We report the effect of UV-B radiation (0.8 ± 0.1 mW cm−2) and UV-B radiation supplemented with low-intensity PAR (∼80 μmol photons m−2 s−1) on the photosynthesis, photosynthetic pigments, phosphoglycolipids, oxidative damage, enzymatic antioxidants, and UV-absorbing compounds in Phormidium tenue, a marine cyanobacterium. UV-B radiation resulted in a decline in photosynthesis and photosynthetic pigments leading to lower biomass. P. tenue synthesized UV-absorbing compounds like mycosporine-like amino acids (MAAs) and scytonemin in response to UV-B radiation. Quantity of MAAs and scytonemin was higher when UV-B was supplemented with low-level PAR. UV-B treatment also resulted in quantitative changes in phosphoglycolipids of the membrane. The UV-B treatment resulted in a slight increase in the level of peroxidation of cell membrane and very little increase in the activity of superoxide dismutase (SOD). Results indicate that UV-B affected photosynthesis and that the main protective system was the synthesis of MAAs and scytonemin-like compounds rather than antioxidant enzymes such as SOD.  相似文献

UV-B radiation (0.8 +/- 0.1 mW cm(-2)) and UV-B radiation supplemented with low intensity PAR (approximately 80 micro mol m(-2) s(-1)) affected photosynthesis at the level of antenna system as well as PS II reaction centre (Fo and Fm declined) in Phormidium corium (Agardh) Gomont. UV-B radiation resulted in decline in sugar content, peroxidation of membrane lipids as well as quantitative and qualitative changes in phosphoglycolipids and neutral lipids. Fatty acid profile did not show any qualitative changes due to the treatment, however, UV-B supplemented with low PAR resulted in slightly higher level of unsaturation. P. corium synthesized MAAs in response to UV-B. Quantity of MAAs increased when UV-B treatment was supplemented with low level PAR.  相似文献

Enzymatic and non-enzymatic defense strategies against ultraviolet-B radiation (UV-B, 280–315 nm) were studied in Anabaena doliolum and Anabaena strain L31, two of the most common strains of Indian cyanobacteria. Upon UV-B irradiation, both strains showed a 2–5-fold increase in antioxidative enzymes, such as superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and peroxidase (POD), as compared to non-irradiated control cultures. These enzymes scavenge damaging reactive oxygen species (ROS), generated by UV-B radiation inside the cells. In addition, these organisms also synthesize mycosporine-like amino acids (MAAs) which are able to carry out UV-screening and/at the same time as UV-quenching. The identification and characterization of three types of MAAs from both Anabaena species were performed using absorption spectroscopy, high performance liquid chromatography (HPLC), electro spray ionization-mass spectrometry (ESI-MS), Fourier Transform Infrared (FTIR) spectroscopy and Nuclear Magnetic Resonance (NMR) spectroscopy. Shinorine was found to be the most common MAA in both Anabaena species while porphyra-334 and mycosporine-glycine were present only in A. doliolum. The results of the present investigation clearly demonstrate that both enzymatic and non-enzymatic defense mechanisms are being employed by A. doliolum and Anabaena strain L31 to counteract the damaging effects of UV-B radiation.  相似文献

Effects of two intensities (1 and 5 W?m?2) of UV-B radiation on the synthesis of UV-absorbing compounds in a terrestrial cyanobacterium Nostoc flagelliforme were investigated. UV-B radiation resulted in lower biomass. Short period (less than 12 h) of UV-B radiation caused an increase of chlorophyll a content, but subsequent duration of treatment (more than 24 h) resulted in a rapid decrease. N. flagelliforme synthesized UV-absorbing compounds such as scytonemin and mycosporine-like amino acids (MAAs) in response to UV-B radiation. Upon 48 h of exposure to UV-B radiation, scytonemin content in cells increased by 103.8 and 164.0 % at 1 and 5 W?m?2, respectively. Oligosaccharide-linked mycosporine-like amino acids increased by 145.5 % after 12 h at 5 W?m?2 and 114.5 % after 48 h at 1 W?m?2 UV-B radiation. HPLC analysis showed that nine MAAs existed in N. flagelliforme cells both from liquid suspension culture and field colony. But the concentration and kinds of them were different. At the two distinct levels of UV-B radiation, the content of particular MAAs increased, declined, or remained unchanged. Moreover, the appearance of two new MAAs was observed.  相似文献

In field studies conducted at the Kongsfjord (Spitsbergen), the effect of filtered natural radiation conditions (solar without ulraviolet [UV]-A+UV-B, solar without UV-B, solar) on photosynthesis and the metabolism of UV-absorbing mycosporine-like amino acids (MAAs) in the marine red alga Devaleraea ramentacea have been studied. While solar treatment without UV-A+UV-B did not affect photosynthesis during the course of a day, solar without UV-B and the full solar spectrum led to a strong inhibition. However, after offset of the various radiation conditions, all algae fully recovered. Isolates collected from different depths were exposed in the laboratory to artificial fluence rates of photosynthetic active radiation (PAR), PAR+UV-A, and PAR+UV-A+UV-B. The photosynthetic capacity was affected in accordance with the original sampling depth, i.e. shallow-water isolates were more resistant than algae from deeper waters, indicating that D. ramentacea is able to acclimate to changes in irradiance. Seven different UV-absorbing MAAs were detected in this alga, namely mycosporine-glycine, shinorine, porphyra-334, palythine, asterina-330, palythinol, and palythene. The total amount of MAAs continuously decreased with increasing collecting depth when sampled in mid June, and algae taken in late August from the same depths contained on average 30–45% higher MAA concentrations, indicating a seasonal effect as well. The presence of increasing MAA contents with decreasing depth correlated with a more insensitive photosynthetic capacity under both UV-A and UV-B treatments. Populations of D. ramentacea collected from 1 m depth, with one fully exposed to solar radiation and the other growing protected as understorey vegetation underneath the kelp Laminaria saccharina, exhibited quantitatively different MAA compositions in the apices. The exposed seaweeds contained 2.5-fold higher MAA values compared with the more shaded algae. Moreover, the exposed isolates showed a strong tissue gradient in MAAs, pigments, and proteins. The green apices contained 5-fold higher MAA contents than the red bases. Transplantation of D. ramentacea from 2 m depth to the surface induced the formation and accumulation of MAAs after 1 week exposure to the full solar spectrum. Control samples which were treated with the solar spectrum without UV-A+B or with solar without UV-B showed unchanged MAA contents, indicating a strong UV-B effect on MAA metabolism. All data well supported the suggested physiological function of MAAs as natural UV sunscreens in macroalgae.  相似文献

The invasive success of Gracilaria vermiculophylla has been attributed to its wide tolerance range to different abiotic factors, but its response to ultraviolet radiation (UVR) is yet to be investigated. In the laboratory, carpospores and vegetative thalli of an Atlantic population were exposed to different radiation treatments consisting of high PAR (photosynthetically active radiation) only (P), PAR+UV-A (PA) and PAR+UV-A+UV-B (PAB). Photosynthesis of carpospores was photoinhibited under different radiation treatments but photosystem II (PSII) function was restored after 12 h under dim white light. Growth of vegetative thalli was significantly higher under radiation supplemented with UVR. Decrease in chlorophyll a (Chl a) under daily continuous 16-h exposure to 300 μmol photons m(-2) s(-1) of PAR suggests preventive accumulation of excited chlorophyll molecules within the antennae to minimize the generation of dangerous reactive oxygen species. Moreover, an increase in total carotenoids and xanthophyll cycle pigments (i.e. violaxanthin, antheraxanthin and zeaxanthin) further suggests effective photoprotection under UVR. The presence of the ketocarotenoid β-cryptoxanthin also indicates protection against UVR and oxidative stress. The initial concentration of total mycosporine-like amino acids (MAAs) in freshly-released spores increased approximately four times after 8-h laboratory radiation treatments. On the other hand, initial specific MAAs in vegetative thalli changed in composition after 7-day exposure to laboratory radiation conditions without affecting the total concentration. The above responses suggest that G. vermiculophylla have multiple UVR defense mechanisms to cope with the dynamic variation in light quantity and quality encountered in its habitat. Beside being eurytopic, the UVR photoprotective mechanisms likely contribute to the current invasive success of the species in shallow lagoons and estuaries exposed to high solar radiation.  相似文献

Caryopteris mongolica is a dwarf shrub mainly found in grassland and desert areas of north-west China, and which can survive severe environmental stress. This study aimed to assess the responses of the flavonoid pathway to UV-B radiation treatments and its correlation to the lipid peroxide and antioxidant systems in C. mongolica. In UV-B radiation experiments, plants were exposed to UV-B radiation treatments with a intensity of 30 J/s for 1, 4 and 24 h, respectively. A control group without UV-B radiation treatment was also used. The chlorophyll fluorescence parameters, contents of chlorophyll and carotenoid, levels of lipid peroxidation, activities of antioxidant system enzymes, accumulations of total flavonoids and anthocyanins, and activities of phenylalanine ammonialyase (PAL) and chalcone isomerase (CHI) under different UV-B radiation treatments were investigated. The correlations between products and key enzymes in the flavonoid pathway and the lipid peroxide and antioxidant systems were also analyzed. The results showed that chlorophyll fluorescence parameters decreased within 24 h of treatment. The chlorophyll contents decreased within 4 h and remained stable after 24 h. Carotenoid content significantly increased. The level of MDA, the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX) and peroxidase (POD) and the contents of total flavonoids and anthocyanidins increased, while catalase (CAT) activity decreased under UV-B stress. The activities of PAL and CHI also increased with the increased content of total flavonoids. The flavonoid products anthocyanidins had a significant positive correlation with MDA level, as well as the activities of antioxidant enzyme SOD. In conclusion, UV-B radiation induced the degradation of photosynthetic pigments and decreased photochemical efficiency of Photosystem II; increased the contents of MDA, total flavonoids and anthocyanidins; and also enhanced activities of antioxidant enzymes (SOD, APX and POD) and key enzymes (PAL and CHI) in the flavonoid pathway in C. mongolica. Thus, we speculate that the flavonoid pathway were involved in the regulation of stress resistance in C. mongolica.  相似文献

Under natural conditions, Chroothece richteriana synthesizes a fairly high proportion of fatty acids. However, nothing is known about how environmental changes affect their production, or about the production of protective compounds, when colonies develop under full sunshine with high levels of UV radiation. In this study, wild colonies of C. richteriana were subjected to increasing temperature, conductivity, ammonium concentrations and photosynthetically active radiation (PAR), and UV radiations to assess the potential changes in lipid composition and mycosporine‐like amino acids (MAAs) concentration. The PERMANOVA analysis detected no differences for the whole fatty acid profile among treatments, but the percentages of α‐linolenic acid and total polyunsaturated fatty acids increased at the lowest assayed temperature. The percentages of linoleic and α‐linolenic acids increased with lowering temperature. γ‐linolenic and arachidonic acids decreased with increasing conductivity, and a high arachidonic acid concentration was related with increased conductivity. The samples exposed to UVB radiation showed higher percentages of eicosapentaenoic acid and total monounsaturated fatty acids, at the expense of saturated fatty acids. MAAs accumulation increased but not significantly at the lowest conductivity, and also with the highest PAR and UVR exposure, while ammonium and temperature had no effect. The observed changes are probably related with adaptations of both membrane fluidity to low temperature, and metabolism to protect cells against UV radiation damage. The results suggest the potential to change lipid composition and MAAs concentration in response to environmental stressful conditions due to climate change, and highlight the interest of the species in future research about the biotechnological production of both compound types.  相似文献

Continuous depletion of the stratospheric ozone layer has resulted in an increase in solar ultraviolet-B (UV-B; 280–315 nm) radiation reaching the Earth's surface. The consequences for aquatic phototrophic organisms of this small change in the solar spectrum are currently uncertain. UV radiation has been shown to adversely affect a number of photochemical and photobiological processes in a wide variety of aquatic organisms, such as cyanobacteria, phytoplankton and macroalgae. However, a number of photosynthetic organisms counteract the damaging effects of UV-B by synthesizing UV protective compounds such as mycosporine-like amino acids (MAAs) and the cyanobacterial sheath pigment, scytonemin. The aim of this contribution is to discuss the responses of algae and cyanobacteria to solar UV-B radiation and the role of photoprotective compounds in mitigating UV-B damage.  相似文献

以"丹麦旺盛菠菜"为材料,通过UV-B和CaCl2复合处理,测定光合色素含量、Hill反应活力、叶绿素荧光、MDA含量和抗氧化酶活性等参数,探讨了CaCl2对UV-B辐射下菠菜叶片电子传递链和光合膜酶保护系统的影响。结果表明,UV-B处理下,光合色素含量、chl/car、类囊体膜上PSII潜在活性(Fv/Fo)、光化学淬灭系数(qP)、非光化学淬灭系数(qN)、PSII光量子产量(ΦPSⅡ)、原初光能转化效率(Fv/Fm),以及Hill反应活力等降低,chla/chlb和MDA含量升高;喷洒CaCl2可不同程度缓解UV-B的伤害。不同处理下,POD、SOD和CAT活性的变化呈现补偿效应。UV-B强度与菠菜叶片PSII功能受损程度呈正相关,CaCl2则主要通过提高chlb含量、类囊体膜上的光量子产量和POD活性,以缓解伤害。重度UV-B辐射下,CaCl2使chlb含量显著提高可能是导致PSII捕光效率提高的重要因素。  相似文献

Microcoleus chthonoplastes constitutes one of the dominant microorganisms in intertidal microbial mat communities. In the laboratory, the effects of repeated daily exposure to ultraviolet radiation (16:8 light:dark cycle) was investigated in unicyanobacterial cultures isolated from three different localities (Baltic Sea = WW6; North Sea = STO and Brittany = BRE). Photosynthesis and growth were measured in time series (12–15 days) while UV-absorbing mycosporine-like amino acids (MAAs) and cellular integrity were determined after 12 and 3 days exposure to three radiation treatments [PAR (22 μmol photon m−2 s−1) = P; PAR + UV-A (8 W m−2) = PA; PAR + UV-A + UV-B (0.4 W m−2) = PAB]. Isolate-specific responses to UVR were observed. The proximate response to radiation stress after 1-day treatment showed that isolate WW6 was the most sensitive to UVR. However, repeated exposure to radiation stress indicated that photosynthetic efficiency (F v/F m) of WW6 acclimated to UVR. Conversely, although photosynthesis in STO exhibited lower reduction in F v/F m during the first day, the values declined over time. The BRE isolate was the most tolerant to radiation stress with the lowest reduction in F v/F m sustained over time. While photosynthetic efficiencies of different isolates were able to acclimate to UVR, growth did not. The discrepancy seems to be due to the higher cell density used for photosynthesis compared to the growth measurement. Apparently, the cell density used for photosynthesis was not high enough to offer self-shading protection because cellular damage was also observed in those filaments under UVR. Most likely, the UVR acclimation of photosynthesis reflects predominantly the performance of the surviving cells within the filaments. Different strategies were observed in MAAs synthesis. Total MAAs content in WW6 was not significantly different between all the radiation treatments. In contrast, the additional fluence of UV-A and UV-B significantly increased MAAs synthesis and accumulation in STO while only UV-B fluence significantly increased MAAs content in BRE. Regardless of the dynamic photosynthetic recovery process and potential UV-protective functions of MAAs, cellular investigation showed that UV-B significantly contributed to an increased cell mortality in single filaments. In their natural mat habitat, M. chthonoplastes benefits from closely associated cyanobacteria which are highly UVR-tolerant due to the production of the extracellular UV-sunscreen scytonemin.  相似文献

Net photosynthesis, growth, and ultraviolet (UV) radiation absorbance were determined for the first leaf of Cucurbita pepo L. exposed to two levels of UV-B irradiation and a UV-B radiation-free control treatment. Absorbance by extracted flavonoid pigments and other UV-B radiation-absorbing compounds from the first leaves increased with time and level of UV-B radiation impinging on leaf surfaces. Although absorbance of UV-B radiation by extracted pigments increased substantially, UV-B radiation attenuation apparently was insufficient to protect completely the photosynthetic apparatus or leaf growth processes. Leaf expansion was repressed by daily exposure to 1365 Joules per meter per day of biologically effective UV-B radiation but not by exposure to 660 Joules per meter per day. Photosynthesis measured through ontogenesis of the first leaf was depressed by both UV-B radiation treatments. Repression of photosynthesis by UV-B radiation was especially evident during the ontogenetic period of maximum photosynthetic activity.  相似文献

The UV-absorbing mycosporine-like amino acids (MAAs) are hypothesized to protect organisms against harmful UV radiation (UVR). Since the physiology and metabolism of these compounds are unknown, the induction and kinetics of MAA biosynthesis by various natural radiation conditions were investigated in the marine red alga Chondrus crispus collected from Helgoland, Germany. Three photosynthetically active radiation (PAR, 400–700 nm) treatments without UVR and three UV-A/B (290–400 nm) treatments without PAR were given. Chondrus crispus collected from 4–6 m depth contained only traces of the MAA palythine. After 24 h exposure to 100% ambient PAR, traces of three additional MAAs, shinorine, palythinol and palythene, were detected, and their concentrations increased strongly during a one-week exposure to all PAR treatments. The concentration of all MAAs varied directly with PAR dose, with palythine and shinorine being four- to sevenfold higher than palythinol and palythene. Likewise, naturally high doses of both UV-A and UV-B resulted in a strong accumulation of all MAAs, in particular shinorine. While shinorine accumulation was much more stimulated by UVR, the content of all other MAAs was more affected by high PAR, indicating an MAA-specific induction triggered by UVR or PAR. Cyanobacteria that form the primary components of microbial mats in freshwater bogs and intertidal marine environments in the Bahamas produce water-soluble brown pigments whose spectral properties imply that they are a type of humic acid. These “humic pigments” are produced by vital processes of living cyanobacteria, not by decomposition of dead ones, as shown by decreases in the concentrations of humic pigments, ultraviolet (UV) radiation-absorbing photoprotective mycosporine-like amino acids (MAAs), and chlorophyll from upper to lower layers of the mats, and by the occurrence of humic pigments in cyanobacterial cultures. Unlike MAAs, which absorb UV radiation only within limited ranges of wavelengths, humic pigments absorb radiation spanning the entire UV spectrum, and absorbance increases with decreasing wavelength. These observations suggest that the biosynthesis of humic pigments originated as a photoprotective adaptation in the early Precambrian, enabling cyanobacteria to colonize shallow-water and terrestrial environments even though the atmosphere was virtually devoid of O2 and O3 and therefore transparent to all solar radiation in the UV region of the spectrum. Moreover, the evolution of this photoprotective mechanism may have been linked to the evolution of photosynthesis.  相似文献

We tested the hypothesis that there is a relation between phylotypes (phylogenetic types, as determined by restriction fragment length polymorphism (RFLP) and partial sequence analysis of the small subunit ribosomal RNA gene (SSUrDNA)) and the synthesis of mycosporine-like amino acids (MAAs) by symbiotic dinoflagellates under the influence of ultraviolet radiation (UV-B/A) and photosynthetically active radiation (PAR). We exposed 27 isolates of symbiotic dinoflagellates simultaneously to UV-B/A and PAR, and subsequently determined the MAAs present in cell extracts and in the media. The algae used included 24 isolates of Symbiodinium spp. originating from jellyfishes, sea anemones, zoanthids, scleractinians, octocorals, and bivalves, and three others in the genera Gymnodinium, Gloeodinium and Amphidinium from a jellyfish, an hydrocoral and a flatworm, respectively. In this study, all of the phylotype A Symbiodinium spp. synthesized up to three identified MAAs. None of the 11 cultured phylotypes B and C Symbiodinium spp. synthesized MAAs. The three non-Symbiodinium symbionts also synthesized up to three MAAs. The results support a conclusion that phylotype A Symbiodinium spp. have a high predilection for the synthesis of MAAs, while phylotypes B and C do not. Synthesis of MAAs by symbiotic dinoflagellates in culture does not appear to relate directly to depths or to the UV exposure regimes from which the consortia were collected.  相似文献

温室种植的吞豆在0(CK),8.82kJ/m2(T1)和12.6kJ/m2(T2)3种剂量的紫外线B(UV-B)辐射引起膜脂变化及其机制的研究结果表明,UV0B处理后,蚕豆叶片中丙二醛(MDA)和H2O2含量升高,膜脂肪酸不饱和度指数(IUFA)降低,脂氧合酶(LOX)活性升高,超氧歧化酶(SOD)活性稍有波动,而3种多胺-腐胺(Put)精胺(Spd)和尸胺(Spn)在照射7天后均有积累,但在处理后期(21d)有所回落,推测由LOX主导的酶促膜过氧化作用和氧自由基引起的非酶促过作用在膜结构的破坏中起重要作用,SOD活性和多胺含量的变化蚕豆对UV-B胁迫的一种适应性生理反应。  相似文献

In the present study, the impact of low fluence rate of UV-B (0.045 W.m?2) on biomass production, photosynthetic pigments (chlorophyll a, carotenoids, and phycobiliproteins), chlorophyll fluorescence, nonenzymatic antioxidants: proline, ascorbate, cysteine, and nonprotein thiols, total phenolic contents, and antioxidant potential (radical scavenging activity) was investigated in three cyanobacteria, viz. Nostoc muscorum, Phormidium foveolarum, and Arthrospira platensis. Selected fluence rate of UV-B caused enhancing effect on these parameters; however, the increased values of these attributes were greater in A. platensis followed by P. foveolarum and N. muscorum. Results indicate that UV-B (at selected fluence rate) could be used as technique that may modify cyanobacterial system for efficient and economic production of natural food supplements and/or natural pharmaceuticals.  相似文献

Arabidopsis thaliana . Three-week-old plants were exposed to a high temperature (30 C), an enhanced light intensity (200 μE/m2/sec), water deficiency (water deprivation for 2 days), a chilling temperature (5 C), or ultraviolet-B (UV-B) radiation (0.25 or 0.094 W/m2) for 1 week (except for water deficiency). The high temperature and enhanced light treatments increased only dehydroascorbate reductase (DHAR) activity. Water deficiency enhanced the activities of DHAR and guaiacol peroxidase (PER). Chilling temperature increased the activities of ascorbate peroxidase (APX) and glutathione reductase (GR), whereas it decreased catalase (CAT) activity. UV-B at an intensity of 0.25 W/m2 elevated the activities of APX, monodehydroascorbate reductase (MDHAR), GR, PER and superoxide dismutase (SOD). It was suggested that the amounts of phenylpropanoid compounds increased during treatments of plants with enhanced light intensity, chilling temperature, and UV-B. These results suggest that some differences exist among the oxidative stress conditions caused by the different treatments, although all of these treatments seem to be related to active oxygen production. We propose that in A. thaliana, environmental stresses may be classified into those which induce DHAR activity and those which induce APX activity. The effects and interaction of drought and UV-B radiation were studied in sunflower plants (Helianthus annuus L. var. Catissol-01), growing in a greenhouse under natural photoperiod conditions. The plants received approximately 1.7 W m(-2) (controls) or 8.6 W m(-2) (+UV-B) of UV-B radiation for 7 h per day. The UV-B and water stress treatments started 18 days after sowing. After a period of 12 days of stress, half of the water-stressed plants (including both UV-B irradiated or non-irradiated) were rehydrated. Both drought and UV-B radiation treatments resulted in lower shoot dry matter per plant, but there was no significant interaction between the two treatments. Water stress and UV-B radiation reduced photosynthesis, stomatal conductance and transpiration. However, the amplitude of the effects of both stressors was dependent on the interactions. This resulted in alleviation of the negative effect of drought on photosynthesis and transpiration by UV-B radiation as the water stress intensified. Intercelluar CO(2) concentration was initially reduced in all treatments compared to control plants but it increased with time. Photosynthetic pigments were not affected by UV-B radiation. Water stress reduced photosynthetic pigments only under high UV-B radiation. The decrease was more accentuated for chlorophyll a than for chlorophyll b. As a measure for the maximum efficiency of photosystem II in darkness F (v)/F (m) was used, which was not affected by drought stress but initially reduced by UV-B radiation. Independent of water supply, UV-B radiation increased the activity of pirogalol peroxidase and did not increase the level of malondialdehyde. On the other hand, water stress did not alter the activity of pirogalol peroxidase and caused membrane damage as assessed by lipid peroxidation. The application of UV-B radiation together with drought seemed to have a protective effect by lowering the intensity of lipid peroxidation caused by water stress. The content of proline was not affected by UV-B radiation but was increased by water stress under both low and high UV-B radiation. After 24 h of rehydration, most of the parameters analyzed recovered to the same level as the unstressed plants.  相似文献

The present investigation is aimed to understand how a bloom-forming cyanobacterium Microcystis aeruginosa adapts to changing climatic conditions. The cyanobacterium was exposed to stresses of UV-B (2 Wm?2) radiation and temperature (45 °C) for desired time intervals. Results showed that both the stresses affect growth and photosynthetic efficiency of M. aeruginosa. More than 50% loss of survival and content of photosynthetic pigments was noted after 4 h treatment of both the above stresses. Such changes were mainly due to the generation of reactive oxygen species which cause damage to proteins, DNA, lipids, and modulation of the membrane stability. An increase in the proline accumulation was noted in the cells which probably negates the harmful effects. In addition, activity of antioxidative enzymes namely, catalase, superoxide dismutase, ascorbate peroxidase, and peroxidase was induced by 1.5–3.0-fold on 3 h of UV-B and temperature treatment indicating their possible role in protection. Interestingly, induction of photoprotective compound, mycosporine-like amino acids (MAAs) were also found under UV-B stress which might be an additional strategy of defense mechanism for the survival of the cyanobacterium. Analysis of photoprotective compound revealed shinorine as the main MAA synthesized by the cyanobacterium.  相似文献
