Summary Root nodule homogenates of actinorhizal plants may representFrankia in a symbiotic stage but released from environmental influence of the host plant. Anaerobic homogenization with a blender in buffer supplied with sucrose, polyvinylpyrrolidone and reducing substances gave three times higher yields of nitrogenase activity (C2H2-reduction) than crushing the nodules in liquid nitrogen. The activity in the homogenates was very reproducible and was, on average, nearly twice as high as the activity in excised nodules and c. 10% of the activity in intact plants. The difference in activity between excised nodules and intact plants was, roughly by halves, due to removal of the root system from the pot and to excision of the nodules. The nitrogenase activity in the homogenates was slightly higher when nodule excision was done in Ar or under water as well as after treatment of the homogenate with toluene or Triton X-100 or osmotic shock. These gains in activity were considered too small to outweigh the increased complications of preparing homogenates for routine use. Due to the reproducible recovery of nitrogenase in the homogenates the technique seems useful for physiological studies on nitrogen fixation inAlnus incana.  相似文献

A flow-through gas system was used to study the effects of disturbanceon nitrogenase (acetylene reduction) activity of nodulated rootsystems of soyabean (Glycine max) and white clover (Trifoliumrepens). Detopping plus removal of the rooting medium (by shaking)produced a substantial decrease in maximum nitrogenase activity. This response is due to a reduction in oxygen flux to the bacteroidscaused by an increase in the oxygen diffusion resistance ofthe nodule. The decrease in maximum nitrogenase activity wasmuch smaller for roots subjected to detopping only. Thus, theeffect of root shaking is more important than that of shootremoval. The effect of detopping plus root shaking on nitrogenase activityoccurred whether the plants were equilibrated and assayed at25°C or 15°C. However, the effect of disturbance onthe oxygen diffusion resistance of the nodules, and thus onnitrogenase activity, was greater at the higher temperature. At the lower temperature the oxygen diffusion resistance ofthe nodules had already been increased in response to the reducedrequirement for oxygen. These nodules were less susceptibleto the effects of disturbance. Thus, comparisons of the effectsof equilibration temperature on nitrogenase activity produceddifferent results depending on whether intact or disturbed systemswere used. With intact systems activity was lower at the lowertemperature but with detopped/shaken roots the lowest activityoccurred at the higher temperature. It is concluded that the use of detopped/shaken roots can producesubstantial errors in the acetylene reduction assay, which makesthe assay invalid even when used for comparative purposes. However, comparisons with rates of 15N2 fixation and H2 production showthat accurate measurements of nitrogenase activity can be obtainedfrom maximum rates of acetylene reduction by intact plants ina flow-through gas system. The continued use of assay proceduresin which cumulated ethylene production from disturbed systemsis measured in closed vessels must be questioned. Screening of Rhizobium leguminosarum bv. phaseoli strains showed some that were able to nodulate common beans (Phaseolus vulgaris L.) at high temperatures (35 and 38°C/8 h/day). The nodulation ability was not related to the capability to grow or produce melanin-like pigment in culture media at high temperatures. However, nodules formed at high temperatures were ineffective and plants did not accumulate N in shoots. Two thermal shocks of 40°C/8 h/day at flowering time drastically decreased nitrogenase activity and nodule relative efficiency of plants otherwise grown at 28°C. Recovery of nitrogenase activity began only after seven days, when new nodules formed; total incorporation of N in tops did not recover for 2 weeks. Non-inoculated beans receiving mineral N were not affected by the thermal shock, and when growing continuously at 35 or 38°C had total N accumulated in shoots reduced by only 18%.  相似文献

Excised roots of Spartina alterniflora Loisel. and corn reduced acetylene in air without the previously reported period of zero activity lasting 8 to 18 hours. The profiles of acetylene-dependent ethylene accumulation by excised roots and intact plants of S. alterniflora were similar. No significant change in the number of bacteria associated with the roots was detectable during the assay. Most of the nitrogenase activity was detected in the roots and rhizomes of the plants. The salt marsh sediment also was capable of reducing acetylene. Additional damage to roots by washing and cutting increased the rate of acetylene reduction with samples incubated in air. Low concentrations of nitrate significantly inhibited the nitrogenase activity associated with the sediment and excised roots, but not with intact plants. Rates of acetylene reduction by excised corn roots were low. Oxidation and endogenous production of ethylene in the absence of acetylene were negligible. Measurements made with excised grass roots as described probably reflect the occurrence and magnitude of nitrogenase activity associated with the plants in the field.  相似文献

Summary Ananas comosus (L.) Merr. var. Smooth Cayenne plants when grown in vitro under different temperature regimes developed as CAM or as C3 plants. The plants used in this study were developed from the lateral buds of the nodal etiolated stem explants cultured on Murashige and Skoog medium for 3 mo. The cultures were maintained under a 16-h photoperiod for different thermoperiods. With 28°C light/15°C dark thermoperiod, as compared with constant 28°C light and dark, pineapple plants had a succulence index two times greater, and also a greater nocturnal titratable acidity and phosphoenolpyruvate carboxylase (PEPCase) activity, indicating CAM-type photosynthesis. The highest abscisic acid (ABA) level occurred during the light period, 8 h prior to maximum PEPCase activity, while the indole-3-acetic acid (IAA) peak was found during the dark period, coinciding with the time of highest PEPCase activity. These plants were also smaller with thicker leaves and fewer roots, but had greater dry weight. Their leaves showed histological characteristics of CAM plants, such as the presence of greater quantities of chlorenchyma and hypoderm. In addition, their vascular system was more conspicuous. In contrast, under constant temperature (28°C light/dark) plants showed little succulence in the leaves. There was no significant acid oscillation and diurnal variation in PEPCase activity in these plants, suggesting the occurrence of C3 photosynthesis. Also, no diurnal variation in ABA and IAA contents was observed. The results of this study clearly indicate a role for temperature in determining the type of carbon fixation pathway in in vitro grown pineapple. Evidence that ABA and IAA participate in CAM signaling is provided.  相似文献

Two pea (Pisum sativum L.) cultivars and a kidney bean (Phaseolus vulgaris L.) cultivars were grown in water cultures at different diurnal temperatures (15, 20, 24, 27, 30°C) or at 10°C night temperature combined with various day temperatures (20, 24, 27, 33 or 35°C) in the root medium. The inoculated plants were, more sensitive to the extreme temperatures than the plants supplied with combined nitrogen (KNO3). The middle-European pea cv. Violetta was adapted to somewhat higher root temperatures than the northern one cv. Torsdag II, the latter showing better growth at lower temperatures, when the plants were inoculated with the same Finnish Rhizobinm strain (HA1). Especially at optimum day temperatures the nitrogen fixation and consequently the dry weights of the inoculated plants were greatly increased when the night temperature was lowered. The optimum temperature for the growth of free-living Rhizobium strains (HA1 and H43) for peus was found to be 25°C and that of a strain (P103) for beans somewhat higher. Effective nitrogen fixation by nodulated legumes without a supply of combined nitrogen is achieved only when the optimum temperature range for root function is very close to the optimum for the rhizobia.  相似文献

The shoots of cultivated tomato (Lycopersicon esculentum cv. T5) wilt if their roots are exposed to chilling temperatures of around 5 °C. Under the same treatment, a chilling‐tolerant congener (Lycopersicon hirsutum LA 1778) maintains shoot turgor. To determine the physiological basis of this differential response, the effect of chilling on both excised roots and roots of intact plants in pressure chambers were investigated. In excised roots and intact plants, root hydraulic conductance declined with temperature to nearly twice the extent expected from the temperature dependence of the viscosity of water, but the response was similar in both species. The species differed markedly, however, in stomatal behaviour: in L. hirsutum, stomatal conductance declined as root temperatures were lowered, whereas the stomata of L. esculentum remained open until the roots reached 5 °C, and the plants became flaccid and suffered damage. Grafted plants with the shoots of one genotype and roots of another indicated that the differential stomatal behaviour during root chilling has distinct shoot and root components.  相似文献

The stored-product bruchid pests,Callosobruchus maculatus (Fabricius) andBruchidius atrolineatus (Pic) cause considerable production losses in cowpea in West Africa.Uscana lariophage Steffan parasitizes the eggs of the bruchids both in the field and in storage. As chemical control of bruchids in traditional granaries is not appropriate for poor farmers, enhancement of the efficacy of the parasitoid by environmental manipulation has been investigated. The effect of temperature on the capacity ofU. lariophaga to parasitize eggs has been studied at eleven constant and three fluctuating temperatures within the range 10 to 45°C. Longevity of the female wasp decreased with increasing temperature. The rate of development increased linearly at temperatures from 17.5 to 35°C, but decreased from 35 to 40°C. Mortality of the developing wasp remained below 20% from 20 to 37.5°C, but outside this range, mortality reached 100% at 15 at 42.5°C. Most parasitization occurred at temperatures of 25 and 30°C. Sex ratio (percentage females) increased with temperature in the high temperature range. The intrinsic rate of increase (rm) forU. lariophaga was highest in the temperature range from 30 to 37.5°C and was higher than that ofC. maculatus at all temperatures. While the rm value ofC. maculatus did not vary much at temperatures from 25 to 35°C, the rm value of the wasp doubled. Relative humidity did not effect longevity, egg-laying capacity, mortality, development time and sex ratio of the wasps withC. maculatus as host. However, withB. atrolineatus as the host, development time and mortality increased at lower relative humidity levels. The results indicate that temperature is the major regulating factor on the parasitoid. As the type of storage structure and its location (sun or shade) affects the temperature inside the store, ways are being investigated of manipulating the storage environment through temperature regulation to increase the impact of the parasitoid.  相似文献

In order to shed new light on the mechanisms of salt-mediated symbiotic N2-fixation inhibition, the effect of salt stress (75 mM) on N2-fixation in pea root nodules induced by R. leguminosarum was studied at the gene expression, protein production and enzymatic activity levels. Acetylene reduction assays for nitrogenase activity showed no activity in salt-stressed plants. To know whether salt inhibits N2-fixing activity at a molecular or at a physiological level, expression of the nifH gene, encoding the nitrogenase reductase component of the nitrogenase enzyme was analyzed by RT-PCR analysis of total RNA extracted from nodulated roots. The nifH messenger RNA was present both in plants grown in the presence and absence of salt, although a reduction was observed in salt-stressed plants. Similar results were obtained for the immunodetection of the nitrogenase reductase protein in Western-blot assays, indicating that nitrogen fixation failed mainly at physiological level. Given that nutrient imbalance is a typical effect of salt stress in plants and that Fe is a prosthetic component of nitrogenase reductase and other proteins required by symbiotic N2-fixation, as leghemoglobin, plants were analyzed for Fe contents by atomic absorption and the results confirmed that Fe levels were severely reduced in nodules developed in salt-stressed plants. In a previous papers (El-Hamdaoui et al., 2003b), we have shown that supplementing inoculated legumes with boron (B) and calcium (Ca) prevents nitrogen fixation decline under saline conditions stress. Analysis of salt-stressed nodules fed with extra B and Ca indicated that Fe content and nitrogenase activity was similar to that of non-stressed plants. These results indicate a linkage between Fe deprivation and salt-mediated failure of nitrogen fixation, which is prevented by B and Ca leading to increase of salt tolerance.  相似文献

Physiology of Root-Associated Nitrogenase Activity in Oryza sativa   总被引:2,自引:1,他引:1       下载免费PDF全文
An intact method for measuring immediately linear rates of acetylene reduction was used to investigate the relationship between temperature, pH, O2 concentration, and light intensity with the rate of root-associated nitrogenase activity in rice (Oryza sativa L.). Nitrogenase activity varied over a temperature range of 10 to 50°C and optimal rates of acetylene reduction were recorded at 35°C. Nitrogenase activity was also influenced by the pH of the liquid surrounding the roots prior to assay. Maximal rates of acetylene reduction were recorded over a pH range from 5.8 to 7.5. Nitrogenase activity was significantly reduced by concentrations of O2 0.5% (v/v) or more when the intact plant assay method was used, and no optimum was detected. However, when the plant tops were removed and the cut ends sealed from the atmosphere for 4 hours, acetylene reduction rates were maximal at 0.25% O2 (v/v). When plants were moved from sunlight (1,400 microeinsteins per square meter per second) to shade (9.6) root-associated nitrogenase activity at 35° C significantly decreased 15 min later to one-fourth the rate and recovered upon return to sunlight. When the light intensity reaching the leaf canopy was progressively reduced from 1,050 to 54 microeinsteins per square meter per second the rate of root-associated nitrogenase activity decreased from 550 ± 135 to 192 ± 55 nanomoles ethylene per gram dry root per hour. The study suggests that the rate of root-associated nitrogenase activity in rice at constant temperature may well be mediated by variations in the concentration of O2 resulting from changes in the rate of photosynthesis as well as variations in the rate of transport of photosynthate.  相似文献

Hydrogen was produced from various marine macro-algae (seaweeds) through anaerobic fermentation using an undefined bacterial consortium. In this study, anaerobic fermentation from various marine macro-algae for Ulva lactuca, Porphyra tenera, Undaria pinnatifida, and Laminaria japonica was studied. From this analysis Laminaria japorica was determined to be the optimum substrate for hydrogen production. When L. japornica was used as the carbon source for enhanced hydrogen production, the optimum fermentation temperature, substrate concentration, initial pH, and pretreatment condition were determined to be 35°C, 5%, 7.5, and BT120 (Ball mill and thermal treatments at 120°C for 30 min), respectively. In addition, hydrogen production was improved when the sludge was heat-treated at 65°C for 20 min. Under these conditions, about 4,164 mL of hydrogen was produced from 50 g/L of dry algae (L. japonica) for 50 h, with a hydrogen concentration around 34.4%. And the maximum hydrogen production rate and yield were found to be 70 mL/L·h and 28 mL/g dry algae, respectively.  相似文献

A leaf spot disease called frosty mildew was observed onChaenomeles sinensis throughout Japan. Small brown spots with white tufts occurred followed by successive defoliation. On the fallen leaves, minute black dots are formed. The causal fungus was regarded as a new species ofMycosphaerella, M. chaenomelis, andCercosporella chaenomelis in anamorph. Pathogenicity of the fungus was confirmed only inC. sinensis by inoculation experiments. Colonies of the fungus grew well on potato sucrose agar, and grew at 10–30°C with an optimum temperature of 25°C. The fungus overwintered on the fallen diseased leaves in the form of pseudothecia, and ascospores served as the primary infection source from April to June.  相似文献

Relative nuclear DNA contents in cortex parenchyma cells in root segments of 3- and 7-d-old soybean seedlings grown at 25 °C and in plants grown for 3 d at 25 °C, and then for 4 d at 10 °C, were determined with cytophotometry. Measurements revealed that in each variant the cortex cell nuclei with DNA content between 2C and 8C were in all the examined segments and nuclei with 8C – 16C DNA appeared in higher parts of roots. However, in chilled plant cells the number of 8C – 16C DNA nuclei was very low. Therefore, chilling inhibited endoreplication in comparison with plants grown at 25 °C for 7 d, and even reduced endopolyploidy level as compared to the initial seedlings, i.e. 3-d-old plants. DNA contents in root hairs grown at 25 °C (control) and in root hairs emerged at 10 °C were also determined. In controls 4C – 8C DNA nuclei predominated while in chilled plants an additional population of 2C – 4C DNA appeared. Thus a reduction of DNA synthesis was brought about by low temperature. The occurrence of an intermediate DNA contents besides those with full endoreplication cycles suggests the possibility of differential DNA replication. This suggestion seems to be supported by the lack of 3H-thymidine incorporation into root hair nuclei at the examined developmental stage both in control and chilled root hairs. The same number, but larger, chromocentric lumps in polyploid cortex cell nuclei of higher root zones, in comparison to meristematic nuclei, suggests that endoreduplication process occurred. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Factorial effects of photoperiod (6, 12 and 18 h) and root-zone temperatures (9, 15 and 21°C) on the growth and mineral nutrient concentration and partitioning in maize (Zea mays L.) were investigated. Strong interactions were observed between photoperiod and root-zone temperature on the growth and concentration of numerous mineral elements in the plant tops and roots. For example, a threefold increase in photoperiod (from 6 to 18 h) did not affect the growth of tops or roots if the root-zone temperature was 9°C but increased them each by eightfold if the root-zone temperature was 21°C. On the other hand, raising the root-zone temperature from 9 to 21°C increased the growth of tops and root each by ca. threefold when plants were grown with 6 h of light. At 18 h photoperiod, however, plant growth was increased 20- to 30-fold by the same rise in the root-zone temperature. The concentrations of different mineral elements in the roots and tops were affected quite differently by the interacting effects of photoperiod and root-zone temperature. In general, increasing the photoperiod at a given root-zone temperature decreased the concentrations of elements while increasing the root-zone temperature at a given photoperiod increased the concentrations of most elements in both roots and tops. The exceptions were K and B which reacted opposite to each other: K concentration in both tops and roots was relatively insensitive to photoperiod but very sensitive to root-zone temperature and the reverse was true for boron. The relative insensitivity of plant growth to increased day length as long as the roots are subjected to suboptimal (low) soil temperatures may have survival significance and point to the predominant role of root temperature over that of day length in the early growth of maize. A possible mechanism by which photoperiod and root-zone temperature might interactively alter the nutrient uptake by the roots is discussed.  相似文献

The effects of three salinities (0, 100 and 500 mM NaCl) and four constant temperatures (10, 20, 30 and 35 °C) on seed germination of Halostachys caspica (M. B.) C. A. Mey., Kalidium foliatum (Pall.) Mop. and Halocnemum strobilaceum (Pall.) Bieb. were investigated. After seeds were treated with different concentrations of NaCl at constant temperatures of 10–35 °C for 16 days, ungerminated seeds were transferred to distilled water for 10 days to investigate the total germination; after this time, the ungerminated seeds from the 10 and 20 °C treatments were then moved to 35 °C for another 5 days to determine the final germination. The three plant species in the present experiment are salt-resistant euhalophytes growing in high saline soils in the Zhungur Basin in Xinjiang, a northwest province of China.Compared with germination under control conditions, germination percentages of all three species were not affected by 100 mM NaCl at 10–35 °C, while severely inhibited by 500 mM NaCl; germination percentages were very low at 10 °C up to 100 mM NaCl for all species; the optimum temperature for germination of H. caspica and K. foliatum was 20–30 °C, while 35 °C for H. strobilaceum, up to 100 mM NaCl; seeds did not suffer ion toxicity for all species, as evidenced by the high total germination after ungerminated seeds pretreated with 500 mM NaCl were transferred to distilled water at constant temperatures of 10–35 °C for 10 days, and the high final germination after the ungerminated seeds from the 10 and 20 °C treatments were subsequently moved to 35 °C for another 5 days; Halostachys caspica had greater sensitivity to increasing temperatures from 10 and 20 °C to 35 °C compared with the other two species.  相似文献

Development, survival, fecundity, progeny sex ratio (PSR) and age-specific life-table parameters of the parasitoid Campoletis chlorideae Uchida (Hymenoptera: Ichneumonidae) were examined at six different constant temperatures (12, 17, 22, 27, 32 and 37°C) in the laboratory [70 ± 10% RH and 10:14 h (light:dark) photoperiod]. Second instar larvae of Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) were reared on chickpea (Cicer arietinum L.) and used as the host. Development times shortened as the temperature increased from 12 to 37°C. The estimated lower developmental threshold (tL) was 3.4°C. The thermal summation for total immature stages was 379.97 degree-days. A reciprocal relationship between temperature and longevity was observed in the range of 12–17°C. The maximum mortality of pupae (71.8%) occurred at 37°C. At 22°C, the yield of a female parasitoid averaged 137.3 ± 14.7 (mean ± SD) progeny, of which 89.6 ± 7.6 were daughters. The number of daughters produced decreased when the females were kept either above or below 22°C, although the PSR was female biased in the range of 17–27°C. The analyses of life-table parameters, developmental rates, reproduction, mortality and PSR suggest that maximum population growth (r m ) is near 27°C. There was little variation observed in most of the desired qualities of C. chlorideae in the range of 17–27°C, and it appears that the parasitoid is adapted to a wide range of temperatures. We suggest that for maximum production the parasitoid should be reared at 22 ± 4°C and be released in areas where the temperature ranges between 17° and 27°C, as in the plains of northern India.  相似文献

The sequence of events leading up to the establishment of symbiotic nitrogen-fixation were studied in two tropical legumes, Centrosema pubescens Benth, and Vigna unguiculata L. Walp. Parameters measured included fresh and dry weights, chlorophyll and leghaemoglobin contents, as well as the activities of NADH-nitrate reductase (EC 1.6.6.1), and nitrogenase (nitric-oxide reductase-EC 1.7.99.2) in plants that were inoculated with suitable rhizobia or which were watered with potassium nitrate. Dry weight and photosynthetic activity of both species followed the sigmoidal pattern which is characteristic of most plants. Growth was little different in either a qualitative or quantitative sense whether nitrogen was supplied as nitrate or through dinitrogen fixation. Although the biochemical sequence of events was dependent on the limiting sensitivities of the individual assays used, the data suggest that nitrate reductase is the first measurable enzymatic activity in the nodules (and roots), followed by acetylene reduction and leghaemoglobin in that order. It is possible therefore, that low levels of symbiotic nitrogen fixation occur in the nodules in the absence of leghaemoglobin. Nitrate reductase activity in C. pubescens nodules was negatively exponentially correlated with nitrogenase activity of the same nodules, suggesting a changing metabolism in old nodules. These data are discussed in terms of environmental and physical factors known to control nitrogen fixation.  相似文献

Summary Nitrogen fixers make up a large percentage of the total microflora in the rhizosphere of lowland rice. There are more aerobic nitrogen fixers than there are anaerobic ones. When soil crumbs from the root zone were placed on a nitrogen free agar medium and inoculated at 0, 5, 10, and 21 percent oxygen concentration, colonies of aerobic nitrogen fixers reached their greatest diameter at 5 and 10 percent oxygen. In acetylene reduction assays rice plants grown in paddy fields and in solution culture were tested for the nitrogenase activities of their roots at different oxygen tensions. Nitrogenase activity was highest at 3 percent oxygen, lower at 0 percent, and far lower at 21 percent. When rice was grown in solution culture the redox potential of the nutrient solution strongly influenced nitrogenase activity. With declining redox potential, nitrogenase activity increased to a maximum value but dropped sharply as redox potential further decreased. Ten ppm of combined nitrogen as urea depressed nitrogenase activity on excised roots. Combined nitrogen applied to one part of the root system affected, to some extent, nitrogen fixation on other roots kept in a solution without nitrogen. Nitrogenase activity in a fertility trial with lowland rice, examined at several dates, showed no inhibitory effect of fertilizer nitrogen, however, presumably because the nitrogen concentration in the soil solution rapidly decreased. Instead, an overall stimulating effect of nitrogen dressing was noticeable. Diurnal fluctuations of nitrogenase activity in the rhizosphere, with a peak in the afternoon and low fixation rates after low solar radiation, suggest a photosynthetic effect on nitrogen fixation. re]19751208  相似文献

The production of transgenic broccoli (Brassica oleracea) with increased shelf-life using an Agrobacterium rhizogenes-mediated co-transformation protocol is reported. An Agrobacterium rhizogenes Ri vector, pRi1855:GFP was constructed to allow expression of the green fluorescent protein to identify insertion of Ri TL-DNA into plant cells. The Brassica oleracea ACC synthase 1 and ACC oxidase 1 and 2 cDNAs in sense and antisense orientations were co-transformed into GDDH33, a doubled haploid calabrese-broccoli cultivar. Transformation efficiency was 3.26%, producing 150 transgenic root lines, of which 18 were regenerated into mature plants. The floral buds from T0 broccoli heads were assayed for post-harvest production of ethylene and chlorophyll levels. Buds from T0 lines transformed with ACC oxidase 1 and 2 constructs produced significantly less post-harvest ethylene at 20 °C than the untransformed plants and chlorophyll loss was significantly reduced over a 96 h post-harvest period. The T0 plants transformed with sense and antisense ACC synthase 1 had a significantly reduced 24 h post-harvest ethylene peak and delayed chlorophyll loss. A positive correlation between post-harvest bud ethylene production and chlorophyll loss was described by a regression. This demonstrates that the shelf-life of a very perishable vegetable may be increased up to 2 days at 20 °C by reducing post-harvest ethylene production.  相似文献

We investigated the influence of root zone temperature (RZT) and the aerial application of paraquat on stress defence mechanisms of Trichosanthes cucumerina L. To achieve this objective, T. cucumerina cv Green was grown with roots at 25 and 30°C root zone temperature and maintained at 20 ± 1°C air temperature in a growth chamber. These RZT and air temperature had earlier been shown to favor growth and fruit production in T. cucumerina. Plants at each RZT were subjected to paraquat treatment (+P) and without paraquat treatment (−P). Paraquat (0.2 mmol/L) was applied as aerial spray. Results showed that the individual main effects of RZT and paraquat treatments significantly affected the chlorophyll fluorescence and gas exchange parameters, while the interaction of both treatments had no significant effect. Results showed that the total phenolics and ascorbic acid contents of T. cucumerina at 30°C were significantly higher than at 25°C. The T. cucumerina plants in +P treatment recorded significantly lower maximum photochemical efficiency (F v/F m), net photosynthesis (A), transpiration rate (E), intercellular CO2 concentration (C i) and stomatal conductance (g 1) compared to untreated plants. Also, plants raised at 30°C recorded significantly higher F v/F m, A, E, C i and g 1 compared to plants raised at 25°C. Plants that were sampled at 48 h after paraquat treatment recorded a higher degree of oxidative damage compared to those sampled at 24 h after treatment. We showed that the degree of damage suffered by T. cucumerina, when treated with paraquat either at 25 or 30°C RZT was similar at 48 h after treatment. We concluded that either at 25 or 30°C, exposure of T. cucumerina to paraquat would impose the same degree of oxidative damage.  相似文献
