Laboratory investigation on vermicomposting efficacy of Perionyx excavatus for recycling of distillation waste biomass of java citronella (Cymbopogon winterianus Jowitt) was carried out in two seasonal trials i.e. summer and winter periods. The experiment was conducted in earthen pots using a mixture of citronella waste material and cowdung in the proportion of 5:1. A control treatment without earthworms was setup for comparison of the results. The vermicompost had shown 5.8 folds reduction in C/N ratio and 5.6 folds enhancement in ash content. The nutrient contents (N, P, K, Ca and Mg) in the vermicompost had increase in the range of 1.2 - 4.1 fold than the initial level. The FT-IR spectra of the vermicompost confirmed increase in nitrogen rich compounds and decrease in aliphatic/aromatic compounds as compared to the initial level of the biowaste materials. The vermicomposting process is influenced by seasonal variation and summer was more productive than winter.  相似文献

Municipal solid waste (MSW) was vermicomposted in combination with cowdung (CD) using Perionyx ceylanensis for 50 days. The decomposition rate of 55-78% was observed in different vermibed substrates, the highest being CD followed by 10:1 ratio of CD + MSW. The C/N ratio was reduced from 41.8 to 17.6 and 38.8 to 15.4 in MSW + CD (10:1) and CD, respectively. The difference in the final C/N ratio between MSW + CD (10:1) and CD vermicompost was not significant (p < 0.05). The important nutrients, NPK showed significantly (p < 0.05) higher contents in vermicomposts than worm-unworked composts. The degradation rate of cellulose and lignin was 37% and 12%, respectively, in 10:1 vermibed mix with P. ceylanensis. The bacterial, fungal and actinomycetes population in vermicompost was significantly higher than in the compost. The biomass, number and cocoons of P. ceylanensis collected after 50 days showed increase with the increase of CD incorporation in MSW.  相似文献

An integrated composting-vermicomposting process has been developed for utilization of primary sewage sludge (PSS). Matured vermicompost was used as bulking material and a source of active microbial culture during aerobic activated composting (AAC). AAC resulted in sufficient enrichment of bulking material with organic matter after 20 cycles of recycling and mixing with PSS and produced materials acceptable for vermicomposting. Vermicomposting caused significant reduction in pH, volatile solids (VS), specific oxygen uptake rate (SOUR), total organic carbon (TOC), C/N ratio and pathogens and substantial increase in electrical conductivity (EC), total nitrogen (TN) and total phosphorous (TP) as compared to compost. Environmental conditions and stocking density have profound effects on vermicomposting. Temperature of 20 °C with high humidity is favorable environmental condition for vermicomposting employing Eisenia fetida. Favorable stocking density range for vermiculture is 0.5-2.0 kg m−2 (optimum: 0.5 kg m−2) and for vermicomposting is 2.0-4.0 kg m−2 (optimum: 3.0 kg m−2), respectively.  相似文献

Solid waste management is a worldwide problem and it is becoming more and more complicated day by day due to rise in population, industrialization and changes in our life style. Transformation of industrial sludges into vermicompost is of double interest: on the one hand, a waste is converted into value added product, and, on the other, it controls a pollutant that is a consequence of increasing industrialization. Garden waste, kitchen waste and cow dung were subjected to recycle through vermicomposting by using the epigeic earthworm Eisenia fetida under field conditions. The pH, moisture content, total organic carbon, humus, nitrogen, phosphorous and potassium in vermicompost was analysed. It was found that moisture content, total organic carbon, humus, nitrogen, phosphorous and potassium was high in cow dung, followed by kitchen waste and garden waste. This study clearly indicates that vermicomposting of garden waste, kitchen waste and cow dung can not only produce a value added produce (vermicomposting) but at the same time reduce the quantity of waste.  相似文献

The aim of this study was to assess the potential of Allolobophora parva Eisen as a candidate for vermicomposting practices. Five organic waste mixtures: cow dung (CD), biogas plant slurry (BGS), cow dung + vegetable waste (CD + VW), BGS + VW and VW + Soil were vermicomposted using A. parva. Vermicomposting showed a decrease in pH, organic C and C:N ratio, but increase total N, available P and exchangeable K at the end. C:N ratio of end material (vermicompost) was within the agronomic acceptable limit (<20). The high level of NPK in worm-processed material indicates the candidature of this species for waste management operations. The earthworm also showed an excellent growth in different wastes. Results thus indicate that A. parva appeared a potential tool for conversion of organic wastes into value added products for sustainable land restoration practices.  相似文献

Flagellated bacteria, such as Escherichia coli, are able to swim up gradients of chemical attractants by modulating the direction of rotation of their flagellar motors, which spin alternately clockwise (CW) and counterclockwise (CCW). Chemotactic behavior has been studied under a variety of conditions, mostly at high loads (at large motor torques). Here, we examine motor switching at low loads. Nano-gold spheres of various sizes were attached to hooks (the flexible coupling at the base of the flagellar filament) of cells lacking flagellar filaments in media containing different concentrations of the viscous agent Ficoll. The speeds and directions of rotation of the spheres were measured. Contrary to the case at high loads, motor switching rates increased appreciably with load. Both the CW → CCW and CCW → CW switching rates increased linearly with motor torque. Evidently, the switch senses stator-rotor interactions as well as the CheY-P concentration.  相似文献

Pineapple wastes, an abundant organic waste in Accra, Ghana, were vermicomposted using native earthworms (Eudrilus eugeniae Kinberg) collected from the banks of streams and around bath houses of this city. Triplicate pilot-scale vermidigesters containing about 90 earthworms and three other control boxes with no earthworms were fed pineapple pulp or peels, and the loss of wet mass was monitored over 20 weeks. In a second experiment, a 1:1 mixture of pineapple peels and pulp (w/w) was fed to triplicate pilot-scale vermicomposters and control boxes during a 20 week period. One month after feeding ended, the vermicompost and composted (control) waste was air dried and analyzed. During the first experiment, the vermicomposted pineapple pulp and peels lost 99% and 87% of their wet mass, respectively, indicating the potential for vermicomposting. Fresh pineapple waste exhibited an initial pH of 4.4, but after 24 weeks, the vermicompost and compost had acquired a neutral to alkaline pH of 7.2–9.2. The vermicompost contained as much as 0.4% total N, 0.4% total P and 0.9% total K, and had a C:N ratio of 9–10. A reduction of 31–70% in the Escherichia coli plus Salmonella loads and 78–88% in the Aspergillus load was observed during vermicomposting. The rapid breakdown of pineapple wastes by E. eugeniae demonstrated the viability of vermicomposting as a simple and low cost technology recycling this waste into a soil amendment that could be used by the 2500 vegetable producers of Accra and its surrounding areas.  相似文献

Chemokine receptors CCR5 and CXCR4 are of major importance in the pathogenesis of HIV-1 infection because they are co-receptors for human immunodeficiency virus (HIV) entry. We examined the frequency of CD3CD56+CCR5+ and CD3CD56+CXCR4+ in HIV-infected long-term slow progressors (SPs), HIV typical progressors (TPs) with or without highly active antiretroviral therapy (HAART), and HIV-seronegative controls. The results showed that the frequency of CD3CD56+CCR5+ was up-regulated, and frequency of CD3CD56+CXCR4+ was down-regulated in HAART-naïve HIV TPs group compared with HIV SPs group and HIV-seronegative controls (P < 0.05). The frequency of CD3CD56+CCR5+ was down-regulated by HAART therapy (P < 0.05). The frequency of CD3CD56+CCR5+ was lower in HIV SPs compared with controls (P < 0.05). Lower frequency of CD3CD56+CXCR4+ and higher frequency of CD3CD56+CCR5+ positively correlated with the level of HIV viral loads and negatively correlated with CD4 T cell counts (P < 0.05). These results indicated that the expression of chemokine receptors on NK cells correlated with HIV disease progression.  相似文献

Many epidemiological studies have investigated IL1α and IL1β polymorphisms with SLE risk, but no conclusions are available because of conflicting results. This meta-analysis was performed to more precisely estimate the relationships. The databases of PubMed updated to September 1st, 2012 were retrieved. Odds ratio (OR) and corresponding 95% confidence interval (95% CI) as effect size were calculated by a fixed- or random-effect model. In total, six case–control studies for IL1β − 511C/T, four studies for IL1β + 3953C/T, three studies for IL1α − 889C/T and three studies for IL1α + 4845G/T were involved in this analysis. The results indicated that for IL1α − 889C/T polymorphism T allele was associated with decreased risk of SLE (OR (95% CI)) (T vs. C: 0.802 (0.679–0.949); TT + CT vs. CC: 0.615 (0.380–0.995); TT vs. CC: 0.679 (0.466–0.989)). However, when analysis for TT vs. CT + CC was conducted, the result indicated that IL1α − 889C/T polymorphism was not associated with SLE (OR (95% CI): 0.847 (0.595–1.205)). Combined analysis indicated that IL1β − 511C/T polymorphism was not overall associated with risk of SLE (OR (95% CI)) (T vs. C: 1.113 (0.954–1.298); TT vs. CT + CC: 1.146 (0.889–1.447); TT + CT vs. CC: 1.145 (0.903–1.452); TT vs. CC: 1.255 (0.928–1.698)). When subgroup analysis for Asian ethnicity was conducted, the results indicated that IL1β − 511C/T polymorphism was associated with SLE only for TT vs. CT + CC (OR (95% CI): 1.468 (1.001–2.152)), but was not associated for T vs. C (OR (95% CI): 1.214 (0.955–1.544)), TT + CT vs. CC (OR (95% CI): 1.112 (0.765–1.615)) and TT vs.CC (OR (95% CI): 1.411 (0.896–2.222)). In addition, overall analyses indicated that IL1β + 3953C/T and IL1α + 4845G/C polymorphisms were also not associated with risk of SLE (OR (95% CI)) (for IL1β + 3953C/T T vs. C: 0.996 (0.610–1.626), TT vs. CT + CC: 0.658 (0.318–1.358), TT + CT vs. CC: 1.021 (0.618–1.687), TT vs. CC: 0.640 (0.309–1.325); for IL1α + 4845G/T T vs. G: 1.067 (0.791–1.440), TT + GT vs. GG: 0.934 (0.646–1.351)).This study inferred that IL1α − 889C/T polymorphism might be moderately associated with SLE, but no sufficient evidence was available to support any associations between IL1β + 3953C/T or IL1α + 4845G/C polymorphisms and SLE. We could not draw a definite conclusion between IL1β − 511C/T polymorphism and risk of SLE owing to the limited data. Further large sample-sized studies should be required.  相似文献

Lactobacillus buchneri is probably the most beneficial microorganism for efficient preservation of animal feed silages made from grass, maize and other plant material against aerobic spoilage. Its obligatory heterofermentative nature, acid resistance and robustness have drawn attention to this species for applications as silage starter culture as well as for genetic engineering. For the first time, two cryptic plasmids present in the same L. buchneri strain, L. buchneri CD034, were isolated, sequenced and characterized. The larger plasmid, designated pCD034-1 was found to be 3424 bp in length with a G + C content of 38.36%. The smaller plasmid, designated pCD034-2 was found to be 2707 bp in length with a G + C content of 38.60%. On both plasmids we predicted three open reading frames. On pCD034-1, ORF 1 encodes a putative replication protein which shares 99% identity with the RepA protein of a Lactobacillus plantarum derived pC194/pUB110-family plasmid. ORF 2 encodes a putative protein of unknown function. ORF 1 and ORF 2 of pCD034-2 correspond to RepA and RepB proteins similar to those of plasmid pLB4 from L. plantarum. ORF 3 of both plasmids encodes a putative mobilization protein similar to that of the pediococcal plasmid pF8801. Double strand origins, putative single strand origins and typical mobilization start signals were identified. Both plasmids were shown to be maintained at relatively high plasmid copy numbers. Two shuttle vectors carrying the origins of replication of pCD034-1 and pCD034-2 were constructed and used to successfully transform two other species isolated from the same environment. Hence, we consider the two novel L. buchneri plasmids a valuable resource for the generation of shuttle and expression vectors for LAB.  相似文献

Antigen-specific regulatory CD4+ T cells have been described but there are few reports on regulatory CD8+ T cells. We generated islet-specific glucose-6-phosphatase catalytic subunit-related protein (IGRP)-specific regulatory CD8+ T cells from 8.3-NOD transgenic mice. CD8+ T cells from 8.3-NOD splenocytes were cultured with IGRP, splenic dendritic cells (SpDCs), TGF-β, and all-trans retinoic acid (ATRA) for 5 days. CD8+ T cells cultured with either IGRP alone or IGRP and SpDCs in the absence of TGF-β and ATRA had low Foxp3+ expression (1.7 ± 0.9% and 3.2 ± 4.5%, respectively). In contrast, CD8+ T cells induced by exposure to IGRP, SpDCs, TGF-β, and ATRA showed the highest expression of Foxp3+ in IGRP-reactive CD8+ T cells (36.1 ± 10.6%), which was approximately 40-fold increase compared with that before induction culture. CD25 expression on CD8+ T cells cultured with IGRP, SpDCs, TGF-β, and ATRA was only 7.42%, whereas CD103 expression was greater than 90%. These CD8+ T cells suppressed the proliferation of diabetogenic CD8+ T cells from 8.3-NOD splenocytes in vitro and completely prevented diabetes onset in NOD-scid mice in cotransfer experiments with diabetogenic splenocytes from NOD mice in vivo. Here we show that exposure to ATRA and TGF-β induces CD8+Foxp3+ T cells ex vivo, which suppress diabetogenic T cells in vitro and in vivo.  相似文献

The complete mitochondrial genome (mitogenome) of a female flightless geometrid moth Apocheima cinerarius was found to be 15,722 bp in length, containing 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and a control region. The A + T content of the complete mitogenome is 80.83%. The AT skew value ([A − T] / [A + T]) is 0.027. The 13 PCGs of the mitogenome start with typical ATN codons, except for cox1 with the start codon CGA. All the tRNA genes have typical cloverleaf secondary structures, except for trnSer(AGN). The secondary structures of rrnL and rrnS were predicted. Six structural domains including conserved regions (IV, V) and variable regions (I, II, III, VI) were identified in the secondary structure of rrnL. The secondary structure of rrnS consists of 3 structural domains. The control region of A. cinerarius begins with conserved motifs of “ATAGA” + 19-bp poly T. It also contains a microsatellite-like (TA)26, a stem-and-loop structure, and a poly-A stretch. Phylogenetic analysis showed that Geometroidea is more closely related to Bombycoidea than to Noctuoidea. A. cinerarius is more closely related to Biston panterinaria than to Phthonandria atrilineata, which is in accordance with the conventional morphology-based classification.  相似文献

Most of the previous studies on vermicomposting have been conducted as lab trials at small-scale (SS) using small quantity of waste mixtures. Efforts were made in this study to stabilize the sewage sludge amended with sugarcane trash using pilot-scale (PS) vermicomposting operation. Results of PS vermireactors were compared with SS trials in terms of quality of ready vermicompost and earthworm production rates. Results thus suggest a clear-cut difference between SS and PS in terms of waste mineralization rate and earthworm production. The waste mineralization rate in PS was significantly lower than SS (P < 0.05). Total N and available P were higher in end product from SS, while exchangeable cations (Ca2+ and K+) showed reverse behavior during the process of waste stabilization. There was significant difference between PS and SS for metal remediation rate in end materials. The growth and reproduction pattern of Eisenia fetida was completely different in PS as compared to lab trials, i.e. SS. Probably, the distinct earthworm stocking density and microclimate conditions in SS and PS were responsible for observed differences in results of waste mineralization rate and earthworm growth. This study suggests that SS laboratory trials may differ in PS field operations due to distinct behavior of earthworm in field conditions. It is concluded that SS laboratory trials should be tested in field at large-scale in order to measure the feasibility of technology for large-scale waste decomposition operations in open conditions.  相似文献

We present the complete mitogenome of a stonefly, Cryptoperla stilifera Sivec (Plecoptera; Peltoperlidae). The mitogenome was a circular molecule consisting of 15,633 nucleotides, 37 genes and a A + T-rich region. C. stilifera mitogenome was similar to Pteronarcys princeps mitogenome (Plecoptera; Pteronarcyidae). All transfer RNA genes (tRNAs) had typical cloverleaf secondary structures except for trnSer (AGN), where the stem-loop structure of the dihydrouridine (DHU) arm was missing. The A + T-rich region of C. stilifera had two stem-loops and each had two interlink. Three conserved sequence blocks (CSBs) were present in the A + T-rich regions of C. stilifera, Peltoperla tarteri and Peltoperla arcuata. Moreover, many polynucleotide stretches (Poly N, N = A, T and C) in the A + T-rich region of C. stilifera Phylogenetic relationships of Polyneopteran species were constructed based on the nucleotide sequences of 13 protein coding genes (PCGs). Both maximum likelihood (ML) and Bayesian inference (BI) analyses supported Grylloblattodea as the sister group to Plecoptera + Dermaptera and Embiidina and Phasmatodea as sister groups.  相似文献

We tested the effects of UV radiation (UVR) and nitrate limitation on the production of dimethylsulfide (DMS), particulate dimethylsulfoniopropionate (DMSPp), and particulate dimethylsulfoxide (DMSOp) in natural seawater from the Gulf of Mexico and in phytoplankton cultures. DMS/Chl a ratios in PAR-only and PAR + UV-exposed seawater were 0.44–2.0 and 0.46–1.9 nmol DMS μg−1 Chl a, respectively, whereas the ratios in cultures of Amphidinium carterae were 1.0–2.2 nmol μg−1 in PAR-exposed samples and 0.91–2.1 nmol μg−1 in PAR + UV-exposed samples. These results suggested that UVR did not substantially affect DMS/Chl a ratios in seawater and A. carterae culture samples. Similarly, UVR had no significant effect on DMSOp/Chl a in seawater samples (0.83–1.6 nmol DMSO μg−1 Chl a for PAR + UV vs. 0.70–1.5 nmol μg−1 for PAR-exposed seawater samples, respectively) or in A. carterae cultures (0.20–1.3 and 0.19–0.88 nmol DMSO μg−1 Chl a in PAR + UV- and PAR-exposed cultures, respectively). In an experiment with the diatom, Thalassiosira oceanica, the culture was grown in high nitrate (30 μM) or low nitrate (6 μM) media and exposed to PAR-only or PAR + UV. The low nitrate, PAR-only samples showed an increase of intracellular dimethylsulfoniopropionate (DMSP) concentration from 2.1 to 15 mmol L−1 in 60 h, but the increase occurred only after cultures reached the stationary phase. Cultures of T. oceanica grown under UVR had lower growth rates than those under PAR-only (μ′ = 0.17 and 0.32 d−1, respectively) and perhaps did not experience severe nitrate limitation even in the low nitrate treatment. These results suggest that the elevated UVR in low nitrate environments could result in reduction of DMSP in some species, whereas DMSP concentrations would not be affected in eutrophic areas.  相似文献

Vermicomposting is a suitable technology for processing different wastes, to produce a valuable end product (vermicompost). However, the pathogenic load of the waste must be greatly reduced in order to prevent risks to human health. Although Eisenia andrei may reduce the levels of several pathogens, the feasibility of vermicomposting, with regard to pathogen reduction, has not been tested on an industrial scale. This work studied whether vermicomposting in a continuous feeding vermireactor, is able to reduce the pathogenic load of cow manure. The effect of E. andrei on pathogens depended on the type of pathogen; thus, levels of Clostridium, total coliforms and Enterobacteria were not modified, but levels of faecal enterococci, faecal coliforms and Escherichia coli were reduced to acceptable levels. Pathogens could have maintained their levels in continuous feeding vermireactors, as fresh layers of manure are added to the top, which allows the vertical spread of pathogens through leaching.  相似文献

Vermicomposting using Lumbricus rubellus for 49 days was conducted after 21 days of pre-composting. Three different combination of treatments were prepared with eight replicates for each treatment namely cow dung: kitchen waste in 30:70 ratio (T(1)), cow dung: coffee grounds in 30:70 ratio (T(2)), and cow dung: kitchen waste: coffee grounds in 30:35:35 ratio (T(3)). The multiplication of earthworms in terms of numbers and weight were measured at the end of vermicomposting. Consequently, only T(2) showed significant increase (from it initial stage) compared to other treatments. The presence of coffee grounds in T(2) and T(3) showed higher percentage of nutrient elements in vermicompost produced. The data reveal that coffee grounds can be decomposed through vermicomposting and help to enhance the quality of vermicompost produced rather than sole use of kitchen waste in vermicomposting.  相似文献

The double-pronged problem of quantity, and disposal of waste streams from a myriad of industries, is becoming increasingly acute, the world over. The use of earthworms as a waste treatment technique for such wastes is gaining popularity. This method is commonly known as vermicomposting. Compared to conventional microbial composting, vermicomposting produces a product that is more or less homogenous, with desirable aesthetics, with reduced levels of contaminants and tends to hold more nutrients over a longer period, without impacting the environment. Like in other related waste treatment techniques, certain parameters need to be established for the design of efficient and economical vermicomposting systems. Specifically, the focus of this study was to investigate and establish an optimal stocking density and an optimal feeding rate for the vermicomposting of biosolids, with paper mulch provided as bedding. A stocking density of 1.60 kg-worms/m2 (0.33 lb-worms/ft2) and a feeding rate of 1.25 kg-feed/kg-worm/day resulted in the highest bioconversion of the substrate into earthworm biomass. The best vermicompost was obtained at the same stocking density and a feeding rate of 0.75 kg-feed/kg-worm/day.  相似文献

Coffee pulp is the main solid residue from the wet processing of coffee berries. Due to presence of anti-physiological and anti-nutritional factors, coffee pulp is not considered as adequate substrate for bioconversion process by coffee farmers. Recent stringent measures by Pollution Control authorities, made it mandatory to treat all the solid and liquid waste emanating from the coffee farms. A study was conducted to evaluate the efficiency of an exotic (Eudrilus eugeniae) and a native earthworm (Perionyx ceylanesis) from coffee farm for decomposition of coffee pulp into valuable vermicompost. Exotic earthworms were found to degrade the coffee pulp faster (112 days) as compared to the native worms (165 days) and the vermicomposting efficiency (77.9%) and vermicompost yield (389 kg) were found to significantly higher with native worms. The multiplication rate of earthworms (280%) and worm yield (3.78 kg) recorded significantly higher with the exotic earthworms. The percentage of nitrogen, phosphorous, potassium, calcium and magnesium in vermicompost was found to increase while C:N ratio, pH and total organic carbon declined as a function of the vermicomposting. The plant nutrients, nitrogen (80.6%), phosphorus (292%) and potassium (550%) content found to increase significantly in the vermicompost produced using native earthworms as compared to the initial values, while the calcium (85.7%) and magnesium (210%) content found to increase significantly in compost produced utilizing exotic worms. Vermicompost and vermicasts from native earthworms recorded significantly higher functional microbial group’s population as compared to the exotic worms. The study reveals that coffee pulp can be very well used as substrate for vermicomposting using exotic (Eudrilus eugeniae) and native earthworm (Perionyx ceylanesis).  相似文献

Induction of stress ethylene production in the plant system is one of the consequences of salt stress which apart from being toxic to the plant also inhibits mycorrhizal colonization and rhizobial nodulation by oxidative damage. Tolerance to salinity in pea plants was assessed by reducing stress ethylene levels through ACC deaminase-containing rhizobacteria Arthrobacter protophormiae (SA3) and promoting plant growth through improved colonization of beneficial microbes like Rhizobium leguminosarum (R) and Glomus mosseae (G). The experiment comprised of treatments with combinations of SA3, G, and R under varying levels of salinity. The drop in plant biomass associated with salinity stress was significantly lesser in SA3 treated plants compared to non-treated plants. The triple interaction of SA3 + G + R performed synergistically to induce protective mechanism against salt stress and showed a new perspective of plant-microorganism interaction. This tripartite collaboration increased plant weight by 53%, reduced proline content, lipid peroxidation and increased pigment content under 200 mM salt condition. We detected that decreased ACC oxidase (ACO) activity induced by SA3 and reduced ACC synthase (ACS) activity in AMF (an observation not reported earlier as per our knowledge) inoculated plants simultaneously reduced the ACC content by 60% (responsible for generation of stress ethylene) in SA3 + G + R treated plants as compared to uninoculated control plants under 200 mM salt treatment. The results indicated that ACC deaminase-containing SA3 brought a putative protection mechanism (decrease in ACC content) under salt stress, apart from alleviating ethylene-induced damage, by enhancing nodulation and AMF colonization in the plants resulting in improved nutrient uptake and plant growth.  相似文献
