Warm temperatures (35°C day/30°C night) which inhibittuberization in potato (Solanum tuberosum L., cv. Sebago) increasedgibberellin activity in crude extracts from buds, but not frommature leaves, as determined by the lettuce hypocotyl bioassay.Changes in the growth of tubers and stolons indicate the occurrenceof basipetal movement of GA3 applied to the terminal bud ora mature leaf. 14C labelling from GA3 or mevalonic acid injectedjust below the terminal bud was recovered in the lower shoot,stolons and tubers, but the amount transported was greater atcool temperatures (20/15°C). Potato shoots were propagated in vitro by placing nodes fromsprouted tubers on Murashige and Skoog type medium without hormones.The vigour of growth and the rate of node production increasedwith both day-length and temperature over the ranges 8–24h and 15–25 °C respectively. Propagation rates ofup to x 10 per month were obtained. In vitro plantlets spontaneouslyformed roots either in agar or liquid cultures. Plantlets leftin the culture jars for 3–4 months eventually senescedand formed small tubers in 16 and 24 h day-lengths. [14C]Sucrose, [14C]glucose and [14C]fructose were introducedinto potato tubers held at 10 °C and the redistributionof label chased over a 65 d period in storage. Respiratory losseswere identical in all treatments, as was the partitioning of14C between soluble and insoluble forms. Sucrose was the predominantlabelled sugar in the tubers after 20 h, regardless of the original[14C]sugar introduced, and was loaded and distributed throughoutthe tubers by the internal phloem system. After 20 h the proportionsof labelled sugars bore no relationship to those of the unlabelledendogenous sugars. However, with time the percentage of 14Cin sucrose fell while that in glucose increased and by 65 dthe proportions of the labelled sugars more closely resembledthe endogenous pools. Fructose represented a consistently lowproportion of both the labelled and unlabelled sugars. By 21d a considerable proportion of the soluble 14C had been convertedto starch (approx. 25% of the total tuber 14C), this value remainingrelatively constant for the remainder of the storage period.Sprouts which formed on the tubers contained up to 6% of thetotal tuber 14C but less than 0.2% of the tuber dry matter.It is suggested that the bulk of the translocated [14C]sucroseentered the symplast and exchanged slowly with the bulk of thesugars in the storage cell vacuoles. Potato plants (Solanum tuberosum L.) were grown in water culturein a controlled environment. Cooling (+8°C) of individualtubers decreased their growth rates and increased the growthrates of non-cooled tubers of the same plant. The carbohydrateconcentration in non-cooled and cooled tubers did not differsignificantly, but 14C-import from labelled photosynthate waslower in cooled than in non-cooled tubers. The markedly lowerconversion rate of ethanol-soluble 14C to starch in cooled,in comparison to non-cooled tubers, was not associated withsignificant differences in the in vitro activities of starchsynthase, ADPG-pyrophosphorylase and starch phosphorylase understandard assay conditions (+30°C). However, the Q10-valuesof the enzymes differed in vitro in the temperature range between30°C and 8°C, leading to a marked decrease in the activityratio of ADPG-pyrophosphorylase/starch phosphorylase in cooledtubers. In tubers differing in growth rates without manipulation, 14d after tuber initiation significant positive correlations werefound between 14C-concentration of tuber tissue and the in vitroactivities of starch synthase and ADPG-pyrophosphorylase anda significant negative correlation between 14C-concentrationand starch phosphorylase. In contrast, in tubers which wereanalysed 5 d after initiation, there were only small differencesbetween tubers in growth rate, 14C import and the activity ratioADPG-pyrophosphorylase/starch phosphorylase. From various directand indirect evidence it is concluded that the growth rate ofindividual tubers, and thus the sink strength, is at least inpart controlled by the activity of starch synthesizing enzymes. Fusarium moniliforme secreted macerating enzymes in liquid mediaonly when these contained certain natural extracts, pectic substances,or galacturonic acid. Apple extract was unsuitable for enzymesecretion and also inhibited enzyme secretion in synthetic mediaotherwise suitable. Protopectinase activity of solutions was highest in the pH range8·0–9·0, was rapidly lost at temperaturesabove 50–60° C., and was reduced by concentrationsof phosphate higher than 0·02 M. The enzyme was partiallypurified by precipitation in 60 per cent. acetone at pH 6·0. Protopectinase solutions also contained an enzyme which reducedthe viscosity of solutions of various pectic substances. Theproperties of this enzyme were, in general, similar to thoseof protopectinase. When activity of enzyme solutions was measured by the liberationof reducing groups, pectate solutions were more rapidly degradedthan were solutions of a high methoxyl pectin, particularlyin the early stages of the reaction. Paper chromatography ofthe products formed showed that pectate and pectin were degradedin different ways. Although the pathogen readily secreted protopectinase in potatoextract, potato tubers were not readily parasitized. The permeability to oxygen of the periderm of the potato tuberhas been measured during its development and storage. When expressedas ml O2, diffusing through 1 cm2 periderm in 1 sec under anO2, pressure of 1 atm, the values for freshly harvested tubersranged from a maximum of 2.4 x 10–4 in immature tubers,to less than 0.7 x 10–4 in tubers harvested mature someweeks after the death of the foliage. In every case, after storagefor up to 10 weeks at 10° C, the permeabilities, as expressedabove, were less than 0.7 x 10–4 (minimum values, c. 0.5x 10–4 It was shown that the oxygen deficit under theperiderm was unlikely to exceed 0.03 atm during development,and the oxygen status of the developing tuber should thus beadequate for the low oxygen-affinity oxidase system (if present)to function.  相似文献

The changes in the components of the ascorbateglutathione systemduring the storage of potato (Solanum tuberosum L. cv. Spunta)tubers for 40 weeks at both 3C and 9C were studied in relationto lipid peroxidation. The level of malondialdehyde was foundto be higher at 3C than at 9C throughout storage. Thus, lipidperoxidation, which is the main cause of membrane deterioration,was accelerated at the lower temperature. Catalase activityincreased throughout storage independently of temperature. Theascorbate content of tubers decreased during storage both at3C and at 9C, as in other ageing processes. However, ascorbateperoxidase activity reached a maximum after about 8 weeks ofstorage, then declined at 9C, but held a higher level at 3C.The dehydroascorbic content also reached a maximum after about8 weeks and was significantly higher in tubers stored at 3C.These findings indicate a greater utilization of ascorbate byascorbate peroxidase at the lower temperature. Ascorbate freeradical reductase, dehydroascorbate reductase and glutathionereductase, the enzymes involved in the regeneration of ascorbate,were not affected by temperature and remained quite unchangedthroughout storage. It can be concluded that the ascorbate systemis involved in the scavenging of the free-radicals responsiblefor lipid peroxidation in stored potato tubers, at least atlow temperatures and in the first period of storage. Second growth is an important physiological disorder of thepotato (Solanum tuberosum L.) plant. A model system to studysecond growth was developed using one-leaf cuttings. Photoperiod,temperature, decapitation and leaf removal treatments were carriedout on the plants from which the cuttings were taken and onthe cuttings themselves. Tuberized, one-leaf cuttings takenfrom moderately-induced plants and exposed to 35 °C afterleaf removal showed 95% second growth within 10 d after treatmentinitiation. Conditions that promoted second growth also reducedstarch and dry-matter content, even in tubers that did not developsecond growth. In potato plants fast and slow growing tubers develop on thesame plant. A hypothetical causality between tuber growth rateand tuber cell number was investigated by determining the tubercell number with the aid of an automatic counting procedure.Our data show a close correlation between tuber size and cellnumber over the whole range of tuber volumes considered (3–28cm3). If the influence of tuber size on cell number is eliminatedby means of a partial correlation analysis, the cell numberof the entire tuber is not significantly correlated with itsgrowth rate. An exclusive consideration of the smaller cells(10–30 µm) in the apical tuber region, where thecell division rate in potato tubers is highest, reveals a loosebut significant partial correlation to tuber growth rate (r= 0.383, P < 0.05). The growth rate of the slow growing tubers of any potato plantmay be enhanced by removing the fast growing tubers. In thefirst few days this enhanced growth rate is not due to a stimulationof cell division rate, but rather due to cell expansion. Effects of dehydration, storage temperature and humidificationon germination of Salix alba andS. matsudana seeds were studied.Newly released seeds showed 100% germination before and afterdehydration to 11–12% moisture content. Germination ofthe high vigour lot (100% initial normal germination) was notaffected by dehydration to 6.7% moisture content but germinationdecreased with further dehydration to 4.3%. The lower vigourlot (75% initial normal germination) was more susceptible todehydration and germination decreased following dehydrationto 6.7% moisture content. Dry seeds of both species survivedimmersion in liquid nitrogen without loss of viability. Thegermination of seeds stored with 9% moisture content decreasedto 35–40% in 5 months at -20°C or in 2 months at 5°C.However, at 25°C seeds entirely lost viability within 2weeks. Seeds showed improved performance when stored at -70°C> - 20°C > 5°C > 25°C and tolerated dehydrationto a moisture content in equilibrium with 15% relative humidity.Results suggest that they are orthodox in storage behaviouralthough they are short-lived. Humidification treatment of lowvigour seed lots resulted in a remarkable increase in germinationpercentage. The relationships between ethylene production, aminocyclopropane-1-carboxylicacid (ACC) content and ethylene-forming-enzyme (EFE) activityduring ageing and cold storage of rose flower petals (Rose hybridaL. cv. Gabriella) were investigated. During flower ageing at20 °C there was a climacteric rise in petal ethylene production,a parallel increase in ACC content, but a continuous decreasein EFE activity. Applied ACC increased petal ethylene productionc. 200-fold. During cold storage of flowers at 1 °C therewere parallel increases in petal ethylene production and ACCcontent, to levels greater than those reached in fresh flowersheld at 20 °C. EFE activity decreased during storage. Immediatelyafter cold-stored flowers were transferred to 20 °C ethyleneproduction and ACC levels were c. four times greater than infreshly cut flowers. These levels increased to maximum valuesof two to four times the maximum values reached during ageingof fresh, unstored, flowers. It was concluded that in rose petalsethylene synthesis is probably regulated by ACC levels and thatcold storage stimulates ethylene synthesis because it increasesthe levels of ACC in the petals. In vitro (mini) tubers were induced within 6–8 weeks inserially propagated potato shoot cultures by subculturing tomedium containing 2.0 mg 1–1 benzylaminopurine (BAP) and6 per cent sucrose in 8- and 24-h days. The effect of BAP inpromoting tubering was greater in short than in long days. Inshort days most of the tubers were formed above the agar, inlong days within the agar. Tubering was promoted less effectivelyby the addition of (2-chloroethyl)-trimethylammonium chloride(CCC) to the medium, but CCC reinforced the effect of BAP leadingto earlier tubering above the agar. Tubering eventually tookplace after 4—5 months on medium without hormones, soonerin short than in long days. Periods of short days and low temperaturesgiven to long-day cultures did not accelerate tubering. Abscisicacid had little effect on, and GA2 strongly inhibited, tubering.Tubering was also inhibited by sealing the culture vessels butnot if ethylene-absorbing agents were included. Seeds of lettuce (Lactuca sativa L.) and sunflower (Helianthusannuus L.) were stored hermetically at 35 °C with 11 differentmoisture contents between 1·3 and 6·9%, and between1·3 and 7·1% of fresh mass, respectively. Germinationand vigour (mean germination time, root length, seedling dryweight) were determined after storage for 0, 8, or 16 weeks(sunflower) or 0, 8, 16, or 48 weeks (lettuce) in these environmentsfollowed by various humidification treatments (to avoid imbibitioninjury). The range of seed storage moisture contents over whichdeterioration was minimized depended upon the criterion of deteriorationused, and varied somewhat between species. Comparison of theseranges for seeds stored for the longest durations showed thatfor some criteria seed performance was poorer (P < 0·05)at both the lowest and highest moisture contents investigatedthan at certain of the intermediate storage moisture contents(e.g, most rapid germination occurred in sunflower followingstorage at 2·2-4·7% moisture content), whereasfor other criteria all the drier storage moisture contents weresuperior to the more moist (e,g. greatest seedling growth occurredin sunflower following storage at 1·3-5·1% moisturecontent). But none of these results suggested that lettuce andsunflower seeds stored hermetically at 2·5-3·0%or 2·2-2·5% moisture content, respectively, wereless vigorous than at any other moisture content tested. Inboth species, these storage moisture contents are in equilibriumwith about 8-10% relative humidity (r.h.) at 20 °C, whichis similar to and indeed marginally less than the 10-13% r.h.recommended following earlier studies on the longevity of seedsin hermetic storage at much warmer temperatures. Thus, theseresults show no evidence that the optimum seed moisture contentfor storage increases with decrease in temperature, at leastover the range 35-65 °C, as has been suggested elsewhere.We conclude that the international recommendation for the long-termseed storage for genetic conservation at 5 ± 1% moisturecontent should not be revised upwardly, and that in situationswhere refrigeration cannot be provided storage at even lowermoisture contents is worthy of further investigation for thoseseeds in which desiccation at 20 °C to equilibrium at 10%r.h. results in moisture contents well below 5%.Copyright 1995,1999 Academic Press Helianthus annuus L., sunflower, Lactuca sativa L., lettuce, desiccation, seed storage, seed vigour  相似文献

In previous work, we demonstrated that there was an optimummoisture level for seed storage at a given temperature (Vertucciand Roos, 1990), and suggested, using thermodynamic considerations,that the optimum moisture content increased as the storage temperaturedecreased (Vertucci and Roos, 1993b). In this paper, we presentdata from a two year study of aging rates in pea (Pisum sativum)seeds supporting the hypothesis that the optimum moisture contentfor storage varies with temperature. Seed viability and vigourwere monitored during storage under dark or lighted conditionsat relative humidities between 1 and 90%, and temperatures between-5 and 65°C. The optimum moisture content varied from 0·015g H2O g-1 d.wt at 65°C to 0·101 g H2O g-1 d.wt at15°C under dark conditions and from 0·057 at 35°Cto 0·092 g H2O g-1 d.wt at -5°C under lighted conditions.Our results suggest that optimum moisture contents cannot beconsidered independently of temperature. Sixteen seed accessions of four Brassica species (B. cretica,B. incana, B. montana and B. oleracea) were examined following5-22 years' storage in the germplasm bank. Germinability andisozyme of seeds stored under long-term (-10 °C and 3% moisturecontent) and short-term (5% °C and 8% moisture content)storage conditions were compared. Long-term storage producedno deterioration and the ability to germinate was satisfactorilymaintained over 8-22 years. Short-term storage conditions maintainedgermination ability up to 10-12 years in all accessions of B.cretica and B. montana. However, seed multiplication might beessential every 10 years for some accessions of B. oleraceastored in this way. In the 8 to 9-year-old accessions of B.cretica, B. incana and B. montana, no significant differenceswere detected between conservation systems for germinabilityand frequency of seeds showing isozyme activity for seven enzymesystems (ACO, IDH, MDH, ME, PGI, PGM and 6-PGD). However, significantdifferences were found for the ADH enzyme system. Floral initiation in seedlings of Stylosanthes guianensis var.guianensis cv. Schofield grown at a photoperiod marginal forflowering (12–11.75 h) was promoted by a combination oflow day (25 °C) and low night (16 or 21 °C) temperatures,and completely inhibited by a 35 °C day temperature. Seeds (caryopses) of North American wild rice (Zizania palustrisvar. interior), a temperate aquatic grass, have been thoughtto require storage at low temperatures and high moisture contentsto preserve viability. The seeds are also deeply dormant atmaturity and require up to 6 months of stratification to breakdormancy. We report here that wild rice seeds can retain viabilityat moisture contents 30% (f. wt. basis) for up to 6 monthsat temperatures as high as 30 °C, and for at least 1 yearat temperatures below 20 °C. Dormancy is not broken at temperaturesabove 10 °C, but subsequent stratification requirementsare unaffected by prior warm storage. Cold storage is thereforenot required to maintain viability of wild rice seeds, but isnecessary to break dormancy. Hydrated wild rice seeds can befrozen to –10 °C without damage, but dormancy is notlost at subfreezing temperatures. These results provide newoptions for long-term storage of wild rice seeds. The effects of storage conditions on the germination of developingmuskmelon (Cucumis melo L.) seeds were tested to determine whetherafter-ripening is required to obtain maximum seed vigour. Seedswere harvested at 5 d intervals from 35 (immature) to 60 (fullymature) days after anthesis (DAA), washed, dried, and storedat water contents of 3·3 to 19% (dry weight basis) at6, 20, or 30°C for up to one year. Germination was testedin water and in polyethylene glycol 8000 solutions ( –0·2to –1·2 MPa osmotic potential) at 15, 20, 25 or30°C. Germination percentages and rates (inverse of meantimes to radicle emergence) were compared to those of newlyharvested, washed and dried seeds. For 40 and 60 DAA seeds,one year of storage at 20°C and water contents <6·5%significantly increased germination percentages and rates at20°C, but had little effect on germination at 25 and 30°C.Storage reduced the estimated base temperature (Tb) and meanbase water potential (b) for germination of both 40 and 60 DAAseeds by approximately 5°C and 0·3 MPa, respectively.Immature 35 DAA seeds showed the greatest benefit from storageat 3 to 5% water content and 30°C, as germination percentagesand rates increased at all water potentials (). Storage underthese same conditions had little effect on the germination ofmature seeds in water, but increased germination percentagesand rates at reduced 's. Accelerated ageing for one month at30°C and water contents from 15 to 19° increased germinationrates and percentages of mature seeds at reduced 's, but longerdurations resulted in sharp declines in both parameters. Immatureseeds lost viability within one month under accelerated ageingconditions. An after-ripening period is required at all stagesof muskmelon seed development to expand the temperature andwater potential ranges allowing germination and to achieve maximumgerminability and vigour. Post-harvest dormancy is deepest atthe point of maximum seed dry weight accumulation and declinesthereafter, both in situ within the ripening fruit and duringdry storage. The effect of light quality on the induction and release ofbud dormancy in aerial tubers of Begonia evansiana ANDR. andthe relation in controlling the mechanism of dormancy betweenlight and temperature were investigated using aerial tubersin different dormant states; immature tubers which were sproutedby irradiation and mature ones which were sprouted only afterchilling. Blue or far-red light induced photo-sprouting in immature tubersat higher temperatures and also promoted release of dormancyin mature tubers caused by chilling (2–5°C). Blueor far-red light, however, failed to break mature tuber dormancyat room temperature. The induction of dormancy in immature tubers was advanced atlow temperatures (15–17°C) which were inoperativefor photo-sprouting. Lower temperatures showed conflicting dualeffects of inducing and breaking dormancy. Red light inhibitednot only photo-sprouting in immature tubers but also dormancyrelease in mature tubers at low temperatures. These light- and temperature-dependent reactions are independentlyinvolved in dormancy regulation, the former is overcome by thelatter with dormancy development. Sequential changes in total activity and molecular forms ofacid phosphatase and ribonuclease from potato tubers were studiedby seasonal assays and Sephadex gel filtration. Ribonucleaseand p-nitrophenyl phosphatase activity fluctuated during storageof tubers, while ß-glycerophosphatase declined toa low level coincident with initiation of sprout growth. Inrecently-lifted tubers acid phosphatase activity occurred ina single high molecular weight peak. Two new forms of lowermolecular weight appeared during ageing of stored tubers. Theinfluence of infection by a tuber-rotting fungus, Phytophthoraerythroseptica,on these seasonal changes was variable. No consistent effectson total hydrolase activities were observed, while post-infectionalchanges in molecular forms included a pronounced shift in themajor acid phosphatase peak. The possible significance of thismolecular weight change in infected samples is discussed inthe light of recent evidence concerning the sub-unit structureof acid phosphatase from potato tubers.  相似文献
