Frankia in acid soils of forests devoid of actinorhizal plants   总被引:1,自引:0,他引:1  
The capacity of some acid forest soils to induce nodulation on a hybrid between Alnus incana (L.) Moench and A. glutinosa (L.) Gaertn. was investigated. Soil was sampled from tree stands devoid for decades of actinorhizal hosts. Seven-week-old Alnus seedlings growing m liquid culture were inoculated with soil dilutions. The nodules were counted after 6 weeks and classified as Sp, if they lacked spores, or as Sp+. if spores were present, according to microscopy of microtome sections. Frankia was found in all the forest soils studied, apart from a soil from a Betula swamp. The highest nodulation capacities on Alnus , caused predominantly by Frankia of the Sp type. were observed in mineral soil sites with Betula stands — even higher than in soil from an A. incana stand. A positive correlation was found between the pH and the noduiation capacity of the soil.  相似文献

Host specificity between local Frankia strains and native alders [Alnus incana (L.) Moench and A. glutinosa (L.) Gaertn.] was evaluated in inoculation experiments. Pure cultures of Frankia , whether originating from A. incana or A. glutinosa , were infective and effective on both host species. These pure cultures were isolated from spore-negative (Sp) nodules. From spore-positive (Sp+) nodules no Frankia isolates were obtained. This strain type resisted all our isolation attempts and therefore crushed nodules had to be used for Sp+ type inoculations.
The Sp+ type Frankia populations differed in their host specificity. Sp+ nodules from A. glutinosa were effective on both alder species, but Sp+ nodules from A. incana induced effective nodules only on the original host; on A. glutinosa only small (1-3mm) prenodule-like structures were found. Such A. glutinosa plants died on N-free medium, thus showing that these nodules were ineffective. In the effective nodules the middle cortex was dominated by infected cells filled with vesicle clusters. In the ineffective nodules only a few cortical cells were infected and sporangia predominated in these cells. Surprisingly enough they also contained vesicle-like structures as demonstrated in electron micrographs.  相似文献

Occurrence and activity of the hydrogen uptake enzyme were studied in root nodule homogenates made from plants of Alnus incana (L.) Moench collected from field sites in the northern part of Sweden. Nitrogenase (EC 1.7.99.2) activity (estimated by acetylene reduction) and hydrogen evolution were studied in excised nodules. All Frankia sources showed acetylene reduction activity, and possessed a hydrogen uptake system. Hydrogen uptake in nodule homogenates from the Frankia sources measured at 23.8 μM H2 ranged from 0.04 to 5.0 μmol H2 (g fresh weight nodule)−1 h−1. The H2 uptake capacity of nodule homogenates from one of the Frankia sources was almost 8 times higher than the hydrogen evolution from nitrogenase, both expressed on a nodule fresh weight basis. Frankia sources from field sites 6 and 11 showed Km for H2 of 13.0 and 23.6 μM H2, respectively. This indicates similarities in the hydrogen uptake enzymes in the two Frankia sources. It is concluded that hydrogen uptake is a common characteristic in Frankia.  相似文献

The responses of three species of nitrogen-fixing trees to CO2 enrichment of the atmosphere were investigated under nutrient-poor conditions. Seedlings of the legume, Robinia pseudoacacia L. and the actinorhizal species, Alnus glutinosa (L.) Gaertn. and Elaeagnus angustifolia L. were grown in an infertile forest soil in controlled-environment chambers with atmospheric CO2 concentrations of 350 μl −1 (ambient) or 700 μl −1. In R. pseudoacacia and A. glutinosa , total nitrogenase (N2 reduction) activity per plant, assayed by the acetylene reduction method, was significantly higher in elevated CO2, because the plants were larger and had more nodule mass than did plants in ambient CO2. The specific nitrogenase activity of the nodules, however, was not consistently or significantly affected by CO2 enrichment. Substantial increases in plant growth occurred with CO2 enrichment despite probable nitrogen and phosphorus deficiencies. These results support the premises that nutrient limitations will not preclude growth responses of woody plants to elevated CO2 and that stimulation of symbiotic activity by CO2 enrichment of the atmosphere could increase nutrient availability in infertile habitats.  相似文献

Variation in stomatal development and physiology of mature leaves from Alnus glutinosa plants grown under reference (current ambient, 360 μmol mol−1 CO2) and double ambient (720 μmol mol−1 CO2) carbon dioxide (CO2) mole fractions is assessed in terms of relative plant growth, stomatal characters (i.e. stomatal index and density) and leaf photosynthetic characters. This is the first study to consider the effects of elevated CO2 concentration on the distribution of stomata and epidermal cells across the whole leaf and to try to ascertain the cause of intraleaf variation. In general, a doubling of the atmospheric CO2 concentration enhanced plant growth and significantly increased stomatal index. However, there was no significant change in relative stomatal density. Under elevated CO2 concentration there was a significant decrease in stomatal conductance and an increase in assimilation rate. However, no significant differences were found for the maximum rate of carboxylation ( V cmax) and the light saturated rate of electron transport ( J max) between the control and elevated CO2 treatment.  相似文献

Hybridization of a 5S rDNA probe to Southern transfers of birch (Betula papyrifera) or alder (Alnus incana) DNA digested with BamH1 reveals similar triple-band "ladder-like" patterns. The sizes of sequenced 5S repeat units from both plants ranges only from 471 to 490 base pairs, suggesting that the complexity detected by Southern analysis is not due to different size classes of 5S repeats as found in other species. Within the intercistronic spacer region, conservation of large blocks of sequence between birch and alder 5S is observed implying a close evolutionary relationship between these two species. In both species, a duplication of part of the coding sequence including a restriction site for BamH1 introduces a second BamH1 site into the repeat unit. Differential methylation of the two BamHI restriction sites can account for the observed triple-band pattern.  相似文献

利用AFLP分子标记结合形态学指标,采用UPGMA法进行聚类分析,对桤木属17个种57份材料进行了亲缘关系研究及一个模糊种鉴定。结果表明:7对引物扩增出369条带,其中346个多态位点,多态位点百分率为93.77%;根据AFLP标记位点聚类分析,在相似系数为0.782时,17种桤木属植物可分为4类,第一类为日本桤木(Alnus japonica);第二类为绿桤木(A.viridis)、意大利桤木(A.cordata)、欧洲桤木(A.glutinosa)、模糊种、四川桤木(A.cremastogyne)、江南桤木(A.trabeculosa)、斑点桤木(A.incana ssp.rugosa)、东北亚灰桤木(A.hirsuta)、台湾桤木(A.formosana)、日本特有桤木(A.firma)和裂叶桤木(A.sinuata);第三类为灰桤木(A.incana)、红桤木(A.rubra)及薄叶桤木(A.tenifolia);第四类喜马拉雅灰桤(A.nitida)和尼泊尔桤木(A.nepalensis)。根据形态学聚类分析,在距离为1.4时可分为三类,意大利桤木(A.cordata)单独为一类;日本桤木(A.japonica)、台湾桤木(A.formosana)、喜马拉雅灰桤木(A.nitida)、江南桤木(A.trabeculosa)和东北亚灰桤木(A.hirsuta);第三类包括模糊种、灰桤木(A.incana)、斑点桤木(A.incana ssp.rugosa)、裂叶桤木(A.sinuata)、红桤木(A.rubra)、欧洲桤木(A.glutinosa)、绿桤木(A.viridis)、四川桤木(A.cremastogyne)、薄叶桤木(A.tenuifolia)和尼泊尔桤木(A.nepalensis)。经形态特征和AFLP分析鉴定模糊种为欧洲桤木。形态学聚类与AFLP聚类结果基本一致,但仍存在一定的差异,说明桤木属植物遗传背景丰富,种的分子分类地位和形态学分类地位具有一定的差异。  相似文献

In vivo nitrate reductase (NR, EC 1.6.6.1.) activity was measured in leaves, branches and trunk of field-grown Alnus glutinosa (L.) Gaertn. All of the assayed tissues enzymatically reduced nitrate with a decreasing activity [μmol NO2 (g dry weight)−1 h−1] in the order: leaves > branch bark > inner branch tissues > trunk xylem. The NR activity of the various tissues of excised branches was inhibited by tungstate added to the transpiration stream. Part of the nitrate added to the feeding solution (0.2, 0.5 or 1 m M KNO3) of excised branches disappeared during its transport via the transpiration stream in the perennial tissues. This disappearance was enzymatic since it was decreased by tungstate.
No evidence was obtained for the presence of nitrate in natural xylem sap nor for a significant correlation between nitrate content of soil and leaf NR activity. These results indicate that in the field-grown black alder, the nitrate not reduced in the roots could be reduced in the perennial tissues of aerial parts. Since the leaf NR activity does not reflect the actual in situ nitrate reduction, the existence of a constitutive NR activity in Alnus leaves is suggested.  相似文献

Presence and activity of the enzymes superoxide dismutase (SOD) and catalase were studied in Frankia in symbiosis with Alnus incana (L.) Moench. Analysis on native PAGE gels indicated that symbiotic Frankia contained an FeSOD and catalase. The activity of the enzymes was in the same range as reported for cultured Frankia . Attempts to characterize SOD by western blots with antisera from Escherichia coli and Azotobacter vinelandii did not give clear-cut results with the antibodies used. Alnus incana plants were grown with the root system in 5, 10, 21 or 40% O2 for up to 6 days. Nitrogenase activity, measured as ARA (acetylene reducing activity) dropped within 3 h when roots were exposed to low or high oxygen. At 40% O2 ARA was almost completely lost while at 5 and 10% O2 ARA decreased to 69 and 74% of the inital value, respectively, Nitrogenase activity recovered at ail oxygen tensions. Recovery rates resembled the continuous increase in ARA in plants continuosly kept at 21% O2, and suggests that new vesicles with envelopes of appropriate thickness were formed. The ARA measurements confirm results from an earlier study where nitrogenase activity was measured as H2 evolution. There was a tendency for increased SOD and catalase activities in Frankia from root systems exposed to 40% O2 for 24 h but not earlier or later than this. When data from all experimental times were pooled. SOD activity increased significantly with increased oxygen tension whereas catalase activity decreased. Although ARA per plant varied with oxygen tension, there was no statistically significant correlation between ARA and SOD or between ARA and catalase. It seems that being linked to nitrogenase activity is only one role of SOD and catalase in this symbiotic Frankia .  相似文献

Regulation of nodulation in Alnus incana-Frankia symbiosis   总被引:1,自引:0,他引:1  
We have studied regulation of nodulation in Alnus incana (L.) Moench using double inoculations in plastic pouches and a slide technique to observe root hair deformation. Initially, the distribution of nodules between main and lateral roots appeared quite constant, independent of the concentration of inoculum (1 to 250 μg of crushed nodules plant−1). Susceptibility to infection after the second inoculation was restricted to lateral roots after the initial infections developed. When pre-existing nodules were excised before the second inoculation, subsequent nodules appeared to arise where infections had arrested at stages earlier than actual nodule emergence. We observed that root hairs formed postinoculation were very crowded and short with a pronounced deformation. No nodules were found later on this region of the root, suggesting a loss of susceptibility in this region. Split-root experiments with delays between inoculation of the first and second side of the root system showed irreversible, systemic inhibition of nodulation on the second side starting between 3 and 6 days after the inoculation of the first side. Only when compatible, infective strains were used in the first inoculation, was nodule formation inhibited after the second inoculation. We conclude that autoregulation of nodulation operates in Alnus incana and on a time scale similar to what is found in some legumes.  相似文献

Pollen analysis was carried out on lacustrine sediment of a small hollow (15 m x 25 m) at the treeless sub-Alpine belt (202 m) of the inner Maurienne valley in the northern French Alps. A 2,500-year-long maple settlement was demonstrared. Three AMS dates of terrestrial plant macroremains support the chronology. First, Betula and Salix spread prior to 9,000 C14 BP. The first pollen grains of Acer, Abies and Pinus cembra are quoted at 8,600 C14 BP. High frequencies of Alnus glutinosa/incana (20%) and Acer (10%) show that mixed communities of Acer and Alnus persisted above the mountainous Abies forest between 7,490 and 5,850 C14 BP. After 5,850 C14 BP, the decrease in Acer stands could be attributed to fire as suggested by the strong increase in Betula and by the delayed expansion of Pinus cembra.  相似文献

Actinorhizal nodules do not usually evolve H2 due to the action of an uptake hydrogenase. We have found that nodules of several Frankia symbioses evolved large amounts of H2 gas when returned to air following exposure to 10 kPa C2HT2 during an acetylene reduction assay. Increased H2 evolution in air persisted for several days when intact root systems of Alnus incana (L.) Moench (inoculated with Frankia UGL 011101) were treated with 10 kPa C.H2 for 1 h. Full recovery of uptake hydrogenase activity required 4 to 8 days. Studies with crude homogenates of nodules of the same plants showed that hydrogenase (measured amperometrically with phenazine metho-sulfate as electron acceptor) was directly affected, since activity in treated nodules was only 10% of that in untreated nodules. A survey of actinorhizal symbioses revealed variation in the effect of an acetylene reduction assay on hydrogen metabolism. Nodules of three species, including Alnus rubra Bong, inoculated with Frankia HFPArD. showed complete inactivation of hydrogenase. H2 evolution in air was 25% of the C2H2 reduction rate and H, evolution in Ar/O2 was equal to the QH2 reduction rate. Two symbioses, Ceanothus americanus L. (soil inoculant) and Batista glomerata Baill. (soil inoculant) showed no change following an acetylene reduction assay. A third group of symbioses showed an intermediate response.  相似文献

Abstract. The principal polyamines in Alnus glutinosa roots, nodules and root pressure sap, putrescine, spermidine and spermine, were quantified by reversed-phase, high-performance liquid chromatography with fluorescence detection following precolumn derivatization with 9-fluorenylmethyl chloroformate and 1-ada-mantanamine. Putrescine was the major component of all tissues and sap. It comprised 70% or more of the polyamine pool except in roots of KNO3-fed plants, in which similar amounts of putrescine and spermidine were present at levels five-fold lower than plants fed (NH4)2SO4. Polyamine levels in nodules were 50% greater than in roots. The polyamine content of roots and nodules was not altered significantly when the nitrogen nutrition was changed from sole reliance on nitrogen fixation to partial or complete utilization of (NH4)2SO4. However, the polyamine content of root pressure sap from nodulated plants increased almost four-fold when they were fed with increasing concentrations of NH4NO3, although the total polyamine content remained low (5mmol m−3 sap). The polyamine content of the Alnus root system changed with plant age. In particular, the spermidine content of both roots and nodules was higher in 10- as compared to 16-week-old plants.  相似文献

A clone of Alnus incana (L.) Moench was grown in symbiosis with a local source of Frankia or with Frankia Ar14. Seven to 9-week-old plants were given 20 m M NH4Cl (20 m M KCl = control) for 3 days. Nitrogenase activity of intact plants decreased gradually within the 3 days of treatment to about 10% of the initial rates. Hydrogen evolution in air and total nitrogenase activity responded similarly to the treatment. Relative efficiency of nitrogenase thus remained the same throughout the study period. Control plants were not affected. Measurements of nitrogenase activity in root nodule homogenates (in vitro measurements) indicated loss of active nitrogenase rather than shortage of energy for nitrogenase activity in Frankia from ammonium-treated plants. Shoots were exposed to 14CO2 and translocation of 14C to Frankia vesicle clusters prepared from root nodules was studied. Frankia vesicle clusters from ammonium-treated plants contained about half as much 14C as those of control plants during all 3 days studied. One explanation for the observed effects is that a reduced supply of carbon to Frankia vesicles in the root nodules caused a reduced metabolic rate, including reduced protein synthesis and synthesis of nitrogenase.  相似文献

Frankia vesicle clusters were prepared from Alnus incana (L.) Moench root nodules containing a local source of Frankia by an improved homogenization-filtration procedure. The capacity of the vesicle clusters to metabolize hexoses was investigated by respirometric and enzymological studies. The vesicle clusters could utilize glucose, glucose-6-phosphate and 6-phosphogluconate provided that appropriate cofactors were added to the preparations. The enzymes hexokinase (EC 2.7.1.1), NADP+: glucose-6-phosphate dehydrogenase (EC 1.1.1.49) and NAD+;6-phosphogluconate dehydrogenase (EC 1.1.1.44) were found in cell-free extracts of the vesicle clusters and kinetic constants for the enzymes were determined. Hexokinase had a lower Km for glucose than for fructose. Extracts from both symbiotic and propionate grown Frankia AvcII also showed activity of these hexose-degrading enzymes, indicating that their presence is not necessarily dependent on sugars as carbon source. The NAD+- dependent 6-phosphogluconate dehydrogenase was only present in Frankia cells and not in alder root cells, which makes this enzyme a useful Frankia -specific marker in these symbiotic systems.  相似文献

Estimations of phylogenies from morphological and molecular data often show contrasting results. We compared morphological and molecular phylogenies in an ancient family of woody dicots, the Betulaceae (birch family). The phylogeny of the family was estimated from parsimony analysis of morphological characters in the genera Alnus, Betula, Carpinus, Corylus, Ostrya, and Ostryopsis and from parsimony and distance-matrix analyses of DNA sequences of the chloroplast gene encoding the large subunit of ribulose-1,5-biphosphate carboxylase (rbcL) in the genera Alnus, Betula, Carpinus, Corylus, and Ostrya and in two outgroups, Quercus and Liquidambar. The topologies obtained by the different methods were completely congruent, and bootstrapping strongly supported the division of the family Betulaceae into two major clades, Betuleae (Alnus and Betula) and Coryleae (other members). Only slightly more homoplasy was present in the rbcL sequence data set than in the morphological set. Relative-rate tests indicated that the Coryleae clade had a faster rate of rbcL evolution than did the Betuleae clade. Heterogeneity of rates of morphological evolution also paralleled those for rbcL.  相似文献

The expression of PR-protein b1' in plants and cell suspension cultures of Nicotiana glutinosa L., Nicotiana debneyi Domin, and an amphidiploid cross of these two species, a hybrid, has been investigated. An enzyme linked immunosorbent assay has been employed to determine the concentration of PR-protein b1' in extracts. The PR-Protein b1' was constitutively produced in intact plants of the hybrid (around 25 μg g−1 leaf tissue), while only trace amounts of the protein (< 50 ng g−1 leaf tissue) were found in plants of the two parents. In suspension culture, the concentrations of PR-protein b1' were 8, 0.4 and less than 0.1 mg l−1 medium for the hybrid. N. debneyi and N. glutinosa , respectively. Only trace amounts of the protein were found in extracts from cells. Seven days after infection by tobacco mosaic virus (TMV) the concentration of PR-protein b1' in leaves of N. glutinosa was 22.5 μg g−1 leaf tissue. In N. debneyi and the hybrid a relatively limited induction of PR-protein b1' by TMV was observed. The influence of various phenoxyacetic acids on the expression of PR-protein b1' in the 3 cell cultures has been investigated. Cultures of N. glutinosa responded to treatments with 2,4-D and 2,4,5-T while cultures of N. debneyi and the hybrid were essentially unaffected. In the former case a concentration of 5–10 mg l−1 2,4,5-T was optimal and cells were most responsive to the treatment 4 days after subcultivation. The concentration of PR-protein b1' in elicited cell cultures of N. glutinosa was 2 to 4 mg l−1 medium.  相似文献

Seasonal changes in the activity of phospho enol pyruvate carboxylase (PEPCase, EC 4.1.1.31), a key enzyme in the interaction of carbohydrate and nitrogen metabolism, were studied in leaves of the C3 semiparasitic mistletoe, Viscum album, growing on different host trees. Maximum extractable PEPCase activities were higher in leaves of mistletoes growing on Betula pendula and Alnus glutinosa hosts compared with those on the conifers, Abies alba and Larix decidua . Independent of host, maximum extractable PEPCase activities were high in spring and autumn while low in summer. Samples with higher PEPCase activities showed higher amounts of PEPCase protein and higher PEPCase mRNA levels. A curvilinear correlation between leaf total nitrogen content and the maximum extractable PEPCase activity as well as PEPCase mRNA level suggested that nitrogen might affect the activity of PEPCase of mistletoe by up-regulating gene expression. In addition to extractable activity, seasonal changes of the PEPCase activation state, the ratio of activities resulting from limited:non-limited assays, were found, which was correlated to the variation of malate content in leaves of mistletoe. ATP-dependent activation of PEPCase was characterized by an increase in I0.5( l -malate), indicating that PEPCase of leaves of mistletoes is probably regulated via phosphorylation.  相似文献

Abstract— The phospholipase A1 (EC 3.1.1.32) and A2 (EC 3.1.1.4) activities of rat sciatic nerve homogenates have been studied. With phosphatidylcholine as substrate normal nerve had significant activity of both types at pH 5.0. Substantial increases occurred in nerve undergoing Wallerian degeneration after transection, beginning as early as 2 days after operation and rising to eight times normal values by the second week.  相似文献

Abstract: Chronic treatment with the adenosine receptor antagonist caffeine evokes an up-regulation of A1 adenosine receptors and increased coupling of the receptor to G proteins in rat brain membranes. However, chronic agonist exposure has not been explored. Primary cultures of cerebellar granule cells were exposed chronically to A1 adenosine receptor agonists and antagonists. Exposure to the A1 adenosine receptor agonist N 6-cyclopentyladenosine resulted in (1) a time- and concentration-dependent reduction in the density of receptors labeled by 1,3-[3H]dipropyl-8-cyclopentylxanthine, (2) an enhanced ability of guanyl nucleotides to decrease the fraction of A1 adenosine receptor sites displaying high affinity for 2-chloroadenosine, and (3) a functional uncoupling of receptors from adenylyl cyclase (EC 4.6.1.1). The adenosine antagonists caffeine and 8- p -sulfophenyltheophylline produced alterations in A1 adenosine receptor homeostasis that were antipodal to those associated with agonist treatment. Antagonist exposure (1) increased the density of A1 adenosine receptors in cerebellar granule cell membranes, (2) blunted the effect of guanyl nucleotides on receptor coupling to G proteins, and (3) increased the functional coupling of receptors to adenylyl cyclase inhibition. Forskolin treatment of cerebellar granule cells did not affect receptor density, suggesting that cyclic AMP is not involved in the regulation of A1 adenosine receptor expression.  相似文献
