolates from different geographical origin were indistinguishable from ALV-I biologically, morphologically, serologically and ultrastructurally. These properties place ALV in the Potyvirus group, but it was serologically unrelated to 12 other potyviruses 10 of which occur commonly in Italy.  相似文献

The ability of globe artichoke to produce inflorescences (capitula) during the autumn when the market price is highest is lost when plants are propagated from seed, as are most F1 hybrid cultivars. To gain an understanding of the phenology of seed‐propagated globe artichoke, both vernalised and non‐vernalised seedlings grown from open pollinated progeny of ‘Spinoso sardo’ plants were transplanted into the field at two monthly intervals covering a whole year. Final leaf number and the date of unfolding of each leaf were used to calculate the phyllochrons. The average final leaf number increased from 24 to 88 in response to increasing daylength above an approximately 11 h threshold during the first 30 days after emergence, and this variate explained about 90% of the variation in the thermal time taken to reach the bolting stage. The average phyllochron ranged from 56 to 96°Cd among the six emergence dates and was the major underlying physiological cause of the loss of early flowering shown by seed‐propagated plants. Genotypic variation for phyllochron within an emergence date was strongly associated with variation for the length of the emergence to bolting stage period. The limited effect of vernalisation on development indicated that the late flowering tendency of seed‐propagated plants cannot be attributed to an unfulfilled cold requirement.  相似文献

An indirect Dot-ELISA was compared with DAS-ELISA for detecting Artichoke Latent Virus (ALV) both in purified preparations and in crude sap from “Spinoso sardo” artichoke leaves. Antigen diluted samples (1 μl) were first spotted on nitrocellulose (NC) and polyvinylidene difluoride (PVDF) membranes. After blocking, the membranes were incubated in rabbit anti-ALV IgG, then in goat anti-rabbit IgG—alkaline phosphatase conjugate, and finally exposed to substrate and examined for a coloured precipitate. The minimum detection levels for ALV by Dot-ELISA were 125 pg of purified virus and 1/2,000 dilution of virus-infected sap on NC, and 83.3 pg of purified virus and 1/4,000 dilution of virus-infected sap on PVDF, as compared with 50 ng of purified virus and 1/1,000 dilution of virus-infected sap detectable by DAS-ELISA. This indirect Dot-ELISA proved to be more sensitive and more economical than DAS-ELISA, and can be completed in as little as 5—6 hours.  相似文献

The role of many insect species in crop pollination has been widely studied. The use of entomophilous pollination, commonly adopted for other crops, may be of key importance in order to improve seed yields in seed-propagated globe artichoke (Cynara cardunculus var. scolymus) cultivars. In this regard, in a 2-year field experiment, the role of two honeybees (Apis mellifera ligustica and A. mellifera siciliana) and one bumblebee (Bombus terrestris) was evaluated on the seed production of two globe artichoke cultivars in caged-protected environments. Overall, the number and weight of seeds plant−1 and head−1, 1,000 seed weight and fruit setting were higher in caged plants with imposed pollination than open field plants. The pollination efficiency was both insect and cultivar dependent. Both honeybee species performed better on the Mediterranean cultivar ‘Violetto di Sicilia’, while the bumblebee performed better on the Brazilian cultivar ‘NP4’. These results could be very useful to modernise the agronomic management of globe artichoke and reduce the costs of cultivation. In addition, such knowledge can be used to improve the seed production in globe artichoke seed-propagated cultivars and cardoon. This will benefit the bioenergy, nutraceutical, cosmetic and ornamental applications.  相似文献

The DNA polymerase from Amherst pheasant virus (APV), a member of the pheasant virus species of retroviruses, was compared to the DNA polymerases of avian leukosis viruses (ALV) and a reticuloendotheliosis virus (spleen necrosis virus (SNV)). Immunoglobulin inhibition tests and competition immunoassays showed that APV and ALV DNA polymerases are closely related at their active sites. The determinants common to their active sites are not shared by SNV DNA polymerase. Bu using a species-specific radioimmunoassay, it was shown that both APV and SNV DNA polymerases are grossly different from ALV DNA polymerase. The specificity of the relationship of the active sites of APV and ALV DNA polymerases was confirmed by a heterologous radioimmunoassay. Our data indicate that pheasant viruses are evolutionarily linked to ALV.  相似文献

Apple latent viruses were eliminated from, the tips of apple shoots by exposure to a temperature of 36 °C for various periods. The length of treatment needed to eliminate a particular virus differed from plant to plant, but viruses were always inactivated in the same order: first chlorotic leaf spot, followed by stem pitting and finally Spy decline. Quince plants developed sooty ring-spot and bark necrosis when inoculated with buds from some heat-treated apple clones infected with Spy decline virus. Only chlorotic leaf spot virus was transmitted to herbaceous hosts by sap extracts from apple leaves, petals and fruits, and returned from herbaceous plants to apple. This virus, isolated from either apple or cherry, caused a dark green mottle on peach leaves.  相似文献

Samples of the aroma volatiles of globe artichoke and Jerusalem artichoke were obtained by well established methods and were analysed by routine GC and GC/MS. Eight sesquiterpene hydrocarbons afforded the major group of components (over 42%) in globe artichoke samples, with β-selinene (ca 32%) as the main constituent. Previously reported caryophyllene could not be detected. α-Cedrene was found to have globe artichoke aroma characteristics on odour evaluation of separated components at an odour port at the exit of the GC column. Jerusalem artichoke samples contained one major component (β-bisabolene, ca 51%) and a range of saturated long-chain hydrocarbons (ca 22%). The sesquiterpene presumably contributes appreciably to the characteristic flavour of Jerusalem artichoke. Both types of artichoke gave a low concentration of total volatiles.  相似文献

The potential importance of the beet ringspot strain of tomato black-ring, a soil-borne virus, was assessed by growing stocks of Kerr's Pink potato for 1 year on infested land and subsequently on uninfested land. The incidence of infection in two stocks was 39 and 8% in the first year on uninfested land, and 29 and 5% after 2 years.
The virus was usually restricted to the roots of plants in the first year of infection, but a few plants showed black rings and spots in their leaves. In the second year, 20–55% of the plants grown from tubers set by symptomless, but infected, mother plants were infected: many of these showed leaf necrosis, others had stunted shoots, and cupped and distorted leaves; some were symptomless although systemically infected. In the third and fourth years, most of the progeny from plants which had symptoms or which were symptomless but systemically infected, contained the virus: nearly all such infected plants were stunted and distorted or were symptomless. Infection decreased the weight of tubers produced by plants with severe necrotic spotting but not the yield of plants with less necrosis. The number and weight of tubers per plant were decreased by 15 and 20% respectively, in symptomless systemically infected plants, and by 20 and 30% in stunted plants.  相似文献

Bacteria have been isolated from shoot tips of symptomless globe artichoke plants. These were identified as Pseudomonas fluorescens, Pseudomonas marginalis, Pseudomonas spp., Serratia liquefaciens, Enterobacter agglomerans/Erwinia, Agrobacterium radiobacter, an unidentified member of Rhizobiaceae and another classified in the “corynebacteria” group. The most frequently isolated species was P. fluorescens, biovars II and III. The endogenous character of these bacteria was studied in plants growing in vitro and in the open field. P. fluorescens, P. marginalis, S. liquefaciens and E. agglomerans/Erwinia caused symptoms in plants growing in vitro, but only P. fluorescens biovar II and P. marginalis produced symptoms in plants growing in open fields. Differences in pathogenicity were observed on inoculated plants growing in vitro or in the open field. This suggests that several endophytic bacterial species may be responsible for the high levels of contaminants found during the micropropagation of globe artichoke.  相似文献

Our objective was to determine whether the cell-associated herpesvirus vaccines used in chickens to control Marek's disease tumors can augment development of lymphoid leukosis (LL) induced by exogenous avian leukosis virus (ALV). Various single or mixed Marek's disease vaccines were inoculated at day 1, and ALV was injected at 1 to 10 days, with chickens of several experimental or commercial strains. Development of LL was monitored at 16 to 48 weeks in various experiments. In several strains of chickens we repeatedly found that the widely used serotype 3 turkey herpesvirus vaccine did not augment LL in comparison with unvaccinated controls. However, LL development and incidence were prominently augmented in several chicken strains vaccinated with serotype 2 vaccines, used alone or as mixtures with other serotypes. In one chicken strain, augmentation was demonstrated after natural exposure to ALV or serotype 2 Marek's disease virus viremic shedder chickens. Augmentation of LL by virulent or attenuated Marek's disease viruses of serotype 1 was intermediate in effect. Serotype 2 Marek's disease virus augmentation of LL was prominent in three laboratory lines and one commercial strain of White Leghorns, but it was not observed in an LL-resistant laboratory line or four commercial strains susceptible to ALV infection. Chickens developed similar levels of viremia and neutralizing antibodies to ALV regardless of the presence of augmentation of LL, suggesting that the mechanism of enhanced LL did not result from differences in susceptibility or immune response to ALV. We postulate that the serotype 2 herpesviruses may augment LL through one of several possible influences on bursal cells that are subsequently transformed by exogenous ALV.  相似文献

A virus with filamentous particles 697 nm in length was isolated from artichoke plants in Southern Italy and identified as a new possible member of Carlavirus group, for which the name artichoke virus M (AVM) is suggested. AVM could not be transmitted by sap inoculation to herbaceous hosts and was always present in artichoke in mixed infections with other viruses. Virus particles had a buoyant density in CsCl of 1.31 g × cm?3 and contained a single species of nucleic acid with an apparent size of 7.5 Kb and a single coat protein species with a mol. wt of 31,000. The virus was distantly related serologically to carnation latent and poplar mosaic carlaviruses but not to other members of the group including the recently described artichoke latent S carlavirus. Cytological alterations consisted of complex cytoplasmic inclusions composed of deranged organelles, lipid droplets and accumulations of membranes.  相似文献

A soil-borne inoculum of Mycosphaerella ligulicola was found to be mildly pathogenic to leaves or stems of globe artichoke, rudbeckia, zinnia, sunflower and dahlia, but severely pathogenic to lettuce. This is the first report of an infection by this fungus of plants other than chrysanthemum or pyrethrum. It was found that, with successive passages of the fungus through lettuce and chrysanthemum respectively, an increase in virulence to these hosts occurred. After a single passage through lettuce a reduction in virulence to chrysanthemum resulted, but with further passages through lettuce there was no further reduction in virulence to chrysanthemum.  相似文献

Scions from red raspberry (Rubus idaeus) plants naturally infected with an aphid- and sap-transmissible virus, code-named 52V, always induced apical necrosis in R. occidentals signifying the presence of black raspberry necrosis virus (BRNV), whereas plants free from 52V did not. These and other findings provide strong circumstantial evidence that 52V is an isolate of BRNV, the heat-labile member of the pair of viruses that together cause raspberry veinbanding mosaic disease. On grafting with R. idaeus scions containing a 52V culture of BRNV free from other detectable viruses, all of twenty-two red raspberry cultivars and four other Rubus species were infected symptomlessly but apical necrosis developed in R. henryi and R. molaccanus. Electron microscopy of thin sections of 52V-infected Chenopodium quinoa, R. henryii and R. occidentalis showed areas of dead cells in the vascular tissue and leaf blade. Some of the cells adjacent to these areas had cell wall outgrowths and many of the plasmodesmata contained virus-like particles c. 25 nm in diameter arranged in a single file.  相似文献

The identification of TVB(S3), a cellular receptor for the cytopathic subgroups B and D of avian leukosis virus (ALV-B and ALV-D), as a tumor necrosis factor receptor-related death receptor with a cytoplasmic death domain, provides a compelling argument that viral Env-receptor interactions are linked to cell death (4). However, other TVB proteins have been described that appear to have similar death domains but are cellular receptors for the noncytopathic subgroup E of ALV (ALV-E): TVB(T), a turkey subgroup E-specific ALV receptor, and TVB(S1), a chicken receptor for subgroups B, D, and E ALV. To begin to understand the role of TVB receptors in the cytopathic effects associated with infection by specific ALV subgroups, we asked whether binding of a soluble ALV-E surface envelope protein (SU) to its receptor can lead to cell death. Here we report that ALV-E SU-receptor interactions can induce apoptosis in quail or turkey cells. We also show directly that TVB(S1) and TVB(T) are functional death receptors that can trigger cell death by apoptosis via a mechanism involving their cytoplasmic death domains and activation of the caspase pathway. These data demonstrate that ALV-B and ALV-E use functional death receptors to enter cells, and it remains to be determined why only subgroups B and D viral infections lead specifically to cell death.  相似文献

An isolate of tomato black ring virus from artichoke (TBRV-A) was compared biologically, physico-chemically and serologically with three strains of the virus, i.e. TBRV-potato bouquet (TBRV-BU), TBRV-beet ringspot (TBRV-W), and TBRV-celery yellow vein (TBRV-Ce). Cytopathic effects of TBRV-A infection in C. quinoa and its relationships with two strains of artichoke Italian latent virus (AILV-S and AILV-G) were also investigated. Physical properties in vitro, sedimentation coefficients and molecular weight of protein subunits and nucleic acid species of TBRV-A were very similar to those known for TBRV. In serological tests, TBRV-A appeared more closely related to TBRV-W (SDI = 1) than to TBRV-Ce and TBRV-BU (SDI = 2–3). Finally, TBRV-A was very, distantly related to AILV-S and AILV-G (SDI = 11–12).  相似文献

Studies on the occurrence of tomato bushy stunt virus in English rivers   总被引:4,自引:0,他引:4  
Tomato bushy stunt virus (TBSV) of unknown source was isolated from water of the River Thames, near Oxford. The isolate designated TBSV-T was mechanically transmissible to several tomato (Lycopersicon esculentum) cvs and to other species including Petunia hybrida, pepper (Capsicum annuum). eggplant (Solanum melongena), Nicotiana clevelandii, Chenopodium amaranticolor and C. quinoa in which it caused systemic symptoms. It caused no infection of globe artichoke (Cynara scolymus) or Pelargonium domesticum. The virus was not adsorbed to soil and could be isolated from leachate of soil in which systemically-infected tomato or C. quinoa plants were grown. Tomato plants became infected when grown in soil watered with virus suspensions. TBSV-T was infective after 10 min at 80°C but not at 90°C and when diluted to 10-5 but not to 10-6. Purified virus preparations contained C. 30 nm isometric particles. In gel-diffusion serological tests, TBSV-T reacted with homologous anti-serum and with antiserum to petunia asteroid mosaic virus but not to pelargonium leaf curl virus. Seed-borne infection (50–65%) of TBSV was demonstrated in plants grown from seed of symptomlessly-infected tomato fruit. TBSV was isolated from symptomlessly-infected tomato fruit imported from Morocco during October-April 1981. One of the isolates (TBSV-M) was indistinguishable from TBSV-T in host range, symptomatology and serological reactions. TBSV was also found in tomato plants growing extraneously in primary settlement beds at sewage works; such plants having been derived from undigested seeds in sewage. Because of its ‘alimentary-resistance’ in man, it is possible that one ecological route whereby TBSV enters rivers is by man's consumption of TBSV-infected tomatoes and eventual sewage dispersal into rivers.  相似文献
