There is a surprising lack of genetic data for the Cuban crocodile (Crocodylus rhombifer), especially given its status as a critically endangered species. Samples from captive individuals were used to genetically characterize this species in comparison with other New World crocodilians. Partial mitochondrial sequence data were generated from cyt-b (843 bp) and the tRNA(Pro)- tRNA(Phe)-D-loop region (442 bp). Phylogenetic analyses were performed by generating maximum parsimony, maximum likelihood, and Bayesian-based topologies. In addition, in an effort to identify species-specific alleles, ten polymorphic microsatellite loci were genotyped. Distance and model-based clustering analyses were performed on microsatellite data, in addition to a model-based assignment of hybrid types. Both mitochondrial and nuclear markers identified two distinct C. rhombifer genetic sub-clades (alpha and beta); and microsatellite analyses revealed that most admixed individuals were F(2) hybrids between C. rhombifer-alpha and the American crocodile (C. acutus). All individuals in the C. rhombifer-beta group were morphologically identified as C. acutus and formed a distinct genetic assemblage. J. Exp. Zool. 309A:649-660, 2008. Globally temperature of marine environments is on the rise and temperature plays an important role in the life-history of reptiles. In this study, we examined the relationship between sea surface temperature and average date of hatching for American crocodiles (Crocodylus acutus) over a 37-year period at two nesting sites, Everglades National Park and Florida Power and Light Turkey Point Power Plant site in southern Florida. Our results indicate that hatch dates are shifting 1.5 days earlier every two years and at half that rate for the Turkey Point site, and with every 1 °C degree increase in temperature, hatching occurs about 10 days earlier in the Everglades and 6 days earlier at Turkey Point. Our results on shifting hatch dates for American crocodiles provide further details about the impacts of temperature change on crocodile life history and suggest that increased temperature may affect their phenology.  相似文献

Hybrid zones represent natural laboratories to study gene flow, divergence and the nature of species boundaries between closely related taxa. We evaluated the level and extent of hybridization between Crocodylus moreletii and Crocodylus acutus using genetic and morphological data on 300 crocodiles from 65 localities. To our knowledge, this is the first genetic study that includes the entire historic range and sympatric zone of the two species. Contrary to expectations, Bayesian admixture proportions and maximum‐likelihood estimates of hybrid indexes revealed that most sampled crocodiles were admixed and that the hybrid zone is geographically extensive, extending well beyond their historical region of sympatry. We identified a few geographically isolated, nonadmixed populations of both parental species. Hybrids do not appear to be F1s or recent backcrosses, but rather are more likely later‐generation hybrids, suggesting that hybridization has been going on for several to many generations and is mostly the result of natural processes. Crocodylus moreletii is not the sister species of C. acutus, suggesting that the hybrid zone formed from secondary contact rather than primary divergence. Nonadmixed individuals from the two species were distinguishable based on morphological characters, whereas hybrids had a complex mosaic of morphological characters that hinders identification in the wild. Very few nonadmixed C. acutus and C. moreletii populations exist in the wild. Consequently, the last nonadmixed C. moreletii populations have become critically endangered. Indeed, not only the parental species but also the naturally occurring hybrids should be considered for their potential conservation value.  相似文献

Identification of units within species worthy of separate management consideration is an important area within conservation. Mitochondrial DNA (mtDNA) surveys can potentially contribute to this by identifying phylogenetic and population structure below the species level. The American crocodile (Crocodylus acutus) is broadly distributed throughout the Neotropics. Its numbers have been reduced severely with the species threatened throughout much of its distribution. In Colombia, the release of individuals from commercial captive populations has emerged as a possible conservation strategy that could contribute to species recovery. However, no studies have addressed levels of genetic differentiation or diversity within C. acutus in Colombia, thus complicating conservation and management decisions. Here, sequence variation was studied in mtDNA cytochrome b and cytochrome oxidase I gene sequences in three Colombian captive populations of C. acutus. Two distinct lineages were identified: C. acutus‐I, corresponding to haplotypes from Colombia and closely related Central American haplotypes; and C. acutus‐II, corresponding to all remaining haplotypes from Colombia. Comparison with findings from other studies indicates the presence of a single “northern” lineage (corresponding to C. acutus‐I) distributed from North America (southern Florida), through Central America and into northern South America. The absence of C. acutus‐II haplotypes from North and Central America indicates that the C. acutus‐II lineage probably represents a separate South American lineage. There appears to be sufficient divergence between lineages to suggest that they could represent two distinct evolutionary units. We suggest that this differentiation needs to be recognized for conservation purposes because it clearly contributes to the overall genetic diversity of the species. All Colombian captive populations included in this study contained a mixture of representatives of both lineages. As such, we recommend against the use of captive‐bred individuals for conservation strategies until further genetic information is available.  相似文献

The reproductive cycle of the Nile crocodile ( Crocodylus niloticus ) was studied in Zimbabwe. Females attained sexual maturity after they had reached a total length of about 262 to 287 cm and males, 270 to 295 cm. Some adult females did not reproduce every year. Follicle growth and vitellogenesis occurred from April to mid August during the dry winter. During this time reproductive females had elevated levels of plasma oestradiol-17β, testosterone, calcium and magnesium, but lowered levels of iron. The measuring of plasma calcium was an 'early pregnancy' test, reliable up to four months before nesting. Elevated levels of plasma testosterone in reproductive females corresponded to the time of courtship and mating. Ovulation occurred during the latter half of August. Males had viable sperm from mid May to mid September, during the winter. Courtship and mating occurred from late June to mid August, when crocodiles were confined to pools. Females nested on the higher sand ridges in the dry river bed from early September to early October, and eggs hatched during December.  相似文献

Genomic elucidation and mapping of novel organisms requires the generation of large genetic resources. In this study, 253 novel and polymorphic microsatellite loci were isolated and characterized for the saltwater crocodile (Crocodylus porosus) by constructing libraries enriched for microsatellite DNA. All markers were evaluated on animals obtained from Darwin Crocodile Farm in the Northern Territory, Australia, and are intended for future use in the construction of a genetic-linkage map for the saltwater crocodile. The 253 loci yielded an average of 4.12 alleles per locus, and those selected for mapping had an average polymorphic information content (PIC) of 0.425.  相似文献

The ultrastructure of Stylodinium littorale Horiguchi et Chihara, a marine, sand-dwelling coccoid dinoflagel-late, was investigated with special emphasis on its stalk and the apical stalk complex. The dinoflagellate alternates between non-motile and motile cells in its life cycle. The non-motile cell possesses a long and distinct stalk. The stalk, consisting of a main cylindrical part and a holdfast, is firmly attached to a thecal plate (the apical pore plate). A part of its proximal portion is hollow and V-shaped in section. The V-shaped hollow space is underlain by a projection from the apical pore plate. An apical stalk complex is present in the motile cells and consists of a large apical pore plate and mucilaginous material. The apical pore plate is depressed into the cell, but has a narrow central tubular projection. The mucilaginous stalk-building material is stored between this plate and the outer plate membrane. The tubular projection of the apical pore plate corresponds to the apical pore of other dinoflagellates and its lumen is filled with electron-dense material. The structure of the apical stalk complex is compared with the homologous structure in Bysmatrum arenicola, the only other example of an apical stalk complex that has been investigated. A general ultrastructural survey revealed that S. littorale possesses a typical dinoflagellate cellular structure.  相似文献

The ultrastructure of spermatozoa from the cauda epididymidis and vas deferens of Octodon degus-a Chilean hystricomorph rodent-is presented. The head of spermatozoa measured 7.7 micrometer long by 5.9 micrometer wide and the tail was 41 micrometer long. The head was flattened dorso-ventrally and ovate in outline. The acrosome was the most distinctive feature of O. degus spermatozoa. In a frontal view of the head, the rim of the acrosome surrounding the nucleus had the shape of an inverted U. The acrosomal region covering the plane of the flattened head exhibited dome-shaped protrusions. Transverse or sagittal sections of acrosomal protrusions showed that the plasma membrane and outer acrosomal membrane were evaginated, while the inner acrosomal membrane followed the contour of the nucleus. The protrusions were not distributed at random and they were absent in the equatorial segment and in the rim of the acrosome. In frontal views, near the boundary between the acrosome and post-acrosomal region, fine rods about 170 nm long ran obliquely on the caudal part of the equatorial segment. Behind the same boundary, the post-acrosomal region showed a serrated border. Phosphotungstic acid treatment at pH 0.3 produced staining at the surface of the sperm as well as within a superficial layer of the marginal thickening of the acrosome and on the acrosomal protuberances.  相似文献

ABSTRACT: BACKGROUND: The Siamese crocodile (Crocodylus siamensis) is a critically endangered species of freshwater crocodiles. Crocodilians live with opportunistic bacterial infection but normally suffer no adverse effects. They are not totally immune to microbial infection, but their resistance thereto is remarkably effective. In this study, crude and purified plasma extracted from the Siamese crocodile were examined for antibacterial activity against clinically isolated, human pathogenic bacterial strains and the related reference strains. METHODS: Crude plasma was prepared from whole blood of the Siamese crocodile by differential sedimentation. The crude plasma was examined for antibacterial activity by the liquid growth inhibition assay. The scanning electron microscopy was performed to confirm the effect of crude crocodile plasma on the cells of Salmonella typhi ATCC 11778. Effect of crude crocodile plasma on cell viability was tested by MTT assay. In addition, the plasma was purified by anion exchange column chromatography with DEAE-Toyopearl 650M and the purified plasma was tested for antibacterial activity. RESULTS: Crude plasma was prepared from whole blood of the Siamese crocodile and exhibited substantial antibacterial activities of more than 40% growth inhibition against the six reference strains of Staphylococcus aureus, Salmonella typhi, Escherichia coli, Vibrio cholerae, Pseudomonas aeruginosa, and Staphylococcus epidermidis, and the four clinical isolates of Staphylococcus epidermidis, Pseudomonas aeruginosa, Salmonella typhi, and Vibrio cholerae. Especially, more than 80% growth inhibition was found in the reference strains of Salmonella typhi, Vibrio cholerae, and Staphylococcus epidermidis and in the clinical isolates of Salmonella typhi and Vibrio cholerae. The effect of the crude plasma on bacterial cells of Salmonella typhi, a certain antibacterial material probably penetrates progressively into the cytoplasmic space, perturbing and damaging bacterial membranes. The effect of the crude plasma was not toxic by the yellow tetrazolium bromide (MTT) assay using a macrophage-like cell, RAW 264.7. The pooled four fractions, designated as fractions D1-D4, were obtained by column chromatography, and only fraction D1 showed growth inhibition in the reference strains and the clinical, human pathogenic isolates. CONCLUSIONS: The crude and purified plasma from the Siamese crocodile significantly showed antibacterial activity against pathogenic bacteria and reference strains by damage cell membrane of target bacterial cells. From the MTT assay, the Siamese crocodile plasma was not cytotoxic to the cells.  相似文献

As part of an ongoing study of the intestinal tract of the Nile crocodile Crocodylus niloticus, the appearance of the small intestinal mucosa was examined using a scanning electron microscope (SEM). The duodenum displayed ridge-like and complex primary folds, both of which were arranged in a longitudinally directed zigzag pattern. The two types of folds alternated with each other, with the second type following the zigzag contours of the first. The folds were covered with polygonal- and dome-shaped epithelial cells. The latter cell type was less common and was restricted in location to the proximal two thirds of the mucosal folds. Both types of epithelial cells were covered with stubby microvilli which displayed a distinct linear arrangement. Goblet cells were present between the absorptive cells. In the jejunum the basic pattern of the folds persisted except that both types of primary folds became tall and leaf-like. The angle of the zigzag pattern was less acute than in the duodenum. In the ileum the two types of alternating folds decreased in height and were arranged in a longitudinally directed wave-like fashion. The zigzag mucosal folds of the small intestine ended abruptly at the ileorectal junction. The rectum displayed low, irregular folds which formed occasional large, puckered, rosette-shaped structures. The particular arrangement of the intestinal folds fulfills the dual function of promoting the absorptive process and facilitating the smooth passage of the intestinal contents. © 1995 Wiley-Liss, Inc.  相似文献

The skins of crocodylids and gavialids can be distinguished from those of alligatorids by the presence of darkly pigmented pits, known as integumentary sense organs (ISOs), on the postcranial scales. The structure of ISOs, in Crocodylus porosus, was studied using light microscopy and scanning and transmission electron microscopy. The stratum corneum of the epidermis in the area of the ISO is thinner, while the stratum germinativum is thicker, relative to other regions of the integument. Beneath the epidermal layer the ISO region has a paucity of collagen fibers relative to the rest of the dermis. Widely dispersed fibrocytes, nerve terminals, and chromatophores occur throughout the ISO region of the dermis, but these elements are concentrated in the area immediately beneath the stratum germinativum in the ISO region. The morphology of the ISOs suggests that they are sensory organs. It has traditionally been assumed that sensory organs on the amniote integument have a mechanosensory function. However, alternate functional interpretations of this structure are possible, and a resolution awaits further work. © 1996 Wiley-Liss, Inc.  相似文献

Summary The ultrastructure of normal neural lobes of adult rabbits is described. The major part consists of non-myelinated fibres containing neural swellings at intervals. These enclose the neurosecretory granules, mitochondria and a few microtubules. The swellings are connected by narrow nerve fibres with parallel neurotubules but no granules. The Herring bodies resemble large neural swellings and may contain many neurotubules. They are sometimes enclosed by multilamellate sheaths. True myelinated nerve fibres full of granules also occur. The relationship between the neural swellings and pituicytes is discussed. The pituicyte cytoplasm contains many delicate fibrils but it does not resemble an active secretory cell.The blood vessels are characterised by an unusually wide perivascular space containing two condensed layers of basement membrane. These layers can be traced into channels which extend from the perivascular spaces and break up the tissue into lobules. It is suggested that these channels form a mucopolysaccharide spongework that may be important in relation to the release of hormones.The common feature of ether-treated, dehydrated and immature animals is the absence of electron-dense cores from the neurosecretory granules. Such glands are not completely depleted of hormones and much of the hormonal activity is still associated with a sedimentable fraction. Loss of electron density may be due to diffusion of the neurophysin carrier or to a change in configuration of the neurophysin molecules resulting in inaccessibility of both osmiophilic and hormone binding sites. Summary Spermiogenesis of the architectonicid Philippia (Psilaxis) oxytropis was studied using transmission electron microscopy. Both spermatids and mature sperm of Philippia show features comparable to sperm/spermatids of euthyneuran gastropods (opisthobranchs, pulmonates) and not mesogastropods (with which the Architectonicidae are commonly grouped). These features include: (1) Accumulation of dense material on the outer membrane of anterior of the early spermatid nucleus — this material probably incorporated into the acrosome; (2) Structure of the unattached and attached spermatid acrosome (apical vesicle, acrosomal pedestal) accompanied by curved (transient) support structures; (3) Formation of the midpiece by individual mitochondrial wrapping around the axonemal complex, and the subsequent fusion and metamorphosis of the mitochondria to form the midpiece; (4) Presence of periodically banded coarse fibres surrounding the axonemal doublets and intra-axonemal rows of granules.
