Previous studies have shown that pituitary growth hormone displays an episodic pattern of evolution, with a slow underlying
evolutionary rate and occasional sustained bursts of rapid change. The present study establishes that pituitary prolactin
shows a similar pattern. During much of tetrapod evolution the sequence of prolactin has been strongly conserved, showing
a slow basal rate of change (approx 0.27 × 109 substitutions/amino acid site/year). This rate has increased substantially (∼12- to 38-fold) on at least four occasions during
eutherian evolution, during the evolution of primates, artiodactyls, rodents, and elephants. That these increases are real
and not a consequence of inadvertant comparison of paralogous genes is shown (for at least the first three groups) by the
fact that they are confined to mature protein coding sequence and not apparent in sequences coding for signal peptides or
when synonymous substitutions are examined. Sequences of teleost prolactins differ markedly from those of tetrapods and lungfish,
but during the course of teleost evolution the rate of change of prolactin has been less variable than that of growth hormone.
It is concluded that the evolutionary pattern seen for prolactin shows long periods of near-stasis interrupted by occasional
bursts of rapid change, resembling the pattern seen for growth hormone in general but not in detail. The most likely basis
for these bursts appears to be adaptive evolution though the biological changes involved are relatively small.

The GH gene cluster in marmoset, Callithrix jacchus, comprises eight GH-like genes and pseudogenes and appears to have arisen as a consequence of gene duplications occurring
independently of those leading to the human GH gene cluster. We report here the complete sequence of the marmoset GH gene
locus, including the intergenic regions and 5′ and 3′ flanking sequence, and a study of the multiple GH-like genes of an additional
New World monkey (NWM), the white-fronted capuchin, Cebus albifrons. The marmoset sequence includes 945 nucleotides (nt) of 5′ flanking sequence and 1596 nt of 3′ flanking sequence that are
“unique”; between these are eight repeat units, including the eight GH genes/pseudogenes. The breakpoints between these repeats
are very similar, indicating a regular pattern of gene duplication. These breakpoints do not correspond to those found in
the much less regular human GH gene cluster. This and phylogenetic analysis of the repeat units within the marmoset gene cluster
strongly support the independent origin of these gene clusters, and the idea that the episode of rapid evolution that occurred
during GH evolution in primates preceded the gene duplications. The marmoset GH gene cluster also differs from that of human
in having fewer and more evenly distributed Alu sequences (a single pair in each repeat unit) and a “P-element” upstream of every gene/pseudogene. In human there is no P-element
upstream of the gene encoding pituitary GH, and these elements have been implicated in placental expression of the other genes
of the cluster. The GH gene clusters in marmoset and capuchin appear to have arisen as the consequence of a single-gene duplication
event, but in capuchin there was then a remarkable expansion of the GH locus, giving at least 40 GH-like genes and pseudogenes.
Thus even among NWMs the GH gene cluster is very variable.

The sequence of growth hormone (GH) is generally strongly conserved in mammals, but episodes of rapid change occurred during the evolution of primates and artiodactyls, when the rate of GH evolution apparently increased substantially. As a result the sequences of higher primate and ruminant GHs differ markedly from sequences of other mammalian GHs. In order to increase knowledge of GH evolution in Cetartiodactyla (Artiodactyla plus Cetacea) we have cloned and characterized GH genes from camel (Camelus dromedarius), hippopotamus (Hippopotamus amphibius), and giraffe (Giraffa camelopardalis), using genomic DNA and a polymerase chain reaction technique. As in other mammals, these GH genes comprise five exons and four introns. Two very similar GH gene sequences (encoding identical proteins) were found in each of hippopotamus and giraffe. The deduced sequence for the mature hippopotamus GH is identical to that of dolphin, in accord with current ideas of a close relationship between Cetacea and Hippopotamidae. The sequence of camel GH is identical to that reported previously for alpaca GH. The sequence of giraffe GH is very similar to that of other ruminants but differs from that of nonruminant cetartiodactyls at about 18 residues. The results demonstrate that the apparent burst of rapid evolution of GH occurred largely after the separation of the line leading to ruminants from other cetartiodactyls. 相似文献

Researchers are still discussing the classification of Nycticebus. We established a molecular phylogeny covering all recognized taxa in Nycticebus to provide information for further evaluation. We sequenced partial D-loop (ca. 390 bp) and cytochrome b genes (425 bp) from 22 specimens. We separated most of the major groups except for some mixing of Nycticebus. coucang coucang and N. bengalensis. Nycticebus pygmaeus diverged earlier from the ancestral stock than the other taxa. Nycticebus coucang menagensis was well discriminated from N. c. coucang. It may be possible to explain the mixing of Nycticebus coucang coucang and N. bengalensis by the hybridization between the 2 groups in the field. Although our data did not provide direct evidence for or against the new proposal that Nycticebus coucang javanicus be raised to the rank of a distinct species (N. javanicus), we have good evidence for regarding N. c. menagensis as a species.Jing-Hua Chen and Deng Pan made equal contributions to the work. 相似文献

Biologists and historians often present natural history and molecular biology as distinct, perhaps conflicting, fields in
biological research. Such accounts, although supported by abundant evidence, overlook important areas of overlap between these
areas. Focusing upon examples drawn particularly from systematics and molecular evolution, I argue that naturalists and molecular
biologists often share questions, methods, and forms of explanation. Acknowledging these interdisciplinary efforts provides
a more balanced account of the development of biology during the post-World War II era.

Zoos and related facilities in North America currently manage five species in the primate family Lorisidae: the greater (Nycticebus coucang), Bengal (N. bengalensis) and pygmy (N. pygmaeus) slow lorises, red slender loris (Loris tardigradus), and potto (Perodicticus potto). We used an online survey to describe institutional housing and husbandry practices for these species and assess the extent to which practices are consistent with established guidelines. Our results show that most captive lorisids are housed solitarily or in pairs. Most individuals occupy a single exhibit space in a building dedicated to nocturnal animals. Facilities are commonly meeting recommendations for abiotic exhibit design and are providing animals with an enriched environment. However, pottos and slender lorises currently occupy exhibit spaces smaller than the recommended minimum, and the impact of cleaning protocols on olfactory communication should be critically evaluated. Few facilities are taking advantage of the benefits of positive reinforcement training for promoting animal welfare. Research is greatly needed on the effects of exhibit lighting on behavior, health, and reproduction; and to determine how best to manage the social needs of lorisids with naturally dispersed social structures. Although captive populations of slender lorises, pottos, and slow lorises are declining, we suggest that improved husbandry knowledge has the potential to positively influence population sustainability and to enhance future efforts to manage the growing pygmy loris population. Cytochrome c oxidase (COX) is a multi-subunit enzyme complex that catalyzes the final step of electron transfer through the respiratory
chain on the mitochondrial inner membrane. Up to 13 subunits encoded by both the mitochondrial (subunits I, II, and III) and
nuclear genomes occur in eukaryotic organisms ranging from yeast to human. Previously, we observed a high number of amino
acid replacements in the human COX IV subunit compared to mouse, rat, and cow orthologues. Here we examined COX IV evolution
in the two groups of anthropoid primates, the catarrhines (hominoids, cercopithecoids) and platyrrhines (ceboids), as well
as one prosimian primate (lorisiform), by sequencing PCR-amplified portions of functional COX4 genes from genomic DNAs. Phylogenetic analysis of the COX4 sequence data revealed that accelerated nonsynonymous substitution rates were evident in the early evolution of both catarrhines
and, to a lesser extent, platyrrhines. These accelerated rates were followed later by decelerated rates, suggesting that positive
selection for adaptive amino acid replacement became purifying selection, preserving replacements that had occurred. The evidence
for positive selection was especially pronounced along the catarrhine lineage to hominoids in which the nonsynonymous rate
was first faster than the synonymous rate, then later much slower. The rates of three types of ``neutral DNA' nucleotide
substitutions (synonymous substitutions, pseudogene nucleotide substitutions, and intron nucleotide substitutions) are similar
and are consistent with previous observations of a slower rate of such substitutions in the nuclear genomes of hominoids than
in the nuclear genomes of other primate and mammalian lineages.

目的:研究泌乳素(PRL)及其受体(PRLR)在乳房肥大和正常体积乳房中的表达情况,探讨乳腺组织中PRL、PRLR的表达与乳房肥大的相关性,为乳房肥大的治疗提供方向。方法:收集2013年1月~2013年4月第四军医大学西京医院26例乳房肥大患者和16例因单纯乳房下垂行乳房悬吊术患者的乳腺组织,采用免疫组织化学法检测乳腺组织标本中PRL和PRLR的表达。结果:PRL在乳房肥大组和正常体积乳房组乳腺组织中的阳性细胞率分别为88.46℅和56.25℅,两组比较差异有统计学意义(P0.05),且两组的PRL阳性的细胞面积、平均光密度值和累积光密度值均有统计学差异(P0.05)。乳房肥大组和正常体积乳房组乳腺组织中PRLR的阳性细胞率分别为76.92℅和75.00℅,两组比较无明显的统计学差异(P0.05),且两组间的PRLR阳性的细胞面积、平均光密度值、累积光密度值均无明显的统计学差异(P0.05)。结论:乳房肥大的发生可能与乳腺组织中泌乳素的表达升高有关,而与乳腺组织中泌乳素受体的表达无关。 相似文献

Cytochrome c (CYC) and 9 of the 13 subunits of cytochrome c oxidase (complex IV; COX) were previously shown to have accelerated rates of nonsynonymous substitution in anthropoid primates. Cytochrome b, the mtDNA encoded subunit of ubiquinol-cytochrome c reductase (complex III), also showed an accelerated nonsynonymous substitution rate in anthropoid primates but rate information about the nuclear encoded subunits of complex III has been lacking.We now report that phylogenetic and relative rates analysis of a nuclear encoded catalytically active subunit of complex III, the ironsulfur protein (ISP), shows an accelerated rate of amino acid replacement similar to cytochrome b. Because both ISP and subunit 9, whose function is not directly related to electron transport, are produced by cleavage into two subunits of the initial translation product of a single gene, it is probable that these two subunits of complex III have essentially identical underlying rates of mutation. Nevertheless, we find that the catalytically active ISP has an accelerated rate of amino acid replacement in anthropoid primates whereas the catalytically inactive subunit 9 does not. 相似文献

In this study, we identified two novel members of prolactin gene family in rat by blast searches against the published genomic database. A further analysis showed that gene duplications leading to PRL gene family in rodents occurred after rodents diverged from other mammals. Major reorganization of the gene loci in rodents was largely completed before the split of rat and mouse. But PL-I and PL-II genes are the exceptions, which have clustered in a species-specific manner in the phylogenetic tree. By combining results from gene conversion testing, relative chromosomal location comparison and estimated time for gene duplication, we believe that rodent PL-I and PL-II genes are species-specific and are the results of serial duplications which occurred after the divergence of mouse and rat. Our analysis also reveals that continual gene duplication and divergence occurred during the evolution of rodent PRL gene family. 相似文献

Evolution of the X-linked Zinc Finger Gene and the Y-linked Zinc Finger Gene in Primates 总被引:1,自引:0,他引:1
We have sequenced the partial exon of the zinc finger genes (ZFX and ZFY) in 5 hominoids, 2 Old World monkeys, 1 New World monkey, and 1 prosimian. Among these primate species, the percentage similarities of the nucleotide sequence of the ZFX gene were 96-100% and 91.2-99.7% for the ZFY gene. Of 397 sites in the ZFX and ZFY gene sequences, 20 for ZFX gene and 42 for ZFY gene were found to be variable. Substitution causes 1 amino acid change in ZFX, and 5 in ZFY, among 132 amino acids. The numbers of synonymous substitutions per site (Ks) between human and the chimpanzee, gorilla and orangutan for ZFY gene were 0.026, 0.033, and 0.085, respectively. In contrast, the Ks value between human and hominoid primates for the ZFX gene was 0.008 for each comparison. Comparison of the ZFX and ZFY genes revealed that the synonymous substitution levels were higher in hominoids than in other primates. The rates of synonymous substitution per site per year were higher in the ZFY exon than in the SRY exon, and higher in the ZFY exon than in the ZFY intron, in hominoid primates. 相似文献

ABSTRACTThe carotenoids constitute the most widespread class of pigments in nature. Most previous work has concentrated on the identification and characterization of their chemical physical properties and bioavailability. In recent years, significant amounts of research have been conducted in an attempt to analyze the genes and the molecular regulation of the genes involved in the biosynthesis of carotenoids. However, it is important not to lose sight of the early evolution of carotenoid biosynthesis. One of the major obstacles in understanding the evolution of the respective enzymes and their patterns of selection is a lack of a well-supported phylogenic analysis. In the present research, a major long-term objective was to provide a clearer picture of the evolutionary history of genes, together with an evaluation of the patterns of selection in algae. These phylogenies will be important in studies characterizing the evolution of algae. The gene sequences of the enzymes involved in the major steps of the carotenoid biosynthetic pathway in algae (cyanobacteria, rhofophyta, chlorophyta) have been analyzed. Phylogenetic relationships among protein-coding DNA sequences were reconstructed by neighbor-joining (NJ) analysis for the respective carotenoid biosynthetic pathway genes (crt) in algae. The analysis also contains an estimation of the rate of nonsynonymous nucleotide substitutions per nonsynonymous site (dN), synonymous nucleotide substitution per synonymous site (dS), and the ratio of nonsynonmous (dN/dS) for the test of selection patterns. The phylogenetic trees show that the taxa of some genera have a closer evolutionary relationship with other genera in some gene sequences, which suggests a common ancient origin and that lateral gene transfer has occurred among unrelated genera. The dN values of crt genes in the early pathway are relatively low, while those of the following steps are slightly higher, while the dN values of crt genes in chlorophyta are higher than those in cyanobacteria. Most of the dN/dS values exceed 1. The phylogenetic analysis revealed that lateral gene transfer may have taken place across algal genomes and the dN values suggest that most of the early crt genes are well conserved compared to the later crt genes. Furthermore, dN values also revealed that the crt genes of chlorophyta are more evolutionary than cyanobacteria. Sequences from nuclear mitochondrial pseudogenes (numts) that originated by transfer of genetic information from mitochondria to the nucleus offer a unique opportunity to compare different regimes of molecular evolution. Analyzing a 1621-nt-long numt of the rRNA specifying mitochondrial DNA residing on human chromosome 3 and its corresponding mitochondrial gene in 18 anthropoid primates, we were able to retrace about 40 MY of primate rDNA evolutionary history. The results illustrate strengths and weaknesses of mtDNA data sets in reconstructing and dating the phylogenetic history of primates. We were able to show the following. In contrast to numt-DNA, (1) the nucleotide composition of mtDNA changed dramatically in the different primate lineages. This is assumed to lead to significant misinterpretations of the mitochondrial evolutionary history. (2) Due to the nucleotide compositional plasticity of primate mtDNA, the phylogenetic reconstruction combining mitochondrial and nuclear sequences is unlikely to yield reliable information for either tree topologies or branch lengths. This is because a major part of the underlying sequence evolution model — the nucleotide composition — is undergoing dramatic change in different mitochondrial lineages. We propose that this problem is also expressed in the occasional unexpected long branches leading to the “common ancestor” of orthologous numt sequences of different primate taxa. (3) The heterogeneous and lineage-specific evolution of mitochondrial sequences in primates renders molecular dating based on primate mtDNA problematic, whereas the numt sequences provide a much more reliable base for dating.[Reviewing Editor: Dr. Rafael Zardoya] 相似文献

为了分析浙江省汉坦病毒分子流行病学的特点,本文应用分子生物学软件对1981~2007年从浙江省不同地区不同宿主分离的12株汉坦病毒的S、M基因序列进行分子进化分析,结果显示:浙江省属汉滩(HTN)型和汉城(SEO)型的混合型疫区,病毒的基因差异和亲缘远近关系主要表现在地区性,而与病毒分离的年代与宿主关系并不大,表现出明显的地理聚集现象,该现象在HTNV中的表现尤为明显,同一地区分离的病毒表现为较高的基因同源性,系统进化树上分布于同一或临近分支。另外发现分离自浙江省建德地区的Gou3和ZJ5株在SEOV进化枝中构成一独立分支,而且与国内外SEOV其他毒株的基因差异均较大,在浙江省建德地区存在着SOEV的特殊亚型病毒。 相似文献
