The Betula pendula Roth. seeds used in this study were collected from three different natural populations in the province of Lugo (NW Spain). Three samplings were obtained from each population during the dispersal period of the species. The objective of our research was to assess the germination response of the seeds from each population and sampling when exposed to some of the most important effects of fires: high temperatures, ash and their joint effect.Significant differences have been found in the germination percentages of the seeds from the different populations and also from the three samplings. However, the tested effects of fire: heat, ash and their joint effect did not significantly affect the germination percentage of this species.  相似文献

This study aims to explore the effect of environmental factors (temperature, light, storage time) on germination response and dormancy patterns in eight Mediterranean native wildplants, belonging to the Euphorbia L. genus. In detail, we considered E. amygdaloides subsp. arbuscula, E. bivonae subsp. bivonae, E. ceratocarpa, E. characias, E. dendroides, E. melapetala, E. myrsinites, and E. rigida. We collected seeds from natural plant populations and performed germination assays in climatic chambers at seven constant temperatures (from 5 to 35°C, with 5°C increments), and four fluctuating temperature regimes (8/15, 8/20, 8/25, and 8/30°C, with a 12/12 hr thermoperiod). Germination assays were set up both in dark (D) and in light/dark conditions (L/D, 12/12 hr photoperiod), after short and long seed storage (SS around 30 days and LS around 150 days). For all these species, except E. amygdaloides subsp. arbuscula, results show that the final germinated proportions were improved by a long storage period (>150 days), which supports the existence of nondeep physiological dormancy. Optimal temperature levels ranged from 14.3 to 21.3°C and base temperatures ranged from 5.6 to 12.1°C, while ceiling temperatures from 25.6 to 34.7°C. For none of these species, germinations were favored by an alternating daily temperature regime, while in several instances, germinations were quicker and more complete in darkness, than in an alternating light/dark regime. In some instances, extreme temperature levels (5 and 30°C) induced dormancy and germinations did not resume when seeds were exposed at optimal temperature levels. Results are discussed in terms of the dynamics of emergences and how this might be affected by climate changes.  相似文献

大丽轮枝菌Verticillium dahliae是一种土传性植物病原真菌,可侵染660多种植物,引发黄萎病。微菌核是大丽轮枝菌在侵染后期形成的特殊休眠结构,可在土壤中存活14年,是黄萎病的主要初侵染来源。本研究在前期大丽轮枝菌萌发与休眠微菌核转录组分析的基础上,选择微菌核休眠状态高表达的VdToxD1VdToxD3基因进行功能研究。结果发现,VdToxD1VdToxD3敲除突变体菌株的微菌核萌发率分别为87.3%和86.0%,显著高于野生型菌株的64.0%;负调控微菌核萌发的转录因子VdCrz1与VdToxD1上游的-937 bp到-349 bp中的特定基序结合,与VdToxD3上游的-752 bp到-496 bp中的特定基序结合,结合位点均位于2个基因的启动子区。  相似文献

Microsclerotium formation by six isolates of Verticillium dahliae was studied at different temperatures both in vitro and in Arabidopsis thaliana . In vitro mycelial growth was optimal at 25°C, but microsclerotium formation was greatest at 20°C (two isolates) or 15–20°C (one isolate). Seedlings of A. thaliana were root-dipped in a conidial suspension, planted, and either placed at 5, 10, 15, or 25°C, or left at 20°C until the onset of senescence, after which some of the plants were placed at 5, 10, 15, or 25°C. The amount of microsclerotia per unit of shoot weight was assessed in relation to isolate and temperature. The optimal temperature for production of microsclerotia was 15–25°C. Two isolates each produced about 10 times more microsclerotia than each of the other four isolates. For these isolates, high R 2adj.-values of 0.77 and 0.66 were obtained, with temperature and its square as highly significant (P   < 0.001) independent variables. R 2adj.-values for the other isolates varied between 0.28 and 0.39. Moving plants to different temperatures at the onset of senescence led to microsclerotial densities that were intermediate between densities on plants that had grown at constantly 20°C and plants grown at other temperatures. This suggests that vascular colonization rate and rate of microsclerotium formation are similarly affected by temperature. The senescence rate of plants appeared unimportant except for plants grown at 25°C, which showed the highest amounts of microsclerotia per unit of plant weight in the most rapidly senescing plants.  相似文献

Conidia were germinated at three temperatures in media in which available water, expressed as water activity, was controlled at three levels. Rate of germination in the basal medium (0.9964 water activity) was most rapid at 25 C and was inhibited at 15 and 30 C. Lowering the water activity at a particular temperature decreased rates of germination. However, at a low water activity (0.9778) germination rate was greater at 30 C than at 25 C. Thus, the effect of temperature on germination appears to be dependent on the water activity of the medium.  相似文献

Under solid-state and liquid-state cultivations, the entomopathogenic fungus Verticillium lecanii F091 produced different types of spores. The aerial spores (AS) on cooked rice formed clusters on the tips of conidiophores, while the submerged spores (SS) were dispersed in the medium. The aerial spore appeared relatively uniform in size, which was 6.1 ± 0.9 m long, and 2.2 ± 0.3 m wide. The submerged spore varied in shape and size, with a mean length of 5.0 ± 1.0 m and width of 1.9 ± 0.5 m. Under scanning electron microscopy, the AS had a tendency to have rough, brittle surface characteristics; however, the SS appeared smooth on the surface. These spores were compared in two different germination media. On SMAY (Sabouraud maltose, agar, yeast extract, and neopeptone) coated coverslips, the AS did not show germ tubes until 8 h of incubation; while the SS showed many germ tubes. However, over 90% spore germination ratio was reached for both types of spores at 18-h of incubation. In the liquid medium, the SS germinated rapidly and many spores even produced spores on the spores; while the AS germinated, grew, and branched in the submerged culture gradually, and some sporulated on the tips of the short branches, or on the mycelia until 18 h of incubation. Evidently, the germination, growth patterns of aerial or submerged spores differed greatly under the different culture conditions. The virulence of the pathogen in relation to the type of spore of V. lecanii is discussed.  相似文献

Abstract. A mathematical model which describes the germination percentage dependency on time and temperature of a seed population was derived from the experimental results with a seed population of Amaranthus patulus Bertol. under sub-optimal temperature conditions (Washitani & Takenaka, 1984). The equation of the model which is a modified thermal time model is where G is germination percentage at a certain time after the start of imbibition ( t ) at a certain temperature ( T ), μ T 1 and σ T 1 are the mean and standard deviation of lower limit temperature among the seeds belonging to the seed population, and Tb, m , and A are the parameters characterizing the linear relationship between the rate and temperature, namely, Tb is the base temperature, m , the median of the required thermal time and A , a parameter determining the pattern of the variation of the required thermal time within seed population, respectively. The equation yields time courses for germination which are very similar to those observed by experiment.  相似文献

In tests using pre-soaked teliospores of Urocystis agropyri, germination was stimulated by tissues from all parts of the host plant (wheat) irrespective of cultivar or resistance to flag smut. Seedling tissues of some non-hosts (maize, rice, sorghum, cotton, mungbean and groundnut) also stimulated germination. Similar stimulatory effects were achieved by brief exposure to volatiles from wheat seedling tissues, and from the known sources of ethylene, ripening bananas, lemon fruit and ethephon (2-chloroethyl phosphonic acid) solution. The possibility that ethylene may have a direct triggering effect on teliospore germination is discussed.  相似文献

The influence of a non-cultivation (herbicide) regime on wilt incidence was compared with that of normal tillage in three observation trials on commercial hop farms. Non-cultivation resulted, on average, in 28% wilt control when wilt incidence was high. When wilt incidence was low there was a tendency for more wilt to occur with non-cultivation. The reduction in wilt under non-cultivation was probably mainly due to the virtually complete eradication throughout the year of alternative weed host plants, but other possible influential factors were: (i) the absence of root severance which could result in increased inoculum potential, (ii) changes in the pattern of hop root distribution, and (iii) the inhibiting action of simazine on root growth in surface soil resulting in reduced contact between roots and surface-borne inoculum. The lack of disease control with non-cultivation when wilt incidence was low was attributed to differential effects of the treatments on nitrogen metabolism or availability. The factors contributing to disease control under non-cultivation were considered to represent aspects of the process of sanitation, and the data were interpreted in terms of the sanitation ratio (sensu van der Plank). When wilt incidence was high the sanitation ratios at the three sites were similar. Modified cultural practices provide an important supplement to genetic resistance in the control of hop wilt. These practices (restricted use of nitrogen fertilizers and efficient sanitation with respect to the crop plant as well as to weeds) have proved economical and beneficial for hop culture in presence of wilt, under the threat of wilt, and in the absence of wilt.  相似文献

To date, studies investigating the consequences of shiftwork have predominantly focused on external (local) time. Here, we report the daily variation in cognitive performance in rotating shiftworkers under real-life conditions using the psychomotor vigilance test (PVT) and show that this function depends both on external and internal (biological) time. In addition to this high sensitivity of PVT performance to time-of-day, it has also been extensively applied in sleep deprivation protocols. We, therefore, also investigated the impact of shift-specific sleep duration and time awake on performance. In two separate field studies, 44 young workers (17 females, 27 males; age range 20-36 yrs) performed a PVT test every 2?h during each shift. We assessed chronotype by the MCTQ(Shift) (Munich ChronoType Questionnaire for shiftworkers). Daily sleep logs over the 4-wk study period allowed for the extraction of shift-specific sleep duration and time awake in a given shift, as well as average sleep duration ("sleep need"). Median reaction times (RTs) significantly varied across shifts, depending on both Local Time and Internal Time. Variability of reaction times around the 24?h mean (≈ ±5%) was best explained by a regression model comprising both factors, Local Time and Internal Time (p < .001). Short (15th percentile; RT(15%)) and long (85th percentile; RT(85%)) reaction times were differentially affected by Internal Time and Local Time. Germination of Onoclea sensibilis L. spores is controlled by light and temperature. Temperatures of 30 C can induce maximal germination in the dark to a level of 60 to 95% of that induced by a saturating dose of red light (0.38 joules/square meter) providing the spores are placed at the elevated temperature immediately after being sown. Maximum dark germination occurs with a minimum exposure of 16 to 24 hours at 30 C, suggesting that the temperature treatment is required for the induction of germination rather than for the germination process per se. Interaction of temperature and light for induction of germination shows nonadditive behavior. Germination induced by light and temperature applied consecutively never exceeded that which could be induced by a saturating dose of red light alone. Imbibition of the spores at 25 C in the dark for 12 or more hours prior to incubation at 30 C results in a loss of thermosensitivity. Dose response curves for red light induction of germination after varying times of imbibition at 25 C show no concomitant loss of sensitivity of the spores to red irradiation. This suggests that the mechanism and/or pathway of thermoinduction of germination differs from that of photoinduction. The loss of thermosensitivity as a result of presoaking at 25 C can be prevented if the spores are imbibed at 25 C in osmotic agents such as 0.3 molar mannitol or 0.1 gram per liter of polyethylene glycol 400 or in 0.08% dimethylsulfoxide or 10 micrograms per milliliter of herbicide SAN 9789 (4-chloro-5-(methylamino)-2-(α,α,α-trifluoro-m-tolyl-3-(2H)pyridazinone). The latter two substances are hypothesized to act upon membranes. These results suggest that the degree of hydration and possibly changes in membrane properties play a role in the change in sensitivity of Onoclea spores to temperature.  相似文献
