Summary Larvae of the marine mosquito,Aëdes togoi Theobald, tolerate environmental salinities ranging from fresh water to 300% sea water. When they were transferred from fresh water to sea water, sodium concentration in the haemolymph increased for the first 2 days and decreased to the seawater-adapted level within 4 days. When transferred from sea water to fresh water, the sodium concentration decreased markedly for the first 2 days and attained the freshwater adapted level after 4 days. When the larvae in sea water were ligated near the anus, they died within 3 days, showing an increased sodium level in the haemolymph. The larvae ligated at the neck lost considerable body weight and died within 4 days.When the anal portion, a terminal portion of the hindgut, was catheterized, the larvae maintained in sea water showed an increase in haemolymph sodium. The anal portion epithelium of the larvae adapted to 100 and 150% sea water demonstrated a strong positive reaction to the histochemical assay for chloride ions, whereas the reaction was negative or weakly positive in freshwater adapted larvae. In the larvae with the anal papillae ligated, a slight increase in haemolymph sodium occurred while in sea water. The anal papillae were weakly positive to chloride ions. Unlike salt-water mosquito larvae of the other species, in which the rectum is considered to be involved in hyperosmotic urine production and the anal papillae appear to be the extrarenal organ, the anal portion inA. togoi larvae seems to play an important role in excretion of excess ions when placed in hyperosmotic media.  相似文献

Artificial selection was carried out on a deviant character of remnants of the anal papillae in pupae of Drosophila melanogaster, i. e. stretched (S) instead of the normal retracted (R) papillae. A chromosomal interchange between the R- and S-selection lines revealed the polygenic determination of the S-character. A comparison of lines with a different number of chromosomes of the S-line shows the importance of properties of the anal papillae for salt adaptation. By a comparison of populations obtained by artificial selection for anal papillae and by natural selection for salt adaptation we could analyse the significance of the deviant character under natural selection on media with increasing percentages of salt. Our results shows that taking into account only one character in natural selection is often a simplification. However, applying artificial selection on components of the phenotype contributing to the adaptation make an important contribution to the analysis of adaptive processes.  相似文献

The ultrastructure of the Malpighian tubules, ileum, rectum, anal canal, and anal papillae of larvae of the mosquito Culiseta inornata was examined. The Malpighian tubules, rectum, and anal papillae have many of the ultrastructural features characteristic of ion transport tissues, i.e., elaboration of the basal and apical membranes and a close association of these membranes with mitochondria. The Malpighian tubules possess two cell types, primary and stellate. The larval rectum of C. inornata is composed of a single segment containing a homogenous population of cells. In this respect, the larval rectum of C. inornata is distinct from that of saline-water species of Aedes. The cells in the larval rectum of C. inornata, however, closely resemble those of one cell type, the anterior rectal cells, of the saline-water mosquito Aedes campestris with regard to cell and nuclear size, the percentage of the cell occupied by apical folds, and mitochondrial density and distribution. No similarities can be found between the rectum of C. inornata and the posterior segment of the saline-water Aedes, which functions as a salt gland. On this basis, we have postulated that the rectum of C. inornata does not function as a site of hyperosmotic fluid secretion. The ultrastructure of the anal papillae of C. inornata is consistent with a role in ion transport. The significance of these findings to comparative aspects of osmoregulatory strategies in mosquito larvae is discussed.  相似文献

In embryological terms the anal papillae are the product of eversion of the hindgut tissues. The rectum and the anal papillae have the same origin and have a marked structural similarity. The insect hindgut is very labile being able to produce salt transporting or ‘chloride cells’ from any of the tissues of which it is composed.The hindgut consists of four distinct regions: the ileum and part of the anal canal have a mechanical function, the rectum and the posterior anal canal contain transporting cells. Two new cell types, ‘interstitial’ and ‘tertiary’ are reported in the rectum.The structure of the anal papillae changes with increased salinity. Changes in the plasma membranes alter the surface area for transport. Changes in the number of mitochondria are not accompanied by changes in oxygen consumption. If mitochondria are the site of oxidative metabolism then their number docs not control the level of oxygen consumption.In Aedes aegypti the papillary epithelium appears to be a syncytium. Across the lumen of the papillae there are cellular sheets supporting the tracheoles. At the base of the papillae there is a cellular transition zone; circular muscles in this region may be used to occlude the papillae. The control of salt transport may be hormonal.  相似文献

盐胁迫对中国柽柳幼苗生理特性的影响   总被引:15,自引:0,他引:15  
以中国柽柳(Tamarix chinensis Lour.)插穗为研究材料,在装有不同土壤盐分梯度(0.4%、0.8%、1.2%、1.6%、2.0%、2.4%)的盆钵中进行扦插试验,测定柽柳扦插成活率、叶绿素含量、超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性及丙二醛(MDA)含量等指标,研究分析柽柳扦插成活率及幼苗生理特性对盐分胁迫的响应特征。结果表明:(1)柽柳扦插成活率随盐胁迫增强逐渐降低,适合扦插繁殖的土壤含盐量低于0.8%;土壤含盐量超过0.8%后,扦插成活率太低,不适合柽柳进行扦插繁殖。(2)柽柳可通过提高叶绿素含量来适应盐胁迫,随着盐胁迫增强,柽柳扦插幼苗叶片中叶绿素含量先升高后降低,但过高的盐胁迫会破坏叶绿素的合成。(3)柽柳扦插幼苗叶片SOD和POD活性随盐胁迫增强先升高后降低,通过提高SOD和POD活性来清除细胞内多余的活性氧自由基,但活性氧自由基积累过多时,膜脂过氧化作用会破坏细胞膜的稳定性,导致抗氧化酶活性降低。(4)柽柳扦插幼苗叶片MDA含量随盐胁迫的增强先升高后降低。在土壤含盐量0.4%—1.2%范围内,MDA含量虽高于CK,但各盐分处理之间差异不显著。在含盐量为1.6%时,叶片细胞受到膜脂过氧化作用加强,MDA含量显著升高,但含盐量为2.0%时,MDA含量降低。  相似文献

NaCl胁迫对6种红花幼苗渗透调节物质及抗氧化系统的影响   总被引:1,自引:0,他引:1  
采用沙培法,对红花(Carthamus tinctorius L.)6个品种幼苗进行不同浓度的NaCl胁迫处理,研究其叶绿素(Chl)、丙二醛(MDA)、可溶性糖(SS)含量及抗氧化酶(SOD、POD和CAT)活性的变化,并用隶属函数法对其耐盐性进行综合评定。结果表明:(1)随着盐浓度的增加,除封丘红花幼苗叶绿素含量逐渐下降之外,其它品种均呈先升高后降低的趋势;(2)盐胁迫下各种红花幼苗丙二醛和可溶性糖含量较对照均有显著增加,可溶性蛋白含量先增加后减少;(3)菏泽红花和川红花SOD活性在盐浓度为50mmol/L时与对照相比升高,随后下降,其它品种均呈下降趋势;NaCl胁迫下,6个红花品种POD和CAT活性都是先升高后降低;(4)应用隶属函数法,得出耐盐性的大小顺序为:封丘红花〉菏泽红花〉川红花〉亳州红花〉云红3号〉新疆红花。  相似文献

以采自甘肃民勤一年生的沙拐枣幼苗为试材,对不同NaCl浓度(0、50、100、200、300mmol·L~(-1))处理下沙拐枣光合生理特性进行分析,并对各生理指标与地上生物量进行灰色关联度分析,以探讨荒漠植物沙拐枣的抗盐机理,为沙拐枣的保护及其恢复荒漠生态系统稳定提供理论依据。结果显示:随着NaCl浓度的升高,沙拐枣同化枝内脯氨酸含量逐渐增大,而其可溶性糖含量逐渐减小;在低浓度NaCl(50mmol·L~(-1) NaCl)处理下,同化枝光合参数均增加,且净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)均达到最大值,比对照分别显著增加了33.3%、68.0%、60.8%;与50mmol·L~(-1) NaCl相比,处理浓度超过50mmol·L~(-1) NaCl时,Pn、Gs、Tr均降低;同化枝叶绿素b含量随着NaCl浓度的增加而降低,而叶绿素a和总叶绿素含量均呈先增加后降低的趋势。灰色关联度分析发现,同化枝的Tr、Gs、Ci以及叶绿素b与地上生物量的关联度较大。研究表明,低盐浓度NaCl激活了沙拐枣的某些生理机制,有利于植株的光合作用和生长,而植物在高盐浓度胁迫时能通过调节脯氨酸和可溶性糖的含量,减少叶绿素含量、Pn和Tr等维持自身的生长。  相似文献

Axillary bud cultures of six low-land tropical varieties of potato Solanum tuberosum (L. T. 2, 5, 6, 7, 8 and 9) were studied for their response to salinity stress using crude sea salt. Concentrations of 0.8% and above severely inhibited the growth, while decreased growth was observed between 0.4% and 0.6% salt in all the varieties, with occasional shoot tip necrosis and compound leaf formation. A lower concentration of salt (0.2%) was beneficial and increased shoot weight in most varieties. Protein content showed a positive correlation with proline levels, indicating that proline accumulation was due to new synthesis rather than breakdown of existing proteins. Soluble carbohydrates increased in the stem with increasing salt. Roots showed an increase in sodium content while shoots showed increased potassium levels, with increasing salt concentrations. There was a positive correlation between dry weight yields and K/Na ratio. It is apparent that the synthesis of proline, accumulation of soluble carbohydrates as well as increase in potassium and sodium content were all used to adjust the physiology in response to salt stress by the different varieties of potato, with varying degrees, in the present study.  相似文献

Soil salinization and alkalinization frequently co-occur in naturally saline and alkaline soils. To understand the characteristics of mixed salt-alkali stress and adaptive response of Medicago ruthenica seedlings to salt-alkali stress, water content of shoots, growth and photosynthetic characteristics of seedlings under 30 salt-alkaline combinations (salinity 24–120 mM and pH 7.03–10.32) with mixed salts (NaCl, Na2SO4, NaHCO3, and Na2CO3) were examined. The indices were significantly affected by both salinity and pH. The interactive effects between salt and alkali stresses were significant, except for photosynthetic pigments. Water content of shoots, relative growth rates of shoots and roots and pigment concentrations showed decreasing trends with increasing salinity and alkalinity. The root activity under high alkalinity and salinity treatments gradually decreased, but was stimulated by the combined effects of low alkalinity and salinity. The survival rate decreased with increased salinity, except at pH 7.03–7.26 when all plants survived. Net photosynthetic rate, stomatal conductance and intercellular CO2 concentration decreased with increased salinity and pH. M. ruthenica tolerated the stress of high salt concentration when alkali concentration was low, and the synergistic effects of high alkali and high salt concentrations lead to the death of some or all seedlings. M. ruthenica appeared to be saltalkali tolerant. Reducing the salt concentration or pH based on the salt components in the soil may be helpful to abate damage from mixed salt-alkaline stress.  相似文献

试验于2011—2012年在江苏南京江苏省农业科学院经济作物研究所试验田进行,采用盆栽方法,以鲁棉研37号和苏棉22号为供试材料,设置土壤盐度降低试验(初始土壤含盐量为0.2%,棉花进入二叶期后每7d加入混合盐1次,每次增加0.1%,使土壤含盐量逐渐达到0.5%,蕾期进行盐度降低处理,使土壤含盐量降低到0.2%左右),研究蕾期土壤盐度降低后棉花叶片的生理代谢动态特征。结果表明:土壤盐度降低后,棉花叶片叶绿素(Chl)、类胡萝卜素(Car)含量和Chl/Car升高;净光合速率和气孔导度升高,且分别在土壤盐度降低后第14天和7天接近于低盐对照;土壤盐度降低后棉花叶片超氧化物歧化酶(SOD)和过氧化物酶(POD)活性升高,过氧化氢酶(CAT)活性和丙二醛(MDA)含量降低,MDA含量在土壤盐度降低后第14天接近于低盐对照;土壤盐度降低后棉花叶片中可溶性糖、游离氨基酸和脯氨酸含量降低,且接近于低盐对照。上述结果表明土壤盐度降低后,棉花叶片生理功能逐渐恢复,进而实现棉花生长发育的恢复补偿。棉花叶片生理功能在土壤盐度降低后的恢复能力存在品种间差异,鲁棉研37号较苏棉22号叶片生理功能表现出更强的恢复能力。  相似文献

In six cultivars of rice (Oryza sativa L.), Pusa Basmati 1, Basmati 370, Type III, Pant Dhan 4, CSR 10 and Pokkali, embryogenic callus growth, plant regeneration, and proline and total protein contents were studied under salt stress (on agar solidified media containing 0, 0.5, 1.0, 1.5 and 2.0 % NaCl). Four weeks after inoculation the callus fresh mass decreased with increasing salt concentration in all the six cultivars. The regeneration frequency in salt stressed callus was also lower as compared to control. 15 d and 30 d after inoculation proline content increased several fold whereas total protein content decreased markedly with increase in salt concentration.  相似文献

The comparative alterations of short term NaCl stress and recovery on growth, water relations, ionic composition, lipid peroxidation and antioxidants in roots of two rice cultivars differing in salt tolerance were studied. Exposed for 24 h to increasing (50, 100 and 150 mmol l−1) concentrations of NaCl, roots of 12D Oryza sativa L. cv. Lunishree and cv. Begunbitchi decreased in fresh weight, dry weight and relative water content. Increased Na+ and decreased K+ ion were determined at increasing NaCl concentrations. Both peroxide content and lipid peroxidation measured in terms of MDA level increased and the ratio was higher in Begunbitchi compared to Lunishree. Recovered roots showed lower peroxide and MDA content. Ascorbate and glutathione contents increased in the stressed and recovered roots of Lunishree, but decreased in Begunbitchi with increasing NaCl concentrations. Although SOD, CAT and GR activities decreased in the stressed roots, CAT activity also increased in recovered roots of both the cultivars. The POX activity increased in stressed and recovered roots of both Lunishree and Begunbitchi. Higher free radicals scavenging capacity and more efficient protection mechanism of Lunishree against salt stress, as revealed by the lower level of lipid peroxidation and improved plant water status as well as activities of some of the antioxidants, suggest that significant cultivar differences in response to salt stress in rice are closely related to differences in the activities of antioxidants and ion content. Another possible conclusion is that improved tolerance to salt stress may be accomplished by increased capacity of antioxidative system.  相似文献

以2年生的流苏播种苗为材料,采用不同浓度(50、100、200、300 mmol·L^-1)NaCl溶液进行胁迫处理,研究盐胁迫对流苏的生长、Na^+和K^+分布格局、渗透调节物质的影响,以明确其耐盐阈值。结果表明:(1)随着NaCl胁迫浓度的增加,流苏幼苗生长量逐渐降低,盐害指数升高、存活率下降;幼苗耐盐阈值为98.693 mmol·L^-1(0.577%W/V)。(2)随着NaCl胁迫浓度的增加,流苏幼苗各器官中的Na^+含量持续增加,并在浓度为50 mmol·L^-1时表现为根>叶>茎,在其余各处理组表现为叶>根>茎;幼苗根、叶中的K^+含量表现为先增后减的变化趋势,茎中K^+含量总体表现为下降趋势,且器官中K^+含量表现为根>叶>茎;幼苗根部到茎部和茎部到叶部的离子选择性运输能力、各器官中的K^+/Na^+比值均呈下降趋势。(3)随着NaCl浓度的增加,流苏幼苗叶片可溶性糖、可溶性蛋白含量总体呈上升趋势,其脯氨酸含量呈先上升后下降的趋势。研究发现,流苏幼苗根系可通过对Na^+的吸收和累积来阻止其向地上部运输进而避免盐害发生;叶片和茎中通过提高对K^+的选择性吸收和累积,从而增大K^+/Na^+比值以减缓盐分对其生理代谢的伤害。  相似文献

为筛选水稻(Oryza sativa)耐盐种质资源,研究了水稻耐盐相关生理指标。结果表明,盐胁迫下,耐盐种质Pokkali、Fl478、JX99的电导率较低,细胞膜透性较小,叶片叶绿素含量较高,受盐害程度较弱。盐胁迫促进了丙二醛的大量积累,加剧了细胞膜脂过氧化程度,而耐盐种质的丙二醛积累较少,但高盐胁迫下3个耐盐种质的丙二醛含量升高,破坏了细胞膜的完整性。感盐种质R29的临界饱和亏的增幅较大,耐盐种质相对含水量降幅小。应用模糊隶属函数法综合评价,4个水稻种质的耐盐性依次为JX99PokkaliFL478IR29。  相似文献

In the present study, we have investigated the effects of NaCl concentrations on the growth and phosphate metabolism of an Anabaena doliolum strain isolated from a paddy field, in order to determine the possible effects of salinization. Growth rate, chlorophyll content, and protein content decreased with increasing salt concentration in the growth medium, while carbohydrate concentration increased. Phosphate content and phosphate uptake rate decreased. There was an increase in total alkaline phosphatase activity, with an approximately 7-fold increase in extracellular activity compensating for an approximately 3-fold decrease in cell-bound activity. NaCl effects on protein and chlorophyll concentrations were greater in P-deficient medium, while presence or absence of P in the medium had little effect on cellular carbohydrate concentrations. It is concluded that growth in high salt likely leads to reduced phosphate uptake in A. doliolum.  相似文献

以一年生灰胡杨幼苗为试验材料,利用田间控盐控水的方法,进行干旱和盐胁迫试验,通过测定生长和生理生化指标探讨幼苗在盐旱交叉胁迫下的生长发育及适应规律,旨在阐明干旱及盐交叉胁迫下植物抗旱抗盐机理。研究结果表明:在盐、旱及交叉胁迫下,灰胡杨幼苗抗氧化酶活性、MDA和脯氨酸含量与对照存在显著差异(P0.05)。(1)在8、11 g/L和15 g/L盐处理下,灰胡杨幼苗相对高生长、相对枝长和冠幅增量均受到抑制,且差异显著(P0.05),而干旱胁迫和盐旱交互胁迫下差异不显著。(2)在盐胁迫、盐旱交叉胁迫下,随着胁迫程度的加重,抗氧化酶SOD、POD、CAT活性表现出先增加后降低的趋势,三者协调一致;仅干旱胁迫时,抗氧化酶SOD、POD、CAT活性显著增加;(3)在盐、旱及其盐旱交叉胁迫下,脯氨酸含量呈上升趋势,MDA含量则表现出先降低后升高趋势,这与抗氧化酶活性先升高后降低的趋势相对应。因此,抗氧化酶活性对缓解脂膜过氧化的伤害具有一定限度,MDA含量与抗氧化酶活性呈负相关,灰叶胡杨幼苗在盐旱交叉胁迫下表现出一定的耐性。  相似文献

The incompatibility of acid gelatin/iota-carrageenan mixtures has been studied. Both these biopolymers undergo a conformational coil-helix transition under suitable conditions of temperature and salt. The aim of this work was to study the concentration at which mixtures are incompatible and the influence of pH, salt and temperature on the phase diagram. Incompatibility occurred over a wide range of concentrations for mixtures prepared in deionized water. Compatibility was increased by increasing the pH or the salt concentration. Temperature did not greatly influence the size of the incompatible region. This is in agreement with the hypothesis that attractive electrostatic interactions lead to associative phase separation (traditionally called complex coacervation).  相似文献

A carboxymethyl starch (CMS) microgel system was prepared for the control of uptaking and releasing proteins (lysozyme). The physicochemical properties of microgels in various degrees of substitution (DS) were determined by thermal gravimetric analysis (TGA), swelling degree, and rheological analysis. The microgel particle size mostly ranged from 25 µm to 45 µm. The result obtained from the TGA studies indicated that carboxymethylation decreased the thermal stability of starch, but crosslinking increased the thermal stability of CMS. The CMS microgels showed typical pH sensitivity, and the swelling degree of microgel increased with the increasing of DS and pH, because of the large amounts of carboxyl group ionization. The samples (2.25%) could behave as viscoelastic solids since the storage modulus was larger than the loss modulus over the entire frequency range. The protein uptake increased with increasing pH and DS at low salt concentration. The optimal pH shifted to lower pH with increasing ionic strength. The saturated protein uptake decreased with increasing ionic strength at each pH. The protein was easily released from the microgel with high pH and high salt concentration.  相似文献

The effect of high salt stress on PS II heterogeneity was investigated in wheat (Triticum aestivum) leaves. On the basis of antenna size, PS II has been classified into three forms, i.e., α, β, and γ centers while on the basis of electron transport properties of the reducing side of the reaction centers, two distinct forms of PS II have been suggested, i.e., QB reducing centers and QB non-reducing centers. The chlorophyll a (Chl a) fluorescence transients, which can quantify PS II behavior, were recorded using PEA to derive OJIP in vivo with high time resolution and further analyzed according to JIP test. Our results showed that with an increase in the salt concentration during growth, the number of QB non-reducing centers increased. In antenna size heterogeneity the number of β and γ centers increased while the number of α centers decreased. A change in the energetic connectivity between the PS II units was also observed. Recovery studies showed that antenna heterogeneity was completely recovered from damage at 0.5 M NaCl concentration and partially recovered at 1 M NaCl concentration while reducing side heterogeneity showed no recovery at all after 0.5 M onwards.  相似文献
