Metapopulation moments: coupling, stochasticity and persistence   总被引:9,自引:1,他引:8  
1.  Spatial heterogeneity has long been viewed as a reliable means of increasing persistence. Here, an analytical model is developed to consider the variation and, hence, the persistence of stochastic metapopulations. This model relies on a novel moment closure technique, which is equivalent to assuming log-normal distributions for the population sizes.
2.  Single-species models show the greatest persistence when the mixing between subpopulations is large, so spatial heterogeneity is of no benefit. This result is confirmed by stochastic simulation of the full metapopulation.
3.  In contrast, natural-enemy models exhibit the greatest persistence for intermediate levels of coupling. When the coupling is too low, there are insufficient rescue effects between the subpopulations to sustain the dynamics, whereas when the coupling is too high all spatial heterogeneity is lost.

Functional connectivity is crucial for the persistence of a metapopulation, because migration among subpopulations enables recolonization and counteracts genetic drift, which is especially important in small subpopulations. We studied the degree and drivers of connectivity among occupied patches of a coastal dune metapopulation of the Natterjack Toad (Epidalea calamita Laurenti), on the basis of microsatellite variation. As spatial landscape heterogeneity is expected to influence dispersal and genetic structure, we analyzed which landscape features affect functional connectivity and to what extent. Sixty different landscape resistance scenarios as well as the isolation-by-distance model were compared using two landscape genetics approaches. We identified three subpopulations with unidirectional levels of gene flow among the two most geographically separated subpopulations, while inferred gene flow into the geographically intermediate subpopulation was limited. Urbanization and vegetation height negatively affected connectivity. Low estimates of genetic diversity and effective population size indicate that conservation measures in the smallest and most isolated subpopulation are required.  相似文献

Fibroblast heterogeneity in collagenolytic response to cyclosporine.   总被引:4,自引:0,他引:4  
To investigate the mechanism of cyclosporine (CS)-induced fibrotic gingival enlargement, the effect of CS on the collagenolytic activities of 14 different human gingival fibroblast strains derived from healthy individuals with non-inflammed gingiva was examined in vitro. There was marked heterogeneity among individuals in basal levels of collagenase activity, and there was also variation among the subpopulations derived from one strain. Fibroblasts from different individuals also varied markedly in their collagenolytic response to CS (0.1 to 0.75 micrograms/ml). In most strains, CS decreased collagenase activity, but in some, the drug caused no change or significantly increased activities. In most of the subpopulations CS significantly decreased collagenolytic activity. Two of the fibroblasts strains and the subpopulations described above were examined for the production of immunoreactive collagenase and tissue inhibitor of metalloproteinase (TIMP). The two strains made similar amounts of collagenase, but differed markedly in TIMP levels; CS affected their collagenase production differently but had similar effects on TIMP. Among the subpopulations there was variation in the production of collagenase, although none made detectable levels of TIMP; they also varied in the production of both proteins in response to CS. In two of the subpopulations and in both strains at some concentrations, the effect of CS on the relative levels of collagenase and TIMP could account for the decreased collagenase activity; i.e., the level of collagenase was unchanged or decreased, and TIMP production was unchanged or increased. This study demonstrates the variation among individuals as well as intrastrain heterogeneity of human gingival fibroblasts with regard to collagenase activity and the production of collagenase and TIMP. The heterogeneity of the collagenolytic response of different gingival fibroblast strains and their subpopulations to CS treatment may partly explain the susceptibility of only some individuals to CS-induced gingival enlargement.  相似文献

Cell population heterogeneity can affect cellular response and is a major factor in drug resistance. However, there are few techniques available to represent and explore how heterogeneity is linked to population response. Recent high-throughput genomic, proteomic, and cellomic approaches offer opportunities for profiling heterogeneity on several scales. We have recently examined heterogeneity in vascular endothelial growth factor receptor (VEGFR) membrane localization in endothelial cells. We and others processed the heterogeneous data through ensemble averaging and integrated the data into computational models of anti-angiogenic drug effects in breast cancer. Here we show that additional modeling insight can be gained when cellular heterogeneity is considered. We present comprehensive statistical and computational methods for analyzing cellomic data sets and integrating them into deterministic models. We present a novel method for optimizing the fit of statistical distributions to heterogeneous data sets to preserve important data and exclude outliers. We compare methods of representing heterogeneous data and show methodology can affect model predictions up to 3.9-fold. We find that VEGF levels, a target for tuning angiogenesis, are more sensitive to VEGFR1 cell surface levels than VEGFR2; updating VEGFR1 levels in the tumor model gave a 64% change in free VEGF levels in the blood compartment, whereas updating VEGFR2 levels gave a 17% change. Furthermore, we find that subpopulations of tumor cells and tumor endothelial cells (tEC) expressing high levels of VEGFR (>35,000 VEGFR/cell) negate anti-VEGF treatments. We show that lowering the VEGFR membrane insertion rate for these subpopulations recovers the anti-angiogenic effect of anti-VEGF treatment, revealing new treatment targets for specific tumor cell subpopulations. This novel method of characterizing heterogeneous distributions shows for the first time how different representations of the same data set lead to different predictions of drug efficacy.  相似文献

Bacterial biofilm formation is an important cause of environmental persistence of food-borne pathogens, such as Salmonella Typhimurium. As the ensemble of bacterial cells within a biofilm represents different physiological states, even for monospecies biofilms, gene expression patterns in these multicellular assemblages show a high degree of heterogeneity. This heterogeneity might mask differential gene expression that occurs only in subpopulations of the entire biofilm population when using methods that average expression output. In an attempt to address this problem and to refine expression analysis in biofilm studies, we used the Differential Fluorescence Induction (DFI) technique to gain more insight in S. Typhimurium biofilm gene expression. Using this single cell approach, we were able to identify 26 genetic loci showing biofilm specific increased expression. For a selected number of identified genes, we confirmed the DFI results by the construction of defined promoter fusions, measurement of relative gene expression levels and construction of mutants. Overall, we have shown for the first time that the DFI technique can be used in biofilm research. The fact that this analysis revealed genes that have not been linked with Salmonella biofilm formation in previous studies using different approaches illustrates that no single technique, in casu biofilm formation, is able to identify all genes related to a given phenotype.  相似文献

采用ISSR分子标记技术对在3个不同地区引起蝉若虫地方病的蝉棒束孢种群遗传结构进行研究.结果表明: 3个地方病种群均表现出较高的遗传多样性,其中敬亭山种群的遗传多样性最高,石台种群最小.UPGMA聚类分析表明,不同地方病种群无优势性,均为多系的异质种群,遗传谱系与地理来源无关.敬亭山不同采集时间的菌株表现出明显的时间异质性.种群(亚种群)间的遗传分化系数Gst为0.2153,而基因流Nm为0.9110(Nm<1), 暗示低水平的基因交流是种群内遗传变异的主要原因之一.因此,蝉棒束孢种群水平的高异质性和低优势性是维持蝉若虫地方病的遗传结构特征.  相似文献

Species often occur in subdivided populations as a consequence of spatial heterogeneity of the habitat. To describe the spatial organization of subpopulations, existing theory proposes three main population models: patchy population, metapopulation and isolated populations. These models differ in their predicted levels of connectivity among subpopulations, and in the risk that a subpopulation will go extinct. However, spatially discrete subpopulations are commonly considered to be organized as metapopulations, even though explicit tests of metapopulation assumptions are rare. Here, we test predictions of the three models on the basis of demographic and genetic data, a combined approach so far surprisingly little used in mobile organisms. From 2002 to 2005, we studied nine subpopulations of the wetland-restricted reed bunting ( Emberiza schoeniclus ) in the southeastern part of the Canton Zurich (Switzerland), from which local declines of this species have been reported. Here, wetlands are as small as 2.7 ha and separated through intensively used agricultural landscapes. Demographic data consisted of dispersal of colour-banded individuals among subpopulations, immigration rates and extinction-/recolonization dynamics. Genetic data were based on the distribution of genetic variability and gene flow among subpopulations derived from the analysis of nine microsatellite loci. Both demographic and genetic data revealed that the patchy population model best described the spatial organization of reed bunting subpopulations. High levels of dispersal among subpopulations, high immigration into the patchy population, and genetic admixture suggested little risk of extinction of both subpopulations and the entire patchy population. This study exemplifies the idea that spatially discrete subpopulations may be organized in ways other than a metapopulation, and hence has implications for the conservation of subpopulations and species.  相似文献

We tested experimentally the effects of genetic variation in host population on the host-parasitoid system persistence. The experimental systems consisted of one parasitoid wasp species (Heterospilus prosopidis), one bean weevil species (Callosobruchus chinensis), and one bean species, of which only the host species (bean weevil) was genetically manipulated. As control treatments with low genetic heterogeneity in the host population, we used two bean weevil strains (Kyoto and Niigata strains) which have several contrasting ecological traits. For the high genetic heterogeneity treatment, hybrid bean weevils which were generated by crossbreeding the strains Kyoto and Niigata were used. In the multiple-generation experiments, all three treatments had different patterns of extinction. The control treatment with the Niigata strain was very prone to system extinction, whereas the treatment with hybrids showed coexistence of constituent species in almost all replicates and also showed stabilized population dynamics. The other control treatment, using the Kyoto strain, showed intermediate proneness. To interpret the results of multiple-generation experiments, we conducted several short-term experiments. The different persistence patterns between the two control treatments were explained by the shapes of the host-finding abilities of wasps and the growth rate of the bean weevils. The mean values of many ecological traits of hybrid lines were not different from those of the Kyoto strain, but their variability increased. These outcomes corresponded well to the prediction of models by Doebeli [J Theor Biol (1997) 188:109–120]. We discuss the mechanisms in which the variability in host species population was effective for the prolonged system persistence.  相似文献

A basic assumption of many epidemic models is that populations are composed of a homogeneous group of randomly mixing individuals. This is not a realistic assumption. Most actual populations are divided into a number of subpopulations, within which there may be relatively random mixing, but among which there is nonrandom mixing. As a consequence of the structuring of the population, there are several sources of heterogeneity within populations that can affect the course of an infection through the population. Two of these sources of heterogeneity are differences in contact number between subpopulations, and differences in the patterns of contact among subpopulations. A model for the spread of a disease in such a population is described. The model considers two levels of interaction: interactions between individuals within a subpopulation because of geographic proximity, and interactions between individuals of the same or different subpopulations because of attendance at common social functions. Because of this structure, it is possible to analyze with the model both heterogeneity in contact number and variation in the patterns of contact. A stability analysis of the model is presented which shows that there is a unique threshold for disease maintenance. Below the threshold the disease goes extinct, and the equilibrium is globally asymptotically stable. Above the threshold, the extinction equilibrium is unstable, and there is a unique endemic equilibrium. The analysis presents a sufficient condition for disease maintenance, which determines critical subpopulation sizes above which the disease cannot go extinct. The condition is a simple inequality relating the removal rate of infectives to the infection rate of susceptibles. In addition, bounds on the actual threshold and the effect of symmetry in the interaction matrix on the threshold are presented.  相似文献

Environmental heterogeneity can promote the emergence of locally adapted phenotypes among subpopulations of a species, whereas gene flow can result in phenotypic and genotypic homogenization. For organisms like amphidromous fishes that change habitats during their life history, the balance between selection and migration can shift through ontogeny, making the likelihood of local adaptation difficult to predict. In Hawaiian waterfall‐climbing gobies, it has been hypothesized that larval mixing during oceanic dispersal counters local adaptation to contrasting topographic features of streams, like slope gradient, that can select for predator avoidance or climbing ability in juvenile recruits. To test this hypothesis, we used morphological traits and neutral genetic markers to compare phenotypic and genotypic distributions in recruiting juveniles and adult subpopulations of the waterfall‐climbing amphidromous goby, Sicyopterus stimpsoni, from the islands of Hawai'i and Kaua'i. We found that body shape is significantly different between adult subpopulations from streams with contrasting slopes and that trait divergence in recruiting juveniles tracked stream topography more so than morphological measures of adult subpopulation differentiation. Although no evidence of population genetic differentiation was observed among adult subpopulations, we observed low but significant levels of spatially and temporally variable genetic differentiation among juvenile cohorts, which correlated with morphological divergence. Such a pattern of genetic differentiation is consistent with chaotic genetic patchiness arising from variable sources of recruits to different streams. Thus, at least in S. stimpsoni, the combination of variation in settlement cohorts in space and time coupled with strong postsettlement selection on juveniles as they migrate upstream to adult habitats provides the opportunity for morphological adaptation to local stream environments despite high gene flow.  相似文献

The ‘environmental heterogeneity hypothesis’ (EHH) has been proposed as a mechanism that enables species coexistence through resource partitioning. In accordance with this hypothesis, plant diversity is predicted to increase with variability in resources, but there has been weak support for this hypothesis from experimental studies. The objectives of this research were to 1) characterize how resource availability and heterogeneity (coefficient of variation) change as plant communities develop using sequentially restored grasslands, 2) determine if resource heterogeneity relates to plant diversity (effective number of species, richness and evenness) and 3) reveal if the strength of resource heterogeneity–diversity relationships is different among levels of resource availability. We quantified means and coefficients of variation in soil nitrate and light availability in grasslands established on former agricultural lands for different times and their relationship to plant diversity using a geostatistically‐informed design. Nitrate availability decreased exponentially with restoration age, but no directional change in nitrate heterogeneity across the chronosequence occurred due to high resource variability in some restorations. Light availability also decreased exponentially across the chronosequence, but there was no directional change in light heterogeneity. Nitrate heterogeneity was positively correlated with both plant richness and plant effective number of species at high levels of nitrate availability. However, no nitrate heterogeneity correlation was detected at low levels of nitrate availability. Light heterogeneity was positively correlated with plant effective number of species at low levels of light availability. However, no light heterogeneity correlation was detected at high levels of light availability. Plant evenness was not correlated with resource heterogeneity at any resource availability level. These results support the positive heterogeneity–diversity relationship predicted by EHH, and uniquely that this relationship develops within a decade of plant community development, but can be obscured by resource availability.  相似文献

The heterogeneity of parasitism risk among host individuals is a key factor for stabilizing or sustaining host–parasitoid interactions. Host maturation variability, or the variation in the maturation times among host individuals, is the simplest source of such heterogeneity, but it has often been neglected in previous theoretical studies. We developed a configuration individual-based model (cIBM) of host–parasitoid interaction to investigate to what degree of host maturation variability promotes the persistence of host–parasitoid interactions. We ran simulations with various degrees of host maturation variability for different lengths of unsusceptible period. The result showed that low host maturation variability could sustain host–parasitoid dynamics when the host-unsusceptible period was short. Conversely, high levels of variability could sustain host–parasitoid dynamics when the host-unsusceptible period was about half of the total larval period. This suggests that the balance between variability and unsusceptible period is important for the persistence of host–parasitoid interaction. We conclude that maturation variability is a factor that can contribute to the sustainment of host–parasitoid interactions.  相似文献

Spatial variation in habitat quality and anthropogenic factors, as well as social structure, can lead to spatially structured populations of animals. Demographic approaches can be used to improve our understanding of the dynamics of spatially structured populations and help identify subpopulations critical for the long-term persistence of regional metapopulations. We provide a regional metapopulation analysis to inform conservation management for Masai giraffes (Giraffa camelopardalis tippelskirchi) in five subpopulations defined by land management designations. We used data from an individual-based mark–recapture study to estimate subpopulation sizes, subpopulation growth rates, and movement probabilities among subpopulations. We assessed the source–sink structure of the study population by calculating source–sink statistics, and we created a female-based matrix metapopulation model composed of all subpopulations to examine how variation in demographic components of survival, reproduction, and movement affected metapopulation growth rate. Movement data indicated no subpopulation was completely isolated, but movement probabilities varied among subpopulations. Source–sink statistics and net flow of individuals indicated three subpopulations were sources, while two subpopulations were sinks. We found areas with higher wildlife protection efforts and fewer anthropogenic impacts were sources, and less-protected areas were identified as sinks. Our results highlight the importance of identifying source–sink dynamics among subpopulations for effective conservation planning and emphasize how protected areas can play an important role in sustaining metapopulations.  相似文献

1. This paper explores the concept of the critical community size for persistence of infection in wildlife populations. We use as a case study the 1988 epidemic of phocine distemper virus in the North Sea population of harbour seals, Phoca vitulina .
2. We summarize the available data on this epidemic and use it to parameterize a stochastic compartmental model for an infection spreading through a spatial array of patches coupled by nearest-neighbour mixing, with replacement of susceptibles occurring as a discrete annual event.
3. A combination of analytical and simulation techniques is used to show that the high levels of transmission between different seal subpopulations, combined with the small annual birth cohort, act to make persistence of infection impossible in this harbour seal population at realistic population levels. The well known mechanisms by which metapopulation structures may act to promote persistence can be seen to have an effect only at weaker levels of spatial coupling, and higher levels of host recruitment, than those empirically observed.  相似文献

Spatial heterogeneity of resources can influence plant community composition and diversity in natural communities. We manipulated soil depth (two levels) and nutrient availability (three levels) to create four heterogeneity treatments (no heterogeneity, depth heterogeneity, nutrient heterogeneity, and depth + nutrient heterogeneity) replicated in an agricultural field seeded to native prairie species. Our objective was to determine whether resource heterogeneity influences species diversity and the trajectory of community development during grassland restoration. The treatments significantly increased heterogeneity of available inorganic nitrogen (N), soil water content, and light penetration. Plant diversity was indirectly related to resource heterogeneity through positive relationships with variability in productivity and cover established by the belowground manipulations. Diversity was inversely correlated with the average cover of the dominant grass, Switchgrass (Panicum virgatum), which increased over time in all heterogeneity treatments and resulted in community convergence among the heterogeneity treatments over time. The success of this cultivar across the wide range of resource availability was attributed to net photosynthesis rates equivalent to or higher than those of the native prairie plants in the presence of lower foliar N content. Our results suggest that resource heterogeneity alone may not increase diversity in restorations where a dominant species can successfully establish across the range of resource availability. This is consistent with theory regarding the role of ecological filters on community assembly in that the establishment of one species best adapted for the physical and biological conditions can play an inordinately important role in determining community structure.  相似文献

Summary Three populations of the grass Danthonia spicata were observed to have different rates of biomass accumulation when grown in common environment treatments. The populations were native to adjacent sites of different successional age and different levels of shading. Twelve individuals from each population were clonally replicated and two replicates were grown in each of two light treatments, 100% and 22% of unshaded sunlight. Following growth in the treatments the populations all exhibited the same mean light-saturated photosynthetic rate of 11.7 mol m-2s-1. This rate is intermediate for published values of sun and shade species and for species from along a successional gradient. There was no difference in photosynthetic rate among treatments. There was significant genetic variation for lightsaturated photosynthetic rate within populations but no significant differences among populations. The populations had similar leaf water potential values of-1.12 MPa in all treatments. There were significant differences among treatments and genotypes for specific leaf weight which resulted in significant differences among treatments and no significant differences among genotypes in light-saturated photosynthetic rate expressed on a leaf weight basis. Lightsaturated photosynthetic rate had a high heritability and low plasticity. We postulate that photosynthetic rate is under strong selection and that the observed rates permit populations of D. spicata to grow in a wide range of habitat light levels.  相似文献

Bacterial persistence is a feature that allows susceptible bacteria to survive extreme concentrations of antibiotics and it has been verified in a number of species, such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus spp., Mycobacterium spp. However, even though Acinetobacter baumannii is an important nosocomial pathogen, data regarding its persistence phenotype are still lacking. Therefore, the aim of this study was to evaluate the persistence phenotype in A. baumannii strains, as well as its variation among strains after treatment with polymyxin B and tobramycin. Stationary cultures of 37 polymyxin B-susceptible clinical strains of A. baumannii were analyzed for surviving cells after exposure to 15 µg/mL of polymyxin B for 6 h, by serial dilutions and colony counting. Among these, the 30 tobramycin-susceptible isolates also underwent tobramycin treatment at a concentration of 160 µg/mL and persister cells occurrence was evaluated equally. A high heterogeneity of persister cells formation patterns among isolates was observed. Polymyxin B-treated cultures presented persister cells corresponding from 0.0007% to 10.1% of the initial population and two isolates failed to produce detectable persister cells under this condition. A high variability could also be observed when cells were treated with tobramycin: the persister fraction corresponded to 0.0003%–11.84% of the pre-treatment population. Moreover, no correlation was found between persister subpopulations comparing both antibiotics among isolates, indicating that different mechanisms underlie the internal control of this phenotype. This is the first report of persister cells occurrence in A. baumannii. Our data suggest that distinct factors regulate the tolerance for unrelated antibiotics in this species, contrasting the multi-drug tolerance observed in other species (eg. dormancy-mediated tolerance). Supporting this observation, polymyxin B – an antibiotic that is believed to act on non-dividing cells as well – failed to eradicate persister cells in the majority of the isolates, possibly reflecting a disconnection between persistence and dormancy.  相似文献

The genetic structure of populations of the ectomycorrhizal basidiomycete Suillus luteus in heavy metal polluted and nonpolluted areas was studied. Sporocarps were collected at nine different locations and genotyped at four microsatellite loci. Six of the sampling sites were severely contaminated with heavy metals and were dominated by heavy metal-tolerant individuals. Considerable genetic diversity was found within the geographical subpopulations, but no reduction of the genetic diversity, current or historic, was observed in subpopulations inhabiting polluted soils. The genetic differentiation between the geographical subpopulations was low, and no evidence for clustering of subpopulations from polluted soils vs. subpopulations from nonpolluted soils was found. These results indicate that heavy metal pollution has a limited effect on the genetic structure of S. luteus populations, and this may be due to the high frequency of sexual reproduction and extensive gene flow in S. luteus, which allows rapid evolution of the tolerance trait while maintaining high levels of genetic diversity.  相似文献

We investigated the distribution of genetic variation within and between seven subpopulations in a riparian population of Silene tatarica in northern Finland by using amplified fragment length polymorphism (AFLP) markers. A Bayesian approach-based clustering program indicated that the marker data contained not only one panmictic population, but consisted of seven clusters, and that each original sample site seems to consist of a distinct subpopulation. A coalescent-based simulation approach shows recurrent gene flow between subpopulations. Relative high FST values indicated a clear subpopulation differentiation. However, amova analysis and UPGMA-dendrogram did not suggest any hierarchical regional structuring among the subpopulations. There was no correlation between geographical and genetic distances among the subpopulations, nor any correlation between the subpopulation census size and amount of genetic variation. Estimates of gene flow suggested a low level of gene flow between the subpopulations, and the assignment tests proposed a few long-distance bidirectional dispersal events between the subpopulations. No apparent difference was found in within-subpopulation genetic diversity among upper, middle and lower regions along the river. Relative high amounts of linkage disequilibrium at subpopulation level indicated recent population bottlenecks or admixture, and at metapopulation levels a high subpopulation turnover rate. The overall pattern of genetic variation within and between subpopulations also suggested a 'classical' metapopulation structure of the species suggested by the ecological surveys.  相似文献

Microhabitat heterogeneity can lead to fine‐scale local adaptation when gene flow is restricted, which may be important for the maintenance of genetic variation within populations. This study tested whether microhabitat heterogeneity was associated with trait differences in a population of Arabidopsis lyrata and studied its impact on the genetic variance–covariance (G) matrix. Maternal seed families were collected from dune tops and bottoms, two microhabitats known to vary significantly in water availability. In a common garden experiment, replicate individuals per family were raised under wet and dry conditions, and physiological, morphological and life‐history traits were assessed. Plants from the two microenvironments differed in their response to treatment in two performance components, in stomata density and most strongly in flowering time. Under wet conditions, plants originating from dune bottoms flowered 4 weeks earlier than those from dune tops. Only one of three G‐matrix comparisons revealed that habitat heterogeneity and evolutionary potential were positively linked. The number of independent trait dimensions was larger in the entire population than within subpopulations separated by microhabitat under wet conditions. However, the size of the G‐matrix was no larger in the entire population than within subpopulations separated by microhabitat, and trait correlation structure between microhabitats and treatments was not significantly different. These results indicate that fine‐scale habitat heterogeneity likely led to local adaptation, which weakly affected levels of across‐trait genetic variation.  相似文献
