The chemical investigation on endophytic fungus Annulohypoxylon cf. stygium in leaves of Anoectochilus roxburghii (Wall.) Lindl . has been performed. Sixteen compounds were isolated and their structures were identified as (?)‐notoamide A, (?)‐notoamide B, (+)‐versicolamide B, notoamide C, notoamide D, stephacidin A, sterigmatocystin, dihydrosterigmatocystin, secosterigmatocystin, versiconol, averufanin, kipukasin D, kipukasin E, diorcinal, palmarumycin CP2 and (?)‐(3R)‐mellein methyl ether, respectively, by spectroscopic analysis and comparison with literature data. All the compounds were isolated from Annulohypoxylon genus for the first time. Sterigmatocystin and palmarumycin CP2 showed selective cytotoxic activities against HepG2, HeLa, MCF‐7 and HT‐29.  相似文献

Two new pairs of enantiomeric butenolides, (+)- and (−)-suberiteslide A, (+)- and (−)-subertieslide B had been obtained from the marine sponge Suberties sp. The structures with absolute configurations of these compounds were unequivocally determined by spectroscopic analyses and ECD (Electronic Circular Dichroism) method. It was the first separation of butenolides from the marine sponges of genus Suberites. Additionally, the anti-inflammatory, antibacterial and cytotoxic activities of these compounds were evaluated. The result indicated that only (−)-subertieslide B showed weak anti-inflammatory activity with the IC50 value of 40.8 μM.  相似文献

Two new butenolides, (4S)‐4,10‐dihydroxydodec‐2‐en‐1,4‐olide ( 1 ) and (4S)‐4,8,10‐trihydroxy‐10‐methyldodec‐2‐en‐1,4‐olide ( 2 ), together with three known compounds, MKN‐003B ( 3 ), MKN‐003C ( 4 ), and cyclo(Ala‐Leu) ( 5 ), were isolated from the culture broth of a bacterium of the genus Streptomyces derived from soil environment. The structures of these compounds were elucidated on the basis of spectroscopic analysis. The inhibitory activities of the butenolides against eight pathogenic fungi were evaluated. All of the butenolides showed moderate‐or‐weak antifungal activities in a broth microdilution assay.  相似文献

Many actinomycetes, newly isolated from soil, converted (?)-carvone to either (?)-trans-carveol or (?)-cis-carveol or to a mixture of both compounds, and known metabolic products, such as (+)-dihydrocarvone, (+)-isodihydrocarvone, (+)-neodihydrocarveol, (?)-dihydrocarveol, (+)-isodihydrocarveol and (+)-neoisodihydrocarveol.

Identification of the metabolic products and the metabolic pathways of (?)-carvone were described in a strain of Streptomyces, A-5–1 and a strain of Nocardia, 1-3-11. A summary shows the reaction mechanism pattern of (?)-carvone conversion by actinomycetes.  相似文献

A new sesquilignan, euonymolin A ( 1 ), and six known lignans, (?)‐de‐O‐methylmagnolin ( 2 ), (+)‐de‐O‐methylepimagnolin A ( 3 ), (+)‐syringaresinol ( 4 ), (+)‐pinoresinol ( 5 ), (+)‐medioresinol ( 6 ), and (+)‐lariciresinol 4′‐Oβ‐d ‐glucopyranoside ( 7 ), were isolated from the twigs of Euonymus alatus (Thunb .) Siebold (Celastraceae). The structures of the isolated compounds were elucidated based on spectroscopic analyses, including extensive 1D‐ and 2D‐NMR techniques, HR‐MS analysis and circular dichroism (CD) data, and the literature data. All of the isolated compounds were evaluated for antiproliferative activity against A549, SK‐OV‐3, SK‐MEL‐2, and HCT‐15 cell lines and inhibition of nitric oxide (NO) production in a lipopolysaccharide (LPS)‐activated BV2 cell line. All compounds showed cytotoxicity against the SK‐MEL‐2 cell line with IC50 values of 23.24 – 48.14 μm and inhibited NO production in LPS‐activated BV‐2 cells with IC50 values of 6.75 – 23.53 μm .  相似文献

Three new indole diketopiperazine alkaloids, 11‐methylneoechinulin E and variecolorin M, and (+)‐variecolorin G, along with 12 known analogs, were isolated from a soft coral‐associated epiphytic fungus Aspergillus sp. EGF 15‐0‐3. The structures of the new compounds were unambiguously established by extensive spectroscopic analyses including HR‐ESI‐MS, 1D and 2D NMR spectroscopy and optical rotation measurements. The absolute configurations of (+)‐ and (?)‐variecolorin G were determined by experimental and quantum‐chemical ECD investigations and single‐crystal X‐ray diffraction analysis. Variecolorin G is a pair of enantiomeric mixtures with a ratio of 1 : 2. Moreover, (+)‐neoechinulin A is firstly reported as a natural product. The cytotoxic activities of all the isolated compounds against NCI‐H1975 gefitinib resistance (NCI‐H1975/GR) cell lines were preliminarily evaluated by MTT method.  相似文献

(+)‐ and (?)‐Xanchryones F and G ((+)‐ and (?)‐ 1 and 2 ) were isolated from the plant Xanthostemon chrysanthus by chiral separation. Compounds 1 and 2 featured a new carbon skeleton with cinnamoyltriketone‐flavone adducts. Their structures with absolute configurations were elucidated by detailed spectroscopic analyses and chemical calculations. Two pairs of new non‐brominated racematic pyrrole derivatives, (±)‐nakamurine D ( 1 ) and (±)‐nakamurine E ( 2 ), two new diterpene alkaloids, isoagelasine C ( 16 ) and isoagelasidine B ( 21 ), together with 13 known pyrrole derivatives ((±)‐ 3  –  15 ), five known diterpene alkaloids ( 17  –  20 , 22 ) were isolated from the South China Sea sponge Agelas nakamurai . The racemic mixtures, compounds 1  –  4 , were resolved into four pairs of enantiomers, (+)‐ 1 and (?)‐ 1 , (+)‐ 2 and (?)‐ 2 , (+)‐ 3 and (?)‐ 3 , and (+)‐ 4 and (?)‐ 4 , by chiral HPLC . The structures and absolute configurations were elucidated on the basis of comprehensive spectroscopic analyses, quantum chemical calculations, quantitative measurements of molar rotations, application of van't Hoff 's principle of optical superposition, and comparison with the literature data. The NMR and MS data of compound 3 are reported for the first time, as the structure was listed in SciFinder Scholar with no associated reference. These non‐brominated pyrrole derivatives were found in this species for the first time. Compound 18 showed valuable cytotoxicities against HL ‐60, K562, and HCT ‐116 cell lines with IC 50 values of 12.4, 16.0, and 19.8 μm , respectively. Compounds 16  –  19 , 21 , and 22 showed potent antifungal activities against Candida albicans with MIC values ranging from 0.59 to 4.69 μg/ml. Compounds 16  –  19 exhibited moderate antibacterial activities against Proteusbacillus vulgaris (MIC values ranging from 9.38 to 18.75 μg/ml).  相似文献

Tetrahydroberberine (THB), a racemic mixture of (+)‐ and (?)‐enantiomer, is a biologically active ingredient isolated from a traditional Chinese herb Rhizoma corydalis (yanhusuo). A chiral high performance liquid chromatography method has been developed for the determination of THB enantiomers in rat plasma. The enantioseparation was carried out on a Chiral®‐AD column using methanol:ethanol (80:20, v/v) as the mobile phase at the flow rate 0.4 ml/min. The ultraviolet detection was set at 230 nm. The calibration curves were linear over the range of 0.01–2.5 μg/ml for (+)‐THB and 0.01‐5.0 μg/ml for (?)‐THB, respectively. The lower limit of quantification was 0.01 μg/ml for both (+)‐THB and (?)‐THB. The stereoselective pharmacokinetics of THB enantiomers in rats was studied after oral and intravenous administration at a dose of 50 and 10 mg/kg racemic THB (rac‐THB). The mean plasma levels of (?)‐THB were higher at almost all time points than those of (+)‐THB. (?)‐THB also exhibited greater Cmax, and AUC0–∞, smaller CL and Vd, than its antipode. The (?)/(+)‐enantiomer ratio of AUC0–∞ after oral and intravenous administration were 2.17 and 1.43, respectively. As a part of a project designed to investigate Colchicum species in Jordan, the chemical constituents of Colchicum crocifolium Boiss. (Colchicaceae) were investigated using LC‐MS and LC–UV/Vis PDA. A decision tree for working with colchicinods has been developed by incorporating data from LC‐UV/PDA and LC‐MS. This dereplication strategy draws upon the UV/PDA spectra to classify compounds into one of four structural groups and combines this with retention time and mass spectra/molecular weight to identify the compounds. This strategy was applied on a small amount of extract (2 mg) of Colchicum crocifolium to dereplicate 10 known compounds from four different structural groups, namely (?)‐demecolcine, 2‐demethyl‐(?)‐colchicine or3‐demethyl‐(?)‐colchicine, N‐deacetyl‐(?)‐colchicine, (?)‐colchiciline, (?)‐colchicine, β‐lumidemecolcine, 2‐demethyl‐β‐lumicolchicine or 3‐demethyl‐β‐lumicolchicine, N,N‐dimethyl‐N‐deacetyl‐β‐lumicornigerine, (?)‐isoandrocymbine and (?)‐autumnaline. Furthermore, a new compound was identi?ed as N,N‐dimethyl‐N‐deacetyl‐(?)‐cornigerine. Three compounds, which had molecular ions at m/z 325, 340 and 374, could not be dereplicated into any obvious structural classes that have been isolated in our laboratories previously or reported in the literature. Compounds that promoted the growth of pollen tubes were isolated from the style of Rhododendron mucronatum and were identified as azalein, (+)-catechin, and (?)epicatechin. Among these compounds, 50–100 ppm of (+)-catechin or (?)-epicatechin increased the growth of pollen tubes in Camelliajaponica, R. mucronatum, Styrax japonica, and Pinus densijlora by 16–20%. These compounds had no significant effects on the growth of pollen tubes in Lilium auratum and Narcissus pseudo-narcissus. Among the phenolic compounds with similar chemical structures as the promoters, catechol-and pyrogallol-type compounds had the activity and the latter types were more effective. As α-tocopherol also showed growth promoting activity, it was thought that the antioxidative activity of the phenolic compounds was responsible for the promotion of the pollen tube growth.  相似文献

The stereoselective metabolism of lactofen in primary rat hepatocytes was studied using a chiral high‐performance liquid chromatographic (HPLC) method. Rac‐lactofen and its two enantiomers, S‐(+)‐ and R‐(?)‐lactofen, as well as two of its major metabolites, acifluorfen, S‐(+)‐ and R‐(?)‐desethyl lactofen, were used as substrates,. The single and joint cytotoxicity of parent compounds and the metabolites were assessed by coincubation with rat hepatocytes as target cells. Cytotoxicity was determined by the methyl tetrazolium (MTT) assay. In hepatocyte incubations, S‐(+)‐lactofen was degraded more rapidly than R‐(?)‐lactofen, and a stereospecific formation of S‐(+)‐desethyl lactofen was detected. Metabolism of lactofen to desethyl lactofen was processed with the retention of configuration, and the achiral compound, acifluorfen, was the shared metabolite generated from both S‐(+)‐ and R‐(?)‐lactofen. There was no chiral conversion of lactofen or desethyl lactofen enantiomers during the incubation. For the cytotoxicity research, the calculated EC50 values indicated that when being applied individually, the parent compound was less toxic than its metabolites, while the combination with metabolites enhanced its cytotoxic effects. The data presented here would be helpful for a more comprehensive assessment of the ecotoxicological and environmental risks of lactofen. The hepatitis B core antigen (HBcAg) is an important target for antiviral response in chronic hepatitis B (CHB) patients. However, the correlation between HBcAg in the hepatocyte nucleus and nucleos(t)ide analogue (NA) therapeutic response is unclear. We sought to evaluate the role of HBcAg by analysing liver biopsies for viral response in NA‐naïve hepatitis B e antigen (HBeAg) positive (+) CHB patients via immunohistochemistry (IHC). A total of 48 HBcAg‐negative (?) patients and 48 HBcAg (+) patients with matching baseline characteristics were retrospectively analysed for up to 288 weeks. Virological response (VR) rates of patients in the HBcAg (?) group were significantly higher at week 48 and 96 than the HBcAg (+) group (77.1% versus 45.8% at week 48, respectively, = 0.002 and 95.3% versus 83.3% at week 96, respectively, = 0.045). The serological negative conversion rate of HBeAg was significantly higher in the HBcAg (?) than in the HBcAg (+) group from week 96 to 288 (35.4 % versus 14.6% at week 96, respectively, = 0.018; 60.4% versus 14.6%, respectively, < 0.001 at week 144; 72.9% versus 35.4%, respectively, < 0.001 at week 288). The cumulative frequencies of VR and lack of HBeAg were higher in the HBcAg (?) group (both P < 0.05). Binary logistic regression analysis showed that HBcAg (?) was the predictor for the lack of HBeAg (OR 4.482, 95% CI: 1.58–12.68). In summary, the absence of HBcAg in the hepatocyte nucleus could be an independent predictor for HBeAg seroconversion rates during NA‐naïve treatment in HBeAg (+) CHB patients.  相似文献

Enantioselectivity in ecotoxicity of chiral pesticides in the aquatic environment has been a subject of growing interest. In this study, the toxicological impacts of hexaconazole enantiomers were investigated with freshwater algae Scenedesmus obliquus. After 96 h of exposure, the EC50 values for rac‐hexaconazole, (+)‐hexaconazole, and (?)‐hexaconazole were 0.178, 0.355, and 0.065 mg l?1, respectively. Therefore, the acute toxicities of hexaconazole enantiomers were enantioselective. In addition, the different toxic effects were evaluated when S. obliquus were exposed to 0.2, 0.5, and 1.0 mg l?1 of rac‐hexaconazole, (+)‐hexaconazole, and (?)‐hexaconazole during 96 h, respectively. The chlorophyll a and chlorophyll b contents of S. obliquus treated by (?)‐hexaconazole were lower than those exposed to (+)‐hexaconazole, whereas the malondialdehyde contents of S. obliquus treated by (?)‐form were higher than those exposed to (+)‐form at higher concentrations. In general, catalase activities were significantly upregulated by exposure to (?)‐enantiomer than (+)‐enantiomer at all three concentrations. However, superoxide dismutase activities exposed to (?)‐hexaconazole were lower than that exposed to (+)‐hexaconazole at 0.2 mg l?1 and 0.5 mg l?1. On the basis of these data, the acute toxicity and toxic effects of hexaconazole against S. obliquus were enantioselective, and such enantiomeric differences must be taken into consideration in pesticide risk assessment. The acetylcholinesterase inhibition by enantiomers of exo‐ and endo‐2‐norbornyl‐Nn‐butylcarbamates shows high stereoselelectivity. For the acetylcholinesterase inhibitions by (R)‐(+)‐ and (S)‐(?)‐exo‐2‐norbornyl‐Nn‐butylcarbamates, the R‐enantiomer is more potent than the S‐enantiomer. But, for the acetylcholinesterase inhibitions by (R)‐(+)‐ and (S)‐(?)‐endo‐2‐norbornyl‐Nn‐butylcarbamates, the S‐enantiomer is more potent than the R‐enantiomer. Optically pure (R)‐(+)‐exo‐, (S)‐(?)‐exo‐, (R)‐(+)‐endo‐, and (S)‐(?)‐endo‐2‐norbornyl‐Nn‐butylcarbamates are synthesized from condensations of optically pure (R)‐(+)‐exo‐, (S)‐(?)‐exo‐, (R)‐(+)‐endo‐, and (S)‐(?)‐endo‐2‐norborneols with n‐butyl isocyanate, respectively. Benalaxyl (BX), methyl‐N‐phenylacetyl‐N‐2,6‐xylyl alaninate, is a potent acylanilide fungicide and consist of a pair of enantiomers. The stereoselective metabolism of BX was investigated in rat and rabbit microsomes in vitro. The degradation kinetics and the enantiomer fraction (EF) were determined using normal high‐performance liquid chromatography with diode array detection and a cellulose‐tris‐(3,5‐dimethylphenylcarbamate)‐based chiral stationary phase (CDMPC‐CSP). The t1/2 of (?)‐R‐BX and (+)‐S‐BX in rat liver microsomes were 22.35 and 10.66 min of rac‐BX and 5.42 and 4.03 of BX enantiomers. However, the t1/2 of (?)‐R‐BX and (+)‐S‐BX in rabbit liver microsomes were 11.75 and 15.26 min of rac‐BX and 5.66 and 9.63 of BX enantiomers. The consequence was consistent with the stereoselective toxicokinetics of BX in vitro. There was no chiral inversion from the (?)‐R‐BX to (+)‐S‐BX or inversion from (+)‐S‐BX to (?)‐R‐BX in both rabbit and rat microsomes. Tetrahydropalmatine (THP), with one chiral center, is an alkaloid that possesses analgesic and many other pharmacological actives. The aim of the present study is to investigate stereoselective metabolism of THP enantiomers in human liver microsomes (HLM) and elucidate which cytochrome P450 (CYP) isoforms contribute to the stereoselective metabolism in HLM. Additionally, the inhibitions of THP enantiomers on activity of CYP enzymes are also investigated. The results demonstrated that (+)‐THP was preferentially metabolized by HLM. Ketoconazole (inhibitor of CYP3A4/5) inhibited metabolism of (?)‐THP or (+)‐THP at same degree, whereas the inhibition of fluvoxamine (inhibitor of CYP1A2) on metabolism of (+)‐THP was greater than that of (?)‐THP; moreover, the metabolic rate of (+)‐THP was 5.3‐fold of (?)‐THP in recombinant human CYP1A2. Meanwhile, THP enantiomers did not show obvious inhibitory effect on the activity of various CYP isoforms (CYP1A2, 2A6, 2C8, 2C9, 2C19, 2E1, and 3A4/5), whereas (?)‐THP, but not (+)‐THP, significantly inhibited the activity of CYP2D6 with the Ki value of 6.42 ± 0.38 μM. The results suggested that THP enantiomers were predominantly metabolized by CYP3A4/5 and CYP1A2 in HLM, and (+)‐THP was preferentially metabolized by CYP1A2, whereas CYP3A4/5 contributed equally to metabolism of (?)‐THP or (+)‐THP.   • 1 Volatiles from the hindgut extracts of males of the Oriental spruce engraver Pseudips orientalis (Wood & Yin) (Coleoptera: Curculionidae, Scolytinae) of different phases of gallery development were analyzed by gas chromatography‐mass spectrometry‐flame ionization detection (GC‐MS/FID) with both polar and enantioselective columns.
  • 2 GC‐MS/FID analyses showed that unmated males or males mated with one female produced approximately 95%‐(?)‐ipsenol and (?)‐cis‐verbenol as major components, as well as (?)‐trans‐verbenol, myrtenol, approximately 70%‐(+)‐ipsdienol and (?)‐verbenone as minor or trace components. The release of these male‐produced compounds was confirmed by GC analysis of an aeration sample of a P. orientalis‐infested spruce log. Mating reduced production of the male‐specific hindgut volatiles.
  • 3 A field‐trapping bioassay in Qinghai, China, showed that a ternary blend containing two major components, 97%‐(?)‐ipsenol (i.e. close to naturally produced enantiomeric ratio) and (?)‐cis‐verbenol, plus a minor component (?)‐trans‐verbenol, caught significantly more P. orientalis beetles (♂: ♀ = 1: 2.7) compared with the unbaited control. Subtraction of (?)‐trans‐verbenol from the active ternary blend had no significant effect on trap catches. The addition of (±)‐ipsdienol (at 0.2 mg/day release) to the active ternary or binary blends significantly interrupted their trap catches. Replacing 97%‐(?)‐ipsenol with (±)‐ipsenol in the ternary blend significantly reduced trap catches to a level that was no different from the blank control.

  • 5 The results obtained suggest that the two major components, 95%‐(?)‐ipsenol and (?)‐cis‐verbenol (at approximately 4–5 : 1), produced by unmated fed males, are probably the primary aggregation pheromone components for P. orientalis. In light of the phylogeny, the use of terpenoid semiochemicals as pheromones probably occurred early in the evolution of Ipini and these semiochemical blends were subsequently modified in the process of speciation.

The chiral inversion and pharmacokinetics of two enantiomers of trantinterol, a new β2 agonist, were studied in rats dosed (+)‐ or (?)‐trantinterol separately. Plasma concentrations of (+)‐ and (?)‐trantinterol were measured by chiral stationary phase liquid chromatography tandem mass spectroscopy (LC‐MS/MS). The apparent inversion ratio was calculated as the ratio of AUC0‐t of (?)‐trantinterol or (+)‐trantinterol inverted from their antipodes to the sum of the AUC0‐t of (?)‐ and (+)‐trantinterol. Following single intravenous administration, both given enantiomers declined in similar plasma concentrations, suggesting that the two enantiomers have approximately the same disposition kinetics by the route of intravenous administration. However, after single oral administration, plasma concentrations of uninverted (?)‐trantinterol at many timepoints were significantly higher than those of uninverted (+)‐trantinterol, suggesting that the two enantiomers undergo apparently different absorption or metabolism after oral administration. Significant bidirectional chiral inversion occurred after intravenous and oral administration of (+)‐ or (?)‐trantinterol. After dosing with optically pure enantiomer, the concentration of the administered enantiomer predominated in vivo. The AUC0‐36 of (+)‐trantinterol after intravenous and oral dosing of (?)‐trantinterol were 16.6 ± 5.2 and 33.3 ± 16%, respectively of those of total [(+) + (?)] trantinterol. The AUC0‐36 of (?)‐trantinterol after intravenous and oral dosing of (+)‐trantinterol were 19.6 ± 8.8 and 37.9 ± 4.5%, respectively, of those of total [(?) + (+)] trantinterol. After intravenous administration of (+)‐ and (?)‐trantinterol the chiral inversion ratios of the two enantiomers were not significantly different and similar results were found for oral administration. The extent of chiral inversion after intravenous administration was apparently lower, indicating that the bidirectional chiral inversion was not only systemic but also presystemic.
