The effects of nitrogen (N) supply restriction on the CO2 assimilation and photosystem 2 (PS2) function of flag leaves were compared between two contrastive Japanese rice cultivars, a low-yield cultivar released one century ago, cv. Shirobeniya (SRB), and a recently improved high-yield cultivar, cv. Akenohoshi (AKN). Both cultivars were solution-cultured at four N supply levels from N4 (control) to N1 (the lowest). With a reduction in N-supply, contents of N (LNC), ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO), and chlorophyll (Chl) in flag leaves decreased in both cultivars. In parallel with this, the net photosynthetic rate (P N), mesophyll conductance (g m), and stomatal conductance (g s) decreased. P N was more dominantly restricted by g m than g s. The values of P N, g m, and RuBPCO content were larger in AKN than SRB at the four N supply levels. The content of Chl greatly decreased with N deficiency, but the reduction in the maximum quantum yield of PS2 was relatively small. Quantum yield of PS2 (ΦPS2) decreased with N deficiency, and its significant cultivar difference was observed between the two cultivars at N1: a high value was found in AKN. The content ratio of Chl/RuBPCO was also significantly low in AKN. The low Chl/RuBPCO is one of the reasons why AKN maintained a comparatively high P N and ΦPS2 at N deficiency. The adequate ratio of N distribution between Chl and RuBPCO is the important prerequisite for the efficient and sustainable photosynthesis in a flag leaf of rice plant under low N-input.  相似文献

Diurnal changes of photosynthesis in the leaves of grapevine (Vitis vinifera × V. labrusca) cultivars Campbell Early and Kyoho grown in the field were compared with respect to gas exchanges and actual quantum yield of photosystem 2 (ΦPS2) in late May. Net photosynthetic rate (PN) of the two cultivars rapidly increased in the morning, saturated at photosynthetic photon flux density (PPFD) from 1200 to 1500 μmol m−2 s−1 between 10:00 and 12:00 and slowly decreased after midday. Maximum PN was 13.7 and 12.5 μmol m−2 s−1 in Campbell Early and Kyoho, respectively. The stomatal conductance (gs) and transpiration rate changed in parallel with PN, indicating that PN was greatly affected by gs. However, the decrease in PN after midday under saturating PPFD was also associated with the observed depression of ΦPS2 at high PPFD. The substantial increase in the leaf to air vapour pressure deficit after midday might also contribute to decline of gs and PN.  相似文献

Response of two spring wheat (Triticum aestivum L.) cultivars, salt tolerant SARC-I and salt sensitive Potohar, to different concentrations of NaCl was examined under glasshouse conditions. Eighteen-day-old plants of both the lines grown in sand culture were irrigated with 0 (control), 80, 160 or 240 mM NaCl in full strength Hoagland's nutrient solution. Shoot fresh and dry masses, and leaf area per plant of SARC-I at the vegetative stage, were significantly greater than those of cv. Potohar at higher salt concentrations, however, relative growth rate (RGR) of cv. Potohar was significantly higher than that of SARC-I. SARC-I had higher net photosynthetic rate (PN), stomatal conductance (gs) and transpiration rate (E) than cv. Potohar at the vegetative stage, but the cultivars did not differ significantly in water-use efficiency (PN/E), intrinsic water use efficiency (PN/gs), and intercellular/ambient CO2 concentration ratio. At the grain development stage, SARC-I had significantly higher PN and gs in the flag leaf than cv. Potohar under salinity. SARC-I was superior to cv. Potohar with respect to number of grains per spike, number of grains per spikelet, mean grain mass, and grain yield per plant at all NaCl concentrations.  相似文献

The effects of iso-osmotic salinity and drought stresses on leaf net photosynthetic rate (P N) in two wheat (Triticum aestivum L.) cultivars BR 8 and Norin 61, differing in drought tolerance, were compared. In drought-sensitive Norin 61, the decline of P N was larger than that in drought-tolerant BR 8. Under NaCl treatment, P N decreased in two phases similarly in both cultivars. In the first phase, photosynthetic depression was gradual without any photochemical changes. In the second phase, photosynthetic depression was rapid and accompanied with a decline of the energy conversion efficiency in photosystem 2 (PS2). Our observations suggest that the osmotic factor may induce a gradual depression of photosynthesis due to stomatal closure under both stress treatments. However, under NaCl treatment, a ionic factor (uptake and accumulation of excess Na+) may have direct effects on electron transport and cause more severe photosynthetic depression. The drought tolerance mechanism of BR 8 was insufficient to maintain single-leaf photosynthesis under salinity.  相似文献

We investigated net photosynthetic rate (PN) of ear and two uppermost (flag and penultimate) leaves of wheat cultivars Hongmangmai (drought resistant) and Haruhikari (drought sensitive) during post-anthesis under irrigated and non-irrigated field conditions. The PNof ear and flag leaf were significantly higher and less affected by drought in Hongmangmai than in Haruhikari. The rate of reduction in stomatal conductance (gs) was similar for the two cultivars, but intercellular CO2concentration (Ci) in the flag leaf of Hongmangmai was lower than that of Haruhikari in non-irrigated treatment. No differences were observed in leaf water potential (ψ1) and osmotic adjustment of the flag leaf of the cultivars. These results imply that differences in photosynthetic inhibition on the flag leaf at low leaf ψ1between the cultivars were primarily due to non-stomatal effects. Hence the main physiological factor associated with yield stability of Hongmangmai under drought stress may be attributed to the capacity for chloroplast activity in the flag leaf, which apparently allows sustained PNof flag leaf during grain filling under drought stress. The higher PNof ear in Hongmangmai under drought could also be related to its drought resistance.  相似文献

Kandelia candel (L.) Druce is the dominant mangrove species on the west coast of northern Taiwan. We have measured the net photosynthetic rate (P N) and chlorophyll (Chl) a fluorescence of seedlings grown at combinations of two nitrogen (0.01 and 0.1 mM) and two NaCl (250 and 430 mM NaCl) controls. With the same nitrogen level, seedlings grown at higher salinity (HS) had a significantly lower P N and stomatal conductance (g s) than those at lower salinity (LS). An increase in nitrogen availability significantly elevated P N and g s of the LS-grown seedlings. Compared to dark adapted leaves, the maximum quantum yield of photosystem 2 (PS2) (Fv/Fm) of leaves exposed to PFDs of 1200 and 1600 μmol m-2 s-1 for 2 h was significantly reduced. The degree of Fv/Fm reduction differed among leaves of the four types of treated plants. Chl fluorescence quenching analysis revealed differences among the examined plants in coefficients of non-photochemical and photochemical quenching. Photosynthetic characteristics of ear and flag leaves of wheat species, tetraploid Triticum dicoccoides Kom and hexaploid Bima1, were studied in plants grown under well-watered (WW) and water-stressed (WS) conditions. Compared to ears, flag leaves exhibited higher photosynthetic rate (P N) at the filling stage, but more severe decrease under WS. P N in the tetraploid wheat ear remained higher than that in the hexaploid wheat during the grain-filling stage. Water stress decreased PN in both the organs; this decline was caused by a reduction in Rubisco activity, not by drought-induced stomatal limitation. Tetraploid wheat ears exhibited higher relative water content and water-use efficiency than that of hexaploid wheat, under WS. The change in phosphoenolpyruvate carboxylase activity and carbon isotope composition indicated the absence of C4 metabolism in the ears of both species under both conditions. The improved performance of the tetraploid wheat ears under WS was associated with better water relations.  相似文献

In some regions of the world, low annual precipitation necessitates irrigation of crop plants which usually leads to soil salinity. Due to climatic changes this effect is also expected in the countries of Central Europe, and so in Poland. The aim of the study was (1) to compare tolerance to salt stress of Polish Triticum aestivum cvs. ‘Bogatka’ and ‘Banderola’ with T. durum cv. ‘Komnata’ and breeding line 121, and (2) to indicate the physiological parameter/parameters most suitable for such comparison. The investigation was performed in two experiments. In the first one, the germination ability of caryopses and coleoptiles’ growth were estimated at 0–250 mM of NaCl. The second experiment was conducted on plants grown in a glasshouse in saline soil at 0–150 mM of NaCl for 6 weeks. Salt tolerance was evaluated on the basis of following parameters: chlorophyll fluorescence, net photosynthesis rate (P N), transpiration rate (E), stomatal conductance (g s), cell membrane permeability (EL), proline content, fresh weight (FW), dry weight (DW), and relative water content (RWC). Highest germination of caryopses of durum cultivars was recorded at all the salinity levels; however, their coleoptiles were shorter than coleoptiles of bread wheat cultivars. Analysis of chlorophyll fluorescence showed that applied salt doses did not disturb the light phase of photosynthesis in all cultivars under study. Plants of durum wheat showed the higher dissipation of energy excess at the level of the antenna chlorophyll (DIo/CSm) under salinity as compared to plants of bread wheat. Both ‘Komnata’ and line 121 showed stronger P N reduction as an effect of salinity. A decline of P N was closely connected with a decrease in g s. The P N correlated with a decrease in DW in all studied cultivars except ‘Bogatka’. Control plants of ‘Komnata’ and line 121 were characterized by higher EL and proline level than bread wheat cultivars. An increasing cell membrane permeability correlated with a decrease of RWC in ‘Banderola’ and ‘Komnata’. The content of proline under the increasing salinity correlated with changes of RWC in ‘Banderola’, ‘Komnata’ and line 121, which indicate protectoral role of proline against dehydration of tissue. Dry weight and RWC seem to be the parameters most useful in the salt-tolerance estimation of wheat plants. Taking into account the studied parameters ‘Banderola’ could be recognized as more salt tolerant, the degree of salinity tolerance of ‘Bogatka’ is the same as line 121, while ‘Komnata’ seems to be the most salt sensitive. Leaf gas exchange, plant growth and leaf ion content were measured in wheat (Triticum durum L. cv. HD 4502) exposed to steady- state salinities (1.6, 12.0 and 16.0 dS nr−1) for 8 weeks. Salinity reduced leaf area and number of tillers, and increased Na+ and Cl concentrations in leaves. Leaf- to- leaf gradients of these ions were observed. The oldest leaf contained 6 to 8 times more Na+ and Cl than the flag leaf. Net photosynthetic rate (PN), transpiration rate (E) and stomatal conductance (gS) were the highest in flag leaf, declined in the middle and fully expanded leaves, and were minimum in the oldest leaves. These processes were reduced by salinity with similar leaf- to- leaf gradients. Intercellular CO2 concentrations in the older leaves were higher than in the flag leaf in non-saline plants, and increased similarly with salinity. Leaf age was the major factor in reducing PN, and senescence processes were promoted by salinity.  相似文献

Seedlings of spring barley, meadow fescue, and winter rape were fumigated with 180 μg kg−1 of ozone for 12 d, and effect of O3 on photosynthesis and cell membrane permeability of fumigated plants was determined. Electrolyte leakage and chlorophyll fluorescence were measured after 6, 9, and 12 d of fumigation, while net photosynthetic rate (P N) and stomatal conductance (g s) were measured 9 d after the start of ozone exposure. O3 treatment did not change membrane permeability in fescue and barley leaves, while in rape a significant decrease in ion leakage was noted within the whole experiment. O3 did not change the photochemical efficiency of photosystem 2 (PS2), i.e., Fv/Fm, and the initial fluorescence (F0). The values of half-rise time (t1/2) from F0 to maximal fluorescence (Fm) decreased in fescue and barley after 6 and 9 d of fumigation. P N decreased significantly in ozonated plants, in the three species. The greatest decrease in P N was observed in ozonated barley plants (17 % of the control). The ozone-induced decrease in P N was due to the closure of stomata. Rape was more resistant to ozone than fescue or barley. Apparently, the rape plants show a large adaptation to ozone and prevent loss of membrane integrity leading to ion leakage. Wheat (Triticum aestivum L.) cv. Jimai22 was used to evaluate the effect of ethylene evolution rate (EER) and 1-aminocyclopropane-1-carboxylic acid (ACC) and their relations with photosynthesis and photochemical efficiency in plants well-watered (WW) and under a severe water deficit (SWD). SWD caused a noticeable reduction in the grain mass. The marked increases in both EER and the ACC concentration were observed under SWD; it was reversed effectively by exogenous spermidine (Spd) or amino-ethoxyvinylglycine (AVG). Thermal images indicated that SWD increased obviously the temperature of flag leaves, mainly due to the decrease in transpiration rate under SWD. Exogenous Spd or AVG decreased to some extent the temperature of the flag leaves. The strong decline in photosynthetic rate (P N) and stomatal conductance as well as the photodamage of PSII were also observed under SWD after 14 and 21 days after anthesis (DAA). Intercellular CO2 concentration was reduced at 7 DAA, but slightly increased at 14 and 21 DAA under SWD, indicating that the decreased P N at 7 DAA might result from stomatal limitations, while the decline after 14 and 21 DAA might be attributed to nonstomatal limitations. Correlation analysis suggested that EER and ACC showed negative relations to photosynthesis and photochemical efficiency. Data obtained suggested that the effects of SWD were mediated predominantly by the increase in EER and ACC concentration, which greatly decreased the leaf photosynthesis and photochemical efficiency, and, therefore, reduced the grain mass. Application of Spd or AVG reduced the EER and ACC, and thus positively influenced photosynthesis and photochemical efficiency under SWD.  相似文献

Co2 exchange and transpiration rates of the flag leaves of four spring wheat (Triticum aestivum L.) cultivars, namely Glenlea, Neepawa, Opal and Kolibri, were compared using infra-red gas-analysis technique. The plants were grown in a controlled environment under an 18-h photoperiod, with day and night temperatures of 20 and 15° C, respectively. The time course of the CO2-exchange rate (CER) of the flag leaf differed among cultivars. CER began to decrease rapidly some 2 weeks after ear emergence in Glenlea, Neepawa and Kolibri, but only after 4 weeks in Opal. The decline in CER of Glenlea, Neepawa and Opal was continuous throughout the period of grain development whereas in Kolibri CER was maintained at a constant level between the 4th and 6th weeks after ear emergence. The transpiration rates of the flag leaves of the 4 cultivars did not change markedly until 6–7 weeks after ear emergence, indicating that the reduction in CER was not primarily a response to increased stomatal resistance to the diffusion of CO2. Removing the ear of the main shoot of intact plants failed to depress CER of the subtending flag leaf until 5 weeks after ear removal. Removing the ears of all the tillers of plants in which all but 3 tillers had been removed at ear emergence did not depress CER until 4 weeks after ear emergence, but removal of the ear of the main shoot of plants where all the tillers had been removed at ear emergence reduced the CER of the flag leaf 2 weeks after ear removal. Removal of tillers at ear emergence had a marked effect on the time course of CER and transpiration rates of the flag leaf. Both CER and transpiration rates of a 4-tiller plant were maintained at a higher level throughout ear development as compared to those of a one-tiller plant. The transpiration rate of the flag leaf of Glenlea increased during the later part of the life of the leaf even for one-tiller plants with no ear, indicating that such a stomatal response may be part of the normal course of leaf aging and not a response to a feedback stimulus from the ear.  相似文献

After 45 d of limited water supply, cassava (Manihot esculenta Crantz) exhibited pronounced reduction in shoot growth, high leaf fall, and decreased stomatal conductance. However, the water status of the remaining leaves was unaffected. This was combined with an amplified heliotropic response and drooping which minimises radiant energy interception at mid-day, suggesting that leaves are sensitive to high irradiance (I). In well-irrigated plants, CO2-saturated oxygen evolution and net photosynthetic rate (P N) in air were markedly higher (5-fold) in young (expanding) leaves than in mature leaves. Water limitation did not strongly modify CO2-saturated oxygen evolution but it altered P N in air for both types of leaves, although differently. The mature leaves of drought-adapted plants displayed residual rate of P N and deteriorated photosystem 2 (PS2) photochemistry estimated from chlorophyll (Chl) a fluorescence measurements. In young leaves at moderate I, P N was depressed by only 66 % in stressed plants. Moreover, the photochemical quenching of Chl a fluorescence and the quantum efficiency of PS2 photochemistry in young leaves were comparable in both control and stressed plants. In contrast at high I, P N was almost null and marked decreases in the two fluorescence parameters were apparent. Hence the strong heliotropic response and drooping displayed by young leaves under water limitation is an important strategy for avoiding inactivation of P N by high I and therefore for cassava tolerance to drought.  相似文献

Trehalose can reduce stomatal aperture by a hydrogen-peroxide-dependent pathway in Vicia faba L. (cv. Daqingpi) resulting in significantly lower values of net photosynthetic rate (PN), stomatal conductance (gs), and transpiration rate (E). At 8 and 24 h, the lower PN in trehalose-treated plants was accompanied by significant decrease in intercellular CO2 concentration (ci) suggesting that the reduction of PN was caused by stomatal limitation. At 48 and 72 h, trehalose decreased apparent carboxylation efficiency (PN/ci) and did not decrease ci and gs compared with controls; therefore the reduction in photosynthesis was caused by non-stomatal limitation. Trehalose treatment resulted in significantly higher effective photochemical efficiency of PS II (ΦPSII) and did not affect maximum photochemical efficiency of PS II (Fv/Fm). At 24, 48, and 72 h, trehalose decreased non-photochemical quenching (NPQ) and increased photochemical quenching (qP). Our results suggest that trehalose did not damage photosynthetic reaction centers.  相似文献

During the grain filling period we followed diurnal courses in leaf water potential (ψ1), leaf osmotic potential (ψπ), transpiration (E), leaf conductance to water vapour transfer (g) and microclimatic parameters in field-grown spring barley (Hordeum distichum L. cv. Gunnar). The barley crop was grown on a coarse textured sandy soil at low (50 kg ha−1) or high (200 kg ha−1) levels of potassium applied as KCl. The investigation was undertaken at full irrigation or under drought. Drought was imposed at the beginning of the grain filling period. Leaf conductance and rate of transpiration were higher in the flag leaf than in the leaves of lower insertion. The rate of transpiration of the awns on a dry weight basis was of similar magnitude to that of the flag leaves. On clear days the rate of transpiration of fully watered barley plants was at a high level during most part of the day. The transpiration only decreased at low light intensities. The rate of transpiration was high despite leaf water potentials falling to rather low values due to high evaporative demands. In water stressed plants transpiration decreased and midday depression of transpiration occurred. Normally, daily accumulated transpirational water loss was lower in high K leaves than in low K leaves and generally the bulk water relations of the leaves were more favourable in high K plants than in low K plants. The factorial dependency of the flag leaf conductances on leaf water potential, light intensity, leaf temperature, and leaf-to-air water vapour concentration difference (ΔW) was analysed from a set of field data. From these data, similar sets of microclimatic conditions were classified, and dependencies of leaf conductance on the various environmental parameters were ascertained. The resulting mathematical functions were combined in an empirical simulation model. The results of the model were tested against other sets of measured data. Deviations between measured and predicted leaf conductance occurred at low light intensities. In the flag leaf, water potentials below-1.6 MPa reduced the stomatal apertures and determined the upper limit of leaf conductance. In leaves of lower insertion level conductances were reduced already at higher leaf water potentials. Leaf conductance was increased hyperbolically as photosynthetic active radiation (PAR) increased from darkness to full light. Leaf conductance as a function of leaf temperature followed an optimum curve which in the model was replaced by two linear regression lines intersecting at the optimum temperature of 23.4°C. Increasing leaf-to-air water vapour concentration difference caused a linear decrease in leaf conductance. Leaf conductances became slightly more reduced by lowered water potentials in the low K plants. Stomatal closure in response to a temperature change away from the optimum was more sensitive in high K plants, and also the decrease in leaf conductance under the influence of lowered ambient humidity proceeded with a higher sensitivity in high K plants. Thus, under conditions which favoured high conductances increase of evaporative demand caused an about 10% larger decrease in leaf conductance in the high K plants than in the low K plants. Stomatal sizes and density in the flag leaves differed between low and high K plants. In plants with partially open stomata, leaf conductance, calculated from stomatal pore dimensions, was up to 10% lower in the high K plants than in the low K plants. A similar reduction in leaf conductance in high K plants was measured porometrically. It was concluded that the beneficial effect of K supply on water use efficiency reported in former studies primarily resulted from altered stomatal sizes and densities.  相似文献

Seedlings of the hypoxia-sensitive cucumber cultivar were hydroponically grown under hypoxia for 7 d in the presence or absence of 24-epibrassinolide (EBR, 2.1 nM). Hypoxia significantly inhibited growth, while EBR partially counteracted this inhibition. Leaf net photosynthetic rate (P N), stomatal conductance, transpiration rate, and water-use efficiency declined greatly, while the stomatal limitation value increased significantly. The maximum net photosynthetic rate was strongly reduced by hypoxia, indicating that stomatal limitation was not the only cause of the P N decrease. EBR markedly diminished the harmful effects of hypoxia on P N as well as on stomata openness. It also greatly stimulated CO2 fixation by the way of increasing the carboxylation capacity of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco), ribulose-1,5-bisphosphate regeneration, Rubisco activity, and the protection of Rubisco large subunit from degradation. Our data indicated that photosystem (PS) II was damaged by hypoxia, while EBR had the protective effect. EBR further increased nonphotochemical quenching that could reduce photodamage of the PSII reaction center. The proportion of absorbed light energy allocated for photochemical reaction (P) was reduced, while both nonphotochemical reaction dissipation of light energy and imbalanced partitioning of excitation energy between PSI and PSII increased. EBR increased P and alleviated this imbalance. The results suggest that both stomatal and nonstomatal factors limited the photosynthesis of cucumber seedlings under hypoxia. EBR alleviated the growth inhibition by improving CO2 asimilation and protecting leaves against PSII damage.  相似文献

Diurnal changes in net photosynthetic rate (P N), chlorophyll (Chl) fluorescence, and stomatal aperture of several hybrid poplar clones subjected to midday light stress were measured in July and August of 1996. Midday depression of P N, photosystem 2 (PS2) efficiency, stomatal conductance (g s), and stomatal aperture was observed in all clones, though at differing rates among them. Non-uniform stomatal closure occurred at noon and at other times, requiring a modification of intercellular CO2 concentration (C 1). A linear relationship was found between g s and stomatal aperture. More than half of the photons absorbed by PS2 centre dissipated thermally when subjected to light stress at noon. There was a linear relationship between the rate of PS2 photochemical electron transport (PxPFD) and P N. There was a consensus for two fluorescence indicators (1 – qP/qN and (Fm' – F)/Fm') in assessment of susceptibility of photoinhibition in the clones. Wheat (Triticum aestivum L.) is the largest cereal crop grown in Western Canada where drought during late vegetative and seed filling stages affects plant development and yield. To identify new physiochemical markers associated with drought tolerance, epidermal characteristics of the flag leaf of two wheat cultivars with contrasting drought tolerance were investigated. The drought resistant ‘Stettler’ had a lower drought susceptibility index, greater harvest index and water‐use efficiency than the susceptible ‘Superb’. Furthermore, flag leaf width, relative water content and leaf roll were significantly greater in Stettler than in Superb at moderate drought stress (MdS). Visible differences in epicuticular wax density on the adaxial flag leaf surfaces and larger bulliform cells were identified in Stettler as opposed to Superb. Mid‐infrared attenuated total internal reflectance spectra revealed that Stettler flag leaves had increased asymmetric and symmetric CH2 but reduced carbonyl esters on its adaxial leaf surface compared to Superb under MdS. X‐ray fluorescence spectra revealed a significant increase in total flag leaf Zn concentrations in Stettler in response to MdS. Such information on the microstructural and chemical features of flag leaf may have potential as markers for drought tolerance and thereby accelerate the selection and release of more drought‐resistant cultivars.  相似文献
