We have tried to study the fate of Salmonella strains in soil. We have looked at their biochemical modifications during their evolution to dormant state and during their reviviscence. The beta galactosidase which is negative in the parent strain became positive after two weeks of cells starvation in soil. Stressed cells became able to produce acetoin. Some stressed cells did not produce the lysine decarboxylase, which is positive in parent cells. These modifications are reversible and depend on cultural conditions. Incubation of stressed cells in nutrient broth for more than four weeks helped them to reverse to normal forms. Simultaneous search for atypical Salmonella was done in dissect sludge of a domestic wastewater treatment plant and in soil irrigated with treated water. Atypical strains of Salmonella are found. We have seen that, after incubation in nutrient broth for more than four weeks, atypical strains characters evolved generally to their parental characters. All modifications of Salmonella in soil samples can make their identification very difficult and perhaps impossible.  相似文献

The objective of this study was to monitor the changes in semen characteristics in vasectomized rams and to determine if infertility was present 14 days after vasectomy. Experiments were performed using five cross-breed rams, aged between 18 and 30 months. Semen was collected weekly by artificial vagina from 2 months before to 5 months after vasectomy. After sexual rest for 10 days, vasectomy was performed by the cranial midscrotal approach. In all ejaculates the volume, concentration, total sperm number, motility and morphology (normal spermatozoa, loose heads) were determined and sperm viability (SYBR-14/PI) was evaluated in all semen samples collected after vasectomy. In the first ejaculate obtained 14 days post vasectomy all rams showed a significant (P < 0.05) drop in mean volume (from 1.2 to 0.5 mL), total sperm count (from 5176.8 to 51.1 x 10(6)) and morphologically normal sperm (from 84.1 to 15.7%), when compared to the last prevasectomy collection. We could also demonstrate a positive correlation (r = 0.89) between the individual cumulative total number of spermatozoa after vasectomy and the scrotal circumference measured before vasectomy. Sperm motility and viability could never be demonstrated after vasectomy and normal spermatozoa continuously decreased concomitant with an increase in loose heads. On post mortem examination 5 months after surgery, spermatocele formation and multiple sperm granulomas were present in all five rams. Our results show that in the first ejaculate collected by artificial vagina 14 days after vasectomy, no motile and viable spermatozoa could be detected. Despite weekly collections during a 5-month period after sterilization, azoospermia could never be achieved.  相似文献

Soil fumigation with dazomet, metam sodium, chloropicrin and chloropicrin + 1.3 D resulted in significant decrease of fungi and increase of bacteria populations in trials carried out in four farms located in different areas. Depending on the farm and the active substance applied, the fungi population was decreased by 1.4- to 3500-fold in comparison to control. Metam sodium and chloropicrin showed the best efficacy, both of them almost totally eliminated the fungi from the soil environment.The total number of bacteria was increased by the chemical fumigation with all tested products. While the population of fluorescent Pseudomonads in all treated plots increased from 2- to 100-fold, depending on the farm, the number of Bacillus spp. was not changed or decreased compared with non fumigated soil.The nematology analysis of the soil indicated that any chemical fumigant significantly limited the population of plant parasitic nematodes, which number was, anyway, below the damage threshold. However, in most cases dazomet and metam sodium reduced the total number of all nematodes present in the soil.The fumigation with chloropicrin and 1,3 D at dose of 30 g/m2 resulted in an increase of the total number of all nematodes in soil.  相似文献

On day 33 of gestation, foetal beagles were irradiated in utero (0.9 Gy of 60Co gamma-irradiation, 0.4 Gy/min). Foetal haematocytopoiesis was studied during the third trimester of gestation (days 42-55). Peripheral blood nucleated cell counts were 33 per cent lower than normal on day 44 and continued to be lower until day 49, when values became higher than normal. Splenic cellularities of irradiated pups on day 44 were more than 3 times those of the nonirradiated, but thereafter they were similar to normal. Differences in haemopoietic progenitor cell activity between irradiated and normal foetuses were observed. In comparison with the other foetal tissues, the foetal liver appeared to experience greater radiation injury. For example, on day 44, the irradiated liver BFU-E, CFU-E, and GM-CFC per 10(5) cells were almost fivefold lower than normal values. Spleens of irradiated foetal beagles contained a marked increase in all haemopoietic progenitor cells (BFU-E, CFU-E, and GM-CFC) and recognizable proliferative granulocytic cells and nucleated erythroid cells. The haemopoietic activity of the irradiated bone marrow during days 42-44 was similar to that of the irradiated spleen, and compensated for the damaged liver. However, unlike the irradiated spleen, the irradiated bone marrow had decreased BFU-E activity compared with the values for the nonirradiated bone marrow during days 48-55. Until day 50, the irradiated marrow contained fewer recognizable proliferative granulocytic cells but more nucleated erythroid cells.  相似文献

The epididymides of Lewis rats were studied at intervals up to 7 months after vasectomy, vasectomy followed 3 months later by vasovasostomy, or sham operations. Epididymal histology was related to testicular alterations and to serum antisperm antibodies as determined with an enzyme-linked immunosorbent assay. In vasectomy and vasovasostomy groups. 13 of 33 rats had testicular alterations, which consisted mainly of pronounced depletion of germ cells. Over half of the rats with testicular alterations also had severe epididymal lesions that included interstitial changes characteristic of an inflammatory response. These consisted of aggregates of mononuclear cells, including lymphocytes, plasma cells, and macrophages. The lumina of epididymides with interstitial changes contained polymorphonuclear leukocytes and/or macrophages. All animals with altered testes had greatly decreased numbers of epididymal sperm. In many instances, the lumen of the proximal cauda epididymidis was collapsed, and columnar cells of the epididymal epithelium contained many very large lysosomes. The distal cauda epididymidis was distended with sperm and debris. None of the rats that lacked testicular alterations showed epididymal changes. Mean serum antisperm antibody levels were significantly higher for rats with epididymal interstitial changes than for animals without such epididymal alterations. Infiltrations of inflammatory cells into the epididymal interstitium and lumen are part of the constellation of changes that occurs after immunization with testicular homogenates to produce experimental allergic orchitis. The observations reported here support the hypothesis that reproductive tract alterations after vasectomy in this model have an immune basis.  相似文献
