The impact of photoperiod on the rate and magnitude of N remobilization relative to uptake of inorganic N during the recovery of shoot growth after a severe defoliation was compared over 18 days in two temperate grass species, timothy (Phleum pratense L. cv. Bodin) and meadow fescue (Festuca pratensis Huds. cv. Salten). Plants were grown in flowing solution culture with N supplied as 20 mM NH4NO3 and pre-treated by extending the 11 h photosynthetically significant light period either by 1 h (short-day or SD plants) or 7 h (long-day or LD plants) of very low light intensity, during the 10 days prior to defoliation. Following a single severe defoliation, 15N-labelled NH4+ or NH4++ NO3? was supplied over a 20-day recovery period under the same SD and LD conditions. Changes in the relative contributions of remobilized N and newly acquired mineral N to shoot regrowth were assessed by sequential harvests. Both absolute and relative rates of N remobilization from root and stubble fractions were higher in LD than SD plants of both species, with the enhancement more acute but of shorter duration in timothy than fescue. Remobilized N was the predominant source of N for shoot regrowth in all treatments between days 0 and 8 after cutting; on average more so for fescue than timothy, because the presence of NO3? reduced the proportional contribution of remobilized N to the regrowth of timothy but not of fescue. Net uptake of mineral N began to recover between days 4 and 6 after cutting, with NO3? uptake restarting 1 or 2 days earlier than NH4+ uptake, even when NH4+ was the only form of N supply. LD timothy plants supplied solely with NH4+ were slowest to resume uptake of mineral N. Supplying NO3? in addition to NH4+ after defoliation promoted shoot regrowth rate but not remobilization of N. Rates of regrowth (shoot dry weight production per plant) were not correlated with rates of N remobilization from stubble either over the short-term (days 0–8) or longer term (days 0–18), interpreted as evidence against a causal dependence of regrowth rate on N remobilization under these conditions. The results are discussed in relation to hypotheses for source/sink-driven rates of N remobilization and their interactions with mineral N uptake following defoliation.  相似文献

Supplying both N forms (NH4 ++NO3 ) to the maize (Zea mays L.) plant can optimize productivity by enhancing reproductive development. However, the physiological factors responsible for this enhancement have not been elucidated, and may include the supply of cytokinin, a growth-regulating substance. Therefore, field and gravel hydroponic studies were conducted to examine the effect of N form (NH4 ++NO3 versus predominantly NO3 ) and exogenous cytokinin treatment (six foliar applications of 22 μM 6-benzylaminopurine (BAP) during vegetative growth versus untreated) on productivity and yield of maize. For untreated plants, NH4 ++NO3 nutrition increased grain yield by 11% and whole shoot N content by 6% compared with predominantly NO3 . Cytokinin application to NO3 -grown field plants increased grain yield to that of NH4 ++NO3 -grown plants, which was the result of enhanced dry matter partitioning to the grain and decreased kernel abortion. Likewise, hydroponically grown maize supplied with NH4 ++NO3 doubled anthesis earshoot weight, and enhanced the partitioning of dry matter to the shoot. NH4 ++NO3 nutrition also increased earshoot N content by 200%, and whole shoot N accumulation by 25%. During vegetative growth, NH4 ++NO3 plants had higher concentrations of endogenous cytokinins zeatin and zeatin riboside in root tips than NO3 -grown plants. Based on these data, we suggest that the enhanced earshoot and grain production of plants supplied with NH4 ++NO3 may be partly associated with an increased endogenous cytokinin supply.  相似文献

Macroalgae has bloomed in the brackish lake of Shenzhen Bay, China continuously from 2010 to 2014. Gracilaria tenuistipitata was identified as the causative macroalgal species. The aim of this study was to explore the outbreak mechanism of G. tenuistipitata, by studying the effects of salinity and nitrogen sources on growth, and the different nitrogen sources uptake characteristic. Our experimental design was based on environmental conditions observed in the bloom areas, and these main factors were simulated in the laboratory. Results showed that salinity 12 to 20 ‰ was suitable for G. tenuistipitata growth. When the nitrogen sources'' (NH4 +, NO3 ) concentrations reached 40 µM or above, the growth rate of G. tenuistipitata was significantly higher. Algal biomass was higher (approximately 1.4 times) when cultured with NH4 + than that with NO3 addition. Coincidentally, macroalgal bloom formed during times of moderate salinity (∼12 ‰) and high nitrogen conditions. The NH4 + and NO3 uptake characteristic was studied to understand the potential mechanism of G. tenuistipitata bloom. NH4 + uptake was best described by a linear, rate-unsaturated response, with the slope decreasing with time intervals. In contrast, NO3 uptake followed a rate-saturating mechanism best described by the Michaelis-Menten model, with kinetic parameters Vmax = 37.2 µM g−1 DM h−1 and Ks = 61.5 µM. Further, based on the isotope 15N tracer method, we found that 15N from NH4 + accumulated faster and reached an atom% twice than that of 15N from NO3 , suggesting when both NH4 + and NO3 were available, NH4 + was assimilated more rapidly. The results of the present study indicate that in the estuarine environment, the combination of moderate salinity with high ammonium may stimulate bloom formation.  相似文献

The effect of NO2 fumigation on root N uptake and metabolism was investigated in 3-month-old spruce (Picea abics L. Karst) seedlings. In a first experiment, the contribution of NO2 to the plant N budget was measured during a 48 h fumigation with 100mm3m?3 NO2. Plants were pre-treated with various nutrient solutions containing NO2 and NH4+, NO3? only or no nitrogen source for 1 week prior to the beginning of fumigation. Absence of NH4+ in the solution for 6d led to an increased capacity for NO3? uptake, whereas the absence of both ions caused a decrease in the plant N concentration, with no change in NO3? uptake. In fumigated plants, NO2 uptake accounted for 20–40% of NO3? uptake. Root NO3? uptake in plants supplied with NH4+plus NO3? solutions was decreased by NO2 fumigation, whereas it was not significantly altered in the other treatments. In a second experiment, spruce seedlings were grown on a solution containing both NO2 and NH4+ and were fumigated or not with 100mm3m?3 NO2 for 7 weeks. Fumigated plants accumulated less dry matter, especially in the roots. Fluxes of the two N species were estimated from their accumulations in shoots and roots, xylem exudate analysis and 15N labelling. Root NH4+ uptake was approximately three times higher than NO3? uptake. Nitrogen dioxide uptake represented 10–15% of the total N budget of the plants. In control plants, N assimilation occurred mainly in the roots and organic nitrogen was the main form of N transported to the shoot. Phloem transport of organic nitrogen accounted for 17% of its xylem transport. In fumigated plants, neither NO3? nor NH4+ accumulated in the shoot, showing that all the absorbed NO2 was assimilated. Root NO3? reduction was reduced whereas organic nitrogen transport in the phloem increased by a factor of 3 in NO2-fimugated as compared with control plants. The significance of the results for the regulation of whole-plant N utilization is discussed.  相似文献

Plants of Lolium perenne L. cv. S23 were grown in sand culturesupplied with either ammonium (NH4+) or nitrate (NO3)in an otherwise complete nutrient solution at 12°C or 20°C.Three weeks after germination, plants were clipped weekly tosimulate grazing. After 10 weeks growth all nitrogen (N) wassupplied enriched with 15N to quantify the effects of form ofN supply and temperature on the relative ability of currentroot uptake and remobilization to supply N for laminae regrowth. The form of N supply had no effect on the dry matter partitioning,while at 20°C more dry weight was allocated to laminae regrowthand less to the remaining plant material. The current root uptakeof N, which subsequently appeared in the laminae regrowth, wassimilar for plants supplied with NH4+ or NO3, and bothwere equally reduced at the lower temperature of growth. Remobilizationof N to laminae regrowth was greater for plants receiving NH4+than NO3; remobilization with either form of N supplywas reduced at the lower temperature of growth. Remobilizationwas reduced to a lesser extent at 12°C than current rootuptake. It was concluded that remobilization became relativelymore important in supplying N for regrowth of laminae at lowertemperatures. A study was designed to (a) identify sources and sinks of N in the maize (Zea mays L.) shoot, by estimating net N fluxes for each of seven parts of the shoot, (b) determine effects of N entering the plant upon fluxes of N absorbed before reproductive growth, and (c) determine the effects of the opaque-2 gene on N fluxes in the maize shoot during early reproductive growth. Plants of a maize hybrid (Pioneer 3369A) and its opaque-2 counterpart (Pioneer L3369) were grown in a greenhouse using nutrient solution/sand culture, with NO3 as the N source during the vegetative growth phase. Beginning at the time of pollination, the same nutrient regime was continued, except that some plants received no N, and others received 3.75 millimolar 15N as NO3-N.

Stalk and leaves were found to be primary N sources for the grain, while shank, husk, and cob acted first as N sinks, then as N sources during reproductive growth. Net fluxes of N for each plant part were estimated by calculating the first derivatives of regression equations used to fit data for N contents of each plant part as functions of time. All parts of the shoot were sinks for exogenous N (absorbed after pollination). Thirty-six days after pollination, the grain contained 60% endogenous N (absorbed before pollination) when 3.75 millimolar NO3-N was supplied after pollination. Rates of total N influx to the grain were identical whether or not N was supplied in the nutrient solution during reproductive growth. At 36 days after pollination, less N had accumulated in the grain of the opaque-2 genotype, but otherwise there were no differences in N contents or dry weights of the shoots due to the opaque-2 gene. Absence of N from the rooting medium significantly affected N fluxes throughout the shoot during reproductive growth, but there were no detectable effects of the opaque-2 gene on N fluxes in parts of the plant other than the grain.

Seven heathland species, four herbaceous plants and three dwarfshrubs, were tested for their capacity to utilize NH4+ or NO3. When cultured in solution at pH 4.0 with 2mol m–3 N,all species showed similar growth responses with respect toN source. Nitrate was assimilated almost equally well as ammonium,with relative growth rate generally averaging 5–8% lowerfor NO3 grown plants, albeit not always significantly.However, N source was significantly and consistently correlatedwith biomass partitioning, as NH4+-fed plants allocated moredry matter to shoots and less to roots when compared to NO3-fed plants. The strong difference in biomass partitioning mayrelate to the relative surplus of carbon per unit plant N (or,alternatively, the relatively suboptimal rate of N assimilationper unit plantC) in NO3-fed plants Inherently slow-growing dwarf shrubs accumulated virtually nofree nitrate in their tissues and reduction of nitrate was strictlyroot-based. Faster-growing herbaceous plants, however, partitionedthe assimilation of nitrate over both shoots and roots, therebyaccumulating relatively high tissue NO3 levels. Ion uptakerates depended clearly on the ‘relative shoot demand’.At similar shoot demands, especially in the herbaceous species,specific uptake rates for N and total inorganic (non-N) anionswere higher in NH4+ -fed plants, whereas the uptake rate fortotal (non-N) cations was higher in NO3-fed plants. Rateof P uptake was enhanced with increasing plant demand, but wasindependent of the N source. Net H+ extrusions ranged from 1.00to 1.34 H+ per NH4+, and from –0.48 to –0.77 H+per NO3 taken up. The influence of NH4+, in the external medium, on fluxes of NO3 and K+ were investigated using barley (Hordeum vulgare cv Betzes) plants. NH4+ was without effect on NO3 (36ClO3) influx whereas inhibition of net uptake appeared to be a function of previous NO3 provision. Plants grown at 10 micromolar NO3 were sensitive to external NH4+ when uptake was measured in 100 micromolar NO3. By contrast, NO3 uptake (from 100 micromolar NO3) by plants previously grown at this concentration was not reduced by NH4+ treatment. Plants pretreated for 2 days with 5 millimolar NO3 showed net efflux of NO3 when roots were transferred to 100 micromolar NO3. This efflux was stimulated in the presence of NH4+. NH4+ also stimulated NO3 efflux from plants pretreated with relatively low nitrate concentrations. It is proposed that short term effects on net uptake of NO3 occur via effects upon efflux. By contrast to the situation for NO3, net K+ uptake and influx of 36Rb+-labeled K+ was inhibited by NH4+ regardless of the nutrient history of the plants. Inhibition of net K+ uptake reached its maximum value within 2 minutes of NH4+ addition. It is concluded that the latter ion exerts a direct effect upon K+ influx.  相似文献

The effect of exogenous NH4+ on NO3 uptake and in vivo NO3 reductase activity (NRA) in roots of Phaseolus vulgaris L. cv Witte Krombek was studied before, during, and after the apparent induction of root NRA and NO3 uptake. Pretreatment with NH4Cl (0.15-50 millimolar) affected neither the time pattern nor the steady state rate of NO3 uptake.

When NH4+ was given at the start of NO3 nutrition, the time pattern of NO3 uptake was the same as in plants receiving no NH4+. After 6 hours, however, the NO3 uptake rate (NUR) and root NRA were inhibited by NH4+ to a maximum of 45% and 60%, respectively.

The response of the NUR of NO3-induced plants depended on the NH4Cl concentration. Below 1 millimolar NH4+, the NUR declined immediately and some restoration occurred in the second hour. In the third hour, the NUR became constant. In contrast, NH4+ at 2 millimolar and above caused a rapid and transient stimulation of NO3 uptake, followed again by a decrease in the first, a recovery in the second, and a steady state in the third hour. Maximal inhibition of steady state NUR was 50%. With NO3-induced plants, root NRA responded less and more slowly to NH4+ than did NUR.

Methionine sulfoximine and azaserine, inhibitors of glutamine synthetase and glutamate synthase, respectively, relieved the NH4+ inhibition of the NUR of NO3-induced plants. We conclude that repression of the NUR by NH4+ depends on NH4+ assimilation. The repression by NH4+ was least at the lowest and highest NH4+ levels tested (0.04 and 25 millimolar).

The influence of various nitrogen (N) and sulphur (S) forms on the uptake of manganese (Mn) in young spring barley (Hordeum vulgare L cv Golf) plants was examined in both a hydroponic system and in a soil-based system. The soil was a typical Danish Mn-deficient soil viz. a sandy loam soil developed on old marine sediments. Plants growing in solution culture with NO3 as the only N source had a higher Mn uptake than plants receiving mixtures of NO3 and NH4+. These findings were opposite to the results obtained in the soil-based experiments, where plants fertilized with NO3 as the only N source accumulated much less Mn than plants fertilized with NH4+. Combining the results of these experiments confirmed that NH4+ acted as a powerful antagonist to Mn2+ during uptake but that this antagonistic effect was more than compensated for by the influence of NH4+ in reducing plant-unavailable Mn(IV) to plant-available Mn(II) in the soil. Furthermore the soil experiments showed that fertilizers containing sulphur in the form of reduced S (thiosulphate) had a strong mobilizing effect on Mn, and enabled the plants to accumulate large amounts of Mn in the biomass compared with oxidized S (sulphate). Thus, fertilization with thiosulphate may be very effective in alleviating Mn-deficiency in soils developed on old marine sediments where Mn availability is limiting plant growth.  相似文献

Assimilation of NO3 and NH4+ by perennial ryegrass (Lolium perenne L.) turf, previously deprived of N for 7 days, was examined. Nitrogen uptake rate was increased up to four- to five-fold for both forms of N by N-deprivation as compared to N-sufficient controls, with the deficiency-enhanced N absorption persisting through a 48 hour uptake period. Nitrate, but not NH4+, accumulated in the roots and to a lesser degree in shoots. By 48 hours, 53% of the absorbed NO3 had been reduced, whereas 97% of the NH4+ had been assimilated. During the early stages (0 to 8 hours) of NO3 uptake by N-deficient turf, reduction occurred primarily in the roots. Between 8 and 16 hours, however, the site of reduction shifted to the shoots. Nitrogen form did not affect partitioning of the absorbed N between roots (40%) and shoots (60%) but did affect growth. Compared to NO3, NH4+ uptake inhibited root, but not shoot, growth. Total soluble carbohydrates decreased in both roots and shoots during the uptake period, principally the result of fructan metabolism. Ammonium uptake resulted in greater total depletion of soluble carbohydrates in the root compared to NO3 uptake. The data indicate that N assimilation by ryegrass turf utilizes stored sugars but is also dependent on current photosynthate.  相似文献

Nitrogen (N) limits plant productivity and its uptake and assimilation may be regulated by N source, N availability, and nitrate reductase activity (NRA). Knowledge of how these factors interact to affect N uptake and assimilation processes in woody angiosperms is limited. We fertilized 1-year-old, half-sib black walnut (Juglans nigra L.) seedlings with ammonium (NH4 +) [as (NH4)2SO4], nitrate (NO3 ) (as NaNO3), or a mixed N source (NH4NO3) at 0, 800, or 1,600 mg N plant−1 season−1. Two months following final fertilization, growth, in vivo NRA, plant N status, and xylem exudate N composition were assessed. Specific leaf NRA was higher in NO3 -fed and NH4NO3-fed plants compared to observed responses in NH4 +-fed seedlings. Regardless of N source, N addition increased the proportion of amino acids (AA) in xylem exudate, inferring greater NRA in roots, which suggests higher energy cost to plants. Root total NRA was 37% higher in NO3 -fed than in NH4 +-fed plants. Exogenous NO3 was assimilated in roots or stored, so no difference was observed in NO3 levels transported in xylem. Black walnut seedling growth and physiology were generally favored by the mixed N source over NO3 or NH4 + alone, suggesting NH4NO3 is required to maximize productivity in black walnut. Our findings indicate that black walnut seedling responses to N source and level contrast markedly with results noted for woody gymnosperms or herbaceous angiosperms.  相似文献

Growth, chemical composition, and nitrate reductase activity (NRA) of hydroponically cultured Rumex crispus, R. palustris, R. acetosa, and R. maritimus were studied in relation to form (NH4 +, NO3 -, or both) and level of N supply (4 mM N, and zero-N following a period of 4mM N). A distinct preference for either NH4 + or NO3 - could not be established. All species were characterized by a very efficient uptake and utilization of N, irrespective of N source, as evident from high concentrations of organic N in the tissues and concurrent excessive accumulations of free NO3 - and free NH4 +. Especially the accumulation of free NH4 + was unusually large. Generally, relative growth rate (RGR) was highest with a combination of NH4 + and NO3 -. Compared to mixed N supply, RGR of NO3 -- and NH4 +-grown plants declined on average 3% and 9%, respectively. Lowest RGR with NH4 + supply probably resulted from direct or indirect toxicity effects associated with high NH4 + and/or low Ca2+ contents of tissues. NRA in NO3 - and NH4NO3 plants was very similar with maxima in the leaves of ca 40 μmol NO2 - g-1 DW h-1. ‘Basal’ NRA levels in shoot tissues of NH4 + plants appeared relatively high with maxima in the leaves of ca 20 μmol NO2 - g-1 DW h-1. Carboxylate to organic N ratios, (C-A)/Norg, on a whole plant basis varied from 0.2 in NH4 + plants to 0.9 in NO3 - plants. After withdrawal of N, all accumulated NO3 - and NH4 + was assimilated into organic N and the organic N redistributed on a large scale. NRA rapidly declined to similar low levels, irrespective of previous N source. Shoot/root ratios of -N plants were 50–80% lower than those from +N plants. In comparison with +N, RGR of -N plants did not decline to a large extent, decreasing by only 15% in -NH4 + plants due to very high initial organic-N contents. N-deprived plants all exhibited an excess cation over anion uptake (net proton efflux), and whole-plant (C-A)/Norg ratios increased to values around unity. Possible difficulties in interpreting the (C-A)/Norg ratio and NRA of plants in their natural habitats are briefly discussed.  相似文献

The effect of ectomycorrhizal association of Pinus pinaster with Hebeloma cylindrosporum was investigated in relation to the nitrogen source supplied as mineral (NH4+ or NO3?) or organic N (L ‐glutamate) and at 5 mol m?3. Plants were grown for 14 and 16 weeks with mineral and organic N, respectively, and samples were collected during the last 6 weeks of culture. Total fungal biomass was estimated using glucosamine amount and its viability was assessed using the glucosamine to ergosterol ratio. Non‐mycorrhizal plants grew better with NH4+ than with NO3? and grew very slowly when supplied with L ‐glutamate. The presence of the fungus decreased the growth of the host plant with mineral N whereas it increased it with L ‐glutamate. Whatever the N source, most of the living fungal biomass was associated with the roots, whereas the main part of the total biomass was assayed outside the root. The form of mineral N did not significantly affect N accumulation rates over the 42 d in control plants. In mycorrhizal plants grown on either N source, the fungal tissues developing outside of the root were always the main N sink. The ectomycorrhizal association did not change 15NH4+ uptake rate by roots, suggesting that the growth decrease of the host‐plant was related to the carbon cost for fungal growth and N assimilation rather than to a direct effect on NH4+ acquisition. In contrast, in NO3?‐grown plants, in addition to draining carbon for NO3? reduction the fungus competed with the root for NO3? uptake. With NH4+ or NO3? feeding, although mycorrhizal association improved N accumulation in shoots, we concluded that it was unlikely that the fungus had supplied the plant with N. In L ‐glutamate‐grown plants, the presence of the fungus increased the proportion of glutamine in the xylem sap and improved both N nutrition and the growth rate of the host plant.  相似文献

The effect of two N-forms (NH4 + and NO3 ) and NaCl on pattern of accumulation of some essential inorganic nutrients was examined in sunflower (Helianthus annuus L.) cv. Hisun 33. Eight-day-old plants of were subjected for 21 d to Hoagland's nutrient solution containing 8 mM N as NH4 + or NO3 ·, and salinized with and addition of NaCl to the growth medium had no significant effect on total leaf N. However, root N of NH4-supplied plants decreased significantly with increase in NaCl concentration, whereas that of NO3-supplied plants remained unaffected. There was no significant effect of NaCl on leaf or root P, but the NO3-supplied plants had significa concentration of leaf P than that of NH4-supplied plants at varying salt treatments. Salinity of the rooting med did not show any significant effect on Na+ concentrations of leaves or roots of plants subjected to two differen N. NH4-treated plants generally had greater concentrations of Cl in leaves and roots and lower K+ content in leaves than NO3-supplied plants. Ca2+ concentrations of leaves and roots and Mg2+ concentrations of leaves decreased in NH4-supplied plants due to NaCl, but they remained unaffected in NO3-treated plants.  相似文献

Several studies have previously shown that shoot removal of forage species, either by cutting or herbivore grazing, results in a large decline in N uptake (60%) and/or N2 fixation (80%). The source of N used for initial shoot growth following defoliation relies mainly on mobilisation of N reserves from tissues remaining after defoliation. To date, most studies investigating N-mobilisation have been conducted, with isolated plants grown in controlled conditions. The objectives of this study were for Lolium perenne L., grown in a dense canopy in field conditions, to determine: 1) the contribution of N-mobilisation, NH4 + uptake and NO3 - uptake to growing shoots after defoliation, and 2) the contribution of the high (HATS) and low (LATS) affinity transport systems to the total plant uptake of NH4 + and NO3 -. During the first seven days following defoliation, decreases in biomass and N-content of roots (34% and 47%, respectively) and to a lesser extent stubble (18% and 43%, respectively) were observed, concomitant with mobilisation of N to shoots. The proportion and origin of N used by shoots (derived from reserves or uptake) was similar to data reported for isolated plants. Both HATS and LATS contributed to the total root uptake of NH4 + and NO3 -. The Vmax of both the NH4 + and NO3 - HATS increased as a function of time after defoliation, and both HATS systems were saturated by substrate concentrations in the soil at all times. The capacity of the LATS was reduced as soil NO3 - and NH4 + concentrations decreased following defoliation. Data from 15N uptake by field-grown plants, and uptake rates of NH4 + and NO3 - estimated by excised root bioassays, were significantly correlated, though uptake was over-estimated by the later method. The results are discussed in terms of putative mechanisms for regulating N uptake following severe defoliation. The effects of nitrogen source NO3 or NH4+ on nitrogen metabolism during the first 2 weeks of germination of the rice seedling (Oryza sativa L., var. IR22) grown in nutrient solution containing 40 μg/ml N were studied. Total, soluble protein, and free amino N levels were higher in the NH4+-grown seedling, particularly during the 1st week of germination. Asparagine accounted for most of the difference in free amino acid level, in both the root and the shoot. Nitrate and nitrite reductase activities were present mainly in the shoot and were higher in the NO3-grown seedling, whereas the activity of glutamate dehydrogenase and glutamine synthetase in the root tended to be lower than that of the NH4+-grown seedling during the 1st week of germination. Glycolate oxidase and catalase activities were present mainly in the shoot. Maximum activity of the above five enzymes occurred 7 to 10 days after germination. Differences in the zymograms of nitrate reductase, glutamate dehydrogenase, and catalase were mainly between shoot and root and not from N source. Nitrite reductase bands were observed only in plants grown in plants grown in NO3.  相似文献

The fern Dicranopteris dichotoma is an important pioneer species of the understory in Masson pine (Pinus massoniana) forests growing on acidic soils in the subtropical and tropical China. To improve our understanding of the role of D. dichotoma in nitrogen (N) uptake of these forests, a short-term 15N experiment was conducted at mountain ridge (MR, with low N level) and mountain foot (MF, with high N level). We injected 15N tracers as 15NH4, 15NO3 or 15N-glycine into the soil surrounding each plant at both MR and MF sites. Three hours after tracer injection, the fern D. dichotoma took up 15NH4 + significantly faster at MF than at MR, but it showed significantly slower uptake of 15NO3 at MF than at MR. Consequently, 15NO3 made greater contribution to the total N uptake (50% to the total N uptake) at MR than at MF, but 15N-glycine only contributed around 11% at both sites. Twenty-four hours after tracer injection, D. dichotoma preferred 15NH4 + (63%) at MR, whereas it preferred 15NO3 (47%) at MF. We concluded that the D. dichotoma responds distinctly in its uptake pattern for three available N species over temporal and spatial scales, but mainly relies on inorganic N species in the subtropical forest. This suggests that the fern employs different strategies to acquire available N which depends on N levels and time.  相似文献

Studies that quantify plant δ15N often assume that fractionation during nitrogen uptake and intra-plant variation in δ15N are minimal. We tested both assumptions by growing tomato (Lycopersicon esculetum Mill. cv. T-5) at NH4+ or NO?3 concentrations typical of those found in the soil. Fractionation did not occur with uptake; whole-plant δ15N was not significantly different from source δ15 N for plants grown on either nitrogen form. No intra-plant variation in δ15N was observed for plants grown with NH+4. In contrast. δ15N of leaves was as much as 5.8% greater than that of roots for plants grown with NO?3. The contrasting patterns of intra-plant variation are probably caused by different assimilation patterns. NH+4 is assimilated immediately in the root, so organic nitrogen in the shoot and root is the product of a single assimilation event. NO?3 assimilation can occur in shoots and roots. Fractionation during assimilation caused the δ15N of NO?3 to become enriched relative to organic nitrogen; the δ15N of NO?3 was 11.1 and 12.9% greater than the δ15N of organic nitrogen in leaves and roots, respectively. Leaf δ15N may therefore be greater than that of roots because the NO?3 available for assimilation in leaves originates from a NO?3 pool that was previously exposed to nitrate assimilation in the root.  相似文献
