Summary Alcohol fixed blood films and fresh blocks of spinal cord were immersed in phosphate buffered neutral 10% formol for graded intervals, the films for 10, 30 min, 1, 2, 4, 8, 24 hr; the blocks for 2, 4, 6, 24 hr at 3 and 24° C; 1, 3, 7, 14, 21, 28, 42, 56 da, 3 and 14 mo at 24–26°. Graded deaminations in 2 N NaNO2/HAc at 3° C were applied: 1, 2, 5, 10, 20, 30 min; 1, 2, 4, 6, 8, 12, 18, 24, 36 hr. Blood films were stained at pH 6 and 6.5, tissue at pH 4.5 and 5.0, both in azure A eosin B. The point at which erythrocytes reached a slightly bluish green was taken as the end point, since no further color change occurred on further exposure and erythrocytes were the last of usually deamination susceptible tissue elements to lose their oxyphilia on deamination. Deamination of alcohol fixed blood films is completed in about 2 min, of sublimate fixed spinal cord in about 1 hr. Progressive formaldehyde exposure increased deamination time of blood films to 10–20 min in 1 hr, to 6–8 hr in 4 hr and to 12 hr in 24 hr. The tissue deamination showed similar progressive increase of deamination time, slower with 3° C fixation than with 24–26°, reaching 18–36 hr by about 3 days formol, and remaining about the same thereafter.Supported by National Cancer Institute Grant No. C-04816, National Institutes of Health.  相似文献

Effects of temperature, pH and water potential on blomass production or hyphal extension of Gliocladium virens (G20) and three Trichoderma isolates were determined in vitro. Optimum blomass production occurred between 20 and 30°C and at pH ranges between 4.6 and 6.8. Two isolates of T. viride grew at 5°C and G. virens grew at 35°C but no isolates grew at 40°C. Hyphal extension rates and conidial germination of all fungi declined with decreasing water potential over the range -0.7 to -14.0 MPa. In general, growth rates for each isolate were lower on potato/dextrose agar with water potential adjusted with polyethylene glycol than when adjusted with NaCl or glycerol. Summary Pollen grains from two hybrids, WF9xH55 (W) and K64xK55 (K) were collected and a sample from each was cultured immediately (0 h). The remainder was subdivided and stored at 2, 20, and 35° C. At 3, 6, 12, 24, 36, 48, 72, 96, and 120 h, a sample was cultured. The culture medium contained 15% sucrose, 0.6% bacto-agar, 0.03% calcium nitrate and 0.01% boric acid. Storage at 2° C resulted in a large increase in germination percentage in both W and K reaching a maximum at 24 h and then slowly decreasing with additional storage. No germination was observed at 96 h with W and at 120 h with K. The complete loss of germination occurred during a 24 h period and was very abrupt. At 20° C, a similar but less pronounced pattern was observed. However, after 24 h, aggregates of 50–1 000 pollen grains developed during storage in both W and K. Storage of W at 35° C slightly decreased the germination percentage at 3 h and eliminated it at 6 h. Storage of K at 35° C substantially increased the germination percentage at 3 h with further increases in storage periods resulting in the aggregation of grains. This general pattern of an increase at shorter storage periods followed by a gradual decrease as the storage period was extended, was found for pollen tube length and growth rate. The kinetics of primary dormancy loss were investigated in seeds of horse chestnut (Aesculus hippocastanum L.) harvested in four different years. Freshly collected seeds from 1991 held for up to 1 year at temperatures between 2C and 42C exhibited two peaks in germination (radicle growth), representing a low temperature (2-8°C) and a high temperature response (31-36°C). Germination at 36°C generally occurred within 1 month of sowing, but was never fully expressed in the seedlots investigated. At low temperatures (2-8°C), germination started after around 4 months. Generally, very low levels of termination were observed at intermediate temperatures (11-26°C). Stratification at 6°C prior to germination at warmer temperatures increased the proportion of seeds that germinated, and the rate of germination for all seedlots. Within a harvest, germination percentage (on a probit scale) increased linearly with stratification time and this relationship was independent of germination temperature (16-26°C). However, inter-seasonal differences in the increases in germination capacity following chilling were observed, varying from 0.044 to 0.07 probits d-1 of chilling at 6°C. Increased sensitivity to chilling was associated with warmer temperatures during the period of seed filling. The estimated base temperature for germination, Tb, for newly harvested seeds varied slightly between collection years but was close to 25°C. For all seedlots, Tb decreased by 1°C every 6 d of chilling at 6°C. This systematic reduction in Tb with chilling ultimately facilitated germination at 6°C after dormancy release.  相似文献

Grape anthracnose, which is caused by Elsinoë ampelina, is a disease that negatively affects grape production. This study aimed to investigate the effects of aeration, temperature, light, and preculture period on the formation of E. ampelina conidia and conidial germination and virulence. The colony morphology on potato dextrose agar (PDA) plates was more diverse than that in PDA bottles. The assessment of different culture methods, temperatures, light conditions, and preculture periods revealed that optimal conidial production occurred on 25‐day‐old colonies grown in PDA bottles at 21°C for 24 hr in the dark. The cultures in PDA bottles consistently produced approximately 5.0 × 106 conidia under these conditions. No conidial formation occurred when the cultures were kept at 25°C in the dark. The highest germination rate of E. ampelina was 80% at 25°C after 24 hr, whereas no germination was observed at 17°C after 12 hr. Pathogenicity tests revealed that symptoms of the disease were observed 4 days postinoculation (dpi) on leaves of Vitis vinifera cv. Red Globe. New conidia were observed on the lesions at 8 dpi. This study provides an effective method for the conidial production of E. ampelina that may also be applicable for other Elsinoë fungal species.  相似文献

Summary Laboratory-stored seeds ofDiamorpha cymosa (Nutt.) Britton (Crassulaceae) were germinated at monthly intervals starting shortly after maturity in late May and ending at approximately the time germination is completed in the field (November). Seeds were placed at 5, 10, 15, 20, 25, 30, 15/6, 20/10, 30/15 and 35/20°C at a 14-hr photoperiod (12/12 hr thermoperiods at the alternating temperature regimes) and in constant darkness. In June, seeds were almost completely dormant and thus germinated poorly or not at all under all conditions. As seeds aged from late May to November 1. germination at the 14-hr photoperiod increased in rate and total percentage, 2. the maximum germination temperature increased from 15 to 25°C at constant temperatures and from 20/10 to 30/15°C at the alternating temperature regimes and 3. the optimum temperature for germination increased from 15 to 15–20°C at constant temperatures but remained at20/10°C at alternating temperature regimes throughout the study. During the same period germination in constant darkness was negligible at constant and alternating temperature regimes. This pattern of physiological after-ripening apparently is an adaptation to summer-dry,winter-wet habitats such as rock outcrops of southeastern United States.A short period of illumination with white light given after a 12-hr imbibition period in darkness promoted germination in the dark at 25/10°C but not at 15 or 25°C. A short period of illumination given during the imbibition period was much less effective in promoting germination in the dark. Drying up to 7 days did not cause light-stimulated seeds to lose their ability to germinate in darkness. Summary In a first experiment, cucumber transplants (cucumis sativa L. cv. Toska) were grwon at five root-zone temperatures (RZT) ranging from 12° to 36°C. Maximum shoot growth and total leaf area were obtained at 24° and 30°C (RZT). In a second experiment, cucumber transplants were submitted to five RZT (12, 18, 24, 30 and 36°C) and five night air temperatures (NAT) that were maintained either constant at 9°, 13° and 17°C or splitted (in two halfs) at midnight (17°/12°C, 17°/9°C). Root-zone warming to 24° or 30°C increased cucumber plant growth and leaf development, but did not compensate completely the loss of productivity induced by low NAT. Split-night temperature had greater effects under the lowest NAT (17°/9°C) and at high RZT (24° or 30°C). In a third experiment, soil warming caused large increase in yields when cucumber plants were grown in the spring, but had very little effects in the fall.  相似文献

Summary The paper deals with some physiological studies of two typical strains ofPhytophthora palmivora Butl. andP. parasitica Dast. var.macrospora Ashby causing severe fruit rots ofAchras sapota L. andAnona squamosa L. respectively. The effect of temperature on the germination of sporangia had a marked effect. Maximum percentage of indirect germination (by formation of zoospores) occurred at 20° C, whereas direct germination of sporangia (by formation of germ tube) was at 30° C, which further continued upto 37° C. Maximum indirect germination at 20° C shows the favourable cool temperature for the epidemic of the diseases (Fruit rots) in nature. The best substrate (medium) for germination of sporangia was found to be tap water. Next to this was the host decoction or extract. Glucose solution also accelerated sporangial germination. The effect of two dyes viz., Malachite green and crystal violet was also studied in relation to growth and sporulation ofPhytophthora isolates. Their addition to the medium in various fractional dilutions had a profound influence in the rate of growth and sporulation. An interesting observation noted was that growth of the isolates was inversely porportional to the various concentrations of the dyes, under study. An attempt was also made to study the influence of various vitamins. In all, six vitamins were included in the study. Out of these, thiamine and riboflavin were found to be the best sources promoting good growth and sporulation of both the species ofPhytophthora under study.Forms a part of Senior author's M. Sc. A thermophilic Bacillus stearothermophilus strain AP-4 excreting a thermostable alkaline protease, was isolated from a local compost. Maximum activity of protease (250 U/ml) was after 36 h growth in broth at pH 9.0 and at 55°C. The protease was optimally active at pH 9.0 and 55°C and was stable in 5 mm CaCl2. The enzyme was completely inactivated by PMSF, EDTA and -mercaptoethanol. It is therefore a metal ion-dependent, alkaline, serine protease.R. Dhandapani and R. Butyrylcholinesterase purified from human serum as 6600-fold was heated at 37°, 40°, 45°, and 50°C for 24 hr. It was observed that the enzyme heated at 45°C for 24 hr converted to a stabilized form and followed Michaelis-Menten kinetics, whereas the enzyme samples, heated at the other temperatures for 24 hr, shown negative cooperativity with respect to its substrate, butyrylthiocholine. Even the sample heated at 45°C for 12 hr shown negative cooperativity. On the contrary to the heated enzyme at 40°C for 24 hr, the heated enzyme at 45°C for 24 hr could not be reactivated when it was kept at 4°C for 24 hr. In the kinetic studies, it was found that substrate analogs choline and benzoylcholine inhibited both the native enzyme and the enzyme heated at 45°C for 24 hr competitively, whereas succinylcholine was the partial competitive inhibitor of native enzyme but the pure competitive inhibitor of the heated enzyme.  相似文献

The effects of water temperature and bottom sediment type were studied on seed dormancy and germination of Zostera japonica Ascherson & Graebner in mesocosm. To test whether the germination rate is affected by cold stratification, seeds were divided into two groups: those exposed to cold (7 °C) and those left untreated (23–15 °C). Additionally, to mimic tidal variation, we used five tidal depth treatments for germination experiments in mesocosm. In mesocosm tanks, there was a wide range of daily fluctuating temperature at datum line +40 cm (17–25 °C), D.L. +20 cm (15 °C), and D.L. +0 cm (4–7 °C). In contrast, the maximum temperature range at D.L. −20 cm and −40 cm was narrow (5–6 °C). In the no cold stratification group, the maximum germination rates on sandy, muddy sand, and muddy bottom sediment were 3%, 11%, and 3%, respectively. In the cold stratification group (7 °C), the maximum germination rates were 40%, 53%, and 54%, respectively. First germination was observed 36 ± 0 days and 43 ± 6 days after the start of the germination experiment in the cold stratification group and the no cold stratification group, respectively. Bottom sediment type and tidal level did not affect seed germination in the both stratification group. Cold stratification strongly increases germination in all sediment types tested and under varying temperature regimes and at different tidal levels. We also tested whether seed germination is affected by daily fluctuations in temperature (10 °C constant, 15 °C/10 °C, and 20 °C/10 °C were compared) in an indoor incubator. Forty-two days after being sown, the maximum seedling emergence rates in the three groups were 3 ± 5%, 21 ± 7%, and 42 ± 26%, respectively. At 20 °C/10 °C, first germination was observed 11 days after the start of incubation, the germination rate rose sharply after 18 day of incubation, and then it leveled off after 32–42 days of incubation. In the no cold stratification group, seed germination was not observed in any of the three treatments. This finding suggests that the breaking of seed dormancy and germination of Z. japonica seeds are determined strongly by cold temperature and daily fluctuations of temperature, respectively.  相似文献

Biology and life table parameters of Brennandania lambi (Krczal) were studied at different temperatures while feeding on white mushroom (Agaricus bisporus) mycelium cultured on mushroom compost. The duration of egg and larva development, preoviposition and oviposition period, female longevity, and the time to 50% mortality declined as temperature increased from 16 to 28°C. The threshold temperature of development (female) was 9°C and the thermal constant for completion of development (female) was 195 day-degrees. At 16, 20, 24 and 28°C, the total fecundity (eggs/female) was 71, 67, 66 and 57, respectively and the daily fecundity rate (eggs/female/day) was 5.6, 8.7, 8.7 and 9.1, respectively. The sex ratio (female/male) ranged from 1.9 to 2.1 at 16–28°C. At 16, 20, 24 and 28°C, the intrinsic rate of natural increase (r m) was 0.11, 0.18, 0.22 and 0.27, respectively, and the population doubling time was 6.1, 3.9, 3.2 and 2.5 days, respectively. All life stages of the mite died when exposed to 35°C constant temperature for 24h, or to 32°C constant temperature for 12 days or to 31–35°C (average 32.9°C) ambient temperature for 4 days. Brennandania lambi completed development only when fed on Ag. bisporus mycelium growing on mushroom compost. It could not survive on mushroom mycelia of Auricularia auricula, Au. polytricha, Ganoderma lucidum, Hericium erinaceus, Lentinus edodes, Pleurotus ostreatus, P. sajor-caju and Tremella fuciformis.  相似文献

Temperature markedly affected germination and germ tube length of A. niger. More than 90% of the spores were germinated in the range 30°–34 °C and formed maximum length of germ tubes. At temperatures from 38° to 43 °C, the proportion of the spores that germinated as well as the germ tube length were both gradually decreased. However, at 47°C germ tube formation was completely inhibited up to 15 hrs. after inoculation.High relative humidity was found necessary for the spore germination of A. niger. Germination failed to occur at 76% relative humidity. At 78 and 81% relative humidity germination was detected 15 hrs. after inoculation while at the higher humidities germination was started after 6 hrs. only.Conidiospores of A. niger were very sensitive to changes in the hydrogen ion concentration, pH. Complete inhibition of germination was found at pH less than 3.5. The germination and the length of the formed germ tubes increased with pH to reach their maximum rates at pH 4.5.  相似文献

Summary The pathogenT. maculans inciting leaf spot of turmeric perpetuates in the form of ascospores and conidia and incites first infection on the lowermost leaves, in October and November when the atmospheric humidity prevails about 80 % with 21°–23° C temperatures. The secondary infection is due to availability of large potential of inoculum of ascospores and conidia frequently and periodically produced by the pathogen under cool temperature conditions (20°–24° C) with about 80 % atmospheric humidity. Oospores germinated in distilled water, but the addition of nutrients increased the percent germinating. A germ tube emerged which quickly elongated into a branched aseptate hypha. Germination occurred over a wide range of temperature. Germinability decreased with age of culture. The incubation temperature and the addition of cholesterol to the medium affected germination percentage. Oospores, formed in cultures on media to which cholesterol was added, were more resistant to heat and to ultraviolet radiation. Oospores survived 2 hours exposure to 50 °C. The effects of stratification temperatures and burial in soil on dormancy levels of Carex pendula L. and C. remota L., two spring-germinating perennials occurring in moist forests, were investigated. Seeds buried for 34 months outdoors, and seeds stratified in the laboratory at temperatures between 3 and 18 °C for periods between 2 and 28 weeks, were tested over a range of temperatures. Seeds of the two species responded similarly to stratification treatments, except for an absolute light requirement in C. pendula. Primary dormancy was alleviated at all stratification temperatures, but low temperatures were more effective than higher ones . (≥ 12 °C). Dormancy induction in non-dormant seeds kept at 5 °C occurred when seeds were subsequently exposed to 18 °C. Dormancy was not induced by a transfer to lower temperatures. Buried seeds of both species exhibited seasonal dormancy cycles with high germination from autumn to spring and low germination during summer. Temperatures at which the processes of dormancy relief and of dormancy induction occurred, overlapped to a high degree. Whether, and when, dormancy changes occurred depended on test conditions. The lower temperature limit for germination (> 10%) was 9 °C in C. remota and 15 °C in C. pendula. Germination ceased abruptly above 36 °C. Germination requirements and dormancy patterns suggest regeneration from seed in late spring and summer at disturbed, open sites (forest gaps) and the capability to form long, persistent seed banks in both species.  相似文献

Polysaccharidases adsorbed on commercial amylodextrins were added to environmentally controlled composts of straw plus poultry manure. After 5 days of composting at 48°C, microbial enzyme activities and numbers of bacteria were higher in the treated compost than in the control. During the next phase at 80°C, between days 5 and 6, more C and N were solubilized in the treated compost. After introducing a microbial inoculum on day 6, and maintaining the substrate at 48°C, colonization by bacteria was faster in the treated compost and consequently, more fibre was degraded. The effect of temperatureon conidial germination, mycelial growth, andsusceptibility of adults of three tephritidfruit flies, Ceratitis capitata(Wiedemann), C. fasciventris (Bezzi) andC. cosyra (Walker) to six isolatesof Metarhizium anisopliae were studied inthe laboratory. There were significantdifferences among the isolates in the effect oftemperature on both germination and growth.Over 80% of conidia germinated at 20, 25 and30°C, while between 26 and 67% conidiagerminated at 35°C and less than 10% at15°C within 24 hours. Radial growth was slowat 15°C and 35°C with all of theisolates. The optimum temperature forgermination and mycelial growth was 25°C. Mortality caused by the six fungal isolatesagainst the three fruit fly species varied withtemperature, isolate, and fruit fly species.Fungal isolates were more effective at 25, 30and 35°C than at 20°C. The LT90values decreased with increasing temperature upto the optimum temperature of 30°C. Therewere significant differences in susceptibilitybetween fly species to fungal infection at allthe temperatures tested.  相似文献
