内生真菌产紫杉醇研究的回顾与展望   总被引:3,自引:1,他引:2  
紫杉醇是一种高效、低毒、广谱的天然抗癌药物,是人类未来20年间最有效的抗癌药物之一。近年来的研究发现,从植物内生真菌中发酵生产紫杉醇被证明是解决药源问题的有效途径。本文概述了微生物发酵生产紫杉醇的研究进展,包括产紫杉醇内生真菌的分离、多样性和真菌产紫杉醇的优势,紫杉醇的提取和检测技术,同时对如何提高内生真菌紫杉醇产量进行了比较全面的综述。随着现代分子生物学技术和微生物发酵工程的发展,工业上大规模发酵生产紫杉醇将有望实现。  相似文献

紫杉醇是重要的抗癌药物之一,已经证明其对多种癌症具有显著疗效。目前,人们主要是从红豆杉的树皮中提取、分离和纯化紫杉醇,但由于红豆杉为生长缓慢、散生、濒危的珍稀植物,且随着紫杉醇临床用途的不断拓宽,市场需求的稳定增长,单纯依靠从红豆杉树皮中提取紫杉醇已经无法满足日益增长的市场需求。为了解决紫杉醇的药源不足,科学家已把目光从红豆杉树分离提取紫杉醇转向了其他替代方法,如化学全合成、半合成、组织培养与细胞培养、微生物发酵法生产紫杉醇等。因此,了解内生真菌紫杉醇生物合成的分子基础和遗传调控机制,对解析内生真菌紫杉醇生物合成机制、构建高产紫杉醇基因工程菌株和早日实现内生真菌紫杉醇工业化生产具有重要的科学意义和现实意义。结合本课题组多年来的科研工作,概述了红豆杉细胞紫杉醇生物合成途径、内生真菌发酵生产紫杉醇的优势、产紫杉醇内生菌的分离研究现状和生物多样性及紫杉醇生物合成相关基因的研究现状。内生真菌生物发酵合成紫杉醇是可以无限生产、大量获取紫杉醇、解决紫杉醇药源短缺问题的很有前景的方法之一。  相似文献

The REMI method was used to introduce the plasmid pV2 harboring the hygromycin B phosphotransferase (hph) gene controlled by the Aspergillus nidulans trpC promoter and the trpC terminator into a taxol-producing endophytic fungus BT2. REMI transformation yielded stable transformants capable of continuing to grow on PDA medium containing 125 mug mL(-1) hygromycin B. The transformation efficiency was about 5-6 transformants mug(-1) plasmid DNA. The presence of hph gene in transformants was confirmed by PCR and Southern blot analyses. To the authors' knowledge, this is the first report on the transformation of taxol-producing endophytic fungi by the REMI technique. This study provides an effective approach for improving taxol production of endophytic fungi by the genetic engineering of taxol biosynthetic pathway genes in the future.  相似文献

一株产紫杉醇罗汉松内生真菌的分离和鉴定   总被引:4,自引:0,他引:4  
[目的]紫杉醇是重要的抗癌药物,主要从罗汉松等植物中提取,为了保护罗汉松等种质资源,本文从罗汉松植株中分离产紫杉醇内生真菌,并对内生真菌所产紫杉醇的抗肿瘤活性进行了分析.[方法]采用组织块法自罗汉松的根、茎、叶等组织中分离内生真菌;通过四唑蓝(Methyl ThiazolylTetrazolium,MTT)比色法筛选有抗肿瘤活性的内生真菌菌株,通过薄层层析(Thin Layer Chro-matography,TLC)和高效液相色谱(High Performance Liquid Chromatography,HPLC)对内生真菌所产活性物质进行鉴定;采用抽提法抽提内生真菌所产紫杉醇,应用Vero细胞对抽提的紫杉醇的活性进行了分析.[结果]从罗汉松属(Podocrapus)植物中分离到155株内生真菌,其中28株内生真菌具有较高的抑癌活性.将其中一株菌株A2命名为EPTP-1,经形态学和分子分类学分析鉴定为烟曲霉(Aspergillus fumigatus).菌株EPTP-1中抽提的紫杉醇5.553μg/L~555.3 μg/L作用24h表现出明显的致细胞凋亡作用.菌株EPTP-1发酵5天时紫杉醇的产率为0.5578±0.0294 mg/L.[结论]从罗汉松中分离到了一株产紫杉醇内生真菌EPTP-1,可作为紫杉醇类药物工业化生产的候选菌株.  相似文献

红豆杉内生真菌产紫杉醇相关基因BAPT的鉴定及初步研究   总被引:2,自引:0,他引:2  
从几种红豆杉中先后分离了30余种内生真菌,深入研究了三种能够产紫杉醇的内生真菌。形态学观察及18srDNA鉴定它们分属于Fusarium(属)和Pestalotiopsis(属),三个菌株均可以在离体培养的条件下产生紫杉醇,经两周培养产量可分别达到8.5,31.5,31.1μg/L。(其中Pestalotiopsis1分离于南方红豆杉,Fusarium1分离于东北红豆杉,Pestalotiopsis2分离于中国红豆杉)。对这些内生真菌产紫杉醇的初步机理作了研究。BAPT(C-13phenylpropanoidsidechain-CoAacyltransferase)是红豆杉中紫杉醇合成途径里支链合成的关键酶之一,我们根据其保守区序列设计了引物,首次在能产生紫杉醇的上述三种红豆杉内生真菌中克隆得到了BAPT基因片段,而分离的其它真菌并没有得到扩增。序列分析表明,来自内生真菌的BAPT基因片段序列与红豆杉BAPT基因片段序列具有非常高的相似性(98.9%)。推测红豆杉内生真菌之所以能够合成紫杉醇,相关基因可能直接源于其宿主植物,即其遗传学起源是基因转移而不是共进化。这同时也建立了一种快速经济的鉴定产紫杉醇真菌的辅助方法。内生真菌的遗传稳定性及改良在进一步研究中。  相似文献

Endophytic fungi represent an under explored resource of novel lead compounds and have the capacity to produce diverse classes of plant secondary metabolites. Here, we investigated the endophytic fungal diversity of taxol-producing endophytes from Taxus baccata L. ssp. wallichiana (Zucc.) Pilger and also tested the antimitogenic effect of fungal taxol using potato disc tumor assay. A total of 60 fungal endophytes were isolated from the inner bark (phloem-cambium) of T. baccata ssp. wallichiana, collected from different locations of the northern Himalayan region. Two key genes, DBAT (10-deacetylbaccatin III-10-O-acetyl transferase) and BAPT (C-13 phenylpropanoid side chain-CoA acyltransferase), involved in taxol biosynthesis were used as molecular markers for the screening of taxol-producing strains. Five representative species gave positive amplification hits by molecular marker screening with the bapt gene. These fungi were characterized and identified based on morphological and molecular identification. The taxol-producing capability of these endophytic fungi was validated by HPLC-MS. Among the five taxol-producing fungi, the highest yield of taxol was found to be 66.25 μg/l by Fusarium redolens compared with those of the other four strains.  相似文献

从云南红豆杉(Taxus yunnanensis)树皮内表皮中分离得到75株内生真菌,采用基于紫杉醇合成关键酶10-去乙酰巴卡亭Ⅲ-10-O-乙酰基转移酶(10-deacetylbaccatin Ⅲ-10-O-acetyl transferase,DBAT)和C-13苯丙氨基侧链CoA乙酰基转移酶(C-13-phenylpropanoid side chain-CoA acyltransferase,BAPT)基因为标志分子的快速筛选方法获得一株可产紫杉醇的内生真菌YN6,并通过高效液相色谱法和质谱法对其紫杉醇进行分析.同时,通过对内生真菌YN6的形态特征分析以及18S rDNA序列分析将其初步鉴定为拟盘多毛孢属(Pestalotiopsis sp.)真菌. 拟盘多毛孢YN6的紫杉醇产量约为120~140 μg/L,是目前已报道的紫杉醇产量较高的野生菌株之一. 拟盘多毛孢YN6的发现为微生物发酵生产紫杉醇提供了潜在的优良种质资源.  相似文献

This study investigated the endophytic fungi diversity of Taxus chinensis and screened the taxol-producing fungi in the host. A total of 115 endophytic fungi isolates obtained from bark segments of T. chinensis were grouped into 23 genera based on the morphological traits and sequence analysis of the internal transcribed spacers (ITS1-5.8S-ITS2), indicating endophytic fungi in T. chinensis are diverse and abundant. Diaporthe, Phomopsis (anamorph of Diaporthe), Acremonium, and Pezicula were the dominant genera, whereas the remaining genera were infrequent groups. The 13 representative species of the distinct genera were capable of producing taxol verified by reverse-phase high performance liquid chromatography (HPLC). Among the taxol-producing fungi, the yield of taxol produced by the Metarhizium anisopliae, H-27 was 846.1 μg l−1 in reformative potato dextrose liquid medium, and the fungal taxol was further validated by mass spectrometry (MS). The taxol-producing fungi (92.3%) were infrequent communities, suggesting that infrequent fungi associated with T. chinensis might be a fascinating reservoir of taxol-generating fungi.  相似文献

紫杉醇是一种高效、低毒、广谱的天然抗癌药物,可以有效地治疗乳腺癌、子宫癌等。近年来的研究发现,从植物内生真菌中发酵生产紫杉醇被证明是解决药源问题的有效途径。从分离到的420株内生真菌中筛选到一株产紫杉醇的内生真菌XC1-07为实验材料进行发酵条件的初步优化。结果表明:最适碳源、氮源分别是麦芽糖和NH4NO3;在含10g/L NH4NO3、90g/L麦芽糖、1.0g/L MgSO4、pH6的优化培养基中培养13d,紫杉醇的产量为1,124.34μg/L。为目前报道的植物内生真菌发酵生产紫杉醇最高的产量。  相似文献

为丰富产紫杉醇植物内生真菌资源库,从曼地亚红豆杉Taxus media茎中分离得到一株产紫杉醇的内生真菌TMS-26。通过对TMS-26的发酵提取物进行高效液相色谱分析,发现其具有与紫杉醇标准品(4.545 min)相近的色谱特征峰。进一步通过液质联用仪检测发现,内生真菌TMS-26的发酵提取物中具有与紫杉醇标准品((M+Na)+=876)相近的质谱特征峰,表明内生真菌TMS-26能够产生紫杉醇。同时通过传统形态学分类鉴定方法和18S r DNA序列分析、Internal-transcribed spacer(ITS)序列分析等现代分子生物学分类鉴定方法,最终将内生真菌TMS-26鉴定为曲霉属烟曲霉Aspergillus fumigatus,并命名为"烟曲霉TMS-26"。  相似文献

分离自太平洋红豆杉的天然抗癌药物紫杉醇(Taxol)已在临床上广泛应用,市场需求日益增强,但工业产量受原料红豆杉树木短缺的制约,供需存在巨大差距。1993年,Stierle等分离到一株与太平洋红豆杉共生的真菌安德烈紫杉菌,证实产生紫杉醇,为利用真菌发酵生产紫杉醇带来可能。通过分析目前工业生产技术存在的问题,总结了产紫杉醇真菌研究的进展及其重要意义。认为利用真菌大规模发酵生产紫杉醇或其中间体,是摆脱制约的一条新思路,具有广阔的应用前景,并且可以带动其他产品的开发。  相似文献

Taxol is the most effective antitumor agent developed in the past three decades. It has been used for effective treatment of a variety of cancers. A taxol-producing endophytic fungus Pestalotiopsis pauciseta (strain CHP-11) was isolated from the leaves of Cardiospermum helicacabum and screened for taxol production. The fungus was identified based on the morphology of the fungal culture and the characteristics of the spores and screened for taxol production. The amount of taxol produced by this endophytic fungus was quantified by HPLC and it produced 113.3 mg/L, thus the fungus can serve as a potential material for fungus engineering to improve taxol production. This fungal taxol also had strong anticancer activity against some cancer cells viz., BT 220, H116, Int 407, HL 251 and HLK 210 tested by Apoptotic assay and it is indicated that with the increase of taxol concentration from 0.005–0.05 mmol/L, taxol induced increased cell death through apoptosis.  相似文献

In this study, a total of 25 endophytic fungi were successfully isolated from the inner bark of Taxus baccata grown in Iran by the aseptic technique. Genomic DNA was extracted from isolated endophytic fungi and subjected to polymerase chain reaction (PCR) analysis for the presence of the Taxus taxadiene synthase (ts) gene, which encodes the enzyme catalyzing the first committed step of taxol biosynthesis. Four of 25 isolated endophytic fungi isolates showed PCR positive for the ts gene. Subsequently, taxol and 10-deacetylbaccatin III (10-DAB III) were extracted from culture filtrates and mycelia of the PCR positive isolates and analyzed by high-performance liquid chromatography and mass spectrometry. The analysis showed that one isolate (SBU-16) produced taxol (6.9?±?0.2?μg?L(-1) ) and its intermediate compound, 10-DAB III (2.2?±?0.1?μg?L(-1) ). The isolate SBU-16 was identified as Stemphylium sedicola SBU-16, according to its morphological characteristics as well as the internal transcribed spacer nuclear rDNA gene sequence analysis. Interestingly, this is the first report of the genus Stemphylium as a taxol-producing taxon.  相似文献

从南方红豆杉(Taxus chinensisvar.mairei)植株分离到内生真菌63株,其中24株经鉴定分属于19个种(属).真子囊菌纲的2个属2株,半知菌纲的14个属22株.真子囊菌纲的无毛毛壳属(Achaetomiumsp.)的1株为新发现种.半知菌纲的葡萄孢属(Botrytissp.)1株和无孢菌群(Mycelia sterlia)2株的培养液,经浓缩萃取,薄层层析检测,Rf值分别为0.404、0.404、0.401,与紫杉醇对照品Rf值一致,为南方红豆杉产紫杉醇内生真菌.  相似文献

The diterpenoid taxol is an important anticancer agent used widely in the clinic. The purpose of this work was to identify a taxol-producing endophytic fungus (strain TF5) isolated from Taxus mairei and study its anticancer activities. Strain TF5 was identified as a Tubercularia sp. according to the morphology of the fungal culture, the mechanism of spore production and the characteristics of the spores. Strain TF5 produced taxol, when grown in potato dextrose liquid medium and analyzed by thin layer chromatography, high performance liquid chromatography, ultraviolet and mass spectrometry. The fungal taxol, which was isolated from the organic extract of the TF5 culture, had strong cytotoxic activity towards KB and P388 cancer cells in vitro, tested by the MTT assay. Observed with immunofluorescence and electron microscopy, the fungal taxol enhanced microtubule stability and bundling in culture cells and induced tubulin polymerization in vitro similar to the authentic taxol.  相似文献

本研究从曼地亚红豆杉(Taxus x media)树皮内表皮分离得到一株产紫杉醇的内生真菌Z58,通过高效液相色谱法、质谱法和核磁共振波谱法对其紫杉醇提取物进行了分析. 结果表明,内生真菌Z58的紫杉醇提取物具有和紫杉醇标准品相近的色谱特征峰,其保留时间为10.2 min;也与紫杉醇标准品具有相同的质谱特征峰((M+Na)+=876)和1H-NMR谱带.并通过形态学特征分析和18S rDNA序列分析,将内生真菌Z58初步鉴定为肉座菌属(Hypocrea sp.)真菌.肉座菌Z58的紫杉醇产量约为2.5~3.0 μg/g(紫杉醇/菌丝干重),是一株具有潜在应用价值的产紫杉醇内生真菌.  相似文献

A total of 38 endophytic fungus strains were isolated from Taxus chinensis var. mairei by aseptic technique. Genomic DNA was extracted from isolated endophytic fungi and subjected to polymerase chain reaction (PCR) analysis for the presence of Taxus taxadiene synthase (TS) gene, a rate-limiting enzyme gene in the taxol biosynthetic pathway. Twelve out of 38 isolated endophytic fungus strains showed PCR positive for the ts gene. Subsequently, taxol and its related compounds were extracted from culture filtrates and mycelia of the PCR positive strains, separated by column chromatography and analyzed by High Performance Liquid Chromatography and Mass Spectrum. The analysis result showed that 3 strains could produce taxol and its related compounds at the detectible level. This study indicates that molecular detection of the ts gene is an efficient method for primary screening of taxol or its related compounds-producing endophytic fungi which can improve prominently screening efficiency.  相似文献
