13C nuclear magnetic resonance spectroscopy (NMR) has been used to study the metabolic flux of carbon through the intracellular pools of the isomeric hexitols d-altritol and d-mannitol in Himanthalia elongata. Natural abundance 13C NMR spectra of freshly collected plant material showed altritol as the dominant intracellular low-molecular-weight organic solute, with mannitol present at less than 20% of the altritol concentration. Plant material incubated in seawater medium with added 13C-enriched bicarbonate showed a rapid increase in the 13C signal due to mannitol, with an increase of more than 12-fold in under 48 h, in contrast to the altritol signal, which increased by approximately 2-fold over the same period. The intracellular mannitol signal decreased rapidly when 13C-enriched plant material was transferred to a non-enriched medium, while altritol showed a slower decline. These results are consistent with previous observations on the effects of salinity on the intracellular hexitol pools of H. elongata, suggesting that mannitol is more rapidly metabolised than altritol. Estimates of the half-time for tracer exchange support this view, with a half-time for the turnover of altritol (> 400 h) which is an order of magnitude greater than that for mannitol turnover (≈ 50h).  相似文献

Natural abundance 13C NMR spectroscopy has shown that altritol (talitol) is a major constituent within vegetative and reproductive tissues of Himanthalia elongata. Quantitative GC studies have confirmed that altritol concentrations are 80–160% higher than for the hexitol mannitol (which occurs throughout the Phaeophyta). The present study is the first comprehensive phylogeny of the class of the Phaeophyceae. For 67 species representing all orders of the class, the sequences of the 3'-end of the small and the 5'-end of the large subunit nrRNA genes were aligned and analysed. A further analysis based on sequences of the 3'-end of the small and of the complete sequences of the 28S gene of the large subunits was also performed, but for only eight taxa. In both analyses, Tribonema marinum (Xanthophyceae) was used as outgroup. The analyses showed the Dictyotales as diverging first, followed by the Sphacelariales, then the Syringodermatales. Most of the orders currently accepted were confirmed as monophyletic groups but the Laminariales and Tilopteridales remained polyphyletic. The relationships of the remaining orders to each other were not resolved with the present data set. Ascoseira, included for the first time in a molecular study, appeared as a separate lineage without any clear relationship with other algae possessing conceptacles (Splachnidium and Fucales). Algae with stellate plastids, never studied in a global context, were polyphyletic; this result is consistent with their plastid ultrastructure and is discussed in detail. As further result of the present study, the South African genus Bifurcariopsis appeared as the sister taxon of the North Atlantic genus Himanthalia, and Xiphophora appeared as the sister taxon of Hormosira rather than as a member of the Fucaceae; the taxonomic position of these genera is discussed.  相似文献

The swimming behaviour of Hormosira banksii spermatozoids wasstudied using videomicroscopy under different conditions, includingmovement on glass surfaces, on solvent droplets containing thespecies-specific gamete attractant at different concentrationsand in relation to eggs. The results indicate a chemotaxis mechanisminvolving phobic responses as well as a kinesis in which theturning rate is reduced upon stimulation by pheromone. This study deals with two range-extending brown algae from Tahitian coral reefs, Sargassum mangarevense and Turbinaria ornata; their alginate properties, mannitol and phenolic contents, antioxidant and antimicrobial activities were determined. Turbinaria ornata showed the richest alginate content with the highest extraction yield (19.2 ± 1.3% dw). Their alginates also exhibited the highest viscosity (50 ± 18 mPa.s), but the M:G ratios (mannuronic acid to glucuronic acid) of alginates (1.25–1.42) were similar in both species. Alginate yield displayed spatial variations, but no significant seasonal changes. The highest mannitol content was found in S. mangarevense (12.2 ± 2.1% dw) during the austral winter. With respect to other tropical Fucales, both algae exhibited also a high phenolic content (2.45–2.85% dw) with significant spatio-temporal variations. Furthermore, high antioxidant activity and activity against Staphylococcus aureus were also detected in extracts. According to these preliminary results, these two range-extending algae are of key interest in numerous industrial areas.  相似文献

There is considerable debate about whether stability (e.g. inertia) of an assemblage, or of individuals in an assemblage, is positively associated with the number of species or whether there are idiosyncratic effects of particular species. We assessed the general model that the loss of an individual alga, caused by trampling, is greater in monospecific than in multi-species stands but that the responses of algae are idiosyncratic, depending on the morphology of the species. The experiment was done on conspicuous and dominant algae with different morphology on temperate Australian rocky shores: the fucalean algae Hormosira banksii and Sargassum sp. and the coralline alga Corallina officinalis. We assessed the relative and interactive effects of the extent of trampling (number of paths) and the localised intensity of trampling (number of travels per path) on the three algae. The number of paths trampled (the extent of disturbance) had more impact on each alga than the number of times paths were travelled (the intensity of disturbance). As predicted, H. banksii was most susceptible to trampling at each level than were the coarser algae Sargassum sp. and C. officinalis. There was a consistent trend for each alga to be more inert to trampling when in the presence of the other two species than when in monospecific stands, but this was only statistically significant (P < 0.05) for the softer alga H. banksii. The responses of H. banksii and Sargassum sp. to disturbance seemed, in many cases, to be due to the presence of C. officinalis rather than to "diversity" per se. The relationship between the number of species and stability is complex in intertidal habitats, depending on the species and the combinations of species with which it grows.  相似文献

Dictyotophycidae is a subclass of brown algae containing 395 species that are distributed worldwide. A complete plastid (chloroplast) genome (ptDNA or cpDNA) had not previously been sequenced from this group. In this study, the complete plastid genome of Dictyopteris divaricata (Okamura) Okamura (Dictyotales, Phaeophyceae) was characterized and compared to other brown algal ptDNAs. This plastid genome was 126,099 bp in size with two inverted repeats (IRs) of 6026 bp. The D. divaricata IRs contained rpl21, making its IRs larger than representatives from the orders Fucales and Laminariales, but was smaller than that from Ectocarpales. The G + C content of D. divaricata (31.19%) was the highest of the known ptDNAs of brown algae (28.94–31.05%). Two protein-coding genes, rbcR and rpl32, were present in ptDNAs of Laminariales, Ectocarpales (Ectocarpus siliculosus), and Fucales (LEF) but were absent in D. divaricata. Reduced intergenic space (13.11%) and eight pairs of overlapping genes in D. divaricata ptDNA made it the most compact plastid genome in brown algae so far. The architecture of D. divaricata ptDNA showed higher similarity to that of Laminariales compared with Fucales and Ectocarpales. The difference in general features, gene content, and architecture among the ptDNAs of D. divaricata and LEF clade revealed the diversity and evolutionary trends of plastid genomes in brown algae.  相似文献

Light microscope and fine structural studies of the gametes of Hormosira banksii show the antherozoids as typical Fucales biflagellate gametes with a posterior whiplash flagellum and an anterior flagellum with mastigonemes. The oospheres are enclosed in a plasma membrane, are highly vacuolate and contain abundant phenolic inclusions.

Antherozoids attach to the surface of the oospheres by their anterior flagellum at conjugation. Following fertilization Golgi derived vesicles underlying the plasma membrane decrease in electron density and an exterior surface coat appears outside this membrane. This may be a “fertilization barrier” which prevents further penetration by other male gametes.

Subsequently a fibrous layer appears which develops into a cell wall around the developing zygote. Histochemical studies show the cell wall of these young zygotes is probably made up of alginic acid. Fine structural and histochemical examination of the young zygotes suggest that whereas there is no preformed adhesive in the oosphere, large reserves of lipid and polysaccharide provide materials necessary for synthesis of wall material and the potential for adhesive production following fertilization.  相似文献

Hybridization is an important evolutionary process, which can have significant effects on biodiversity. While hybridization is well known in plants, less is known about the prevalence of hybridization in other kingdoms. Hybridization in the field has been confirmed in brown algae in a few cases, mainly in the northern hemisphere genus Fucus. Putative hybrids have been found in the New Zealand endemic species Carpophyllum angustifolium and Carpophyllum maschalocarpum. We used nuclear‐encoded molecular data (ITS2) and morphometrics to confirm hybridization between C. angustifolium and C. maschalocarpum. Putative hybrid thalli were collected that had heterozygous ITS2, each copy corresponding to one of the parental species from that population. Morphological analysis also showed that the three classes (two parental species and hybrids) were easily distinguishable in these populations. It was found that the hybrids had an intermediate morphology to the parent species. Some individuals with C. angustifolium morphology had hybrid ITS2 ribotypes suggestive of backcrossing between the hybrids and C. angustifolium. Our data reveal another case of hybridization within the Fucales and suggests that further research on how these species remain separate is needed.  相似文献

1984~2000年间,对娘子关泉域的藻类植物进行了15次采集,共采得藻类植物标本200余号。经鉴定,有107种,隶属7门,32科,49属。其中,以绿藻门的种类最多,有13科,19属,45种。其次是裸藻门,有2科,7属,26种。排在最后的是轮藻门,有1科,1属,3种。群落主要包括8类,即刚毛藻-红毛菜群落(Cladophora Bangia community),刚毛藻群落(Cladophora community),拖拉藻-轮藻群落(Chara Thorea community),弯枝藻-拖拉藻群落(Composopogon-Thorea community),水绵-双星藻群落(Spirogyra Zygnema community),无隔藻群落(Vaucheria community),串珠藻群落(Batrachospermum community)和泡状饶氏藻群落(Jaoa community)。其中,有的群落类型已趋于消失或消失,许多种类已成为濒危物种,其原因主要是人为因素造成藻类植物生存环境的改变或破坏。本文也提出了相应的保护对策。  相似文献

The metabolism of [1-13C]glucose in the vegetative mycelium of the ectomycorrhizal ascomycete Tuber borchii was studied in order to characterize the biochemical pathways for the assimilation of glucose and amino acid biosynthesis. The pathways were characterized using nuclear magnetic resonance spectroscopy in conjunction with [1-13C]glucose labeling. The enzymes of mannitol cycle and ammonium assimilation were also evaluated. The majority of the 13C label was incorporated into mannitol and this polyol was formed via a direct route from absorbed glucose. Amino acid biosynthesis was also an important sink of assimilated carbon and 13C was mainly incorporated into alanine and glutamate. From this intramolecular 13C enrichment, it is concluded that pyruvate, arising from [1-13C]glucose catabolism, was used by alanine aminotransferase, pyruvate dehydrogenase and pyruvate carboxylase before entering the Krebs cycle. The transfer of 13C-labeled mycelium on [12C]glucose showed that mannitol, alanine, and glutamate carbon were used to synthesize glutamine and arginine that likely play a storage role.  相似文献

Fifteen species from different genera of blue-green algae have been examined for their formation of plastocyanin (PC) and cytochrome c-553 (cyt c-553) in high or low Cu media. In addition to species which contain only cyt c-553 and those which completely exchange their cyt c-553 by PC, a new regulatory type was detected in which this exchange was incomplete. By comparing different species, it could be shown that either this incomplete exchange of cyt c-553 by PC as well as lack of PC in some other blue-green algae is not caused by restricted Cu uptake but is due to different biosynthetic and regulatory properties. Occurrence of PC and cyt c-553 cannot be used as a taxonomic criterium to classify blue-green algae. However, formation of either one or both of these redox components fits well into a line of evolution of the photosynthetic apparatus from the blue-green algae via green algae to higher plants.Abbreviations PC plastocyanin; cyt c-553, cytochrome c-553  相似文献

Application of phylogenetic species recognition to morphologically recognized species in the genera Cystoseira Agardh and Halidrys Lyngbye on North American west coasts revealed little genetic variation despite a remarkable degree of morphological variation currently used to recognize and delineate species. Whereas morphological characteristics allow recognition of two genera, four morphological species and three informal forms, maximum genetic variation among them was similar to that characteristic of the intraspecific level in European congeners and other Fucales. Among morphological species and forms, nucleotide variation in a combined 26S (large subunit (LSU)) and internal transcribed spacer (ITS) ribosomal DNA analysis was below 3% while it was 1% or less for the RUBISCO spacer of the chloroplast DNA. Comparison of the LSU data to available data for European congeners showed that the genera Cystoseira and Halidrys are not monophyletic and that the previously recognized Cystoseiraceae should be included within the family Sargassaceae. These observations suggest that the current taxonomy for the Sargassaceae fails to reflect evolutionary history because Atlantic and Pacific Cystoseira and Halidrys appear to have arrived at similar morphologies independently. Our results indicate a comparatively recent establishment on the west coast of North America of a sargassacean progenitor whose descendant taxa have experienced limited genetic divergence and are characterized by a high capacity for phenotypic variation despite their overall genetic similarity.  相似文献

Experiments were performed to determine whether mannitol occurs as a native constituent of marine and freshwater Rhodophyceae. Those red algae which had previously been reported to contain mannitol were tested. In none of these species could mannitol or any other hexitol be detected, either as 14C-assimilate after photosynthetic assimilation of 14C from H14CO3 or in trace amounts of the EtOH-soluble fraction. Attempts to qualify the action of a specific mannitol synthesizing enzyme (mannitol-1-phosphate dehydrogenase, EC 1.1.1.17) also failed. Though mannitol [14C] is taken up when exogenously supplied by a seawater medium, less than 5% of 14C from mannitol [14C] taken up after 3 hr is recovered in other compounds. Mannitol is therefore not regarded as a natural metabolite (assimilate) of Rhodophyceae.  相似文献

Of 70 pure microbial cultures isolated from aquatic habitats, soil, and air according to the ability to lyse live blue-green algae, 62 were fungi representing the genera Acremonium, Emericellopsis, and Verticillium. Algal-lysing fungi were isolated from all habitat types sampled. The remaining isolates comprised four bacteria and four streptomycetes. All isolates lysed Anabaena flos-aquae and, in most cases, several other filamentous and unicellular blue-green algae. The fungi generally showed greater activity than most other isolates towards a wider range of susceptible algae, including green algae in some cases. Acremonium and Emericellopsis isolates, but not Verticillium, also inhibited the growth of blue-green algae and gram-positive bacteria, but did not lyse the latter. Lysis of blue green algae by Acremonium and Emericellopsis spp. was associated with the formation of diffusible heat-stable extracellular factors which, evidence suggests, could be cephalosporin antibiotic(s). Blue-green algae were also lysed by pure cephalosporin C. The frequent isolation of lytic fungi from algal habitats suggests a possible natural algal-destroying role for such fungi, which might be exploitable for algal bloom control.  相似文献

Global warming and the loss of sea ice threaten to alter patterns of productivity in arctic marine ecosystems because of a likely decline in primary productivity by sea ice algae. Estimates of the contribution of ice algae to total primary production range widely, from just 3 to >50%, and the importance of ice algae to higher trophic levels remains unknown. To help answer this question, we investigated a novel approach to food web studies by combining the two established methods of stable isotope analysis and fatty acid (FA) analysis--we determined the C isotopic composition of individual diatom FA and traced these biomarkers in consumers. Samples were collected near Barrow, Alaska and included ice algae, pelagic phytoplankton, zooplankton, fish, seabirds, pinnipeds and cetaceans. Ice algae and pelagic phytoplankton had distinctive overall FA signatures and clear differences in delta(13)C for two specific diatom FA biomarkers: 16:4n-1 (-24.0+/-2.4 and -30.7+/-0.8 per thousand, respectively) and 20:5n-3 (-18.3+/-2.0 and -26.9+/-0.7 per thousand, respectively). Nearly all delta(13)C values of these two FA in consumers fell between the two stable isotopic end members. A mass balance equation indicated that FA material derived from ice algae, compared to pelagic diatoms, averaged 71% (44-107%) in consumers based on delta(13)C values of 16:4n-1, but only 24% (0-61%) based on 20:5n-3. Our estimates derived from 16:4n-1, which is produced only by diatoms, probably best represented the contribution of ice algae relative to pelagic diatoms. However, many types of algae produce 20:5n-3, so the lower value derived from it likely represented a more realistic estimate of the proportion of ice algae material relative to all other types of phytoplankton. These preliminary results demonstrate the potential value of compound-specific isotope analysis of marine lipids to trace C flow through marine food webs and provide a foundation for future work.  相似文献

The structure and dynamics of periphytic algae in Jinyang Lake, the largest man-made lake in Taiyuan, Shanxi, have been studied by examining the periphytic algae samples collected from four sampling stations, during March 2006 to February 2007. In total, 227 species of periphytic algae (including varieties), belonging to three divisions, 27 families, 62 genera, have been identified. Diatoms were the dominant periphytic algae group. Taken the Shannon–Weaver, Margalef and Simpson diversity index together, it showed obvious temporal and spatial dynamics among different seasons and different sampling stations. The highest periphytic community structure evenness appeared at sampling station I in spring, summer and autumn, while at sampling station III in winter. The average biomass of periphytic algae varied from seasons. Our data showed that major factors affecting the periphytic algae in Jinyang Lake was water temperature, nutrient levels as well as human activities. In addition, the main effect caused by thermal power plant has been discussed.  相似文献

Summary A complex system of growth inhibition is present in the green algae (Volvocaceae). Inhibitors are found in the culture filtrates of some genera which limit their own growth (autoinhibition) while others in the family produce inhibitors which check the growth of other genera (heteroinhibition).
