Amino acid sequences of the two major isoforms of troponin C from crayfish   总被引:1,自引:0,他引:1  
The primary structure of the two major isoforms (alpha and gamma) of troponin C (TnC) from crayfish tail muscle has been determined by the application of manual and automated Edman degradation procedures to fragments generated by suitable chemical and proteolytic cleavages. Both amino acid sequences commence with an acetylated methionyl residue and contain 150 amino acid residues, including a single proline residue at position 29 and 2 residues of tyrosine at positions 95 and 102. No cysteine or tryptophan are present. The molecular weights calculated for alpha- and gamma-TnC are 17,157 and 16,974, respectively. The two crayfish proteins are invariable at 129 positions and conserved at 11 others. Pairwise comparisons show that the two sequences are 33-39% identical with those of seven TnCs reported so far and 39% identical with that of bovine brain calmodulin. The N-terminal end of about 10 residues, found in vertebrate TnCs, is absent in crayfish TnCs. In the latter proteins, domains I and III appear as abortive Ca2+-binding sites due to nonconservative amino acid replacements at the key Ca2+-coordinating positions in their loops. The remaining two Ca2+-binding loops (II and IV) show a remarkable similarity with the Ca2+-specific loops (I and II) found in vertebrate TnCs. These findings are consistent with the Ca2+-binding data (Wnuk, W. (1989) J. Biol. Chem. 264, 18240-18246) which indicate the presence of two Ca2+-specific sites in crayfish TnCs. These two sites display the same affinity for Ca2+ (log KCa = 4.3) on gamma-TnC but differ in their affinity (log KCa = 6.0 and 4.1) on alpha-TnC. The only structural difference between the dodecapeptide loops II and IV in both alpha- and gamma-TnC, which correlates with the existence of the high affinity (log KCa = 6.0) Ca2+-specific site on alpha-TnC, is position 11 occupied by a methionyl residue in the loop IV of alpha-TnC as opposed to negatively charged residues found in the other three loops. This suggests that the high affinity Ca2+-specific site on alpha-TnC is located in domain IV. Since the Ca2+-binding studies show that the formation of the complex of crayfish troponin I (TnI) with alpha- and gamma-TnC increases significantly the affinity of only one of their two Ca2+-specific sites and this TnI-sensitive site is not the high affinity Ca2+-specific site on alpha-TnC, we conclude that the binding of Ca2+ to site II controls the Ca2+-dependent interaction between crayfish TnCs and TnI.  相似文献

The amino terminal sequence of the 40,000 dalton polypeptide subunit of Torpedocalifornica acetylcholine receptor has been determined for twenty-five cycles using automatic microsequencing procedures. The results demonstrate a unique polypeptide sequence for this receptor subunit and quantitation of amino acid recoveries shows that no significant amounts of polypeptides with blocked amino terminii are present.  相似文献

The amino acid sequences of the two major isozymes of rhizopuspepsin, an aspartic proteinase from Rhizopus chinensis, were determined by analyzing the tryptic peptides derived from the reduced and carboxymethylated (RCm-) derivative of each isozyme. Amino acid substitutions were shown to occur at eight positions. Rhizopuspepsin I, with an isoelectric point of 5.1, had Ile-15, Asn-61, Ser-116, Lys-162, Ile-230, Tyr-241, Asp-293, and Glu-325, whereas rhizopuspepsin II, with an isoelectric point of 5.8, had Val-15, Lys-61, Asn-116, Ser-162, Val-230, Ser-241, Asn-293, and Gln-325, the other parts of the two isozymes being identical with each other. Thus, rhizopuspepsin I had two more net negative charges than rhizopuspepsin II. This is consistent with the difference in isoelectric point of these two isozymes.  相似文献

By means of reverse-phase HPLC, 2 different proteins were obtained from apparently purified pig eosinophil major basic protein (MBP) and these proteins were named GMPB1 and GMBP2. It was revealed that these 2 components of MBP have similar molecular weights and pI values, although the amino acid compositions were slightly different. In the previous study, we cloned and sequenced GMPB1 cDNA. Here we obtained another clone by plaque hybridization using a screening probe synthesized by means of polymerase chain reaction. After sequencing, it became apparent that this clone corresponded to GMBP2. As in the case of GMBP1, the cDNA of GMBP2 encoded pre-proGMBP2 with 3 domains; signal peptide, acidic pro-portion, and mature GMBP2. By comparing the sequences of GMBP1 and GMBP2, it was revealed that the proteins were quite similar to each other. In addition, their sequences also resembled those of human MBP, especially in the basic domain of mature protein; but no such similarity existed in the pro-portion. Although the molecular weights determined by SDS-PAGE of guinea pig and human MBPs were 11,000 and 9,300, respectively, the calculated molecular weights of these 3 MBPs were all 13.8 kDa. The calculated pI values of GMBP1, GMBP2 and human MBP were 11.7, 11.3 and 11.6, respectively. By means of Harr plot analysis, it was revealed that the amino acid sequences, not only in signal peptides but also in the basic domains of mature proteins, were well conserved between guinea pig and human MBPs.  相似文献

The N-terminal amino acid sequences of the hemopexins purified from the plasma of rat, rabbit and chicken were compared with each other and with that of human hemopexin. Although the N-terminal sequences differ among these species, residues 2, 3 and 14 are identical in all four hemopexins. Ten of the first 28 residues are identical in all but the chicken protein. When introducing gaps into the sequence, a much greater homology is observed between the human and rat or rabbit hemopexins (60%) than when the sequences were compared directly (40%).  相似文献

1. The nature of the subunits in rabbit muscle triose phosphate isomerase has been investigated. 2. Amino acid analyses show that there are five cysteine residues and two methionine residues/subunit. 3. The amino acid sequences around the cysteine residues have been determined; these account for about 75 residues. 4. Cleavage at the methionine residues with cyanogen bromide gave three fragments. 5. These results show that the subunits correspond to polypeptide chains, containing about 230 amino acid residues. The chains in triose phosphate isomerase seem to be shorter than those of other glycolytic enzymes.  相似文献

The amino acid sequences of fragments from light meromyosin and heavy meromyosin subfragment-2 have been analysed and structural features noted. As with other α-fibrous protein sequences, there is a regular disposition of apolar residues in positions a and d of the heptapeptide-type repeat characteristic of the coiled-coil conformation. The common occurrence of acidic and basic residues in the e and g positions, respectively, give rise to a maximum number of interchain ionic interactions when the two parallel chains of myosin are in axial register. Although the quasi-repeating heptapeptides in the sequences both have two points of discontinuity (unlike that in most other α-fibrous proteins), secondary structure prediction methods indicate that the fragments will be 90 to 100% α-helical. Fast Fourier transform techniques have revealed a significant periodicity of about 27.4 ± 0.3 residues (~41 Å) in the linear disposition of the acidic residues and the basic residues in both of the fragments. This period is compatible with similarly directed myosin molecules in the thick filament being axially staggered with respect to one another by an odd multiple of 143 Å. Preliminary evidence is also presented to show that the sequence of the rod region of myosin may have a 28 residue gene duplication repeat.  相似文献

alpha-1-Antitrypsin is found in hepatocytes as a high-mannose glycoprotein (Mr 49 000), extracellularly as a complex-type glycoprotein (Mr 54 000). Deglycosylation of both forms with peptide: N-glycosidase led to proteins of identical app. Mr (41 000). The sequence of 26 N-terminal amino acids of rat alpha 1-antitrypsin was determined. A high content of polar amino acids was found. The partially characterized presequence of in vitro synthesized alpha 1-antitrypsin showed a cluster of hydrophobic amino acids. A pre-peptide of 24 amino acids is proposed. There is no evidence for the existence of a propeptide.  相似文献

Forty-five amino acids at the amino terminus of macromomycin, a protein antitumor antibiotic, have the following sequence: Ala-Pro-Gly-Val-Thr-Val-Thr-Pro-Ala-Thr-Gly-Leu-Ser- Asn-Gly-Glu-Thr-Val-Thr-Val-Ser-Ala-Thr-Gly-Leu-Thr- Pro-Gly-Thr-Val-Tyr-His-Gly-Glu-Ser-Ala-Val-Ala-Glu- Pro-Gly-Val-Ile-Gly-Pro- ..... Of the first 31 residues in this sequence 16 are homologous with the amino terminal sequence in neocarzinostatin, a protein antitumor antibiotic which also degrades DNA. The amino acid sequence of the N-terminal 139 residues of the L (light) chain derived from a homogeneous rabbit antibody to type III pneumococci was determined. This L chain, designated BS-5, exhibits a greater degree of homology with the basic sequence of human kappa chains of subgroup I (72%) than with subgroups II and III. L-chain BS-5 differs from another L chain (BS-1), also derived from an antibody to type III pneumococci (Jaton, 1974), by eight amino acid residues, even though the chains are identical within the N-terminal 30 residues. Six of these eight substitutions are located within the three hypervariable sections of the variable half: Asn/Ser in position 31, Glu/Ala in position 55, Asx/Thr, Thr/Gly, Thr/Gly and Val/Tyr in positions 92, 94, 96 and 97 respectively. The two anti-pneumococcal L chains BS-1 and BS-5 are much more similar to each other than to an anti-azobenzoate L chain (Appella et al., 1973), from which they differ by 30 and 29 residues respectively. Of these interchanges 13-15 are confined to the three hypervariable sections, and 11 occur within the N-terminal 27 positions. The three chains have an identical sequence from residue 98 to residue 139, except for a possible inversion of two residues in positions 130-131 of the anti-azobenzoate chain.  相似文献
