紫外激光消融主动脉斑块和感生荧光光谱观测   总被引:1,自引:0,他引:1  
利用XeCI准分子激光辐照主动脉血管正常组织和斑块组织,观察到组织被激光消融。消融所产生的凹坑与辐射时间呈对数直线关系。308nm激光感生的血管壁荧光光谱在可见波段出现二个荧光极大值,用二极值的相对强度之比可以判断正常或斑块组织。  相似文献

研究了Er:YAG激光消融离体牙本质过程中激光脉宽对消融阈值、消融深度以及消融后牙硬组织微结构的影响。使用50μs到1 ms脉宽的Er:YAG激光对人牙本质在无喷水冷却下消融实验,通过体式光学显微镜观察样品消融切口进而测量消融坑洞深度,利用扫描电镜(SEM)观察消融后牙本质的形态。实验结果表明,随着激光脉宽的增加,Er:YAG激光对人牙本质的消融阈值、消融深度呈现逐渐增大的趋势,同时还观察到高能量密度消融实验中牙小管封闭以及牙本质熔融现象。该实验结果对了解Er:YAG激光消融牙硬组织的微观过程及牙科治疗应用具有参考价值。  相似文献

激光消融技术在生物骨硬组织领域具有广泛的应用前景.选用脉冲CO2激光对离体牛胫骨组织进行消融,以电钻作为参照组,体视显微镜和扫描电子显微镜观察骨组织弹坑的形态学变化,光学相干层析成像(OCT)技术测量弹坑消融深度.比较结果表明:脉冲CO2激光辐照骨硬组织后产生的消融凹陷在几何形状和形态学方面与电钻的磨削效果相类似.此外,喷水速率为31 mL/min时可减少热损伤,并提高消融效率.  相似文献

研究正常人膀胱和膀胱癌组织在Kube lka-Munk二流模型下对476.5 nm,514.5 nm和808 nm波长的激光的光学特性的差异。采用双积分球系统和Kube lka-Munk二流模型进行测量研究。实验结果表明,正常膀胱和膀胱癌组织在Kube lka-Munk二流模型下对476.5 nm,514.5 nm和808 nm波长的每一个波长的激光的吸收、散射、总衰减、有效衰减系数都有非常显著性的差异(P<0.01)。膀胱癌组织对476.5 nm,514.5 nm和808nm波长的激光的吸收系数明显地较正常膀胱组织对相应波长的激光的吸收系数要大(P<0.01),膀胱癌组织对476.5 nm和514.5 nm波长的激光的散射系数明显地较正常膀胱组织对相应波长的激光的散射系数要小(P<0.01),而膀胱癌组织对808 nm波长的激光的散射系数明显地较正常膀胱组织对同一波长的激光的散射系数要大(P<0.01)。膀胱癌组织对476.5 nm,514.5 nm和808 nm波长的激光的总衰减系数明显地较正常膀胱组织对相应波长的激光的总衰减系数要大(P<0.01),膀胱癌组织对476.5 nm,514.5 nm和808 nm波长的激光的有效衰减系数明显地较正常膀胱组织对相应波长的激光的有效衰减系数要大(P<0.01)。提示使用双积分球系统和Kube lka-Munk二流模型来确定离体的正常膀胱组织和膀胱癌组织对476.5 nm,514.5 nm和808nm波长的激光的光学特性的差异鉴别诊断病变的膀胱组织是一个有效的方法。  相似文献

目的探讨miR-143在人颈动脉粥样硬化患者斑块组织中的表达变化及可能的作用机制。方法收集我院血管外科因颈动脉狭窄行颈动脉内膜剥脱术(CEA)切除的颈动脉斑块组织及斑块旁内膜组织56例,根据术前颈动脉超声检查及术后斑块病理学检查将所取得的斑块组织又分为稳定斑块组26例和不稳定斑块组30例。另外选择同期年龄、性别相匹配的在我院因外伤、门脉高压或原发性血小板增多症而行脾脏切除术患者的正常的脾动脉内膜组织20例作为正常对照组,通过实时荧光定量PCR(qRT-PCR)检测组织中miR-143的表达水平。体外培养人主动脉血管平滑肌细胞(HAVSMCs),分别转染mi R-143模拟物、模拟物阴性对照、miR-143抑制物、抑制物阴性对照,采用qRT-PCR检测转染后HAVSMC细胞miR-143表达水平的变化,CCK-8法和Transwell迁移实验分别检测转染后细胞增殖活性和迁移能力的变化。通过生物信息学预测miR-143的可能靶基因,双荧光素酶报告基因实验观察miR-143与ADAMTS4的靶向调控关系,Western blot观察miR-143对ADAMTS4表达水平的影响。结果颈动脉粥样硬化斑块组织中miR-143表达水平明显低于斑块旁内膜组织和正常动脉血管内膜组织,并且不稳定斑块组患者斑块组织中miR-143的表达水平明显低于稳定斑块组斑块组织。转染miR-143模拟物后,HAVSMCs中miR-143表达水平明显升高,增殖活性和迁移能力明显降低;转染miR-143抑制物后,HAVSMCs中miR-143表达水平明显降低,增殖活性和迁移能力明显升高。双荧光素酶报告基因显示ADAMTS4是miR-143的直接靶基因,Western blot显示miR-143能够在转录后水平负向调控ADAMTS4表达。结论 miR-143在颈动脉粥样硬化斑块组织中表达降低,其可能通过靶向调控ADAMTS4影响血管平滑肌细胞增殖和迁移能力,与颈动脉斑块的形成和不稳定性有关。  相似文献

采用双积分球系统和光辐射测量技术的基本原理 ,以及运用生物组织的光学模型 ,研究了 5 32nm和80 8nm激光及其线偏振激光辐照人正常膀胱和膀胱癌组织的光学特性 .结果表明 :膀胱癌组织对同一波长的激光或其线偏振激光的衰减明显较正常膀胱组织的要大 ,膀胱癌组织对 5 32nm和 80 8nm激光的衰减均较其线偏振激光的要略大一些 .膀胱癌组织对 5 32nm和 80 8nm激光及其线偏振激光的衰减明显较正常膀胱组织的要大 .正常膀胱或膀胱癌组织对同一波长的激光及其线偏振激光的折射率均没有明显的差异 ,膀胱癌组织对 5 32nm和80 8nm激光的折射率比正常膀胱的明显要大 .Kubelka Munk二流模型下 ,两种组织对同一波长的激光或其线偏振激光的光学特性均有显著性差异 (P <0 0 1) .同一组织对不同波长的激光及其线偏振激光的光学特性也有显著性差异 (P <0 0 1) ,正常膀胱组织对同一波长的激光及其线偏振激光的光学性有明显差异 ,而膀胱癌组织对同一波长的激光及其线偏振激光的光学特性则没有明显差异 .膀胱癌组织对 5 32nm和 80 8nm激光及其线偏振激光的前向散射通量i (x)、后向散射通量 j (x)、总散射通量I (x)的衰减均较正常膀胱组织的明显要大得多 ,且其i (x)均明显较j (x)要强  相似文献

动脉粥样硬化斑块的微区拉曼光谱检测   总被引:1,自引:0,他引:1  
为探讨动脉粥样硬化斑块的微区拉曼光谱特征,以球囊损伤日本长耳白兔右侧颈总动脉后予以高脂饮食喂养,在实验过程中监测体重和血脂变化情况。3个月后,以中国斑点蝰蛇毒和组胺加以触发使斑块破裂,将动物处死并查找动脉硬化斑块,动脉组织经大体病理分类后,进行微区拉曼光谱及病理检测。结果显示:动脉粥样硬化斑块的拉曼光谱图在1450及1660cm-1处均有明显的胆固醇等脂质特征峰。特征峰曲线下相对面积统计结果表明:明显动脉粥样硬化斑块的谱峰下相对面积(5.80×10-3±3.51×10-3)显著高于轻度动脉粥样硬化组织(2.01×10-3±1.49×10-3)及正常动脉组织(1.01×10-3±0.94×10-3),P<0.05。正常动脉组织拉曼光谱曲线较光滑,无明显特征峰。血栓形成处拉曼光谱图荧光背底较强,未见特征谱峰。该研究结果证明微区拉曼光谱可以对动脉粥样硬化斑块的胆固醇等脂质含量进行特异性定量检测,表明微区拉曼光谱是评估动脉粥样硬化程度及斑块稳定性的可行方法。  相似文献

人体鼻咽组织的时间分辨自体荧光光谱研究   总被引:1,自引:1,他引:0  
实验研究了在397 nm半导体脉冲激光激发下,人体离体鼻咽正常和癌变组织在600 nm荧光发射波长处的时间分辨自体荧光光谱特性。利用双指数衰减方程对时间分辨自体荧光光谱进行拟合后,获得相应的荧光强度随时间的指数衰减方程以及荧光平均寿命。人体鼻咽癌变和正常组织在600 nm处的自体荧光平均寿命分别为(2.94±0.51)ns和(4.29±0.71)ns,两者之间存在显著的差异。应用时间分辨光谱技术的诊断灵敏度和特异性分别为75%和100%。初步表明了时间分辨自体荧光光谱在早期鼻咽癌诊断的应用价值,该方法可望与传统的稳态荧光光谱结合起来,进一步提高早期鼻咽癌荧光诊断的准确率。  相似文献

The response of living tissue to surgical lasers was studied numerically. An algorithm computed the crater boundaries formed by a single laser pulse and the thermochemical damage around this crater. Heat conduction and beam attenuation by tissue vapors were found to be the major factor in the reduction of cutting efficiency.  相似文献

Summary Further investigations on the applicability of the laser technique for qantitative purposes in histochemistry were conducted. Special attention has been paid to the relationship between the volume of the crater burnt out in the tissue by laser ray treatment and the photometric readings of the spectral bands. A statistical evalution of the measurements of crater volumes for quantitative histochemistry as well as determination of the error of the former method used and previously described (Kozik et al., 1970b) have been performed. Special agar standards containing known amounts of Pb(NO3)2 and appropriately prepared photographic plates were used. From the results obtained it appeared that the variances calculated for the differences between density of spectral bands for the Pb present in the investigated sample and the density of the respective band for the Ag present in the photographic plate fall in the range of between 14.1 and 19.7%, whereas the error of the method in which the density of spectral bands was related to the crater volumes covered a range from 3.45–7.7%.Based on the discussion of results the authors conclude that both methods of acP assay in histological slices by means of laser microanalysis may be successfully used.  相似文献

Encephalomyocarditis (EMC) virus (K-2 strain) was separated into two distinct peaks of optical density by chromatography on calcium phosphate, eluting at 0.17 and 0.31 m, respectively. The virus in these peaks could not be distinguished by infectivity (plaque-forming units), hemagglutinin activity, electron microscopic appearance, sedimentation coefficient, or base ratios. However, variants producing large plaques predominated in the first peak eluted, whereas variants producing small plaques were concentrated in the second peak. When plaque-purified large and small plaque variants were chromatographed individually, each gave a single peak eluting at 0.16 and at 0.43 m, respectively. However, one small plaque type isolated was eluted in the 0.17 m region expected for large plaque variants, and not with the bulk of the small plaque variants. There were indications that EMC virus (K-2 strain) could be separated into more than two peaks by use of selected elution gradients. The possibility that there may be a relationship between plaque size and point of elution from calcium phosphate is discussed.  相似文献

The stigma of Caesalpinia pulcherrima is crateriform. The crater continues as a slit-like canal through the style and into the ovary. Both crater and canal are lined by several layers of fusiform and thin-walled cells which are continuous in two narrow regions in the ovary. Postanthesis and before pollination, the middle lamella of cells lining the stigmatic crater and stylar transmitting tissue undergoes dissolution. This occurs in a progression down the style with cells separating partially or wholly from neighbours. Dissolution is initiated at intercellular junctions where wall fibrils loosen and variously-sized and -shaped holes appear. Cytoplasmic changes include increased dictyosome activity, increased rough and smooth endoplasmic reticulum at the periphery of cells and accumulation of electron opaque deposits at the plasma membrane. The crater fills with stigmatic fluid and the diameter of the stylar canal increases. Pollen germinates in the secretion-filled crater, and pollen tubes grow down the style between the cells of the transmitting tissue but do not enter the canal. They emerge at the entrance to the ovary cavity and grow over one or two narrow strips of ovarian transmitting tissue cells which are present throughout the length of the ovary close to the ovules. This ensures that tubes grow in close proximity to the micropyles.  相似文献

Serum was obtained from 25 male renal cell carcinoma (RCC) patients and 21 healthy males. O-glycans were released by a β-elimination reaction and purified by graphitized carbon cartridge solid phase extraction, then profiled by matrix-assisted laser desorption/ionisation-time of flight mass spectrometry. After noise removal and peak alignment, 1372 peaks were extracted from 200000 data points. Feature peaks were analyzed by calculation of differential sensitivity and specificity. The combination of two feature peaks was chosen as a biomarker and could clearly differentiate RCC and normal samples in our study group.  相似文献

无损光声成像技术结合了纯光学成像高选择特性和纯超声成像中深穿透特性的优点,克服了光散射限制,实现了对活体深层组织的高分辨、高对比度成像。该成像技术对内源物质例如脱氧血红蛋白、含氧血红蛋白、黑色素、脂质等进行成像,提供了活体生物组织结构和功能信息,已经在生物医学领域表现出巨大的应用前景。然而,很多与病理过程相关的特征分子的光吸收能力较弱,在活体环境中难以被光声成像系统所识别,从而限制了光声成像技术的应用范围。基于功能纳米探针的光声成像-光声分子成像极大拓展光声成像的应用范围,可以在活体层面对病理过程进行分子水平的定性和定量研究,将为实现目标疾病的早期诊断提供强大的技术支持。本文发展在近红外具有窄吸收线宽(半高宽仅为60 nm)的纳米金锥作为新型的光声探针。通过选择不同径长比的纳米金锥,可以任意调节纳米金锥的吸收峰。通过调谐激光器的波长,可实现对不同吸收峰纳米金锥的选择性激发。纳米金锥将有可能用于多光谱光声成像,实现对不同靶标的目标分子探测。  相似文献

It has recently been shown that bimodal histograms with false aneuploid peaks may be obtained by DNA flow cytometry from histologically normal tissue allowed to autolyze. To investigate if such peaks can be generated from surgically excised archival tissue, 198 paraffin blocks from 179 patients containing histologically normal spleen (n = 65), liver (n = 26), thyroid (n = 32), pancreas (n = 19), salivary gland (n = 49), or lymph node tissue (n = 7), obtained from the archives of two university pathology departments, were analyzed for nuclear DNA content. The great majority (n = 160, 83.8%) of the 191 interpretable histograms had a single symmetrical G1 peak; and 8 histograms, all produced from liver tissue had a tetraploid pattern. A slight or a prominent repeatable deviation in the G1 peak outline was present in 14 (7.3%) cases. A peak resembling an aneuploid G1 peak with a DNA index (DI) ranging from 1.14 to 1.38 was repeatedly produced from 9 (4.7%) blocks containing histologically normal or inflamed splenic (n = 3), pancreatic (n = 3), liver (n = 1), thyroid (n = 1), or lymph node (n = 1) tissue. The three abnormal peaks produced from pancreatic tissue were rounded in shape and resembled closely the ones that can be obtained from autolytic pancreatic tissue, and the six remaining extra peaks were all fused with the "diploid" peak. In conclusion, false peaks, probably caused by degradation of the nuclear contents during formalin fixation or before it, may rarely be obtained from surgical paraffin-embedded samples.  相似文献

目的:利用表面增强激光解吸电离飞行时间质谱技术(SELDI-TOF-MS)筛选慢性阻塞性肺疾病(COPD)血清特异标志物。方法:应用SELDI-TOF-MS技术检测30例COPD稳定期患者和30例健康对照者血清蛋白指纹图谱,采用Biomarker pattern软件进行分析,建立COPD的诊断模型。结果:COPD患者血清蛋白图谱与对照组相比,在相对分子质量2000-15 000范围内共检测到75个蛋白峰,发现19个有统计学差异的蛋白峰(P0.05)。通过对COPD组与对照组间的数据作进一步分析,经BPS软件分析,建立质荷比(M/Z)3 167、4 645的差异蛋白组成的诊断模型,其诊断敏感度为96.67%,特异度为96.67%。结论:SELDI-TOF-MS技术是一种快速、简单易行、用量少和高通量的分析方法。能直接筛选出COPD血清中特异表达标志物,用特异表达标志物建立的诊断模型能有效区分COPD患者与健康对照者,有望成为COPD诊断的辅助指标。  相似文献
