Liver tumors, ranging from benign nodules to carcinomas, developed spontaneously in 115 (87%) of 132 germfree Wistar rats beyond the age of 30 months. In addition, the rats developed a high incidence of benign adenomas of endocrine glands and of the breasts. 相似文献

Although Slc:Wistar rats are used widely in biomedical research as outbred rats, close
similarities in growth curves, survival rates, and immunological and biochemical
phenotypes have been reported between Slc:Wistar and F344 inbred rats. We reported
previously that nine genetic variations that were fixed in Slc:Wistar rats had identical
genotypes in F344 rats. Here, we examined the genetic characteristics of Slc:Wistar rats
using 27 simple-sequence length polymorphism (SSLP) markers and compared them with other
Wistar stocks available in Japan and with some F344 strains. Among 27 SSLP loci, 23 (85%)
were fixed in the Slc:Wistar rats, which was the highest among the other Wistar stocks.
The 23 fixed loci shared identical genotypes with corresponding loci in F344 rats.
Further, the predominant allele types in the unfixed loci had allele frequencies as high
as 80%, and these alleles were identical in the F344 rats. When the nine genetic
variations reported previously are added, a total of 32 (89%) out of the 36 loci examined
were fixed and identical in the Slc:Wistar and F344 rat genomes. These findings indicate
the low genetic variation in Slc:Wistar rats and the high genetic similarity between the
Slc:Wistar and F344 inbred rats. This study demonstrates the importance of characterizing
outbred rats and the need to pay ample attention to the genetic characteristics the
Slc:Wistar rats for their proper use. 相似文献

F344/Slc and F344/JCL rats 2 years of age were histologically examined for the incidence and distribution of calcification. In the male rats of both strains, calcification was observed in the testis, lung, brain, kidney, heart, aorta, cornea, prostate and seminal vesicle respectively. In the female F344/JCL rats, calcification appeared in the kidney, lung, cornea, brain, stomach, ovary and heart. Among these of both sexes, the lung was one of the most affected organs for calcification. On the other hand, calcification in the kidney was more severe and frequent in the females than in the males, suggesting that the sex may be one of enhancing factors for calcification. 相似文献

Spontaneous behavior (locomotion, feeding, drinking, and exploration in a two box apparatus) as well as conditioned behavior (passive and active avoidance responding, and freezing in the light-dark box apparatus) were studied in naive male Wistar and Long Evans rats. Concerning spontaneous behavior, Long Evans rats were more active during both light and dark periods, and showed better exploratory performance than Wistar rats. Concerning conditioned behavior, Long Evans rats acquired and retained better active and passive avoidance responses, and exhibited longer initial freezing than Wistar rats in the range of 0.6-1.4 mA footshocks. The results better define the important behavioral differences existing between the two strains, Long Evans rats showing consistently a higher level of alertness and a better conditioned performance. 相似文献

The rat is commonly employed in otological research, but spontaneous ear infections can confound the results of experimental procedures--wasting time, money, and animals. The authors focus on the incidence of spontaneous otitis media in Wistar rats. They compare disease incidence in animals housed in standard cages with those housed in barrier units, showing that 20% of their conventionally housed animals developed spontaneous otitis media, whereas only 5% of their animals housed in isolated units were infected. These results underscore the importance of strict control of the shipping, housing conditions, and manipulation of animals to be used in otological research. 相似文献

Neoplastic lesions were observed in untreated aged Sprague Dawley (SD) rats throughout their lifespan starting at 5 weeks. Their mean survival times were 89 to 105 weeks of age. The total tumor incidences were 70 to 76.7% and 87 to 95.8% in males and females, respectively. The common neoplasmas were pituitary adenoma and adrenal pheochromocytoma in both sexes, testicular Leydig cell tumor in males and mammary gland tumors, thyroidal C-cell adenoma and uterine stromal polyp in females. 相似文献

The present investigations were performed to assess the differences among rat colonies commonly used for neurophysiological research regarding the development of complete Freund's adjuvant (CFA)-induced arthritis. Inflammatory signs including edema in the paw fluctuated remarkably among individual Wistar (Slc/Wistar/ST) and Sprague-Dawley (Slc/SD) rats, while the inflammatory signs of Lewis (LEW/Crj) rats appeared earlier and was severer and more consistent than Slc/Wistar/ST and Slc/SD rats. Edema in the hind paw developed in 100% of LEW/Crj rats with the lowest dose of CFA (0.6 mg/rat) used as compared with 64% of Slc/Wistar/ST (CFA 1 mg/rat) and 38% of Slc/SD rats (CFA 1.2 mg/rat). Retardation of weight gain was observed in Slc/Wistar/ST and Slc/SD rats in contrast to a severe weight decrease in inflamed LEW/Crj rats after the development of arthritis. 相似文献

Incidence of spontaneous tumors was examined in 77 male and 73 female SD-JCL rats reared throughout their lifespan of 5 to 36 months. Total tumor incidences were 60% and 95% for males and females, respectively. The sex difference was mostly attributable to a high incidence of mammary tumor in females. The pituitary tumor was found in 30% of males and 66% of females aged 5 to 36 months while islet cell tumor of the pancreas and thyroid tumor occurred later and the incidence of latter was particularly high in rats lived more than 2 years. Focal epithelial hyperplasia and metaplasia were observed in the intralobular ducts of the sublingual salivary glands of 302 out of 1142 Wistar rats. These rats (aged 16-145 weeks) served as controls in toxicological experiments. The ductal changes varied from small groups of irregular cylindrical cells with basophilic cytoplasm and vesicular nuclei, with often prominent nucleoli, to large areas of non-keratinized stratified squamous epithelium with marked mitotic activity and necrotic superficial cells being released in the lumen. Mononuclear inflammatory cells were often present in fibrous tissue surrounding altered ducts. Within certain limits the degree and incidence of the changes increased with increasing age; their aetiology is unknown. 相似文献

Neural lobe function in male rats of the Wistar/Tw strain was studied at 3, 7 and 16-18 months of age. A significant rise in the serum arginine vasopressin (AVP) level was noted in 16-18-month-old rats showing polydipsia and polyuria. The content and concentration of AVP in the neural lobe of aged rats were significantly less than those of younger animals (3 and 7 months). These results point out an enhancement of AVP release from the neural lobe of aged rats. The reduction in urinary volume in aged rats subjected to 24 hours of water deprivation was less than those in younger animals. No increase in urinary sodium, potassium and chloride concentrations was observed in aged rats, and the decrease in electrolyte excretion from urine during the dehydration period was less in aged rats than younger ones. These results suggest that the antidiuretic response to osmotic stimuli was reduced in aged rats. The administration of AVP to aged rats resulted in a significant decrease in water intake and urinary volume, but AVP administration did not induce any change in the electrolyte balance. Therefore, it is concluded that the main cause of the development of polydipsia and polyuria is the decline in renal function but not in neurosecretory activity, although exogenous AVP can effectively reduce water intake and urinary output in aged rats. 相似文献

The kynurenine pathway of tryptophan catabolism plays an important role in several biological systems affected by aging. We quantified tryptophan and its metabolites kynurenine (KYN), kynurenine acid (KYNA), picolinic acid (PIC) and quinolinic acid (QUIN), and activity of the kynurenine pathway enzymes indoleamine 2,3-dioxygenase (IDO), tryptophan 2,3-dioxygenase (TDO) and quinolinic acid phosphoribosyltransferase (QPRTase), in the brain, liver and kidney of young, middle-aged and old female Wistar rats. Tryptophan levels and TDO activity decreased in all tissues with age. In contrast, brain IDO activity increased with age, while liver and kidney IDO activity decreased with age. The levels of KYN, KYNA, QUIN and PIC in brain all increased with age, while the levels of KYN in the liver and kidney showed a tendency to decrease. The levels of KYNA in the liver did not change, but the levels of KYNA in the kidney increased. The levels of PIC and QUIN increased significantly in the liver but showed a tendency to decrease in the kidney. QPRTase activity in both brain and liver decreased with age but was elevated in the kidney in middle-aged (12-month-old) rats. These age-associated changes in tryptophan metabolism have the potential to impact upon major biological processes, including lymphocyte function, pyridine (NAD(P)(H)) synthesis and N-methyl-d-aspartate (NMDA)-mediated synaptic transmission, and may therefore contribute to several degenerative changes of the elderly. 相似文献

Although accumulating evidence has shown crucial roles of ghrelin and insulin in food intake and energy metabolism, the exact relationship between these hormones remains unclear. In this study, we determined the in vivo effect of ghrelin on insulin secretion. We demonstrated that ghrelin inhibited the glucose-stimulated release of insulin when infused into the portal vein of Wistar rats. However, ghrelin infusion into the femoral vein did not induce such an inhibitory effect. Hepatic vagotomy or coinfusion with atropine methyl bromide diminished the inhibitory effect of ghrelin on glucose-stimulated insulin secretion. In conclusion, ghrelin exerts an inhibitory effect on glucose-stimulated insulin secretion via the hepatic portal system and the vagus nerve. The decrease in ghrelin level after a meal is important for the occurrence of the incretin effect in rats. 相似文献

目的了解Wistar大鼠心脏自发性病变发病情况,为长期致癌性研究、老年病学研究及毒性病理学提供背景资料。方法采用160只清洁级Wistar大鼠,雌雄各半,常规饲养,分别在9月龄、12月龄、18月龄、24月龄时处死40只大鼠,HE及Masson三色法染色,观察心脏的病理改变。结果 9月龄Wistar大鼠心脏未见明显病理改变;12月龄Wistar大鼠月龄心脏病变的发病率为2.5%(1/40),表现为少数心肌细胞变性坏死伴少量以单核细胞为主的炎细胞浸润;18月龄大鼠心脏病变的发病率为57.5%(23/40),表现为轻至中度心肌病,雄性发病率高于雌性。24月龄大鼠100%(40/40)出现不同程度的心肌病,并有2.5%(1/40)发生心内膜下纤维组织增生。Masson染色显示9月龄大鼠心脏血管周围及心脏瓣膜环下有少量胶原纤维,随年龄增长,血管周围及心脏瓣膜环下胶原纤维逐渐增多,并延伸入心肌细胞间。结论随年龄增长,大鼠心脏自发病变比率升高,主要病变为心肌病,偶尔可发生心内膜下纤维组织增生;胶原纤维沉积首先发生于血管周围及心脏瓣膜环下,随年龄增长而增多,可能与大鼠心肌病的的发生密切相关。 相似文献

A high incidence (8/892) of urinary tract calculi was encountered during a study involving rats being fed diets supplemented with retinoids. When the incidence was compared to that observed in earlier studies without retinoid supplements and spanning several years (2/2998), the incidence in the current study was significantly higher. Possible causative factors such as age, sex, strain, diet and carcinogen or retinoid treatment have been analyzed without a clear explanation for the increased incidence. The data suggest that retinoid treatment increased the risk of stone formation, but that retinoids were less important than some other unidentified factor(s) that operated during the recent period. 相似文献

Kidneys from 1806 Wistar rats were examined grossly for hydronephrosis and ureteral dilation. Hydronephrosis was seen more often on the right side (11%) than the left (0.3%). Structural characteristics (survival, growth, connections) have been studied in the transplant of the cerebral cortex tissue in Wistar rat embryos (18-day-old), implanted into the brain of mature rats of the same line at various time after a partial lesion of the sensomotor cortex. In 3-5 months after transplantation the light microscopy methods demonstrate that spatial interconnections of the transplant and the injured brain of the recipient depend on time interval between the cerebral lesion and transplantation of the embryonal nervous tissue. Horseradish peroxidase (HP) is ionophoretically injected into the recipient's cerebral tissue away from the place of transplantation. In the transplant retrogradely labelled HP neurons are revealed. This demonstrates efferent connections of the implanted tissue with the host's brain. Presence of the anterogradely labelled nervous terminals in the transplant tissue demonstrates existence of afferent connections of the transplant with the recipient's tissue. Possible mechanisms of survival, growth and formation of connections of the transplant in the injured brain of the mature animal are discussed. 相似文献
