The effects of both 3,5,3'-triiodo-L-thyronine and spontaneous metamorphosis on Rana catesbeiana liver mRNA were studied using in vitro translation of isolated liver poly(A)+ RNA in a rabbit reticulocyte lysate system. Conventional phenol extraction methods yielded degraded RNA due to high levels of endogenous ribonucleases released upon homogenization of Rana catesbeiana liver. Isolation of intact total RNA was achieved using the potent ribonuclease denaturant, guanidinium thiocyanate. Adult bullfrog serum albumin was purified to homogeneity and a monospecific antibody was elicited against it. A serum protein of 23,000 daltons that migrated near serum albumin on a 6% native gel was also purified to homogeneity. A monospecific antibody was also raised against this protein. Both antibodies were used to quantitatively immunoprecipitate the in vitro translation products of poly(A)+ RNA isolated at intervals following a single injection of triiodothyronine or during various stages of spontaneous amphibian metamorphosis. Triiodothyronine caused a sevenfold increase in translatable albumin mRNA and a threefold increase in translatable mRNA for the 23,000 dalton protein. These increases are consistent with a nuclear initiated mechanism for thyroid hormone action during amphibian metamorphosis.  相似文献

The heterogeneity of liver mitochondria of the tadpoles, Rana catesbeiana, undergoing metamorphosis was investigated by a combination of pulse-chase labeling of mitochondria with [methyl-3H]thymidine and centrifugation of mitochondria on a density gradient of metrizamide. 2. The liver mitochondria of tadpole at premetamorphic stage are separated into two populations with mean densities of 1.128 (M2) and 1.112 (M3). 3. At metamorphic stage a population with mean densities of 1.174 (M1) appears additionally. 4. The activity of mitochondria to take up [methyl-3H]thymidine in vivo is 2-3 times higher at metamorphic stage than at premetamorphic stage. 5. The M1 population has a prominently high activity to take up L-[4.5-3H]leucine in vitro and also a high specific activity of cytochrome c oxidase. 6. These findings suggest that the mitochondrial populations found are of alternate stages in the mitochondrial maturation.  相似文献

1. Intestinal brush border membrane vesicles have been isolated form Rana catesbeiana tadpole. 2. Electron microscopy of brush border membrane vesicles demonstrates a fairly homogenous preparation of vesicles, some of them still containing electron dense material. 3. The dense vesicles probably comprise both microvillus core and membrane. 4. Negative staining of vesicles reveals the presence of knob-like structures (particles) covering the outer surface of the membrane. 5. The membranous fraction is characterized by a high specific activity of alkaline phosphatase, trehalase, glucoamylase, maltase and gamma-glutamyltranspeptidase.  相似文献

The effect of griseofulvin treatment on the synthesis of cytochrome c oxidase was studied with the liver of the tadpole, Rana catesbeiana. (1) In the liver of tadpole treated with griseofulvin, a ferrochelatase inhibitor, the synthesis of heme a, but not cytochrome c oxidase protein, is inhibited. (2) The apocytochrome c oxidase which is formed in the liver of tadpole treated with griseofulvin is converted to the active holoenzyme by exogenously added heme a.  相似文献

Studies of insulin producing β-cells have reported conflicting responses to NF-κB activation, encompassing both pro- and anti-apoptotic effects, possibly reflecting the use of β-cells from different species. Therefore, the aim of this study was to compare the temporal activation of NF-κB in rat and human insulin producing cells and relate this to the dynamics of cell death, STAT-1 activation and the production of nitric oxide (NO). Rat RIN5AH and human islet cells were exposed to the cytokines IL-1β and IFN-γ and the NOS inhibitor aminoguanidine. Cell death, NO production, IκBα phosphorylation, p65 methylation, STAT-1 phosphorylation and cIAP-2 levels were analyzed at different time-points. Cytokine-induced RIN5AH cell death occurred on day 1, and this was paralleled by NF-κB activation, STAT-1 phosphorylation and production of NO. On the other hand, the human islet cells instead died by an NO-independent mechanism on day 3 and 5. This later occurring cell death was associated with a gradual decrease in IκBα phosphorylation and p65 methylation, and a lowered expression of the NF-κB target genes IκBα and cIAP-2. STAT-1 phosphorylation was persistently high during the entire cytokine exposure period in human islet cells. The results favor a pro-survival role of NF-κB and a pro-apoptotic role of STAT-1 in human islet cells. Thus, rodent insulin producing cells may not be suitable as models for human β-cells in the context of cytokine-induced damage.  相似文献

Spike frequency was recorded in the nerve of the isolated superfused first gill arch of the bullfrog larva, Rana catesbeiana and the response to different superfusate PO2 was evaluated. In the metamorphic tadpole, spike frequency increased significantly when the superfusate PO2 was decreased (mean +/- SEM): 8.5 +/- 1.6 Hz at 650 Torr, 11.7 +/- 1.9 Hz at 140 Torr, 13.3 +/- 1.8 Hz at 65 Torr, 14.8 +/- 2.4 Hz at 0 Torr (ANOVA, p = 0.0002). The O2 sensitive chemoreceptor stimulants NaCN and almitrine also increased the spike frequency. This study demonstrates the presence of O2 sensitive chemoreceptors in the first gill arch of the tadpole.  相似文献

Heme oxygenase in the liver microsomes of tadpole and adult bullfrog, Rana catesbeiana, was studied in relation to hemoglobin metabolism during tadpole development. The results obtained are as follows. 1. The specific activity of the enzyme of premetamorphic tadpole liver was comparable to that of adult frog liver. The apparent Km for methemalbumin was about 25 microM for both tadpole and frog enzymes. 2. The enzyme activity was stimulated in vivo by the injection of methemalbumin, phenylhydrazine and triiodothyronine to the animals. 3. A marked increase in the enzyme activity was found in the liver of tadpole during prometamorphic stage and metamorphic climax.  相似文献

Four major components of the hemoglobin of the bullfrog tadpole, Rana catesbeiana, have been isolated and characterized structurally and functionally. These components fall into two clear functional classes. Components I and II have substantially higher affinities for oxygen than do components III and IV. Components I and II predominate in very young tadpoles and are largely replaced by components III and IV in older tadpoles. The data (Broyles, R.H., and Frieden, E. (1973) Nature New Biol. 241, 207-209) indicate that component I arises in the kidney and components III and IV in the liver. The synchrony of appearance and functional similarity o components I and II suggest that component II probably also arises in the kidney. Amphibian metamorphosis affords a useful experimental system in which to study thyroid hormone regulation of gene expression during postembryonic vertebrate development. In order to isolate gene-specific cDNA probes which correspond to thyroid hormone-responsive mRNAs, we employed differential colony hybridization of a cDNA library constructed from poly(A)+ RNA of thyroxine-treated premetamorphic tadpole liver. From an initial screening of about 6000 transformants, 32 "potentially positive" colonies were obtained. The recombinant cDNA-plasmids from 13 of these colonies plus two "potentially negative" colonies were purified for further study. Southern blot analysis of the plasmid DNA was employed to determine whether different cDNAs encoded for the same mRNA. The effect of thyroid hormone on the relative levels of specific mRNA species was examined by Northern analysis of liver RNA from premetamorphic tadpoles, thyroxine-treated tadpoles, and adult bullfrogs. Three independent cDNA clones were obtained which encoded thyroid hormone-enhanced mRNAs. We also obtained two independent cDNA clones encoding thyroid hormone-inhibited mRNAs and three independent clones encoding thyroid hormone-unresponsive mRNAs. The levels of two thyroid hormone-enhanced mRNAs and one thyroid hormone-inhibited mRNA were essentially the same in the thyroid hormone-treated tadpole liver and adult liver, suggesting that thyroid hormone induces stable changes in liver gene expression during spontaneous metamorphosis. Using selected cDNAs, RNA dot blot analysis of liver mRNA from tadpoles at different stages of metamorphosis showed that the level of one thyroid hormone-enhanced mRNA increased during late prometamorphosis and metamorphic climax. Similarly, a mRNA which was strongly inhibited by thyroid hormone treatment was observed to decline during prometamorphosis and reach undetectable levels during metamorphic climax. One mRNA was detected which was reproducibly inhibited by thyroid hormone treatment but which remained essentially unchanged during spontaneous metamorphosis. These results provide the first direct evidence for the coordinate and selective pretranslational regulation by thyroid hormone of several liver genes during the developmental process of metamorphosis.  相似文献

During both spontaneous and thyroid hormone (TH)-induced metamorphosis, the Rana catesbeiana tadpole undergoes postembryonic developmental changes in its liver which are necessary for its transition from an ammonotelic larva to a ureotelic adult. Although this transition ultimately results from marked increases in the activities and/or de novo synthesis of the urea cycle enzymes, the precise molecular means by which TH exerts this tissue-specific response are presently unknown. Recent reports, using RNA from whole Xenopus laevis tadpole homogenates and indirect means of measuring TH receptor (TR) mRNAs, suggest a correlation between the up-regulation of TR beta-mRNAs and the general morphological changes occurring during amphibian metamorphosis. To assess whether or not this same relationship exists in a TH-responsive tissue, such as liver, we isolated and characterized a cDNA clone containing the complete nucleotide sequence for a R. catesbeiana urea cycle enzyme, ornithine transcarbamylase (OTC), as well as a genomic clone containing a portion of the hormone-binding domain of a R. catesbeiana TR beta gene. Through use of these homologous sequences and a heterologous cDNA fragment encoding rat carbamyl phosphate synthetase (CPS), we directly determined the relative levels of the TR beta, OTC, and CPS mRNAs in liver from spontaneous and TH-induced tadpoles. Our results establish that TH affects an up-regulation of mRNAs for its own receptor prior to up-regulating CPS and OTC mRNAs. Moreover, results with cultured tadpole liver demonstrate that TH, in the absence of any other hormonal influence, can affect an up-regulation of both the TR beta and OTC mRNAs.  相似文献
