The study addressed to understand whether or not lipoproteins at low concentrations could modulate Receptor-C dependent platelet signalling revealed that LDL, like exogneous cholesterol, had the capacity to initiate PLD-dependent platelet signalling in a dose dependent fashion and this effect was inhibited in presence of HDL; cAMP; DTT; Zn++ and butanol whereas cGMP had no effect upon this PLD-dependent signalling. Further Receptor C from platelet in the purified-form displayed LDL-or cholesterol-dependent autophosphorylation at the tyrosine residues and this Receptor-C tyrosine kinase (Receptor-Ck) activity contributed to the observed LDL-or cholesterol-dependent PLD activity in human platelets. Based upon these results coupled with earlier results, an attempt was made to define the lipoprotein-dependent platelet signalling pathway.  相似文献

The restriction landmark genomic scanning (RLGS) method was applied to rice, using two Japanese cultivars, Nipponbare and Koshihikari, and a Chinese landrace, LiuZhou'Bao'Ya'Zao'. More than 3000 landmarks were detected as spots on the individual autoradiograms of each cultivar. Nipponbare and LiuZhou' Bao'Ya'Zao' showed apparently different RLGS profiles, from which the genetic similarity (GS) between them was estimated as 0.344. Although the two Japanese cultivars, Nipponbare and Koshihikari showed quite similar RLGS profiles, they were easily distinguished on the basis of the presence or absence of specific spots; the GS value between them was calculated as 0.980. The RLGS method is shown to be a powerful fingerprinting technique, especially for the classification and identification of cultivars in rice and probably in other crops as well.  相似文献

During the 19th and early 20th centuries, ideas related to the possible origin in space of bioorganic molecules, or seeds, or even germs and organisms (and how they reached the Earth) included the Panspermia theory. Based on the idea of the eternity of life proposed by eminent physicists – such as Arrhenius and Kelvin – Panspermia is mainly divided into two branches: lithopanspermia (transport of germs inside stones traveling in space) and radiopanspermia (transport of spores by radiative pressure of stellar light). We point out some arguments to help to understand whether Panspermia could exist nowadays as the same theory defined one century ago. And we wonder about the kind of evolution Panspermia could have undergone during only a few decades. This possible evolution of the Panspermia concept takes place in the framework of the emergence of a new field, Bioastronomy. We present how this discipline has emerged during a few decades and how it has evolved. We consider its relationship with the progression of other scientific fields, and finally we examine how it is now included in different projects of space agencies. Bioastronomy researches having become more and more robust during the last few years, we emphasize several questions about new ideas and their consequences for the current hypothesis of Panspermia and of universal life.  相似文献

Summary The total nitrogen uptake of wheat in a pot experiment was found to be linearly related to the amounts of applied nitrogen in the range of 0 to 120 pounds N per acre. The N-value of the soil used in the pot experiment was estimated by extrapolation of the regression line of total nitrogen uptakevs rates of added nitrogen. The N-value was compared with the values of available soil nitrogen as estimated by three chemical methods, two of which used alkaline KMnO4 and the third, dilute H2SO4 as hydrolysing agents of soil organic nitrogen. Results obtained by none of these methods agreed closely with the N-value. The amounts of nitrogen estimated by Shihata's method and Purvis and Leo's method were practically same and were appreciably lower than N-value whereas the value obtained by Subbiah and Asija's method was much higher than the N-value. However, Purvis and Leo's method deserves further study as it is well suited for mass-scale analysis.  相似文献

As part of a program to explore the structural requirement of N-glycans in the carbohydrate-mediated biological interactions, N-linked pentasaccharide core structure was stereochemically modified in terms of glycosidic linkage. Three isomers, -D-Man-(13)-[-D-Man-(16)]--D-Man-(14)--D-GlcNAc-(14)--D-GlcNAc-L-Asn, -D-Man-(13)-[-D-Man-(16)]--D-Man-(14)--D-GlcNAc-(14)--D-GlcNAc-L-Asn, and -D-Man-(13)-[-D-man-(16)]--D-Man-(14)--D-GlcNAc-(14)--D-GlcNAc-L-Asn, were synthesized. Summary 2-Amino-2-deoxyuridine reacts efficiently with nucleoside 5-phosphorimidazolides in aqueous solution. The dinucleoside monophosphate analogues were obtained in yields exceeding 80% under conditions in which little reaction occurs with the natural nucleosides.In a similar way, the 5-phosphorimidazolide of 2-amino-2-deoxyuridine undergoes self-condensation in aqueous solution to give a complex mixture of oligomers.The phosphoramidate bond in the dinucleoside monophosphate analogues is stable for several days at room temperature and pH 7. Summary The zymogram phenotypes that Chinese Spring-Imperial, Holdfast-King II and Kharkov-Dakold wheat-rye addition lines presented for esterase isozymes were determined using polyacrylamide gel ectrophoresis. The analyses were carried out with different parts of the dry kernel, namely embryo plus scutellum and endosperm, leaves and roots. In all cases, embryo plus scutellum, endosperm and leaf presented different patterns of esterases. The patterns of leaves and roots were the same. Results indicate that rye esterases exist as monomers and dimers. Dimeric esterases are controlled by one locus located on the 3R chromosomes of Imperial, King II and Dakold rye cultivars. Five loci involved in the production of monomeric esterases have been located on the 6R chromosomes of these cultivars, specifically on the long arm of the King II 6R chromosome. On the basis of these results, considerations concerning chromosome homoeology and homology are made.  相似文献

Summary The purified bacteriophage replication proteins O and P sediment separately in metrizamide gradients of low ionic strength as dimers. Together they interact with each other forming an oligomer, composed of two molecules of O and one molecule of P. The O-P oligomer is active in the in vitro replication of ori-containing DNA.Equilibrium sedimentation in preformed metrizamide density gradients under conditions that separate DNA-protein complexes from free proteins was employed in order to study possible interactions among the replication proteins and ori DNA. It was found that the P protein binds specifically to ori-containing plasmid DNA only in the presence of O protein. About 100 molecules of O and 10 molecules of P form a complex with the ori DNA. The DNA-O-P complex was shown to be active in an in vitro replication system.Since the physical interactions between ori and O and between P and the Escherichia coli dnaB replication protein are well documented, the evidence for a O-P interaction presented in this paper provides the missing link in the molecular mechanism that enables to direct the host replication machinery to the replication of its own DNA.  相似文献

Summary The establishment and development of vesicular-arbuscular mycorrhizal (VAM) fungi were studied in three cultivars of potato, which differed in susceptibility to Late blight, in a field experiment on a lateritic sandy-loam during two growing seasons (1980 and 1981). The cultivars SSC 1174 (highly resistant) and Kufri Jyoti (resistant) showed an earlier establishment and more rapid development of VAM fungi than up-to-date (highly susceptible). The first mycorrhizal infection in both SSC 1174 and Kufri Jyoti was observed after 12 days in 1980 and 8 days in 1981, whereas in up-to-date it was observed after 19 and 12 days respectively. The mycorrhizal infection increased with the age of the plants in all the three cultivars.  相似文献

Thidiazuron substitution for chilling requirement in three apple cultivars   总被引:2,自引:0,他引:2  
Thidiazuron [(TDZ)N-phenyl-N-1,2,3-thidiazol-5-ylurea] at 750 M was applied to buds of apple trees to determine if it could substitute for the chilling requirement to induce bud break. Shoots of cv. Anna (low chill), Delicious cv. Redchief (medium chill), and Northern Spy (high chill) were untreated, treated with TDZ prior to chilling (before-chill), and treated with TDZ at various intervals after the accumulation of specific amounts of chilling (after-chill). Shoots were stored in a cold room at 4°C. TDZ applied prior to chilling reduced the chill unit (CU) requirement (1 CU = 1 h at 4°C) for the promotion of bud break on 1-year-old shoots of Anna and Northern Spy and 1- and 2-year-old wood of Delicious. TDZ applied after-chill promoted bud break only for Anna and buds on 2-year-old wood of Delicious. While accumulating CUs, untreated buds or buds treated with TDZ on 1-year Delicious and Northern Spy did not respond to the cold treatment even after 1848 h of CU accumulation. For all three cultivars, TDZ treatment was more effective in promoting bud break when applied before the initiation of chilling.The use of a company or product name does not constitute an endorsement by USDA or the University of Maryland nor imply approval to the exclusion of other suitable products.  相似文献

Summary A mutant strain of Rhodococcus equi accumulates three metabolites from the androst-4-ene-3,17-dione or from its degradation intermediate, 3a-H-4(3'-propionic acid)-7a-methylhexahydro-1,5-indanedione (MEPHIP). These three metabolites are: 3a-H-4a(3'-propionic acid)-5-hydroxy-7a-methylhexahydro-1-indanone--lactone (HIL); 3a-H-4(3'-trans acrylic acid)-5-hydroxy-7a-methylhexahydro-1-indanone (2'-5-hydroxy-MEPHIP); and 3a-H-4(3'-hydroxy-3'-propionic acid)-5-hydroxy-7a-methylhexahydro-1-indanone (3'-hydroxy-HIL). The behaviour of this mutant allows us to propose a pathway for degradation of the intermediates, methylperhydroindanone propionates. Summary We have examined the globin gene complex for 49 individuals with -thalassemia-2 (–3.7). Crossovers resulting in -thalassemia-2 (type I) were observed in all 57 chromosomes with the –3.7 defect. Except for one -thalassemia-2 chromosome, all were linked to the absence of an Rsa I restriction site located 0.7 kb 5 to the 2-globin gene; this polymorphic site was observed for 10 of 38 non--thalassemia chromosomes from Black Americans. In four Black families with a heterozygous -thalassemia-2 [–3.7 (I)], an Apa I restriction site has been identified in the IVS-2 of the 2 gene of the normal chromosome (labeled the *2 gene). The *2 gene of one Black subject was cloned and a segment located 5 to the Cap site as well as the IVS-2, exon 3, and a 3 segment were sequenced. The data show that the *2 gene is an 2 gene except for a segment between nucleotides (nts) 580–81 and nt 509 (Cap site=nt 1), and perhaps as far upstream as nt-634, which has an 1 sequence. This *2 hybrid gene probably originated through a double crossover; the structural identity of its IVS-2 with that of the 1 gene adequately explains the presence of the Apa I restriction site.  相似文献

Photosynthetic potential of isolated chloroplasts was investigated during in situ water deficits. An eight day stress cycle imposed on spinach plants reduced leaf w by 0.57MPa, and leaf by 0.50MPa, resulting in partial turgor maintenance during the stress cycle. Pressure/volume curves confirmed the occurrence of osmotic adjustment. Leaf depression was associated with an altered response of chloroplasts to low in vitro. Optimum reaction medium for photosynthesis shifted from –1.04 to –1.57MPa, and low was not as inhibitory to photosynthesis of plastids pre-exposed to stress in situ. These data indicate that chloroplasts acclimate to low external in response to leaf water deficits. This response was still evident four days after a stress cycle ended, but was nearly reversed eight days after stress. Repeated stress cycles in situ did not increase the degree of chloroplast acclimation to low in vitro. Summary We have synthesized 2(3)-O-(glycyl)-adenosine-5-(O-methylphos-phate), an analogue of the 3-terminus of aminoacylated tRNA. A 0.4M solution of this compound maintained at pH 8.2, yields 5.5% of diglycine and 11.5% of diketopiperazine, in addition to the hydrolysis products glycine and adenosine-5-(O-methylphosphate). Under the same conditions, glycine ethyl ester reacts much more slowly, but ultimately gives similar yields of diglycine and diketopiperazine.The aminolysis of 2(3)-O-(glycyl)-adenosine-5-(O-methylphosphate) by free glycine is relatively inefficient, but serine reacts 20 times more rapidly and yields up to 50% of N-glycylserine. Fowl plague virus, strain Dutch, was metabolically labeled withd-[2-3H]mannose, or withd-[6-3H]glucosamine, and the small subunit (HA2; 0.8 mg in total) of the viral hemagglutinin was isolated by preparative sodium dodecylsulfate-polyacrylamide gel electrophoresis. After proteolytic digestion, the radioactive oligosaccharides were sequentially liberated from the glycopeptides by treatment with different endo--N-acetylglucosaminidases and with peptide:N-glycosidase or, finally, by hydrazinolysis. In this manner, four groups of glycans could be obtained by consecutive gel filtrations and were subfractionated by HPLC. The structures of the individual oligosaccharides were analyzed by micromethylation, by acetolysis or by digestion with exoglycosidases. Based on different marker information content mapping of QTLs for Fusarium head blight resistance in wheat was compared with regard to number and consistency of detected QTLs as well as QTL positions and effects. Therefore, two linkage maps, obtained by dominant and codominant genotyping of hemizygous markers, were constructed with 211 AFLPs, 37 SSRs and the barley RGA marker XaACT/CAA. The codominant marker set comprised 59% codominant markers, whereas the dominant map consisted of only 13%. A segregating wheat population of 94 F4-RILs was used for QTL analysis. Fusarium head blight resistance was estimated in field trials in six environments. Conventional dominant marker scoring found seven QTLs. The phenotypic variations explained by QTLs detected in single environment analyses ranged from 11.1 to 44.6%. QTL analysis performed with the codominant marker set confirmed not only all QTL positions as revealed by dominant QTL analysis', but also 12 additional QTLs were found. QTLs in single environments explained 36.3 up to 55.7% of the phenotypic variation. In the QTL analysis across all environments, none of the QTLs could be confirmed using dominant marker scoring. However, by codominant QTL analysis' environment-specific QTLs were retrieved. STS marker XaACT/CAA was found to be significantly associated with FHB resistance only by codominant scoring. Support intervals of QTLs commonly found in both marker sets averaged to 10.3 cM in the dominant QTL analysis', whereas the length was shortened to 8.9 cM by codominant genotyping. The advantages of extracting codominant information from dominant markers are discussed.  相似文献

The sialidase of swine influenza A viruses of N1 and N2 subtypes, isolated from 1930 to 1992, was studied for substrate specificity with ganglio-series, lacto-series type II and GM3 gangliosides containing Neu5Ac2-3Gal, Neu5Gc2-3Gal and Neu5Ac2-6Gal linkages. All viral sialidases tested showed that the activity for hydrolysing substrates with Neu5Ac2-3Gal was higher than the activities with Neu5Gc2-3Gal and Neu5Ac2-6Gal linkages. When GM1b, GM3 and sialylparagloboside were used as substrates, the earliest strain (A/Wisconsin/15/30 H1N1, isolated in 1930) showed the activity ratio of Neu5Ac2-6Gal to Neu5Ac2-3Gal to be 0.13:0.2, and the ratio Neu5Gc2-3Gal/Neu5Ac2-3Gal to be 0.19:0.37, while those strains isolated from 1978 to 1992 exhibited ratios of 0.29:0.58 for Neu5Ac2-6Gal/Neu5Ac2-3Gal and 0.51:0.76 for Neu5Gc2-3Gal/Neu5Ac2-3Gal. The concentration of airborne spores of Cladosporium and Alternaria has been investigated at five monitoring stations situated in cities from the foot of the Tatra Mountains to central Poland along a south-north transect (Zakopane, Kraków, Ostrowiec witokrzyski, Warszawa, Pozna) i.e. from a height of 900 m to 80 m above sea level. The aerobiological monitoring of fungal spores was performed by means of five Burkard volumetric spore traps.Cladosporium spores were dominant at all the stations. The highest Cladosporium and Alternaria spore concentrations were observed at all the sites in July and August, except at Warszawa in both years and at Pozna in 1995 where the maximum of Cladosporium spores occurred in June and July, and at Ostrowiec witokrzyski in 1995 where the maximum was found in July, August and September.Fungal spore concentrations in Zakopane and Kraków were significantly lower than those in Ostrowiec witokrzyski, Warszawa and Pozna and periods of abundant Cladosporium spore occurrence were different in these two groups of monitoring stations.  相似文献
