The oxygen consumption of Nothobranchius korthausae eggs in different developmental stages, including diapause II and III, was measured. Oxygen consumption increases exponentially during embryonic development. In diapause II and III there is a drop in oxygen consumption, which attains a minimal level in diapause II after 3 weeks and in diapause III after 2 weeks. During early development the embryos can escape from hypoxic stress by entering diapause I and II. During late embryogenesis embryos in diapause III can escape from hypoxic stress by hatching. We conclude that survival of annual fish embryos is enhanced during conditions of low oxygen concentration by reduced oxygen consumption rates during diapause.  相似文献

Analysis over the first 48 h of development in vitro from the one-cell stage to the early four-cell stage indicated that (i) ethylenediaminetetraacetic acid (EDTA) exerts the major beneficial effect on culture to the blastocyst stage of F1 and MF1 embryos, (ii) glutamine assists development of MF1, but not F1, embryos to the blastocyst stage and probably functions as part of a metabolic response to oxidative damage to mitochondria and (iii) exposure to glucose at some time during early cleavage is essential for full development to blastocysts. None of the culture conditions examined affected significantly the increase in concentration of reactive oxygen species in late two-cell embryos in vitro, although F1 embryos in vitro often had lower peroxide concentrations than MF1 embryos. A decline in oxygen tension from 20 to 50% had no consistent effect on culture to the blastocyst stage or production of reactive oxygen species. Aminooxyacetate, an inhibitor of transaminase activity, prevented non-blocking embryos from developing beyond G2 of the second cell cycle. It is concluded that the chelation of transitional metals provides the most effective method of overcoming the block to development in vitro.  相似文献

We studied properties of lactate dehydrogenase isozymes expressed at different stages of Danio rerio ontogenesis. H4-LDH and a minor fraction H3M1 are expressed during embryonic development. The muscle isozyme (M4) appears after the beginning of muscle contractions in the embryo. H4 and M4 isozymes isolated from the heart and skeletal muscle of the adult fish, respectively, show significant differences in terms of Km, activation energy (AE), and inactivation temperature. H4-LDH isozymes isolated from unfertilized eggs, the skeletal muscle of larvae, and the heart of the adult fish differ in Km and activation energy, as well as in inactivation temperature. We propose that these differences may be associated with a ligand interacting with the H4 isozyme at different steps of ontogenesis.  相似文献

It is shown that the level of information richness of the environment in which the juvenile fish were raised at early stages of ontogenesis is one of the determining factors for the development of the most important adaptive patterns of behavior. Minimal values of fullness index, lower body weight, and shorter length, along with a feeding spectrum strongly differing from that in natural conditions, are characteristics of juvenile fish kept preliminarily in the hatchery. The food of this group of fish is dominated by organisms of epibenthos, which may have been detected by the fish using gustatory and tactile receptors. At the same time, the proportion of infaunal invertebrates, the detection of which is only possible using olfaction and electric reception, is minimal. The prolonged development of juvenile fish under conditions of sensory deprivations leads to inadequate behavioral skills, making the process of conventionally reflective “switching” upon the release of such juvenile fish in the wild more difficult.  相似文献

In previous studies of nuclear transplantation, most cloned animals were obtained by intraspecies nuclear transfer and are phenotypically identical to their nuclear donors; furthermore, there was no further report on successful fish cloning since the report of cloned zebrafish. Here we report the production of seven cross-genus cloned fish by transferring nuclei from transgenic common carp into enucleated eggs of goldfish. Nuclear genomes of the cloned fish were exclusively derived from the nuclear donor species, common carp, whereas the mitochondrial DNA from the donor carp gradually disappeared during the development of nuclear transfer (NT) embryos. The somite development process and somite number of nuclear transplants were consistent with the recipient species, goldfish, rather than the nuclear donor species, common carp. This resulted in a long-lasting effect on the vertebral numbers of the cloned fish, which belonged to the range of goldfish. These demonstrate that fish egg cytoplasm not only can support the development driven by transplanted nuclei from a distantly related species at the genus scale but also can modulate development of the nuclear transplants.  相似文献

The spawning areas and early development of long spiky-head carp, Luciobrama macrocephalus (Lacépède), an endemic fish species in China, were investigated in the Yangtze River and Pearl River of central and southeastern China between 1961 and 1993. The potamodromous fish migrated upstream to spawn between May and July as the floodwater began to rise. The water-hardened eggs drifted down the river, and the embryos and larvae developed in the course of drifting. The spawning areas of the fish were widely found in the upper and middle main channels and large tributaries. Two large dams (Gezhouba dam and Danjiangkou dam) did not significantly impact on the reproduction of the fish. Fifty stages of the early development from one cell to the juvenile with fully formed fins were observed and characterized pictorially. The larvae of long spiky-head carp could be distinguished from the larvae of other co-occurring species by counting the number of somites and comparing the proportion of sizes of eye to otic capsule.  相似文献

In poikilothermic animals body temperature varies with environmental temperature, and this results in a change in metabolic activity (Q10 of enzymatic reactions typically is around 2-3). Temperature changes also modify gas transport in body fluids. While the diffusion coefficient increases with increasing temperatures, physical solubility and also hemoglobin oxygen affinity decrease. Therefore, an increase in temperature typically requires adjustments in cardiac activity because ventilatory and convectional transport of respiratory gases usually are tightly coupled in adults in order to meet the oxygen demand of body tissues. Hypoxic conditions also provoke adaptations in the central circulatory system, like the hypoxic bradycardia, which has been described for many adult lower vertebrates, combined with an increase in stroke volume and peripheral resistance. In embryos and larvae the situation is much more complicated, because nervous control of the heart is established only late during development, and because the site of gas exchange changes from mainly cutaneous gas exchange during early development to mainly pulmonary or branchial gas exchange in late stages. In addition, recent studies in amphibian and fish embryos and larvae reveal, that at least in very early stages convectional gas transport of the hemoglobin is not essential, which means that in these early stages ventilatory and convectional gas transport are not yet coupled. Accordingly, in early stages of fish and amphibians the central cardiac system often does not respond to hypoxia, although in some species behavioral adaptations indicate that oxygen sensors are functional. If a depression of cardiac activity is observed, it most likely is a direct effect of oxygen deficiency on the cardiac myocytes. Regulated cardiovascular responses to hypoxia appear only in late stages and are similar to those found in adult species.  相似文献

用抑制性差减杂交结合SMART cDNA合成和RACE—PCR技术克隆到雌核发育银鲫(Carassius auratus gibelio)肌酸激酶M3-CK基因的全长cDNA。银鲫M3-CK cDNA全长1551bp,编码380个氨基酸,与普通鲤鱼(cyprinus carpio)M3-CK的氨基酸序列同源性高达95%。种系分析表明,银鲫M3-CK与其它脊椎动物的肌肉型肌酸激酶聚为较近的一支,与鲤鱼的M3-CK聚在一起,与脑特异型肌酸激酶及线粒体型肌酸激酶分歧较大。虚拟Northern杂交显示银鲫M3-CK基因在胚胎发育中差异表达。RT—PCR表明,银鲫M3-CK基因在成熟卵母细胞和胚胎发育早期可检测到少量的转录产物,在胚胎发育期间从肌肉效应期开始转录,并一直持续表达。组织RT—PCR表明,银鲫M3-CK基因只在心脏和肌肉表达。  相似文献

用RACE-PCR方法从原肠期SMART文库中扩增到银鲫pou2基因的全长cDNA,其全长为2421bp,开放阅读框为1416bp,编码471个氨基酸,与斑马鱼pou2基因的氨基酸序列一致性高达91.0%。我们用RT-PCR和整体原位杂交的方法研究了银鲫pou2基因在胚胎发育过程中的时空表达图式。RT-PCR结果显示,银鲫pou2基因有母源转录本,其合子基因在高囊胚期强烈表达,在50%下包期和90%下包期也有高量的转录本,但在100%下包期表达量急剧降低,至体节期时已经完全检测不到其转录本。胚胎整体原位杂交结果显示其母源转录本在所有的胚盘细胞中。在高囊胚期和50%下包期,高度表达的合子转录本仍在所有的胚盘细胞中,但至90%下包期时,pou2的表达向胚胎背部的正中线汇聚,集中在神经板的两侧区域和脑部的两条横向条带。在100%下包期时,pou2的表达集中在神经板的中间区域以及预期形成的中后脑区域。至体节期时,转录本消失,这与RT-PCR结果高度一致。银鲫pou2基因的表达图式提示该基因在胚胎发育的早期具有重要作用,它可能参与调控神经板的形成和中后脑细胞的发育命运。  相似文献

Changes in the water quality and temperature relationships of ponds may affect the structure of amphibian assemblages. The survival, time to hatching, hatching size and hatching stage of newt embryos were studied in the three British species ( Triturus cristatus, T. helveticus and T. vulgaris ), at two temperatures and two pHs. All T. cristatus embryos failed to hatch at pH 4.5, whereas over 80% of T. helveticus and T. vulgaris embryos hatched successfully at the same pH. At pH 7.5, T. cristatus survival was the same as the other two species, after the 50% mortality due to the homomorphism of chromosome 1 was taken into account. Temperature had no effect on survival of embryos. Time to hatching was two to four times longer at 12°C than at 17°C. Low pH shortened development time in T. vulgaris but not in T. helveticus . Low pH, but not temperature, affected size at hatching, with T. helveticus and T. vulgaris embryos emerging at a smaller size and earlier stage of development under acidic conditions. This reduction of size at low pH affected T. vulgaris more than T. helveticus . We predict that T. cristatus embryos will be the most vulnerable of the three species to acidification in nature. Warm ponds will result in rapid embryonic development, but T. helveticus and T. vulgaris larvae hatching in acid ponds will do so at a smaller size and earlier stage of development. The pattern of vulnerability to acidification within amphibian assemblages may change during ontogeny.  相似文献

We studied growth and respiration rate during early ontogenesis of the axolotl Ambystoma mexicanum, Bosca's newt Triturus waltlii, the green toad Bufo viridis, and the smooth clawed frog Xenopus laevis. The respiration rate in these amphibian species increases during embryonal and larval development, peaks after transition to active feeding, and decreases at later stages of ontogenesis. The patterns of dynamics of this energy metabolism index in tailed and tailless amphibians have some differences related to their specific development. The changes in respiration rate in the embryos and larvae are correlated with the concentrations of mitochondria.  相似文献

High performance liquid chromatography (HPLC) was used to assess the uptake dynamics of the cryoprotectant DMSO by intact unfertilized eggs (stage 0), 8-cell (stage 5) and eyed embryos (stage 30) of medaka, Oryzias latipes, the relation of the internal concentration (Cin) of DMSO with fertilization and survival rates, and the effects of several factors on these processes. The factors examined were: cryoprotectant concentration (0.6, 1.2, 1.9 and 2.5 M), impregnation time (1, 3, 5, 10, 15 and 20 min), temperature (0, 5 and 20 degrees C), hydrostatic pressure (0 and 50 atm), and the osmotic conditions of the materials (normal or partially dehydrated). Cryoprotectant permeation, estimated from the initial rates of DMSO uptake, was higher in embryos than in eggs and increased with embryonic development; however, the DMSO Cin in eyed embryos reached a plateau at 1-5 min and could not be increased by prolonging impregnation. The highest fertilization and survival rates for any given DMSO Cin were obtained with high concentrations and short times of impregnation rather than low concentrations and long impregnation times. Application of hydrostatic pressure (50 atm) and exposure for 3 min to a 1 M trehalose solution prior to impregnation induced a substantial increase in the DMSO Cin of 8-cell embryos in comparison to untreated controls with no significant effect on survival. Hydrostatic pressure also promoted DMSO uptake in unfertilized eggs, but with rapid loss of viability, and was ineffective in eyed embryos. The uptake of DMSO and its toxicity to 8-cell embryos were directly proportional to the temperature of impregnation. The results of this study reveal important interactions between cryoprotectant concentration, impregnation time and the developmental stage (or type) of the materials and provide evidence that hydrostatic pressure, temperature of impregnation and the osmotic conditions of the materials can be manipulated to increase the uptake of cryoprotectant by fish eggs and embryos.  相似文献

Investigations of the erythrocyte count, haemoglobin content and M index of 7 fish species were performed under equivalent environmental conditions in the same period of the year. The species were: bighead Hypophthalmichthys nobilis (Richardson), carp Cyprinus carpio L., silver carp Hypophthalmichthys molitrix (Val.), grass carp Ctenopharyngodon idella (Val.), sheat-fish Silurus glanis L. , pike Esox Indus L., pike-perch Lucioperca lucioperca (L.). Bighead and carp are carnivorous and omnivorous, silver carp and grass carp are herbivorous and sheat-fish, pike and pike-perch are predatory. Significant differences were demonstrable between M values of fish living on different type of food, where the M index is: It is concluded that M index values are useful as one parameter differentiating between fish with distinctly different food habits.  相似文献

Exposure of embryos of Chondrostoma nasus to a strongly reduced oxygen content (about 10% of air saturation) enhanced mortality and depressed hatching success. Viable larvae hatched only from the controls and from the group exposed to reduced oxygen during the earliest embryogenesis (from fertilization to gastrula). In the groups exposed from gastrula to eyed stage and from eyed stage to hatching, a high percentage of fish hatched, but all were deformed and almost all died during the following 1–5 days. Hatching period was prolonged in the groups exposed to reduced oxygen content. At a spawning area C. nasus had deposited a large portion of eggs in areas of reduced oxygen content. On the other hand, eggs placed at the substratum surface were exposed to high predation pressure from Barbus barbus, Lota lota and ducks ( Anas platyrhynchos ). The trade-off between predation pressure at the substratum surface and reduced oxygen conditions in the interstices resulted in a narrow space for successful development of embryos of this endangered rheophilic fish species under natural conditions.  相似文献

泥鳅和白鲢胚胎发育时的耗氧量是随着发育进程而以指数函数方式在增长,其耗氧曲线显示出卵裂期、原肠期、神经胚期、肌肉效应期以及出膜期是5个呼吸代谢水平较高的时期,其中又以原肠期与出膜期对孵化环境溶氧的需求尤为突出。胚胎在发育过程中纳入氧的快慢与强弱,都直接或间接地反映出新陈代谢的速度以及营养物质不断地被氧化、消耗和能量传递的状况。因此,鱼类胚胎发育时耗氧的高低,无疑是其胚胎发育状况的一种生理指标。  相似文献

An analysis of carp blastoderm development was carried out in culture after isolation from the yolk cell and its yolk syncytial layer (YSL). The blastoderms were separated from the YSL at four different stages of embryogenesis: the blastula, early epiboly, early gastrula and late gastrula stages. Absence of the YSL in explants was checked by scanning electron microscopy. From observations of living embryos and histological examination of tissues which were formed in explants from all stages studied it was observed that they contained notochordal, muscle and neural tissue as signs of dorsal types of differentiation. Only in explants from the early and late gastrula stages were histotypical tissues organized in an embryonic-like body pattern. The data indicate that mesoderm differentiation in fish embryos is independent from the YSL, contrary to normal pattern formation which needs the presence of the YSL before the onset of gastrulation.  相似文献

A mechanistic model of an energy budget in fish embryos and yolk-sac larvae was developed using data for five freshwater fish species: rainbow trout Oncorhynchus mykiss , nase Chondrostoma nasus , carp Cyprinus carpio , tench Tinca tinca and African catfish Clarias gariepinus , based on the existing models for adult and juvenile fishes. The model simulates changes in the components of the budget under various conditions. Besides the effects of body mass and temperature on consumption and metabolic rate, the dependence of ration size on amount of available yolk and initial egg size was implemented in the model. The model parameters were found through optimization. A sensitivity analysis of the model was conducted by varying its parameters and observing changes in the output. A comparative analysis showed that the values generated by the model closely approximated independent empirical observations. Simulations of energy budgets demonstrated that the overall pattern of energy partitioning was the same for different species, irrespective of egg size and temperature preferences. Most energy was allocated to body growth. Clarias gariepinus showed the fastest growth and had the highest yolk conversion efficiency.  相似文献

Developmental expression of CagMdkb during gibel carp embryogenesis   总被引:3,自引:0,他引:3  
Midkine (Mdk) genes have been revealed to have different expression patterns in vertebrates and therefore, additional studies on Mdk expression patterns are required in more species. In this study, CagMdkb has been cloned and characterized from a SMART cDNA library of 10-somite stage embryos of Carassius auratus gibelio. Its full length cDNA is 1091 bp and encodes a sequence of 147 amino acids, which shows 97.3% identity to zebrafish Mdkb on the amino acid level. RT-PCR analysis reveals that CagMdkb is first transcribed in gastrula embryos and maintains a relatively stable expression level during subsequent embryogenesis. Western blot analysis reveals a 19 kDa maternal CagMdkb protein band and the zygotic CagMdkb protein is expressed from gastrula stage. At around 10 somite stage, the 19 kDa CagMdkb is processed to another protein band of about 17 kDa, which might be the secreted form with the 21-residue signal peptide removed. With immunofluorescence analysis, maternal CagMdkb protein was found to be localized in each blastamere cell of early embryos. The zygotic CagMdkb positive fluorescence signal was detected from a pair of large neurons at 18-somite stage. At the later stages, CagMdkb protein was also extended to numerous small neurons in the forebrain, midbrain and hindbrain, as well as to nerve fibers in the spinal cord. Co-localization with 3A10 antibody revealed CagMdkb immunoreactivity on developing Mauthner neurons, a member of reticulospinal neurons. In addition, ectopic expression of CagMdkb in early embryos of gibel carp and zebrafish suppressed head formation and CagMdkb function was found to depend on secretory activity. All these findings indicate that CagMdkb plays an important role in neural development during gibel carp embryogenesis and there is functional conservation of Mdkb in fish head formation.  相似文献
