The induction of synthesis of the secreted enzymes endo-1,4--xylanase (EC 3.2.1.8) and -galactosidase (EC 3.2.1.23) in original and recombinant Penicillium canescens strains has been studied. In all producer strains, the synthesis of these enzymes was induced by arabinose and its metabolite arabitol. The two enzymes differed in the concentration of arabinose required for induction: the synthesis of -galactosidase was most pronounced at 1 mM, whereas maximum synthesis of endo-1,4--xylanase was observed at 5–10 mM. An increase in the number of endo-1,4--xylanase copies in the high-copy-number strain of the fungus suppressed the synthesis of -galactosidase; the synthesis of endo-1,4--xylanase in the high-copy-number recombinant producing -galactosidase was affected to a lesser extent. The amount of enzymes synthesized did not depend on the saccharide used as the sole source of carbon for growing the mycelium prior to its transfer to the inducer-containing medium.  相似文献

Summary Microfluorimetrically recorded fluorescence bands of vitally stained nuclei and protoplasts from the inner epidermis of yellow onion(Allium cepa) scales are compared with fluorescence bands of Tarions model solutions. The strong fluorescent staining of the nucleus and the weaker hue of the cytoplasm after application of acridine orange is mainly due to the accumulation of monomeric dye cations in polar cytoplasmic lipids. Vital fluorescent staining with neutral red shows similar effects, but in addition an accumulation of the dye base in apolar lipids can be ascertained without doubt. On the other hand, the major accumulation of the acid fluorochrome uranin (sodium fluorescein) does not take place in polar cytoplasmic lipids but, apparently, rather in the form of anions in the water phase of the ground cytoplasm and inner nuclear plasm owing to a mechanism of plasmatic ion trap.The proof that dye ions are present in vitally stained protoplasm suggests the possibility, that the familiar phenomenon of vacuole contraction may depend on a Donnan effect in the case of basic dyes and might be a consequence of the raising of osmotic values by dye anions in the case of acid dyes.Experiments with three phases (dye solution-oil-blood plasma) yield fluorescent stainings of blood plasma with acridine orange, neutral red and uranin, which may be compared to vital staining of nuclei and cytoplasm. The well-known properties of blood plasma-lipids suggest a similar function of cytoplasmic lipids, viz. as a vehicle of lipid transport in a mainly aquatic, molecular-disperse phase. A diagram (Fig. 15) illustrates the cooperation between sheetlike lipoproteid complexes (boundary layers) and lipoproteid complexes of the ground cytoplasm dispersed as particles.  相似文献

Summary Fluorescence spectra of leukocytes stained by both mithramycin and DAPI showed that the fluorescence of the two dyes can be separated efficiently by using different excitation wavelengths, for instance the 435 nm and the 365 nm mercury lines. In human chromosomes the complementary (reverse) banding pattern produced by these dyes may thus be observed on double stained chromosome spreads. In plants, for instance in Anemone blanda, the two dyes may reveal two different banding patterns. The results of absorption and fluorescence measurements suggest the existence of at least two binding sites, or types, for each dye, with different fluorescent yields and binding strengths.  相似文献

Biology cannot accommodate all aspects of culture. Aspects of culture that a biological approach can take into account can be covered by the biological categories of phenotype and environment. There is no need to treat culture as a separate category. Attempts to elaborate biological explanations of cultural variation will meet with success only if biologists expand theories of development, and integrate them in evolutionary biology. The alternative — elaborating the idea of so-called cultural inheritance — makes little sense from a biological point of view.  相似文献

Zusammenfassung In dieser ersten von insgesamt drei Studien über die Beziehungen zwischen Substantia granulofilamentosa und Krinom wurden am Blut Phenylhydrazin-vergifteter Meerschweinchen 57 kationische Farbstoffe verschiedener chemischer Struktur und ausgewählte basische Stoffe ohne Farbstoffcharakter auf ihre Eignung geprüft, die Substantia granulofilamentosa auszufällen und gegebenenfalls hinsichtlich der Art der Ausfällung individuelle, vor allem morphologische Merkmale erkennen zu lassen.Von den 57 Farbstoffen waren 36, vorwiegend die hauptsächlich untersuchten Derivate von Heterocyclen des Anthracens, zur Darstellung der Substantia granulofilamentosa geeignet. Zu den erstmals für die Rc-Darstellung als brauchbar erkannten Farbstoffen gehören Methylengrün, Toluylenblau, Pseudoisocyanin, Neutralviolett und Amethystviolett.Auch in einigen Alkaloiden in hoher Konzentration, insbesondere mit Chinin, gelingt die Präzipitation einer netzförmigen, basophilen Struktur, die allerdings nicht die gleiche Dichte und Farbstoffaffinität aufweist, wie die von guten Farbstoffen ausgefällte Substantia granulofilamentosa.Bei intensiv mit Phenylhydrazin behandelten Meerschweinchen wurde Spontanentmischung der basophilen Substanz der Rc bereits in den Kontrollpräparaten beobachtet.Die Mehrzahl der positiven Farbstoffe gehört ihrer chemischen Struktur nach zu den Heterocyclen des Anthracens. — Ursachen negativer Resultate der Rc-Markierung werden besprochen.Quantitative Beziehungen zwischen Änderungen der Farbstoffkonzentration und den Rc-Zahlen wurden vergleichend am Beispiel äquimolarer Lösungen von Acridinorange, 9-Aminoacridin, Neutralrot und Chinin untersucht. In dieser Reihenfolge geht die Wirksamkeit der vier Substanzen bei abnehmender Konzentration zurück.Nach Einwirkung konzentrierter (1100) geeigneter Farbstoffe auf Blutzellen zeigt die Struktur der in Rc ausgefällten Substantia granulofilamentosa gewisse farbstoffabhängige Eigenarten.Einige dieser Eigentümlichkeiten in der Darstellung der Substantia granulofilamentosa werden beschrieben. Typisch z. B. für Derivate des Acridins ist das Bild der dichten zentralen Zusammenballung der basophilen Substanz im May-Grünwald-Giemsa-nachgefärbten Präparat.Mit einigen positiven Farbstoffen (Konzentration 12000) wird in Rc das Phänomen der Vakuolenbildung, welches direkte Beziehungen zur Krinombildung hat, hervorgerufen. Die meisten Farbstoffe werden bei dieser Konzentration in Vakuolen konzentriert. Pyronin ist nur zur Darstellung der Substantia granulofilamentosa geeignet.In Froscherythrocyten vermögen Farbstoffe der Gentianaviolett-Gruppe die Substantia granulofilamentosa nicht auszufällen.Aus Vergleichsgründen wird die gleiche Farbstoffauswahl auch hinsichtlich ihrer Affinität zu den chemisch andersartig aufgebauten Heinz-Körpern untersucht. Die Heinz-Körper werden besonders intensiv mit Farbstoffen der Gentianaviolett-Gruppe und mit Oxazin angefärbt. Sie zeigen ziemlich intensiv Primärfluoreszenz, nach Einwirkung verschiedener Fluorochrome Sekundärfluoreszenz.Gesichtspunkte der Beziehungen zwischen chemischer Struktur der untersuchten Farbstoffe und ihren Wirkungen auf Rc werden besprochen.
Summary This is the first of three studies on the relationship between Substantia granulofilamentosa and crinoma. Using the blood of phenylhydrazine-poisened guinea pigs, 57 cationic dyes of various chemical structures and selected basic substances of nondye character, are tested for their suitability to precipitate the substantia granulofilamentosa. Attention is paid to the various types of precipitates in order to find out whether they show individual morphologic features. Out of 57 dyes, 36—most of which are derivatives of heterocyclic anthracene — are suitable for the demonstration of the substantia granulofilamentosa. The group of dyes, which for the time ever have been used successfully for the demonstration of reticulocytes, includes: methylene green, toluylene blue, pseudo-isocyanine, neutral violet, and amethyst violet.After treatment with certain alkaloids in high concentrations (especially quinine) a reticular basophil structure is precipitated. It shows, however, not the same density and affinity for dyes as the precipitates, obtained with good dyes. In guinea pigs, treated with high doses of phenylhydrazine, a spontaneous segregation of the basophil substance of the reticulocytes is already observed in the controls. According to their chemical structures, the majority of posiive dyes belong to the heterocyclic anthracenes. — The reasons for negative results in the demonstration of reticulocytes are discussed. Quantitative relationships between dye concentration and number of reticulocytes are tested with equimolar solutions of acridine-orange, 9-amino acridine, neutral red, and quinine. The efficacy of these four substances decreases with decreasing concentration in the order given above. If blood cells are treated with a concentrated solution (1100) of suitable dyes, certain dye-dependent peculiarities are observed in the precipitated substantia granulofilamentosa, some of which are described in detail. A typical picture after treatment with acridine derivatives is — for example — a dense central conglomerate of the basophil substance, found after additional staining with May-Grünwald-Giemsa. Some positive dyes (concentration 12000) cause the formation of vacuoles in reticulocytes, which stands in direct relation to the formation of the crinoma. If used at this concentration most of the dyes are concentrated in vacuoles. Pyronine is suitable only for the demonstration of the substantia granulofilamentosa.Dyes that belong to the gentian violet group, are not suitable for the precipitation of the substantia granulofilamentosa in frog erythrocytes.For comparative reasons the same group of dyes is tested for its affinity to the Heinz bodies, which chemically are very different from the substantia granulofilamentosa. Heinz bodies are intensely stained with dyes belonging to the gentian violet group and by oxazine dyes. A rather intensive primary fluorescence is observed in Heinz bodies; if several fluorochromes are used, they show secondary fluorescence. The relation between the chemical structure of the dyes used in this study and their effect on reticulocytes is discussed.

Preincubation of lymphocytes with des-Tyr1--endorphin (DTE) inhibits the reaction between some HLA alloantisera and their corresponding antigens. One HLA-A2-specific antiserum was found which could detect a subtype of the HLA-A2 antigen on DTE-treated lymphocytes from some donors. Comparison with the HLA-A2 subtypes as defined by a combination of cytotoxic T lymphocyte typing and biochemistry showed a complete correlation with the previously described HLA-A2.3 subtype.  相似文献

We describe a novel compound ciliary structure (g-cilium) from the food groove of the lobate ctenophore Mnemiopsis mccradyi. G-cilia are small, flat compound ciliary organelles that are oriented with their tips pointing toward the mouth. Typically three to four rows of g-cilia line the inner surface of the tentacular groove, which together with the transport groove, make up the food groove. G-cilium cells are 11.4 m long and 4.2 m wide at the g-cilium base. The g-cilium itself is 3.4 m long and tapers to a flat, sharp tip. G-cilia are not motile but are surrounded by many hundreds of smaller, actively motile cilia that beat with orally-directed effective strokes. G-cilia contain 50 conventional `9+2' cilia embedded in a fibrous core that arises from the cell body. In addition, g-cilia contain mitochondria, thousands of small membrane-bounded vesicles and rod bacteria. G-cilia basal bodies are anchored by large, strongly-banded rootlets that extend approximately the entire length of the cell. G-cilia may have organizational, sensory and/or secretory function within the feeding apparatus. Their placement strongly suggests that they play critical roles in feeding. They may enhance the efficiency of prey capture and so contribute to M. mccradyi's well-known voracious appetite. By enhancing prey capture they probably play a critical role in the capacity of this organism to follow prey dynamics, so contributing to dense blooms in mid-late summer in coastal regions.  相似文献

The usefulness of 4,4-diisothiocyanatostilbene-2,2-disulfonic acid (DIDS) for in-situ studies of the chloroplast phosphate translocator was evaluated by fluorescence microscopy and radiolabeling of spinach (Spinacia oleracea L.) (C3 plant) and maize (Zea mays L.) (C4 plant) chloroplasts. In maize mesophyll and bundle-sheath chloroplasts and in spinach chloroplasts that were either intact, broken or swollen, DIDS fluorescence was only associated with the chloroplast envelope. Intact chloroplasts often had fluorescent patches corresponding to concave regions of the chloroplast which we assume to be regions enriched in DIDS-binding sites.Incubation of intact or broken spinach chloroplasts or maize mesophyll chloroplasts with [3H2]DIDS resulted in the labeling of a single polypeptide (relative molecular mass, Mr, 30 kDa) in the envelope fraction, in each case. Label in the stromal fraction was not detected when intact chloroplasts were incubated with [3H2]DIDS. However, when broken chloroplasts were incubated with [3H2]DIDS, several polypeptides of various molecular masses were labeled, but not the 30×31-kDa polypeptide. In thylakoid fractions from both broken and intact chloroplasts, a single 30×31-kDa polypeptide was labeled inconsistently. There are important genetic influences on the tendency to dizygotic (DZ) twinning and it is a plausible hypothesis that these reside in one or more of the genes coding for the major reproductive hormones. We used Southern analysis of DNA from 50 young (<32) mothers of DZ twins, who also had a family history of DZ twinning, and 50 controls, to examine allele frequencies of five restriction fragment length polymorphisms (RFLPs) in four hormone genes coding for follicle stimulating hormone (FSH), chorionic gonadotropin (CGB), inhibin B and gonadotropin releasing hormone (GnRH). Comparison of allele frequencies revealed no significant differences between DZ twin mothers and controls. However this does not rule out the role of these genes in the hereditary tendency of multiple ovulation in humans, since absence of linkage disequilibrium does not imply absence of linkage.  相似文献

Summary In biomedical studies, dyes are divided into acid and basic dyes. This classification cannot be reconciled with current chemical definitions of acids and bases. Brönsted-Lowry acids are compounds that can donate protons; bases are proton acceptors. The definition of acids and bases is independent of the electric charge, i.e. acids and bases can be neutral, anionic or cationic. Reactions between acids and bases result in formation of new acid-base pairs. Lewis acids and bases do not depend on a particular element, but are characterized by their electronic configurations. Lewis bases are electron donors; Lewis acids are electron acceptors. This classification is also unrelated to the electric charge. Lewis acids and bases interact by formation of coordinate covalent bonds.In histochemistry and histology, dyes containing SO 3 , –COO and/or –O groups are classified as acid dyes. However, such compounds are electron pair donors and hence Brönsted-Lowry and Lewis anionic bases. Dyes carrying a positive charge are termed basic dyes. Chemically, many cationic dyes are Lewis acids because they can add a base, e.g. OH, acetate, halides. The hypothesis that transformation of –NH2 into ammonium groups imparts basic properties to dyes is untenable; ammonium groups are proton donors and hence acids. Furthermore, conversion of an amino into an ammonium group blocks a lone electron pair and the color of the dye changes drastically, e.g. from violet to green and yellow. It appears therefore highly unlikely that ammonium groups are responsible for binding of cationic (basic) dyes. In histochemistry, it is usually not of critical importance whether anionic or cationic dyes are chemically acids or bases. It is therefore suggested to substitute the terms anionic for acid and cationic for basic dyes; this nomenclature will always be chemically correct.  相似文献

Cerebellar granule cells (CGC) at different stages of maturation in vitro (1 or 6 DIV), were treated with 25–35 and acetyl-L-carnitine arginine amide (ST857) in presence of 25 mM KC1 in the culture medium, and neuronal viability was assessed. Three days of treatment slightly modified the survival of 1 DIV-treated cells, which degenerate and die five days later -amyloid matching. Similarly, a significative neurotoxic effect was observed on 6 DIV treated-cells after 5 days of exposure to the peptide, while the death occurred within 8 days. ST857 coincubated with 25–35 was able to rescue neurons from 25–35-induced neurotoxicity. We also studied the changes in Ca2+ homeostasis following glutamate stimulation, in control and -amyloid treated single cells, either in presence or in absence of ST857. 25–35 did not affect basal [Ca2+]i, while modified glutamate-induced [Ca2+]i increase, causing a sustained plateau phase of [Ca2+]i, that persisted after the removal of the agonist. ST857 pretreatment completely reverted this effect suggesting that, in CGC chronically treated with 25–35, ST857 could protect the cells by neurotoxic insults of the peptide likely interfering with the cellular mechanisms involved in the control of Ca2+ homeostasis.  相似文献

From 100 g sunflower seeds, 1.2 mg purified -galactosidase was obtained with an overall yield of 51%. The -galactosidase acted on both terminal -galactosyl residues and side-chain -galactosyl residues of the galactomanno-oligosaccharides and galactomannans. The cDNA coding for sunflower -galactosidase was cloned and the deduced amino acid sequence revealed that the mature enzyme consisted of 363 amino acid residues with a molecular weight of 40263. Seven cysteine residues were found but no putative N-glycosylation sites were present in the sequence. The deduced amino acid sequences of mature enzyme and -galactosidases from coffee, guar and Mortierella vinacea -galactosidase II showed over 81%, 77%, and 47% homology, respectively. These enzymes are classified into the third group in which the enzyme has no insertion sequence and a broad specificity on galactomanno-oligosaccharides compared to the other groups.  相似文献

Synopsis A fluorescence metachromatic modification of the masked basophilia method is described. It is based on the acridine dye Coriphosphine O. Excitation and emission spectra of green (orthochromatic) and red (metachromatic) fluorescent tissue components are presented.When the method is applied to suitably fixed sections, metachromasia is demonstrable in cells of the polypeptide hormone-secreting APUD series, and in a few other situations.The view that side-chain carboxyl groups are demonstrated by the masked basophilia technique is considered to be accurate but inadequate. It is proposed that the technique, and its fluorescence modification, are influenced more by secondary than by primary protein structure. In particular, it is suggested that the conformation of the protein precursors of polypeptide hormones, in the storage granules of endocrine cells, is predominantly random-coil.  相似文献

Zusammenfassung In 2 Versuchsserien wurden Kohlmeisen(Parus major) und japanische Möwchen(Lonchura striata var.domestica) einzeln und schallisoliert gehalten. In der ersten Versuchsserie, in der alle Vögel einen dunklen Schlafkasten hatten, wurde der Einfluß der Beleuchtungsstärke auf die Periode () der Hüpfaktivität und auf das Verhältnis von Aktivitätszeit zu Ruhezeit ( : -Verhältnis) untersucht. Sowhol Kohlmeisen als auch japanische Möwchen folgen der Regel, daß mit wachsender Beleuchtungsstärke die Periode kürzer und das : -Verhältnis größer wird.In der 2. Serie wurde der Einfluß Ruhe im dunklen Schlafkasten auf die Periodenlänge und auf das : -Verhältnis untersucht. Es wurden die Messungen aus Bedingung 1 (der Vogel hat einen dunklen Schlafkasten zur Verfügung) mit den Messungen aus Bedingung 2 (der Vogel hat keinen oder einen hellen Schlafkasten zur Verfügung) verglichen. Das Ergebnis bei Kohlmeisen entspricht den Befunden bei konstantem Licht verschiedener Intensität. Unter Bedingung 1 ist länger und das : -Verhältnis kleiner als in Bedingung 2. Das Ausmaß der Änderung von nach Fortnahme des dunklen Kastens ist unabhängig von der Periodenlänge in Bedingung 1. Das Ausmaß der änderung von : ist unabhängig von a : in Bedingung 1, jedoch schwach negativ korreliert mit der Periodenlänge in Bedingung 1.Bei japanischen Möwchen entsprechen die Ergebnisse dieser Versuchsserie nicht der Regel für tagaktive Vögel. Mit Benützen des dunklen Schlafkastens ist kürzer als ohne den Schlafkasten. Ohne den Schlafkasten ist etwa 24 Std. Das : -Verhältnis ist in Bedingung 1 unter bestimmten Voraussetzungen kleiner als in Bedingung 2. Das Ausmaß der Änderung von nach Fortnahme des Kastens ist mit der dazugehörigen Periode in Bedingung 1 hochsignifikant korreliert (Regressionskoeffizient b=-1.01, Korrelationskoeffizient r=0.89). Ebenfalls ist das Ausmaß der Änderung von : nach Fortnahme des Kastens mit : aus Bedingung 1 korreliert; es scheint, als würde ein bevorzugtes : -Verhältnis von etwa 2.0 eingeregelt.Die Ergebnisse werden im Hinblick auf 4 Punkte diskutiert: 1) Das circadiane System arbeitet innerhalb eines engen Bereiches von - und : -Werten optimal. 2) Der Optimalbereich wird bevorzugt unter ungünstigen Bedingungen angestrebt. 3) Der Entzug des dunklen Schlafkastens belastet japanische Möwchen mehr als Kohlmeisen. 4) Bei japanischen Möwchen wird in Bedingung 1 durch fortplfanzungsphysiologischen Einfluß verkürzt.
Circadian activity rhythms of birds with and without a dark nest box
Summary Perch-hopping activity of Great tits(Parus major) and Bengales finches(Lonchura striata domestica), housed individually in soundproof boxes, was studied in two series of experiments. In the first series all birds had access to a dark nest box, in which they retired during their subjective night. In this experiment the effect of light intensity on the freerunning circadian activity rhythm was investigated. Both Great tits and Bengalese finches obey the circadian rule by responding to an increase in light intensity with shortening the circadian period () and with an increase of the ratio of activity time and rest time ( : ).In the second series of experiments the influence of sleeping in the dark nest box on both circadian period and : -ratio was studied. The results of two experimental conditions — without and with access to a dark nest box — were compared. In the Great tits, the results are in agreement with the effect of light intensity: when a dark nestbox is available, is longer and the : -ratio is smaller than in the absence of a nest box. The magnitude of the change in free-running period after removal of the nest box is independent of the original value of ; the amount of change : -ratio is likewise independent of the original : -ratio, but is weakly correlated to the original .InLonchura striata var. domestica, removal of the dark nest box leads to a lenghtening of the free-running period to about 24 hours; the : -ratio is smaller in the presence of a dark nestbox, if certain other conditions are fulfilled. The magnitude of the change in after removal of the nest box is highly correlated to the original free-running period (r=-0.89) in such a way that, without nest box, the period approaches a value of 24 hours. Also, the amount of change in : -ratio due to nest box removal is negatively correlated to the original : -ratio. A probably preferred : -ratio of 2.0 is adopted.These results are discussed in the view of 4 points: 1) The circadian system operates at its optimum within a narrow range of - and : -values. 2) This optimal range is especially adopted when conditions become adverse. 3) Removal of the dark nest box results in a more stressful situation for Bengalese finches than for Great tits. 4) In the Bengalese finches, is shortened in the presence of a nest box due to effects on reproductive physiology.

Summary We have measured the potential-dependent light absorption changes of 43 impermeant oxonol dyes with an oxidized cholesterol bilayer lipid membrane system. The size of the signal is strongly dependent on the chain length of alkyl groups attached to the chromophore. Dye molecules with intermediate chain lengths give the largest signals. To better understand the dependence of the absorbance signal on alkyl chain length, a simple equilibrium thermodynamic analysis has been derived. The analysis uses the free energy of dye binding to the membrane and the on-off model (E.B. George et al.,J. Membrane Biol.,103:245–253, 1988a) for the potential-sensing mechanism. In this model, a population of dye molecules in nonpolar membrane binding sites is in a potential-dependent equilibrium with a second population of dye that resides in an unstirred layer adjacent to the membrane. Dye in the unstirred layer is in a separate equilibrium with dye in the bulk bathing solution. The equilibrium binding theory predicts a sigmoidally shaped increase in signal with increasing alkyl chain length, even for very nonpolar dyes. We suggest that aggregation of the more hydrophobic dyes in the membrane bathing solution may be responsible for their low signals, which are not predicted by the theory.  相似文献

Within a 30-ha permanent plot of a tropical evergreen forest in the Anamalais, Indian Western Ghats, all the 13415 trees 30cmgbh (belonging to 153 species) were identified and tagged during 1997–1998. This communication reports the results of tree population changes based on two annual censuses (1999 and 2000) of the plot after the initial census, and compares the results with other tropical forest sites. One species, Ficus beddomei, disappeared from the plot and there was no new addition of species. Of the 96 species that showed changes in their population density, for 12 species recruitment and mortality matched. Tree recruitment (5 treesha–1year–1) exceeded mortality (4 treesha–1year–1) during the two-year period. Four modes of tree deaths were recorded in our site. The per cent mortality of trees differed between tree size classes during each recensus. The plot tree density increased progressively in the two-year period, indicating that shifts in species composition and density in natural forests, without major catastrophic disturbance, occur slowly.  相似文献

Mannans of the yeast Saccharomyces cerevisiae have been implicated as containing the allergens to which bakers and brewers are sensitive and also the antigen recognized by patients with Crohn's disease. A fraction of S. cerevisiae mannan, Sc500, having high affinity for antibodies in Crohn's patients has been characterized by NMR spectroscopy followed by fragmentation using alkaline elimination, partial acid hydrolysis and acetolysis. The released oligosaccharides were separated by gel filtration on a Biogel P4 column and analyzed by fluorescence labeling, HPLC and methylation analysis. The relationship between structure and antigen activity was measured by competitive ELISA. The antigenic activity of the original high molecular weight mannan could be ascribed to terminal Man13Man12 sequences which are rarely found in human glycoproteins but were over-represented in Sc500 compared to other yeast mannans.  相似文献

Embryo suspensor masses (ESMs) were induced by culture of isolated mature zygotic embryos of balsam fir [Abies balsamea (Mill.)] on media containing 10 M cytokinin [6-(--dimethylallylamino)purine (2iP), 6-benzyladenine (BA), or thidiazuron (TDZ)]. Once induced, ESMs proliferated on media containing 2iP, BA or TDZ (10 M) or on 4.5 M BA in combination with 10 M naphthyl-1-acetic acid. When ESMs were transferred to media containing 5–80 M abscisic acid, cotyledonary-stage embryos were formed. The rate of accumulation of conjugated dienes of polyunsaturated fatty acids was measured during free-radical oxidation of linoleic acid (18:2n-6, LA), -linolenic acid (18:3n-3, -LNA), and -linolenic acid (18:3n-6, -LNA) initiated by 2,2"-azo-bis-(2-amidinopropane) hydrochloride in aqueous micellar solutions of sodium dodecyl sulfate and sodium cholate. It was shown that, unlike homogeneous solutions, the oxidative stability of PUFAs in aqueous dispersions increased with an increase in the extent of unsaturation. The rate of LA oxidation was more than tenfold greater than that of - and -LNA. The antioxidant activity of -carotene, in contrast to homogeneous solutions, in both micellar systems studied depended on the degree of PUFA unsaturation. We found that 5 M -carotene effectively inhibited the LA oxidation (almost by 90%), whereas the oxidation of -LNA and -LNA was not inhibited by -carotene even at much greater concentration (30 M). The paradoxical discrepancy between the extent of unsaturation and the PUFA oxidation rate, as well as a decrease in the efficiency of -carotene-dependent inhibition of oxidation of more polyunsaturated fatty acids in reactions conducted in aqueous dispersions is consistent with the model according to which the peroxyl radicals of LA and fatty acids with the doublebond number greater than two exhibit different polarity.  相似文献

Vesicular fragments of sarcoplasmic reticulum (SR) were spin labelled with 2,2,6,6-tetramethyl, 4-isothiocyanate piperidine-1-oxyl (probe A) and 2,2,6,6-tetramethyl, 4-amino (N-iodoacetamide) piperidine-1-oxyl (probe B). Two to five moles of probe A or B were covalently bound to 106g of membrane protein, with minimal loss of activity (ATPase, Ca2+, uptake). The EPR spectra of labelled SR were then studied in various experimental conditions.Strongly acid or alkaline pH, protein denaturation with ura, and membrane solubilization with deoxycholate produced marked alterations of the EPR spectra of spin-labelled SR, indicating changes in the local environment surrounding the probes, and the occurrence of conformational changes.A reversible modification of the EPR spectra of probe A and an accelerated efflux of accumulated Ca2+ were produced by increasing the temperature of SR suspensions from 30° to 40° C. Such a parallel behavior indicates that reversible structural transitions may control membrane permeability and Ca2+ efflux.ATP modifies the EPR spectra of probe B, suggesting that ATP binding to the membrane induces a structural change involving the local environment of certain sulfhydryl groups. The ATP concentration required for this effect is comparable to that requied for activation of ATPase. ADP and ITP are also effective, while pyrophosphate, AMP, and cyclic AMP are not. The effect of ATP is reversible.In other experiments, 2,2,6,6-tetramethylpiperidine-1-oxyl (probeC) was equilibrated with concentrated suspensions of SR. The EPR spectra obtained thereafter indicate that probe C binds to the membrane fragments, still maintaining a high degree of motional freedom. These spectra were markedly changed by deoxycholate solubilization of the membrane fragments, while they were little affected by protein denaturation with guanidine.
