南海半红树植物黄槿内源真菌GT20036029代谢产物研究   总被引:1,自引:0,他引:1  
应用多种色谱技术从半红树药用植物黄槿内生真菌GT20036029中分离得到9个化合物,通过波谱学方法并与已知化合物数据作比较,鉴定它们分别为N-(2-羟基苯乙基)乙酰胺(1)、环(L-脯氨酸-D-异亮氨酸)(2)、环(L-亮氨酸-L-脯氨酸)(3)、环(D-亮氨酸-L-脯氨酸)(4)、环(亮氨酸-酪氨酸)(5)、环(苯丙氨酸-丝氨酸)(6)、脑苷脂B(7)、(25S)-纽替皂苷元-3-O-α-L-鼠李糖-(1→2)-β-D-葡萄糖苷(8)和(25S)-异纽替皂苷元-3-O-α-L-鼠李糖-(1→2)-β-D-葡萄糖苷(9)。化合物1为首次从海洋真菌代谢产物中分离得到。化合物8显示了较好的肿瘤细胞生长抑制活性。  相似文献

Two new benzopyran derivatives, (2R,4S)‐5‐methoxy‐2‐methyl‐3,4‐dihydro‐2H‐1‐benzopyran‐4‐ol and (2S,4R,2′S,4′R)‐4,4′‐oxybis(5‐methoxy‐2‐methyl‐3,4‐dihydro‐2H‐1‐benzopyran), and a new aliphatic compound, (3E,5Z,8S,10E)‐8‐hydroxytrideca‐3,5,10,12‐tetraen‐2‐one, together with three known benzopyran derivatives, were obtained from a mangrove endophytic fungus Penicillium citrinum QJF‐22 collected in Hainan island. Their structures were determined by analysis of spectroscopic data and the relative configuration of (2R,4S)‐5‐methoxy‐2‐methyl‐3,4‐dihydro‐2H‐1‐benzopyran‐4‐ol was also confirmed by single‐crystal X‐ray diffraction. The absolute configurations of four compounds were established by comparison of ECD spectra to calculations. The configuration of (3E,5Z,8S,10E)‐8‐hydroxytrideca‐3,5,10,12‐tetraen‐2‐one was confirmed by comparison of optical value to the similar compound. The configurations of the compounds (2S,4S)‐5‐methoxy‐2‐methyl‐3,4‐dihydro‐2H‐1‐benzopyran‐4‐ol and (2R,4R)‐5‐methoxy‐2‐methyl‐3,4‐dihydro‐2H‐1‐benzopyran‐4‐ol were first determined. (3R,4S)‐3,4,8‐Trihydroxy‐3,4‐dihydronaphthalen‐1(2H)‐one exhibited moderate inhibitory effects on LPS‐induced NO production in RAW264.7 cells with IC50 of 44.7 μM, and without cytotoxicity to RAW264.7 cells within 50 μM.  相似文献

Acetylcholinesterase (AChE) inhibitory activity-guided studies on the mangrove-derived endophytic fungus Penicillium citrinum YX-002 led to the isolation of nine secondary metabolites, including one new quinolinone derivative, quinolactone A ( 1 ), a pair of epimers quinolactacin C1 ( 2 ) and 3-epi-quinolactacin C1 ( 3 ), together with six known analogs ( 4 – 9 ). Their structures were elucidated based on extensive mass spectrometry (MS) and 1D/2D nuclear magnetic resonance (NMR) spectroscopic analyses, and compared with data in the literature. The absolute configurations of compounds 1 – 3 was determined by combination of electronic circular dichroism (ECD) calculations and X-Ray single crystal diffraction technique using CuKα radiation. In bioassays, compounds 1 , 4 and 7 showed moderate AChE inhibitory activities with IC50 values of 27.6, 19.4 and 11.2 μmol/L, respectively. The structure-activity relationships (SARs) analysis suggested that the existence of carbonyl group on C-3 and the oxygen atom on the five-membered ring were beneficial to the activity. Molecular docking results showed that compound 7 had a lower affinity interaction energy (−9.3 kcal/mol) with stronger interactions with different sites in AChE activities, which explained its higher activities.  相似文献

利用刚果红-CMC平板染色法、DNS法从槲寄生内生真菌中筛选获得1株高效纤维素酶产生菌H-3-3,通过分析其菌落形态特征和18S rDNA序列的同源性,将菌株H-3-3鉴定为拟茎点霉属Phom opsissp.;对其酶学性质研究得到:槲寄生内生真菌H-3-3中CMC酶、β-葡糖苷酶、滤纸酶这3种组分酶最适温度均为50℃,β-葡糖苷酶活和滤纸酶活最适pH值均为4.8,CMC酶最适pH值为4.4;3种酶在30~45℃,pH4.0~8.0稳定性较强。金属离子Mn^2+、Co^2+、Mg^2+和Ca^2+对3组分酶都有不同程度的促进作用,其中以Co^2+效果最佳;而Ba^2+、Zn^2+、Cu^2+对3组分酶分均表现出明显的抑制作用。  相似文献

研究植物内生真菌布雷青霉菌(Penicillium brefeldianum) F4a次级代谢产物的提取分离方法、结构鉴定及其降血糖和抗氧化活性。采用液体发酵培养,大孔吸附树脂HP20提取后,经硅胶柱色谱、Sephadex LH-20凝胶柱色谱、ODS反相开放柱色谱和高效液相色谱等手段进行分离,应用核磁共振等技术进行结构鉴定;采用紫外分光光度吸收法进行降血糖和抗氧化活性筛选。结果表明,分离得到6个化合物,分别鉴定为环(L-色氨酸-L-脯氨酸)(1)、3,3′-Methylenebis(4-hydroxy-6-methyl-2H-pyran-2-one)(2)、2-(2′S-Hydroxypropyl)-5-methyl-7-hydroxychromone(3)、染料木素(4)、大豆素(5)和苯酚(6)。化合物1和2具有一定的抗氧化活性,化合物4和5具有较强的抗氧化活性和降血糖活性。化合物2和3为首次从青霉属真菌中分离得到。化合物1和2的ABTS自由基清除活性为首次报道。  相似文献

Four previously unreported chromones, 5‐hydroxy‐2‐(hydroxymethyl)‐8‐methoxy‐4H‐chromen‐4‐one ( 1 ), (5R,7S)‐5,7‐dihydroxy‐2‐propyl‐5,6,7,8‐tetrahydro‐4H‐chromen‐4‐one ( 2 ), (5R,7S)‐5,7‐dihydroxy‐2‐methyl‐5,6,7,8‐tetrahydro‐4H‐chromen‐4‐one ( 3 ), and (5R,7S)‐5,7‐dihydroxy‐2‐[(E)‐prop‐1‐en‐1‐yl]‐5,6,7,8‐tetrahydro‐4H‐chromen‐4‐one ( 4 ), as well as one known analogue 5‐hydroxy‐2‐methyl‐4H‐chromen‐4‐one ( 5 ) were isolated from the fermentation broth of the endophytic fungus Colletotrichum gloeosporioides derived from the mangrove Ceriops tagal. Their structures were elucidated based on extensive spectroscopic analyses. The absolute configurations of 2 – 4 were determined by comparison the experimental and calculated electronic circular dichroism (ECD) spectra. Compound 2 showed cytotoxic activity against A549 cell line with the IC50 value of 0.094 mm .  相似文献

One natural p‐terphenyl glycoside, gliocladinin C, and two furano‐polyene derivatives, chaetominins A and B, were isolated from potato endophytic fungus Chaetomium subaffine. The absolute configurations of these compounds were elucidated by HR‐ESI‐MS, NMR, the DP4+ probabilities and electronic circular dichroism (ECD) spectra. Furthermore, gliocladinin C and chaetominin A showed cytotoxic activity against two selected human tumor cell lines (Hep‐2 and HepG‐2).  相似文献

从银杏叶柄分离筛选到具抗菌活性的内生真菌Colletotrichum.SP NTB-2菌株,利用硅胶柱色谱、制备高效液相色谱等方法在其发酵产物中分离到抗枯草芽孢杆菌、鼠伤寒沙门氏菌等具有广谱抑菌活性的化合物,经MS、NMR等波谱数据确认该活性成分为芹菜素-8-C-葡萄糖苷(apigenin-8-C-β-D-glucopyranoside),该化合物首次从真菌中分离得到。  相似文献

The chemical investigation on endophytic fungus Annulohypoxylon cf. stygium in leaves of Anoectochilus roxburghii (Wall.) Lindl . has been performed. Sixteen compounds were isolated and their structures were identified as (?)‐notoamide A, (?)‐notoamide B, (+)‐versicolamide B, notoamide C, notoamide D, stephacidin A, sterigmatocystin, dihydrosterigmatocystin, secosterigmatocystin, versiconol, averufanin, kipukasin D, kipukasin E, diorcinal, palmarumycin CP2 and (?)‐(3R)‐mellein methyl ether, respectively, by spectroscopic analysis and comparison with literature data. All the compounds were isolated from Annulohypoxylon genus for the first time. Sterigmatocystin and palmarumycin CP2 showed selective cytotoxic activities against HepG2, HeLa, MCF‐7 and HT‐29.  相似文献

由杜仲植株的根、茎、叶分离到内生真菌,分别对各菌株进行液体培养,筛选能产生与杜仲药材相同或相似黄酮类化合物的内生真菌。将培养物适当处理后,根据黄酮类化合物所特有的颜色反应进行初筛,然后根据TLC分析和紫外-可见光分光光度法测定结果进行复筛,结果从杜仲植株共分离得到44株内生真菌,其中1株(DZY5)能产出与其宿主相同或相似的黄酮类物质。  相似文献

One new naphthalenone, didymelol A, and three new naphthols, didymelol B–D, together with three known naphthalenones, (3S,4R)‐3,4,6‐trihydroxy‐3,4‐dihydronaphthalen‐1(2H)‐one, (4S)‐4,6‐dihydroxy‐3,4‐dihydronaphthalen‐1(2H)‐one, (4S)‐4‐hydroxy‐3,4‐dihydronaphthalen‐1(2H)‐one, were isolated from the Saussurea laniceps endophytic fungus Didymella glomerata X223. The structures of the isolates were elucidated based on extensive spectroscopic data analysis. The absolute configuration of didymelol A was established through single‐crystal X‐ray diffraction data and didymelols B–D were identified through comparisons of experimental and calculated ECD spectra. All compounds were evaluated for cytotoxic activity against human non‐small cell lung cancer A549 cells and human breast carcinoma MDA‐MB‐435 cells.  相似文献

Two new nonenolides named diaportheolides A ( 1 ) and B ( 2 ) were isolated from the endophytic fungus Diaporthe sp. SXZ-19 of Camptotheca acuminata. The chemical structures of 1 and 2 were elucidated by spectroscopic analyses, including 1D- and 2D-NMR experiments and HR-ESI-MS data analysis. 对大花金挖耳内生真菌H-18菌株的菌丝提取物进行了抑菌活性研究并对菌株进行了鉴定.离体试验结果表明,H-18菌株菌丝体丙酮提取物在500mg/L浓度下对玉米大斑病菌、番茄灰霉病菌和辣椒疫霉病菌的菌丝生长抑制率均大于80%;组织法和盆栽试验结果表明,H-18菌株菌丝体丙酮提取物在2000mg/L浓度下对番茄灰霉病和辣椒疫霉病的保护效果和治疗效果均在50%以上.结合形态分类学和ITS-5.8S rDNA序列分析结果鉴定菌株H-18为同色镰孢菌(Fusarium concolor).  相似文献

从野生的霍山石斛(Dendrobium huoshanense)根中分离出1株内生真菌SH-01,纯化后以生物量为主要指标对培养条件进行优化,并将内生真菌SH-01与霍山石斛试管苗共生培养。结果表明:与对照相比,该菌株对石斛试管苗的鲜重、株高及分蘖都有一定的促进作用,对株高的增长有显著促进作用(P<0.05),但对鲜重和分蘖的促进作用不显著(P>0.05)。在离体条件下,选择适当的内生真菌与石斛共生培养可以促进石斛试管苗的生长。  相似文献

通过大量筛选从银杏茎中分离到一株内生真菌(编号为No.1028),其代谢产物对重要作物真菌病害病原菌具有明显的抑制作用。结果表明:离体条件下内生真菌发酵液对番茄早疫病菌、黄瓜枯萎病菌、菜豆炭疽病菌、葡萄炭疽病菌、水稻纹枯病菌、黄瓜立枯病菌均有较强的抑制作用,对上述不同病原菌的抑制率分别为:66.7%、48.3%、64.6%、36.5%、57.1%和23%。同时也研究了不同碳源、氮源、无机离子对内生真菌No.1028生物学特性和生长的影响。结果发现玉米粉、黄豆饼粉比较适合其生长和抑菌代谢产物的合成,Na+对抑菌代谢产物的合成有较明显的促进作用。  相似文献

为了探讨影响红树林淡紫拟青霉胞外多糖提取的因素,确定最佳提取方案,设置不同的发酵液浓缩倍数、三氯乙酸用量等因素,设计单因素实验测定多糖提取最佳条件。然后设计正交试验,检测多糖在不同条件下的提取率,以获得最佳提取工艺。结果发现,发酵液浓缩3~5倍时多糖提取率最高,10%的三氯乙酸对蛋白质脱除效果最好;正交试验表明影响多糖提取的因素依次为乙醇用量、沉淀时间和温度,最优方案为3倍95%乙醇、4 ℃沉淀24 h。该条件下,多糖提取率可达57.835%±1.206%。研究结果为红树林淡紫拟青霉胞外多糖的提取研究提供了参考。  相似文献
