Verticillium agaricinum (Link) Corda, grown in a yeast extract-sucrose medium, conidiated abundantly in darkness after irradiation with near ultraviolet (290–400 nm) for 15 min or blue light (400–550 nm) for 60 min. Few conidia were formed in total darkness. Exposure to 30 min of near ultraviolet light suppressed conidiation. Conidiation was also suppressed by phosphate in excess of 10−4 M irrespective of light condition. After irradiation with near ultraviolet light for more than 30 min, there was a cessation of growth and a change in colony color from yellow to reddish. The color does not appear to be due to a carotenoid because the colonies turned from red to yellow when covered with acid. At pH lower than 6.0 the pigment has an absorption maximum around 390 nm, whereas at higher pH it is around 540 nm. Thus, it appears that irradiation of V. agaricinum with near ultraviolet may cause an increase in pH, which in turn produces the change of colony color from yellow to reddish.  相似文献

Induction of conidiation in Alternaria tomato (Cke.) Weber and Helminthosporium oryzae V. Breda de Haan was repeatedly controlled by ulternating doses of near ultraviolet light and blue light. These fungi did not form conidia in total darkness. However, when dark-grown colonies were exposed to near altraviolet light, conidiophore formation was induced, and conidia were then produced upon return to darkness. Conidiation was inhibited by blue light applied immediately after near ultraviolet light. Furthermore, the final response depended on the last kind of light received. The action spectrum for induction of conidiation showed a peak near 300 nm with a shoulder between 310 nm and 330 nm, as is common in photoconidiation of various fungi. The action spectrum for the reversion of induction of conidiation showed a peak near 447 nm with a shoulder between 400 nm and 440 nm, one minor peak around 478 nm and another one near 385 nm. This is very similar to the spectrum for the ubiquitous blue light receptor (cryptochrome).  相似文献

The effect of temperature and light conditions (spectral quality, intensity and photoperiod) on germination, development and conidiation of tomato powdery mildew (Oidium neolycopersici) on the highly susceptible tomato cv. Amateur were studied. Conidia germinated across the whole range of tested temperatures (10–35°C); however, at the end‐point temperatures, germination was strongly limited. At temperatures slightly lower than optimum (20–25°C), mycelial development and time of appearance of the first conidiophores was delayed. Conidiation occurred within the range of 15–25°C, however was most intense between 20–25°C. Pathogen development was also markedly influenced by the light conditions. Conidiation and mycelium development was greatest at light intensities of approximately 60 μmol/m2 per second. At lower intensities, pathogen development was delayed, and in the dark, conidiation was completely inhibited. A dark period of 24 h after inoculation had no stimulatory effect on later mycelium development. However, 12 h of light after inoculation, followed by continuous dark, resulted in delayed mycelium development and total restriction of pathogen conidiation (evaluated 8 days postinoculation). When a longer dark period (4 days) was followed by normal photoperiod (12 h/12 h light/dark), mycelium development accelerated and the pathogen sporulated normally. When only inoculated leaf was covered with aluminium foil while whole plant was placed in photoperiod 12 h/12 h, the intensive mycelium development and slight subsequent sporulation on covered leaf was recorded.  相似文献

Summary Conidiation of Aspergillus niger was studied in carbon-limited and nitrogen-limited chemostat culture. Under citrate-limitation conidiation intensity varied inversely with dilution rate. Conidiophores were less complex than in aerial conidiation and at high dilution rates conidia occasionally developed from modified hyphal tips. Conidiation was difficult to achieve under glucose-limitation. At the low dilution rates that allowed limited conidiation steady state could not be maintained due to onset of autolysis. At higher dilution rates when steady state was readily obtained conidiation did not occur. The maximum yield constants under citrate-limitation and glucose-limitation were respectively 0.145 and 0.4 mg dry weight/mg substrate, while the relative specific maintenance values were 0.045 and 0.018 mg substrate/mg dry weight/h. Under ammonium-limitation with citrate as the carbon source there was no conidiation. When nitrate became the limiting nitrogen source conidiophore initiation occurred but biomass production was low and wash-out occurred at D=0.034 h-1.  相似文献

Penicillium cyclopium growing in a surface culture with 2-ketoglutarate or glutamate as a sole carbon source produced ethylene in two phases. The first peak of ethylene production (EP 1) was associated with aerial mycelium growth whereas the second peak of ethylene production (EP 2) occurred with formation and maturation of conidia. Conidiation was induced by blue light between 120 and 172 h after the culture was started and depended on the presence of a carbon source at the stage of conidiophore initiation. Exogenous phosphate content dropper rapidly before the onset of conidiation. The EP 2 was connected with conidiation via this drop. Addition of phosphate prior to the conidiophore initiation and during conidiation inhibited EP 2 without affecting conidiation, but conidia lacked a green pigment and their germination ability decreased by 905. Exogenous ethylene did not restore normal development. The EP 2 in asporogenic cultures was evoked by incubation in the dark and by phosphate removal. The EP 2 and conidiation were accompanied by an increased oxygen consumption. The EP 1 yield of ethylene depended only on biomass growth and was unaffected by any treatment mentioned above.  相似文献

Conidiation in Botrytis fabae was stimulated by irradiating 1 to 3 day old, but not 4 to 5 day old mycelium. Three cycles of 12 h irradiation + 12 h darkness stimulated the production of about twice as many spores compared with only 12 h irradiation. At 18°C all the spores had been produced within 3 days but not within 2 days from the start of irradiation. Near-u.v. irradiation at wavelengths of 375–400 nm induced most sporulation. Red light at 600–650 nm also stimulated conidiation but irradiation at other wavelengths from 300 to 700 nm was ineffective. Fewer conidia were produced when the fungus was kept in darkness at 4°C between periods of irradiaton at 18°C compared with continuous 18°C. The optimum osmotic potential of the culture, medium for conidiation was about-27 bar although more mycelium grew at even lower osmotic potentials. Abundant spore production occurred when the fungus was grown in media with a wide range of pH values.  相似文献

The filamentous fungus Penicillium cyclopium conidiates in the presence of calcium ions in submerged culture without nutrient limitation according to a precisely timed program. Conidiation could be prematurely induced in a nutritionally sufficient medium which had previously supported growth, suggesting that a metabolite which influenced the process was produced. A diterpenoid with conidiation-inducing activity, conidiogenone, was purified from the culture medium, along with conidiogenol, a putative derivative with delayed activity. Contrary to previous thought, the presence of calcium was demonstrated to only decrease the threshold concentration of conidiogenone required for the induction to proceed. In light of these results, a mechanism of conidiation induction is presented according to which the mycelium produces a conidiation inducer (conidiogenone) that accumulates extracellularly. When a threshold concentration is reached, induction likely takes place by interaction with a specific cellular receptor. The results indicate that conidiogenone is both sufficient and necessary to induce conidiation.  相似文献

Conidiation of the entomopathogenic fungus Metarhizium acridum on agar media was investigated. M. acridum CQMa102 exhibits two different conidiation patterns on agar media: normal conidiation in which conidia are formed on extended hyphae and microcycle conidiation in which conidiation occurs directly after conidia germination. Microcycle conidiation resulted in a mass of conidia produced via budding by accelerated development at the inoculation site. The mean total conidial yield (conidiation at day 10) was 4–5-fold greater after microcycle conidiation than during normal conidiation. Insect pathology assays indicated that microcycle conidia produced on SYA agar were as effective as normal aerial conidia against the locust. Ultraviolet (UV)-resistance tests showed no significant differences between the two types of cell propagules. However, microcycle conidia were more heat resistant than normal aerial conidia, and accumulated higher levels of trehalose in response to heat induction compared to normal aerial conidia.  相似文献

Derivatives of cyclic-3′,5′-adenosine monophosphate, N6,2′-O-dibutyryl-cAMP, 8-(chlorophenylthio)-cAMP and 8-bromoadenosine-cAMP added at 0.1–10 μmol/L concentrations into the growth medium have markedly stimulated conidiation inTrichoderma viride. Their stimulatory effect on conidiation was best observed in colonies that were constantly kept in the dark but was also marked in colonies illuminated by sub-saturating doses of light. The relative stimulation of conidiation depended not only on the concentration of exogenous cyclic nucleotides but also on the concentration of glucose in the medium. It was more pronounced in glucose-rich media than in media where the concentration of the sugar was low. Concentrations of cAMP analogues equal to 50 μmol/L and higher inhibited the conidiation.  相似文献

For the purpose of studying the mechanism of conidiogenesis in Piricularia oryzae, methods were developed to separate the phase of mycelial growth from that of conidiation and also to evaluate them quantitatively.

When P. oryzae was grown on media with low carbon : nitrogen ratio and high nitrogen concentration, conidiation did not take place, in spite of its vegetative growth. Conidiation occurred in a very short period of time when the above mycelia were replaced on nutritionally poor media. Cellophane membrane was overlaid on solid medium and conidia were spread uniformly. Evenly grown mycelial mat which could be easily transferred onto the post-culture medium was obtained. As the preculture medium, MSA medium with carbon: nitrogen ratio of 6.3 and nitrogen concentration of 1.5 g/liter was used. The evenly grown mycelial mat was cut into small square mats of 1.44 cm2 each and the small square mycelial mats were transferred onto the post-culture medium together with the cellophane membrane. The conidiation took place in the post-culture and the vegetative growth in the preculture and the conidiation in the post-culture could be observed separately and quantitatively.

Conidiation did not occur at all in the preculture and the degree of conidiation which took place in the post-culture varied according to the precultural conditions. This means that it is a certain state of physiological condition in the preculture which determines the degree of conidiation in the post-culture. The authers designated this state of physiological condition as the “latent activity of conidiation” (LAC). For the purpose of the quantitative estimation of this activity, we expressed LAC in terms of the degree of conidiation in the post-culture under a defined cultural condition. The LAC was subject to change very easily, declined rapidly and disappeared upon prolonged preculture. Only young mycelia showed to have this activity.

The influences of the precultural condition on the development of the LAC and vegetative growth were generally parallel. However, the LAC was generally more sensitive to the environmental condition than the vegetative growth, especially to the temperature change.

The conidia formed were uniform in size and had high rate of germination. Several strains of P. oryzae tested showed very similar behavior.  相似文献

Conidiation in Trichoderma has been demonstrated to be favoured by a low ambient pH and more recently PacC (Pac1) mediated pH-regulation has been implicated in the control of conidiation. In this study, ambient pH effects on conidiation were investigated in three isolates (Trichoderma hamatum, Trichoderma atroviride and Trichoderma pleuroticola) exposed to a single blue-light burst or to mycelial injury. Disks of conidiation were observed for T. atroviride in response to a single light exposure, which clearly demonstrates that all cells are potentially competent for photoconidiation. Previous studies have suggested T. hamatum does not conidiate in response to mycelial injury, however, in this study a clear injury response was observed from pH 2.8 to 3.2. T. pleuroticola displayed three distinct pH-dependent colony morphologies from pH 2.8 to 5.2. Conidiation was strictly low pH-dependent on buffered media and observed at all pH values on unbuffered media. The dependence of the conidial phenotype on the buffering state of the medium rather than the pH per se, was unexpected as it has been suggested that conidiation is PacC regulated. Conversely, excretion of an anthraquinone was strictly pH-dependent regardless of the buffering state. These studies highlight the complexity of ambient pH effects on Trichoderma spp. and demonstrate a need to widen the scope of research to multiple species.  相似文献

Conidiation is important in the life cycles of mitosporic fungi for survival and transmission. A full-length cDNA of one gene named CMCPS1 encoding L-arginine-specific carbamoyl-phosphate synthase was obtained from Coniothyrium minitans, a sclerotial parasite of the plant pathogenic fungus Sclerotinia sclerotiorum. T-DNA insertional disruption of CMCPS1 resulted in conidiation deficiency of mutant ZS-1T2029, and this was confirmed with the RNAi technique. The phenotype was restored by complementation with L-arginine, and the effect of L-arginine on conidiation may be mediated by nitric oxide, which is catalyzed by nitric oxide synthase (NOS). Conidiation of ZS-1T2029 was restored by sodium nitroprussiate, a NO donor; and conidiation of wild type strain ZS-1 could be suppressed by L-NAME, an inhibitor of NOS. The highest amount of NO in mycelia was detected at an early stage of conidiation (72 hpi) in liquid shake culture medium. Staining with the NO-sensitive fluorescent probe, DAF-FM DA, gave strong fluorescent signals in primordia and young pycnidia. This work presents the first report that L-arginine is involved in conidiation of C. minitans, and the possibility of L-arginine-derived nitric oxide-mediated conidiation among fungi and possible modes of action are discussed.  相似文献

Summary The production of - and -galactosidases, proteinase and -amylase and also conidiation of Aspergillus oryzae were examined in liquid soybean meal culture. In a culture of soybean meal only, conidiation of the fungus was not induced and the production of the enzymes was not significant, although fungal growth was abundant. When phosphate was added to the medium at concentrations above 0.2 M, enzyme production was significantly increased and the cells formed conidiophores after enzyme production had attained maximum level. Increase in production of galactosidases was the most marked.K- or Na-salts other than phosphate were not effective stimulants for enzyme production, while conidiation was not induced under these growth conditions. Conidiation and production of enzymes were repressed by the addition of glucose or casamino acids to the soybean meal medium containing KH2PO4.Conidiation and enzyme production were also studied in modified Czapek media in which sucrose was replaced by other carbon sources. Lactose, lactulose, melibiose and polysaccharides composed of galactosyl linkage such as arabinogalactan induced both conidiation and production of the enzymes.  相似文献

Nonsteroid antiinflammatory agents (NSAIA's) (inhibitors of cyclooxygenase and lipoxygenase) of several structural series inhibited growth ofTrichoderma viride. The most potent growth inhibitors were indomethacin and its derivative repanidal which inhibited in the range of 0.1 mmol/L. The weakest inhibitors were acetylosalicylic acid and lysine salicylate which exerted only a weak effect at concentrations above 1 mmol/L. The inhibition of growth was accompanied by a stimulation of conidiation in the dark. A light pulse increased the efficiency of these drugs to stimulate the conidiation.Saccharomyces cerevisiae was as sensitive to NSAIA's asT. viride whileBotrytis cinerea was less sensitive. The results indicate that arachidonate metabolism may play a role in the growth of fungi and may participate also in the process of conidiation.  相似文献

After a short irradiation at 366 nm with 200 lx, the intensity of conidiation ofTrichoderma viride colonies grown in the dark increased for the first 10 s proportionally with time. The increase slowed down after 10 s—6 min of exposure and after 10–60 min of irradiation the conidiation intensity began to decrease. When photo-induced by daylight, the conidiation started at a high rate after 25 h and persisted even after 48 h. The conidiation had no circadian character and its periodicity depended on the periodicity of photo-induction. Its intensity was also influenced by the carbon sources used, a maximum being reached with glucose (1–2 %). Higher glucose concentrations inhibited conidiation but had no influence on growth of colonies.  相似文献

Effects of respiratory inhibitors on the circadian clock, respiratory activity, and ATP content were examined in Neurospora crassa. All inhibitors, potassium cyanide, sodium azide, antimycin A, and carbonyl cyanide m-chlorophenyl hydrazone (CCCP), shifted the phase of the conidiation rhythm. All the phase response curves were similar and resembled that for cycloheximide, but were different from the phase response curve for light. Phase shifting by azide and CCCP was proportional to the lowering of respiratory activity and ATP content, but such a correlation was not observed for cyanide and antimycin A. In particular, cyanide at a concentration of 0.5 millimolar completely depleted ATP of the cultures but did not significantly shift their phase. Their results suggest that large shifts caused by these inhibitors are not due to a decrease in energy from respiratory activity.  相似文献

Asexual reproduction (conidiation) in Aspergillus nidulans is induced by environmental signals like exposure to air or nutrient starvation, and depends on brlA gene activation. The study of 'fluffy' mutants showing delayed asexual development and reduced brlA expression has defined the fluG pathway, involved in regulation of this differentiation process. Genetic characterization of a 'fluffy' mutant identified tmpA as a new gene involved in regulation of conidiation. TmpA defines a new family of putative transmembrane proteins of unknown function, widespread in filamentous fungi and plants, with homologues showing similarity to non-ribosomal peptide synthetases. The deletion of tmpA resulted in decreased brlA expression and conidiation in air-exposed colonies. This defect was suppressed when DeltatmpA mutants were grown next to wild-type or DeltafluG mutant colonies, even without direct contact between hyphae. In liquid culture, tmpA was essential for conidiation induced by nitrogen but not by carbon starvation, whereas the overexpression of different tmpA tagged alleles resulted in conidiation. The overexpression of fluG-induced conidiation independently of tmpA and DeltatmpADeltafluG double mutants showed an additive 'fluffy' phenotype, indicating that tmpA and fluG regulate asexual sporulation through different pathways. TmpA and its homologues appear to have diverged from the ferric reductase family, retaining overall transmembrane architecture, NAD(P), flavin adenine dinucleotide (FAD) and possibly haem-binding domains. Based on our results, we propose that TmpA is a membrane oxidoreductase involved in the synthesis of a developmental signal.  相似文献
